A layer of smear that coats the walls of root canals is produced by root canal instrumentation, which could be unfavorable to endodontic therapy. The endodontic irrigant SmearOFF is designed to effectively remove both the smear layer and bacteria concurrently. The objective of this study was to evaluate and compare the efficacy of SmearOFF and 17% EDTA in removing the smear layer across the coronal, middle, and apical thirds of root canals. Sixty‐four single‐canal mandibular premolar roots were chosen. Two irrigant protocols were separated into two sets of thirty‐two teeth, respectively, Group 1 (6% NaOCL/SmearOFF) and Group 2 (6% NaOCl/17% EDTA.) Until X2, the ProtaperNext rotary system (Dentsply, Maillefer, Switzerland), with a COXO C‐SMART Endomotor (Foshan COXO Medical instrument Co., Ltd., China) was utilized for the shaping of all teeth, the equipment settings were tuned to 300 revolutions per minute (rpm) and a torque of 3 Newton‐centimeters (Ncm). Before applying the final irrigants, an initial irrigation with 6% sodium hypochlorite (NaOCl) was performed using a 27‐G side‐vented needle. An ultrasonic gadget, EndoUltra, was utilized to activate the irrigation. After that, the determination of how well the proposed solutions worked on the prepared teeth was conducted by scanning electron microscopy. The mean smear layer scores were lower in all three regions (coronal, middle, and apical) using 17% EDTA in comparison with the samples treated with SmearOFF. Despite that, there were no significant differences between G1, 6% NaOCL/SmearOFF and G2, 6% NaOCL/17% EDTA in smear layer removal according to Kruskal–Wallis tests and Mann–Whitney U‐tests (p < 0.05). Considering the findings of this investigation, both 17% EDTA and SmearOFF serve as chelating agents, demonstrating the capability to effectively remove the smear layer. We previously examined the diachronic shifts in the narrative structure of research articles (RAs) and review manuscripts using abstract corpora from MEDLINE. This study employs Nini's Multidimensional Analysis Tagger (MAT) on the same datasets to explore five linguistic dimensions (D1–5) in these two sub-genres of biomedical literature, offering insights into evolving writing practices over 30 years. Analyzing a sample exceeding 1.2 million abstracts, we observe a shared reinforcement of an informational, emotionally detached tone (D1) in both RAs and reviews. Additionally, there is a gradual departure from narrative devices (D2), coupled with an increase in context-independent content (D3). Both RAs and reviews maintain low levels of overt persuasion (D4) while shifting focus from abstract content to emphasize author agency and identity. A comparison of linguistic features underlying these dimensions reveals often independent changes in RAs and reviews, with both tending to converge toward standardized stylistic norms. Systematic reviews are cumbersome yet essential to the epistemic process of medical science. Finding significant reports, however, is a daunting task because the sheer volume of published literature makes the manual screening of databases time-consuming. The use of Artificial Intelligence could make literature processing faster and more efficient. Sentence transformers are groundbreaking algorithms that can generate rich semantic representations of text documents and allow for semantic queries. In the present report, we compared four freely available sentence transformer pre-trained models (all-MiniLM-L6-v2, all-MiniLM-L12-v2, all-mpnet-base-v2, and All-distilroberta-v1) on a convenience sample of 6110 articles from a published systematic review. The authors of this review manually screened the dataset and identified 24 target articles that addressed the Focused Questions (FQ) of the review. We applied the four sentence transformers to the dataset and, using the FQ as a query, performed a semantic similarity search on the dataset. The models identified similarities between the FQ and the target articles to a varying degree, and, sorting the dataset by semantic similarities using the best-performing model (all-mpnet-base-v2), the target articles could be found in the top 700 papers out of the 6110 dataset. Our data indicate that the choice of an appropriate pre-trained model could remarkably reduce the number of articles to screen and the time to completion for systematic reviews. Granular cell tumors of the neurohypophysis (GCT) are rare benign neoplasms belonging, along with pituicytoma and spindle cell oncocytoma, to the family of TTF1‐positive low‐grade neoplasms of the posterior pituitary gland. GCT usually present as a solid sellar mass, slowly growing and causing compressive symptoms over time, occasionally with suprasellar extension. They comprise polygonal monomorphous cells with abundant granular cytoplasm, which is ultrastructurally filled with lysosomes. Here we report the case of a GCT presenting as a third ventricle mass, radiologically mimicking chordoid glioma, with aberrant expression of GFAP and Annexin‐A, which lends itself as an example of an integrated diagnostic approach to sellar/suprasellar and third ventricle masses. Aim: Bibliometric surveys are time-consuming endeavors, which cannot be scaled up to meet the challenges of ever-expanding fields, such as bone regeneration. Artificial intelligence, however, can provide smart tools to screen massive amounts of literature, and we relied on this technology to automatically identify research topics. Materials & methods: We used the BERTopic algorithm to detect the topics in a corpus of MEDLINE manuscripts, mapping their similarities and highlighting research hotspots. Results: Using BERTopic, we identified 372 topics and were able to assess the growing importance of innovative and recent fields of investigation such as 3D printing and extracellular vescicles. Conclusion: BERTopic appears as a suitable tool to set up automatic screening routines to track the progress in bone regeneration. [ABSTRACT FROM AUTHOR]

