4. Plasma Total Antioxidant Capacity and Activities of Blood Glutathione Peroxidase and Superoxide Dismutase Determined in Healthy Dogs by Using Commercially Available Kits

Data on the values of selected blood antioxidant parameters, i.e. total antioxidant capacity, glutathione peroxidase, and superoxide dismutase in healthy dogs, are lacking. There are no published accepted standard reference methods for their determination. The aim of this study was to determine the values of plasma total antioxidant capacity and the activities of whole blood glutathione peroxidase and erythrocyte superoxide dismutase in 30 healthy client-owned dogs (19 females, 11 males). The effect of age and sex on the measured antioxidant parameters was also investigated. Antioxidant parameters were determined with an automated biochemical analyser, using the commercially available Randox kits. No significant difference in age, weight, and antioxidant parameters was determined between females and males. A significant positive effect of age (p = 0.002, r2 = 0.284) on superoxide dismutase activity was confirmed. There was no effect of sex on any of the antioxidant parameters measured. However, we observed a tendency of the effect of sex (p = 0.063, r2 = 0.118), as well as age (p = 0.073, r2 = 0.111), on the activity of glutathione peroxidase. Our results are in part comparable with the results of other studies in which the same types of methods and samples were used to determine antioxidant parameters. In conclusion, the sex and age of dogs should be taken into consideration when planning a study on antioxidant status parameters.

Abstract Immunotherapeutic drugs are promising medicines for cancer treatment. A potential candidate for immunotherapy is interleukin-12 (IL-12), a cytokine well known for its ability to mediate antitumor activity. We developed a plasmid encoding human IL-12 devoid of an antibiotic resistance gene (phIL12). For the approval of phase I clinical trials in basal cell carcinoma (BCC), the regulatory agency requires non-clinical in vivo testing of the pharmacodynamic, pharmacokinetic and toxicological properties of the plasmid. As human IL-12 is not biologically active in mice, a mouse ortholog of the plasmid phIL12 (pmIL12) was evaluated. The evaluation demonstrated the antitumor effectiveness of the protein accompanied by immune cell infiltration. The plasmid was distributed throughout the body, and the amount of plasmid diminished over time in all organs except the skin around the tumor. The therapy did not cause any detectable systemic toxicity. The results of the non-clinical evaluation demonstrated the safety and efficacy of the pmIL12/phIL12 GET, and on the basis of these results, approval was obtained for the initiation of a phase I clinical study in BCC.

Abstract Background The aim of this double-blind, placebo-controlled study was to investigate the effect of vitamin E supplementation as an addition to a commercial renal diet on survival time of cats with different stages of chronic kidney disease (CKD). In addition, we were interested whether vitamin E supplementation affects selected oxidative stress and clinical parameters. Thirty-four cats with CKD and 38 healthy cats were included in the study. Cats with CKD were classified according to the IRIS Guidelines; seven in IRIS stage 1, 15 in IRIS stage 2, five in IRIS stage 3 and seven in IRIS stage 4. Cats with CKD were treated according to IRIS Guidelines. Cats with CKD were randomly assigned to receive vitamin E (100 IU/cat/day) or placebo (mineral oil) for 24 weeks in addition to standard therapy. Plasma malondialdehyde (MDA) and protein carbonyl (PC) concentrations, DNA damage of peripheral lymphocytes and plasma vitamin E concentrations were measured at baseline and four, eight, 16 and 24 weeks thereafter. Routine laboratory analyses and assessment of clinical signs were performed at each visit. Results Vitamin E supplementation had no effect on the survival time and did not reduce the severity of clinical signs. Before vitamin E supplementation, no significant differences in vitamin E, MDA and PC concentrations were found between healthy and CKD cats. However, plasma MDA concentration was statistically significantly higher (p = 0.043) in cats with early CKD (IRIS stages 1 and 2) than in cats with advanced CKD (IRIS stages 3 and 4). Additionally, DNA damage was statistically significantly higher in healthy cats (p ≤ 0.001) than in CKD cats. Plasma vitamin E concentrations increased statistically significantly in the vitamin E group compared to the placebo group four (p = 0.013) and eight (p = 0.017) weeks after the start of vitamin E supplementation. During the study and after 24 weeks of vitamin E supplementation, plasma MDA and PC concentrations and DNA damage remained similar to pre-supplementation levels in both the placebo and vitamin E groups. Conclusions Vitamin E supplementation as an addition to standard therapy does not prolong survival in feline CKD.

Chronic kidney disease (CKD) is characterized by chronic inflammation, which mediates the progressive replacement of functional nephrons by fibrotic tissue. Hemogram-derived inflammatory markers are known to serve as markers of pathological conditions; however, their diagnostic value in feline CKD is still unknown. The aim of this retrospective study was to investigate selected hemogram-derived inflammatory markers (neutrophil-to-lymphocyte ratio (NLR), monocyte-to-lymphocyte ratio (MLR), platelet-to-lymphocyte ratio (PLR) and the systemic immune-inflammatory index (SII)) in cats at different clinical stages of CKD. Eighty-eight client-owned cats with CKD and thirty-two healthy control cats were included. Cats with CKD were divided into two groups: early CKD (IRIS stage 1 and 2; 62 cats) and progressed CKD (IRIS stage 3 and 4; 26 cats). The values of inflammatory markers were compared between the two CKD groups and the control group. All investigated hemogram-derived inflammatory markers were significantly (p < 0.05) greater in cats with advanced CKD than in those in the other two groups. Additionally, we demonstrated a statistically significant weak to moderate correlation between serum urea, creatinine, selected hematologic and urinary parameters, and the investigated inflammatory markers in cats with CKD. Chronic inflammation can be easily and inexpensively assessed with hemogram-derived markers.

11. Thrombocytosis in brachycephalic dogs with brachycephalic obstructive airway syndrome

Brachycephalic dogs are prone to a conformation-related respiratory disorder known as brachycephalic obstructive airway syndrome (BOAS). Due to its systemic consequences, BOAS should be considered a systemic disease. The aim of the present study was to investigate platelet count (PLT), mean platelet volume (MPV), and plateletcrit (PCT) in patients with various grades of BOAS and non-brachycephalic dogs. The latter served as a control group. We included 106 patients with BOAS and 41 non-brachycephalic dogs. According to the severity of the disease, BOAS patients were classified into grade 1 (17 dogs), grade 2 (42 dogs), and grade 3 (47 dogs). Thrombocytosis was found in 46% (46/106) of all BOAS patients. High platelet mass was found in 62% (66/106) of all BOAS patients. We found significantly (P < 0.05) higher PLT, MPV, and PCT in BOAS patients of all grades compared to non-brachycephalic dogs. Our results suggest that platelets may play an important role in BOAS. Our results suggest that platelets may play an important role in BOAS. However, further studies are needed to clarify the role of PLT and platelet indices in BOAS and their relationship with inflammation and hypercoagulability.

