Introduction: About 50% of cutaneous melanoma (CM) patients present activating BRAF mutations that can be effectively targeted by BRAF inhibitors (BRAFi). However, 20% of CM patients exhibit intrinsic drug resistance to BRAFi, while most of the others develop adaptive resistance over time. The mechanisms involved in BRAFi resistance are disparate and globally seem to rewire the cellular signaling profile by up-regulating different receptor tyrosine kinases (RTKs), such as the epidermal growth factor receptor (EGFR). RTKs inhibitors have not clearly demonstrated anti-tumor activity in BRAFi resistant models. To overcome this issue, we wondered whether the shared up-regulated RTK phenotype associated with BRAFi resistance could be exploited by using immune weapons as the antibody-dependent cell cytotoxicity (ADCC)-mediated effect of anti-RTKs antibodies, and kill tumor cells independently from the mechanistic roots. Methods and results: By using an in vitro model of BRAFi resistance, we detected increased membrane expression of EGFR, both at mRNA and protein level in 4 out of 9 BRAFi-resistant (VR) CM cultures as compared to their parental sensitive cells. Increased EGFR phosphorylation and AKT activation were observed in the VR CM cultures. EGFR signaling appeared dispensable for maintaining resistance, since small molecule-, antibody- and CRISPR-targeting of EGFR did not restore sensitivity of VR cells to BRAFi. Importantly, immune-targeting of EGFR by the anti-EGFR antibody cetuximab efficiently and specifically killed EGFR-expressing VR CM cells, both in vitro and in humanized mouse models in vivo, triggering ADCC by healthy donors’ and patients’ peripheral blood cells. Conclusion: Our data demonstrate the efficacy of immune targeting of RTKs expressed by CM relapsing on BRAFi, providing the proof-of-concept supporting the assessment of anti-RTK antibodies in combination therapies in this setting. In this letter, the implementation of a system-on-chip for the documentation of road accidents is presented. Key features of the system are the implementation on a programmable architecture of a compression algorithm capable of encoding up to 20 black and white QCIF frames/s, and the computation of a digital signature performed every frame which is applied to the encoded bitstream certifying the source of the video sequence. The acquired images are then stored on a battery of on-chip flash memories that can be used to retrieve accident information to be used in legal confrontations. Having to perform in critical energy conditions (e.g., continued image acquisition for up to 10-20 s after an accident has occurred), the system was designed to minimize energy consumption at all levels. The system-on-chip has been implemented in 6 x 6 [mm.sup.2] on a 0.25-[micro]m 6-metal standard-cell CMOS technology and works at 40 MHz with a 2.5-V power supply. Performance can be decreased to 12 QCIF frames/s, 24 MHz, and 1.3-V power supply in order to achieve 30-mW power consumption. Background: At present, the prognostic significance of programmed cell death receptor ligand 1 (PD-L1) expression in oropharyngeal squamous cell carcinoma (OPSCC) patients is still controversial. In this study, we aim to synthesize relevant studies that have assessed the prognostic value of PD-L1 in patients with primary OPSCC treated according to the current standard-of-care. Methods: A systematic search of Medline/PubMed, Cochrane, Embase, Web of Science, and Scopus was conducted to define the prognostic role of PD-L1 expression in OPSCC. All studies published before July 31, 2021 were screened. Summary hazard ratios (sHR) with 95% confidence intervals (CIs) were calculated using a random-effects model. Results: A total of 1522 OPSCC patients from 12 studies were included. PD-L1 expression in OPSCC tumor cells (TCs) was significantly associated with longer overall survival (sHR=0.63, 95% CI 0.50-0.79), and progression-free survival (sHR=0.62, 95% CI 0.49-0.79). A benefit in survival was also observed in PD-L1-positive OPSCC patients who underwent surgery (sHR=0.34, 95% CI 0.18-0.65). Finally, although PD-L1-positive expression was related to better outcomes both in HPV-negative and HPV-positive OPSCC, the difference reached the statistical significance only in the HPV-positive subgroup (sHR=0.37, 95% CI 0.19-0.73). No heterogeneity emerged between studies for all considered outcomes, with I 2 ranging from 0% for progression-free survival to 11% for overall survival. Conclusions: PD-L1 expression on TCs associated with improved survival in OPSCC. In particular, HPV-positive OPSCC most benefited from PD-L1 expression when compared to the PD-L1 negative counterpart. In this work we investigate techniques for embedding domain-specific spatial invariances into highly-constrained neural networks. This information is used to drastically reduce the number of weights which have to be determined during the learning phase, thus allowing us to apply artificial neural networks to problems characterized by a relatively small number of available examples. As an application of the proposed methodology, we study the problem of optical inspection of machined parts. More specifically, we have characterized the performance of a network created according to this strategy, which accepts images of parts under inspection at its input and issues a flag at its output which states whether the part is defective. The results obtained so far show that the proposed methodology provides a potentially relevant approach for the quality control of industrial parts, as it offers both accuracy and short software development time, when compared with a classifier implemented using a standard approach.

