The study aimed to assess the effects of dietary black soldier fly oil (BSFO) on the growth performance, flesh quality, and health status of largemouth bass (Micropterus salmoides). Six iso-nitrogenous and isolipid diets were formulated by substituting fish oil and soybean oil (1/2, wt/wt) with BSFO in percentages of 0%, 20%, 40%, 60%, 80%, and 100%, respectively. The diets were fed to 960 fish (initial body weight = 16.5 g) in four replicates for 8 weeks. Indicators related to growth performance, body composition, hematology, flesh quality, expression of genes related to inflammatory cytokines and apoptosis, and the response of fish to Aeromonas veronii challenge were analyzed. The results showed that the weight gain rate was numerically improved in all BSFO substitution groups, ranging from 9.3% to 44.0% compared to the control group. The highest survival rate and the lowest hepatosomatic index and condition factor were observed in the BFSO20 group. In terms of flesh quality, the water-holding capacity of the dorsal muscle was elevated with higher levels of dietary BSFO. Abstract Background Acute retinal necrosis (ARN) is a relatively rare but highly damaging and potentially sight-threatening type of uveitis caused by infection with the human herpesvirus. Without timely diagnosis and appropriate treatment, ARN can lead to severe vision loss. We aimed to develop a deep learning framework to distinguish ARN from other types of intermediate, posterior, and panuveitis using ultra-widefield color fundus photography (UWFCFP). Methods We conducted a two-center retrospective discovery and validation study to develop and validate a deep learning model called DeepDrARN for automatic uveitis detection and differentiation of ARN from other uveitis types using 11,508 UWFCFPs from 1,112 participants. Model performance was evaluated with the area under the receiver operating characteristic curve (AUROC), the area under the precision and recall curves (AUPR), sensitivity and specificity, and compared with seven ophthalmologists. Results DeepDrARN for uveitis screening achieved an AUROC of 0.996 (95% CI: 0.994–0.999) in the internal validation cohort and demonstrated good generalizability with an AUROC of 0.973 (95% CI: 0.956–0.990) in the external validation cohort. DeepDrARN also demonstrated excellent predictive ability in distinguishing ARN from other types of uveitis with AUROCs of 0.960 (95% CI: 0.943–0.977) and 0.971 (95% CI: 0.956–0.986) in the internal and external validation cohorts. DeepDrARN was also tested in the differentiation of ARN, non-ARN uveitis (NAU) and normal subjects, with sensitivities of 88.9% and 78.7% and specificities of 93.8% and 89.1% in the internal and external validation cohorts, respectively. The performance of DeepDrARN is comparable to that of ophthalmologists and even exceeds the average accuracy of seven ophthalmologists, showing an improvement of 6.57% in uveitis screening and 11.14% in ARN identification. Conclusions Our study demonstrates the feasibility of deep learning algorithms in enabling early detection, reducing treatment delays, and improving outcomes for ARN patients.

Abstract Dual emission (DE) in nanoclusters (NCs) is considerably significant in the research and application of ratiometric sensing, bioimaging, and novel optoelectronic devices. Exploring the DE mechanism in open-shell NCs with doublet or quartet emissions remains challenging because synthesizing open-shell NCs is difficult due to their inherent instability. Here, we synthesize two dual-emissive M1Ag13(PFBT)6(TPP)7 (M = Pt, Pd; PFBT = pentafluorobenzenethiol; TPP = triphenylphosphine) NCs with a 7-electron open-shell configuration to reveal the DE mechanism. Both NCs comprise a crown-like M1Ag11 kernel with Pt or Pd in the center surrounded by five PPh3 ligands and two Ag(SR)3(PPh3) motifs. The combined experimental and theoretical studies revealed the origin of DE in Pt1Ag13 and Pd1Ag13. Specifically, the high-energy visible emission and the low-energy near-infrared emission arise from two distinct quartet excited states: the core-shell charge transfer and core-based states, respectively. Moreover, PFBT ligands are found to play an important role in the existence of DE, as its low-lying π* levels result in energetically accessible core-shell transitions. This novel report on the dual-quartet phosphorescent emission in NCs with an open-shell electronic configuration advances insights into the origin of dual-emissive NCs and promotes their potential application in magnetoluminescence and novel optoelectronic devices.

According to studies by relevant scholars, the ground subsidence rate in Sanhe City, Hebei, increased gradually from 2003 to 2016, with the most severe subsidence observed in the Yanjiao area , which has become contiguous with the subsidence area in Tongzhou, Beijing. However, the development trend of ground subsidence disasters in Sanhe City after 2016 remains unclear. With the implementation of national policies such as the integration of Beijing-Tianjin-Hebei region and the construction of Beijing’s sub-center, it is crucial to identify the development and evolution characteristics of land subsidence disasters in Sanhe City and analyze their causes to ensure the urban safety and sustainable development of Sanhe City. In this context, the authors used SBAS-InSAR technology to interpret the evolution characteristics of ground subsidence in Sanhe City from 2018 to 2020, and analyzed several inducing factors that led to land subsidence in Sanhe City, summarizing the main causes. Through this study, the spatial distribution and evolutionary characteristics of land subsidence disasters in Sanhe City were grasped: the western part of the city exhibits severe subsidence, while the eastern part is less affected, with a focus on the overall characteristics of key subsidence areas. Two main subsidence areas were identified, namely, the Yanjiao Town subsidence area with three subsidence funnels and the DuanJialing town subsidence area with one subsidence funnel, with the former being the most severely affected. From 2018 to 2020, the overall trend of ground subsidence disasters in Sanhe City showed a slowdown. Through comparative analysis, it was determined that severe overexploitation of groundwater, soil properties, urbanization development, and population growth are the main factors contributing to the occurrence and development of ground subsidence disasters in Sanhe City. The research results of this paper will provide reference for the prevention and control of ground subsidence disasters in the region.

