Ulcerative colitis is an immune-mediated inflammatory bowel disease characterized by recurrent diarrhea, abdominal pain, and mucous, pus, and bloody stools. Both Traditional Chinese Medicine and western medicine have their own strengths in the prevention and control of ulcerative colitis. There is an urgent need of developing effective combination of Traditional Chinese Medicine and western medicine therapy, as well as integrative nursing interventions, for the treatment of ulcerative colitis. By searching the literature at home and abroad, this paper summarized the current research status of Traditional Chinese Medicine and western medicine treatment in the diagnosis and treatment of UC and related clinical nursing practice, in order to provide a certain theoretical basis for clinical treatment and nursing of ulcerative colitis. Hierarchical and periodic nanostructures of dielectrics or metals are highly demanded for wide applications in optical, electrical, biological, and quantum devices. In this work, programmable plasmonic–photonic hierarchical nanostructures are fabricated using a facile and effective method with high controllability and stable reproducibility. The fabrication involves colloidal self-assembly, metal film deposition, and pulsed laser-induced dewetting in sequence for controllably pairing metal nanostructures on dielectric nanospheres in either large area or a local precision. Au nanostructures including Au nanocrown (AuNC), large Au nanosphere (AuNS), and multiple small Au nanoparticles (AuNPs) have been paired one-on-one on assembled SiO2 nanosphere (SiO2NS) arrays, with size and shape controlled by correlating the laser fluence and irradiation time, and the Au film thickness. The fabricated hierarchical nanostructures demonstrate synergistic effect of the photonic effects from the monolayer SiO2NS arrays and the surface plasma resonance effect from the Au nanostructures. The dewetting induced metal film reshaping has been modeled theoretically corresponding to observed experimental results. We can directly “write” the plasmonic Au nanostructures on the photonic crystal array using a focused laser beam to form encode patterns, showing angle-dependent structural colors for anti-counterfeiting information storage and display in rigid/flexible and opaque/transparent devices. It provides a promising path to actively construct on-demand pixelated plasmonic–photonic arrays for optical multiplexing technology in sensing, information encryption, and display.

Bursting is a crucial form of firing in neurons, laden with substantial information. Studying it can aid in understanding the neural coding to identify human behavioral characteristics conducted by these neurons. However, the high-dimensionality of many neuron models imposes a difficult challenge in studying the generative mechanisms of bursting. On account of the high complexity and nonlinearity characteristic of these models, it becomes nearly impossible to theoretically study and analyze them. Thus, this paper proposed to address these issues by focusing on the midbrain dopamine neurons, serving as the central neuron model for the investigation of the bursting mechanisms and bifurcation behaviors exhibited by the neuron. In this study, we considered the dimensionality reduction of a high-dimensional neuronal model and analyzed the dynamical properties of the reduced system. To begin, for the original thirteen-dimensional model, using the correlation between variables, we reduced its dimensionality and obtained a simplified three-dimensional system. Then, we discussed the changing characteristics of the number of spikes within a burst by simultaneously varying two parameters. Finally, we studied the co-dimension-2 bifurcation in the reduced system and presented the bifurcation behavior near the Bogdanov-Takens bifurcation. [ABSTRACT FROM AUTHOR]

