The magnetic dipole moments (μ) of 209Tlg (N=128) and 207Tlm (N=126) have been measured for the first time using the in-source laser resonance-ionization spectroscopy technique with the Laser Ion Source and Trap (LIST) at ISOLDE (CERN). The application of the LIST suppresses the usually overwhelming background of the isobaric francium isotopes and allows access to heavy thallium isotopes with A⩾207. The self-consistent theory of finite Fermi systems based on the energy density functional by Fayans et al. well describes the N dependence of μ for 1/2+ thallium ground states, as well as μ for the 11/2− isomeric states in europium, gold and thallium isotopes. The inclusion of particle-vibration coupling leads to a better agreement between the theory and experiment for μ(Tlg, Iπ=1/2+). It is shown that beyond mean-field contributions to μ cannot be neglected at least for thallium isotopes with Iπ=1/2+.

Two nearly degenerate positive-parity bands with the πg9/22⊗νg9/2−1 configuration and three nearly degenerate negative-parity bands with the πg9/2(p3/2,f5/2)⊗νg9/2−1 configuration have been identified in 81Kr. They are interpreted as chiral doublet bands and pseudospin-chiral triplet bands, which is supported by the constrained covariant density functional theory and the multiparticle plus rotor model calculations. The present work reports two new chiral configurations πg9/22⊗νg9/2−1 and πg9/2(p3/2,f5/2)⊗νg9/2−1, and the first example of pseudospin-chiral triplet bands involving the π(p3/2,f5/2) pseudospin doublet.

Excited states in 98Ru were investigated using γ-ray spectroscopy following the β-decay of 98Rh, and via the 100Ru(p,t) reaction. Combining the results from the two experiments, two states were revised to have spin-parity of 4+ and subsequently assigned to the 02+ and “γ” bands, respectively. The observed structures in 98Ru are suggested to be deformed and rotational, rather than spherical and vibrational, and fit well into the systematics of these excitations in the Ru isotopes. The 02+ excitation is suggested as a shape coexisting configuration. This observation eliminates some of the last remaining candidates for nearly harmonic vibrational nuclei in the Z≈50 region. Beyond-mean-field calculations are presented that support shape coexistence throughout the Ru isotopes with N=52–62, and suggest a smooth evolution of the shapes.

We report for the first time two distinctive features in the odd–odd nucleus 120I: a pair of doublet bands and a high-spin isomer built on the πh11/2νh11/2 configuration. For producing the excited states of 120I, a fusion-evaporation reaction 118Sn(6Li, 4n) at E=lab48 MeV was employed. The beam was provided by the 14UD tandem accelerator of the Heavy Ion Accelerator Facility at the Australian National University. The observed doublet structure built on the positive-parity states is the first case and unique in isotopes with Z=53. The emerging properties are indicative of the known chiral characteristics, leading to a doubling of states for the πh11/2νh11/2 configuration. In contrast, the high-spin isomer with a half-life of 49(2) ns at spin-parity Jπ=25+ can be explained in terms of a noncollective oblate structure with the full alignment of six valence nucleons outside the 114Sn core: three protons (g7/2)1(d5/2)1(h11/2)1 and three neutrons (h11/2)3. This is an outstanding case that reveals a pure single-particle structure consisting of equal numbers of valence protons and neutrons outside the semi-double shell closure of 114Sn with Z=50 and N=64. Objective: Current research does not provide a clear explanation for why some patients with Parkinson’s Disease (PD) develop psychotic symptoms. The ‘aberrant salience hypothesis’ of psychosis has been influential and proposes that dopaminergic dysregulation leads to inappropriate attribution of salience to irrelevant/non-informative stimuli, facilitating the formation of hallucinations and delusions. The aim of this study is to investigate whether non-motivational salience is altered in PD patients and possibly linked to the development of psychotic symptoms. Methods: We investigated salience processing in 14 PD patients with psychotic symptoms, 23 PD patients without psychotic symptoms and 19 healthy controls. All patients were on dopaminergic medication for their PD. We examined emotional salience using a visual oddball fMRI paradigm that has been used to investigate early stages of schizophrenia spectrum psychosis, controlling for resting cerebral blood flow as assessed with arterial spin labelling fMRI. Results: We found significant differences between patient groups in brain responses to emotional salience. PD patients with psychotic symptoms had enhanced brain responses in the striatum, dopaminergic midbrain, hippocampus and amygdala compared to patients without psychotic symptoms. PD patients with psychotic symptoms showed significant correlations between the levels of dopaminergic drugs they were taking and BOLD signalling, as well as psychotic symptom scores. Conclusion: Our study suggests that enhanced signalling in the striatum, dopaminergic midbrain, the hippocampus and amygdala is associated with the development of psychotic symptoms in PD, in line with that proposed in the ‘aberrant salience hypothesis’ of psychosis in schizophrenia.