Abstract The design and preparation of efficient catalysts for ammonia production under mild conditions is a desirable but highly challenging target. Here, we report a series of single-atom catalysts [M-SACs, M = Mn(II), Fe(III), Co(II), Ni(II), Cu(II), Zn(II), Mo(II)] derived from UiO-66 containing structural defects and their application to electrochemical reduction of nitrate (NO3 -) to ammonia (NH3). Cu-SAC and Fe-SAC exhibit remarkable yield rates for NH3 production of 30.0 and 29.0 mg h−1 cm−2, respectively, with a high Faradaic efficiency (FENH3) of over 96% at −1.0 V versus the reversible hydrogen electrode. Importantly, their catalytic performance can be retained in various simulated wastewaters. Complementary experiments confirmed the nature of single-atom sites within these catalysts and the binding domains of NO3 - in UiO-66-Cu. In situ spectroscopic techniques, coupled with density functional theory calculations confirm the strong binding of NO3 - and the formation of reaction intermediates, thus facilitating the catalytic conversion to NH3.

A significant clinical challenge in large-to-massive rotator cuff tendon injuries is the need for sustaining high mechanical demands despite limited tissue regeneration, which often results in clinical repair failure with high retear rates and long-term functional deficiencies. To address this, an innovative tendon substitute named “BioTenoForce” is engineered, which uses (i) tendon extracellular matrix (tECM)'s rich biocomplexity for tendon-specific regeneration and (ii) a mechanically robust, slow degradation polyurethane elastomer to mimic native tendon's physical attributes for sustaining long-term shoulder movement. Comprehensive assessments revealed outstanding performance of BioTenoForce, characterized by robust core-shell interfacial bonding, human rotator cuff tendon-like mechanical properties, excellent suture retention, biocompatibility, and tendon differentiation of human adipose-derived stem cells. Importantly, BioTenoForce, when used as an interpositional tendon substitute, demonstrated successful integration with regenerative tissue, exhibiting remarkable efficacy in repairing large-to-massive tendon injuries in two animal models. Noteworthy outcomes include durable repair and sustained functionality with no observed breakage/rupture, accelerated recovery of rat gait performance, and >1 cm rabbit tendon regeneration with native tendon-like biomechanical attributes. The regenerated tissues showed tendon-like, wavy, aligned matrix structure, which starkly contrasts with the typical disorganized scar tissue observed after tendon injury, and was strongly correlated with tissue stiffness. Our simple yet versatile approach offers a dual-pronged, broadly applicable strategy that overcomes the limitations of poor regeneration and stringent biomechanical requirements, particularly essential for substantial defects in tendon and other load-bearing tissues.

Some structures or components of coastal buildings are often in an unsaturated state between drying and wetting conditions, and their performance deterioration is extremely serious. This paper mainly studied Cl− diffusion of coral aggregate concrete under the coupled erosion of drying-wetting cycles, sulfate and chloride salts. Based on the microscopic morphology of corroded concrete, the erosion mechanism of the concrete under the coupled erosion of drying-wetting cycles sulfate and chloride salt was elaborated. The results indicated that, under the coupling erosion of sulfate and chloride salts, a large amount of Na2SO4 crystals will precipitate on the surface of concrete. The addition of mineral admixtures can not only improve the relative dynamic elastic modulus and relative compressive strength of coral aggregate concrete, but also improve its resistance to Cl− corrosion. SO42− reacts with cement hydration products to produce ettringite and gypsum, which hinder further Cl− corrosion during the early stage of erosion. However, as the erosion time increases, large numbers of erosion products lead to continuous expansion of cracks and accelerate Cl− transport. Finally, based on the transport mechanism of Cl− in the concrete, machine learning prediction models for Cl− content were established using CART, SVR, and XGBoost, respectively. It was found that the predicted values of the XGBoost model were in good agreement with the measured values, and the goodness of fit of the test set was R2 = 0.98, RMSE = 0.021%, MAE = 0.013%. This work can provide a new method for predicting chloride concentration in concrete under the coupling effect of multiple factors.

In this study, Al2O3–Cu/35Mo composites were prepared by rapid hot-press sintering-internal oxidation method, and the materials were modified by adding trace amounts of graphene oxide (GO). The results show that the relative densities of the composites are above 99%, the γ-Al2O3 generated by internal oxidation is diffusely distributed on the copper matrix. The incorporation of GO produced a small number of hard MoC particles at the interface, which facilitated the interfacial bonding of the composites. In the 30 V DC, 10–30 A electrical contact test, the anode mass of the composites increases, while the cathode mass decreases. And GO reduces the total mass loss of the electric contact by 63.6%, 38.9%, 51.3% and 20.9%, respectively. A comparison of the electrical contact properties of the two materials showed that the addition of GO dispersed the concentrated erosion of the arc. At 10 A and 30 A, the arc energy is reduced by 75.5% and 12.5%, respectively. With the gradual increase in electric current, GO makes the Al2O3–Cu/35Mo composites more stable during the electric contact process, improves the anti-welding ability of the contact and reduces contact resistance.

