We report on a study bearing implications for ethical learning in schoolchildren during social interaction. The study was conducted as part of a project aimed at promoting ethical learning of socially-oriented values within the context of dialogic education. 172 fourth graders from 7 classes participated in an 8-session series designed to foster empathy, inclusion, and tolerance. Two of these sessions (3 and 8) were pre-selected for analysis. We investigated (1) whether students' discussion of ethical issues and the ethical aspects of their actual in-class interaction with each other can be reliably measured, and (2) what relation holds between students' ethical thinking during classroom discussions and the ethical aspects of their behavior. We thus developed an analytical framework comprising two tools for appraising ethical thinking and behavior in in-class interaction: dialogue on ethics (DoE) and ethics of dialogue (EoD). This framework was applied to the dialogues taken from the two sessions. The DoE and EoD tools proved reliable, as inter-rater agreement was substantial. Moreover, the relation between children's DoE and their EoD was positive where the topic posed for discussion presented a dilemma and students' interaction proceeded under moderate teacher guidance. In contrast, it was negative when the discussion was conceptual, and the teacher was dominant. We conclude that (1) DoE/EoD is a suitable framework for studying children's ethical learning and development in social interaction, and (2) ethical learning, in its epistemological and behavioral dimensions, can be boosted or inhibited in a context of dialogic education, depending on design principles.

The copy number of a plasmid is linked to its functionality, yet there have been few attempts to optimize higher-copy-number mutants for use across diverse origins of replication in different hosts. We use a high-throughput growth-coupled selection assay and a directed evolution approach to rapidly identify origin of replication mutations that influence copy number and screen for mutants that improve Agrobacterium-mediated transformation (AMT) efficiency. By introducing these mutations into binary vectors within the plasmid backbone used for AMT, we observe improved transient transformation of Nicotiana benthamiana in four diverse tested origins (pVS1, RK2, pSa and BBR1). For the best-performing origin, pVS1, we isolate higher-copy-number variants that increase stable transformation efficiencies by 60-100% in Arabidopsis thaliana and 390% in the oleaginous yeast Rhodosporidium toruloides. Our work provides an easily deployable framework to generate plasmid copy number variants that will enable greater precision in prokaryotic genetic engineering, in addition to improving AMT efficiency.

BackgroundMercury (Hg) is highly toxic and has the potential to cause severe health problems for humans and foraging animals when transported into edible plant parts. Soil rhizobia that form symbiosis with legumes may possess mechanisms to prevent heavy metal translocation from roots to shoots in plants by exporting metals from nodules or compartmentalizing metal ions inside nodules. Horizontal gene transfer has potential to confer immediate de novo adaptations to stress. We used comparative genomics of high quality de novo assemblies to identify structural differences in the genomes of nitrogen-fixing rhizobia that were isolated from a mercury (Hg) mine site that show high variation in their tolerance to Hg.ResultsOur analyses identified multiple structurally conserved merA homologs in the genomes of Sinorhizobium medicae and Rhizobium leguminosarum but only the strains that possessed a Mer operon exhibited 10-fold increased tolerance to Hg. RNAseq analysis revealed nearly all genes in the Mer operon were significantly up-regulated in response to Hg stress in free-living conditions and in nodules. In both free-living and nodule environments, we found the Hg-tolerant strains with a Mer operon exhibited the fewest number of differentially expressed genes (DEGs) in the genome, indicating a rapid and efficient detoxification of Hg from the cells that reduced general stress responses to the Hg-treatment. Expression changes in S. medicae while in bacteroids showed that both rhizobia strain and host-plant tolerance affected the number of DEGs. Aside from Mer operon genes, nif genes which are involved in nitrogenase activity in S. medicae showed significant up-regulation in the most Hg-tolerant strain while inside the most Hg-accumulating host-plant. Transfer of a plasmid containing the Mer operon from the most tolerant strain to low-tolerant strains resulted in an immediate increase in Hg tolerance, indicating that the Mer operon is able to confer hyper tolerance to Hg.ConclusionsMer operons have not been previously reported in nitrogen-fixing rhizobia. This study demonstrates a pivotal role of the Mer operon in effective mercury detoxification and hypertolerance in nitrogen-fixing rhizobia. This finding has major implications not only for soil bioremediation, but also host plants growing in mercury contaminated soils.

