Low-frequency peaks in the Raman spectra of amorphous poly(ether ether ketone) (PEEK) were investigated. An amorphous sample with zero crystallinity, as confirmed by wide-angle X-ray diffraction, was used in this study. In a previous study, two peaks were observed in the low-frequency Raman spectra of the crystallized samples. Among these, the peaks at 135 cm−1 disappeared for the amorphous sample. Meanwhile, for the first time, the peak at 50 cm−1 was observed in the crystallized sample. Similar to the peak at 135 cm−1, the peak at 50 cm−1 disappeared in the amorphous state, and its intensity increased with increasing crystallinity. The origins of the two peaks were associated with the Ph-CO-Ph-type intermolecular vibrational modes in the simulation. This suggests that the Ph-CO-Ph vibrational mode observed in the low-frequency region of PEEK was strongly influenced by the intermolecular order.

Low-frequency Raman spectroscopy (LFRS) is a valuable tool to detect the solid state of amorphous and crystalline drugs in solid dosage forms and the transformation of drugs between different polymorphic forms. It has also been applied to track the solubilisation of solid drugs as suspensions in milk and infant formula during in vitro digestion. This study reports the use of LFRS as an approach to probe drug precipitation from a lipid-based drug delivery system (medium-chain self-nanoemulsifying drug delivery system, MC-SNEDDS) during in vitro digestion. Upon lipolysis of the digestible components in MC-SNEDDS containing fenofibrate as a model drug, sharp phonon peaks appeared at the low-frequency Raman spectral region (−1), indicating the precipitation of fenofibrate in a crystalline form from the formulation. Two multivariate data analysis approaches (principal component analysis and partial least squares discriminant analysis) and one univariate analysis approach (band ratios) were explored to track these spectral changes over time. The low-frequency Raman data produces results in good agreement with in situ small angle X-ray scattering (SAXS) measurements with all data analysis approaches used, whereas the mid-frequency Raman requires the use of PLS-DA to gain similar results. This suggests that LFRS can be used as a complementary, and potentially more accessible, technique to SAXS to determine the kinetics of drug precipitation from lipid-based formulations.

The solid-state landscape of carbamazepine during its dehydration was explored using Raman spectroscopy in the low- (−300 to −15, 15 to 300) and mid- (300 to 1800 cm−1) frequency spectral regions. Carbamazepine dihydrate and forms I, III, and IV were also characterized using density functional theory with periodic boundary conditions and showed good agreement with experimental Raman spectra with mean average deviations less than 10 cm−1. The dehydration of carbamazepine dihydrate was examined under different temperatures (40, 45, 50, 55, and 60 °C). Principal component analysis and multivariate curve resolution were used to explore the transformation pathways of different solid-state forms during the dehydration of carbamazepine dihydrate. The low-frequency Raman domain was able to detect the rapid growth and subsequent decline of carbamazepine form IV, which was not as effectively observed by mid-frequency Raman spectroscopy. These results showcased the potential benefits of low-frequency Raman spectroscopy for pharmaceutical process monitoring and control.

Raman and infrared spectroscopy, used as individual and low-level fused datasets, were evaluated to identify and quantify the presence of adulterants (palm oil, PO; ω-3 concentrates in ethyl ester, O3C and fish oil, FO) in krill oil. These datasets were qualitatively analysed with principal component analysis (PCA) and classified as adulterated or unadulterated using support vector machines (SVM). Using partial least squares regression (PLSR), it was possible to identify and quantify the adulterant present in the KO mixture. Raman spectroscopy performed better (r2 = 0.98; RMSEP = 2.3%) than IR spectroscopy (r2 = 0.91; RMSEP = 4.2%) for quantification of O3C in KO. A data fusion approach further improved the analysis with model performance for quantification of PO (r2 = 0.98; RMSEP = 2.7%) and FO (r2 = 0.76; RMSEP = 9.1%). This study demonstrates the potential use of Raman and IR spectroscopy to quantify adulterants present in KO.

The charge transfer (CT) properties of photosensitizers largely determine the photovoltaic performances of dye sensitized solar cells (DSSCs). Thus, understanding the CT properties of photosensitizers is key to further improving the performances of DSSCs. We herein investigated the underlying relationship between the molecular structures and CT properties of the photosensitizers using resonance Raman (RR) spectroscopy and density functional theory (DFT) calculations. RR spectroscopy combined with DFT calculations showed that the presence of a triple bond (T-D1, T-D2, and T-D3) enhanced the degree of CT from the donor to the acceptor. In addition, the presence of electron donating groups (EDGs) on the donor (T-D2 and T-D3) further increased the CT properties of the donor. Moreover, DFT analysis based on the harmonic oscillator model of aromaticity revealed that the presence of a triple bond and an EDG increased the quinoidal character of the photosensitizer in the excited state. Finally, it was found that the degree of CT properties exhibited by each photosensitizer was in good agreement with the order of the DSSC performances.

