Abstract Background Equine degenerative suspensory ligament desmitis (DSLD) is a systemic connective tissue disorder first identified in Peruvian Paso horses but afflicting other horse breeds as well. Inappropriate accumulation of proteoglycans in connective tissues, most prominently in tendons and ligaments, leads to progressive and debilitating lameness and pain. It is largely unknown what drives the overproduction of proteoglycans, but our previous studies suggest involvement of bone morphogenetic protein 2 (BMP2), a member of the transforming growth factor-β (TGFβ) family, impacting synthesis of proteoglycans. To identify potential players in pathogenesis of DSLD a new approach utilizing next generation sequencing was undertaken. Methods Next generation sequencing was performed using RNA extracted from skin biopsies of six control Peruvian Pasos and six horses with DSLD (4 Peruvian Pasos and 2 warmbloods). The CuffDiff result sets were validated with algorithms used to run them. This was based on the determined false discovery rates derived from the P values adjusted for multiple testing for any given result. Results Bioinformatics analysis of transcriptomes revealed differential expression of over 1500 genes, including increased expression of genes for several growth factors (most prominently BMP2, FGF5, CTGF, many members of the EGF family), and mediators of signaling (Fos, Myc, MAPK system), and keratins. Two genes encoding for enzymes involved in synthesis of hyaluronan were also overexpressed. Gene expression was decreased for protein cores of many proteoglycans, several growth factors, most collagens, and many peptides with immune function. Conclusions The overexpression of BMP2 correlates well with our previous data. However, the decrease in expression of numerous proteoglycans was unexpected. A mutation in a gene of a less characterized proteoglycan and/or glycosyltransferase with subsequent increased production of hyaluronan and/or a proteoglycan(s) undetected in our study could account for the systemic proteoglycan deposition. Decreased collagen gene expression indicates abnormal connective tissue metabolism. The increased expression of keratin genes and FGF5 supports reports of skin abnormalities in DSLD. Underexpression of immune function genes corresponds with lack of inflammation in DSLD tissues. Finally, though the proteoglycan and/or glycosaminoglycan abundant in DSLD has not been identified, we validated our previous data, including overexpression of BMP2, and systemic nature of DSLD due to disturbed metabolism of the extracellular matrix.

Barriers exist at a variety of levels that either impede or prohibit persons with disabilities from full participation within the worship, education, and mission of congregations. Placing the emerging body of literature on inclusion in congregations in conversation with a case study of one particular congregation, the author examines the sources and strategies of one church's efforts at becoming more inclusive. The role of the Director of Christian Education, Martha Bess DeWitt, is presented, with the focus on her participation, leadership, and reflection on hospitality as central to the church's move toward intentional inclusion of people with disabilities.

The article provides information on a study that investigated a specific case of the interaction between leaders' and followers' personalities in the determination of behavior in groups. It describes the results of a study of the behavior of authoritarian and equalitarian personalities in small groups. The California F scale was used to measure authoritarian personality. It reveals the results of the study. It discusses the implications of these results for an interaction theory of leadership, and for the prediction of behavior from personality measures of individuals.

A rapid gas chromatographic method for the determination of underivatized theophylline in whole blood is described. Theophylline is extracted from acidified blood and chromatographed directly using cyheptamide as an internal standard. Concentrations of the drug down to 2 mug/ml in blood could be determined with recoveries ranging from 90 to 110%.

A combined thin-layer and gas-liquid chromatographic (TLC-GLC) procedure for determination of tricyclic antidepressant drugs in urine is described. GLC on 1.5% OV-17 was utilized to analyzed basic urine extracts directly for the tricyclics and to confirm the identity of spots extracted from thin-layer chromatograms, and TLC was used to confirm the results of these GLC screens. Lower limits of sensitivity for the thin-layer spot extraction procedure were found to range from 2 mug/ml for amitriptyline and imipramine to 10 mug/ml for desipramine and nortriptyline. Turn-around-times for the GLC tricyclic screens were usually less than 1 h. The finding that nortriptyline and desipramine were chemically altered under various extraction conditions was used as a means of confirming the identity of these drugs, and the identity of these chemically altered derivatives was discussed with reference to their low resolution mass spectra.

A prospective study of 181 patients suspected of having liver disease was carried out to determine the relative efficiencies of serum bilirubin (total and direct), alkaline phosphatase (AP), gamma glutamyl transferase (GGT), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) with respect to diagnosis. Liver biopsies, liver scans, abdominal ultrasound, and clinical parameters were also tabulated and used independently to evaluate the patient's hepatic status and to determine the final diagnoses in each case. From the results of these tests for the 60 patients who were diagnosed as having liver disease, and the 87 patients who were felt to be free of liver disease, predictive values of the above tests were established. Data from this study suggests that while direct bilirubin is the most specific test, GGT is the most sensitive and has the fewest false negatives in the diagnosis of liver disease.
