Abstract To gain a deeper understanding of the metabolic differences within and outside the body, as well as changes in transcription levels following estrus in yaks, we conducted transcriptome and metabolome analyses on female yaks in both estrus and non-estrus states. The metabolome analysis identified 114, 13, and 91 distinct metabolites in urine, blood, and follicular fluid, respectively. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis highlighted an enrichment of pathways related to amino acid and lipid metabolism across all three body fluids. Our transcriptome analysis revealed 122 differentially expressed genes within microRNA (miRNA) and 640 within long non-coding RNA (lncRNA). Functional enrichment analysis of lncRNA and miRNA indicated their involvement in cell signaling, disease resistance, and immunity pathways. We constructed a regulatory network composed of 10 lncRNAs, 4 miRNAs, and 30 mRNAs, based on the targeted regulation relationships of the differentially expressed genes. In conclusion, the accumulation of metabolites such as amino acids, steroids, and organic acids, along with the expression changes of key genes like miR-129 during yak estrus, provide initial insights into the estrus mechanism in yaks.

This study investigated the effects of defective pear fermentation (DPF) diets on growth performance and gastrointestinal microbial communities in 60 healthy male small-tailed Han sheep, aged 90 days. The sheep were randomly divided into four groups, each consisting of three replicates with five sheep per replicate. Initially, all groups received a basal diet for seven days during the adaptation stage. Subsequently, for 60 days, group C (control) was fed a basal diet, group X received a basal diet with 2% DPF, group Y had a basal diet with 4% DPF, and group Z was fed a basal diet with 6% DPF. The results indicated that group Y experienced a significant increase in average daily gain (ADG) and average daily feed intake (ADFI). The addition of DPF significantly elevated the levels of GSH-Px and notably reduced MDA content compared to group C. Analysis of gastrointestinal microbiota showed that groups receiving DPF had increased relative abundances of Lachnospiraceae_NK3A20_group, norank_f p-2534-18B5_gut_group, Acetitomaculum, Actinobacteriota, Bacteroidota and Ruminococcus_gauvreauii_group, and decreased abundances of Proteobacteria, Prevotella, Staphylococcus, and Psychrobacter compared to group C. Group X exhibited the highest relative abundance of Olsenella, while group Y showed a significant increase in unclassified_f Lachnospiraceae compared to the other groups. Bacterial function prediction indicated that pathways related to energy metabolism were more prevalent in group X and Y. This study preliminarily confirms the feasibility of using DPF as feed additives, providing a foundation for further research and evaluation of DPF's application in animal production.

Polyphenol-rich grape pomace (GP) represents a valuable processing by-product with considerable potential as sustainable livestock feed. This study aimed to investigate the effects of different levels of GP on the growth performance and nitrogen utilization efficiency, antioxidant activity, and rumen and rectum microbiota of Angus bulls. Thirty Angus bulls were allocated three dietary treatments according to a completely randomized design: 0% (G0), 10% (G10), and 20% (G20) corn silage dry matter replaced with dried GP dry matter. The results showed that the average daily gain (ADG) of the G0 group and G10 group was higher than that of the G20 group (p < 0.05); urinary nitrogen levels decreased linearly with the addition of GP (linear, p < 0.05). In terms of antioxidants, the levels of catalase (CAT) in the G10 group were higher than in the G0 and G20 groups (p < 0.05), and the total antioxidative capacity (T-AOC) was significantly higher than that in the G20 group (p < 0.05). In addition, in the analysis of a microbial network diagram, the G10 group had better microbial community complexity and stability. Overall, these findings offer valuable insights into the potential benefits of incorporating GP into the diet of ruminants.

ABSTRACT: This study aimed to investigate the effects of compound probiotics (consisting of 108 cfu/g of Lactobacillus plantarum, 108 cfu/g of Pediococcus acidilactici, 108 cfu/g of Pediococcus pentosaceus, 107 cfu/g of and Bacillus subtilis) on growth performance, rumen fermentation, bacteria community, blood parameters, and health status of Holstein calves at the first 3 mo of age. Forty-eight newborn calves were randomly divided into the following 3 groups: control group (milk replacer with no compound probiotics), low compound probiotics group (milk replacer + 0.12 g of compound probiotics per head per day), and high compound probiotics group (HP; milk replacer + 1.2 g of compound probiotics per head per day). Starter pellets of the low compound probiotics and HP groups were coated with 0.05% compound probiotics. Milk replacer was provided from 2 to 63 d of age (6 L at 2–10 d, 8 L at 11–42 d, 6 L at 43–49 d, 4 L at 50–56 d, and 2 L at 57–63 d), and starter pellets were provided ad libitum from 7 to 90 d of age. Body weight and body size (d 1, 30, 60, and 90), blood (d 40 and 80), and rumen fluid (d 90) were analyzed using the one-way ANOVA procedure; fecal score was recorded daily and analyzed as repeated measures using the mixed model procedure. Results showed that diet supplemented with compound probiotics had no effects on the body weight, average daily gain, dry matter intake, and feed efficiency. At 90 d of age, diet supplemented with compound probiotics decreased the withers height. Immunity activities increased in the HP group, supported by the increased concentrations of serum total protein and immunoglobulins at 40 d of age, and by the increased activity of superoxide dismutase at 80 d of age. Diet supplemented with compound probiotics altered rumen fermentation, indicated by the decreased rumen acetic acid and propionic acid, and the increased butyric acid concentrations. Diet supplemented with compound probiotics improved the health status of calves, indicated by the decreased fecal score at 3 wk of age and the decreased medicine treatments. In summary, although diet supplemented with HP decreased the withers height, this level of probiotics is recommended to improve rumen development and health status of newborn Holstein calves.

The objective of this study was to evaluate the effects of potato vine and leaf mixed silage (PVS) on rumen fermentation and the microbe in ruminants and to improve the utilization of PVS resources in ruminants through in vitro gas production and feeding trials. The experiment was divided into three groups: PVS1 (50% corn + 50% potato vine and leaf silage), PVS2 (75% potato vine and leaf + 15% rice straw + 10% cornmeal silage) and whole-plant corn silage (CS). The in vitro gas production results showed that there was a significant reduction in PVS groups in the indexes of total gas (p < 0.05) and CH4 production (p < 0.05). The digestibility of dry matter (p < 0.05), neutral detergent fiber (NDF) (p < 0.05), and acid detergent fiber (ADF) (p < 0.05) at 48 h were decreased in the PVS group. For the rumen fermentation indexes, the pH (p < 0.05), microbial crude protein (MCP) (p < 0.05), and acetate to propionate (p < 0.05) showed an increase in the PVS group, but a decrease in the total volatile fatty acid concentration (p < 0.05). In the feeding trial, different silages in diets had no significant effect on the rumen fermentation indexes (p > 0.05). In the rumen microbe composition, the PVS diet significantly reduced the abundance of Prevotella (p < 0.05) compared with the CS diet group. The PVS2 diet significantly increased the abundance of the Lachnospiraceae_XPB1014_group (p < 0.05) and Bacteroidales_bacterium_Bact_22 (p < 0.05) compared with the CS diet group. In conclusion, PVS had no negative effect on rumen fermentation characteristics and rumen microbial flora and could significantly reduce rumen gas production compared to CS, positively mitigating animal husbandry CH4 emissions and environmental protection.

