Abstract

Up to 80% of Parkinson's disease patients develop dementia, but time to dementia varies widely from motor symptom onset. Dementia with Lewy bodies presents with clinical features similar to Parkinson's disease dementia, but cognitive impairment precedes or coincides with motor onset. It remains controversial whether dementia with Lewy bodies and Parkinson's disease dementia are distinct conditions or represent part of a disease spectrum. The biological mechanisms underlying disease heterogeneity, in particular the development of dementia, remain poorly understood, but will likely be the key to understanding disease pathways and, ultimately, therapy development. Previous genome-wide association studies in Parkinson's disease and dementia with Lewy bodies/Parkinson's disease dementia have identified risk loci differentiating patients from controls. We collated data for 7804 patients of European ancestry from Tracking Parkinson's, The Oxford Discovery Cohort, and Accelerating Medicine Partnership-Parkinson's Disease Initiative. We conducted a discrete phenotype genome-wide association study comparing Lewy body diseases with and without dementia to decode disease heterogeneity by investigating the genetic drivers of dementia in Lewy body diseases. We found that risk allele rs429358 tagging APOEe4 increases the odds of developing dementia, and that rs7668531 near the MMRN1 and SNCA-AS1 genes and an intronic variant rs17442721 tagging LRRK2 G2019S on chromosome 12 are protective against dementia. These results should be validated in autopsy-confirmed cases in future studies.

9. Characterizing Interoceptive Differences in Autism: A Systematic Review and Meta-Analysis of Case-Control Studies

Abstract

Interoception, the body's perception of its own internal states, is thought to be altered in autism, though results of empirical studies have been inconsistent. The current study systematically reviewed and meta-analyzed the extant literature comparing interoceptive outcomes between autistic (AUT) and neurotypical (NT) individuals, determining which domains of interoception demonstrate robust between-group differences. A three-level Bayesian meta-analysis compared heartbeat counting performance, heartbeat discrimination performance, heartbeat counting confidence ratings, and self-reported interoceptive attention between AUT and NT groups (15 studies; n[subscript AUT] = 467, n[subscript NT] = 478). Autistic participants showed significantly reduced heartbeat counting performance [g = - 0.333, CrI[subscript 95%] (- 0.535, - 0.138)] and higher confidence in their heartbeat counting abilities [g = 0.430, CrI[subscript 95%] (0.123, 0.750)], but groups were equivalent on other meta-analyzed outcomes. Implications for future interoception research in autism are discussed.

Abstract

Although TP53 is the most commonly mutated gene in human cancers, the p53-dependent transcriptional programs mediating tumor suppression remain incompletely understood. Here, to uncover critical components downstream of p53 in tumor suppression, we perform unbiased RNAi and CRISPR-Cas9-based genetic screens in vivo. These screens converge upon the p53-inducible gene Zmat3, encoding an RNA-binding protein, and we demonstrate that ZMAT3 is an important tumor suppressor downstream of p53 in mouse KrasG12D-driven lung and liver cancers and human carcinomas. Integrative analysis of the ZMAT3 RNA-binding landscape and transcriptomic profiling reveals that ZMAT3 directly modulates exon inclusion in transcripts encoding proteins of diverse functions, including the p53 inhibitors MDM4 and MDM2, splicing regulators, and components of varied cellular processes. Interestingly, these exons are enriched in NMD signals, and, accordingly, ZMAT3 broadly affects target transcript stability. Collectively, these studies reveal ZMAT3 as a novel RNA-splicing and homeostasis regulator and a key component of p53-mediated tumor suppression.

