Abstract

Results are presented for the measurement of large-scale anisotropies in the arrival directions of ultra–high-energy cosmic rays detected at the Pierre Auger Observatory during 19 yr of operation, prior to AugerPrime, the upgrade of the observatory. The 3D dipole amplitude and direction are reconstructed above 4 EeV in four energy bins. Besides the established dipolar anisotropy in R.A. above 8 EeV, the Fourier amplitude of the 8–16 EeV energy bin is now also above the 5 σ discovery level. No time variation of the dipole moment above 8 EeV is found, setting an upper limit to the rate of change of such variations of 0.3% yr ^−1 at the 95% confidence level. Additionally, the results for the angular power spectrum are shown, demonstrating no other statistically significant multipoles. The results for the equatorial dipole component down to 0.03 EeV are presented, using for the first time a data set obtained with a trigger that has been optimized for lower energies. Finally, model predictions are discussed and compared with observations, based on two source emission scenarios obtained in the combined fit of spectrum and composition above 0.6 EeV.

Abstract

Abstract LOPES, the LOFAR prototype station, was an antenna array for cosmic-ray air showers operating from 2003 to 2013 within the KASCADE-Grande experiment. Meanwhile, the analysis is finished and the data of air-shower events measured by LOPES are available with open access in the KASCADE Cosmic Ray Data Center (KCDC). This article intends to provide a summary of the achievements, results, and lessons learned from LOPES. By digital, interferometric beamforming the detection of air showers became possible in the radio-loud environment of the Karlsruhe Institute of Technology (KIT). As a prototype experiment, LOPES tested several antenna types, array configurations and calibration techniques, and pioneered analysis methods for the reconstruction of the most important shower parameters, i.e., the arrival direction, the energy, and mass-dependent observables such as the position of the shower maximum. In addition to a review and update of previously published results, we also present new results based on end-to-end simulations including all known instrumental properties. For this, we applied the detector response to radio signals simulated with the CoREAS extension of CORSIKA, and analyzed them in the same way as measured data. Thus, we were able to study the detector performance more accurately than before, including some previously inaccessible features such as the impact of noise on the interferometric cross-correlation beam. These results led to several improvements, which are documented in this paper and can provide useful input for the design of future cosmic-ray experiments based on the digital radio-detection technique.

Abstract

A search for time-directional coincidences of ultra-high-energy (UHE) photons above 10 EeV with gravitational wave (GW) events from the LIGO/Virgo runs O1 to O3 is conducted with the Pierre Auger Observatory. Due to the distinctive properties of photon interactions and to the background expected from hadronic showers, a subset of the most interesting GW events is selected based on their localization quality and distance. Time periods of 1000 s around and 1 day after the GW events are analyzed. No coincidences are observed. Upper limits on the UHE photon fluence from a GW event are derived that are typically at ∼7 MeV cm ^−2 (time period 1000 s) and ∼35 MeV cm ^−2 (time period 1 day). Due to the proximity of the binary neutron star merger GW170817, the energy of the source transferred into UHE photons above 40 EeV is constrained to be less than 20% of its total GW energy. These are the first limits on UHE photons from GW sources.

Abstract

A catalog containing details of the highest-energy cosmic rays recorded through the detection of extensive air showers at the Pierre Auger Observatory is presented with the aim of opening the data to detailed examination. Descriptions of the 100 showers created by the highest-energy particles recorded between 2004 January 1 and 2020 December 31 are given for cosmic rays that have energies in the range 78–166 EeV. Details are also given on a further nine very energetic events that have been used in the calibration procedure adopted to determine the energy of each primary. A sky plot of the arrival directions of the most energetic particles is shown. No interpretations of the data are offered.

