Islet encapsulation devices serve to deliver pancreatic beta cells to type 1 diabetic patients without the need for chronic immunosuppression. However, clinical translation is hampered by mass transport limitations causing graft hypoxia. This is exacerbated in devices relying only on passive diffusion for oxygenation. Here, we describe the application of a cylindrical in vitro perfusion system to study oxygen effects on islet-like clusters immobilized in alginate hydrogel. Mouse insulinoma 6 islet-like clusters were generated using microwell plates and characterized with respect to size distribution, viability, and oxygen consumption rate to determine an appropriate seeding density for perfusion studies. Immobilized clusters were perfused through a central channel at different oxygen tensions. Analysis of histological staining indicated the distribution of viable clusters was severely limited to near the perfusion channel at low oxygen tensions, while the distribution was broadest at normoxia. The results agreed with a 3D computational model designed to simulate the oxygen distribution within the perfusion device. Further simulations were generated to predict device performance with human islets under in vitro and in vivo conditions. The combination of experimental and computational findings suggest that a multichannel perfusion strategy could support in vivo viability and function of a therapeutic islet dose.

Abstract: Objective: To investigate the effect of omega-3 PUFAs, eicosapentanoic acid (EPA) and docosohexanoic acid (DHA) on inflammatory cytokine production in the amnion. Study design: Amnion explants were obtained at elective caesarean sections and cultured in vitro with EPA and DHA. IL-8 and IL-6 secretion was determined by ELISA, the role of PPARγ was investigated using specific agonists and antagonists and activity of MMP assessed by gelatin zymography. Results: A combination of EPA and DHA significantly reduced the concentration of IL-8 and IL-6 released into the supernatant compared to untreated controls (p<0.001). Stimulation of PPARγ with troglitazone reduced IL-8 production, and the PPARγ antagonist GW9662 partially reversed this effect. The activity of MMP-9 was also significantly reduced by treatment with EPA and DHA in combination compared to untreated control (p<0.05). Conclusion: The omega-3 PUFAs EPA and DHA decrease the inflammatory response of the amnion, and this may be partially mediated through PPARγ. [Copyright &y& Elsevier]