This study aims to explore the different growth performances of the Angus bull on potato vine and leaf mixed silage in the early fattening period and to provide a reference animal production trial. Thirty-six 13-month-old Angus bulls were divided into three groups with 403.22 ± 38.97 kg initial body weight and fed with three different silage diets: (1) control: whole-plant corn silage as control (CS); (2) treatment 1: 50% whole-plant corn +50% potato vine and leaf silage (PVS1); and (3) treatment 2: 75% potato vine and leaf +15% rice straw +10% cornmeal silage (PVS2). After the 14 days pre-feeding, the formal experiment was carried out for 89 days. The result showed that the ash content of the potato vine and leaf mixed silage (PVS) in the treatment groups was higher than that in control group, and the ash content of PVS1 and PVS2 even reached 10.42% and 18.48% (DM%), respectively, which was much higher than that of the CS group at 4.94%. The crude protein content in silage also increased with the additional amount of potato vine and leaf. The apparent crude protein digestibility of the PVS groups was also significantly higher than that of the CS group (p < 0.05). In terms of serum biochemical indexes, blood urea nitrogen (BUN) in the experimental groups was significantly higher than in the control group (p < 0.05). Compared with PVS2, cholesterol (CHO) was significantly lower in the CS and PVS1 groups (p < 0.05). Moreover, the high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) of PVS2 were significantly higher than those of the CS and PVS1 group (p < 0.05), and daily gain (ADG) as a key production index had a significantly negative correlation with the CHO (r = −0.38, p < 0.05) and HDL-C (r = −0.40, p < 0.05) of cattle. In conclusion, PVS had higher crude protein content and ash but less starch than whole-corn silage. The PVS could replace whole-plant corn silage at the same dry matter status and did not affect the weight gain in this trial.

The objective of this study was to evaluate the effects of isopropyl ester of 2-hydroxy-4-(methylthio)-butyrate acid (HMBi) on ruminal and cecal fermentation, microbial composition, nutrient digestibility, plasma biochemical parameters, and growth performance in finishing beef cattle. The experiment was conducted for 120 days by a complete randomized block design. Sixty 24-month-old Angus steers (723.9 ± 11.6 kg) were randomly assigned to one of the flowing three treatments: basal diet (the concentrate: 7.6 kg/head·d−1, the rice straw: ad libitum) supplemented with 0 g/d MetaSmart® (H0), a basal diet supplemented with 15 g/d of MetaSmart® (H15), and a basal diet supplemented with 30 g/d of MetaSmart® (H30). Results showed that the average daily gain (ADG) increased linearly (P = 0.004) and the feed conversion ratio (FCR) decreased linearly (P < 0.01) with the increasing HMBi supplementation. Blood urea nitrogen (BUN) concentration significantly decreased in the H30 group (P < 0.05) compared with H0 or H15. The ruminal pH value tended to increase linearly (P = 0.086) on day 56 with the increased HMBi supplementation. The concentrations of ammonia–nitrogen (NH3-N), propionate, isobutyrate, butyrate, isovalerate, valerate, and total volatile fatty acid (VFA) were linearly decreased in the cecum (P < 0.05). The results of Phylogenetic Investigation of Communities by Reconstruction of Unobserved States (PICRUSt) showed that the abundance of most pathways with a significant difference was higher in the rumen and lower in the cecum in the H30 group compared to the H0 group, and those pathways were mainly related to the metabolism of amino acids, carbohydrates, and lipids. Correlation analysis showed that ADG was positively associated with the ratio of firmicutes/bacteroidetes both in the rumen and cecum. Additionally, the abundance of Lachnospiraceae, Saccharofermentans, Lachnospiraceae_XPB1014_group, and Ruminococcus_1 was positively correlated with ADG and negatively correlated with FCR and BUN in the rumen. In the cecum, ADG was positively correlated with the abundances of Peptostreptococcaceae, Romboutsia, Ruminococcaceae_UCG-013, and Paeniclostridium, and negatively correlated with the abundances of Bacteroidaceae and Bacteroides. Overall, these results indicated that dietary supplementation of HMBi can improve the growth performance and the feed efficiency of finishing beef cattle by potentially changing bacterial community and fermentation patterns of rumen and cecum.

Objective This study investigated the effect of dietary crude protein (CP) supplementation on nutrient intake, nitrogen (N) utilisation, blood metabolites, ruminal fermentation and growth performance of young Holstein bulls. Methods Twenty-one young bulls weighing 277±11.2 kg were equally divided into three groups and were offered diets formulated with low CP (LCP; 10.21% CP and 4.22% rumen degradable protein [RDP]), medium CP (MCP; 12.35% CP and 5.17% RDP) and high CP (HCP; 14.24% CP and 6.03% RDP). Hybridization in bovines is practiced with the main aim of improving production performance, which may imply the microbial variations in the rumen from the parental breed cross to their progeny. Besides, the interactions of offspring breed with sex in terms of rumen bacteria are not clear. This study aims to evaluate the variations in rumen bacterial communities in different breeds and sexes, and the correlations among fattening performance, serum biochemical parameters, and rumen fermentation. Forty-two 19.2 ± 0.67-month-old beef cattle (390 ± 95 kg of initial body weight) comprising two genetic lines (Yiling and Angus × Yiling) and two sexes (heifers and steers) were raised under the same high-grain diet for 120 d. On the last two days, blood samples were collected from each animal via the jugular vein before morning feeding for analyzing serum biochemical parameters; rumen fluid samples were obtained via esophageal intubation 2 h after morning feeding for analyzing rumen fermentation parameters and bacterial communities. The results show that both breed and sex had a certain impact on fattening performance, serum biochemical parameters, and rumen fermentation. No differences in the diversity and structure of rumen bacterial communities were observed. Significant interactions (p < 0.05) of breed and sex were observed for Succinivibrionaceae UCG-002 and Prevotellaceae UCG-001. The relative abundances of the Rikenellaceae RC9 gut group, Prevotellaceae UCG-003, and Succinivibrio were different (p < 0.05) between breeds. Heifers had a higher (p = 0.008) relative abundance of the Rikenellaceae RC9 gut group than steers. Correlation analysis showed a significant relationship (p < 0.05) of rumen bacteria with serum biochemical parameters, rumen pH, and rumen fermentation patterns. Additionally, only two genera, Prevotellaceae UCG-003 and Prevotellaceae UCG-001, had positive correlations with feed efficiency. In conclusion, serum biochemical parameters, rumen fermentation, and rumen bacterial communities are partly driven by the breed and sex of cattle fed a high-grain diet.