Abstract Background Zonal application maps are designed to represent field variability using key variables that can be translated into tailored management practices. For cotton, zonal maps for crop growth regulator (CGR) applications under variable-rate (VR) strategies are commonly based exclusively on vegetation indices (VIs) variability. However, VIs often saturate in dense crop vegetation areas, limiting their effectiveness in distinguishing variability in crop growth. This study aimed to compare unsupervised framework (UF) and supervised framework (SUF) approaches for generating zonal application maps for CGR under VR conditions. During 2022–2023 agricultural seasons, an UF was employed to generate zonal maps based on locally collected field data on plant height of cotton, satellite imagery, soil texture, and phenology data. Subsequently, a SUF (based on historical data between 2020–2021 to 2022–2023 agricultural seasons) was developed to predict plant height using remote sensing and phenology data, aiming to replicate same zonal maps but without relying on direct field measurements of plant height. Both approaches were tested in three fields and on two different dates per field. Results The predictive model for plant height of SUF performed well, as indicated by the model metrics. However, when comparing zonal application maps for specific field-date combinations, the predicted plant height exhibited lower variability compared with field measurements. This led to variable compatibility between SUF maps, which utilized the model predictions, and the UF maps, which were based on the real field data. Fields characterized by much pronounced soil texture variability yielded the highest compatibility between the zonal application maps produced by both SUF and UF approaches. This was predominantly due to the greater consistency in estimating plant development patterns within these heterogeneous field environments. While VR application approach can facilitate product savings during the application operation, other key factors must be considered. These include the availability of specialized machinery required for this type of applications, as well as the inherent operational costs associated with applying a single CGR product which differs from the typical uniform rate applications that often integrate multiple inputs. Conclusion Predictive modeling shows promise for assisting in the creation of zonal application maps for VR of CGR applications. However, the degree of agreement with the actual variability in crop growth found in the field should be evaluated on a field-by-field basis. The SUF approach, which is based on plant heigh prediction, demonstrated potential for supporting the development of zonal application maps for VR of CGR applications. However, the degree to which this approach aligns itself with the actual variability in crop growth observed in the field may vary, necessitating field-by-field evaluation.

Universities have consistently struggled with undergraduate student retention. However, one variable that has been shown to be an important factor related to undergraduate retention and success has been college student sense of belonging. The purpose of this study was to examine freshmen students' perceived sense of belonging to the University of Arkansas and the College of Agricultural, Food, and Life Sciences. First-year freshmen at the University of Arkansas (n = 233) were asked to provide responses on a survey measuring perceived sense of belonging, peer support, faculty support, empathy of faculty, class comfort, and isolation. Results showed respondents generally had a strong sense of belonging to the university and college of agriculture. Most students perceived faculty to be empathic and understanding, but also reported slightly lower levels of comfort when seeking help. Most students were neutral in perceptions of classroom comfort, meaning they may be less confident speaking in class, volunteering ideas, asking questions, and contributing to discussions. Just over half agreed they had peer support, while about a third perceived feeling isolated. Peer support, faculty support, and empathetic faculty were all predictors of sense of belonging in the college. Recommendations include using cooperative learning activities and continuing to provide opportunities for student organization participation to help students establish a sense of belonging.

The integration of behavioral health care within primary care settings, otherwise known as integrated care, has emerged as a treatment modality for counselors to reach a wide range of clients. However, previous counseling scholars have noted the lack of integrated care representation in counseling journals. In this scoping review, we identified 27 articles within counseling journals that provide integrated care implications. These articles appeared in 10 unique counseling journals, and the publication years ranged from 2004-2023. Articles were classified as: (a) conceptual, (b) empirical, or (c) meta-analyses and systematic reviews. The data extracted from the articles focused on the implications for integrated care training and practice for the next generation of counselors, evidence-based treatment approaches, and future research directions.

