38. Accelerating the discovery of materials for clean energy in the era of smart automation

Abstract

The discovery and development of novel materials in the field of energy are essential to accelerate the transition to a low-carbon economy. Bringing recent technological innovations in automation, robotics and computer science together with current approaches in chemistry, materials synthesis and characterization will act as a catalyst for revolutionizing traditional research and development in both industry and academia. This Perspective provides a vision for an integrated artificial intelligence approach towards autonomous materials discovery, which, in our opinion, will emerge within the next 5 to 10 years. The approach we discuss requires the integration of the following tools, which have already seen substantial development to date: high-throughput virtual screening, automated synthesis planning, automated laboratories and machine learning algorithms. In addition to reducing the time to deployment of new materials by an order of magnitude, this integrated approach is expected to lower the cost associated with the initial discovery. Thus, the price of the final products (for example, solar panels, batteries and electric vehicles) will also decrease. This in turn will enable industries and governments to meet more ambitious targets in terms of reducing greenhouse gas emissions at a faster pace.

Activating mutations in NRAS are found in 20% of melanomas. NRAS-mutant melanoma is more aggressive and, therefore, has poorer outcomes, compared to non-NRAS-mutant melanoma. Despite promising preclinical data, to date immune checkpoint inhibitors remain the standard of care for locally advanced unresectable or metastatic NRAS melanoma. Data for efficacy of immunotherapy for NRAS melanoma mainly come from retrospective cohorts with divergent conclusions. MEK inhibitors have been the most developed targeted therapy approach. Although associated with an increase in progression-free survival, MEK inhibitors do not provide any benefit in terms of overall survival. Combination strategies with PI3K-AKT-mTOR pathway and CDK4/6 inhibitors seem to increase MEK inhibitors’ benefit. Nevertheless, results from clinical trials are still prelaminar. A greater comprehension of the biology and intracellular interactions of NRAS-mutant melanoma will outline novel impactful strategies which could improve prognosis of these subgroup of patients.Keywords: metastatic melanoma, NRAS mutation, MEK inhibitor, immunotherapy

Abstract

Background and objectivesObesity is a global epidemic which increases the risk of the metabolic syndrome. Cathelicidin (LL-37 and mCRAMP) is an antimicrobial peptide with an unknown role in obesity. We hypothesize that cathelicidin expression correlates with obesity and modulates fat mass and hepatic steatosis.Materials and methodsMale C57BL/6 J mice were fed a high-fat diet. Streptozotocin was injected into mice to induce diabetes. Experimental groups were injected with cathelicidin and CD36 overexpressing lentiviruses. Human mesenteric fat adipocytes, mouse 3T3-L1 differentiated adipocytes and human HepG2 hepatocytes were used in the in vitro experiments. Cathelicidin levels in non-diabetic, prediabetic and type II diabetic patients were measured by enzyme-linked immunosorbent assay.ResultsLentiviral cathelicidin overexpression reduced hepatic steatosis and decreased the fat mass of high-fat diet-treated diabetic mice. Cathelicidin overexpression reduced mesenteric fat and hepatic fatty acid translocase (CD36) expression that was reversed by lentiviral CD36 overexpression. Exposure of adipocytes and hepatocytes to cathelicidin significantly inhibited CD36 expression and reduced lipid accumulation. Serum cathelicidin protein levels were significantly increased in non-diabetic and prediabetic patients with obesity, compared with non-diabetic patients with normal body mass index (BMI) values. Prediabetic patients had lower serum cathelicidin protein levels than non-diabetic subjects.ConclusionsCathelicidin inhibits the CD36 fat receptor and lipid accumulation in adipocytes and hepatocytes, leading to a reduction of fat mass and hepatic steatosis in vivo. Circulating cathelicidin levels are associated with increased BMI. Our results demonstrate that cathelicidin modulates the development of obesity.

Abstract

Introducción. El tabaquismo es una enfermedad adictiva y crónica. Es el principal problema de salud pública y la patología más prevalente de las sociedades desarrolladas, siendo la principal causa de morbimortalidad. En España, la media es de casi 52000 muertes al año y la prevalencia de fumadores mayores de 15 años es del 24.4%. El objetivo del presente estudio es conocer la prevalencia de tabaquismo entre los docentes de educación secundaria, saber su opinión sobre la influencia de su hábito tabáquico en los alumnos, así como quién y en qué cursos deben iniciarse las intervenciones educativas. Material y métodos. Se realizó un estudio descriptivo transversal mediante un cuestionario anónimo autoadministrado compuesto por 17 ítems al personal docente de los centros de Educación Secundaria Obligatoria (ESO) de Alcázar de San Juan (Ciudad Real). Resultados. Se administraron 131 encuestas, 66 docentes habían fumado alguna vez (50.4%) y 33 fumaban en el momento de realizar la encuesta. Casi la mitad de los profesores consideraban que el hábito de fumar influía poco en sus alumnos. La mayor parte afirma que la prevención debe realizarse por profesionales sanitarios y que ésta debe iniciarse antes de llegar a la educación secundaria (en torno a los 10 años de edad). Conclusiones. Los resultados de la encuesta a profesores de Enseñanza Secundaria resultan una herramienta útil para obtener información sobre las percepciones y opiniones de los profesores. El realizar encuestas de manera simultánea a profesores y alumnos de ESO permite obtener información complementaria y una visión global del tabaquismo en Centros Educativos. La información obtenida de esta encuesta debería contribuir a diseñar y planificar intervenciones específicas en programas de prevención de tabaquismo.
