In Taiwan, fish farmers buying aquaculture insurance are subsidized by over 50% of the premiums, but the demand is still low. Also, there is clear pattern that if they got little claim payments this year, their incentive to insure would decrease next year, and vice versa. This paper investigates the factors that affect aquaculture insurance purchase insurance in Taiwan from a behavioural economics perspective. By the survey sample data of 343 fish farmers from a research project of Council of Agriculture, we provide the evidence of the farmers’ narrow framing bias, and, by using a logistic regression, the results shows that those who are less willingness to buy, because the claims did not occur, less likely buy the insurance. It also supports the hypothesis that fish farmers with higher narrow framing bias and shortsightedness would buy less aquaculture insurance. Thus, it leads to the low demand for the aquaculture insurance.

Abstract We utilized a case control study to determine if novel oral anticoagulants were associated with a higher risk of inpatient epistaxis admission. Adult patients admitted with a principal diagnosis of epistaxis in 2019–2021 were identified as well as a control group of patients matched 1:1 for age, sex, race, and medical comorbidities. For both cohorts, the presence or absence of an oral anticoagulant, classified as vitamin K inhibitors, direct oral anticoagulants (DOAC) or platelet inhibitors, was identified. 158 adult unique inpatient admissions with a principal diagnosis of epistaxis were identified. Vitamin K inhibition was present in 5.7% of cases versus 0.6% of controls (p = 0.02; OR 9.48, range 1.19‐75.77), DOACs in 4.4% of cases versus 5.1% of controls (p = 1.0) and platelet inhibitors in 2.5% of cases versus 3.8% of controls (p = 0.75). We concluded vitamin K inhibitors, compared to DOACs and platelet inhibitors, may be associated with higher likelihood of epistaxis admission.

Abstract The pathological changes in epigenetics and gene regulation that accompany the progression of low-grade to high-grade gliomas are under-studied. The authors use a large set of paired atac-seq and RNA-seq data from surgically resected glioma specimens to infer gene regulatory relationships in glioma. Thirty-eight glioma patient samples underwent atac-seq sequencing and 16 samples underwent additional RNA-seq analysis. Using an atac-seq/RNA-seq correlation matrix, atac-seq peaks were paired with genes based on high correlation values (|r2| > 0.6). Samples clustered by IDH1 status but not by grade. Surprisingly there was a trend for IDH1 mutant samples to have more peaks. The majority of peaks are positively correlated with survival and positively correlated with gene expression. Constructing a model of the top six atac-seq peaks created a highly accurate survival prediction model (r2 = 0.68). Four of these peaks were still significant after controlling for age, grade, pathology, IDH1 status and gender. Grade II, III, and IV (primary) samples have similar transcription factors and gene modules. However, grade IV (recurrent) samples have strikingly few peaks. Patient-derived glioma cultures showed decreased peak counts following radiation indicating that this may be radiation-induced. This study supports the notion that IDH1 mutant and IDH1 wildtype gliomas have different epigenetic landscapes and that accessible chromatin sites mapped by atac-seq peaks tend to be positively correlated with expression. The data in this study leads to a new model of treatment response wherein glioma cells respond to radiation therapy by closing open regions of DNA.

This paper studies why the demand for Agricultural revenue insurance is so low in Taiwan even the government subsidize almost half of the premium. We look into how risk attitude, probability ambiguity and claim payment ambiguity can affect the lower demand of the insurance, and control factors like age, education, farming experience, scale, and loan borrowing. By the survey data of 199 farmers, we find that half of the sample farmers are kind of risk lovers, and, about half of them haver problem of probability ambiguity or claim payment ambiguity. By using the two-class logistic model of insurance buying or no buying, we find that the risk attitude as risk aversion has significant positive impacts on the insurance buying, or the risk lover has less demand for the insurance. However, the probability ambiguity and claim payment ambiguity, which are expected to have negative impacts on the insurance demand, is not statistically significant.

The regeneration potential of the mammalian heart is incredibly limited, as cardiomyocyte proliferation ceases shortly after birth. β-adrenergic receptor (β-AR) blockade has been shown to improve heart functions in response to injury; however, the underlying mechanisms remain poorly understood. Here, we inhibited β-AR signaling in the heart using metoprolol, a cardio-selective β blocker for β1-adrenergic receptor (β1-AR) to examine its role in heart maturation and regeneration in postnatal mice. We found that metoprolol enhanced cardiomyocyte proliferation and promoted cardiac regeneration post myocardial infarction, resulting in reduced scar formation and improved cardiac function. Moreover, the increased cardiomyocyte proliferation was also induced by the genetic deletion of Gnas, the gene encoding G protein alpha subunit (Gαs), a downstream effector of β-AR. Genome wide transcriptome analysis revealed that the Hippo-effector YAP, which is associated with immature cardiomyocyte proliferation, was upregulated in the cardiomyocytes of β-blocker treated and Gnas cKO hearts. Moreover, the increased YAP activity is modulated by RhoA signaling. Our pharmacological and genetic studies reveal that β1-AR-Gαs-YAP signaling axis is involved in regulating postnatal cardiomyocyte proliferation. These results suggest that inhibiting β-AR-Gαs signaling promotes the regenerative capacity and extends the cardiac regenerative window in juvenile mice by activating YAP-mediated transcriptional programs.