This study uses Raman and IR spectroscopic methods for the detection of adulterants in marine oils. These techniques are used individually and as low-level fused spectroscopic data sets. We used cod liver oil (CLO) and salmon oil (SO) as the valuable marine oils mixed with common adulterants, such as palm oil (PO), omega-3 concentrates in ethyl ester form (O3C), and generic fish oil (FO). We showed that support vector machines (SVM) can classify the adulterant present in both CLO and SO samples. Furthermore, partial least squares regression (PLSR) may be used to quantify the adulterants present. For example, PO and O3C adulterated samples could be detected with a RMSEP value less than 4%. However, the FO adulterant was more difficult to quantify because of its compositional similarity to CLO and SO. In general, data fusion improved the RMSEP for PO and O3C detection. This shows that Raman and IR spectroscopy can be used in concert to provide a useful analytical test for common adulterants in CLO and SO.

Different formulation techniques have been investigated to prepare highly aerosolizable dry powders to deliver a high dose of antibiotics to the lung for treating local infections. In this study, we investigated the influence of the co-amorphization of a model drug, kanamycin, with selected amino acids (valine, methionine, phenylalanine, and tryptophan) by co-spray drying on its aerosolization. The co-amorphicity was confirmed by thermal technique. The physical stability was monitored using low-frequency Raman spectroscopy coupled with principal component analysis. Except for the kanamycin-valine formulation, all the formulations offered improved fine particle fraction (FPF) with the highest FPF of 84% achieved for the kanamycin-methionine formulation. Variation in animal coloration is often viewed as the result of chemically distinct pigments conferring different hues. The role of molecular environment on hue tends to be overlooked as analyses are mostly performed on free pigments extracted from the integument. Here we analysed psittacofulvin pigments within parrot feathers to explore whether the in situ organization of pigments may have an effect on hue. Resonance Raman spectra from a red region of a yellow-naped amazon Amazona auropalliata tail feather show frequency dispersion, a phenomenon that is related to the presence of a range of molecular conformations (and multiple chromophores) in the pigment, whereas spectra from a yellow region on the same feather do not show the same evidence for multiple chromophores. Our findings are consistent with non-isomeric psittacofulvin pigments behaving as a single chromophore in yellow feather barbs, which implies that psittacofulvins are dispersed into a structurally disordered mixture in yellow feathers compared with red feathers. Frequency dispersion in red barbs may instead indicate that pigments are structurally organized through molecule–molecule interactions. Major differences in the hues of parrot feathers are thus associated with differences in the organization of pigments within feathers.

Valorization of agricultural waste has become increasingly important. Wastes generated by wineries are high in phenolic compounds with antioxidant and antibacterial properties, which contribute to phytotoxicity, making their immediate use for agricultural means limited. Utilizing a water-based extraction method, the phenolic compounds from winery waste were extracted and purified. The resulting extract was characterized for phenolic composition using high-pressure liquid chromatography-ultraviolet/visible and electrochemical detectors (HPLC-UV/Vis, ECD) for monomers, and spectral assessment of the tannins present using attenuated total reflectance- Fourier transform infrared (ATR-FTIR), FT-Raman, and solid-state nuclear magnetic resonance (SSNMR) spectroscopies. The extract’s antioxidant activity was assessed by the scavenging of the 2,2-diphenyl-1−picrylhydrazyl (DPPH) radical and Folin-Ciocalteu total phenolic assay, and was found to be as effective as a commercially obtained grape extract. The extract’s antimicrobial efficacy was tested for minimum bactericidal concentration using Candida albicans, Escherichia coli 25922, and Staphylococcus aureus 6538, which resulted in greater efficacy against gram-positive bacteria as shown over gram-negative bacteria, which can be linked to both monomeric and tannin polyphenols, which have multiple modes of bactericidal action.