Abstract: Much research has gone into developing a sustainable management tool for haemonchosis, namely the FAMACHA© system, which has been validated by numerous studies worldwide. However, the financial implications on production of implementing this system have not been fully investigated in sheep previously. A trial was conducted on a farm comprising a flock of approximately 300 Mutton Merinos on which the FAMACHA© system was in use. Seventy five maiden and multiparous ewes were randomly allocated to the following three trial groups: (i) FAMACHA© in which ewes judged to be in categories 4 or 5 were dosed with levamisole HCl 2.5% (Nemasol NF, Intervet); (ii) Conventionally dosed in which all ewes were dosed every six weeks with levamisole HCl 2.5% (Nemasol NF, Intervet); (iii) Suppressively dosed in which all ewes were injected every six weeks with moxydectin 1%m/v (Cydectin, Bayer AH). There were no significant differences in body weight gains, FAMACHA© and body condition scores amongst the three treatment regimens for ewes that did not conceive. For those that became pregnant during the trial there was a significant difference when analysed by calendar date but not when analysed by reproductive stage. There were also no significant differences between trial groups in lamb birth weight and, after slow growers had been sold, the average daily gain of the lambs born to ewes in the trial. The cost of anthelmintics was considerably lower for the FAMACHA© group. In conclusion there were no significant differences in selected production parameters when using the FAMACHA© system as opposed to other methods of anthelmintic use in a Mutton Merino flock in a semi-intensive farming system and that the gestation status should be taken into account when measuring these parameters in future studies. The atmospheric pressure glow discharge torch (APGD-t) is a miniature pulsed RF (13.56 MHz) plasma source specifically designed to efficiently produce and transport reactive species to and interact with biological samples. When applied to adherent cell cultures, the APGD-t is capable of inducing temporary cell permeabilization. In this work, we evaluate the maximum radius of macromolecules able to enter into HeLa cells following a plasma treatment using a size exclusion approach with dextran molecules. We observe that this maximum radius is below 6.5 nm. We also show that no degradation occurs when a plasmid DNA suspended in culture media is plasma-treated at the operating conditions leading to cell permeabilization. The plasma-assisted DNA transfection of the HeLa cells with the hrGFP-II-1 plasmid using the APGD-t is demonstrated. Local transfection efficiencies obtained are as high as 35%. Interleukin-8 (IL-8) attracts neutrophils into tissues and causes them to degranulate. Both menstruation and parturition involve neutrophil migration into uterine tissues and therefore IL-8 is a likely mediator of the tissue re-arrangements that accompany these events. We have examined the ability of endometrium explants and chorion cells in culture to synthesize and release IL-8 and the ability of progesterone, a synthetic progestin [medroxyprogesterone acetate (MPA)] and dexamethasone to inhibit this production. In endometrium, the stage of the menstrual cycle did not affect IL-8 production but a 10(-6) M concentration of progesterone or dexamethasone significantly reduced the concentration of IL-8 in medium after 24 h. After a further 24 h with lipopolysaccharide (LPS) stimulation, only MPA and dexamethasone inhibited production significantly. In chorion cells, IL-8 production was significantly decreased by both MPA and dexamethasone in the LPS stimulated cells but the reduction in the first 24 h was not significant. The IL-8 produced in uterine tissues might act synergistically with prostaglandin E (PGE), a likely site for this interaction being blood vessels where PGE production is also repressed by progesterone. Such a cooperative action would maintain low leukocyte entry into uterine tissues in the presence of progesterone and falling steroid levels would induce leukocyte immigration and activation with consequent tissue destruction. Such steroid-dependent interactions are important in our understanding of the mechanisms of menstruation and parturition and could allow new approaches to the treatment of uterine pathology. Dietary potassium intake was estimated in 46 men and 88 women randomly selected from people aged 65 and over living in their own homes, and 24-hour urinary potassium, creatinine, and urea excretions were determined. The mean dietary potassium intake was 71 mEq/day in men and 54 mEq/day in women. There was a decline with age in both sexes. The sex and age differences were eliminated by expression of dietary potassium intake per unit lean body mass. Urinary potassium excretion averaged 51 mEq/day in men and 44 mEq/day in women, and again the sex difference and decline with age were abolished by expression of urinary potassium per unit lean body mass. There were highly significant relationships between dietary and urinary potassium. Consideration of the regression lines of these correlations suggested that if faecal potassium losses were 5 mEq/day, a negative potassium balance would be inevitable if dietary potassium fell below 27 mEq/day in men and 35 mEq/day in women. Sixteen common biochemical estimations have been performed on nearly 500 people randomly selected from those over 65 living at home. The mean values and ranges found for serum sodium, potassium, chloride, bicarbonate, magnesium, bilirubin, inorganic phosphate, total protein, albumin, and globulin, were identical, and those of urea, creatinine, cholesterol, uric acid, calcium, and alkaline phosphatase were higher than those considered normal in younger people. Sex differences were demonstrable for serum urea, creatinine, bilirubin, phosphate, cholesterol, calcium, and alkaline phosphatase; in the last four instances these seemed likely to be of clinical importance. In some pathological conditions like aortic stiffening and calcific aortic stenosis (CAS), the microstructure of the aortic root and the aortic valve leaflets are altered in response to stress resulting in changes in tissue thickness, stiffness, or both. This aortic stiffening and CAS are thought to affect coronary blood flow. The goal of the present paper was to include the flow in the coronary ostia in the previous fluid structure interaction model we have developed and to analyze the effect of diseased tissues (aortic root stiffening and CAS) on coronary perfusion. Results revealed a significant impact on the coronary perfusion due to a moderate increase in the aortic wall stiffness and CAS (increase of the aortic valve leaflets thickness). A marked drop of coronary peak velocity occurred when the values of leaflet thickness and aortic wall stiffness were above a certain threshold, corresponding to a threefold of their normal value. Consequently, mild and prophylactic treatments such as smoking cessation, exercise, or diet, which have been proven to increase the aortic compliance, may significantly improve the coronary perfusion. Objectives: We studied collagen plugging of the fetoscopic access site in an in vivo fetal lamb model for fetoscopic surgery and possible role for matrix metalloproteinase (MMP)-2 and -9 and tissue inhibitors (TIMPs).Methods: Eight ewes had fetoscopic balloon occlusion of the trachea as an experimental treatment for congenital diaphragmatic hernia between days 88 and 99 of gestation (term 145 days) with sampling of amniotic, allantoic, and tracheal fluid. Nonoperated cotwins were used as controls. The fetoscopy port was closed using a collagen plug. Ten days (range 9–12) later, fluids were sampled and plug sites collected for histologic analysis. Activity of MMP-2 (72 kDa, gelatinase A) and MMP-9 (92 kDa, gelatinase B) was determined in the fluids by zymography and secretion of TIMPs (27–30 kDa; TIMP-1, glycosylated TIMP-3 and TIMP-4, 24 kDa; unglycosylated TIMP-3, 21 kDa; TIMP-2) by reverse zymography and quantified by densitometric analysis.Results: No pregnancy was complicated by amniorhexis or preterm labor. At cesarean, normal volumes of amniotic and allantoic fluid were present in all cases. Histology of the plug sites revealed good integration of the collagen plug without complete restoration of membrane integrity. MMP-2, MMP-9, and TIMPs were detected in all fluids. In the operated animals, significantly (P < .05) higher activity of MMP-9 was found in amniotic fluid, with lower concentrations of TIMPs in allantoic fluid (P < .01). Tracheal occlusion was associated with a significant (P < .02) increase in both MMP-2 and -9 in tracheal fluid.Conclusion: Collagen plugging of the fetoscopic access port sites in sheep resulted in functionally effective sealing of the fetal membranes. Phthalate plasticizers are incorporated into plastics to make them soft and malleable, but are known to leach out of the final product into their surroundings with potential detrimental effects to human and ecological health. The replacement of widely-used phthalate plasticizers, such as di-ethylhexyl phthalate (DEHP), that are of known toxicity, by the commercially-available alternative Tris(2-ethylhexyl) tri-mellitate (TOTM) is increasing. Additionally, several newly designed "green" plasticizers, including di-heptyl succinate (DHPS) and di-octyl succinate (DOS) have been identified as potential replacements. However, the impact of plasticizer exposure from medical devices on patient recovery is unknown and, moreover, the safety of TOTM, DHPS, and DOS is not well established in the context of patient recovery. To study the direct effect of clinically based chemical exposures, we exposed C57bl/6 N male and female mice to DEHP, TOTM, DOS, and DHPS during recovery from cardiac surgery and assessed survival, cardiac structure and function, immune cell infiltration into the cardiac wound and activation of the NLRP3 inflammasome. Male, but not female, mice treated in vivo with DEHP and TOTM had greater cardiac dilation, reduced cardiac function, increased infiltration of neutrophils, monocytes, and macrophages and increased expression of inflammasome receptors and effectors, thereby suggesting impaired recovery in exposed mice. In contrast, no impact was detected in female mice and male mice exposed to DOS and DHPS. To examine the direct effects in cells involved in wound healing, we treated human THP-1 macrophages with the plasticizers in vitro and found DEHP induced greater NLRP3 expression and activation. These results suggest that replacing current plasticizers with non-phthalate-based plasticizers may improve patient recovery, especially in the male population. Background: There are challenges around the practicality of conventional standard setting methods for student assessment. Furthermore, accuracy of absolute methods of standard setting is difficult to achieve.The aim was to determine which group of judges is most accurate at establishing the minimum level required to pass questions in order to ensure an appropriate standard (cut-scores), and how the Bloom's level of each question affected the correlation of cut-scores to student performance., Methods: The modifications to the classical Angoff method where a group of judges convene and discuss cut-scores was that, in this study, the judges set cut-scores independently and did not receive the answers to the questions that they were assessing. Computer-based multiple choice and multiple response type questions were compiled, and allocated Bloom's levels. Judges answered the questions, determined cut-scores and completed a questionnaire. Simple linear regression was used to determine whether number of years' experience, proportion of time spent in small ruminant practice or specialisation in the field resulted in the most accurate comparison to student performance., Results: Individuals spending the greatest proportion of time in small ruminant practice demonstrated greater accuracy in determining cut-scores. Although theoretical training of veterinary students is uncomplicated even for larger groups, practical training remains a challenge. Much has been said about the value of practical training in curriculum design. Yet, the impact of practical training on theoretical knowledge needs further research. A cohort of 89 students with very limited clinical practical experience completed an assessment at the end of their theoretical training in small ruminants. The scores obtained by the students were compared with those obtained by a group of 35 veterinarians who volunteered to participate in the study. In addition to comparing the scores between students and practitioners, the cognitive level of each of the questions was considered. Overall, veterinarians achieved higher test scores than did the students. The veterinarians outperformed the students in all cognitive levels except for 'applying' type questions where there was no difference. Different levels of experience, namely young veterinarians (n = 11), established veterinarians (n = 13) and veterinarians approaching retirement (n = 11), were evaluated against the revised Bloom's cognitive levels. When modelling congress attendance frequency, years' experience, proportion of time spent with ruminants and revised Bloom's levels, congress attendance was not a significant variable, and thus, only the other three variables remained. This investigation found that practical experience has a positive effect on theoretical knowledge. The type of practical experience and where such practical experience is included in a curriculum need further research. Working for a number of years in a specific discipline will provide the best support for theoretical knowledge.