Abstract

This trial examined whether a stepped care program for depression, which initiated treatment with internet cognitive behavioral therapy, including telephone and messaging support, and stepped up non-responders to telephone-administered cognitive behavioral therapy (tCBT), was noninferior, less costly to deliver, and as acceptable to patients compared to tCBT alone. Adults with a diagnosis of major depressive episode (MDE) were randomized to receive up to 20 weeks of stepped care or tCBT. Stepped care (n = 134) was noninferior to tCBT (n = 136) with an end-of-treatment effect size of d = 0.03 and a 6-month post-treatment effect size of d = -0.07 [90% CI 0.29 to 0.14]. Translation abstract LOIN for elements of the geological‐geotechnical ground modelThe ÖNORM EN 17412‐1 defines the Level of Information Need (LOIN) through clear specifications on requirements and responsibilities, as well as the integration of geometric and alphanumeric information and as well as documentation requirements. For geological‐geotechnical ground models with the aim of defining ground elements, the LOIN has not yet been described in more detail. Previous work focused mainly on the geometric realisation of ground models, in this paper the focus of the LOIN is set on the alphanumeric information. It is explained how geological information of the four planning phases the basic survey, exploration planning, exploration and interpretation can be implemented in digital models in a structured and comprehensive way to ensure a transfer without loss of information to geotechnical planning. A project‐related definition of the LOIN for selected ground elements, for example the element “borehole”, enables a precise and structured provision of data, avoids information overload and supports well‐founded decisions and efficient planning over the entire life cycle of a building. It emphasizes the need for a well‐defined LOIN, as well as early involvement and coordination with project stakeholders for effective geotechnical planning and ongoing modelling of geological data. Abstract

Background: Marrow stimulation is a common reparative approach to treat injuries to cartilage and other soft tissues (e.g., rotator cuff). It involves the recruitment of bone marrow elements and mesenchymal stem cells (MSCs) into the defect, theoretically initiating a regenerative process. However, the resulting repair tissue is often weak and susceptible to deterioration with time. The populations of cells at the marrow stimulation site (beyond MSCs), and their contribution to inflammation, vascularity, and fibrosis, may play a role in quality of the repair tissue. Summary: In this review, we accomplish three goals: (1) systematically review clinical trials on the augmentation of marrow stimulation and evaluate their assumptions on the biological elements recruited; (2) detail the cellular populations in bone marrow and their impact on healing; and (3) highlight emerging technologies and approaches that could better guide these specific cell populations towards enhanced cartilage or soft tissue formation. Key Messages: We found that most clinical trials do not account for cell heterogeneity, nor do they specify the regenerative element recruited, and those that do typically utilize descriptions such as "clots," "elements," and "blood." Furthermore, our review of bone marrow cell populations demonstrates a dramatically heterogenous cell population, including hematopoietic cells, immune cells, fibroblasts, macrophages, and only a small population of MSCs. Finally, the field has developed numerous innovative techniques to enhance the chondrogenic potential (and reduce the anti-regenerative impacts) of these various cell types. Abstract

The evolution of oxygenic photosynthesis during the Archean (4–2.5 Ga) required the presence of complementary reducing pathways to maintain the cellular redox balance. While the timing of the evolution of superoxide dismutases (SODs), enzymes that convert superoxide to hydrogen peroxide and O2, within bacteria and archaea is not resolved, the first SODs appearing in cyanobacteria contained copper and zinc in the reaction center (CuZnSOD). Here, we analyse growth characteristics, SOD gene expression (qRT‐PCR) and cellular enzyme activity in the deep branching strain, Pseudanabaena sp. PCC7367, previously demonstrated to release significantly more O2 under anoxic conditions. The observed significantly higher growth rates (p < 0.001) and protein and glycogen contents (p < 0.05) in anoxically cultured Pseudanabaena PCC7367 compared to control cultures grown under present‐day oxygen‐rich conditions prompted the following question: Is the growth of Pseudanabaena sp. PCC7367 correlated to atmospheric pO2 and cellular SOD activity? Expression of sodB (encoding FeSOD) and sodC (encoding CuZnSOD) strongly correlated with medium O2 levels (p < 0.001). Expression of sodA (encoding MnSOD) correlated significantly to SOD activity during the day (p = 0.019) when medium O2 concentrations were the highest. The cellular SOD enzyme activity of anoxically grown cultures was significantly higher (p < 0.001) 2 h before the onset of the dark phase compared to O2‐rich growth conditions. The expression of SOD encoding genes was significantly reduced (p < 0.05) under anoxic conditions in stirred cultures, as were medium O2 levels (p ≤ 0.001), compared to oxic‐grown cultures, whereas total cellular SOD activity remained comparable. Our data suggest that increasing pO2 negatively impacts the viability of early cyanobacteria, possibly by increasing photorespiration. Additionally, the increased expression of superoxide‐inactivating genes during the dark phase suggests the increased replacement rates of SODs under modern‐day conditions compared to those on early Earth. Abstract