Abstract

Abstract The ‘KASCADE Cosmic ray Data Centre’ is a web portal (https://kcdc.ikp.kit.edu), where the data of the astroparticle physics experiment KASCADE-Grande are made available for the interested public. The KASCADE experiment was a large-area detector for the measurement of high-energy cosmic rays via the detection of extensive air showers. The multi-detector installations KASCADE and its extension KASCADE-Grande stopped the active data acquisition in 2013 after more than 20 years of data taking. In several updates since our first release in 2013 with KCDC we provide the public measured and reconstructed parameters of more than 433 million air showers. In addition, KCDC provides meta data information and documentation to enable a user outside the community of experts to perform their own data analysis. Simulation data from three different high energy interaction models have been made available as well as a compilation of measured and published spectra from various experiments. In addition, detailed educational examples shall encourage high-school students and early stage researchers to learn about astroparticle physics, cosmic radiation as well as the handling of Big Data and about the sustainable and public provision of scientific data.

Abstract

The radio technique is a promising method for detection of cosmic-ray air showers of energies around 100PeV and higher with an array of radio antennas. Since the amplitude of the radio signal can be measured absolutely and increases with the shower energy, radio measurements can be used to determine the air-shower energy on an absolute scale. We show that calibrated measurements of radio detectors operated in coincidence with host experiments measuring air showers based on other techniques can be used for comparing the energy scales of these host experiments. Using two approaches, first via direct amplitude measurements, and second via comparison of measurements with air shower simulations, we compare the energy scales of the air-shower experiments Tunka-133 and KASCADE-Grande, using their radio extensions, Tunka-Rex and LOPES, respectively. Due to the consistent amplitude calibration for Tunka-Rex and LOPES achieved by using the same reference source, this comparison reaches an accuracy of approximately 10% – limited by some shortcomings of LOPES, which was a prototype experiment for the digital radio technique for air showers. In particular we show that the energy scales of cosmic-ray measurements by the independently calibrated experiments KASCADE-Grande and Tunka-133 are consistent with each other on this level. Abstract

KASCADE-Grande is an extensive air shower experiment devoted to the study of cosmic rays in the 1016 ÷ 1018 eV energy range. The array is composed by different detectors allowing independent measurements of the number of muons (Nμ) and charged particles (Nch) of extensive air showers (EAS). These two observables are then used to study the primary energy spectrum, separating the events in two samples, on the basis of the shower size ratio, corrected for attenuation in the atmosphere, lnNμ/ lnNch. The two samples represent the light and heavy mass groups of the primaries. In the studied energy range only the spectrum of heavy primaries shows a significant change of slope, the energy (estimated using the QGSJET II hadronic interaction model) of this feature is in agreeement with the expectations of a rigidity dependent knee feature.

Using CORSIKA, we determine typical energies and phase space regions of secondary particles which are important for muon production in extensive air showers and confront the results with existing fixed target measurements. Abstract

Abstract  The electron and muon number of extensive air showers (EAS) measured at ground level are the main observables of the detector array of the KASCADE experiment. The two-dimensional frequency distribution of these observables is used for reconstructing the energy spectra of five elemental groups representing the chemical composition of primary cosmic rays in the energy range from 1015 eV to 1017 eV. The results of the presented unfolding analysis depend crucially on simulations of EAS, and therefore on the hadronic interaction models used for their generation. As it turns out, the results for the individual energy spectra show to be sensitive to the characteristics of the interaction model used. Furthermore, none of the models discussed (QGSJet and SIBYLL) is capable to describe the data consistently over the whole measurement range with discrepancies appearing in different energy regions. Abstract  KASCADE-Grande is a multi-detector setup to get redundant information on single air shower basis. The information is used to perform multi-parameter analyses to solve the threefold problem of the reconstruction of the unknown primary energy, the primary mass, and to quantify the characteristics of the hadronic interactions in the air-shower development. This contribution discusses the various ways of testing the hadronic interaction mechanisms with data of the original KASCADE experiment and their results, as well as the capabilities in testing the models with the extension of KASCADE, the KASCADE-Grande experiment. Though no hadronic interaction model is fully able to describe the multi-parameter data of KASCADE consistently, the more recent models or improved versions of older models reproduce the data better than a few years ago. Abstract