Background: There is currently no scientific evidence regarding the current climatic or other epidemiological factors that could influence the occurrence of heartwater in South Africa., Objectives: The objective was to determine whether climatic changes or other epidemiological factors influence the occurence of heartwater in South Africa., Method: A survey was conducted to scrutinise these factors using both veterinarians and farmers working in known areas in which heartwater had previously been confirmed to establish the value of each of these factors. Based on the observations, meaningful tendencies were noted, and conclusions drawn., Results: These include changes in the spatial distribution of heartwater in many areas, with serious expansion, in some instances, of up to 150 km. In total, 48% of veterinarians and 42% of farmers reported seeing increase in the number of farms affected by heartwater. Climate change as a causative factor indicated by observations of increased average temperatures, milder frosts, less rain and shorter rainy seasons was identified by the majority of farmers but not by as many veterinarians. Respondents in both groups considered vegetation change an important factor. Increasing number of wildlife, especially antelope, was seen as a major factor by most veterinarians and also by many farmers. Both groups identified the movement of livestock and wildlife as an increasingly important factor that should be of major concern for both industries because it leads to the avoidable spread of many diseases apart from heartwater., Conclusion: Movement controls should be reinstated and reinforced by vigorously enforced legislation. The role of genetically determined resistance or resilience to heartwater infection in ruminants should be investigated. Breeding better adapted animals could provide part of a sustainable approach to the disease.