Interactions between proteins and glycans are critical to various biological processes. With databases of carbohydrate-interacting proteins and increasing amounts of structural data, the three-sided right-handed β-helix (RHBH) has emerged as a significant structural fold for glycan interactions. In this review, we provide an overview of the sequence, mechanistic, and structural features that enable the RHBH to interact with glycans. The RHBH is a prevalent fold that exists in eukaryotes, prokaryotes, and viruses associated with adhesin and carbohydrate-active enzyme (CAZyme) functions. An evolutionary trajectory analysis on structurally characterized RHBH-containing proteins shows that they likely evolved from carbohydrate-binding proteins with their carbohydrate-degrading activities evolving later. By examining three polysaccharide lyase and three glycoside hydrolase structures, we provide a detailed view of the modes of glycan binding in RHBH proteins. The 3-dimensional shape of the RHBH creates an electrostatically and spatially favorable glycan binding surface that allows for extensive hydrogen bonding interactions, leading to favorable and stable glycan binding. The RHBH is observed to be an adaptable domain capable of being modified with loop insertions and charge inversions to accommodate heterogeneous and flexible glycans and diverse reaction mechanisms. Understanding this prevalent protein fold can advance our knowledge of glycan binding in biological systems and help guide the efficient design and utilization of RHBH-containing proteins in glycobiology research. Introduction: The objective of this study was to examine the utility of protein kinase N1 (PKN1) as a biomarker of cardiac surgery-associated AKI (CSA-AKI). Methods: A prospective cohort study of 110 adults undergoing on-pump cardiac surgery was conducted. The associations between post-operative PKN1 and CSA-AKI, AKI severity, need for renal replacement therapy (RRT), duration of AKI, length of ICU stay, and post-operative hospital stay were evaluated. Results: Patients were categorized into three groups according to PKN1 tertiles. The incidence of CSA-AKI in the third tertile was 3.4-fold higher than that in the first. PKN1 was an independent risk factor for CSA-AKI. The discrimination of PKN1 to CSA-AKI assessed by ROC curve indicated that the AUC was 0.70, and the best cutoff was 5.025 ng/mL. This group (>5.025 ng/mL) was more likely to develop CSA-AKI (p < 0.001). The combined AUC of EuroSCORE, aortic cross-clamp time, and PKN1 was 0.82 (p < 0.001). A higher level of PKN1 was related to increased need for RRT, longer duration of AKI, and length of ICU and post-operative hospital stays. Conclusions: PKN1 could be a potential biomarker for the prediction of CSA-AKI. The combination of PKN1, EuroSCORE, and aortic cross-clamp time was likely to predict the occurrence of CSA-AKI. Y chromosomes control essential male functions in many species, including sex determination and fertility. However, because of obstacles posed by repeat-rich heterochromatin, knowledge of Y chromosome sequences is limited to a handful of model organisms, constraining our understanding of Y biology across the tree of life. Here, we leverage long single-molecule sequencing to determine the content and structure of the nonrecombining Y chromosome of the primary African malaria mosquito, Anopheles gambiae We find that the An. gambiae Y consists almost entirely of a few massively amplified, tandemly arrayed repeats, some of which can recombine with similar repeats on the X chromosome. Sex-specific genome resequencing in a recent species radiation, the An. gambiae complex, revealed rapid sequence turnover within An. gambiae and among species. Exploiting 52 sex-specific An. gambiae RNA-Seq datasets representing all developmental stages, we identified a small repertoire of Y-linked genes that lack X gametologs and are not Y-linked in any other species except An. gambiae, with the notable exception of YG2, a candidate male-determining gene. YG2 is the only gene conserved and exclusive to the Y in all species examined, yet sequence similarity to YG2 is not detectable in the genome of a more distant mosquito relative, suggesting rapid evolution of Y chromosome genes in this highly dynamic genus of malaria vectors. The extensive characterization of the An. gambiae Y provides a long-awaited foundation for studying male mosquito biology, and will inform novel mosquito control strategies based on the manipulation of Y chromosomes.

The Asian tiger mosquito, Aedes albopictus, is a highly successful invasive species that transmits a number of human viral diseases, including dengue and Chikungunya fevers. This species has a large genome with significant population-based size variation. The complete genome sequence was determined for the Foshan strain, an established laboratory colony derived from wild mosquitoes from southeastern China, a region within the historical range of the origin of the species. The genome comprises 1,967 Mb, the largest mosquito genome sequenced to date, and its size results principally from an abundance of repetitive DNA classes. In addition, expansions of the numbers of members in gene families involved in insecticide-resistance mechanisms, diapause, sex determination, immunity, and olfaction also contribute to the larger size. Portions of integrated flavivirus-like genomes support a shared evolutionary history of association of these viruses with their vector. The large genome repertory may contribute to the adaptability and success of Ae. albopictus as an invasive species.

Variation in vectorial capacity for human malaria among Anopheles mosquito species is determined by many factors, including behavior, immunity, and life history. To investigate the genomic basis of vectorial capacity and explore new avenues for vector control, we sequenced the genomes of 16 anopheline mosquito species from diverse locations spanning ~100 million years of evolution. Comparative analyses show faster rates of gene gain and loss, elevated gene shuffling on the X chromosome, and more intron losses, relative to Drosophila. Some determinants of vectorial capacity, such as chemosensory genes, do not show elevated turnover but instead diversify through protein-sequence changes. This dynamism of anopheline genes and genomes may contribute to their flexible capacity to take advantage of new ecological niches, including adapting to humans as primary hosts.