Particle size reduction is a simple means to enhance the dissolution rate of poorly water soluble BCS-class II and IV drugs. However, the major drawback of this process is the possible introduction of process induced disorder. Drugs with different molecular arrangements may exhibit altered properties such as solubility and dissolution rate and, therefore, process induced solid state modifications need to be monitored. The aim of this study was two-fold: firstly, to investigate the dissolution rates of milled and unmilled simvastatin; and secondly, to screen for the main milling factors, as well as factor interactions in a dry ball milling process using simvastatin as model drug, and to optimize the milling procedure with regard to the opposing responses particle size and process induced disorder by application of a central composite face centered design. Particle size was assessed by scanning electron microscopy (SEM) and image analysis. Process induced disorder was determined by partial least squares (PLS) regression modeling of respective X-ray powder diffractograms (XRPD) and Raman spectra. Valid and significant quadratic models were built. The investigated milling factors were milling frequency, milling time and ball quantity at a set drug load, out of which milling frequency was found to be the most important factor for particle size as well as process induced disorder. Milling frequency and milling time exhibited an interaction effect on the responses. The optimum milling settings using the maximum number of milling balls (60 balls with 4 mm diameter) was determined to be at a milling frequency of 21 Hz and a milling time of 36 min with a resulting primary particle size of 1.4 μm and a process induced disorder of 6.1% (assessed by Raman spectroscopy) and 8.4% (assessed by XRPD), at a set optimization limit of < 2 μm for particle size and < 10% for process induced disorder. This optimum was tested experimentally and the process induced disorder was determined to be 6.9% (± 2.2) by Raman spectroscopy and 7.8% (± 2.3) by XRPD. Subsequent intrinsic dissolution testing revealed that the process induced disorder was negligible with regard to the dissolution rate. The predicted primary particle size of 1.4 μm could be confirmed experimentally, but due to agglomeration of the primary particles a dissolution rate advantage was not shown, highlighting the importance of dissolution testing at an early stage of drug development.

To realise useful control over molecular motion in the future an extensive toolbox of both actionable molecules and stimuli-responsive units must be developed. Previously, our laboratory has reported 1,1′-disubstituted ferrocene (Fc) rotor units which assume a contracted/π-stacked conformation until complexation of cationic metal ions causes rotation about the Ferrocene (Fc) molecular ‘ball-bearing’. Herein, we explore the potential of using the photochemical ejection of [Ru(2,2′-bipyridyl)2]2+ units as a stimulus for the rotational contraction of new ferrocene rotor units. Fc rotors with both ‘regular’ and ‘inverse’ 2-pyridyl-1,2,3-triazole binding pockets and their corresponding [Ru(2,2′-bipyridyl)2]2+ complexes were synthesised. The rotors and complexes were characterised using nuclear magnetic resonance (NMR) and ultraviolet (UV)-visible spectroscopies, Electro-Spray Ionisation Mass Spectrometry (ESI–MS), and electrochemistry. The 1,1′-disubstituted Fc ligands were shown to π-stack both in solution and solid state. Density Functional Theory (DFT) calculations (CAM-B3LYP/6-31G(d)) support the notion that complexation to [Ru(2,2′-bipyridyl)2]2+ caused a rotation from the syn- to the anti-conformation. Upon photo-irradiation with UV light (254 nm), photo-ejection of the [Ru(2,2′-bipyridyl)2(CH3CN)2]2+ units in acetonitrile was observed. The re-complexation of the [Ru(2,2′-bipyridyl)2]2+ units could be achieved using acetone as the reaction solvent. However, the process was exceedingly slowly. Additionally, the Fc ligands slowly decomposed when exposed to UV irradiation meaning that only one extension and contraction cycle could be completed.

A series of eight carbazole-cyanoacrylate based donor-acceptor dyes were studied. Within the series the influence of modifying the thiophene bridge, linking donor and acceptor and a change in the nature of the acceptor, from acid to ester, was explored. In this joint experimental and computational study we have used electronic absorbance and emission spectroscopies, Raman spectroscopy and computational modeling (density functional theory). From these studies it was found that extending the bridge length allowed the lowest energy transition to be systematically red shifted by 0.12 eV, allowing for limited tuning of the absorption of dyes using this structural motif. Using the aforementioned techniques we demonstrate that this transition is charge transfer in nature. Furthermore, the extent of charge transfer between donor and acceptor decreases with increasing bridge length and the bridge plays a smaller role in electronically mixing with the acceptor as it is extended.