Although nasal masses are uncommon in sheep and may have several causes, including neoplasia and bacterial, fungal and viral infections, these lesions may lead to economic losses resulting from weight loss and even death. It is therefore important to differentiate between various categories of upper respiratory tract obstructions and lower respiratory tract infections. The correct aetiological diagnosis of obstructive masses is essential for appropriate treatment and management to be given or action to be taken. The presentation, clinical signs, treatment and pathology of a case of suspected mycotic rhinitis in a 6-year-old Mutton Merino ewe, are described.

Objectives: The aim of this study is to determine whether patients undergoing the Ross procedure with bicuspid aortic valves have pulmonary artery biomechanical properties different from those with tricuspid valves., Methods: Thirty-two pulmonary arteries and 20 aortas were obtained from patients undergoing the Ross procedure at the time of surgery, from a cohort of 32 patients. The aortic valve was tricuspid in 5 patients (16%), bicuspid in 18 patients (56%) and unicuspid in 9 patients (28%). Histological analysis and ex vivo equi-biaxial tensile testing completed within 8 hours of surgery were used to evaluate differences in patient groups and between the pulmonary artery and the ascending aorta., Results: There was no difference in thickness among pulmonary arteries when compared according to aortic valve phenotype (P = 0.94). There was no difference in the tensile tissue properties among aortas and pulmonary arteries when compared according to aortic valve phenotype, in either the circumferential or longitudinal axis. When compared according to the main surgical indication, pulmonary artery walls from patients with pure aortic regurgitation were less stiff than their counterparts (aortic regurgitation: 0.055 ± 0.037 MPa, aortic stenosis: 0.103 ± 0.051 MPa, mixed disease: 0.110 ± 0.044 MPa and aortic valve endocarditis: 0.216 ± 0.033 MPa, P = 0.002). There was no difference in the number of elastic lamellae in pulmonary artery specimens from the three different aortic valve phenotypes, as well as in the aortic specimens., Conclusions: No significant differences were observed in the biomechanical properties of pulmonary arteries when compared according to aortic valve phenotype., (© The Author 2016. A questionnaire of 15 questions was completed by four categories of respondents with the aim of establishing the experience and opinions of these groups on the constraints including animal health problems for communal, small-scale sheep and goat farming in the Eastern Cape province of South Africa. The questionnaires were completed independently and categories were representative of the areas investigated. Analysis of responses was done by means, ranges, votes and clusters of responses. Comparisons between the responses of the four categories were made to identify similarities or contrasts. The results revealed that of non-veterinary concerns, stock theft was the major problem for these farms. Nutrition was a further major constraint. A third area of significant concern was the provision or availability of facilities like fences, water troughs, dips and sheds. Lack of marketing and business skills were also seen as important deficiencies to be rectified so as to promote profitable farming. Of the most important veterinary problems identified, the provision, availability, cost and care of drugs and vaccines were seen as major stumbling blocks to effective disease control, as well as lack of access to veterinary services. The most important diseases that constrain small-ruminant livestock farming in the farming systems investigated were sheep scab and other ectoparasites, heart water, enterotoxaemia, internal parasites and bluetongue. A lack of knowledge in key areas of small-stock farming was revealed and should be rectified by an effective training and support programme to improve the contribution of small-ruminant farming to livelihoods in these communities.