12. Echocardiographic parameters in French Bulldogs, Pugs and Boston Terriers with brachycephalic obstructive airways syndrome

Abstract Background In this prospective study, we hypothesized that dogs with signs of brachycephalic obstructive airway syndrome (BOAS) would show differences in left and right heart echocardiographic parameters compared with brachycephalic dogs without signs of BOAS and non-brachycephalic dogs. Results We included 57 brachycephalic (30 French Bulldogs 15 Pugs, and 12 Boston Terriers) and 10 non-brachycephalic control dogs. Brachycephalic dogs had significantly higher ratios of the left atrium to aorta and mitral early wave velocity to early diastolic septal annular velocity; smaller left ventricular (LV) diastolic internal diameter index; and lower tricuspid annular plane systolic excursion index, late diastolic annular velocity of the LV free wall, peak systolic septal annular velocity, late diastolic septal annular velocitiy, and right ventricular global strain than non-brachycephalic dogs. French Bulldogs with signs of BOAS had a smaller diameter of the left atrium index and right ventricular systolic area index; higher caudal vena cava at inspiration index; and lower caudal vena cava collapsibility index, late diastolic annular velocity of the LV free wall, and peak systolic annular velocity of the interventricular septum than non-brachycephalic dogs. Conclusions The differences in echocardiographic parameters between brachycephalic and non-brachycephalic dogs, brachycephalic dogs with signs of BOAS and non-brachycephalic dogs, and brachycephalic dogs with and without signs of BOAS indicate higher right heart diastolic pressures affecting right heart function in brachycephalic dogs and those with signs of BOAS. Most changes in cardiac morphology and function can be attributed to anatomic changes in brachycephalic dogs alone and not to the symptomatic stage.

14. Haematological profiles of pigs of different age in relation to the presence or absence of porcine reproductive and respiratory virus, porcine circovirus type 2 and hepatitis E virus

This study was aimed to assessing haematological parameters in pigs of different age categories from six farms differing in the presence or absence of selected pathogens. The following categories were included: 5 age groups of growers, fatteners and breeding sows. Individual blood samples for determining complete blood count and white blood cell differential count were taken. Group samples of oral fluid and faeces were collected from each farm and tested for detection of Porcine Reproductive and Respiratory Virus (PRRSV), Porcine Circovirus Type 2 (PCV2), and Hepatitis E Virus (HEV) using PCR, RT-PCR, and qRT-PCR protocols. On farms free of PRRSV, PCV2 and HEV, the age of pigs had significant effect on: white blood cell count (WBC), haemoglobin concentration (Hb), mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin concentration, platelet count (PLT), percentage of neutrophils, lymphocytes and eosinophils and absolute numbers of neutrophils, lymphocytes, monocytes, basophils and ‘large unstained cells’ (LUCs). On positive farms all observed blood parameters except the percentage of LUCs were significantly affected by the age. The percentages of lymphocytes, mean corpuscular volume (MCV) and haematocrit were significantly lowered by PRRSV, while WBC, PLT, percentage and absolute numbers of neutrophils, basophils and LUCs were increased. Nine-week-old pigs with PCV2 had significantly lower red blood cell count, PLT, percentage and absolute numbers of monocytes and LUCs, and significantly higher MCV, MCH, percentage of neutrophils and percentage and absolute number of eosinophils than 9-week-old pigs with PCV2 and HEV. Age-related changes in haematological parameters occurred in PCR negative and positive farms.HIGHLIGHTS Age-related changes in haematological parameters were present in PCR negative farms and in PCR positive farms in relation to selected pathogens. Values for several haematological parameters in 9-week-old pigs with a monoinfection differed significantly from those in pigs of the same age with two viral infections. To interpret results of haematological analyses correctly, it is important to consider the haematological analyser used, the pig age and, health history, and clinical data.

15. The effect of the surgical treatment of brachycephalic obstructive airway syndrome on the thermoregulatory response to exercise in French bulldogs: a pilot study

IntroductionDue to altered anatomy of the upper respiratory tract, brachycephalic dogs exhibit increased airway resistance and reduced surface area for evaporative heat loss, predisposing them to respiratory and thermoregulatory problems, a syndrome referred to as Brachycephalic Obstructive Airway Syndrome (BOAS). Compared to non-brachycephalic dogs, brachycephalic breeds are more susceptible to heat-related injuries even at low ambient temperatures and relatively low physical activity levels. Surgical treatment alleviates clinical signs, potentially improving dogs’ thermoregulatory ability with BOAS. Our study aimed to investigate the thermoregulatory response in French bulldogs before and after the surgical correction of BOAS, respectively.MethodsThirteen dogs were exposed to dynamic exercise on a treadmill and the dynamics of their rectal temperature (RT) and heart rate (HR) was measured. The experiment was performed in two independent sessions, before and after the surgical treatment. The test consisted of two consecutive 5-min walks at a speed of 2.5 km/h, first at an inclination of 0% and the second at an inclination of 5%, and a 30-min recovery period. Rectal temperature and HR were measured before the start of the test (t0), at the end of the first (t1) and the second part (t2), and 15 min (t3) and 30 min (t4) in the recovery.ResultsA significant increase in RT and HR was observed during exercise and recovery compared to the resting values, implying that the exercise intensity was sufficient to challenge the thermoregulatory response. The increase of RT was significantly lower during both parts (t1: p = 0.004; t2: p < 0.001) of exercise after the surgical treatment than before the treatment. Although a trend of lower RT after recovery was observed compared to before the surgery, it did not reach statistical significance. Similarly to RT, the HR was significantly lower during exercise after the first (p = 0.020) and the second part (p = 0.011) of exercise after the surgery compared to before the surgery but did not reach significance in the recovery.ConclusionSurgical treatment of BOAS can improve thermoregulation during exercise in French bulldogs with BOAS.

Myxomatous mitral valve degeneration (MMVD) is the most common naturally occurring heart disease in dogs. There is a lack of data on antioxidant status and oxidative damage in dogs with MMVD stage B1 according to the American College of Veterinary Internal Medicine (ACVIM B1). The aim of this study was to investigate antioxidant status (plasma vitamin E, lipid-standardized vitamin E (LS-VitE), antioxidant capacity of lipid-(ACL) and water-soluble antioxidants, whole blood glutathione peroxidase and erythrocyte superoxide dismutase), and lipid peroxidation [malondialdehyde (MDA)] in dogs with MMVD ACVIM B1. Serum cholesterol and triglyceride concentrations were measured to calculate LS-VitE. Fourteen dogs with MMVD ACVIM B1 and 12 control dogs were included in the study. Dogs with MMVD had significantly higher vitamin E, ACL, MDA, and cholesterol concentrations and significantly higher LS-VitE values than control dogs. No significant correlations between MDA and antioxidant parameters were determined in either group. In conclusion, oxidative damage to lipids is already present and the antioxidant status is altered but not depleted in dogs with MMVD ACVIM B1. The antioxidant response to increased oxidative damage consists mainly of the activation of fat-soluble antioxidants. Further research is needed to evaluate the efficacy and targets of early antioxidant supplementation to prevent or ameliorate oxidative stress and mitigate disease progression in dogs with early-stage MMVD.