This paper presents an open source framework called Creamino. It consists of an Arduino-based cost-effective quick-setup EEG platform built with off-the-shelf components and a set of software modules that easily allow users to connect this system to Simulink or BCI-oriented tools (such as BCI2000 or OpenViBE) and set up a wide number of neuroscientific experiments. Creamino is capable of processing multiple EEG channels in real-time and operates under Windows, Linux, and Mac OS X in real-time on a standard PC. Its objective is to provide a system that can be readily fabricated and used for neurophysiological experiments and, at the same time, can serve as the basis for development of novel BCI platforms by accessing and modifying its open source hardware and software libraries. Schematics, gerber files, bill of materials, source code, software modules, demonstration videos, and instructions on how to use these modules are available free of charge for research and educational purposes online at https://github.com/ArcesUnibo/creamino. Electrical impedance tomography is a noninvasive and cost-effective imaging method that is increasingly attractive in the field of medical diagnostics. Several health conditions, such as stroke and solid tumors, are characterized by compact conductivity anomalies surrounded by a fairly regular background. Commonly employed voxel-by-voxel reconstruction methods for impedance imaging share the disadvantages of high computational cost and substantial sensitivity to measurement noise and imperfections in the electrical model describing the domain of interest. We present a special purpose algorithm for automatic detection and identification of compact conductivity variations. The technique exploits a priori structural information and, by reconstructing only the limited number of parameters required to describe a compact conductivity contrast, does not depend on a critical regularization parameter. The most demanding kernels are implemented to run on graphics processing units to accelerate computation. The parametric reconstruction is quicker and more robust than widely employed approaches with respect to measurement noise and imperfections in the electrical model, as shown by computational analysis performed on a segmented head domain and experimental measurements acquired on a cylindrical phantom. When the goal is quick detection of compact conductivity contrasts in complex 3-D domains, the inclusion of specific constraints relating to the problem considered leads to enhanced quality of reconstruction, making the presented technique a promising alternative to common voxel-by-voxel reconstruction methods. The paper presents a novel Driving Right Leg (DgRL) circuit designed to mitigate the effect of common mode signals deriving, say, from power line interferences. The DgRL drives the isolated ground of the instrumentation towards a voltage which is fixed with respect to the common mode potential on the subject, therefore minimizing common mode voltage at the input of the front-end. The paper provides an analytical derivation of the common mode rejection performances of DgRL as compared to the usual grounding circuit or Driven Right Leg (DRL) loop. DgRL is integrated in a bio-potential acquisition system to show how it can reduce the common mode signal of more than 70 dB with respect to standard patient grounding. This value is at least 30 dB higher than the reduction achievable with DRL, making DgRL suitable for single-ended front-ends, like those based on active electrodes. EEG signal acquisition is performed to show how the system can successfully cancel power line interference without any need for differential acquisition, signal post-processing or filtering. This paper presents a power-aware scheduling algorithm based on efficient distribution of the computing workload to the resources on heterogeneous CPU-GPU architectures. The scheduler manages the resources of several computing nodes with a view to reducing the peak power. The algorithm can be used in concert with adjustable power state software services in order to further reduce the computing cost during high demand periods. Although our study relies on GPU workloads, the approach can be extended to other heterogeneous computer architectures. The algorithm has been implemented in a real CPU-GPU heterogeneous system. Experiments prove that the approach presented reduces peak power by 10 percent compared to a system without any power-aware policy and by up to 24 percent with respect to the worst case scenario with an execution time increase in the range of 2 percent. The