Featured Application: A polynucleotide-hyaluronic acid compound represents a promising tool to improve soft tissue regeneration. More advanced 3D culture models are required to better characterize its effects on target cells. (1) Background: Three-dimensional cultures are useful tools to evaluate regenerative approaches in vitro, as they may mimic the spatial arrangement of cells more closely to natural tissues than routine 2D culture methods. (2) Methods: We investigated the effects of a polynucleotide, hyaluronic acid (PN, HA) compound on 3D spheroid cultures of primary gingival fibroblasts, by measuring their morphology over time, cell viability with Calcein-AM, a fluorescent marker, and cell growth potential by re-plating spheroids in attachment-permissive regular culture plates under routine conditions and following them up for 15 days. (3) Results: PN + HA induced an increase in spheroid size and perimeter and a decrease in spheroid circularity, as cells tended to grow and form small peripheral stacks around the spheroid. Levels of cell viability were also higher in this group. After re-plating, only the spheroids previously stimulated with PN + HA dissolved completely during the second week of culture and colonized the plate, thus indicating the retention of a higher level of viability by the cells forming the whole spheroid with this stimulus. (4) Conclusions: Taken together, our data support the idea that the combination of PN and HA has synergic effects on primary fibroblasts and promotes their viability, the growth of 3D cellular constructs, and the retention of a remarkable proliferative potential over the course of the experimental period, making it a promising compound for further investigations. Primary large B-cell lymphoma of immune-privileged sites (IP-LBCL) is a rare malignant hematological neoplasm. Involvement of the cerebellum is even rarer and its diagnosis is often difficult to make due to its non-specific clinical and radiological presentation. Methods: We reported 3 cases of cerebellar IP-LBCL followed at our hospital and reviewed the medical literature to unravel the peculiarities of this poorly studied entity. Outcomes: Analyzing our cases and reviewing the literature, we could collect and study 26 cases of cerebellar IP-LBCL. To the best of our knowledge, this is the largest cohort of such patients currently published. Conclusion: Cerebellar IP-LBCL presents more often in adult females with cerebellum-related focal neurological signs such as ataxia, headache, and nausea. Histological confirmation is mandatory for a correct diagnosis and treatment and all cases feature diffuse large B-cell lymphoma histopathology. Compared to other encephalic IP-LBCL, cerebellar cases seem to include a higher number of cases with germinal center B-cell phenotype and better survival. These differences may be related to a different immune microenvironment and especially immunoregulation that distinguishes the cerebellum from other areas of the CNS. [ABSTRACT FROM AUTHOR]

The aminoxyl radical benzotriazole-N-oxyl (BTNO) is generated by the oxidation of 1-hydroxybenzotriazole (HBT) with a Ce(super IV) salt and characterized by spectrophotometry, EPR, cyclic voltammetry and laser flash photolysis. Rate constants of H-abstraction (k(sub h)) from a number of H-donor substrates by BTNO are determined spectroscopically.

