Bile acids, the products of concerted host and gut bacterial metabolism, have important signaling functions within the mammalian metabolic system and a key role in digestion. Given the complexity of the mega-variate bacterial community residing in the gastrointestinal tract, studying associations between individual bacterial genera and bile acid processing remains a challenge. Here, we present a novel in vitro approach to determine the bacterial genera associated with the metabolism of different primary bile acids and their potential to contribute to inter-individual variation in this processing. Anaerobic, pH-controlled batch cultures were inoculated with human fecal microbiota and treated with individual conjugated primary bile acids (500 μg/ml) to serve as the sole substrate for 24 h. Samples were collected throughout the experiment (0, 5, 10, and 24 h) and the bacterial composition was determined by 16S rRNA gene sequencing and the bile acid signatures were characterized using a targeted ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) approach. Data fusion techniques were used to identify statistical bacterial-metabolic linkages. An increase in gut bacteria associated bile acids was observed over 24 h with variation in the rate of bile acid metabolism across the volunteers (n = 7). Correlation analysis identified a significant association between the Gemmiger genus and the deconjugation of glycine conjugated bile acids while the deconjugation of taurocholic acid was associated with bacteria from the Eubacterium and Ruminococcus genera. A positive correlation between Dorea and deoxycholic acid production suggest a potential role for this genus in cholic acid dehydroxylation. A slower deconjugation of taurocholic acid was observed in individuals with a greater abundance of Parasutterella and Akkermansia. This work demonstrates the utility of integrating compositional (metataxonomics) and functional (metabonomics) systems biology approaches, coupled to in vitro model systems, to study the biochemical capabilities of bacteria within complex ecosystems. Characterizing the dynamic interactions between the gut microbiota and the bile acid pool enables a greater understanding of how variation in the gut microbiota influences host bile acid signatures, their associated functions and their implications for health.

A newly-described syndrome called Aneurysm-Osteoarthritis Syndrome (AOS) was recently reported. AOS presents with early onset osteoarthritis (OA) in multiple joints, together with aneurysms in major arteries, and is caused by rare mutations in SMAD3. Because of the similarity of AOS to idiopathic generalized OA (GOA), we hypothesized that SMAD3 is also associated with GOA and tested the hypothesis in a population-based cohort.Study participants were derived from the Chingford study. Kellgren-Lawrence (KL) grades and the individual features of osteophytes and joint space narrowing (JSN) were scored from radiographs of hands, knees, hips, and lumbar spines. The total KL score, osteophyte score, and JSN score were calculated and used as indicators of the total burden of radiographic OA. Forty-one common SNPs within SMAD3 were genotyped using the Illumina HumanHap610Q array. Linear regression modelling was used to test the association between the total KL score, osteophyte score, and JSN score and each of the 41 SNPs, with adjustment for patient age and BMI. Permutation testing was used to control the false positive rate.A total of 609 individuals were included in the analysis. All were Caucasian females with a mean age of 60.9±5.8. We found that rs3825977, with a minor allele (T) frequency of 20%, in the last intron of SMAD3, was significantly associated with total KL score (β = 0.14, Ppermutation = 0.002). This association was stronger for the total JSN score (β = 0.19, Ppermutation = 0.002) than for total osteophyte score (β = 0.11, Ppermutation = 0.02). The T allele is associated with a 1.47-fold increased odds for people with 5 or more joints to be affected by radiographic OA (Ppermutation = 0.046).We found that SMAD3 is significantly associated with the total burden of radiographic OA. Further studies are required to reveal the mechanism of the association.