Objective Wheat is an alternative to corn silage for ruminant feeding in northern China. This study examined the effects of harvest time and added molasses on nutritional content, ensiling characteristics and in vitro degradation of whole crop wheat (WCW). Methods Fresh WCW at the milk-ripe stage was harvested at 0700 h (i.e., in the morning [Mo]) and 1700 h (i.e., in the afternoon [Af]), and then immediately used to prepare silage and make hay. Twenty-eight male, weaned Chinese Holstein calves ((156.8±33.4) kg) were used to investigate the effects of dietary forage to concentrate ratio (F:C) and forage length on nutrient digestibility, plasma metabolites, ruminal fermentation, and fecal microflora. Animals were randomly allocated to four treatments in a 2×2 factorial arrangement: whole-length forage (WL) with low F:C (50:50); WL with high F:C (65:35); short-length forage (SL) with high F:C (65:35); and SL with low F:C (50:50). Chinese wildrye was used as the only forage source in this trial. The grass in the SL treatments was chopped using a chaff cutter to achieve small particle size (∼50% particles >19 mm). Dry matter intake (DMI) and organic matter (OM) intake was increased by increasing both F:C (P0.05). Microflora similarity was 50–73% among the different treatments. It is concluded that the WL with high F:C (65:35) diet is suitable for weaned calves.

This study was conducted to explore the proper time required to achieve stabilization in digestibility, serum metabolism, and rumen fermentation characteristics when different diets shift, thus providing decision-making of practical sampling frequency for basal nutritional research. For these purposes, 12 Holstein steers (body weight 467 ± 34 kg, age 14 ± 0.5 months) were equally assigned to two dietary treatments: high-density (metabolizable energy (ME) = 2.53 Mcal/kg and crude protein (CP) = 119 g/kg; both ME and CP were expressed on a dry matter basis) or low-density (ME = 2.35 Mcal/kg and CP = 105 g/kg). The samples of feces, serum, and rumen contents were collected with a 30-day interval. All data involved in this study were analyzed using the repeated measures in mixed model of SPSS. Results showed that nutrient apparent digestibility and serum metabolic parameters were stable across each monthly collection, while most rumen fermentation characteristics, namely concentrations of acetate, propionate, isobutyrate, and valerate, were affected by the interaction effects between collection period and dietary density. These findings indicate that rumen fermentation characteristics require more time to stabilize when diet shifts. It is recommended to collect ruminal digesta monthly to evaluate rumen fermentation characteristics, while unnecessary to sample monthly for digestion trials and blood tests in the long-term fattening of Holstein steers. This study may provide insights into exploring the associations between detected parameters and stabilization time, and between diet type and stabilization time when diet shifts.

The objectives of this study were to investigate the effects of age and dietary straw levels on growth performance, carcass and meat traits, as well as tissue antioxidant status of Yiling cull cows. Twenty-four Yiling cull cows were arranged in a 2 × 2 factorial design: two age classes consisting of younger cull cows (YCC; appearing with three or four pairs of permanent teeth) and older cull cows (OCC; worn out teeth); two dietary treatments consisting of lower and higher rice straw levels (LRS and HRS; providing 0.7 kg/d and 1.2 kg/d rice straw per head based on air-dry basis, respectively). Cows were fed twice a day. Straw was offered at half of the predetermined weight each meal; concentrate was separately supplied ad libitum. After 300 d of feeding, final body weight (BW), total BW gain, average daily gain and gain:feed intake were higher (p < 0.01) in the YCC group than in the OCC group. Total dry matter intake was higher (p = 0.03) in the HRS group than in the LRS group, but neutral detergent fiber apparent digestibility was negatively affected (p = 0.01) by increased straw levels. Decreased C15:0, C17:0, C20:5n3c, and saturated fatty acids (SFAs) proportion as well as increased C18:1n9c and monounsaturated fatty acids (MUFAs) proportion in meat from YCC with HRS diet were observed as compared to that in meat from YCC with LRS diet (p < 0.05). Meat from HRS group had higher (p = 0.04) C18:3n3c proportion than meat from LRS group. No significant differences (p > 0.05) were found for meat quality attributes except for cooking loss, which was higher (p = 0.02) in the HRS group than in the LRS group. Both YCC group and HRS group had higher (p < 0.05) cold carcass weight compared to OCC group and LRS group. Moreover, catalase activity of liver tissue was higher (p = 0.045) in YCC than in OCC, while superoxide dismutase activity of muscle tissue was higher (p = 0.01) in LRS than in HRS. Based on results, we concluded that younger age and feeding high-level straw can improve the finishing performance of Yiling cull cows.

This study determined the effects of wheat stage, preservation treatment, and harvest time on the fermentation characteristics, methane production, and bacterial diversity. In this study, processing wheat into hay can reduce methane production. The MWS7 (wheat harvested at 7:00 in milk stage and preserved as silage) group had a significantly lower CO2 compared with the DWS15 (wheat harvested at 15:00 in dough stage and preserved as silage) group. Neutral detergent fiber degradation in the hay treatment harvesting at 7:00 was significantly higher than that in other treatments. The butyrate proportion in the DWH7 (wheat harvested at 7:00 in dough stage and preserved as hay) group was higher than that in the MWS7 group. Results from high-throughput sequencing showed that there were differences in the relative abundance of some minor rumen microbiota among the treatments. The MWS7 group had greater microbial diversity and the MWH7 group (wheat harvested at 7:00 in milk stage and preserved as hay) had higher species richness. In addition, the MWH7 group had a lower Methanobrevibacter abundance and methane production. Overall, the MWH7 group may have advantages of rumen fermentation and reduce methane production.

Roughage particle size can influence rumen development, which is also determined by rumen microorganisms and their metabolic end-products. Therefore, the aim of this study was to evaluate the comprehensive effects of roughage length and rumen bacterial community on the rumen development of weaned calves. A total of thirty-six weaned Angus female calves (125 ± 3 d; 161.2 ± 13.0 kg) were randomly assigned to three diets differing in roughage particle size: 4 cm (short length); 24 cm (medium length); and 44 cm (long length). Results showed that chopping roughage increased dry matter intake and organic matter apparent digestibility; altered rumen fermentation indicated by the increased rumen butyrate and valerate concentrations; and increased plasma glucose, cholesterol, and total protein. Chopping roughage affected rumen bacterial community, as indicated by altering the diversity indices; by increasing ruminal bacteria Papillibacter and Eubacterium_hallii_group, which are involved in butyrate production; and by increasing Synergistetes and Mogibacterium, which are involved in bacterial colonization. In conclusion, chopping roughage at 4 cm was shown to improve the rumen bacterial community, alter rumen fermentation, eventually promote the development of rumen.