Readily synthesised and functionalised di-1,2,3-triazole “click” ligands are shown to self-assemble into coordinatively saturated, quadruply stranded helical [Pd2L4](BF4)4cages with Pd(ii) ions. The cages have been fully characterised by elemental analysis, HR-ESMS, IR, 1H, 13C and DOSY NMR, DFT calculations, and in one case by X-ray crystallography. By exploiting the CuAAC “click” reaction we were able to rapidly generate a small family of di-1,2,3-triazole ligands with different core spacer units and peripheral substituents and examine how these structural modifications affected the formation of the [Pd2L4](BF4)4cages. The use of both flexible (1,3-propyl) and rigid (1,3-phenyl) core spacer units led to the formation of discrete [Pd2L4](BF4)4cage complexes. However, when the spacer unit of the di-1,2,3-triazole ligand was a 1,4-substituted-phenyl group steric interactions led to the formation of an oligomeric/polymeric species. By keeping the 1,3-phenyl core spacer constant the effect of altering the “click” ligands’ peripheral substituents was also examined. It was shown that ligands with alkyl, phenyl, electron-rich and electron-poor benzyl substituents all quantitatively formed [Pd2L4](BF4)4cage complexes. The results suggest that a wide range of functionalised palladium(ii) “click” cages could be rapidly generated. A spectroscopic and computational study of a series of 2,5-bis(2-thien-2-ylethenyl) thiophene-based oligomers with a para-R-arylethenyl substituent is reported. The primary aim of this investigation is to increase understanding of how charge moves through these molecules by comparing the neutral and oxidized structures for each molecule. To this end, the B3LYP/6-31G(d) computational method was used to calculate the geometry and vibrational spectra for all molecules considered and their oxidation products. For vibrational data, mean absolute deviations for frequencies between experimental and theoretical results ranging from 2 to 18 cm-1were obtained. Experimental Raman spectroscopy, in conjunction with calculated bond length analyses, was used to gain an insight into the position and delocalization of the charged defect on the oxidized oligomers. The relative frequencies of different ethylene stretching modes served as a particularly useful probe in this regard. It was found that the ethenyl spacers do not impede -electron delocalization and, therefore, give rise to a longer conjugation length relative to the corresponding terthiophenes. Furthermore, the para-R-arylethenyl substituent was found to orientate the charged defect toward a specific region of the 2,5-bis(2-thien-2-ylethenyl)thiophene conjugation path. The structural and electronic properties of neutral and oxidized terthiophenes substituted with polyaromatic systems have been investigated using a combination of both Raman and electronic absorption spectroscopy in conjunction with density functional theory calculations. Naphthylethenyl terthiophene exhibits structural and electronic properties, in both the neutral and oxidized species, that are dominated by the terthiophene backbone, in a manner similar to that previously reported for phenylethenylterthiophene. Anthracenylethenyl terthiophene, on the other hand, displays properties that are dominated by the anthracene group. Unlike both phenylethenyl and naphthylethenyl terthiophene, which have electronic absorption spectra dominated by transitions between molecular orbitals that are delocalized throughout the molecules, the absorption spectrum of anthracenylethenyl terthiophene consists of a simple addition of the absorption bands of the separate terthiophene and anthracenylethene chromophores. This is the result of a spatial partitioning of its molecular orbitals that effectively electronically decouples the anthracene and terthiophene moieties. Upon oxidation, the naphthylethenylterthiophene -dimerizes to form sexithiophene charged species and spectral signatures of the sexithiophene backbone are evident in both the electronic absorption and resonance Raman spectra. In contrast, these signatures are absent in the corresponding spectra of the oxidized anthracenylethenylterthiophene, suggesting that the anthracene group is the primary site of the structural changes induced by oxidation. A number of case studies that illustrate how quantum chemistry may be used in studying pharmaceutical systems are reviewed. A brief introduction to quantum methods is provided and the use of these methods in understanding the structure and properties of indometacin and carbamazepine is discussed. The use of calculated structures and molecular electrostatic potentials in developing quantitative structure–activity relationships is discussed along with the use of computation chemistry to predict spectroscopic properties. Terahertz pulsed spectroscopy (TPS) and terahertz pulsed imaging (TPI) are two novel techniques for the physical characterization of pharmaceutical drug materials and final solid dosage forms, utilizing spectral information in the far infrared region of the electromagnetic spectrum. This review focuses on the development and performance of pharmaceutical applications of terahertz technology compared with other tools for physical characterization. TPS