Objectives: Topical treatment with acyclovir cream has shown low efficacy in recent studies. Nano drug delivery systems, have received much attention in recent decades. The aim of this study was to compare the efficacy of acyclovir nanofiber patch with acyclovir cream. Material and Methods: In this double‐blind three‐armed randomized clinical trial, a total of 60 patients with recurrent labial herpes, were randomly divided into three groups, each consisting of 20. The patients in the first, second, and third groups were treated with acyclovir nanofiber patch, placebo nanofiber patch, and acyclovir cream, respectively. A numerical scale was used by the patients to record the self‐reported symptoms. Symptoms score, crusting time and healing time were assessed by the clinician. Kruskal‐Wallis test was used to compare the symptoms between the three groups, a survival test was also performed to evaluate the crusting and healing time. Data were analyzed in SPSS V22 at P‐value < 0.05. Results: The mean scores of symptoms at baseline were 1.6, 1.5, and 1.4 in the first, second, and third groups, respectively. The symptoms were not significantly different between the three groups on different treatment days. The mean crusting time was 2.3, 2.4, and 2.6 days in the three groups, and the mean healing time was 7.4, 7.2, and 7.7 days, respectively. Crusting time and healing time were not significantly different between the three groups. Conclusions: Acyclovir nanofiber patches are recommended for accelerating symptom relief in recurrent labial herpes, however, they are not effective in shortening the crusting or healing time. A simple 2‐step method, consisting of film casting and polyvinyl alcohol leaching, is proposed to prepare magnesium oxide (MO) nanoparticle‐reinforced sodium alginate scaffolds with right properties for bone tissue engineering. The cytocompatibility of the as‐prepared scaffolds was also evaluated using the 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium‐bromide yellow tetrazole assay test, wherein chondrocyte cells had been considered as target cells. According to the results, the ensuing sodium alginate nanocomposites, containing 4‐wt% MO nanoparticles, demonstrated the highest physical and mechanical properties after leaching step. The Young modulus of sodium alginate/4‐wt% MO was improved about 44%, in comparison with that of the pure alginate sample. Furthermore, incorporating MO nanoparticles up to 4 wt% controlled the liquid uptake capacity of scaffolds vis‐à‐vis the resultant pure sodium alginate sample. Moreover, with increasing the nanoparticle content, the antibacterial properties of scaffolds enhanced, but their degradation rates under in vitro conditions tapered off. With the introduction of 3‐ and 4‐wt% MO, the average diameter of the bacterial zone of the scaffold samples reduced to less than 10 mm2, suggesting an insensitive antimicrobial performance, compared with the pure sodium alginate and the samples with 1‐ and 2‐wt% MO content, which exhibit antimicrobial sensitivity. 3‐(4,5‐Dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium‐bromide assay test also revealed the cultivated chondrocyte cells on the 4‐wt% MO nanoparticle‐reinforced scaffold possessed better interaction as well as appropriate cell attachment and proliferation than the pristine sodium alginate sample. Background: Wound healing is a complex biological process. Some injuries lead to chronic nonhealing ulcers, and healing process is a challenge to both the patient and the medical team. We still look forward an appropriate wound dressing. Materials and Methods: In this study, starch-based nanocomposite hydrogel scaffolds reinforced by zeolite nanoparticles (nZ) were prepared for wound dressing. In addition, a herbal drug (chamomile extract) was added into the matrix to accelerate healing process. To estimate the cytocompatibility of hydrogel dressings, fibroblast mouse cells (L929) were cultured on scaffolds. Then, 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium-bromide assay test and interaction of cells and scaffolds were evaluated. For evaluating healing process, 48 male rats were randomly divided into four groups of four animals each (16 rats at each step). The ulcers of the first group were treated with the same size of pure hydrogels. The second group received a bandage with the same size of hydrogel/extract/4 wt% nZ (hydrogel NZE). The third group was treated with chamomile extract, and the fourth group was considered as control without taking any medicament. Finally, the dressings were applied on the chronic refractory ulcers of five patients. Results: After successful surface morphology and cytocompatibility tests, the animal study was carried out. There was a significant difference between starch/extract/4 wt% nZ and other groups on wound size decrement after day 7 (P < 0.05). At the clinical pilot study step, the refractory ulcers of all five patients were healed without any hypersensitivity reaction. The present study describes a novel and very sensitive electrochemical assay for determination of hydrogen peroxide (H2O2) based on synergistic effects of reduced graphene oxide- magnetic iron oxide nanocomposite (rGO-Fe3O4) and celestine blue (CB) for electrochemical reduction of H2O2. rGO-Fe3O4 nanocomposite was synthesized and characterized by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), X-ray diffraction (XRD), electrochemical impedance spectroscopy and cyclic voltammetry. Chitosan (Chit) was used for immobilization of amino-terminated single-stranded DNA (ss-DNA) molecules via a glutaraldehyde (GA) to the surface of rGO-Fe3O4. The MTT (3-(4,5-Di methyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide) results confirmed the biocompatibility of nanocomposite. Experimental parameters affecting the ss-DNA molecules immobilization were optimized. Finally, by accumulation of the CB on the surface of the rGO-Fe3O4-Chit/ssDNA, very sensitive amperometric H2O2 sensor was fabricated. The electrocatalytic activity of the rGO-Fe3O4-Chit/DNA-CB electrode toward H2O2 reduction was found to be very efficient, yielding very low detection limit (DL) of 42 nM and a sensitivity of 8.51 μA/μM. Result shows that complex matrices of the human serum samples did not interfere with the fabricated sensor. The developed sensor provided significant advantages in terms of low detection limit, high stability and good reproducibility for detection of H2O2 in comparison with recently reported electrochemical H2O2 sensors. Cartilage is under extensive investigation in tissue engineering research. Herein, we evaluated scaffolds prepared by composites of polyvinyl alcohol (PVA) and collagen incorporated with zeolite and silica nanoparticles (nZe and nSi). The scaffolds were prepared by the electrospinning method. The mean diameters of nanofibers were 0.61 ± 0.34 µm for PVA/collagen versuss 0.62 ± 0.22 µm and 0.66 ± 0.25 µm for the PVA/collagen/nZe and the PVA/collagen/nSi scaffolds, respectively. DAPI staining results revealed that cell proliferations on the PVA/collagen/nZe and PVA/collagen/nSi were strikingly higher than on the pure PVA/collagen. The results encouraged further investigation of PVA/collagen/nSi scaffolds as biomimetic platform for chondrocyte cells in tissue engineering. [ABSTRACT FROM AUTHOR]