IC presented integrates the front-end for EEG and Electrical Impedance Tomography (EIT) acquisition on the electrode, together with electrode-skin contact impedance monitoring and EIT current generation, so as to improve signal quality and integration of the two techniques for brain imaging applications. The electrode size is less than 2 cm^2 and only 4 wires connect the electrode to the back-end. The readout circuit is based on a Differential Difference Amplifier and performs single-ended amplification and frequency division multiplexing of the three signals that are sent to the back-end on a single wire which also provides power supply. Since the system's CMRR is a function of each electrode's gain accuracy, an analysis is performed on how this is influenced by mismatches in passive and active components. The circuit is fabricated in 0.35 \mu m CMOS process and occupies 4 mm^2, the readout circuit consumes 360 \mu W, the input referred noise for bipolar EEG signal acquisition is 0.56 \mu VRMS between 0.5 and 100 Hz and almost halves if only EEG signal is acquired. The computing demand of many signal processing algorithms is dramatically growing because of the increasing complexity of embedded software applications. Concurrently, as process technology scales, the design effort for realizing very large scale integrated circuits and the associated costs are becoming critically high. A possible solution to address this performance/costs challenge is given by customizable multiprocessor system-on-chips. The approach proposed in this paper leads to the customization of multi/many processor system-on-chip at two levels of abstraction: 1) customization through application-specific hardware accelerators implemented on configurable datapath that can target three kinds of structured application-specific integrated circuit technologies: metal, via, and runtime programmable and 2) customization of the architectural parameters of the platform. The proposed platform is equipped with a design framework that assists the user in the high-level design-space exploration of signal processing applications described using the Open Computing Language (OpenCL) language. A peculiar added value of the flow is to support the migration of OpenCL kernels and tasks into pipelined hardware accelerators described using a C-level language. The platform is able to provide an average performance of 90 GOPS on a set of reference signal processing applications, and an average computational energy efficiency of 130 GOPS/W in its metal-programmable configuration. This result shows the benefits in terms of energy efficiency of hardware customization applied to multiprocessor systems with respect to many core devices such as general-purpose graphic processing units, able to provide on average 2.5 GOPS/W for the applications under analysis. This paper describes the application space exploration of a heterogeneous digital signal processor with dynamic reconfiguration capabilities. The device is built around three reconfigurable engines featuring different flavours and computation granularities that make it suitable for a wide range of signal processing application domains such as video coding, image processing, telecommunications, and cryptography. Performance of signal processing applications is evaluated from measurements performed on a CMOS 90 nm prototype. In order to characterize the application space of the processor, performance is compared with state-of-the-art devices, taking programmability, computational capabilities, and energy efficiency as the main metrics. The device exploits performance and energy efficiency significantly more than general purpose processors, while still maintaining a user-friendly programming approach that mainly relies on software-oriented languages. The device is able to achieve 1.2 to 15 GOPS with an energy efficiency from 2 to 50 GOPS/W when running the selected applications. [ABSTRACT FROM PUBLISHER]