Urbanization is related to the loss of biodiversity and alteration of ecosystem processes. However, long-term changes in the southern hemisphere’s bird communities have been scarcely analyzed in urban areas. The general objective of this work was to analyze changes in bird species richness, diversity, and composition in parks located in the urban and suburban landscapes of Mar del Plata City (Argentina) between 2003 and 2018. Additionally, we aimed to analyze whether these changes were associated with an increase in urbanization or with alterations in the parks’ habitat structure. Tree cover showed significantly lower coverage during 2018. The results showed increased species richness and diversity in all parks by 2018. Rarefaction curves showed that the park bird richness did not differ between the landscapes surrounding parks, but the Shannon and Simpson diversities were higher in parks in suburban landscapes than in parks in urban landscapes. The Shannon and Simpson diversities showed higher values in suburban parks during 2018. However, the diversity values were similar between years in parks in urban landscapes. The species composition was significantly different between landscapes and years. Species turnover was the most predominant pattern of species dissimilarity between landscapes, and species nestedness explained the most dissimilarity between years. Nestedness between years was related to the population increase in many bird species in 2018. In conclusion, we observed differences in the park bird communities between years and landscape types. However, the long-term community changes were not associated with an urbanization increase. Instead, the changes in the local bird community were more related to regional bird species population changes associated with climate warming and increased crop cover.

The process by which infants learn verbs through daily social interactions is not well-understood. This study investigated caregivers' use of verbs, which have highly abstract meanings, during unscripted toy-play. We examined how verbs co-occurred with distributional and embodied factors including pronouns, caregivers' manual actions, and infants' locomotion, gaze, and object-touching. Object-action verbs were used significantly more often during caregiver-infant joint attention interactions. Movement and cognition verbs showed distinct co-occurrences with different contexts. Cognition and volition verbs were differentiated by pronouns. These findings provide evidence for how verb acquisition may be supported by the distributional and embodied contexts in caregiver-infant interactions.

Peer mentoring has shown beneficial effects in facilitating academic and social integration among first-year students in higher education. Previous research is, however, limited by the exclusion of a comparison group, to examine whether the integration process still occurs among non-mentored students, independently of peer mentoring. By using Tinto´s theoretical framework, we examine the effects of a formal peer mentoring program, comparing these effects on integration with non-mentored students. Data was gathered from focus group interviews and analyzed using thematic analysis. Our findings show that peer mentoring supports the integration process among first-year students, although the results suggest that it is not a necessary condition to ensure that integration occurs. Future studies should further extend the research to also include an evaluation of other activities taking place at the institution, to expand the knowledge of which measures facilitates the integration process.

This study evaluated the use of two different gonadotropin-releasing hormone (GnRH) compounds in estrus synchronization protocols with or without presynchronization. Cows (n = 1,585) were enrolled in a completely randomized design with a 2×2 factorial arrangement of treatments. Within each location, cows were randomly assigned to one of four treatments: (1 and 2) cows were estrus synchronized using the 7-day CO-Synch + CIDR protocol (7D) wherein they received an injection of one of two GnRH compounds [gonadorelin hydrochloride (GH) or gonadorelin diacetate tetrahydrate (GDT)] and a controlled internal drug release (CIDR) device on day 0, an injection of prostaglandin F2α (PG) at CIDR removal on day 7, and a second injection of their respective GnRH compound at fixed-time artificial insemination (TAI) on day 10 (7D-GH and 7D-GHT); (3 and 4) cows were treated with the 7&7 Synch (7&7) protocol, wherein they were treated the same as treatments 1 and 2 but received their CIDR inserts on day -7 in conjunction with an injection of PG (7&7-GH and 7&7-GDT). Breeding indicator patches were applied to all cows at CIDR removal and were evaluated for activation at TAI. Follicle diameter differed by ovulation synchronization protocol on days 0, 7, and 10, where cows enrolled in the 7&7 had greater (P ≤ 0.008) dominant follicle diameters than those in the 7D. No differences (P ≥ 0.19) in ovarian parameters were determined between GH and GDT cows. Estrus expression differed (P < 0.001) by synchronization protocol and was greater in cows enrolled in the 7&7 than the 7D (80.4 ± 2.8 vs. 55.5 ± 4.1%, respectively), yet no differences (P = 0.32) in estrus expression were determined between GH- and GDT-treated cows. Pregnancy rates to TAI did not differ (P = 0.57) by GnRH compound but differed (P = 0.01) by synchronization protocol, where cows enrolled in the 7&7 had greater PR/AI when compared to those in the 7D (60.9 ± 2.5 vs. 53.9 ± 2.6%, respectively). In conclusion, fertility was improved through the use of presynchronization; however, no differences in fertility parameters were determined between GH and GDT in either synchronized or presynchronized beef cows.