Preclinical models of osteochondral defects (OCDs) are fundamental test beds to evaluate treatment modalities before clinical translation. To increase the rigor and reproducibility of translational science for a robust "go or no‐go," we evaluated disease progression and pain phenotypes within the whole joint for two OCD rat models with same defect size (1.5 x 0.8 mm) placed either in the trochlea or medial condyle of femur. Remarkably, we only found subtle transitory changes to gaits of rats with trochlear defect without any discernible effect to allodynia. At 8‐weeks post‐surgery, anatomical evaluations of joint showed early signs of osteoarthritis with EPIC‐microCT. For the trochlear defect, cartilage attenuation was increased in trochlear, medial, and lateral compartments of the femur. For condylar defect, increased cartilage attenuation was isolated to the medial condyle of the femur. Further, the medial ossicle showed signs of deterioration as indicated with decreased bone mineral density and increased bone surface area to volume ratio. Thus, OCD in a weight‐bearing region of the femur gave rise to more advanced osteoarthritis phenotype within a unilateral joint compartment. Subchondral bone remodeling was evident in both models without any indication of closure of the articular cartilage surface. We conclude that rat OCD, placed in the trochlear or condylar region of the femur, leads to differing severity of osteoarthritis progression. As found herein, repair of the defect with fibrous tissue and subchondral bone is insufficient to alleviate onset of osteoarthritis. Abstract

As biology education is obliged to enable students to acquire scientific literacy, long-lasting and sustainable learning is required. According to self-determination theory, such learning is based on self-determined motivation. Therefore, motivating students is a key issue of biology teaching. The present study investigated whether students' quality of motivation depends on the extent to which their basic needs are satisfied. Initial studies confirmed a positive impact on biology learning in terms of perceived autonomy, competence, and relatedness. However, these studies (1) do not consider all three basic needs, (2) nor do they distinguish between all motivational regulation styles separately. A more fine-grained investigation is required. Thus, a cross-sectional study with 738 students was conducted. Structural analyses showed differing effects of the students' perceived autonomy, competence, and relatedness on their motivational regulation. The perception of autonomy and competence were important predictors of intrinsic and identified regulation. However, while intrinsic regulation required mainly the perception of autonomy, identified regulation depended mainly on competence. Perceived relatedness had no predictive power. When it came to controlled regulations, only the perceived relatedness was a negative predictor of students' introjected regulation. Regarding external regulation, neither perceived autonomy and competence nor relatedness were negative predictors.

Abstract

There is considerable evidence that the immune system plays a role in hypertension, however this role is not fully characterized. Our previous studies demonstrated that mesenteric perivascular adipose tissue (mPVAT) harbors a large T cell population, which is a cell type identified as contributing to hypertension. In the present study, we tested the hypothesis that soluble mediators in mPVAT influence T cell function just prior to the development of hypertension. Toward this end, we utilized a unique model of hypertension in which Dahl S rats on a high fat (HF) diet develop hypertension. We found that conditioned media (CM) from mPVAT from healthy Dahl S rats on control diet buffers T cell activation, however, mPVAT-CM from Dahl S rats on a HF diet markedly increased inflammatory cytokine induction (IFNγ, GM-CSF and IL-17a) by activated T cells. These cytokines are known to promote activation of macrophages and neutrophils, among other effects. Conversely, the anti-inflammatory cytokine, IL-10, was not different between the groups, suggesting the effect is selective for inflammatory cytokines. Furthermore, we conducted bulk RNA-seq on activated T cells cultured in mPVAT-CM from Dahl S rats on either control (CTL) or HF diet for 10 weeks. In accordance with the cytokine analysis, mPVAT-CM from HF diet-fed rats significantly upregulated many genes associated with IFNγ/IL-17 induction, whereas Th2/Treg-associated genes were downregulated. Taken together, these data strongly suggest soluble mediators from mPVAT influence T cell inflammatory status and may promote Th1/Th17 differentiation preceding the development of hypertension triggered by HF diet.