Extracellular nanoparticles (EPs) are a subject of increasing interest for their biological role as mediators in cell–cell communication; however, their harvesting and assessment from bodily fluids are challenging, as processing can significantly affect samples. With the aim of minimizing processing artifacts, we assessed the number density (n) and hydrodynamic diameter (D
h ) of EPs directly in diluted plasma and blood using the following recently developed technique: interferometric light microscopy (ILM). We analyzed 613 blood and plasma samples from human patients with inflammatory bowel disease (IBD), collected in trisodium citrate and ethylenediaminetetraacetic acid (EDTA) anticoagulants, and 163 blood and plasma samples from canine patients with brachycephalic obstructive airway syndrome (BOAS). We found a highly statistically significant correlation between n in the plasma and n in the blood only in the human (i.e., but not canine) blood samples, between the samples with trisodium citrate and EDTA, and between the respective Dh for both species (all p < 10−3 ). In the human plasma, the averageh> was 139 ± 31 nm; in the human blood, h> was 158 ± 11 nm; in the canine plasma, h> was 155 ± 32 nm; and in the canine blood, h> was 171 ± 33 nm. The differences within species were statistically significant (p < 10 −2 ), with sufficient statistical power (P > 0.8). For, we found no statistically significant differences between the human plasma and blood samples or between the samples with trisodium citrate and EDTA. Electrochemotherapy (ECT) in combination with the gene electrotransfer of interleukin 12 (IL-12 GET) has been successfully used in veterinary medicine for the treatment of mast cell tumours (MCT), but the biomarkers that could predict response to this treatment have not yet been investigated. The aim of this study was to determine the plasma nucleosome and serum ferritin concentrations, as well as the lactate dehydrogenase (LDH) activity, in the serum of treated patients before and one and six months after treatment to evaluate their utility as potential biomarkers that could predict response to the combined treatment. The study was conducted in 48 patients with a total of 86 MCTs that we treated with the combined treatment. The blood samples used for analysing the potential predictive biomarkers were taken before treatment and one and six months after treatment, when the response to treatment was also assessed. The Nu. Q® Vet Cancer Test, the Canine Ferritin ELISA Kit, and the RX Daytona+ automated biochemical analyser were used to analyse the blood samples. The results showed that the plasma nucleosome concentration (before treatment (BT): 32.84 ng/mL (median); one month after treatment (1 M AT): 58.89 ng/mL (median); p = 0.010) and serum LDH activity (BT: 59.75 U/L (median); 1 M AT: 107.5 U/L (median); p = 0.012) increased significantly one month after treatment and that the increase correlated significantly with the presence of a more pronounced local reaction (necrosis, swelling, etc.) at that time point for both markers (nucleosome: BT (necrosis): 21.61 ng/mL (median); 1 M AT (necrosis): 69.92 ng/mL (median), p = 0.030; LDH: BT (necrosis): 54.75 U/L (median); 1 M AT (necrosis): 100.3 U/L (median), p = 0.048). Therefore, both the plasma nucleosome concentration and serum LDH activity could serve as early indicators of the effect of the treatment. In this context, the serum ferritin concentration showed no significant predictive potential for treatment response (p > 0.999 for all comparisons). In conclusion, this study provides some new and important observations on the use of predictive biomarkers in veterinary oncology. Furthermore, it emphasises the need for the continued identification and validation of potential predictive biomarkers in dogs with MCT and other malignancies undergoing ECT treatment in combination with IL-12 GET to ultimately improve treatment outcomes.

The aim of the study was to investigate the effect of a therapeutic renal diet on selected clinical, biochemical, and urinary parameters and on selected parameters of oxidative stress in cats with early stages of chronic kidney disease (CKD). A prospective study of a 3-month duration was conducted to evaluate the effect of renal diet on selected clinical and laboratory parameters in client-owned cats with early stages of CKD. Of a total of 29 enrolled client-owned cats, nineteen (19) cats completed the study, ten receiving renal diet and nine receiving a diet of the owner’s choice. A clinical examination was performed, and blood and urine samples were collected on the day of presentation and at regular check-ups after 3-4, 7-8, and 10-12 weeks. Serum creatinine and symmetric dimethylarginine (SDMA) concentrations and selected parameters of oxidative stress (plasma glutathione peroxidase (GPX) activity and plasma malondialdehyde (MDA) and serum selenium concentrations), were measured and electrophoresis of urinary proteins was performed. At inclusion, a significant positive correlation (p < 0.001) was found between serum selenium concentration and plasma GPX activity (Pearson correlation coefficient 0.83 (95% CI: [0.65 - 0.92]) and a significant negative correlation (p < 0.001) between serum SDMA and urine specific gravity (Pearson correlation coefficient -0.70 (95% CI: [-0.87 - (-0.38)]). At the end of the 3-month feeding trial no significant difference was found in SDMA and creatinine concentrations. UČINEK MONOTERAPEVTSKE LEDVIČNE DIETE NA KLINIČNE, BIOKEMIJSKE, URINSKE IN SERUMSKE PARAMETRE OKSIDATIVNEGA STRESA PRI MAČKAH S CDK STOPNJE 1 IN 2 Izvleček: Namen študije je bil raziskati učinek terapevtske ledvične diete na izbrane klinične, biokemijske in urinske parametre ter parametre oksidativnega stresa pri mačkah v začetnih stopnjah kronične ledvične bolezni (KLB). Raziskava je bila zasnovana kot prospektivna, tri mesece trajajoča klinična študija, v katero je bilo vključenih 29 lastniških mačk. Devetnajst mačk je zaključilo študijo, od teh jih je deset prejemalo ledvično dieto, devet pa vzdrževalno dieto po izboru lastnika. Pri vseh mačkah smo izvedli klinični pregled in odvzem krvnih ter urinskih vzorcev na dan vključitve v študijo in pri treh kontrolnih pregledih, ki so bili izvedeni 3–4, 7–8 in 10–12 tednov kasneje. Določili smo serumsko koncentracijo kreatinina, simetričnega dimetilarginina (SDMA) in izbrane parametre oksidativnega stresa (aktivnost plazemske glutation peroksidaze (GPX) in plazemska koncentracija malondialdehida (MDA) ter serumska koncentracija selena). Poleg tega smo izvedli elektroforezo urinskih proteinov. Ob vključitvi mačk v raziskavo smo ugotovili značilno pozitivno korelacijo (p < 0,001) med serumsko koncentracijo selena in aktivnostjo plazemske GPX (Pearsonov korelacijski koeficient 0,83, 95 % CI: [0,65–0,92]) ter značilno negativno korealcijo (p < 0,001) med koncentracijo SDMA in specifično težo urina (Pearsonov korelacijski koeficient –0,70 (95 % CI: [–0,87–(–0,38)]). Trimesečno hranjenje s terapevtsko hrano ni privedlo do značilnih sprememb v serumski koncentraciji SDMA in kreatinina pri vključenih mačkah. Optimal heart function depends on perfect synchronization between electrical and mechanical activity. In this pilot study, we aimed to investigate the electromechanical activity of the heart in healthy cats and cats with cardiomyopathy with phonocardiography (PCG) synchronized to an electrocardiography (ECG) pilot device. We included 29 cats (12 healthy cats and 17 cats diagnosed with cardiomyopathy) and performed a clinical examination, PCG synchronized with ECG and echocardiography. We measured the following durations with the pilot PCG device synchronized with ECG: QRS (ventricular depolarization), QT interval (electrical systole), QS1 interval (electromechanical activation time (EMAT)), S1S2 (mechanical systole), QS2 interval (electrical and mechanical systole) and electromechanical window (end of T wave to the beginning of S2). The measured parameters did not differ between healthy cats and cats with cardiomyopathy; however, in cats with cardiomyopathy, EMAT/RR, QS2/RR and S1S2/RR were significantly longer than in healthy cats. This suggests that the hypertrophied myocardium takes longer to generate sufficient pressure to close the mitral valve and that electrical systole, i.e., depolarization and repolarization, and mechanical systoles are longer in cats with cardiomyopathy. The PCG synchronized with the ECG pilot device proved to be a valuable tool for evaluating the electromechanical activity of the feline heart.