BackgroundThe integration of information technology into health care has created opportunities to address diagnostic challenges. Internet searches, representing a vast source of health-related data, hold promise for improving early disease detection. Studies suggest that patterns in search behavior can reveal symptoms before clinical diagnosis, offering potential for innovative diagnostic tools. Leveraging advancements in machine learning, researchers have explored linking search data with health records to enhance screening and outcomes. However, challenges like privacy, bias, and scalability remain critical to its widespread adoption. ObjectiveWe aimed to explore the potential and challenges of using internet search data in medical diagnosis, with a specific focus on diseases and conditions such as cancer, cardiovascular disease, mental and behavioral health, neurodegenerative disorders, and nutritional and metabolic diseases. We examined ethical, technical, and policy considerations while assessing the current state of research, identifying gaps and limitations, and proposing future research directions to advance this emerging field. MethodsWe conducted a comprehensive analysis of peer-reviewed literature and informational interviews with subject matter experts to examine the landscape of internet search data use in medical research. We searched for published peer-reviewed literature on the PubMed database between October and December 2023. ResultsSystematic selection based on predefined criteria included 40 articles from the 2499 identified articles. The analysis revealed a nascent domain of internet search data research in medical diagnosis, marked by advancements in analytics and data integration. Despite challenges such as bias, privacy, and infrastructure limitations, emerging initiatives could reshape data collection and privacy safeguards. ConclusionsWe identified signals correlating with diagnostic considerations in certain diseases and conditions, indicating the potential for such data to enhance clinical diagnostic capabilities. However, leveraging internet search data for improved early diagnosis and health care outcomes requires effectively addressing ethical, technical, and policy challenges. By fostering interdisciplinary collaboration, advancing infrastructure development, and prioritizing patient engagement and consent, researchers can unlock the transformative potential of internet search data in medical diagnosis to ultimately enhance patient care and advance health care practice and policy.

Abstract The exploration of extraterrestrial environments has become a focal point of scientific inquiry, driven by advancements in technology and a growing interest in the potential for life beyond Earth. This study investigates the adaptability of Antarctic nematodes, known for thriving in extreme cold and isolation, to simulated Martian (MGS-1) and Lunar (LMS-1) soils. The experiment revealed differential responses in nematode survivability to the two simulants, with Lunar soil demonstrating better adaptability compared to Martian soil. Our study provides valuable insights into the potential survivability of Antarctic nematodes on real Martian and Lunar substrates with the use of the MGS-1 and the LMS-1 under terrestrial conditions. The observed results suggest differential survival responses to Lunar (6.92 ± 0.74) and Martian (5.58 ± 1.91) regolith simulants and highlight the complexity of factors influencing organismal survival. Considering their crucial role in soil food webs, and their relation with nutrient cycling and carbon dioxide emissions, the observed capacity of nematodes to thrive in Martian and Lunar regoliths positions them as potential candidates to sustain human habitats, helping on the development of in-situ resource utilization (ISRU) systems, and forming the basis for a biological life-support system to facilitate human survival in extraterrestrial environments.

