Background: Standard methods for analysing data from large-scale assessments (LSA) cannot merely be adopted if hierarchical (or multilevel) regression modelling should be applied. Currently various approaches exist; they all follow generally a design-based model of estimation using the pseudo maximum likelihood method and adjusted weights for the corresponding hierarchies. Specifically, several different approaches to using and scaling sampling weights in hierarchical models are promoted, yet no study has compared them to provide evidence of which method performs best and therefore should be preferred. Furthermore, different software programs implement different estimation algorithms, leading to different results. Objective and method: In this study, we determine based on a simulation, the estimation procedure showing the smallest distortion to the actual population features. We consider different estimation, optimization and acceleration methods, and different approaches on using sampling weights. Three scenarios have been simulated using the statistical program R. The analyses have been performed with two software packages for hierarchical modelling of LSA data, namely Mplus and SAS. Results and conclusions: The simulation results revealed three weighting approaches performing best in retrieving the true population parameters. One of them implies using only level two weights (here: final school weights) and is because of its simple implementation the most favourable one. This finding should provide a clear recommendation to researchers for using weights in multilevel modelling (MLM) when analysing LSA data, or data with a similar structure. Further, we found only little differences in the performance and default settings of the software programs used, with the software package Mplus providing slightly more precise estimates. Different algorithm starting settings or different accelerating methods for optimization could cause these distinctions. However, it should be emphasized that with the recommended weighting approach, both software packages perform equally well. Finally, two scaling techniques for student weights have been investigated. They provide both nearly identical results. We use data from the Programme for International Student Assessment (PISA) 2015 to illustrate the practical importance and relevance of weighting in analysing large-scale assessment data with hierarchical models.

Over the course of the COVID-19 pandemic, Generalized Additive Models (GAMs) have been successfully employed on numerous occasions to obtain vital data-driven insights. In this article we further substantiate the success story of GAMs, demonstrating their flexibility by focusing on three relevant pandemic-related issues. First, we examine the interdepency among infections in different age groups, concentrating on school children. In this context, we derive the setting under which parameter estimates are independent of the (unknown) case-detection ratio, which plays an important role in COVID-19 surveillance data. Second, we model the incidence of hospitalizations, for which data is only available with a temporal delay. We illustrate how correcting for this reporting delay through a nowcasting procedure can be naturally incorporated into the GAM framework as an offset term. Third, we propose a multinomial model for the weekly occupancy of intensive care units (ICU), where we distinguish between the number of COVID-19 patients, other patients and vacant beds. With these three examples, we aim to showcase the practical and 'off-the-shelf' applicability of GAMs to gain new insights from real-world data. [ABSTRACT FROM AUTHOR]

Our objective is the estimation of voter transitions between two consecutive parliamentary elections. Usually, such analyses have been based either on individual survey data or on aggregated data. To move beyond these methods and their respective problems, we propose the application of so-called hybrid models, which combine aggregate and individual data. We use a Bayesian approach and extend a multinomial-Dirichlet model proposed in the ecological inference literature. Our new hybrid model has been implemented in the R-package eiwild (= Ecological Inference with individual-level data). Based on extensive simulations, we are able to show that our new estimator exhibits a very good estimation performance in many realistic scenarios. Application case is the voter transition between the Bavarian Regional election and the German federal elections 2013 in the Metropolitan City of Munich. Our approach is also applicable to other areas of electoral research, market research, and epidemiology.