General anesthesia increases the production of reactive oxygen species (ROS), which can exacerbate or increase oxidative stress and thus affect the prognosis of surgical procedures. Oxidative stress has been implicated in the development of cardiovascular, dermatologic, oncologic, and other diseases in dogs, as well as ischemia and reperfusion injury. Some anesthetics, such as halogenated anesthetics, have been shown to stimulate the production of ROS, while others, such as propofol, have antioxidant properties. However, the antioxidant effects of these anesthetics may not be sufficient to counteract oxidative damage at the doses used clinically. Nevertheless, the effects of anesthetics should be considered to minimize oxidative damage during anesthesia in dogs to improve the outcome of procedures requiring general anesthesia. This mini-review addresses the current knowledge on oxidative stress during inhalational and intravenous anesthesia in dogs. There is still a lack of information on the management of anesthesia in dogs with respect to oxidative stress. Further research, including comprehensive clinical studies is needed to better understand oxidative injury mechanisms and improve perioperative protocols during anesthesia in dogs.

23. Inflammation and its association with oxidative stress in dogs with heart failure

Abstract Background Inflammation and oxidative stress can contribute to the development and progression of heart failure. This study aimed to investigate the association between inflammatory and oxidative stress markers in dogs with congestive heart failure (CHF). Associations between the disease severity marker N-terminal pro-B-type natriuretic peptide (NT-proBNP) and markers of inflammation and oxidative stress were also determined. Results Thirty-seven dogs with cardiovascular diseases (dilated cardiomyopathy, DCM (16 dogs), myxomatous mitral valve disease, MMVD (21 dogs)) and ten healthy dogs were included in this prospective study. The patients were further divided into groups with (26) and without CHF (11). We found a significantly higher serum concentration of C-reactive protein (P = 0.012), white blood cell (P = 0.001), neutrophil (P = 0.001) and monocyte counts (P = 0.001) in patients with CHF compared to control dogs. The concentration of tumor necrosis factor-alpha (TNF-α) was significantly higher in patients with CHF compared to patients without CHF (P = 0.030). No significant difference was found in most of the measured parameters between MMVD and DCM patients, except for glutathione peroxidase (GPX) and NT-proBNP. In patients with CHF, TNF-α correlated positively with malondialdehyde (P = 0.014, r = 0.474) and negatively with GPX (P = 0.026, r = − 0.453), and interleukin-6 correlated negatively with GPX (P = 0.046, r = − 0.412). NT-proBNP correlated positively with malondialdehyde (P = 0.011, r = 0.493). In patients without CHF none of the inflammatory and oxidative stress markers correlated significantly. Furthermore, in the group of all cardiac patients, GPX activity significantly negatively correlated with NT-proBNP (P = 0.050, r = − 0.339) and several markers of inflammation, including TNF-α (P = 0.010, r = − 0.436), interleukin-6 (P = 0.026, r = − 0.382), white blood cell (P = 0.032, r = − 0.369), neutrophil (P = 0.027, r = − 0.379) and monocyte counts (P = 0.024, r = − 0.386). Conclusion Inflammatory and oxidative stress markers are linked in canine CHF patients, but not in patients without CHF. These results suggest complex cross communication between the two biological pathways in advanced stages of CHF.

Genska terapija postaja čedalje bolj zanimiva tudi v onkologiji. Med aplikacijami je morda najzanimivejša imunostimulacija. Pripravimo lahko plazmidno DNA, ki nosi zapis za različne imunostimulatorne molekule, ki jih vnesemo v celice tumorjev ali normalnih tkiv. Ta tkiva postanejo proizvajalci teh molekul, ki lahko delujejo lokalno ali pa se izločajo tudi sistemsko v krvni obtok. Ker plazmidna DNA ne prehaja celične membrane, so potrebni dostavni sistemi, virusni ali nevirusni. V naših študijah uporabljamo predvsem nevirusni dostavni sistem – elektroporacijo. Interlevkin 12 (IL-12) je eden od zanimivih citokinov, za katerega je znano protitumorsko delovanje s spodbujanjem imunskega odziva in antiangiogenim delovanjem. Namen projekta SmartGene.si je bil pripraviti plazmid z zapisom za interlevkin 12 (plazmid phIL12) in pripraviti vse potrebno za njegovo klinično testiranje za zdravljenje kožnih tumorjev. V konzorciju smo združili moči s partnerji z akademskega in industrijskega področja. Treba je bilo pripraviti plazmid za uporabo v humani onkologiji po zahtevah Evropske agencije za zdravila (EMA). Za prijavo klinične študije na Javno agencijo za zdravila in medicinske pripomočke (JAZMP) smo morali izvesti tudi vse neklinične raziskave o varnosti in učinkovitosti zdravila. Nato je bilo treba razviti postopek priprave zdravila, zagotoviti primerne prostore za pripravo in izvedbo postopka priprave zdravila. V treh letih smo dosegli vse te zastavljene cilje in dobili dovoljenje za izvajanje klinične študije na kožnih tumorjih, ki ga je izdala JAZMP na osnovi pozitivnega mnenja Komisije Republike Slovenije za medicinsko etiko. Zdaj poteka klinična študija faze I preizkušanja plazmida phIL12 na kožnih tumorjih glave in vratu z namenom preveriti varnost in sprejemljivost genskega elektroprenosa plazmida v tumorje. Cilj študije je prav tako določiti primeren odmerek zdravila, ki bi ga v nadaljnji klinični študiji uporabili kot adjuvantno zdravljenje k ablativnim terapijam, kot sta radioterapija ali elektrokemoterapija.