Background: The redox-silent vitamin E analog αtocopheryl succinate (αTOS) was found to synergistically cooperate with vitamin K3 (VK3) plus ascorbic acid (AA) in the induction of cancer cell-selective apoptosis via a caspase-independent pathway. Here we investigated the molecular mechanism(s) underlying cell death induced in prostate cancer cells by αTOS, VK3 and AA, and the potential use of targeted drug combination in the treatment of prostate cancer. Methodology/Principal Findings: The generation of ROS, cellular response to oxidative stress, and autophagy were investigated in PC3 prostate cancer cells by using drugs at sub-toxic doses. We evaluated whether PARP1-mediated apoptosis-inducing factor (AIF) release plays a role in apoptosis induced by the combination of the agents. Next, the effect of the combination of αTOS, VK3 and AA on tumor growth was examined in nude mice. VK3 plus AA induced early ROS formation associated with induction of autophagy in response to oxidative stress, which was reduced by αTOS, preventing the formation of autophagosomes. αTOS induced mitochondrial destabilization leading to the release of AIF. Translocation of AIF from mitochondria to the nucleus, a result of the combinatorial treatment, was mediated by PARP1 activation. The inhibition of AIF as well as of PARP1 efficiently attenuated apoptosis triggered by the drug combination. Using a mouse model of prostate cancer, the combination of αTOS, VK3 and AA was more efficient in tumor suppression than when the drugs were given separately, without deleterious side effects. Conclusions/Significance: αTOS, a mitochondriαtargeting apoptotic agent, switches at sub-apoptotic doses from autophagy-dependent survival of cancer cells to their demise by promoting the induction of apoptosis. This work deals with the solution of large non-Hermitian linear systems on desktop workstations with multiple graphics processing units (GPUs). While our implementation is motivated by the need to accelerate volume conductor modeling for bioelectrical brain imaging, the problem itself is common in scientific computing. Whenever a complex partial differential equation is numerically solved, a typically non-Hermitian sparse complex linear system needs to be solved. For problem sizes in the millions, this can take a long time even with highly optimized CPU-based solvers. Our GPU-accelerated solver outperforms an optimized OpenMP-based reference running on two quad-core CPUs by a factor of up to 31× in single precision and up to 7× in double precision, at the cost of a very modest hardware upgrade of two dual-GPU GTX 295 graphics cards. Electrical impedance tomography (EIT) is an imaging technology based on impedance measurements. To retrieve meaningful insights from these measurements, EIT relies on detailed knowledge of the underlying electrical properties of the body. This is obtained from numerical models of current flows therein. The nonhomogeneous and anisotropic electric properties of human tissues make accurate modeling and simulation very challenging, leading to a tradeoff between physical accuracy and technical feasibility, which at present severely limits the capabilities of EIT. This work presents a complete algorithmic flow for an accurate EIT modeling environment featuring high anatomical fidelity with a spatial resolution equal to that provided by an MRI and a novel realistic complete electrode model implementation. At the same time, we demonstrate that current graphics processing unit (GPU)-based platforms provide enough computational power that a domain discretized with five million voxels can be numerically modeled in about 30 s. A four-shell head phantom has been built and characterized. Its structure is similar to that of nonhomogeneous concentric shell domains used by numerical solvers that better approximate current distribution than phantoms currently used to validate electrical impedance tomography systems. Each shell represents a head tissue, namely, skin, skull, cerebrospinal fluid, and brain. A novel technique, which employs a volume conductive impermeable film, has been implemented to prevent ion diffusion between different agar regions without affecting current distribution inside the phantom. Comparisons between simulations and phantom measurements performed over four days are given to prove both the adherence to the model in the frequency range between 10 kHz and 1 MHz and its long-term stability. An architecture to compensate the voltage attenuation introduced by 3-D capacitive coupling is proposed. The scheme is based on a calibration channel which sets the gain of the variable gain amplifiers of the signal channels in such a way as to compensate for the voltage attenuation. Based on this architecture, a prototype has been designed aimed at demonstrating that 3-D technology based on capacitive coupling allows one to transmit analog signals as well as digital ones. CMOS 90 nm technology was used and 3-D assembly is done at die level using a face to face stacking procedure. The area of each signal channel and of the calibration channel is 90\, \times \,30 \mum^2 and 138\,\times \,191 \mum^2, respectively, with a power consumption of 1 mW and 3.6 mW. This paper describes a System on Chip implementation of