Huntington's disease (HD) is an inherited neurodegenerative disorder that is classically defined by a triad of movement and cognitive and psychiatric abnormalities with a well-established pathology that affects the dopaminergic systems of the brain. This has classically been described in terms of an early loss of dopamine D2 receptors (D2R), although interestingly the treatments most effectively used to treat patients with HD block these same receptors. We therefore sought to examine the dopaminergic system in HD not only in terms of striatal function but also at extrastriatal sites especially the hippocampus, given that transgenic (Tg) mice also exhibit deficits in hippocampal-dependent cognitive tests and a reduction in adult hippocampal neurogenesis. We showed that there was an early reduction of D2R in both the striatum and dentate gyrus (DG) of the hippocampus in the R6/1 transgenic HD mouse ahead of any overt motor signs and before striatal neuronal loss. Despite downregulation of D2Rs in these sites, further reduction of the dopaminergic input to these sites by either medial forebrain bundle lesions or receptor blockade using sulpiride was able to improve both deficits in motor performance and adult hippocampal neurogenesis. In contrast, a reduction in dopaminergic innervation of the neurogenic niches resulted in impaired neurogenesis in healthy WT mice. This study therefore provides evidence that D2R blockade improves hippocampal and striatal deficits in HD mice although the underlying mechanism for this is unclear, and suggests that agents working within this network may have greater effects than previously thought.

The use of allogeneic fetal neural precursor cells (NPCs) as a cell replacement therapy in neurodegenerative disorders holds great promise. However, previous studies concerning the possibility of alloimmune rejection of the transplanted cells have been inconclusive. Here, we used flow cytometry to quantify the expression of major histocompatibility complex (MHC) molecules by human NPCs, obtained from the cortex or ventral mesencephalon of fetuses with gestational ages between 7 and 11 weeks. MHC class I was undetectable on the surface of freshly isolated primary fetal tissue from either location, but increased over time in proliferating NPC cultures; after 7 days in vitro, MHC class I was detectable on most cells. Following differentiation, MHC class I expression persisted on non-neuronal cells. MHC class II levels remained low at all time points but were inducible by pro-inflammatory cytokines, whereas the co-stimulatory molecules, CD80 and CD86, remained undetectable. Nonetheless, CD4+ and CD8+ T cells proliferated when peripheral blood mononuclear cells (PBMCs) were cultured with allogeneic NPCs. Weaker responses were obtained when NPCs were co-cultured with purified allogeneic responder T cells, suggesting that indirect allorecognition contributed significantly to PBMC responses. In conclusion, differentiating human NPCs are immunogenic in vitro, suggesting that they may trigger immune rejection unless transplant recipients are immunosuppressed.

Only a small proportion of dopamine neurons in nigral grafts typically survive transplantation into the adult striatum. Since many anaesthetics reduce blood flow and disturb a variety of brain metabolites, surgical anaesthesia may be one of the factors that compromise graft survival. Conversely, the lowered core body temperature induced by some anaesthetics might promote the survival of grafted cells by slowing their metabolism. In an initial screen, the widely-used surgical anaesthetic, equithesin, was found to reduce core temperature, mean arterial blood pressure, and to increase the partial pressure of oxygen in arterial blood without producing any significant alteration in arterial pH or the partial pressure of carbon dioxide. In the main experiment, rats with unilateral 6-hydroxydopamine lesions of the nigrostriatal bundle received dopamine-rich embryonic nigral grafts injected into the deafferented neostriatum via previously implanted guide cannulae, which allowed comparison to be made of graft survival after transplantation into awake and in re-anaesthetised animals. There were no significant differences between groups in either the functional effects of the grafts to compensate amphetamine-induced rotation, or in the survival and growth of the grafts as measured in post mortem histology. We therefore conclude that anaesthesia per se is not a major contributory factor in the relatively poor survival of dopamine neurons following transplantation into the rat striatum.