Abstract Background Antioxidants located in both the hydrophilic and lipophilic compartments of plasma act as a defence system against reactive oxygen species (ROS). Excessive production of ROS during anaesthesia affects the antioxidant capacity of plasma and may result in oxidative stress. The aim of this study was to evaluate the antioxidant capacity of lipid- (ACL) and water-soluble (ACW) antioxidants in client-owned dogs diagnosed with periodontal disease and early-stage myxomatous mitral valve degeneration (MMVD) and anaesthetised for a dental procedure with propofol and sevoflurane or with propofol only. Results Dogs with MMVD were anaesthetised with propofol and sevoflurane (MMVD/PS, n = 8) or with propofol only (MMVD/P, n = 10). Dogs with no evidence of MMVD (PS, n = 12) were anaesthetised with propofol and sevoflurane. Blood samples for determination of ACL and ACW were collected before and 5 min, 60 min and 6 h after induction to anaesthesia. In MMVD/PS dogs, ACL was significantly higher at all sampling times when compared to PS dogs. Compared to basal values, only anaesthesia maintained with propofol significantly increased ACL at 60 min in dogs with MMVD. In MMVD/P dogs, ACW increased after induction to anaesthesia and remained elevated up to 6 h after anaesthesia. Compared to basal values, anaesthesia maintained with sevoflurane significantly increased ACW only at 60 min in both dogs with and without MMVD. The only difference between propofol and propofol/sevoflurane anaesthesia in dogs with MMVD was significantly higher ACW at 60 min after induction to anaesthesia in the propofol group. Conclusions Regarding antioxidant capacity, propofol could be a better choice than sevoflurane for anaesthesia of dogs with early-stage MMVD, although further studies are necessary to clarify the advantage of this antioxidant capacity.

Our study aimed to investigate the blood variables: coenzyme Q10, N-terminal pro-B-type natriuretic peptide (NT-proBNP), electrolytes, cholesterol, triglyceride, glutathione peroxidase, white blood cell, neutrophil, lymphocyte and monocyte counts and relative numbers of neutrophils, lymphocytes and monocytes that may be associated with survival of dogs in congestive heart failure (CHF). Twenty-one client-owned dogs in CHF were included in the study. Cox regression analysis showed significant association only between NT-proBNP blood concentration and survival.

Simple Summary: Chronic kidney disease is a progressive disease affecting middle-aged and older cats. Recent research has linked chronic inflammatory processes to disease progression. Hemogram-derived inflammatory markers have been studied in many pathological conditions; however, their diagnostic value in feline chronic kidney disease is unknown. The aim of the present study was to investigate selected hemogram-derived inflammatory markers in cats at different clinical stages of chronic kidney disease. The results showed that the values of all the studied inflammatory markers were greater in cats with advanced chronic kidney disease than in healthy cats and in cats with early disease. These findings indicate that cats with advanced chronic kidney disease are in a state of chronic systemic inflammation, which is detectable by simple hematological testing. However, the prognostic value of hemogram-derived inflammatory markers should be assessed in further studies. Chronic kidney disease (CKD) is characterized by chronic inflammation, which mediates the progressive replacement of functional nephrons by fibrotic tissue. Hemogram-derived inflammatory markers are known to serve as markers of pathological conditions; however, their diagnostic value in feline CKD is still unknown. The aim of this retrospective study was to investigate selected hemogram-derived inflammatory markers (neutrophil-to-lymphocyte ratio (NLR), monocyte-to-lymphocyte ratio (MLR), platelet-to-lymphocyte ratio (PLR) and the systemic immune-inflammatory index (SII)) in cats at different clinical stages of CKD. Eighty-eight client-owned cats with CKD and thirty-two healthy control cats were included. Cats with CKD were divided into two groups: early CKD (IRIS stage 1 and 2; 62 cats) and progressed CKD (IRIS stage 3 and 4; 26 cats). The values of inflammatory markers were compared between the two CKD groups and the control group. All investigated hemogram-derived inflammatory markers were significantly (p < 0.05) greater in cats with advanced CKD than in those in the other two groups. Additionally, we demonstrated a statistically significant weak to moderate correlation between serum urea, creatinine, selected hematologic and urinary parameters, and the investigated inflammatory markers in cats with CKD. Scarce data exist on the effects of coenzyme Q10 (CoQ10) supplementation in dogs with myxomatous mitral valve disease (MMVD). The purpose of this study was to investigate the effect of CoQ10 supplementation on oxidative stress markers (glutathione peroxidase, F2-isoprostanes), markers of inflammation (tumor necrosis factor-α, TNF soluble receptor II, leucocytes, and their subtypes), lymphocyte subpopulations (T helper and cytotoxic T lymphocytes, including activated T lymphocytes, and B lymphocytes), and echocardiographic and clinical parameters in dogs with MMVD. In this randomized, controlled, double-blind, longitudinal study, 43 MMVD dogs in stages ACVIM (American College of Veterinary Internal Medicine classification) B2 and ACVIM C and D (congestive heart failure (CHF)) received water-soluble coenzyme Q10 (100 mg twice daily) or placebo for 3 months, and 12 non-supplemented healthy dogs served as controls. All parameters were measured before and after supplementation in MMVD dogs and once in healthy dogs. CoQ10 supplementation had a positive impact on neutrophil percentage, lymphocyte percentage, and lymphocyte concentration in our cohort of dogs with CHF (ACVIM C and D). Conclusion: CoQ10 as an oral supplement may have benefits in terms of decreasing inflammation in dogs with MMVD and CHF.

