Abstract Background Inflammatory bowel disease (IBD) is a chronic condition influenced by diet, which affects gut microbiota and immune functions. The rising prevalence of IBD, linked to Western diets in developing countries, highlights the need for dietary interventions. This study aimed to assess the impact of white kidney beans (WKB) on gut inflammation and microbiota changes, focusing on their effects on enteric glial cells (EGCs) and immune activity in colitis. Methods Male C57BL/6 mice were divided into four groups: normal diet (ND), ND with 2.5% dextran sulfate sodium (DSS) for colitis induction, ND with 20% WKB, and WKB with 2.5% DSS. The dietary intervention lasted 17 weeks, with DSS given in the final week. Colonic inflammation was assessed by body weight, disease activity index, and histopathology. Epithelial barrier integrity was evaluated using immunofluorescence, transmission electron microscopy, and permeability assays. EGCs activity was analyzed via immunofluorescence and quantitative real-time PCR. Immune responses were measured using flow cytometry and cytokine profiling, while gut microbiota changes were examined through metagenomic sequencing. Results WKB supplementation significantly alleviated DSS-induced colitis in mice, evidenced by reduced weight loss, disease activity, and improved colonic histology. This effect was linked to enhanced mucosal barrier integrity, seen through increased tight junction protein and Muc2 expression, accompanied by favorable ultrastructural changes. WKB modulated EGCs activity via TNF-like cytokine 1 A inhibition, resulting in reduced glial fibrillary acidic protein expression. Immunologically, it downregulated Th1 and Th17 pro-inflammatory cells, increased Treg cells, and altered cytokine profiles (reduced TNF-α, IFN-γ, IL-17; increased IL-10). Metagenomic analysis showed that WKB restored gut microbiota balance, particularly enhancing beneficial bacteria like Akkermansia. KEGG pathway analysis further indicated that WKB supplementation improved key metabolic pathways, notably those related to phenylalanine, tyrosine, and tryptophan biosynthesis, thereby countering DSS-induced metabolic disruptions. Conclusions WKB shows promise for treating IBD by enhancing mucosal barriers, inhibiting EGCs activity, balancing Th1/Th17/Treg cells, and restoring gut microbiota and metabolic homeostasis, thereby alleviating colitis symptoms.

Summary: A major side effect of reperfusion therapy following myocardial infarction is myocardial ischemia-reperfusion injury (MIRI). Electroacupuncture preconditioning (EA-pre) has a long history in the treatment of cardiovascular diseases. Here, we demonstrate how EA-pre attenuates MIRI by affecting the phagocytosis of neuronal dendritic spines of microglia of the fastigial nucleus (FNmicroglia). We observed that EA-pre increased activity in FNGABA and then improved myocardial injury by inhibiting abnormal activities of glutaminergic neurons of the FN (FNGlu) during MIRI. Interestingly, we observed changes in the quantity and shape of FN microglia in mice treated with EA-pre and a decrease in the phagocytosis of FNGABA neuronal dendritic spines by microglia. Furthermore, the effects of improving MIRI were reversed when EA-pre mice were chemically activated by intra-FN lipopolysaccharide injection. Overall, our results provide new insight indicating that EA-pre regulates microglial engulfment capacity, thus promoting the improvement of cardiac sympathetic nervous disorder during MIRI.

AIM:To investigate the short-term effectiveness of orthokeratology(ortho-k)on controlling low-to-moderate myopic in Chinese children.METHODS: There were 271 subjects(271 eyes)enrolled in this study. In total, there were 141 cases aged 9.43±1.10 in the ortho-k group and the spherical equivalent refractive errors(SER)were -2.74±1.15D. The patients were examined at 1, 7, 30, and 90d after they started wearing the ortho-k lenses. There were 130 cases aged 9.37±1.00 enrolled in the control group, with the SER -2.88±1.39D, and the examinations occurred at 6, 12mo after they started wearing single-vision spectacles. Myopic progression was estimated from changes of axial length in both groups. The chi-square test, independent samples t-tests, paired t-tests, Spearman analysis were used to compare the data of the two groups. RESULTS: The axial elongation was 0.27±0.17mm in the ortho-k group after 1a which was significantly longer than that before wearing(PPPt-test). There was significant negative correlation between axial elongation and initial age in both groups during the one-year period(ortho-k group: rs=-0.309, Prs=-0.472,P0.36mm in 1a)were 38% in the ortho-k group and 76.5% in the control group, respectively; whereas those of the older group(aged 9.4～12.0)were lower, reaching 24.3% and 12.9% in the ortho-k and control groups. In patients with SER 5.00～6.00D, the axial elongation in ortho-k group was 57.1% lower than that in control group. CONCLUSION: Ortho-k lens is effective to control myopic progression in children with low-to-moderate myopia. In particular, it reduces the percentage of younger children with fast progression and has a better effect of controlling myopic progression with higher degrees of myopia.