Abstract Activation of the NF-κB pathway is strictly regulated to prevent excessive inflammatory and immune responses. In a well-known negative feedback model, IκBα-dependent NF-κB termination is a delayed response pattern in the later stage of activation, and the mechanisms mediating the rapid termination of active NF-κB remain unclear. Here, we showed IκBα-independent rapid termination of nuclear NF-κB mediated by CLK2, which negatively regulated active NF-κB by phosphorylating the RelA/p65 subunit of NF-κB at Ser180 in the nucleus to limit its transcriptional activation through degradation and nuclear export. Depletion of CLK2 increased the production of inflammatory cytokines, reduced viral replication and increased the survival of the mice. Mechanistically, CLK2 phosphorylated RelA/p65 at Ser180 in the nucleus, leading to ubiquitin‒proteasome-mediated degradation and cytoplasmic redistribution. Importantly, a CLK2 inhibitor promoted cytokine production, reduced viral replication, and accelerated murine psoriasis. This study revealed an IκBα-independent mechanism of early-stage termination of NF-κB in which phosphorylated Ser180 RelA/p65 turned off posttranslational modifications associated with transcriptional activation, ultimately resulting in the degradation and nuclear export of RelA/p65 to inhibit excessive inflammatory activation. Our findings showed that the phosphorylation of RelA/p65 at Ser180 in the nucleus inhibits early-stage NF-κB activation, thereby mediating the negative regulation of NF-κB.

Compression tests of the Cu-1.5Ti and Cu-1.5Ti-0.5Fe alloys were carried out using the Gleeble-1500D hot deformation simulator. The deformation temperature was 750–900 °C and the strain rate was 0.1–0.003 s−1. The Cu-1.5Ti-0.5Fe alloy clearly shows higher rheological stress and displays dynamic recovery and dynamic recrystallization characteristics under the same deformation conditions. The constitutive equations of the alloys were established. The activation energy of the Cu-1.5Ti alloy was 341.76 kJ/mol, which increased to 378.03 kJ/mol with the addition of 0.5 wt% Fe. After 750 °C and 0.003 s−1 deformation the Cu4Ti phase precipitated in Cu-1.5Ti alloy. Adding 0.5 wt% Fe caused dynamic recrystallization during hot deformation. The hot-deformed Cu-1.5Ti-0.5Fe alloy had more dislocations and finer grains with Cu4Ti and Fe2Ti precipitated phases.

Magnetic reconnection and plasma waves share a critical connection. On the one hand, quasi-steady wave structures (shock, KAW eigenmode) play a pivotal role in the formation and structure of magnetic reconnection. On the other hand, higher-frequency and small-scale waves may influence energy conversion, particle heating, and anomalous resistivity. Waves of various scales dictate the multi-scale coupling characteristics and energy conversion within the reconnection process. This review focuses on plasma waves during the Earth's magnetosphere magnetic reconnection, seeking to elucidate the characteristics and roles of kinetic Alfven waves (KAW), low hybrid waves, whistler waves, electrostatic solitary waves (ESWs), ion acoustic waves, and electron-scale high-frequency electrostatic waves. The KAW eigenmode is capable of describing various phenomena in the diffusion region such as Hall magnetic fields and Hall electric fields, significantly impacting the reconnection rate. Lower hybrid waves are easily excited in the strong density gradient in the current layer and found to heat electrons in the parallel direction, while whistler waves are excited through Landau resonance and cyclotron resonance. Both whistler waves and lower hybrid waves are found to exhibit minimal influence on the reconnection rate through the anomalous resistivity. ESWs predominantly emerge within the magnetic reconnection boundary region, necessitating further investigation into their heating effects. Finally, the review delves into high-frequency electrostatic waves, emphasizing their importance in the magnetic reconnection diffusion region, specifically for electron diffusion and scattering processes. The insights gained from these research advancements serve as a foundation for future studies and advancements in the field of magnetic reconnection and plasma wave interactions.

Abstract Small cell lung cancer (SCLC) is a highly aggressive malignancy characterized by rapid growth and early metastasis and is susceptible to treatment resistance and recurrence. Understanding the intra‐tumoral spatial heterogeneity in SCLC is crucial for improving patient outcomes and clinically relevant subtyping. In this study, a spatial whole transcriptome‐wide analysis of 25 SCLC patients at sub‐histological resolution using GeoMx Digital Spatial Profiling technology is performed. This analysis deciphered intra‐tumoral multi‐regional heterogeneity, characterized by distinct molecular profiles, biological functions, immune features, and molecular subtypes within spatially localized histological regions. Connections between different transcript‐defined intra‐tumoral phenotypes and their impact on patient survival and therapeutic response are also established. Finally, a gene signature, termed ITHtyper, based on the prevalence of intra‐tumoral heterogeneity levels, which enables patient risk stratification from bulk RNA‐seq profiles is identified. The prognostic value of ITHtyper is rigorously validated in independent multicenter patient cohorts. This study introduces a preliminary tumor‐centric, regionally targeted spatial transcriptome resource that sheds light on previously unexplored intra‐tumoral spatial heterogeneity in SCLC. These findings hold promise to improve tumor reclassification and facilitate the development of personalized treatments for SCLC patients.