Prisons are increasingly looking for localised, innovative and collaborative approaches to address rehabilitation and full recovery from substance misuse. This article presents the findings from an evaluation of the Master Gardener (MG) programme, a gardening intervention with substance misuse offenders at HMP Rye Hill. Whilst the extension of the MG programme to a prison setting recognises a range of positive outcomes associated with the role of horticulture in supporting wellbeing, it also reflects Rye Hill's move towards the development of a dedicated Recovery Unit, offering a suite of interventions to support substance misusing offenders. The MG programme at Rye Hill demonstrates an innovative and successful partnership, working with the charity Garden Organic, Public Health Northamptonshire and the Drug and Alcohol Recovery Team (DART), using horticulture as a means to address recovery. This paper sets out the evaluation's aims and objectives, methodological approach, key findings and conclusions which include a number of recommendations. The approach taken has allowed for an examination of the process and experiences from multiple perspectives of the MG programme within a prison setting. As well as focusing on the impacts of the programme, the article reflects on gardening as an embodied practice and the garden as a space that promotes humanisation and self-worth, community, a connection to nature and a longer term, holistic approach to recovery. Processed potato (Solanum tuberosum L.) products, such as chips and French fries, contribute to the dietary intake of acrylamide, a suspected human carcinogen. One of the most promising approaches for reducing its consumption is to develop and commercialize new potato varieties with low acrylamide-forming potential. To facilitate this effort, a National Fry Processing Trial (NFPT) was conducted from 2011 to 2013 in five states. More than 140 advanced breeding lines were evaluated for tuber agronomic traits and biochemical properties from harvest through 8 mo of storage. Thirty-eight and 29 entries had significantly less acrylamide in French fries than standard varieties Russet Burbank and Ranger Russet, with reductions in excess of 50%, after one and 8 mo of storage, respectively. As in previous studies, the glucose content of raw tubers was predictive of acrylamide in finished French fries (R2 = 0.64-0.77). Despite its role in acrylamide formation, tuber free asparagine was not significant, potentially because it showed relatively little variation in the NFPT population. Even when glucose was included in the model as a covariate, genotype was highly significant (p = 0.001) for predicting acrylamide, indicating there may be yet-unidentified genetic loci to target in breeding. The NFPT has demonstrated that there exist many elite US breeding lines with low acrylamide-forming potential. Purpose – Desistance is increasingly conceptualised as a theoretical construct which is used to explain how offenders orient themselves away from committing crimes. Previous studies suggest that successful desistance occurs due to one or a number of factors. These factors include things such as: faith (Giordano et al., 2007); a rite of passage (Maruna, 2010); gender (Giordano et al., 2002); psychosocial processes (Healey, 2010); personal and social circumstances which are space and place specific (Flynn, 2010); ethnicity and faith (Calverley, 2013); race and racialisation (Glynn, 2014). However, to date there has been little work undertaken to examine how notions of “intersectionality” may be a more appropriate theoretical lens through which to locate and contextualise the understandings of desistance when looking at marginalised populations such as black offenders. Intersectionality is an understanding of human beings as shaped by the interaction of different social locations. These interactions occur within a context of connected systems and structure of power. Through such processes independent forms of privilege and oppression are created. The paper aims to discuss these issues. Design/methodology/approach – The paper concludes with a perspective that envisions moving towards an “intersectional model of desistance for marginalised groups such as black offenders”. Findings – It is the author’s view that the development of a intersectional model of desistance for black offenders may begin a dialogue that further pushes the study of re-entry and desistance into an area that transcends the criminal justice system and locates itself firmly within the civil and human rights of black offenders, and indeed, offenders as a whole. It is hoped that by using intersectional approaches when conducting inquiries we will be able to lead towards eradicating multiple oppressions faced by so many sections of the offender populations and the communities they come from. Research limitations/implications – This paper is positional inasmuch as it attempts to establish some, principled arguments to advance the study of desistance. Therefore, a testing of the views expressed in the paper is required. Practical implications – It is the author’s contention that for those black offenders who desire to quit crime, there needs to be networks and activities that not only support their desire to desist, but a radical reframing of how interlocking oppressions that render them subordinate must become a key driver for the desistance project. How can/do black offenders acquire and tell their own authentic narrative when it has been shaped by a history of oppression? It is therefore right to assume that meaningful reintegration of black offenders back into communities requires a deeper commitment to culturally competent rehabilitative processes, that could lead towards a culture of desistance. Social implications – An “Intersectional Model of Desistance” also needs to challenge some white criminologists’ claims by validating the black contribution to criminological theorising. This position should embrace and include perspectives which unify theoretical positions that validate interlocking oppressions; racism, poverty, ethno-cultural group membership, etc., where the broader distribution of opportunities across society, and the ability to recognise them as such as opportunities for black men to desist are taken into consideration. As part of a process of rehabilitation, black offenders need to be engaged with intersectional institutional processes and practices that will lead to a challenge of their criminal values and behaviour, designed to increase their capacity to consider desistance. It is hoped that by using intersectional approaches when conducting inquiries we will be able to eradicate multiple oppressions faced by so many sections of the offender populations and the communities they come from. Originality/value – Understanding how the experiences of black offenders, are impacted by examining interlocking oppressions of criminal justice processes; police, courts, incarceration, probation, etc., would critically assess how these intersections enhances or impedes the desistance trajectories of black offenders, in relation to offenders as a whole. As much of black offender lived reality centres on having to contend, not just with criminal justice process, but the additional oppression of racialisation, the outcomes become more heavily context dependent and driven.