The implementation of biodegradable stents has the potential to revolutionize obstructive coronary artery disease treatment. Limitations still currently exist, however, that prevent biodegradable stents from replacing permanent metallic stents in the global market. The ideal combination of stent properties, including sufficient mechanical strength, controlled degradation, and biocompatibility, has yet to be realized. A novel manufacturing process is proposed that utilizes cold gas-dynamic spraying to fabricate a metal structure with significantly reduced grain size. Iron and stainless steel 316L are combined to form a novel amalgamate with enhanced mechanical strength and a controllable degradation rate, due to the resulting microgalvanic reaction. Flat specimens composed of iron and 316L are fabricated in various compositions, and mechanical and degradation tests were conducted. Femto laser techniques are utilized to produce stents composed of 80% Fe and 20% stainless steel 316L. The in vitro degradation behaviour of the stent is investigated using static and dynamic corrosion tests. It is shown that the corrosion rate can be adjusted to desired values, by varying the weight percentage of iron and stainless steel 316L within the amalgamate.

Nature shows many examples of surfaces with extraordinary wettability,which can often be associated with particular air-trapping surface patterns. Here,robust non-wetting surfaces have been created by femtosecond laser ablation of polytetrafluoroethylene (PTFE). The laser-created surface structure resembles a forest of entangled fibers, which support structural superhydrophobicity even when the surface chemistry is changed by gold coating. SEM analysis showed that the degree of entanglement of hairs and the depth of the forest pattern correlates positively with accumulated laser fluence and can thus be influenced by altering various laser process parameters. The resulting fibrous surfaces exhibit a tremendous decrease in wettability compared to smooth PTFE surfaces; droplets impacting the virgin or gold coated PTFE forest do not wet the surface but bounce off. Exploratory bioadhesion experiments showed that the surfaces are truly air-trapping and do not support cell adhesion. Therewith, the created surfaces successfully mimic biological surfaces such as insect wings with robust anti-wetting behavior and potential for antiadhesive applications. In addition, the fabrication can be carried out in one process step, and our results clearly show the insensitivity of the resulting non-wetting behavior to variations in the process parameters,both of which make it a strong candidate for industrial applications.

Congenital malformations of the branchial arches, clefts and grooves have not been previously reported in sheep. These defects may be due to infectious agents (especially viruses), toxins or genetic abnormalities. Defects were reported in two of a set of quadruplet lambs born prematurely to an eight-tooth Mutton Merino ewe. The lambs weighed between 2.0 kg and 2.5 kg; this is below the normal expected birth weight of 3.5 kg for quadruplet lambs, below which viability is compromised. The firstborn lamb was severely affected by bilateral oroauricular fistulae. The second lamb was unilaterally affected on the right, less severely than the first. The third lamb was normal and the fourth was mummified. The occurrence of another case in this small flock almost a decade earlier indicates that there could be genetic involvement.