High-throughput phenotypic analysis plays an increasingly important role in crop breeding. In such research, the breeder usually establishes hundreds to thousands of plots, with each plot having its independent genetic breeding sources. The breeding plot extraction of genetic sources is usually outlined manually on RGB UAV imagery, which is time-consuming and subject to human bias. Therefore, a rapid method to extract the breeding plot for each genetic source in high-throughput phenotypic analysis would be very significant. In this paper, we propose a transferable method for extracting breeding plots from UAV RGB imagery. We utilized the fully convolutional neural network model A-UNet with an attention gate. After obtaining binary raster data from deep learning, we introduced post-processing. Subsequently, the raster data after post-processing were converted to vector data to obtain geographical coordinates. Finally, the UAV imagery was masked by the vector data to obtain the extraction results for each plot. The results showed that A-UNet achieved accuracies of over 90 % in precision, recall, and F1 score. Post-processing resulted in a 93 % average IoU in breeding plot extraction in the main study area. The average IOU achieved over 86 % in different spatial resolutions (1.6 cm and 0.4 cm), plot sizes (1 m × 1.5 and 2 m × 5 m), and crop types (rice). In summary, this study developed a method for extracting breeding plots in high-throughput phenotype analysis, which would help to be used as a high-throughput screening technique for accelerating crop breeding.

Intraglandular dissemination is an important pathological feature of thyroid cancer, yet the biological characteristics of this phenomenon remain relatively underexplored. This paper aims to provide a comprehensive overview of its biological behaviors, protein expressions, and identification methods. Several retrospective studies have found that thyroid cancers with intraglandular dissemination have higher rates of lymph node metastasis, capsule invasion, and vascular invasion, exhibiting more aggressive biological behavior. Immunohistochemistry results show abnormal expression of proteins such as FKBP5, CENPF, CX26, KIF11, PTK7, which are associated with poor prognosis in thyroid cancers with intraglandular dissemination, offering potential guidance for specific targeted therapy in the future. Moreover, adjunctive techniques including ultrasound, fine-needle aspiration, and genetic testing offer valuable support in accurately identifying these cases, facilitating moreproactive treatment and closer follow-up.

ObjectiveThe objective of this research is to investigate the relationship between dietary glycine consumption and the prevalence of hypertension, hyperlipidemia, and overweight or obesity in economically disadvantaged areas of northern China using a cross-sectional study design.MethodsA cross-sectional study involving 774 participants utilized a web-based dietary questionnaire (IDQC) and underwent physical measurements. Data analysis was conducted using IBM SPSS Statistics software (Version 21). Participants were stratified into four groups based on quartiles of their dietary glycine intake: Q1 (2.26). Continuous variables were reported as mean ± standard deviation and compared using ANOVA or the Kruskal-Wallis test, while categorical variables were presented as frequencies (%) and compared using the chi-square test. Finally, multivariable logistic regression with p-value of less than 0.05 was considered statistically significant.ResultsSignificant differences in dietary glycine intake were observed between the highest quartile group (Q4) and the lowest quartile group (Q1), with corresponding dominance ratios of 0.590 (95% CI, 0.360–0.966), 0.547 (95% CI, 0.327–0.913), and 0.547 (95% CI, 0.353–0.850) for the risk of hypertension, hyperlipidemia, and overweight/obesity, respectively. Furthermore, no significant correlation was found between dietary glycine intake and hypertension or hyperlipidemia within each sex and age subgroup.ConclusionThere exists a potential correlation between increased dietary glycine intake and reduced prevalence of hypertension, hyperlipidemia, and overweight/obesity. However, additional research is necessary to validate this finding through larger-scale studies conducted at a population level.

The Amundsen Sea of West Antarctica is one of the regions of particularly interest to the oceanography community due to accelerated basal melting of its ice shelves. Quantitative estimates of the composition of water masses and their sources are essential for understanding the heat and salt budgets and the stability of ice shelves in this region. In this study, the water mass distributions on the Amundsen Sea continental shelf are estimated based on FOAM-GLOSEA5v13, a global oceanographic reanalysis product, using a modified statistical based optimal water mass analysis method. The reanalysis product effectively captures the signal of primary water masses, including the Winter Water (WW) and the Circumpolar Deep Water (CDW) below the Antarctic Summer Surface Water (SSW). The horizontal and vertical distributions and temporal evolution of three water mass endmembers (WW, CDW and DSW) and their mixing ratios are calculated, based on which an intrusion location detection method was established. Using the reanalysis product and intrusion detection method, all potential CDW intrusion sites along the shelf break are identified. Based on the reanalysis product, an intrusion site over the shelf break, away from major deep troughs was captured and confirmed by numerical particle tracking experiments.

Abstract Small cell lung cancer (SCLC) is a highly aggressive subtype of lung cancer characterized by rapid tumor growth and early metastasis. Accurate prediction of prognosis and therapeutic response is crucial for optimizing treatment strategies and improving patient outcomes. In this study, we conducted a deep-learning analysis of Hematoxylin and Eosin (H&E) stained histopathological images using contrastive clustering and identified 50 intricate histomorphological phenotype clusters (HPCs) as pathomic features. We identified two of 50 HPCs with significant prognostic value and then integrated them into a pathomics signature (PathoSig) using the Cox regression model. PathoSig showed significant risk stratification for overall survival and disease-free survival and successfully identified patients who may benefit from postoperative or preoperative chemoradiotherapy. The predictive power of PathoSig was validated in independent multicenter cohorts. Furthermore, PathoSig can provide comprehensive prognostic information beyond the current TNM staging system and molecular subtyping. Overall, our study highlights the significant potential of utilizing histopathology images-based deep learning in improving prognostic predictions and evaluating therapeutic response in SCLC. PathoSig represents an effective tool that aids clinicians in making informed decisions and selecting personalized treatment strategies for SCLC patients.