The aim of this study is to track the dynamic alterations in nutrient intake and digestion, rumen fermentation and plasma metabolic characteristics, and rumen bacterial community of Holstein finishing steers in response to three nutrient density diets as fattening phases advanced. A total of eighteen Holstein steers were randomly allocated into three nutrient density groups and steers in each group were fed under a three-phase fattening strategy, with nutrient density increased in each group when fattening phase advanced. Results showed that both fattening phase and dietary nutrient density significantly influenced the nutrient digestion, most of the rumen fermentation parameters, and part of bacteria at phylum and genus levels. Individually, dietary nutrient density affected the concentrations of plasma alanine aminotransferase and urea N, bacterial richness and evenness. All determined nutrient intake and plasma biochemical parameters, except for alanine aminotransferase and triglyceride, differed among fattening phases. Spearman correlation analysis revealed strong correlations between fiber intake and bacterial richness and evenness, rumen fermentation characteristics and certain bacteria. Moreover, Patescibacteria abundance was positively correlated with ambient temperature and plasma total protein. These results indicate that rumen fermentation and nutrient digestion were influenced by both dietary nutrient density and fattening phase, and these influences were regulated by certain rumen bacterial community and ruminal bacteria may be affected simultaneously by ambient temperature. This study may provide insights into diet optimization and potentially adaptive mechanism of rumen bacterial community in response to fattening phases and gradually climatic change.

The objective of this study was to explore whether collecting rumen samples of finishing steers at monthly intervals differed, and whether this difference or similarity varied with diets. For these purposes, 12 Chinese Holstein steers were equally divided into two groups. The dietary treatments were either standard energy and standard protein (C) or low energy and low protein (L). Rumen samples were obtained on day 30, day 60 and day 90 from both dietary treatments and were analyzed by using 16S rRNA gene sequencing. The results showed that monthly intervals had no effect on the richness and evenness of the rumen bacterial community in the two diets. However, taxonomic difference analysis (relative abundance >0.5%) revealed that the relative abundance of three phyla (Proteobacteria, Fibrobacteres and Cyanobacteria) and six genera (Rikenellaceae_RC9_gut_group, Ruminococcaceae_NK4A214_group, Fibrobacter, Eubacterium_coprostanoligenes_group, Ruminococcaceae_UCG-010 and Ruminobacter) were significantly different between monthly sampling intervals, and the difference was prominent between sampling in the first month and the subsequent two months. Moreover, the differences in abundances of phyla and genera between monthly sampling intervals were affected by diets. Analysis of similarity (ANOSIM) showed no significant differences between monthly sampling intervals in the C diet. However, ANOSIM results revealed that significant differences between the first month and second month and between the first month and third month were present in the L diet. These results indicated that temporal dynamics in rumen bacterial community composition did occur even after an adaptation period of three months. This study tracked the changes in rumen bacterial populations of finishing cattle after a shift in diet with the passage of time. This study may provide insight into bacterial adaptation time to dietary transition in finishing steers.

Background Freemartinism generally occurs in female offspring of dizygotic twins in a mixed-sex pregnancy. Most bovine heterosexual twin females are freemartins. However, about 10% of bovine heterosexual twin females are fertile. Farmers mostly cull bovine fertile heterosexual twin females due to the lack of a practical diagnostic approach. Culling of such animals results in economic and genetic-material losses both for dairy and beef industry. Methods In this study, a comparative test, including qualitative detection of SRY gene by polymerase chain reaction (PCR), quantitative detection of relative content of SRY by real-time quantitative polymerase chain reaction (qPCR), and quantitative detection of H-Y antigen, was performed to establish the most accurate diagnosis for freemartin. Twelve Holstein heterosexual twin females were used in this study, while three normal Holstein bulls and three normal Holstein cows were used as a positive and negative control, respectively. Results Polymerase chain reaction results revealed that SRY gene were absent in three heterosexual twin females and only two of them were verified as fertile in later age. The qPCR results showed that relative content of SRY was more than 14.2% in freemartins and below 0.41% in fertile heterosexual twin females. The H-Y antigen test showed no significant numerical difference between freemartin and fertile heterosexual twin female. Discussion Our results show that relative content of SRY quantified by qPCR is a better detection method for diagnosis of freemartin in Holstein cattle as compare to qualitative detection of SRY gene by PCR or quantitative detection of H-Y antigen. To the authors’ knowledge, this is the first time we applied qPCR to diagnosing freemartin by quantifying SRY gene and got relative SRY content of each freemartin and fertile heterosexual twin female. We concluded that low-level of SRY would not influence fertility of bovine heterosexual twin female.

Antibiotically disturbed gastrointestinal microbiota needs a long period time to be restored to normal, which may cause a series of problems to the host. The understanding of restoration of the biased microbiota by antibiotics remains largely unknown. Here, we investigated the microbiota shift in foregut (rumen) and hindgut (rectum) of lactating cows after antibiotics exposure as well as after antibiotics withdrawal with (Microbiota transplantation, MT group) or without (Control, CON group) microbiota transplantation. We were able to demonstrate that microbiota in both foregut and hindgut significantly changed after 3 or 14 days of antibiotics exposure, and the changes persisted over long period of time (>18 days) after withdrawing the antibiotics. We further observed a faster restoration of microbiota in both foregut and hindgut of MT group than CON group, microbiota in foregut was mainly benefited from microbiota transplantation by restoring the alpha-diversity as well as within-group similarity, while microbiota in hindgut was primarily benefited from microbiota transplantation by reestablishing the co-occurrence network (nodes number, edges number, density, modularity as well as closeness centrality). These results together expanded our understanding of restoration of the biased microbiota by antibiotics, and may also be instructive to deal with the delayed microbiota restoration at least in cows.