Simple Summary: This study focused on identifying and evaluating the use of selected potential biomarkers in predicting the treatment response of mast cell tumours (MCT) in dogs using a combination of electrochemotherapy (ECT) and interleukin 12 gene electrotransfer (IL-12 GET). In the study of 48 patients with 86 MCTs, blood samples were taken before, one month and six months after treatment to identify reliable biomarkers that predict response to treatment. An increased plasma nucleosome concentration and increased serum lactate dehydrogenase (LDH) activity one month after treatment correlated with a more effective local response, such as necrosis and swelling. These biomarkers proved to be potential early indicators of treatment success. However, the serum ferritin concentration did not prove to be meaningful. The results of this study provide crucial insights into the use of the plasma nucleosome concentration and serum LDH activity as potential indicators of treatment efficacy in veterinary oncology. Ultimately, this study emphasises the need to identify and validate predictive biomarkers to improve treatment outcomes in dogs with MCT and other malignancies treated similarly. Electrochemotherapy (ECT) in combination with the gene electrotransfer of interleukin 12 (IL-12 GET) has been successfully used in veterinary medicine for the treatment of mast cell tumours (MCT), but the biomarkers that could predict response to this treatment have not yet been investigated. The aim of this study was to determine the plasma nucleosome and serum ferritin concentrations, as well as the lactate dehydrogenase (LDH) activity, in the serum of treated patients before and one and six months after treatment to evaluate their utility as potential biomarkers that could predict response to the combined treatment. The study was conducted in 48 patients with a total of 86 MCTs that we treated with the combined treatment. The blood samples used for analysing the potential predictive biomarkers were taken before treatment and one and six months after treatment, when the response to treatment was also assessed. The Nu. ® Vet Cancer Test, the Canine Ferritin ELISA Kit, and the RX Daytona+ automated biochemical analyser were used to analyse the blood samples. The results showed that the plasma nucleosome concentration (before treatment (BT): 32.84 ng/mL (median); one month after treatment (1 M AT): 58.89 ng/mL (median); p = 0.010) and serum LDH activity (BT: 59.75 U/L (median); 1 M AT: 107.5 U/L (median); p = 0.012) increased significantly one month after treatment and that the increase correlated significantly with the presence of a more pronounced local reaction (necrosis, swelling, etc.) at that time point for both markers (nucleosome: BT (necrosis): 21.61 ng/mL (median); 1 M AT (necrosis): 69.92 ng/mL (median), p = 0.030; LDH: BT (necrosis): 54.75 U/L (median); 1 M AT (necrosis): 100.3 U/L (median), p = 0.048). Therefore, both the plasma nucleosome concentration and serum LDH activity could serve as early indicators of the effect of the treatment. In this context, the serum ferritin concentration showed no significant predictive potential for treatment response (p > 0.999 for all comparisons). In conclusion, this study provides some new and important observations on the use of predictive biomarkers in veterinary oncology. Furthermore, it emphasises the need for the continued identification and validation of potential predictive biomarkers in dogs with MCT and other malignancies undergoing ECT treatment in combination with IL-12 GET to ultimately improve treatment outcomes. The effects of antioxidant supplements on exercise-induced oxidative stress have not been investigated in untrained leisure horses. We investigated the effects of 14-day supplementation with vitamin E (1.8 IU/kg/day), coenzyme Q10 (CoQ10; ubiquinone; 800 mg/day), and a combination of both (the same doses as in mono-supplementation) on the blood levels of CoQ10, vitamin E, and oxidative stress parameters in untrained leisure horses subjected to acute moderate exercise. Correlations between lipid peroxidation and muscle enzyme leakage were also determined. Forty client-owned horses were included in the study, with 10 horses in each of the antioxidant and placebo (paraffin oil) groups. Blood parameters were measured before supplementation, before and immediately after exercise, and after 24 h of rest. The differences in individual parameters between blood collection times and groups were analysed with linear mixed models (p ˂ 0.05). None of the supplemented antioxidants affected vitamin E and CoQ10 concentrations, oxidative stress parameters, or serum muscle enzymes. Lipid peroxidation occurred in horses supplemented with placebo and CoQ10 but not in horses supplemented with vitamin E or the combination of both antioxidants. These results suggest that vitamin E alone or in combination with CoQ10 prevented lipid peroxidation in untrained leisure horses subjected to acute moderate exercise.

The objective of this study was to compare the effects of >Your< Iron Syrup, a novel oral liquid iron-containing food supplement, with the commonly prescribed iron sulphate (Fe-sulphate) in a mouse model of diet-induced iron deficiency. Standard inbred BALB/cOlaHsd mice were fed low-iron diet for 11 weeks to induce significant decrease in blood haemoglobin and haematocrit and were then supplemented by gavage with either >Your< Iron Syrup or Fe-sulphate for two weeks. In >Your< Iron Syrup group, several markers of iron deficiency, such as serum iron concentration, transferrin saturation and ferritin level were significantly improved in both female and male mice. Fe-sulphate induced similar responses, except that it did not significantly increase iron serum in females and serum ferritin in both sexes. Fe-sulphate significantly increased liver-iron content which >Your< Iron Syrup did not. Transcription of Hamp and selected inflammatory genes in the liver was comparable between the two supplementation groups and with the Control diet group. Some sex-specific effects were noted, which were more pronounced and less variable in males. In conclusion, >Your< Iron Syrup was efficient, comparable and in some parameters superior to Fe-sulphate in improving iron-related parameters without inducing a response of selected liver inflammation markers in a mouse model of diet-induced iron deficiency.

The urothelium is a vital permeability barrier that prevents the uncontrolled flow of urinary components into and out of the bladder interstitium. Our study addressed the question of possible sex-specific variations in the urothelium of healthy mice and their impact on chronic bladder inflammation. We found that healthy female bladders have a less robust barrier function than male bladders, as indicated by significant differences in transepithelial electrical resistance (TEER) values. These differences could be attributed to detected higher claudin 2 mRNA expression and a less pronounced glycocalyx in females than in males. In addition, TEER measurements showed delayed barrier recovery in chronically inflamed female bladders. We found subtle differences in the expressions of genes involved in the regulation of the actin cytoskeleton between the sexes, as well as pronounced urothelial hyperplasia in females compensating for attenuated barrier function. The identified genetic variations in glycosylation pathways may also contribute to this divergence. Our findings add to the growing body of literature on the intricate sex-specific nuances of urothelial permeability function and their implications for chronic bladder inflammation. Optimal heart function depends on perfect synchronization between electrical and mechanical activity. In this pilot study, we aimed to investigate the electromechanical activity of the heart in healthy cats and cats with cardiomyopathy with phonocardiography (PCG) synchronized to an electrocardiography (ECG) pilot device. We included 29 cats (12 healthy cats and 17 cats diagnosed with cardiomyopathy) and performed a clinical examination, PCG synchronized with ECG and echocardiography. We measured the following durations with the pilot PCG device synchronized with ECG: QRS (ventricular depolarization), QT interval (electrical systole), QS1 interval (electromechanical activation time (EMAT)), S1S2 (mechanical systole), QS2 interval (electrical and mechanical systole) and electromechanical window (end of T wave to the beginning of S2). The measured parameters did not differ between healthy cats and cats with cardiomyopathy; however, in cats with cardiomyopathy, EMAT/RR, QS2/RR and S1S2/RR were significantly longer than in healthy cats. This suggests that the hypertrophied myocardium takes longer to generate sufficient pressure to close the mitral valve and that electrical systole, i.e., depolarization and repolarization, and mechanical systoles are longer in cats with cardiomyopathy. The PCG synchronized with the ECG pilot device proved to be a valuable tool for evaluating the electromechanical activity of the feline heart. [ABSTRACT FROM AUTHOR]