BackgroundAnopheles stephensi is the key vector of malaria throughout the Indian subcontinent and Middle East and an emerging model for molecular and genetic studies of mosquito-parasite interactions. The type form of the species is responsible for the majority of urban malaria transmission across its range.ResultsHere, we report the genome sequence and annotation of the Indian strain of the type form of An. stephensi. The 221 Mb genome assembly represents more than 92% of the entire genome and was produced using a combination of 454, Illumina, and PacBio sequencing. Physical mapping assigned 62% of the genome onto chromosomes, enabling chromosome-based analysis. Comparisons between An. stephensi and An. gambiae reveal that the rate of gene order reshuffling on the X chromosome was three times higher than that on the autosomes. An. stephensi has more heterochromatin in pericentric regions but less repetitive DNA in chromosome arms than An. gambiae. We also identify a number of Y-chromosome contigs and BACs. Interspersed repeats constitute 7.1% of the assembled genome while LTR retrotransposons alone comprise more than 49% of the Y contigs. RNA-seq analyses provide new insights into mosquito innate immunity, development, and sexual dimorphism.ConclusionsThe genome analysis described in this manuscript provides a resource and platform for fundamental and translational research into a major urban malaria vector. Chromosome-based investigations provide unique perspectives on Anopheles chromosome evolution. RNA-seq analysis and studies of immunity genes offer new insights into mosquito biology and mosquito-parasite interactions.

Intrinsic bursting neurons represent a common neuronal type that displays bursting patterns upon depolarization stimulation. These neurons can be described by a system of seven-dimensional equations, which pose a challenge for dynamical analysis. To overcome this limitation, we employed the projection reduction method to reduce the dimensionality of the model. Our approach demonstrated that the reduced model retained the inherent bursting characteristics of the original model. Following reduction, we investigated the bi-parameter bifurcation of the equilibrium point in the reduced model. Specifically, we analyzed the Bogdanov-Takens bifurcation that arises in the reduced system. Notably, the topological structure of the neuronal model near the bifurcation point can be effectively revealed with our proposed method. By leveraging the proposed projection reduction method, we could explore the bursting mechanism in the reduced Pospischil model with greater precision. Our approach offers an effective foundation for generating theories and hypotheses that can be tested experimentally. Furthermore, it enables links to be drawn between neuronal morphology and function, thereby facilitating a deeper understanding of the complex dynamical behaviors that underlie intrinsic bursting neurons. Soil ecosystem provides a guarantee for the ecological environment. Soil salinization is a typical land degradation problem. In order to explore the applicability of GF-5, Sentinel-1, and Sentinel-2 remote sensing data in soil salinity research, this study selected Golmud of Qaidam Basin in China as the study area, using particle swarm optimization-artificial neural network (PSO-ANN), partial least squares regression (PLSR), and whale optimization algorithm-extreme learning machine (WOA-ELM) to predict soil salinity. The research results indicated that the correlation between Sentinel-2 and electric conductivity (EC) was higher than GF-5 and Sentinel-1. The simulation accuracy of models based on Sentinel-2 was slightly higher than that based on GF-5 and Sentinel-1. The three models' mean values of root mean square error (RMSE), R2, and mean absolute error (MAE) of Sentinel-2 were slightly better than those of GF-5 and Sentinel-1, and the fitting accuracy of GF-5 was similar to that of Sentinel-1 on the whole. The result proved that Sentinel-1, GF-5, and Sentinel-2 were suitable for salinity monitoring because the strong penetrability and sensitivity of Sentinel-1, the high spectral resolution of GF-5, and the high spatial resolution of Sentinel-2. The performance of PSO-ANN was the best, and that of PLSR was the worst. The salinity mapping result of Sentinel-2 coupled with PSO-ANN proved that machine learning methods and spaceborne data could be used to study large-scale soil salinization. [ABSTRACT FROM AUTHOR]