Background: Identifying and eliminating health disparities is a public health priority. The goal of this analysis is to determine whether cardiac testing or outcome disparities exist by race or sex in patients with detectable to mildly elevated serum troponin. Methods: We conducted a secondary analysis of the CMR-IMPACT trial that randomized patients with symptoms suggestive of acute coronary syndrome and a detectable or mildly elevated troponin measure from 4 US hospitals to an early invasive angiography or cardiac MRI strategy. The primary endpoint was the composite of all-cause mortality, myocardial infarction, cardiac hospital readmission, and repeat cardiac ED. Secondary outcomes were components of the composite and revascularization. Results: Participants (n = 312, mean age 61 ± 11 years) were 36.2 % non-white and 40.1 % female. The composite outcome occurred in 63.7 % of non-white vs. 49.8 % of white patients (aHR 1.50, 95 % CI 1.08–2.09) and 53.6 % of female vs. 55.6 % of male patients (aHR 0.93, 95 % CI 0.68–1.28). Hirschsprung’s disease (HSCR) is characterized by congenital absence of ganglion cells in the gastrointestinal tract, which leads to impaired defecation, constipation and intestinal obstruction. The current diagnosis of HSCR is based on Rectal Suction Biopsies (RSBs), which could be complex in newborns. Occasionally, there is a delay in diagnosis that can increase the risk of clinical complications. Consequently, there is room for new non-invasive diagnostic methods that are objective, more logistically feasible and also deliver a far earlier base for a potential surgical intervention. In recent years, microRNA (miRNA) has come into the focus as a relevant early marker that could provide more insights into the etiology and progression of diseases. Therefore, in the search of a non-invasive HSCR biomarker, we analyzed miRNA expression in urine samples of HSCR patients. Results from 5 HSCR patients using microarrays, revealed hsa-miR-378 h, hsa-miR-210-5p, hsa-miR-6876-3p, hsa-miR-634 and hsa-miR-6883-3p as the most upregulated miRNAs; while hsa-miR-4443, hsa-miR-22-3p, hsa-miR-4732-5p, hsa-miR-3187-5p, and hsa-miR-371b-5p where the most downregulated miRNAs. Further search in miRNAwalk and miRDB databases showed that certainly most of these dysregulated miRNAs identified target HSCR associated genes, such as RET, GDNF, BDNF, EDN3, EDNRB, ERBB, NRG1, SOX10; and other genes implied in neuronal migration and neurogenesis. Finally, we could also validate some of these miRNA changes in HSCR urine by RT-qPCR. Altogether, our analyzed HSCR cohort presents a dysregulated miRNA expression presents that can be detected in urine. Our findings open the possibility of using specific urine miRNA signatures as non-invasive HSCR diagnosis method in the future.

Tin dioxide (SnO2) is an oxide semiconductor with n-type characteristics, with high transparency in the UV–Vis, where the donors are usually associated with oxygen vacancies and interstitial tin ions. Quinoline derivatives (QD) are usually p-type semiconductors with emission in the blue range. We report photo-induced properties of the QD 4-(6-(diethylamino)-4-phenylquinolin-2-yl)benzoic acid and the combination with the inorganic semiconductor oxide SnO2, both layers in the form of thin film, which forms a heterostructure. Thin film is a very convenient format for integration in optoelectronics. Emission of the QD takes place in blue range (470–485 nm) and depends on the solvent when in solution, being used acetone and tetrahydrofuran (THF). However, when in the form of thin film, it does not depend on the solvent. Concerning the heterostructure, it is explored under distinct device architecture: 1) combination in a transport profile perpendicular to the films (transverse contacts) leading to a rectifying behavior similar to a p-n junction, which is evidence of the p-type-like electrical behavior of the QD; 2) in parallel conduction profile, where there seems to exist some sort of interfacial phenomenon similar to a two-dimensional electron gas (2-DEG), a property that can be explored in transparent high-mobility transistors.

The pGinger suite of expression plasmids comprises 43 plasmids that will enable precise constitutive and inducible gene expression in a wide range of Gram-negative bacterial species. Constitutive vectors are composed of 16 synthetic constitutive promoters upstream of red fluorescent protein (RFP), with a broad-host-range BBR1 origin and a kanamycin resistance marker. The family also has seven inducible systems (Jungle Express, Psal/NahR, Pm/XylS, Prha/RhaS, LacO1/LacI, LacUV5/LacI, and Ptet/TetR) controlling RFP expression on BBR1/kanamycin plasmid backbones. For four of these inducible systems (Jungle Express, Psal/NahR, LacO1/LacI, and Ptet/TetR), we created variants that utilize the RK2 origin and spectinomycin or gentamicin selection. Relevant RFP expression and growth data have been collected in the model bacterium Escherichia coli as well as Pseudomonas putida. All pGinger vectors are available via the Joint BioEnergy Institute (JBEI) Public Registry. IMPORTANCE Metabolic engineering and synthetic biology are predicated on the precise control of gene expression. As synthetic biology expands beyond model organisms, more tools will be required that function robustly in a wide range of bacterial hosts. The pGinger family of plasmids constitutes 43 plasmids that will enable both constitutive and inducible gene expression in a wide range of nonmodel Proteobacteria.