can be used to characterize crystalline properties of drugs and excipients. Different polymorphic forms of a drug can be readily distinguished and quantified. Recent developments towards a better understanding of the fundamental theory behind spectroscopy in the far infrared have been discussed. Applications for TPI include the measurement of coating thickness and uniformity in coated pharmaceutical tablets, structural imaging and 3D chemical imaging of solid dosage forms. The influence of various excipients on the conversion of carbamazepine polymorphs to the dihydrate in aqueous suspension has been investigated. Ten excipients having functional groups which were potentially able to form hydrogen bonds with carbamazepine (group 1: methylcellulose, hypromellose (hydroxypropyl methylcellulose), hydroxypropylcellulose (HPC), 2-hydroxyethylcellulose (HEC), carmellose sodium (sodium carboxymethylcellulose), cellobiose; group 2: povidone (polyvinylpyrrolidone), povidone-vinyl acetate copolymer (povidone/VA) and N-methyl-2-pyrrolidone; group 3: macrogol (polyethylene glycol) and polyethylene oxide–polypropylene oxide copolymer (PEO/PPO)) were selected. Carbamazepine polymorphic forms III and I were dispersed separately into each aqueous excipient solution (0.1%, w/v) for 30 min at room temperature. The inhibition effect of each excipient was quantified using Raman spectroscopy combined with multivariate analyses. The solubility parameter of each excipient was calculated and used for categorizing excipients. Excipients in groups 1 and 2, which had both low solubility parameters (< 27.0 MPa1/2) and strong hydrogen bonding groups, inhibited the conversion completely. With increasing solubility parameter, the inhibition effect decreased for group 1 excipients, especially for carbamazepine form I, which had a higher specific surface area. Also, the excipients of group 3, lacking strong hydrogen bonding groups, showed poor inhibition although they had low solubility parameters (< 21.0 MPa1/2). This study indicated the importance of both hydrogen bonding interaction and a suitable hydrophobicity (expressed by the solubility parameter) in the inhibition of the conversion of carbamazepine to the dihydrate. Raman spectroscopy is experiencing a surge in interest in solid-state pharmaceutical applications. It is rapid, non-destructive, no sample preparation is required and measurements can be made in aqueous environments. It can be used for not only qualitative, but also quantitative, analysis. In this paper, the use of Raman spectroscopy for quantitative analysis of pharmaceutical solids is reviewed. The technique has been used for chemical and physical form analysis. Physical form analysis has involved quantification of polymorphism, hydrates, the amorphous form and, recently, protein conformation. Initially, simple powder systems were quantified, although this has since extended to complex pharmaceutical formulations, including tablets, capsules, microspheres and suspensions. Formulations have also been analysed through packaging. The characteristics of the technique make it ideal for process monitoring and it has been used to quantify changes in-situ during processes such as wet granulation and batch crystallisation. The theoretical basis of quantitative Raman spectroscopy, common data analysis approaches, including multivariate analysis, and sources of error in quantitative analysis are also discussed. Amorphous materials are prevalent in the pharmaceutical setting. Whether they are a help or hindrance, their physico-chemical characteristics must be investigated. However, the amorphous form remains a challenge to characterise with many of its properties poorly understood. In this study, γ-crystalline and amorphous indometacin are investigated using vibrational spectroscopy and quantum chemical calculations. The structure of the single indometacin molecule and the dimer in the γ-form were optimised using density functional theory calculations. The optimised structures were similar to the conformations in the crystal form, suggesting that conformation of the molecules in the crystal may be close to the average molecular structure in less-ordered states. Infrared and Raman spectra were calculated from the optimised structures. Many modes in the calculated spectra could be matched with the experimental spectra of the γ-crystalline and amorphous forms, and a description of the matched modes has been provided. By analysis of the theoretical vibrational modes it was confirmed that the amorphous form of indometacin produced by quench cooling the melt consists predominantly of dimers, similar in structure to in the γ-crystalline form. In addition, differences in intermolecular bonding between the two forms were identified. The lowest energy transition of Ru(CN)4(ppb)2-(ppb dipyrido2,3-a:3‘,2‘-cphenazine), a metal-to-ligand charge transfer, has been probed using resonance Raman spectroscopy with excitation wavelengths (488, 514, 530, and 568 nm) spanning the lowest energy absorption band centered at 522 nm. Wave packet modeling was used to simultaneously model this lowest energy absorption band and the cross sections of the resonance Raman bands at the series of excitation wavelengths across this