We present the results of the search for large-scale anisotropies in the arrival directions of cosmic rays performed with the KASCADE-Grande experiment at energies higher than eV. To eliminate spurious anisotropies due to atmospheric or instrumental effects we apply the east–west method. We show, using the solar time distribution of the number of counts, that this technique allow us to remove correctly the count variations not associated to real anisotropies. By applying the east–west method we obtain the distribution of number of counts in intervals of 20 minutes of sidereal time. This distribution is then analyzed by searching for a dipole component; the significance of the amplitude of the first harmonic is 3.5σ, therefore, we derive its upper limit. The phase of the first harmonic is determined with an error of a few hours and is in agreement with the measurements obtained in the 1014 < E < 2 × 1015 eV energy range by the EAS-TOP, IceCube, and IceTop experiments. This supports the hypothesis of a change of the phase of the first harmonic at energies greater than ∼2 × 1014 eV. [ABSTRACT FROM AUTHOR]

Abstract

We show, for the first time, radio measurements of the depth of shower maximum (X&#95;{max}) of air showers induced by cosmic rays that are compared to measurements of the established fluorescence method at the same location. Using measurements at the Pierre Auger Observatory we show full compatibility between our radio and the previously published fluorescence dataset, and between a subset of air showers observed simultaneously with both radio and fluorescence techniques, a measurement setup unique to the Pierre Auger Observatory. Furthermore, we show radio X&#95;{max} resolution as a function of energy and demonstrate the ability to make competitive high-resolution X&#95;{max} measurements with even a sparse radio array. With this, we show that the radio technique is capable of cosmic-ray mass composition studies, both at Auger and at other experiments.

Abstract

Instantons, which are nonperturbative solutions to Yang-Mills equations, provide a signal for the occurrence of quantum tunneling between distinct classes of vacua. They can give rise to decays of particles otherwise forbidden. Using data collected at the Pierre Auger Observatory, we search for signatures of such instanton-induced processes that would be suggestive of super-heavy particles decaying in the Galactic halo. These particles could have been produced during the post-inflationary epoch and match the relic abundance of dark matter inferred today. The nonobservation of the signatures searched for allows us to derive a bound on the reduced coupling constant of gauge interactions in the dark sector: α&#95;{X}≲0.09, for 10^{9}≲M&#95;{X}/GeV<10^{19}. Conversely, we obtain that, for instance, a reduced coupling constant α&#95;{X}=0.09 excludes masses M&#95;{X}≳3×10^{13}  GeV. In the context of dark matter production from gravitational interactions alone, we illustrate how these bounds are complementary to those obtained on the Hubble rate at the end of inflation from the nonobservation of tensor modes in the cosmological microwave background.

Abstract

Although our understanding of centromere maintenance, marked by the histone H3 variant CenH3(CENP-A) in most eukaryotes, has progressed, the mechanism underlying the de novo formation of centromeres remains unclear. We used a synthetic system to dissect how CenH3(CENP-A) contributes to the accumulation of CENP-C and CENP-T, two key components that are necessary for the formation of functional kinetochores. We find that de novo CENP-T accumulation depends on CENP-C and that recruitment of these factors requires two domains in CenH3(CENP-A): the HJURP-binding region (CATD) and the CENP-C-binding region (CAC). Notably, HJURP interacts directly with CENP-C and is critical for de novo accumulation of CENP-C at synthetic centromeres. Abstract

We report the first direct measurement of the overall characteristics of microwave radio emission from extensive air showers. Using a trigger provided by the KASCADE-Grande air shower array, the signals of the microwave antennas of the Cosmic-Ray Observation via Microwave Emission experiment have been read out and searched for signatures of radio emission by high-energy air showers in the GHz frequency range. Microwave signals have been detected for more than 30 showers with energies above 3×10^{16}  eV. The observations presented in this Letter are consistent with a mainly forward-directed and polarized emission process in the GHz frequency range. The measurements show that microwave radiation offers a new means of studying air showers at E≥10^{17}  eV.