SARS-CoV-2 antibody quantity and quality are key markers of humoral immunity. However, there is substantial uncertainty about their durability. We investigated levels and temporal change of SARS-CoV-2 antibody quantity and quality. We analyzed sera (8 binding, 4 avidity assays for spike-(S-)protein and nucleocapsid-(N-)protein; neutralization) from 211 seropositive unvaccinated participants, from the population-based longitudinal TiKoCo study, at three time points within one year after infection with the ancestral SARS-CoV-2 virus. We found a significant decline of neutralization titers and binding antibody levels in most assays (linear mixed regression model, p<0.01). S-specific serum avidity increased markedly over time, in contrast to N-specific. Binding antibody levels were higher in older versus younger participants – a difference that disappeared for the asymptomatic-infected. We found stronger antibody decline in men versus women and lower binding and avidity levels in current versus never-smokers. Our comprehensive longitudinal analyses across 13 antibody assays suggest decreased neutralization-based protection and prolonged affinity maturation within one year after infection. Real-time surveillance is a crucial element in the response to infectious disease outbreaks. However, the interpretation of incidence data is often hampered by delays occurring at various stages of data gathering and reporting. As a result, recent values are biased downward, which obscures current trends. Statistical nowcasting techniques can be employed to correct these biases, allowing for accurate characterization of recent developments and thus enhancing situational awareness. In this paper, we present a preregistered real-time assessment of eight nowcasting approaches, applied by independent research teams to German 7-day hospitalization incidences during the COVID-19 pandemic. This indicator played an important role in the management of the outbreak in Germany and was linked to levels of non-pharmaceutical interventions via certain thresholds. Due to its definition, in which hospitalization counts are aggregated by the date of case report rather than admission, German hospitalization incidences are particularly affected by delays and can take several weeks or months to fully stabilize. For this study, all methods were applied from 22 November 2021 to 29 April 2022, with probabilistic nowcasts produced each day for the current and 28 preceding days. Nowcasts at the national, state, and age-group levels were collected in the form of quantiles in a public repository and displayed in a dashboard. Moreover, a mean and a median ensemble nowcast were generated. We find that overall, the compared methods were able to remove a large part of the biases introduced by delays. Most participating teams underestimated the importance of very long delays, though, resulting in nowcasts with a slight downward bias. The accompanying prediction intervals were also too narrow for almost all methods. Averaged over all nowcast horizons, the best performance was achieved by a model using case incidences as a covariate and taking into account longer delays than the other approaches. For the most recent days, which are often considered the most relevant in practice, a mean ensemble of the submitted nowcasts performed best. We conclude by providing some lessons learned on the definition of nowcasting targets and practical challenges. Author summary: Current trends in epidemiological indicators are often obscured by the fact that recent values are still incomplete. This is due to reporting delays and other types of delays. Statistical nowcasting methods can be used to account for these biases and reveal yet unobserved trends, thereby improving situational awareness and supporting public health decision-making. While numerous methods exist for this purpose, little is known about their behavior in real-time settings and their relative performance. In this paper, we compared eight different nowcasting methods in an application to COVID-19 hospitalization incidences in Germany from November 2021 to April 2022. Additionally, we combined the predictions of these methods to create so-called ensemble nowcasts. Our findings indicate that while all methods yielded practically useful results, some systematic biases in nowcasts occurred and the remaining uncertainty was generally underestimated. Antibody studies analyze immune responses to SARS-CoV-2 vaccination and infection, which is crucial for selecting vaccination strategies. In the KoCo-Impf study, conducted between 16 June and 16 December 2021, 6088 participants aged 18 and above from Munich were recruited to monitor antibodies, particularly in healthcare workers (HCWs) at higher risk of infection. Roche Elecsys® Anti-SARS-CoV-2 assays on dried blood spots were used to detect prior infections (anti-Nucleocapsid antibodies) and to indicate combinations of vaccinations/infections (anti-Spike antibodies). The anti-Spike seroprevalence was 94.7%, whereas, for anti-Nucleocapsid, it was only 6.9%. HCW status and contact with SARS-CoV-2-positive individuals were identified as infection risk factors, while vaccination and current smoking were associated with reduced risk. Older age correlated with higher anti-Nucleocapsid antibody levels, while vaccination and current smoking decreased the response. Vaccination alone or combined with infection led to higher anti-Spike antibody levels. Increasing time since the second vaccination, advancing age, and current smoking reduced the anti-Spike response. The cumulative number of cases in Munich affected the anti-Spike response over time but had no impact on anti-Nucleocapsid antibody development/seropositivity. Due to the significantly higher infection risk faced by HCWs and the limited number of significant risk factors, it is suggested that all HCWs require protection regardless of individual traits.