The objective of this study was to track the dynamic variations in fecal bacterial composition and fermentation profile of finishing steers in response to three stepwise diets varied in energy and protein density. A total of 18 Holstein steers were divided into three groups in such a way that each group contained six animals and received one of three stepwise dietary treatments. Dietary treatments were C = standard energy and protein diet, H = high energy and protein diet, and L = low energy and protein diet. Animals were fattened for 11 months with a three-phase fattening strategy. Fecal samples were collected to evaluate the dynamics of fecal fermentation and bacterial composition in response to dietary treatments and fattening phases using 16S rRNA gene sequencing. Fecal acetate, propionate, and butyrate increased with increasing density of diet and as the fattening phase continued. The relative abundances of Firmicutes and Bacteroidetes dominated and showed 56.19% and 33.58%, respectively. Higher dietary density decreased the fecal bacterial diversity, Firmicutes to Bacteroidetes ratio, and the relative abundances of Ruminococcaceae_UCG-005, Rikenellaceae_RC9_gut_group, and Bacteroides, whereas higher dietary density increased the abundance of Prevotella_9. Our results indicated that both fecal fermentation profile and bacterial composition share a time-dependent variation in response to different dietary densities. This knowledge highlights that both diet and fattening phase impact fecal fermentation profile and bacterial composition, and may provide insight into strategies to reduce fecal contamination from the origin by optimizing diet and fattening time.

This essay aims to comparatively study the microscopic morphology and grinding properties of brazed diamonds with amorphous and crystalline filler alloy in the case of rare-earth Nd-doped modified Ni-based filler alloy. The results show that at the same content of Nd, Cr7C3 is mainly generated at the brazing interface of crystalline samples. In contrast, Cr27C6 is mainly generated at the brazing interface of amorphous samples. Amorphous solder material achieves a robust chemical metallurgical union, producing a dense and homogeneous carbide layer. The bonding strength between amorphous solder material and diamond is further enhanced. The addition of rare earth element Nd contributes greatly to the forming ability of amorphous filler alloy. Nd atoms in the amorphous solder material readily bond with Ni atoms, reducing the chemical corrosion of Ni atoms on diamonds and resulting in a lower degree of graphitization. When the amount of Nd added is 1.0 wt%, the morphology of amorphous filler alloy brazing diamond is more complete than the effect of the crystalline state, the degree of thermal damage is lower, and the surface of amorphous filler alloy brazing is smoother. Frictional wear experiments show that the amorphous filler alloy holds diamond better than the crystalline filler alloy, and the diamond specimens prepared with the amorphous filler alloy demonstrate a significantly superior grinding performance in comparison to the diamond specimens prepared with the crystalline filler alloy.

In this study, the properties of Ni-based filler alloy are improved by adding rare earth Sm to them and brazing them at a temperature of 1080 °C. Relevant tests are carried out on filler alloy and brazed joints by means of electron microscopy (SEM), energy spectroscopy (EDS) and X-ray diffractometry (XRD). The results show that the addition of rare earth Sm can refine the grain of the filler alloy, improve the hardness of the filler alloy and reduce the thermal damage of the diamond after welding. The addition of rare earth Sm affects the diffusion of Cr, which reacts with the diamond at the interface to form Cr3C2 and Cr7C3, and the formation of these two carbides improves the bonding strength between the filler alloy and the diamond. When the addition of rare earth Sm is 1.5 wt%, the melting point of the filler alloy is the lowest, the grain refinement is the best, the hardness is the highest, and the thermal damage of the brazed sample is the lowest. In addition, the filler alloy of 1.5 wt%Sm has the greatest effect on the diffusion of the Cr element, the bonding strength between the filler alloy and diamond is the highest, and the mechanical properties of the brazed specimens are the best.

Assessing the safety of amphibious aircraft hinges significantly on two key factors: wave-added resistance and motion stability during takeoff and landing on water surfaces. To tackle this, we employed the Reynolds-averaged Navier–Stokes (RANS) equations solved via the finite volume method. We utilized the volume fraction method to accurately capture the free surface and employed the overset grid technique to manage the relative motion between the aircraft and the liquid surface. Our approach involves establishing a numerical simulation scheme to investigate the water-planing motion of amphibious aircraft across varying wave heights, wavelengths, speeds, and center-of-gravity positions. The computational findings demonstrate a close match between calculated forces and aircraft motion compared to experimental values. Notably, we observed pronounced nonlinearity in wave-added resistance. Under high sea conditions, operating in a short-wavelength environment or with a rearward center-of-gravity position proves advantageous for reducing wave-added resistance. Conversely, poor longitudinal stability is evident during planing in long waves.

Free radicals, including reactive oxygen species (ROS) and reactive nitrogen species (RNS), play critical roles in various physiological activities such as cell differentiation, apoptosis, and vascular tension when existing in cells at low levels. However, excessive amounts of free radicals are harmful, causing DNA damage, lipid peroxidation, protein degeneration, and abnormal cell death. Certain viral infections induce cells to produce excessive free radicals, which in multiple ways help the virus to replicate, mature, and exit. Iron is a necessary element for many intracellular enzymes, involved in both cellular activities and viral replication. Ferroptosis, a programmed cell death mode distinct from apoptosis, necrosis, and pyroptosis, is characterized by lipid peroxide accumulation and damage to the antioxidant system, affecting many cellular processes. Viral infection commonly manifests as decreased glutathione (GSH) content and down-regulated glutathione peroxidase 4 (GPX4) activity, similar to ferroptosis. Recent studies have suggested a possible relationship among free radicals, viral infections and ferroptosis. This review aims to elucidate the molecular mechanism linking free radicals and ferroptosis during viral infections and provide a new theoretical basis for studying viral pathogenesis and control.

Objective: Wheat is an alternative to corn silage for ruminant feeding in northern China. This study examined the effects of harvest time and added molasses on nutritional content, ensiling characteristics and in vitro degradation of whole crop wheat (WCW). Methods: Fresh WCW at the milk-ripe stage was harvested at 0700 h (i.e., in the morning [Mo]) and 1700 h (i.e., in the afternoon [Af]), and then immediately used to prepare silage and make hay. Commercial molasses was added to Af WCW at 0%, 2%, 4%, and 6% (fresh weight) proportions. The WCW treated with molasses was mixed thoroughly prior to ensiling. Results: Dry matter (DM), neutral detergent fiber, water soluble carbohydrate (WSC) content (p<0.01), accumulative gas production in 72 h (GP72h, 77.46 mL/g vs 95.15 mL/g) and dry matter disappearance in vitro (69.15% vs 76.77%) were lower (p<0.05), while crude protein (CP) content was higher for WCW silage (WCWS) compared to WCW (p<0.01). The propionic acid and butyric acid concentrations in WCWS from Mo WCW were 1.47% and 0.26%, respectively. However, the propionic and butyric acid concentrations were negligible, while the ammonia nitrogen/total nitrogen (NH3-N/TN, p<0.01) concentration was lower and the rate of gas production at 50% of the maximum (17.05 mL/h vs 13.94 mL/h, p<0.05) was higher for Af WCWS compared to Mo WCWS. The incubation fluid's NH3-N concentration was lower in WCWS and Af WCW compared to Mo WCW (p<0.05). The CP and WSC content increased with increasing molasses levels (p<0.05). Furthermore, the pH (p<0.01) and time when gas production was 50% of the maximum (2.78 h vs 3.05 h, p<0.05) were lower in silage treated with 4% molasses than silage without molasses. Conclusion: Harvesting wheat crops in the afternoon and adding molasses at 4% level to WCW optimally improved ensiling characteristics, leading to well-preserved silage. [ABSTRACT FROM AUTHOR]