Abstract

Eukaryotic cells critically depend on the correct regulation of intracellular vesicular trafficking to transport biological material. The Rab subfamily of small guanosine triphosphatases controls these processes by acting as a molecular on/off switch. To fulfill their function, active Rab proteins need to localize to intracellular membranes via posttranslationally attached geranylgeranyl lipids. Each member of the manifold Rab family localizes specifically to a distinct membrane, but it is unclear how this specific membrane recruitment is achieved. Here, we demonstrate that Rab-activating guanosine diphosphate/guanosine triphosphate exchange factors (GEFs) display the minimal targeting machinery for recruiting Rabs from the cytosol to the correct membrane using the Rab-GEF pairs Rab5A-Rabex-5, Rab1A-DrrA, and Rab8-Rabin8 as model systems. Specific mistargeting of Rabex-5/DrrA/Rabin8 to mitochondria led to catalytic recruitment of Rab5A/Rab1A/Rab8A in a time-dependent manner that required the catalytic activity of the GEF. Therefore, RabGEFs are major determinants for specific Rab membrane targeting.

Abstract

Despite over two decades of research, the mechanism of Rab targeting to specific intracellular membranes is still not completely understood. Present evidence suggests that the original hypothesis that the message for targeting resides solely in the hypervariable C-terminus is incorrect, and a second mechanism involving a GDF [GDI (guanine-nucleotide-dissociation inhibitor) displacement factor] to disrupt stable Rab-GDI complexes has only been shown to apply in one case, despite the need for targeting over 60 human Rab proteins. Evidence for the involvement of Rab-effector interactions has only been presented for a few cases or in a very specific context. There is mounting evidence that GEFs (guanine-nucleotide-exchange factors) are essential for membrane targeting, although contributions from additional factors are likely to be of importance, at least in specific cases.

Abstract

We report the observation of a steepening in the cosmic ray energy spectrum of heavy primary particles at about 8×10(16) eV. This structure is also seen in the all-particle energy spectrum, but is less significant. Whereas the "knee" of the cosmic ray spectrum at 3-5×10(15) eV was assigned to light primary masses by the KASCADE experiment, the new structure found by the KASCADE-Grande experiment is caused by heavy primaries. The result is obtained by independent measurements of the charged particle and muon components of the secondary particles of extensive air showers in the primary energy range of 10(16) to 10(18) eV. 25. The versatile Legionella effector protein DrrA.

Abstract

The human pathogen Legionella pneumophila is a bacterium that infects human cells and interferes with intracellular signaling. The Legionella protein DrrA is one of the numerous effectors that the bacterium translocates into the host cytosol. DrrA binds to the Legionella containing vacuole (LCV), an organelle in which Legionella survives and replicates, and recruits and activates the vesicular trafficking regulator Rab1 to redirect vesicular trafficking between the endoplasmatic reticulum and the Golgi. After depositing Rab1 at the LCV, DrrA covalently modifies Rab1 with an AMP moiety at a specific tyrosine residue (Tyr77), which is centrally located in the functionally important switch II region. This adenylylation reaction interferes with the deactivation of Rab1 by GTPase activating proteins (GAPs), thereby presumably prolonging the active state of the protein at the LCV. Here, we summarize the versatile properties of DrrA and speculate on the effects of Rab1-adenylylation.