Abstract Morbidity from pulmonary arterial hypertension (PAH) ensues when the pulmonary pressure reaches suprasystemic levels. A transcatheter alternative to the Potts shunt would allow decompression of the right heart without the surgical risks. To aid development of a transcatheter aortopulmonary shunt (TAPS) device, we described the anatomic relationship between the left pulmonary artery (LPA) and the descending aorta (dAO) in adults with severe pulmonary hypertension. Adults with severe PAH (peak systolic pulmonary arterial pressure [PASP] ≥80 mmHg) who had computed tomography of the chest were enrolled. Measurements were taken on the axial plane at the level of the pulmonary artery bifurcation. Forty patients (male sex, 9 patients; median age ± standard deviation [SD], [Formula: see text] years; median PASP ± SD, [Formula: see text] mmHg) were identified. The mean distance (±SD) between the LPA and dAO was [Formula: see text] mm. The mean luminal dAO and LPA diameters (±SD) were [Formula: see text] mm and [Formula: see text] mm, respectively. The LPA and dAO approximated in 93% of patients, with 38% having aortic calcification at the contact site. The mean "landing zone" width and height (defining an area with distance <4 mm between the outer borders) of the two arteries were [Formula: see text] mm and [Formula: see text] mm, respectively, at a mean distance of [Formula: see text] mm from the main pulmonary artery bifurcation. This study shows that the landing zone is able to accommodate a TAPS device of up to 15 mm in diameter in the majority of patients with severe PAH.

In July 2006, a case of two out of three lambs born to one ewe in a flock of 45 had signs that, in retrospect, were possibly consistent with Schmallenberg virus infection. This occurred in the Onderstepoort area (Gauteng Province) but a definitive diagnosis was not made. Then, in May 2008, a farmer in the Delmas area (Mpumalanga Province) reported that deformed lambs had been born to several ewes in the flock. Six of the approximately 50 mated ewes gave birth to lambs showing varying degrees of arthrogryposis, torticollis, kyphosis, mandibular brachygnathia and hydrocephalus. Of these, only two were born alive but they died within a few hours. Blood was collected from the ewes with deformed lambs, a random sample of ewes that had given birth to normal lambs and a lamb that was normal but had a twin that was deformed. The samples were tested for Wesselsbron and Akabane antibodies using a complement fixation test and a haemagglutination/haemagglutination inhibition test that were available at that time. Bluetongue virus antibodies were also tested for using a commercial Enzyme-linked immunosorbent assay (ELISA) test. All samples showed negative results for all diseases tested. At the time Rift Valley fever virus had not been diagnosed in that region for many years and so it was not included in the testing. It is unlikely that this was the cause as no liver pathology was detected on postmortem examination of the lambs and no adult ewes had died. The farmer reported that another farm just a few kilometres away experienced the same deformities in some of their lambs but this farm was not investigated. During investigation it was thought that the cause was possibly a new strain of Akabane virus, although there was no way to confirm it. However, with the recent discovery of the Schmallenberg virus, it is possible that this virus has been present in South Africa for at least the last four years without being identified.

During a routine flock visit, a farmer observed that one of the eight tooth Bapedi rams had been losing body condition despite being separated from the flock and fed supplementary feed. The ram's body condition score was assessed as 2 out of 5 (one point less than the average of the rest of the rams) and the teeth appeared normal with no excessive wear. The rumen was assessed by auscultation, palpation and ballottement where a foreign body (approximately 20 cm × 5 cm - 10 cm) was clearly palpated and ballotted. A rumenotomy was performed and a large mass of tightly compacted foreign matter and plant material was removed. The mass consisted of synthetic fibre, plant material and calcium phosphate (50.5%). It appeared to have formed as the result of the ingestion of a synthetic fibre which formed the nidus of a concretion. This was probably the result of deficient nutrition, with the rams eating the synthetic fibre in an attempt to increase feed intake. The ram recovered uneventfully after the rumenotomy was performed and supplementary feeding.