Fish protein hydrolysates (FPH) were applied in a 8-week feeding experiment with juvenile common carp (Cyprinus carpio) (initial body weight 8.75±0.05 g), to preliminarily investigate their effects on growth performance and feed utilization and potential influence on intestinal antioxidant status, expressions of protein synthesis gene and microflora. Common carp were fed with 0%, 0.5%, 1%, 1.5%, and 2% FPH (named as FPH-0, FPH-0.5, FPH-1, FPH-1.5, and FPH-2) respectively, substituting 0%, 13.33%, 28.33%, 41.66%, and 55.00% of dietary fish meal. Each diet was randomly fed to triplicate groups of 25 fish per aquarium. Feeding experimental results revealed that final body weight, weight gain rate and protein efficiency ratio in the FPH-0.5 treatment were significant higher than those in other treatments. Abstract Migrasomes represent a recently uncovered category of extracellular microvesicles, spanning a diameter range of 500 to 3000 nm. They are emitted by migrating cells and harbour a diverse array of RNAs and proteins. Migrasomes can be readily identified in bodily fluids like serum and urine, rendering them a valuable non‐invasive source for disease diagnosis through liquid biopsy. In this investigation, we introduce a streamlined and effective approach for the capture and quantitative assessment of migrasomes, employing wheat germ agglutinin (WGA)‐coated magnetic beads and flow cytometry (referred to as WBFC). Subsequently, we examined the levels of migrasomes in the urine of kidney disease (KD) patients with podocyte injury and healthy volunteers using WBFC. The outcomes unveiled a substantial increase in urinary podocyte‐derived migrasome concentrations among individuals with KD with podocyte injury compared to the healthy counterparts. Notably, the urinary podocyte‐derived migrasomes were found to express an abundant quantity of phospholipase A2 receptor (PLA2R) proteins. The presence of PLA2R proteins in these migrasomes holds promise for serving as a natural antigen for the quantification of autoantibodies against PLA2R in the serum of patients afflicted by membranous nephropathy. Consequently, our study not only pioneers a novel technique for the isolation and quantification of migrasomes but also underscores the potential of urinary migrasomes as a promising biomarker for the early diagnosis of KD with podocyte injury.

An efficient method was discovered for catalyzing the esterification under air using Novozym 435 to obtain pyridine esters. The following conditions were found to be optimal: 60 mg of Novozyme 435, 5.0 mL of n-hexane, a molar ratio of 2:1 for nicotinic acids (0.4 mmol) to alcohols (0.2 mmol), 0.25 g of molecular sieve 3A, a revolution speed of 150 rpm, a reaction temperature of 50 °C, and reaction time of 48 h. Under nine cycles of Novozym 435, the 80 % yield was consistently obtained. Optimum conditions were used to synthesize 23 pyridine esters, including five novel compounds. Among them, gas chromatography-mass spectrometry-olfactometry (GC-MS-O) showed phenethyl nicotinate (3g), (E)-hex-4-en-1-yl nicotinate (3m), and octyl nicotinate (3n) possessed strong aromas. Thermogravimetric analysis (TG) revealed that the compounds 3g, 3m and 3n exhibited stability at the specified temperature. This finding provides theoretical support for adding pyridine esters fragrance to high-temperature processed food.

Osteoarthritis is a chronic degenerative disease based on the degeneration and loss of articular cartilage. Inflammation and aging play an important role in the destruction of the extracellular matrix, in which microRNA (miRNA) is a key point, such as miRNA-34a-5p. Upregulation of miRNA-34a-5p was previously reported in a rat OA model, and its inhibition significantly suppressed interleukin (IL)-1β-induced apoptosis in rat chondrocytes. However, Oxidative stress caused by reactive oxygen species (ROS) can exacerbate the progression of miRNA regulated OA by mediating inflammatory processes. Thus, oxidative stress effects induced via tert-butyl hydroperoxide (tBHP) in human chondrocytes were assessed in the current research by evaluating mitochondrial ROS production, mitochondrial cyclooxygenase (COX) activity, and cell apoptosis. We also analyzed the activities of antioxidant enzymes including glutathione peroxidase (GSH-Px), catalase (CAT), and superoxide dismutase (SOD). Additionally, inflammatory factors, such as tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-6, IL-8, and IL-24, which contribute to OA development, were detected by enzyme-linked immunosorbent assay (ELISA). The results of this study indicated that miR-34a-5p/silent information regulator 1 (SIRT1)/p53 axis was involved in the ROS-induced injury of human chondrocytes. Moreover, dual-luciferase assay revealed that SIRT1 expression was directly regulated by miR-34a-5p, indicating the presence of a positive feedback loop in the miR-34a-5p/SIRT1/p53 axis that plays an important role in cell survival. However, ROS disrupted the miR-34a-5p/SIRT1/p53 axis, leading to the development of OA, and articular injection of SIRT1 agonist, SRT1720, in a rat model of OA effectively ameliorated OA progression in a dose-dependent manner. Our study confirms that miRNA-34a-5p could participate in oxidative stress responses caused by ROS and further regulate the inflammatory process via the SIRT1/p53 signaling axis, ultimately affecting the onset of OA, thus providing a new treatment strategy for clinical treatment of OA.