absorption band. A fit to within ±20% was obtained for the Raman cross sections, close to the experimental uncertainty which is typically 10−20%. values of 0.1−0.4 were obtained for modes which were either localized on the ppb ligand (345−1599 cm-1) or the CN modes (2063 and 2097 cm-1). The steady-state and transient emission properties of unsubstituted terthiophene and a series of 3‘-[E-2-(4-R-phenyl)ethenyl]-2,2‘:5‘,2‘ ‘-terthiophenes (where R = H, MeO, NH2, CN, NMe2, NO2) have been examined. The R = NO2 compound is nonemissive at room temperature in all solvents but cyclohexane. All of the other compounds show measurable steady-state emission in a variety of solvents. The behavior of these spectra may be split into two groups. The first group, those substituted compounds with R = CN, NH2 and NMe2, show solvatochromic behavior, where their Lippert−Mataga plots suggest changes in dipole upon photoexcitation ranging from 12.5 to 16.0 D. For the second group, where R = H and MeO (and unsubstituted terthiophene as well), the Lippert−Mataga plots indicate dipole moment changes ranging from 0 to 7.9 D. The difference in behavior between the two groups of emissive compounds can be attributed to a charge-transfer character of the emitting state in the first group. This conclusion is supported by density functional theory calculations, which show that the frontier MOs in the group one compounds are spatially separated whereas those of group two have frontier MOs that are delocalized over both the styryl and terthiophene moieties. Picosecond time-resolved fluorescence spectroscopy reveals that unsubstituted terthiophene has the shortest emission lifetime of 140 ps in acetonitrile. For the styryl substituted terthiophenes, the lifetimes are much longer and range from 320 to 670 ps for R = CN and NMe2 respectively, a result that can be explained in terms of a smaller rate of intersystem crossing in these compounds. IR spectroscopy in concert with DFT calculations and resonance Raman spectroelectrochemistry has been used to identify the molecular orbital nature of the singly occupied molecular orbital (SOMO) in reduced Re(CO)3Cl(L) and Re(CO)3(4-Mepy)(L)complexes, where L 1,10-phenanthroline and its 4,7-diphenyl- and 3,4,7,8-tetramethyl-substituted analogues. The SOMO of each reduced species considered was found to be of b1symmetry, rather than the close lying orbital of a2symmetry (within a C2vsymmetry description of the phenanthroline moiety). This was deduced in a number of ways. First, the average carbonyl band force constants (kavkavreduced complex − kavparent complex) range from −57 to −41 N m-1for the series of compounds studied. The value of kavrelates to the extent of orbital overlap between the ligand MO and the metal d MO. These values are consistent with population of a b1MO because the wave function amplitude at the chelating nitrogens for this MO is significantly greater than that for a2MO. Second, calculations on singly reduced Re(CO)3(4-Mepy)(phen)and Re(CO)3(4-Mepy)(tem)predict population of a b2SOMO. The spectra predicted for these species are in close agreement with the vibrational spectroscopic data; for the IR data the shifts in the CO bands are predicted to 6 cm-1and the mean absolute deviation between calculated and measured Raman bands was found to be 10 cm-1. Stable isotope analyses for paleodiet investigations require good preservation of bone protein, the collagen, to obtain reliable stable isotope values. Burial environments cause diagenetic alterations to collagen, especially in the leaching of the organic bone content. The survival of bone protein may be assessed by the weight % collagen, % carbon and % nitrogen yields, but these values are achieved only after destructive chemical processing. A non-destructive method of determining whether bone is suitably preserved would be desirable, as it would be less costly than chemical processing, and would also preserve skeletal collections. Raman analysis is one such potential non-destructive screening method. In previous applications, Raman spectroscopy has been used to test both the alteration of the mineral portion of bone, as well as to indicate the relative amount of organic material within the bone structure. However, there has been no research to test the relationship between the Raman spectroscopic results and the survival of bone protein. We use a set of 41 bone samples from the prehistoric archaeological site of Ban Non Wat, Northeast Thailand, to assess if Raman spectroscopy analysis of the organic-phosphate ratio has a significant correlation with the weight % collagen, and carbon and nitrogen yields obtained by isotopic analysis. The correlation coefficients are highly statistically significant in all cases (r = 0.716 for collagen, r = 0.630 for carbon and r = 0.706 for nitrogen, p≤0.001 for all) with approximately or close to half of the variation in each explained by variation in the organic-phosphate ratio (51.2% for collagen, 39.6% for carbon, and 49.8% for nitrogen). Although the Raman screening method cannot directly quantify the extent of collagen survival, it could be of use in the selection of bone most likely to have viable protein required for reliable results from stable isotope analysis.