During late gestation in the mare, rapid fetal growth is accompanied by considerable placental growth and further invasion of the endometrium by microvilli. This growth requires extensive remodeling of the extracellular matrix (ECM). In early pregnancy, we know that matrix metalloproteinase (MMP)-9 and -2 are involved in the endometrial invasion during endometrial cup formation. The present study investigated whether MMPs are found in fetal fluids later in gestation and during parturition, and if there was a difference in their activities between normal and preterm delivery. Amniotic fluids were collected from pony mares during the latter half of gestation, and amniotic and allantoic fluids from pony and thoroughbred mares at foaling. The fluids were analysed for the activity of MMP-9 and -2, and TIMPs using zymography techniques. There was an increase (P = 0.002) in activity of latent MMP-9 when approaching normal foaling, and a decrease (P < 0.001) during foaling. MMP-2 activity did not change through gestation, or during foaling. When comparing samples from pregnancies resulting in preterm deliveries with samples from foaling mares, the activity of MMP-9 was lower (P < 0.001) and MMP-2 activity was higher (P = 0.004) during foaling than preceding preterm delivery. The activity of MMP-9 was lower (P = 0.002) prior to preterm delivery than before delivery of a live foal at term, whereas no difference (P = 0.07) was demonstrated for latent MMP-2 activity when comparing the same groups. The activity of TIMP-2 was higher (P < 0.001) in the pre-parturient period before normal foaling than preceding preterm delivery. Objective: To investigate whether pregnancies with development of subsequent pre-eclampsia and intra-uterine growth restriction are associated with altered levels of kisspeptin in maternal serum in the second trimester., Study Design: Retrospective case-control study of 16-20 week serum samples matched for duration of storage at -70 degrees C. Levels of kisspeptin were measured in serum from women with pregnancies with subsequent development of pre-eclampsia (n = 57), intra-uterine growth restriction (n = 118), and matched controls (n = 317)., Results: Serum kisspeptin levels were significantly lower in those women who subsequently developed pre-eclampsia than in controls [median (quartile range) 1109 (449) vs 1188 (365) pg/mL, p = 0.029] and in those with intra-uterine growth restriction [1164 (386) vs 1188 (365) pg/mL, p = 0.016]., Conclusions: Kisspeptin levels are lower in maternal serum in the second trimester, in pregnancies associated with placental dysfunction. The differences in kisspeptin are modest, so although not forming a single screening marker in pre-eclampsia and intra-uterine growth restriction, measurement of kisspeptin may be useful in combination with other markers. Understanding the role of kisspeptin in the establishment of the placenta may further our knowledge of the mechanisms underlying placental function.

Background: In-stent restenosis rates have been closely linked to the wall shear stress distribution within a stented arterial segment, which in turn is a function of stent design. Unfortunately, evaluation of hemodynamic performance can only be evaluated with long term clinical trials. In this work we introduce a set of metrics, based on statistical moments, that can be used to evaluate the hemodynamic performance of a stent in a standardized way. They are presented in the context of a 2D flow study, which analyzes the impact of different strut profiles on the wall shear stress distribution for stented coronary arteries., Results: It was shown that the proposed metrics have the ability to evaluate hemodynamic performance quantitatively and compare it to a common standard. In the context of the simulations presented here, they show that stent's strut profile significantly affect the shear stress distribution along the arterial wall. They also demonstrates that more streamlined profiles exhibit better hemodynamic performance than the standard square and circular profiles. The proposed metrics can be used to compare results from different research groups, and provide an improved method of quantifying hemodynamic performance in comparison to traditional techniques., Conclusion: The strut shape found in the latest generations of stents are commonly dictated by manufacturing limitations. This research shows, however, that strut design can play a fundamental role in the improvement of the hemodynamic performance of stents. Present results show that up to 96% of the area between struts is exposed to wall shear stress levels above the critical value for the onset of restenosis when a tear-drop strut profile is used, while the analogous value for a square profile is 19.4%. The conclusions drawn from the non-dimensional metrics introduced in this work show good agreement with an ordinary analysis of the wall shear stress distribution based on the overall area exposed to critically low wall shear stress levels. The proposed metrics are able to predict, as expected, that more streamlined profiles perform better hemodynamically. These metrics integrate the entire morphology of the shear stress distribution and as a result are more robust than the traditional approach, which only compares the relative value of the local wall shear stress with a critical value of 0.5 Pa. In the future, these metrics could be employed to compare, in a standardized way, the hemodynamic performance of different stent designs.

Background: Several contrast media (CM) are used for diagnostic angiography and coronary percutaneous interventions. Catheter miniaturization allows performance of most diagnostic studies using 4-5 F catheters and interventions using 5-6 F catheters. As a result of catheter lumen downsizing and viscosity of CM, the operators are sometimes required to forcefully inject to produce adequate images., Methods and Results: The aim of the study is to perform a comparative rheology analysis between three different commonly used CM: iso-osmolar, nonionic iodixanol, Visipaque, (GE Healthcare); low-osmolar, nonionic ioversol, Optiray; and low-osmolar, ionic ioxaglate, Hexabrix, (Tyco Healthcare, US). The viscosity was experimentally assessed for temperature varying from 14 to 40 degrees C. To reproduce clinical use, an experimental set-up was used and the pressure developed to inject CM was evaluated at different temperatures and compared between the three CM. All three agents demonstrated a nonlinear inverse relationship between temperature and viscosity. At 14 degrees C iodixanol showed a twofold increase in viscosity compared with ioversol and ioxaglate. At 40 degrees C, the difference was reduced to 27%. At room temperature (20 degrees C), the difference in pressure needed to inject CM was 10% between iodixanol and ioxaglate and 6% between iodixanol and ioversol. As the temperatures increased, the differences in pressure became less important, becoming negligible (1%) at 37 degrees C., Conclusion: The viscosity of the iso-osmolar nonionic contrast agent iodixanol showed a stronger dependence on temperature compared with ioversol and ioxaglate. The impact of differences in viscosity and pressure to inject between CM were minimized at 37 degrees C. This emphasizes the importance of temperature control when using current low-osmolar CM and iso-osmolar CM with smaller sized catheters., (Copyright 2008 Wiley-Liss, Inc.)