Mycoplasma synoviae (MS) is an important avian pathogen causing considerable economic hardship in the poultry industry. A major inflammation caused by MS is synovitis that occurs in the synovial tendon sheath and joint synovium. However, the overall appearance of pathological changes in the tendon sheath and surrounding tissues caused by MS infection at the level of pathological tissue sections was poor. Studies on the role of MS and synovial sheath cells (SSCs) interaction in the development of synovitis have not been carried out. Through histopathological observation, our study found that a major MS-induced pathological change of the tendon sheath synovium was extensive scattered and focal inflammatory cell infiltration of the tendon sheath synovial layer. In vitro research experiments revealed that the CFU numbers of MS adherent and invading SSC, the levels of expression of various pattern recognition receptors, inflammatory cytokines, and chemokines coding genes, such as IL-1β, IL-6, IL-8, CCL-20, RANTES, MIP-1β, TLR7, and TLR15 in SSCs, and chemotaxis of macrophages were significantly increased when the multiplicity of infection (MOI) of MS to SSC were increased tenfold. The expression level of IL-12p40 in SSC was significantly higher when the MOIs of MS to SSC were increased by a factor of 100. The interaction between MS and SSC can activate macrophages, which was manifested by a significant increase in the expression of IL-1β, IL-6, IL-8, CCL-20, RANTES, MIP-1β, and CXCL-13. This study systematically demonstrated that the interaction of MS with chicken SSC contributes to the inflammatory response caused by the robust expression of related cytokines and macrophage chemotaxis. These findings are helpful in elucidating the molecular mechanism of MS-induced synovitis in chickens.

Feeding high-quality forage diets may lead to excessive weight gains and over-conditioning for dairy heifers. Restriction of energy density and dry matter intake by using low-energy forages, such as straw, is a good approach for controlling this problem. Alfalfa stems contain high fiber and moderate protein content and have the potential to be used to replace straw to reduce dietary energy. The objective of this study was to compare nutrient intakes, digestibilities, growth performance, and feeding behaviors of dairy heifers offered an alfalfa silage/corn silage high-energy diet (HE; 13.1% crude protein, 65.4% total digestible nutrients, 39.7% neutral detergent fiber) with 2 energy-diluted diets that replaced various proportions of the corn or alfalfa silages with either alfalfa stemlage (STM; 12.6% crude protein, 59.1% total digestible nutrients, 46.4% neutral detergent fiber) or chopped wheat straw (WS; 12.6% crude protein, 61.9% total digestible nutrients, 43.7% neutral detergent fiber). Seventy-two pregnant Holstein heifers (16.8 ± 1.3 mo) were stratified into 3 blocks (24 heifers/block) by initial body weight (light, 440 ± 18.0 kg; medium, 486 ± 18.6 kg; heavy, 534 ± 25.1 kg), with each block composed of 3 pens (8 heifers/pen), with diets assigned randomly to 1 pen within the block. Diets were offered in a 56-d feeding trial. Both dry matter intake and energy intake were decreased with the addition of low-energy forages to the diets, but no differences in dry matter intake were observed across diluted diets. Digestibility of dry matter, organic matter, neutral detergent fiber, and apparent N were greater for HE compared with diluted diets, and for WS compared with STM. Total body weight gain (74 vs. 56 kg) and average daily gain (1.32 vs. 1.00 kg/d) were greater for heifers offered HE compared with diluted diets. Feed efficiency tended to be less for heifers offered the diluted diets compared with HE (10.7 vs. 8.6 kg of feed/kg of gain). Heifers did not sort for or against particles when offered HE. However, increased sorting behavior was observed for diluted diets. Compared with ad libitum feeding dairy heifers a diet with high nutrient content forages (corn silage and alfalfa silage), use of diet diluted with alfalfa stemlage or wheat straw is an effective feeding management strategy to control total daily dry matter and energy intake by increasing gut fill, and maintain desirable body condition and growth rates, even though the diluted diets had greater sortability. Abstract Background Duck hepatitis A virus type 3 (DHAV-3) is one of the most harmful pathogens in the duck industry. However, the molecular mechanism underlying DHAV-3 infection in ducklings remains poorly understood. To study the genetic regulatory network for miRNA-mRNA and the signaling pathways involved in DHAV-3 infection in ducklings, we conducted global miRNA and mRNA expression profiling of duckling liver tissues infected with lethal DHAV-3 by high-throughput sequencing. Results We found 156 differentially expressed miRNAs (DEMs) and 7717 differentially expressed genes (DEGs) in livers of mock-infected and DHAV-3-infected duckling. A total of 19,606 miRNA-mRNA pairs with negatively correlated expression patterns were identified in miRNA-mRNA networks constructed on the basis of these DEMs and DEGs. Moreover, immune-related pathways, including the cytokine-cytokine receptor interaction, apoptosis, Toll-like receptor, Jak-STAT, and RIG-I-like receptor signaling pathway, were significantly enriched through analyzing functions of mRNAs in the network in response to DHAV-3 infection. Furthermore, apl-miR-32-5p, apl-miR-125-5p, apl-miR-128-3p, apl-miR-460-5p, and novel-m0012-3p were identified as potential regulators in the immune-related signaling pathways during DHAV-3 infection. And some host miRNAs were predicted to target the DHAV-3 genome. Conclusions This is the first integrated analysis of miRNA and mRNA in DHAV-3-infected ducklings. The results indicated the important roles of miRNAs in regulating immune response genes and revealed the immune related miRNA-mRNA regulation network in the DHAV-3-infected duckling liver. These findings increase our knowledge of the roles of miRNAs and their target genes in DHAV-3 replication and pathogenesis. They also aid in the understanding of host-virus interactions.

In order to solve the interception problem of escaping targets under environmental interference and inaccurate target perception, this paper introduces the guidance interception algorithm and introduces a Kalman filter to predict the trajectory of escaping targets. In order to make a USV pursuit under a disturbance in a time-varying real marine environment, a disturbance observer is proposed to estimate the external disturbance and model uncertainty. The simulation of pursuit–evasion results shows that the interception algorithm designed enables the pursuer to intercept the escaping target in a time-varying, disturbed marine environment. Both the simulation and experimental results are provided to validate the effectiveness of the method.