Aiming at the university management information system security threats and challenges,a security architecture named 1C4GS was proposed.First connotation and function of five important components of 1C4GS was expounded,which named security management center,security communication network,security region boundary,security computing environment and security application.Then we used “basic personal data reporting system” as an example to construct the management information system security arrangement application based on 1C4AS,this arrangement application integrated a variety of security technologies and strategies such as transparent data encryption,user Identify,form edit cache as a whole to,and achieved the management information system’s network security,border security,computing environment security and application security.

CUEDC2, a CUE-domain-containing protein, modulates inflammation, but its involvement in tumorigenesis is still poorly understood. Here, we report that CUEDC2 is a key regulator of macrophage function and critical for protection against colitis-associated tumorigenesis. CUEDC2 expression is dramatically upregulated during macrophage differentiation, and CUEDC2 deficiency results in excessive production of proinflammatory cytokines. The level of CUEDC2 in macrophages is modulated by miR- 324-5p. We find that Cuedc2 KO mice are more susceptible to dextran-sodium-sulfate-induced colitis, and macrophage transplantation results suggest that the increased susceptibility results from the dysfunction of macrophages lacking CUEDC2. Furthermore, we find that Cuedc2 KO mice are more prone to colitis-associated cancer. Importantly, CUEDC2 expression is almost undetectable in macrophages in human colon cancer, and this decreased CUEDC2 expression is associated with high levels of interleukin-4 and miR-324-5p. Thus, CUEDC2 plays a crucial role in modulating macrophage function and is associated with both colitis and colon tumorigenesis.

In order to adapt to the demand of statisting and analyzing huge-data in universities under current situation,an overall programme was designed and implemented to construct data statistical service platform based on data warehouse.The overall framework of university data warehouse and the modeling method of data warehouse were researched,and a statistical data model based on data star-schema model was designed.Furthermore,the key technology in the process of implementing statistical service platform like ETL(Extraction-Transformation-Loading) and UI interface design were explored.It provides practical basis and theoretical support for the next step to build the management and decision support platform of universities.

Big data management gives challenges to data manage and service platform based on traditional relational database management system,and research on big data management and service platform supporting both structured and unstructured data has become a very urgent and important task.4 key technologies for big data management service platform was summarized,and the configurable generalization table model was adopt.The data service center system prototype BIG-DMS based on Hadoop platform and the application scheme of data integration services platform based on BIG-DMS were also designed to support both structured and unstructured data storage,management and access services.

Myocardial ischemia-reperfusion injury (MIRI) has high morbidity and mortality worldwide. Increasing evidence has shown that electroacupuncture (EA) plays a critical role in alleviating MIRI. The aim of this study is to investigate whether glutamatergic neurons in the lateral hypothalamus (LH) have vital effect on MIRI as well as the underlying mechanism during the EA pretreatment. The MIRI model was established by ligating the left anterior descending (LAD) coronary artery for 30 min followed by reperfusion for 2 h. Chemogenetics, electrocardiogram (ECG) recording, ELISA, multichannel physiology recording, and immunofluorescence staining methods were combined to demonstrate that firing frequencies of neurons in the LH and expression of c-Fos decreased by EA pretreatment. Meanwhile, EA preconditioning significantly reduced the percentage of infarct size and the levels of cardiac troponin I (cTnI) and creatine kinase isoenzymes (CK-MB) were similar to inhibition of glutamatergic neurons in LH, also attenuated morphology of myocardial tissue was induced by MIRI. However, activation of glutamatergic neurons in LH weakened the above effects of EA pretreatment. [ABSTRACT FROM AUTHOR]