To address the alerting issue of energy demand, lithium-ion capacitors (LICs) have been widely studied as promising electrochemical energy storage devices, which can deliver higher energy density than supercapacitors (SCs), and have higher power density with longer cycling life than lithium-ion batteries (LIBs). In this work, the active material lithium nickel cobalt manganese oxide LiNi0.5Co0.2Mn0.3O2 (NCM523) is grown on a cotton textile template and building a 3-dimensional (3D) integrity to improve capacitance and energy density of LICs by enhancing the interfacial ion-exchange process. With the 3D structure, the specific discharge capacitance is increased to 718.67 F g−1 at 0.1 A g−1 from that of non-textile NCM523 (265.97 F g−1), and remains a high capacitance of 254.48 Fg−1 at 10 A g−1 in the half-cell capacitors. In addition, the energy density can achieve up to 36.17 Wh kg−1 at the power density of 1,200 W kg−1 in the full-cell capacitor. The textile NCM can maintain an energy density of 28.26 Wh kg−1 at the current density of 10 A g−1 and power density of 6,000 W kg−1. Our results present promising applications of electrodes with the 3D porous structure for high energy density LICs.

Background: The management of primary hypothyroidism demands a comprehensive approach that encompasses both the implications of autoimmune thyroid disease and the distinct effects posed by obesity and metabolic irregularities. Despite its clinical importance, the interplay between obesity and hypothyroidism, especially in the context of metabolic perspectives, is insufficiently explored in existing research. This study endeavors to classify hypothyroidism by considering the presence of autoimmune thyroid disease and to examine its correlation with various metabolic obesity phenotypes.Method: This research was conducted by analyzing data from 1,170 individuals enrolled in the Thyroid Disease Database of Shandong Provincial Hospital. We assessed four distinct metabolic health statuses among the participants: Metabolically Healthy No Obese Metabolically Healthy Obese Metabolically Unhealthy No Obese and Metabolically Unhealthy Obese Utilizing logistic regression, we investigated the association between various metabolic obesity phenotypes and hypothyroidism.Results: The study revealed a significant correlation between the Metabolically Unhealthy Obese (MUO) phenotype and hypothyroidism, particularly among women who do not have thyroid autoimmunity. Notably, the Metabolically Unhealthy No Obese (MUNO) phenotype showed a significant association with hypothyroidism in individuals with thyroid autoimmunity, with a pronounced prevalence in women. Furthermore, elevated levels of triglycerides and blood glucose were found to be significantly associated with hypothyroidism in men with thyroid autoimmunity and in women without thyroid autoimmunity.Conclusion: Effective treatment of hypothyroidism requires a thorough understanding of the process of thyroid autoimmune development. In patients without concurrent thyroid autoimmunity, there is a notable correlation between obesity and metabolic issues with reduced thyroid function. Conversely, for patients with thyroid autoimmunity, a focused approach on managing metabolic abnormalities, especially triglyceride levels, is crucial.

Abstract Ultrasmall metal nanoclusters (NCs) exhibit a quantized conduction band, hence, a distinct HOMO‐LUMO gap (Eg). Such a quantized electronic structure gives rise to multiple discrete peaks in the optical absorption spectrum of the NCs. As the size grows to 130 gold atoms (Au130 protected by ligands), electrical charging and optical behaviors seem to show certain metal‐like features (hence, transition‐sizes). To probe such behaviors, especially the potential ligand effect, we have devised the synthesis of Au130 NCs protected by phenylethanethiolate and naphthalenethiolate, respectively, with the former having a nonconjugated separation between the aromatic molecular group and the metal core while the latter being in direct bonding. A careful comparison of these two Au130 nanoclusters with the earlier reported analogues is carried out, including the structurally characterized Au130(pMBT)50 and the aqueous counterpart. While all of these nanoclusters possess the same 80 free electron counts in the core, some notable differences in electrochemical and optical properties are found, which are attributed to the ligand effects. The obtained insights may stimulate further interest in the transition‐sized nanoclusters and also promote their applications in optics, energy conversion, and biomedicine.

ABSTRACTAzole drugs are the main therapeutic drugs for invasive fungal infections. However, azole-resistant strains appear repeatedly in the environment, posing a major threat to human health. Several reports have shown that mitochondria are associated with the virulence of pathogenic fungi. However, there are few studies on the mechanisms of mitochondria-mediated azoles resistance. Here, we first performed mitochondrial proteomic analysis on multiple Candida species (Candida albicans, Nakaseomyces glabrata, Pichia kudriavzevii, and Candida auris) and analyzed the differentially expressed mitochondrial proteins (DEMPs) between azole-sensitive and azole-resistant Candida species. Subsequently, we performed Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis, gene ontology analysis, and protein-protein interaction network analysis of DEMPs. Our results showed that a total of 417, 165, and 25 DEMPs were identified in resistant C. albicans, N. glabrata, and C. auris, respectively. These DEMPs were enriched in ribosomal biogenesis at cytosol and mitochondria, tricarboxylic acid cycle, glycolysis, transporters, ergosterol, and cell wall mannan biosynthesis. The high activations of these cellular activities, found in C. albicans and C. auris (at low scale), were mostly opposite to those observed in two fermenter species—N. glabrata and P. kudriavzevii. Several transcription factors including Rtg3 were highly produced in resistant C. albicans that experienced a complex I activation of mitochondrial electron transport chain (ETC). The reduction of mitochondrial-related activities and complex IV/V of ETC in N. glabrata and P. kudriavzevii was companying with the reduced proteins of Tor1, Hog1, and Snf1/Snf4.IMPORTANCECandida spp. are common organisms that cause a variety of invasive diseases. However, Candida spp. are resistant to azoles, which hinders antifungal therapy. Exploring the drug-resistance mechanism of pathogenic Candida spp. will help improve the prevention and control strategy and discover new targets. Mitochondria, as an important organelle in eukaryotic cells, are closely related to a variety of cellular activities. However, the role of mitochondrial proteins in mediating azole resistance in Candida spp. has not been elucidated. Here, we analyzed the mitochondrial proteins and signaling pathways that mediate azole resistance in Candida spp. to provide ideas and references for solving the problem of azole resistance. Our work may offer new insights into the connection between mitochondria and azoles resistance in pathogenic fungi and highlight the potential clinical value of mitochondrial proteins in the treatment of invasive fungal infections.