Abstract

In the course of Legionnaires' disease, the bacterium Legionella pneumophila affects the intracellular vesicular trafficking of infected eukaryotic cells by recruiting the small guanosine triphosphatase (GTPase) Rab1 to the cytosolic face of the Legionella-containing vacuole. In order to accomplish this, the Legionella protein DrrA contains a specific guanine nucleotide exchange activity for Rab1 activation that exchanges guanosine triphosphate (GTP) for guanosine diphosphate on Rab1. We found that the amino-terminal domain of DrrA possesses adenosine monophosphorylation (AMPylation) activity toward the switch II region of Rab1b, leading to posttranslational covalent modification of tyrosine 77. AMPylation of switch II by DrrA restricts the access of GTPase activating proteins, thereby rendering Rab1b constitutively active.

Abstract

An enhanced chip-based detection platform was developed by integrating a surface acoustic wave biosensor of the Love-wave type with protein identification by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-ToF MS). The system was applied to characterize the interaction of aptamers with their cognate HIV-1 proteins. The aptamers, which target two proteins of HIV-1, were identified using an automated in vitro selection platform. For aptamers S66A-C6 and S68B-C5, which target the V3 loop of the HIV-1 envelope protein gp120, KD values of 406 and 791 nM, respectively, were measured. Aptamer S69A-C15 was shown to bind HIV-1 reverse transcriptase (HIV-1 RT) with a KD value of 637 nM when immobilized on the biosensor surface. HIV-1 RT was identified with high significance using MALDI-ToF MS even in crude protein mixtures. The V3-loop of gp120 could be directly identified when using chip-bound purified protein samples. From crude protein mixtures, it could be identified indirectly with high significance via its fusion-partner glutathione-S-transferase (GST). Our data show that the combination of the selectivity of aptamers with a sensitive detection by MS enables the reliable and quantitative analysis of kinetic binding events of protein solutions in real time.

Abstract

The nature of ultrahigh-energy cosmic rays (UHECRs) at energies >10(20) eV remains a mystery. They are likely to be of extragalactic origin, but should be absorbed within approximately 50 Mpc through interactions with the cosmic microwave background. As there are no sufficiently powerful accelerators within this distance from the Galaxy, explanations for UHECRs range from unusual astrophysical sources to exotic string physics. Also unclear is whether UHECRs consist of protons, heavy nuclei, neutrinos or gamma-rays. To resolve these questions, larger detectors with higher duty cycles and which combine multiple detection techniques are needed. Radio emission from UHECRs, on the other hand, is unaffected by attenuation, has a high duty cycle, gives calorimetric measurements and provides high directional accuracy. Here we report the detection of radio flashes from cosmic-ray air showers using low-cost digital radio receivers. We show that the radiation can be understood in terms of the geosynchrotron effect. Our results show that it should be possible to determine the nature and composition of UHECRs with combined radio and particle detectors, and to detect the ultrahigh-energy neutrinos expected from flavour mixing.

29. Automated high-throughput purification of 6xHis-tagged proteins.

Abstract

Methods based on immobilized-metal affinity chromatography (IMAC) technology for the isolation of 6xHis-tagged proteins offer a one-step purification process that is both robust and meets the challenge of quantitatively purifying thousands of proteins with differing structures and characteristics. To perform this method in a high-throughput, automated format, protocols have been developed that can be run on lab automation workstations. Ready-to-run protocols covering a wide range of protein purification and assay applications, and which rely on the well-established 6xHis-Ni-NTA IMAC technology, are available. An Ni-NTA magnetic bead-based protocol allows micro-scale purification of up to 15 microg of 6xHis-tagged protein per well. If larger amounts of purified protein are needed, a vacuum-controlled Ni-NTA resin-based process provides a convenient medium-scale method for purification of up to 300 microg of 6xHis-tagged protein in a 96-well format. This protocol has been adapted to further increase the yield of 6xHis-tagged proteins allowing purification of up to several milligrams of highly pure protein per well. The protocol can process 96 samples, each derived from up to 25 mL culture volume (E. coli), in less than 3 h. Examples of purification and assay applications using the protocols mentioned above, and data on reproducibility and cross-contamination-free processing are shown.