Purpose: Corneal wound healing involves a cascade of interactions between the epithelium and stroma. Pax6 is upregulated, and early events include epithelial cell migration and apoptosis of superficial keratocytes. The mouse heterozygous Pax6 (Pax6+/-) corneal phenotype mimics human aniridia-related keratopathy (ARK), and some aspects of wound healing have been shown to be abnormal, including matrix metalloproteinase (MMP)-9 expression. The purpose of this study was to test whether the Pax6+/- genotype affects corneal wound-healing responses, including stromal cell apoptosis, epithelial cell migration rate, and MMP secretion in culture., Method: Pax6+/- and wild-type (Pax6+/+) mice were killed and their corneas wounded by epithelial debridement. Whole eyes were cultured in organ culture and corneal epithelial healing rates and keratocyte apoptosis were quantified by topical fluorescein staining and TUNEL, respectively. Dissociated corneal epithelial cells from Pax6+/- and wild-type mice were cultured, and the activities of secreted MMP-9 were determined by zymography., Results: Wound-healing rates during the first 6 hours were significantly faster for larger wounds and for Pax6+/- corneas. Compared with wild-type, wounded Pax6+/- eyes showed significantly more stromal cell apoptosis, and cultured Pax6+/- corneal epithelial cells produced lower MMP-9 activity., Conclusions: The cumulative effect of abnormal wound-healing responses, characterized by increased stromal cell apoptosis and reduced levels of MMP-9 secretion may contribute to the corneal changes in the Pax6+/- mice. Possible contributions of elevated stromal cell apoptosis and other abnormal wound-healing responses to ARK are discussed.

One of the principal therapies considered for the control of in-stent restenosis is the use of drug loaded polymer-coated stents for local delivery. We present two-dimensional and three-dimensional numerical models to study local delivery of drug eluting stents. The impact of various stent and flow parameters on the concentration distribution in the wall are investigated including the effect of the strut size, coating thickness, strut inter-distance and strut embedment in the vascular wall, blood flowing speed and the respective diffusion coefficients in the blood, wall and polymer. We also present criteria to assess the drug delivery efficiency based of the concept of the therapeutic window which aims at an spatial homogeneous concentration distribution and we introduce the variables to assess the amount of drug delivered in the wall. The results suggest that advection have a much stronger effect compared to diffusion in the blood media and that drug diffusivity in the arterial wall and in the polymer coating significantly affects the drug distribution. It is also shown that fully-embedded struts provide better spatial drug concentration uniformity after a short period of time and the half-embedded struts have a better temporal uniformity.

Local mass transfer coefficients and flow patterns were examined in an idealized human aortic bifurcation model. The objectives of this study are to gain further insights on the convective mass transfer process and its possible role in the localization of atherosclerotic lesions. The laser photochromic tracer method provided velocity and wall shear stress estimates in the plane of symmetry of a UV-transparent Plexiglas bifurcation model. Steady flow data were acquired at Reynolds numbers of 500, 600, and 750. A novel copper electrodeposition technique was used to obtain time-averaged convective local mass transfer coefficients in a model identical to that used in the flow experiments. The laminar flow mass transfer data for the trunk of the bifurcation are in good agreement with the analytical Levesque solution. At the bifurcation, higher mass transfer coefficients along the inner wall and lower ones along the outer wall were observed. Further, mass transfer and wall shear stress follow similar patterns both on the inner and outer walls in that StSc 2/3 and C(f)/2 demonstrate analogous behavior. Lower transfer rates of momentum and mass occurred along the outer wall of the branches where lesions tend to develop.