38. THE INFLUENCE OF ISOFLURANE ANAESTHESIA ON INTESTINAL PERMEABILITY IN HEALTHY DOGS

This study investigated the most appropriate blood sampling time for determining the lactulose/mannitol (L/M) index, and whether isoflurane anaesthesia increases intestinal permeability in dogs, in terms of changes in the L/M index. Six dogs were given 100 ml of sugar solution (3.6 g lactulose and 3.4 g mannitol) with orogastric tube. Blood samples for determination of basal plasma L/M index were taken 90, 120, and 180 minutes later. The next day, the dogs were administered methadone, induced with midazolam mixed with ketamine, followed immediately by propofol, and anaesthesia maintained with isoflurane in oxygen for 200 minutes. The same sugar solution as the day before was administered at the end of anaesthesia and 12 and 24 hours post-anaesthesia. Blood samples were taken 90, 120 and 180 minutes after administration of sugar solution. HPLC-MS was used for plasma determination of lactulose and mannitol and the results expressed as L/M index. The highest concentrations of lactulose and mannitol were detected 120 minutes after administration of sugar solution. Lactulose/mannitol index significantly increased at the end of anaesthesia, regardless the sampling time, when compared to basal values from non-anaesthetized dogs. The increase of L/M index due to anaesthesia with isoflurane was short-lived as there was no significant difference between L/M index at 12 hours after the anaesthesia and basal values. The most appropriate blood sampling time for determining the L/M index is 120 minutes after oral administration of dual sugar solution. Key words: dog; isoflurane; intestinal permeability; L/M index VPLIV ANESTEZIJE Z IZOFLURANOM NA PREPUSTNOST ČREVESJA PRI ZDRAVIH PSIH Namen raziskave je bil ugotoviti najprimernejši čas odvzema krvi za določanje razmerja med laktulozo in manitolom (indeks L/M) in za ugotavljanje ali anestezija z izofluranom zveča indeks L/M in s tem prepustnost črevesne bariere pri psih. Šestim psom smo z orogastrično sondo dali 100 ml sladkorne raztopine, in sicer 3,6 g laktuloze in 3,4 g manitola. Za določitev bazalne vrednosti indeksa L/M smo psom odvzeli kri 90, 120 in 180 minut po dajanju sladkorne raztopine. Naslednji dan smo jih premedicirali z metadonom, v anestezijo pa uvedli z midazolamom s ketaminom in propofolom. Anestezijo smo vzdrževali z izofluranom v kisiku 200 minut. Enako sladkorno raztopino smo psom dali še trikrat, in sicer na koncu anestezije ter 12 in 24 ur po anesteziji. Po vsakem dajanju sladkorne raztopine smo po 90, 120 in 180 minutah odvzeli kri za določitev indeksa L/M. Laktulozo in manitol v plazmi smo določali z metodo HPLC-MS in rezultat izrazili kot indeks L/M. Najvišjo koncentracijo laktuloze in manitola smo zaznali 120 minut po dajanju sladkorne raztopine. Indeks L/M se je, v primerjavi z bazalnimi vrednostmi pri neanesteziranih psih, na koncu anestezije značilno zvišal ne glede na čas odvzema krvi. Anestezija z izofluranom kratkotrajno zveča indeks L/M, saj že 12 ur po anesteziji ni bilo značilne razlike v primerjavi z bazalno vrednostjo. 39. Antioxidative Effects of Olive Polyphenols Compared to Vitamin E in Piglets Fed a Diet Rich in N-3 PUFA

The aim of the current study was to assess the antioxidative effects of the dietary supplementation of olive leaf extract (OLE) in different concentrations compared to those of vitamin E in piglets under conditions of dietary n-3 PUFA-induced oxidative stress. Forty-eight castrated male piglets (10.6 ± 0.99 kg) were fed the following experimental diets: Cont− (low-fat diet, no supplement), Cont+ (high linseed oil diet, no supplement), Vit-E (as Cont+, 105 IU vitamin E/day), OLE-1 (as Cont+, 3.84 mg hydroxytyrosol equivalents (HEQs)/day), OLE-2 (as Cont+, 38.4 mg HEQ/day), and OLE-3 (as Cont+, 96 mg HEQ/day). After 21 days of feeding, the experimental diets, blood and urine samples were collected to assess the extent of the oxidative stress. Results indicated that diet OLE-1 lowered the activity of gamma-glutamyl transferase, protected DNA (measured as DNA tail %) and altered urinary 8-hydroxy-2’-deoxyguanosine (8-OHdG). Dietary vitamin E lowered the levels of urinary F2-isoprostanes, as well as of plasma malondialdehyde and γ-tocopherol, but raised the plasmatic α-tocopherol and altered the level of urinary 8-OHdG. In conclusion, only minor positive effects of dietary OLE on the oxidative stress parameters were observed. Additionally, OLE did not show concentration dependence.

Despite numerous studies on haemorrhagic shock treatment, the choice of fluid and the rate of administration is a subject of controversy. Early haemodynamic effects, neuroendocrine response and renal function following acute sodium load with either hypertonic saline (HS) or Lactated Ringer's (LRS) were investigated in a model of controlled haemorrhagic shock. Six anaesthetized dogs were bled 30% of calculated blood volume, i.e., 27 ml/kg b.w. over 15 min and left untreated for 30 min. Afterward, the dogs were treated with either HS 4 ml/kg b.w. or LRS 81 ml/kg b.w., followed by whole blood. After a resting period of at least 30 days, the dogs underwent a similar second shock experiment, but with the second solution. Haemodynamics, renal effects and neuroendocrine response were studied during shock and at 15, 30, 45 and 60 min after administration of HS or LRS and 30 and 60 min after the addition of whole blood. LRS treatment resulted in significantly higher cardiac filling pressures and cardiac performance indexes than HS but, following whole blood administration, there were no differences between treatments, except that pulmonary capillary wedge pressure was higher in the LRS group. Urinary output and glomerular filtration rate index were restored to normal values by both treatments; however, they were significantly higher in the LRS group until whole blood was administered. LRS and HS restored plasma norepinephrine to baseline values with no difference between the treatments. Both solutions improved haemodynamics and renal function but their effects were short lived and whole blood was required for a favourable outcome, regardless of the initial solution used. In contrast to LRS, HS did not overload the cardiovascular system.