a reconfigurable digital signal processor. The device is suitable for execution of a wide range of applications exploiting a balanced mix of heterogeneous reconfigurable fabrics merged together by a flexible and efficient communication infrastructure based on a 64-bit Network On Chip. The SoC combines a fine grain embedded FPGA, a mid grain configurable processor and a coarse grain reconfigurable array. An ARM processor featuring a resident operating system is the SoC supervisor, managing communication, synchronization and reconfiguration mechanisms. This computational model enables the programmer to manage the high level synchronization and global data of complex signal processing applications through the ARM processor, while allocating most critical computational kernels to the most suitable reconfigurable engines. The SoC has been fabricated in 90-nm technology, the die area being 110 mm²; it integrates 97 million transistors and has a peak power consumption of 2.5 W. In order to demonstrate the proposed computational model and the reconfigurable signal processor capabilities in a real test case, a video surveillance motion detection application was implemented in the SoC. This review paper summarizes some of the most challenging issues regarding the development of electronic microsystems for the isolation, manipulation, and characterization of rare cells. Two relevant areas, namely, immunology for cancer therapy and monitoring of microbial contaminants in water, are presented as example applications which require handling of rare cells. Starting from these applications, the state of the art in electronic microsystem research is presented, and various solutions are discussed, highlighting the advantages and disadvantages of each technology and suggesting the need to integrate multiple solutions to meet the application requirements. This article discusses a study on the development of laboratory-on-a-chip (lab-on-a-chip) devices that are able to manipulate microparticles and cells. The study used laboratory-on-a-chip (lab-on-a-chip) to manipulate microparticles and cells. It revealed the characterization of microparticles that show the differential ability to be DEP-caged, levitated and moved. It also assessed the use of the SmartSlide to levitate and move tripalmitin-based lipospheres.

A wireless interconnection scheme based on capacitive coupling provides mono- and bi-directional transmission capabilities for 3-D system integration. Chips are implemented in 0.13 μm CMOS technology and assembled face-to-face at die-level. RX-TX circuits are specifically designed for low-power functionality and the implementation takes advantage of the two different voltage thresholds that are available for the standard transistors in the CMOS process we used. The communication circuits are coupled via electrodes with an area down to 8 x 8 μm² and this enables the vertical propagation of clock signals at 1.7 GHz, a propagation delay of 420 ps for general purpose signals and a throughput of more than 22 Mb/s/μm² with 0.08 pJ/b energy consumption. This paper presents a 3-D interconnection scheme based on capacitive coupling. We propose synchronous communication circuits, based on a precharge and transmission approach, that provide an optimization of interconnection sensitivity. Measurements on a 0.13 µm CMOS implementation demonstrate working connections with an area occupation of 8 × 8 µm². Experimental results are presented for both die-to-die and wafer-to-wafer assembly techniques. They show a maximum communication bandwidth of 1.23 Gb/s, leading to a throughput per area of 19 Mb/s/µm² with an energy consumption of 0.14 mW/Gb/s. Quantitative evaluation of the changes in skin topographic structures are of great importance in the dermocosmetic field to assess subjects response to medical or cosmetic treatments. Although many devices and methods are known to measure these changes, they are not suitable for a routine approach and most of them are invasive. Moreover, it has always been difficult to give a measure of the skin health status as well as of the human aging process by simply analyzing the skin surface appearance. This work describes how a portable capacitive device could be utilized to achieve measurements of skin ageing in vivo and routinely. The capacitive images give a high resolution representation of the skin micro-relief, both in terms of skin surface tissue and wrinkles. In a previous work we dealt with the former; here we have addressed the latter. The algorithm we have developed allowed us to extract two original features from wrinkles: the first