Background: Safety and efficacy data for dupilumab beyond 1 year are lacking for patients from Japan with moderate-to-severe asthma. Methods: The TRAVERSE open-label extension (OLE) study (NCT02134028) assessed the safety and efficacy of dupilumab 300 mg every 2 weeks up to 96 weeks in 2282 patients who completed a previous dupilumab asthma study. The primary endpoint was incidence of treatment-emergent adverse events (TEAEs). Secondary endpoints included annualized severe exacerbation rate and change from parent study baseline in pre-bronchodilator forced expiratory volume in 1 second (FEV1), asthma control, quality of life, and blood eosinophil levels. Anti-drug antibodies (ADA) were evaluated. We report results in 160 (7.8% of exposed population) patients recruited from Japanese centers with non-oral corticosteroid (OCS)-dependent asthma rolled over from two parent studies, and in subgroups with a type 2 inflammatory phenotype. Results: TEAEs were consistent with the parent studies and the known safety profile of dupilumab. One patient permanently discontinued treatment due to TEAEs. Exacerbation rates remained low and were sustained to Week 96, as were improvements in pre-bronchodilator FEV1. Rapid, sustained improvements were observed in dupilumab-treated patients who previously received placebo in a parent study, while further improvements in exacerbation rates, asthma control, and asthma-related quality of life were observed in those continuing dupilumab. Blood eosinophil levels decreased progressively while on treatment. Treatment-emergent ADA responses were highest in patients who had previously received placebo. Efficacy results were consistent in patients with a type 2 phenotype. Conclusions: Long-term dupilumab treatment was well tolerated and efficacious in patients with non–OCS-dependent, moderate-to-severe asthma recruited from Japan.(Funded by Sanofi and Regeneron Pharmaceuticals, Inc.; ClinicalTrials.gov identifier, NCT02134028)

Background Indication of omalizumab in the United States was recently extended to include pediatric (6–11 years) uncontrolled moderate-to-severe allergic asthma patients. Objective The purpose of this study was to describe baseline characteristics of this population from a real-world dataset. Methods Allergic asthma patients and uncontrolled moderate-to-severe allergic asthma patients, aged 6–11 years, were identified in the Allergy Partners Network Electronic Medical Records (2007–2016). The index date for allergic asthma patients was the latest between the second asthma-related visit and the allergic status confirmation. Uncontrolled moderate-to-severe allergic asthma patients were stratified into omalizumab-exposed (index date) or omalizumab-unexposed (index date randomly generated) groups. Characteristics were evaluated during the 12-month preindex period. Results A total of 5806 allergic asthma, 37 omalizumab-exposed, and 2620 omalizumab-unexposed patients were selected (mean age approximately 9 years). Allergic asthma and omalizumab-unexposed patients were predominantly white (70.2% and 61.2%) whereas the majority of omalizumab-exposed were African Americans (62.2%). Mean immunoglobulin E was 782.0 IU/ml in allergic asthma patients (available in 2.2%), 1134.4 IU/ml in omalizumab-exposed (available in 100.0%), and 746.1 IU/ml in omalizumab-unexposed (available in 3.1%). Allergic asthma patients were less severe than omalizumab-exposed and omalizumab-unexposed based on the forced expiratory volume in 1 s as a percentage of predicted value (FEV 1 % predicted) and the Childhood Asthma Control Test (C-ACT). Es incuestionable que sus comentarios, críticas e informaciones permiten en cierta forma la educación permanente de los públicos que leen esos contenidos, obligándoles al análisis y reflexión sobre los temas de actualidad. El problema se plantea en relación a la utilización de la prensa como medio de educación de los sectores de bajos niveles educativos y económicos, o sea precisamente de quienes están más necesitados de aprovechar los sistemas de difusión colectiva, para desarrollar un conocimiento de sí mismos y del ambiente circundante, lo cual podría entenderse como elevación del nivel cultural. Por lo tanto, la prensa en función de su papel propio, es medio de educación permanente. Si ahora no alcanza el ámbito que podría lograr se debe a las distorsiones de contenido y lenguaje.

To examine the association between body mass index (BMI) percentile and asthma in children 2–11 years of age, we performed a cross-sectional analysis of 853 Black and Hispanic children from a community-based sample of 2- to 11-year olds with measured heights and weights screened for asthma by the Harlem Children’s Zone Asthma Initiative. Current asthma was defined as parent/guardian-reported diagnosis of asthma and asthma-related symptoms or emergency care in the previous 12 months. Among girls, asthma prevalence increased approximately linearly with increasing body mass index (BMI) percentile, from a low of 12.0% among underweight girls (BMI ≤5th percentile) to a high of 33.3% among girls at risk for overweight (BMI 85th–94th percentile). Among boys, asthma prevalence was associated in a U-shaped curve with the extremes of BMI percentile, that is, 36.4% among underweight boys, 19.1% among normal weight boys (BMI 6th–84th percentile), and 34.8% among overweight boys (>95th percentile). After adjusting for age, race/ethnicity, and household smoking, among girls, having asthma was associated with being at risk for overweight (odds ratio [OR], 2.6; 95% confidence interval [CI], 1.4–5.0) and being overweight (OR, 2.1; 95% CI, 1.2–3.8) compared to normal weight; among boys, having asthma was associated both with overweight (OR, 2.4; 95% CI, 1.4–4.3) and with underweight (OR, 2.9; 95% CI, 1.1–7.7). Large, prospective studies that include very young children are needed to further explore the observed association between underweight and asthma among boys. The diseases caused byseveral Salmonella serovars in poultry results in important aconomical lossesfor this industrie in developing countries. The consumption of Salmonella contaminated poultry products by humanscan produce diarrhea; therefore Salmonella contaminated poultry products has become an important public health problem revealing the necesity to develop a vaccine against the different Salmonella serovars for poultry . In order to develope this vaccine will be necesary to obtein immunogens capable of inducing a protective respònse aginst these bacteria. In the present work we describe the purification and immunochemical characteristics of porins from Salmonella enterica serovar gallinarum, Salmonella enterica serovar pullorum and Salmonella enterica serovar enteritidis. The porins are proteins highly conserved of gram negative bacteria. They are found like homotrimers in the outer membrane, acting like difussion channels. Several experimental models had showned that these proteins are important antigens in the induction of host protective immune responses. The outer membrane proteins (OMP's) were isolated by solubilization of the cellular envelopes in a buffer of high ionic strength and detergent (0.3M NaCl, 2%SDS). This enriched porin preparation was further purified by gel filtration using a Sephacryl S-200 column; we obtained an Absorbance peak (280) that was analyzed for porin pressence by discontinuous poliacrylamide-SDS gels and by western blot ussing specific porin antibodies. We obtained two bands with 35 and 37 Kda that reacted with the specific antibodies. The S.enterica serovars porins showed homology between them and with the previously characterized porins fro S.typhi and S.typhimurium. By bidimensional electrophoresis we found out that each serovar porin has a particular bidimentional pattern, these porins were acidic proteins with isoelectric points between 4.7-5.5; this distinctive bidimentional pattern of each porin will allow us to issolate and sequence them in the near future, So far we have been able to purified and characterized porins from important poultry Salmonella enterica serovars than are being used as antigens to develope an specific vaccine.