This experiment was conducted to investigate the effect of energy density (ED) in the close-up period and supplementation of extruded full-fat soybean (ESB) during the first 4 wk after parturition on intake, body weight (BW), metabolic status, and performance of dairy cows. Fifty-seven Chinese Holstein cows with similar parity, previous 305-d milk yield, and expected calving date were dried off at -60 d relative to parturition and fed the standard herd dry-cow diet until -21 d relative to parturition. Energy density at low (LED, 1.25 Mcal/kg), medium (MED, 1.41 Mcal/kg) or high (HED, 1.55 Mcal/kg) levels of the close-up diets and postpartum supplementation of ESB at 0 kg/d (control, CON) or 1.5 kg/d (TRT) were used in a 3 x 2 factorial arrangement. All cows received the same CON diet from wk 5 to wk 8. As ED increased in the close-up diet, cows had higher dry matter intake and gained more BW and body condition score, and consequently were in improved energy balance status during the prepartum period, but lost more BW and more body condition score during the first 8 wk of lactation. Compared with LED cows, HED cows had higher blood concentrations of insulin and glucose, and lower nonesterified fatty acids (NEFA) prepartum, but had lower insulin concentration, higher leptin concentration and tended to have higher NEFA concentration postpartum. Milk production was not affected by the prepartum ED, although HED cows produced approximately 2 kg/d less milk than MED and LED cows during early lactation. Postpartum ESB supplementation elevated blood glucagon concentration regardless of prepartum ED during the first 4 wk of lactation. Interactive effects between prepartum ED and postpartum ESB supplementation were observed in blood concentrations of insulin, NEFA, bilirubin, total protein, albumin, and globulin. During early lactation, TRT decreased globulin concentrations in MED cows, and reduced NEFA and bilirubin concentrations in HED cows. Compared with CON, TRT cows had higher yields of milk (34.32 vs. 36.53 kg/d), milk lactose (1.63 vs. 1.74 kg/d), and solids-nonfat (2.98 vs. 3.18 kg/d), tended to have a greater yield of milk protein (1.11 vs. 1.17 kg/d), but tended to have lower milk fat percentage (4.18 vs. 3.94%) during the first 4 wk of lactation. In conclusion, compared with feeding the HED diet, feeding the LED diet during the close-up dry period had positive carryover effects on metabolism and production during early lactation, and the MED diet showed no advantage over the LED diet. Extruded full-fat soybean supplementation during the first 4 wk of lactation had positive effects on postpartum metabolic status, especially for those receiving the MED or HED diet prepartum, and resulted in an overall improved milk production during early lactation. Mycoplasma synoviae is an important pathogen of poultry, causing significant economic losses in this industry. Analysis of the unique genes and shared genes among different M. synoviae strains and among related species is helpful for studying the molecular pathogenesis of M. synoviae and provides valuable molecular diagnostic targets to facilitate the identification of M. synoviae species. We selected a total of 46 strains, including six M. synoviae strains, from 25 major animal (including avian) Mycoplasma species/subspecies that had complete genome sequences and annotation information published in GenBank, and used them for comparative genomic analysis. After analysis, 16 common genes were found in the 46 strains. Thirteen single-copy core genes and the 16s rRNA genes were used for genetic evolutionary analysis. M. synoviae was found to have a distant evolutionary relationship not only with other arthritis-causing mycoplasmas, but also with another major avian pathogen, Mycoplasma gallisepticum, that shares the major virulence factor vlhA with M. synoviae. Subsequently, six unique coding genes were identified as shared among these M. synoviae strains that are absent in other species with published genome sequences. Two of the genes were found to be located in the genetically stable regions of the genomes of M. synoviae and were determined to be present in all M. synoviae isolated strains (n = 20) and M. synoviae-positive clinical samples (n = 48) preserved in our laboratory. These two genes were used as molecular diagnostic targets for which SYBR green quantitative PCR detection methods were designed. The two quantitative PCR methods exhibited good reproducibility and high specificity when tested on positive plasmid controls and genomic DNA extracted from different M. synoviae strains, other major avian pathogenic bacteria/mycoplasmas, and low pathogenic Mycoplasma species. The detection limit for the two genes was 10 copies or less per reaction. The clinical sensitivity and specificity of the quantitative PCR methods were both 100% based on testing chicken hock joint samples with positive or negative M. synoviae infection. This research provides a foundation for the study of species-specific differences and molecular diagnosis of M. synoviae.

A total of 182 beef samples were minced and divided into calibration set (n=140) and independent validation set (n=42). Calibration models of NIRS (1000-1800 nm) were built using partial least squares regression (PLSR) on the calibration set of samples. Both the coefficient of determination in calibration (R2C) and the coefficient of determination in prediction (R2P) were over 0.98 for all chemical compositions. The ratio performance deviation (RPD) was 17.37, 5.12 and 10.43 for fat, protein and moisture, respectively. Mycoplasma synoviae (MS), which causes respiratory disease, eggshell apex abnormalities, infectious synovitis, and arthritis in avian species, has become an economically detrimental poultry pathogen in recent years. In China, the disease is characterized by infectious synovitis and arthritis. However, the mechanism by which MS causes infectious synovitis and arthritis remains unknown. Increasing evidence suggests that synovial fibroblasts (SF) play a key role in the pathogenesis of arthritis. Here, both RNA sequencing and tandem mass tag analyses are utilized to compare the response of primary chicken SF (CSF) following infection with and without MS. The host response between non-infected and infected cells was remarkably different at both the mRNA and protein levels. In total, 2,347 differentially expressed genes (DEGs) (upregulated, n = 1,137; downregulated, n = 1,210) and 221 differentially expressed proteins (DEPs) (upregulated, n = 129; downregulated, n = 92) were detected in the infected group. A correlation analysis indicated a moderate positive correlation between the mRNA and protein level changes in MS-infected CSF. At both the transcriptomic and proteomic levels, 149 DEGs were identified; 88 genes were upregulated and 61 genes were downregulated in CSF. Additionally, part of these regulated genes and their protein products were grouped into seven categories: proliferation-related and apoptosis-related factors, inflammatory mediators, proangiogenic factors, antiangiogenic factors, matrix metalloproteinases, and other arthritis-related proteins. These proteins may be involved in the pathogenesis of MS-induced arthritis in chickens. To our knowledge, this is the first integrated analysis on the mechanism of CSF-MS interactions that combined transcriptomic and proteomic technologies. In this study, many key candidate genes and their protein products related to MS-induced infectious synovitis and arthritis were identified.