Based on the Michaelis-Menten reaction model with catalytic effects, a more comprehensive one-dimensional stochastic Langevin equation with immune surveillance for a tumor cell growth system is obtained by considering the fluctuations in growth rate and mortality rate. To explore the impact of environmental fluctuations on the growth of tumor cells, the analytical solution of the steady-state probability distribution function of the system is derived using the Liouville equation and Novikov theory, and the influence of noise intensity and correlation intensity on the steady-state probability distributional function are discussed. The results show that the three extreme values of the steady-state probability distribution function exhibit a structure of two peaks and one valley. Variations of the noise intensity, cross-correlation intensity and correlation time can modulate the probability distribution of the number of tumor cells, which provides theoretical guidance for determining treatment plans in clinical treatment. Furthermore, the increase of noise intensity will inhibit the growth of tumor cells when the number of tumor cells is relatively small, while the increase in noise intensity will further promote the growth of tumor cells when the number of tumor cells is relatively large. The color cross-correlated strength and cross-correlated time between noise also have a certain impact on tumor cell proliferation. The results help people understand the growth kinetics of tumor cells, which can a provide theoretical basis for clinical research on tumor cell growth.

Abstract Uveal melanoma (UM) is the most frequent primary intraocular malignancy with high metastatic potential and poor prognosis. Macrophages represent one of the most abundant infiltrating immune cells with diverse functions in cancers. However, the cellular heterogeneity and functional diversity of macrophages in UM remain largely unexplored. In this study, we analyzed 63,264 single-cell transcriptomes from 11 UM patients and identified four transcriptionally distinct macrophage subsets (termed MΦ-C1 to MΦ-C4). Among them, we found that MΦ-C4 exhibited relatively low expression of both M1 and M2 signature genes, loss of inflammatory pathways and antigen presentation, instead demonstrating enhanced signaling for proliferation, mitochondrial functions and metabolism. We quantified the infiltration abundance of MΦ-C4 from single-cell and bulk transcriptomes across five cohorts and found that increased MΦ-C4 infiltration was relevant to aggressive behaviors and may serve as an independent prognostic indicator for poor outcomes. We propose a novel subtyping scheme based on macrophages by integrating the transcriptional signatures of MΦ-C4 and machine learning to stratify patients into MΦ-C4-enriched or MΦ-C4-depleted subtypes. These two subtypes showed significantly different clinical outcomes and were validated through bulk RNA sequencing and immunofluorescence assays in both public multicenter cohorts and our in-house cohort. Following further translational investigation, our findings highlight a potential therapeutic strategy of targeting macrophage subsets to control metastatic disease and consistently improve the outcome of patients with UM.

Abstract Objectives The public health system faces major challenges due to the double burden of diabetes mellitus (DM) and tuberculosis (TB) in China. We aimed to investigate the prevalence and impact of diabetes on patients with TB. Methods Stratified cluster sampling was used to select 13 counties as study sites in the Zhejiang province. Patients who visited designated TB hospitals in these areas participated in this study between 1 January 2017 and 28 February 2019. Multiple logistic regression models were performed to investigate the association between DM and bacteriological and imaging results. A decision tree was used to predict the bacteriology and imaging results under the influence of DM. Results Of 5920 patients with newly diagnosed pulmonary tuberculosis, 643 (12.16%) had DM. Patients with pulmonary TB and DM were more likely to have pulmonary cavities (adjusted odds ratio [aOR], 2.81; 95% confidence intervals [95% CI]: 2.35–3.37) and higher rates of positive bacteriological tests (aOR, 2.32; 95% CI:1.87–2.87). Decision‐tree analysis showed similar results. Conclusions Concurrence of DM and pulmonary TB makes patients more likely to have positive bacteriological results and pulmonary cavities. Therefore, appropriate measures are necessary to promptly identify and manage patients with TB and DM.

Introduction: CircRNAs are closely related to many human diseases; however, their role in acne remains unclear. This study aimed to determine the role of hsa_circ_0102678 in regulating inflammation of acne. Methods: First, microarray analysis was performed to study the expression of circRNAs in acne. Subsequently, RNase R digestion assay and fluorescence in situ hybridization assay were utilized to confirm the characteristics of hsa_circ_0102678. Finally, qRT-PCR, Western blotting analysis, immunoprecipitation, luciferase reporter assay, circRNA probe pull-down assay, biotin-labeled miRNA pull-down assay, RNA immunoprecipitation assay, and m6A dot blot assay were utilized to reveal the functional roles of hsa_circ_0102678 on inflammation induced by C. acnes biofilm in human primary keratinocytes. Results: Our investigations showed that the expression of hsa_circ_0102678 was significantly decreased in acne tissues, and hsa_circ_0102678 was a type of circRNAs, which was mainly localized in the cytoplasm of primary human keratinocytes. Moreover, hsa_circ_0102678 remarkably affected the expression of IL-8, IL-6, and TNF-α, which induced by C. acnes biofilm. Importantly, mechanistic studies indicated that the YTHDC1 could bind directly to hsa_circ_0102678 and promote the export of N6-methyladenosine-modified hsa_circ_0102678 to the cytoplasm. Besides, hsa_circ_0102678 could bind to miR-146a and sponge miR-146a to promote the expression of IRAK1 and TRAF6. Conclusion: Our findings revealed a previously unknown process by which hsa_circ_0102678 promoted keratinocyte inflammation induced by C. acnes biofilm via regulating miR-146a/TRAF6 and IRAK1 axis.