is based on photometric properties while the second has been achieved through the multiresolution analysis of the wavelet transform. Reconfigurable architectures are well suited for wireless applications since they provide high performance computation together with the capability to adapt to changing communication protocols. Moving to 90·nm technology and below, FPGAs could suffer from leakage energy consumption due to the large number of inactive transistors. This paper presents an extensive study on the application of different low-leakage techniques to the design of FPGAs. The approaches are compared and mixed to find an implementation of switch blocks and look-up tables which reduces leakage without affecting delay and area. The circuits we propose achieve an 86% stand-by energy saving and 46% active leakage reduction with respect to standard implementations. In the nanometer era, the increase in nonrecurring engineering costs is a challenge for SoCs that can be faced through a standardization process. Hardware specialization of a standard platform to a given application can be achieved by exploiting reconfigurable technology. This paper presents a XiSystem SoC, which integrates two different field-programmable devices to provide application-specific computing blocks and IOs. A XiRisc reconfigurable processor is exploited to achieve more than one order of magnitude speed-up and energy consumption reduction vis-à-vis a DSP-like processor, while an eFPGA is integrated in the system in order to make it flexible enough to support various IO ports and protocols. Manipulation of populations of living cells on an individual basis is essential for the investigation of complex interactions among cells. We present a new approach to the integration on silicon of dielectrophoretic actuators and optical sensors that allow us to carry out this task. The device presented in this paper is an 8 × 8 mm² chip implemented in a two-poly three-metal 0.35-µm CMOS technology, featuring 102,400 actuation electrodes, arranged in an array of 320 × 320, 20 µm × 20 µm microsites each comprising addressing logic, an embedded memory for electrode programming, and an optical sensor. The chip enables software-controlled displacement of more than 10,000 individual living cells, allowing biologists to devise complex interaction protocols that are impossible to manage otherwise. This paper describes a new architecture for embedded reconfigurable computing, based on a very-long instruction word (VLIW) processor enhanced with an additional run-time configurable datapath. The reconfigurable unit is tightly coupled with the processor, featuring an application-specific instruction-set extension. Mapping computation intensive algorithmic portions on the reconfigurable unit allows a more efficient elaboration, thus leading to an improvement in both timing performance and power consumption. A test chip has been implemented in a standard 0.18-µm CMOS technology. This paper presents a mixed-signal system-on-chip (SOC) for sensing capacitance variations, enabling the creation of pressure-sensitive fabric. The chip is designed to sit in the corner of a smart fabric such as elastic foam overlaid with a matrix of conductive threads. Background: The Prognostic Nutritional Index (PNI) is a parameter of nutritional and inflammation status related to toxicity in cancer treatment. Since data for head and neck cancer are scanty, this study aims to investigate the association between PNI and acute and late toxicity for this malignancy. Methods: A retrospective cohort of 179 head and neck cancer patients treated with definitive radiotherapy with induction/concurrent chemotherapy was followed-up (median follow-up: 38 months) for toxicity and vital status between 2010 and 2017. PNI was calculated according to Onodera formula and low/high PNI levels were defined according to median value. Odds ratio (OR) for acute toxicity were calculated through logistic regression model; hazard ratios (HR) for late toxicity and survival were calculated through the Cox proportional hazards model. Results: median PNI was 50.0 (interquartile range: 45.5–53.5). Low PNI was associated with higher risk of weight loss > 10% during treatment (OR = 4.84, 95% CI: 1.73–13.53 for PNI < 50 versus PNI ≥ 50), which was in turn significantly associated with worse overall survival, and higher risk of late mucositis (HR = 1.84; 95% CI:1.09–3.12). PNI predicts acute weight loss >10% and late mucositis. Conclusions: PNI could help clinicians to identify patients undergoing radiotherapy who are at high risk of acute and late toxicity.