This work focused on the influence of hydrogen content on the microstructure and mechanical properties of ER5183 Al-Mg-Mn alloy wires for aluminum alloy welding. The hydrogen content of the ER5183 wires was measured, the macroscopic and microscopic morphologies of fractures were observed as well as the microstructure of the wires, and the tensile strength of the wires was also tested and investigated. The experimental results demonstrated three typical irregular macroscopic fractures of the wires appeared during the drawing process when the hydrogen content exceeded 0.23 μg/g. In the meantime, the aggregated pores were observed in the microstructure of the ϕ5.2 mm wire with the hydrogen content of 0.38 μg/g. Such defects may become the origin of cracks in subsequent processing and tensile tests. Moreover, higher hydrogen content in the ϕ5.2 mm welding wire will bring obvious changes in the fracture surface, which are internal cracks and micropores replacing the original uniform and compact dimples. With the higher hydrogen content, the tensile strength and plastic strain rate of ϕ1.2 mm wires would decrease. At the same time, unstable crack propagation would occur during the process of plastic deformation, leading to fracture. Considering the mechanical properties and microstructure, the hydrogen content of the ER5183 wires should be controlled below 0.23 μg/g.

The microstructure and properties of a Cu/304 stainless steel dissimilar metal joint brazed with a low silver Ag16.5CuZnSn-xGa-yCe braze filler after aging treatment were investigated. The results indicated that the addition of Ce could reduce the intergranular penetration depth of the filler metal into the stainless steel during the aging process. The minimum penetration depth in the Ag16.5CuZnSn-0.15Ce brazed joint was decreased by 48.8% compared with the Ag16.5CuZnSn brazed joint. Moreover, the shear strength of the brazed joint decreased with aging time while the shear strength of the AgCuZnSn-xGa-yCe joint was still obviously higher than the Ag16.5CuZnSn joint after a 600 h aging treatment. The fracture type of the Ag16.5CuZnSn-xGa-yCe brazed joints before aging begins ductile and turns slightly brittle during the aging process. Compared to all the results, the Ag16.5CuZnSn-2Ga-0.15Ce brazed joints show the best performance and could satisfy the requirements for cost reduction and long-term use.

California is the leading dairy state in the United States, producing 21% of the nation's milk supply. The state's highest concentration of dairies is in the San Joaquin Valley, a region that violates federal limits for ozone and particulate matter in the air. Volatile organic compounds and greenhouse-gas emissions from dairies contribute to regional air-quality challenges and also play a role in climate change. We used an environmentally controlled chamber to monitor greenhouse-gas emissions from dairy cattle over a 24-hour period, and we measured the emissions from waste slurry using a simulated dairy waste lagoon. This research helps to quantify emissions from dairies in California and suggests possible approaches for their mitigation.

The aim of the study was to evaluate the variations in the rumen ecosystem at genus level and predicted functions of rumen bacteria by using whole crop corn silage (WCS), whole crop rice silage (WRS) or rice straw (RS) as forage sources in beef cattle ration. Ruminal digesta samples from 10 bulls per treatment were collected at day 60 of experimental period. The PCoA plots based on the Bray-Curtis distance matrix (BDM) expressed separation between WRS and WCS, WRS and RS using PC1 (P<0.05, 45.56%). The PCoA plots based on BDM also expressed separation among WRS, WCS and RS group using PC2 (P<0.05, 11.95%). Microbiota composition resutls at genus level showed that the most abundant genera were Prevotella (13.37%) and Ruminococcus (4.00%). Comparison of treatments represented that Prevotella, Treponema and Anaerostipes were higher in bulls fed WCS forage. Clostridium, Anaeroplasma and RFN20 were higher in bulls fed RS forage than animals fed WCS and WRS. Butyrivibrio was higher in WRS and RS treatments than WCS treatment group. Pseudobutyrivibrio was higher in bulls fed WRS forage than animals fed RS and WCS forage. Fibrobacter was higher in RS and WCS as compared to WRS. The results of functional alteration of rumen microbiota in different experimental groups represented that the leading modified function of the microbiome was the transporter. Transient receptor potential melastatin 7 (TRPM7), a non-selective cation channel, is highly expressed expressed in the brain and plays a critical role in ischemic neuronal death. Astrocyte, the most abundant cell type in central nervous system (CNS), exerts many essential functions in the physiological and pathological conditions. Here we investigated the expression and functions of the TRPM7 channel in mouse cortical astrocytes. Using reverse transcription (RT)-PCR, immunostaining, western blot and patch clamp recording, we showed that functional TRPM7 channel is expressed in cultured mouse cortical astrocytes. Knocking down TRPM7 with specific siRNA impairs the proliferation and migration of astrocytes by 40.2% ± 3.9% and 40.1% ± 11.5%, respectively. Consistently, inhibition of TRPM7 with 2-aminoethoxydiphenyl borate (2-APB) also decreases astrocyte proliferation and migration by 46.1% ± 2.5% and 64.2% ± 2.4%. MAPKs and Akt signaling pathways have been shown to be implicated in TRPM7-mediated responses including cell proliferation and migration. Our data show that suppression of TRPM7 in astrocytes reduces the phosphorylation of extracellular signal-regulated kinases (ERK) and c-Jun N-terminal kinases (JNK), but not p38 mitogen-activated protein kinase and Akt. In addition, TRPM7, as a cation channel, has been involved in the Ca²⁺ and Mg²⁺ homeostasis in several types of cells. In our study, we found that silencing TRPM7 decreases the intracellular basal Mg²⁺ concentration without affecting Ca²⁺ concentration in astrocytes. However, an addition of Mg²⁺ to the growth medium could not rescue the impaired proliferation of astrocytes. Together, our data suggest that TRPM7 channel may play a critical role in the proliferation and migration of astrocytes via the ERK and JNK pathways.

This study investigated the change of transient receptor potential melastatin 7 (TRPM7) expression by high glucose and its role in hyperglycemia induced injury of vascular endothelial cells. Human umbilical vein endothelial cells (HUVECs) were incubated in the presence or absence of high concentrations of D-glucose (HG) for 72 h. RT-PCR, Real-time PCR, Western blotting, Immunofluorescence staining and whole-cell patch-clamp recordings showed that TRPM7 mRNA, TRPM7 protein expression and TRPM7-like currents were increased in HUVECs following exposure to HG. In contrast to D-glucose, exposure of HUVECs to high concentrations of L-glucose had no effect. HG increased reactive oxygen species (ROS) generation, cytotoxicity and decreased endothelial nitric oxide synthase protein expression, which could be attenuated by knockdown of TRPM7 with TRPM7 siRNA. The protective effect of silencing TRPM7 against HG induced endothelial injury was abolished by U0126, an inhibitor of the extracellular signal-regulated kinase signaling pathway. These observations suggest that TRPM7 channels play an important role in hyperglycemia-induced injury of vascular endothelial cells.