Following a request from five European Nordic countries, the EFSA Panel on Nutrition, Novel Foods and Food Allergens (NDA) was tasked to provide scientific advice on a tolerable upper intake level (UL) or a safe level of intake for dietary (total/added/free) sugars based on available data on chronic metabolic diseases, pregnancy‐related endpoints and dental caries. Specific sugar types (fructose) and sources of sugars were also addressed. The intake of dietary sugars is a well‐established hazard in relation to dental caries in humans. Based on a systematic review of the literature, prospective cohort studies do not support a positive relationship between the intake of dietary sugars, in isocaloric exchange with other macronutrients, and any of the chronic metabolic diseases or pregnancy‐related endpoints assessed. Based on randomised control trials on surrogate disease endpoints, there is evidence for a positive and causal relationship between the intake of added/free sugars and risk of some chronic metabolic diseases: The level of certainty is moderate for obesity and dyslipidaemia (> 50–75% probability), low for non‐alcoholic fatty liver disease and type 2 diabetes (> 15–50% probability) and very low for hypertension (0–15% probability). Health effects of added vs. free sugars could not be compared. A level of sugars intake at which the risk of dental caries/chronic metabolic diseases is not increased could not be identified over the range of observed intakes, and thus, a UL or a safe level of intake could not be set. Based on available data and related uncertainties, the intake of added and free sugars should be as low as possible in the context of a nutritionally adequate diet. Decreasing the intake of added and free sugars would decrease the intake of total sugars to a similar extent. This opinion can assist EU Member States in setting national goals/recommendations. Periodontal regeneration is a complex goal, which is commonly pursued with a combination of surgical techniques, biomaterials, and bioactive compounds. One such compound is enamel matrix derivative (EMD), a medical substance that is extracted from porcine tooth germs and which contains several protein fractions with BMP- and TGF-β-like action. Activation of TGF-β signaling is required for EMD activity on cells and tissues, and a growing body of evidence indicates that EMD largely relies on this pathway. As low frequency electromagnetic fields (EMFs) have long been investigated as a tool to promote bone formation and osteoblast activity, and because recent studies have reported that the effects of EMFs on cells require primary cilia, by modulating the presence of membrane-bound receptors (e.g., for BMP) or signal mediators, it can be hypothesized that the application of EMFs may increase cell sensitivity to EMD: as TGFBR receptors have also been identified on primary cilia, EMFs could make cells more responsive to EMD by inducing the display of a higher number of receptors on the cellular membrane. A study was conducted to analyze competition mechanisms in the photochemical cleavage of the C-X bond of aryl-substituted vinyl halides. The Rayonet RPR-100 reactor was utilized to carry out the photochemical reactions. Reactions at 254 nm were performed in a quartz cuvette supporting a magnetic ministirrer. In addition, an analysis of the stabilizing influence resulting from alpha-substituents in ethenyl cation structures is presented.

The stereochemical characterisitics of the vinylic SRN1(V) reaction of 2-anisyl-1,2-diphenylvinyl bromide was analyzed to determine the properties of the vinyl radical intermediate. The photostimulated SRn1(V) reaction is characteized by electron-transfer steps that forms an intermediate radical with a bridged or bent and configurationally stable structure. Furthermore, SRN1(V) propagation also involves the electron-transfer-induced hydrodehalogenation during the formation of the vinyl radical.

The structure of alpha-substituted vinyl radicals was analyzed via ab initio DFT molecular calculations to characterize the effects of substituents on the linear or bent configuration of the radicals. DFT analysis of alpha-substituted vinyl radical bearing sigma-type substituents indicated the presence of linear pi-type substituents. The electronegativity of the substituents was also enhanced by the inversion barrier of the E and Z forms of vinyl radicals bearing sigma-type substituents.

Stereochemical results of a SRN1(V) reaction were observed to determine properties of the vinyl radical intermediate using E/Z isomeric precursors. Such an intermediate can be found in the vinylic SRN1 reaction (SRN1(V)) of 2-anisyl-1,2-diphenylvinyl bromide 2. The reaction establishes the presence of a beta-substituted alpha-phenylvinyl radical as a reactive intermediate with specific linear structure.