This study was aimed to assessing haematological parameters in pigs of different age categories from six farms differing in the presence or absence of selected pathogens. The following categories were included: 5 age groups of growers, fatteners and breeding sows. Individual blood samples for determining complete blood count and white blood cell differential count were taken. Group samples of oral fluid and faeces were collected from each farm and tested for detection of Porcine Reproductive and Respiratory Virus (PRRSV), Porcine Circovirus Type 2 (PCV2), and Hepatitis E Virus (HEV) using PCR, RT-PCR, and qRT-PCR protocols. On farms free of PRRSV, PCV2 and HEV, the age of pigs had significant effect on: white blood cell count (WBC), haemoglobin concentration (Hb), mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin concentration, platelet count (PLT), percentage of neutrophils, lymphocytes and eosinophils and absolute numbers of neutrophils, lymphocytes, monocytes, basophils and 'large unstained cells' (LUCs). On positive farms all observed blood parameters except the percentage of LUCs were significantly affected by the age. The percentages of lymphocytes, mean corpuscular volume (MCV) and haematocrit were significantly lowered by PRRSV, while WBC, PLT, percentage and absolute numbers of neutrophils, basophils and LUCs were increased. Nine-week-old pigs with PCV2 had significantly lower red blood cell count, PLT, percentage and absolute numbers of monocytes and LUCs, and significantly higher MCV, MCH, percentage of neutrophils and percentage and absolute number of eosinophils than 9-week-old pigs with PCV2 and HEV. Age-related changes in haematological parameters occurred in PCR negative and positive farms. Age-related changes in haematological parameters were present in PCR negative farms and in PCR positive farms in relation to selected pathogens. Values for several haematological parameters in 9-week-old pigs with a monoinfection differed significantly from those in pigs of the same age with two viral infections. Background: Data on alterations in peripheral blood lymphocyte (PBL) subtypes in dogs with myxomatous mitral valve disease (MMVD) is lacking. Objectives: To investigate PBL subtypes and their correlation with parameters of inflammation and MMVD progression markers in dogs with different stages of MMVD. Animals: Seventy‐eight client‐owned dogs: 65 with MMVD (American College of Veterinary Internal Medicine [ACVIM] classification stages B2, C, and D) and 13 healthy controls. Methods: Prospective cross‐sectional study. Complete cardiac assessment, flow cytometry (T lymphocytes [CD3+], their subtypes [CD3+CD4+, CD3+CD8+, CD3+CD4+CD8+, CD3+CD4−CD8−], and B lymphocytes [CD45+CD21+]) and measurement of N‐terminal pro B‐type natriuretic peptide, cardiac troponin I, and C‐reactive protein concentrations were performed. Results: The percentage of CD3+CD4+ lymphocytes was significantly lower in stable ACVIM C patients (P =.01) and unstable ACVIM C and D patients (P =.003), the percentage of CD3+CD8+ lymphocytes was significantly higher in stable ACVIM C patients (P =.01) and unstable ACVIM C and D patients (P =.01), CD3+CD8+ lymphocyte concentration was significantly higher in unstable ACVIM C and D patients (P =.05), and the CD3+CD4+/CD3+CD8+ ratio was significantly lower in stable ACVIM C patients (P =.01) and unstable ACVIM C and D patients (P =.01) compared with healthy controls. Conclusions and Clinical Importance: The percentages of CD3+CD4+ and CD3+CD8+ PBL and CD4+/CD8+ ratio were altered in MMVD dogs with congestive heart failure (ACVIM C, D), but not in ACVIM B2, suggesting involvement of these PBL subtypes in the pathogenesis of congestive heart failure in dogs with MMVD. Our study aimed to investigate the blood variables: coenzyme Q10, N-terminal pro-B-type natriuretic peptide (NT-proBNP), electrolytes, cholesterol, triglyceride, glutathione peroxidase, white blood cell, neutrophil, lymphocyte and monocyte counts and relative numbers of neutrophils, lymphocytes and monocytes that may be associated with survival of dogs in congestive heart failure (CHF). Twentyone client-owned dogs in CHF were included in the study. Cox regression analysis showed significant association only between NT-proBNP blood concentration and survival. [ABSTRACT FROM AUTHOR]

The aim of this study was to investigate the effects of total intravenous anaesthesia with propofol and anaesthesia induced with propofol and maintained with sevofl urane on oxidative stress parameters in dogs with early-stage myxomatous mitral valve degeneration (MMVD). Sixteen client-owned dogs with early stage MMVD that required periodontal treatment were included in the study. After induction with propofol, anaesthesia was maintained with propofol (group P) or sevoflurane (group PS). Blood samples for determination of vitamin E, superoxide dismutase, glutathione peroxidase and malondialdehyde were collected before premedication, 5 and 60 minutes and 6 hours after induction to anaesthesia. There were no significant differences between groups in any of the oxidative stress parameters at each sampling time. Compared to basal values, vitamin E concentration decreased significantly during anaesthesia in both groups and glutathione peroxidase activity increased 60 minutes after induction to anaesthesia in PS group. Anaesthesia with propofol or with propofol and sevofl urane did not have any significant impact on oxidative stress parameters in dogs with early stage MMVD. In terms of oxidative stress, both protocols may be equally safely used in dogs with early stage MMVD. [ABSTRACT FROM AUTHOR]

We tested the hypothesis that indirect measures of oxidative stress (vitamin E, glutathione peroxidase, and malondialdehyde) differ in dogs in heart failure resulting from either myxomatous mitral valve disease or dilated cardiomyopathy. Dogs were classified according to the International Small Animal Cardiac Health Council (ISACHC) classification. Additionally, the effect of cardiac therapy on oxidative stress parameters and N-terminal pro–B-type natriuretic peptide (NT-proBNP) in advanced stages of congestive heart failure was investigated. There were no significant differences in oxidative stress parameters between healthy dogs and the individual groups of cardiac patients. Significantly lower malondialdehyde (MDA) was observed in the ISACHC II group in comparison to ISACHC groups III and I. A significant positive correlation in treated patients was observed between NT-proBNP and MDA, NT-proBNP and vitamin E, as well as between MDA and vitamin E (and lipid-standardized vitamin E). No significant differences in any of the measured parameters were found between treated and non-treated cardiac patients. Our results suggest an association between MDA (the extent of lipid peroxidation) and NT-proBNP, vitamin E and NT-proBNP, as well as between MDA and vitamin E in treated canine patients. Low levels of plasma vitamin E concentrations were found in canine atopic dermatitis (CAD). The present study was aimed at determining the effect of an eight-week vitamin E supplementation on clinical response (Canine Atopic Dermatitis Extent and Severity Index (CADESI-03) scores and pruritus intensity) in dogs with atopic dermatitis. Levels of oxidative stress markers (plasma malondialdehyde and total antioxidant capacity (TAC), blood glutathione peroxidase and erythrocyte superoxide dismutase, plasma and skin vitamin E concentrations) were also determined. Twenty-nine dogs with CAD were included in the study. Fourteen received vitamin E (8.1 IU/kg once daily, orally) and 15 received mineral oil as placebo (orally). All dogs were treated with antihistamine fexofenadine. Levels of oxidative stress markers (with the exception of skin vitamin E), CADESI-03 and pruritus intensity were determined at the beginning, then every two weeks. Skin vitamin E was determined at the beginning and at the end of the treatment. Significantly higher plasma levels of vitamin E and TAC were observed in the vitamin E group than in the placebo group. CADESI-03 scores determined throughout the treatment in the vitamin E group were significantly lower than in the placebo group. The findings of this study support the supplementation of vitamin E in dogs with atopic dermatitis. [ABSTRACT FROM AUTHOR]

The article talks about a study to compare various endocrine, antioxidant, haematological and biochemical parameters between Coxiella burnetii positive and negative ewes to examine the possible effects of infection on selected blood parameters. Topics discussed include determination of serum Coxiella antibodies by enzyme-linked immunosorbent assay, determination of red blood count, haematocrit and mean corpuscular volume for the study, and leptin hormone produced by white adipose tissue.