Abstract Background The utilization of hospital delivery and antenatal care (ANC) is essential for improving maternal and newborn outcomes. However, social and cultural barriers in underdeveloped rural areas hindered maternal care utilization. This study aims to design and evaluate the effectiveness of a culturally adapted digital-platform intervention to promote maternal care utilization among women in ethnic minority communities in China. Methods From January 1st, 2020, to December 31st, 2021, all pregnant women in Mianshan town, Liangshan Autonomous Prefecture, were invited to participate in the intervention. The multifaceted intervention included participatory and cultural-tailored health education on a popular social media platform, transportation subsidies, and capacity building and economic incentives for healthcare providers. The effectiveness of the intervention was evaluated by comparing two groups: mothers who gave live birth before the intervention (January 1st to December 31st, 2019) and mothers whose entire pregnancy period was covered by the intervention. The primary outcomes were the rate of hospital delivery and ANC utilization. Data on pregnant women were retrospectively collected through telephone surveys and the maternal and newborn’s health monitoring system. The present research examined the impact of L-glutamic acid (Glu) supplementation on the growth performance, muscle composition, gene expression correlated with muscle growth, and intestinal health of largemouth bass. There were 525 fish in total, which were distributed randomly into five groups. Each group had three replicates, and each replicate consisted of 35 fish. Groups with control and experimental diets were assigned glutamic acid amounts of 0.2%, 0.4%, 0.6%, and 0.8%. The findings demonstrated that glutamic acid supplementation enhanced growth performance, feed intake (FI), and condition factor (CF), with the best value being attained at 0.4% Glu. The mean muscle fiber area was increased and the muscle fiber density was decreased in the 0.6% Glu group. The levels of total amino acids and specific amino acids, such as glutamic acid, aspartic acid, leucine, valine, alanine, and glycine, were shown to be higher in the 0.6% Glu group. In the 0.6% Glu group, the mRNA expression levels of atrogin-1, murf-1, foxo3a, and 4e-bp1 were decreased compared to the control group. Conversely, the mRNA expression levels of myf5, myog, myod, s6k1, tor, akt, and pi3k were increased in the 0.6% Glu group compared to the control group. The 0.4% Glu group had higher intestinal amylase, lipase, and protease activities and greater villus height, villus width, and muscle thickness. In summary, Glu can support largemouth bass growth, muscular development, intestinal digestion, and absorption.

Ni-rich Li(NixCoyMnz)O2 (x ≥ 0.8)-layered oxide materials are highly promising as cathode materials for high-energy-density lithium-ion batteries in electric and hybrid vehicles. However, their tendency to undergo side reactions with electrolytes and their structural instability during cyclic lithiation/delithiation impairs their electrochemical cycling performance, posing challenges for large-scale applications. This paper explores the application of an Al2O3 coating using an atomic layer deposition (ALD) system on Ni-enriched Li(Ni0.8Co0.1Mn0.1)O2 (NCM811) cathode material. Characterization techniques, including X-ray diffraction, scanning electron microscopy, and transmission electron microscopy, were used to assess the impact of alumina coating on the morphology and crystal structure of NCM811. The results confirmed that an ultrathin Al2O3 coating was achieved without altering the microstructure and lattice structure of NCM811. The alumina-coated NCM811 exhibited improved cycling stability and capacity retention in the voltage range of 2.8–4.5 V at a 1 C rate. Specifically, the capacity retention of the modified NCM811 was 5%, 9.11%, and 11.28% higher than the pristine material at operating voltages of 4.3, 4.4, and 4.5 V, respectively. This enhanced performance is attributed to reduced electrode–electrolyte interaction, leading to fewer side reactions and improved structural stability. Thus, NCM811@Al2O3 with this coating process emerges as a highly attractive candidate for high-capacity lithium-ion battery cathode materials.

Traffic sign detection plays a pivotal role in autonomous driving systems. The intricacy of the detection model necessitates high-performance hardware. Real-world traffic environments exhibit considerable variability and diversity, posing challenges for effective feature extraction by the model. Therefore, it is imperative to develop a detection model that is not only highly accurate but also lightweight. In this paper, we proposed YOLO-ADual, a novel lightweight model. Our method leverages the C3Dual and Adown lightweight modules as replacements for CPS and CBL modules in YOLOv5. The Adown module effectively mitigates feature loss during downsampling while reducing computational costs. Meanwhile, C3Dual optimizes the processing power for kernel feature extraction, enhancing computation efficiency while preserving network depth and feature extraction capability. Furthermore, the inclusion of the CBAM module enables the network to focus on salient information within the image, thus augmenting its feature representation capability. Our proposed algorithm achieves a mAP@0.5 of 70.1% while significantly reducing the number of parameters and computational requirements to 51.83% and 64.73% of the original model, respectively. Compared to various lightweight models, our approach demonstrates competitive performance in terms of both computational efficiency and accuracy.