This paper uses Fratini’s (2017) actuarial approach and the traditional risk neutral approach to price the revenue insurance under the framework of Black Scholes put option pricing. Then, we extend the pricing to include a coverage limit. Further, we also decompose the revenue into price and yield and develop another pricing formula using the price and yield information. The risk neutral approach assumes that the underlying revenue is tradable while the actuarial approach assumes the revenue risk is diversifiable. We compute the premiums of the revenue insurance of sugar apple and banana in Taiwan. The results show that the premiums of the actuarial approach are generally lower than the risk neutral approach. Because revenue is often non-tradable, thus, if the individual revenue risk can be fully diversified, then the actuarial approach should be suggested for premiums calculation.

BACKGROUND: Obturator hernia (OH) usually presents as a surgical emergency, with open primary repair most commonly performed. Given the morbidity and high recurrence of this approach, we present a case and review the literature to evaluate the influence of the operative approach on OH repair. METHODS: A literature search via PubMed was performed. Inclusion criteria were studies that: (1) were written in English and published within 10 years; (2) included as keywords “obturator hernia” and/or “incarcerated” and/or “strangulated”; (3) reported the operative approach; and (4) reported postoperative outcomes. RESULTS: Overall, 225 studies were identified, and 53 met the inclusion criteria. Data from 425 patients were pooled. Open repair without mesh was performed in 239 (56.2%) patients, 121 (28.5%) had open repair with mesh, 44 (10.4%) had laparoscopic repair with mesh, and 21 (4.9%) had laparoscopic repair without mesh. Open repair had a mean hospital length of stay (LOS) of 13.4 days, 40.3% postoperative complications, and 9.7% 30-day mortality rate whereas laparoscopic repair had a mean LOS of 7.9 days, 3.1% postoperative complications, and no deaths. Small bowel resection (SBR) was performed in 44.7% of open and 15.4% of laparoscopic repairs. Patients with SBR demonstrated higher morbidity and mortality compared with patients without SBR. In patients without SBR, laparoscopy had advantages over open surgery in LOS, complications, and mortality rate. The overall recurrence rate was 7.7%, with a mean follow-up of 20.4 months. One (0.7%) recurrence was reported in mesh repair, whereas 28 (12.1%) recurrences were reported with tissue repair. CONCLUSION: OHs are the most common open repair without mesh. Our literature review showed that laparoscopic OH repair is associated with enhanced postoperative recovery and the use of mesh was associated with less recurrence. Further studies are still necessary to determine the optimal approach for OH repair, but laparoscopic repair with mesh should be performed when possible.

Objective: OsteoDIP aims to collect and provide, in a simple searchable format, curated high throughput RNA expression data related to osteoarthritis. Design: Datasets are collected annually by searching “osteoarthritis gene expression profile” in PubMed. Only publications containing patient data and a list of differentially expressed genes are considered. From 2020, the search has expanded to include non-coding RNAs. Moreover, a search in GEO for “osteoarthritis” datasets has been performed using ‘Homo sapiens' and ‘Expression profiling by array’ filters. Annotations for genes linked to osteoarthritis have been downloaded from external databases. Results: Out of 1204 curated papers, 63 have been included in OsteoDIP, while GEO curation led to the collection of 28 datasets. Literature data provides a snapshot of osteoarthritis research derived from 1924 human samples, while GEO datasets provide expression for additional 1012 patients. Similar to osteoarthritis literature, OsteoDIP data has been created mostly from studies focused on knee, and the tissue most frequently investigated is cartilage. GEO data sets were fully integrated with associated clinical data. We showcase examples and use cases applicable for translational research in osteoarthritis. Conclusions: OsteoDIP is publicly available at http://ophid.utoronto.ca/OsteoDIP. The website is easy to navigate and all the data is available for download. Data consolidation allows researchers to perform comparisons across studies and to combine data from different datasets. Our examples show how OsteoDIP can integrate with and improve osteoarthritis researchers’ pipelines.

The intracellular bacterial pathogen Shigella infects and spreads through the human intestinal epithelium. Effector proteins delivered by Shigella into cells promote infection by modulating diverse host functions. We demonstrate that the effector protein OspB interacts directly with the scaffolding protein IQGAP1, and that the absence of either OspB or IQGAP1 during infection leads to larger areas of S. flexneri spread through cell monolayers. We show that the effect on the area of bacterial spread is due to OspB triggering increased cell proliferation at the periphery of infected foci, thereby replacing some of the cells that die within infected foci and restricting the area of bacterial spread. We demonstrate that OspB enhancement of cell proliferation results from activation of mTORC1, a master regulator of cell growth, and is blocked by the mTORC1-specific inhibitor rapamycin. OspB activation of mTORC1, and its effects on cell proliferation and bacterial spread, depends on IQGAP1. Our results identify OspB as a regulator of mTORC1 and mTORC1-dependent cell proliferation early during S. flexneri infection and establish a role for IQGAP1 in mTORC1 signaling. They also raise the possibility that IQGAP1 serves as a scaffold for the assembly of an OspB-mTORC1 signaling complex.

Oligodendrocytes are the critical cell types giving rise to the myelin nerve sheath enabling efficient nerve transmission in the central nervous system (CNS). Oligodendrocyte precursor cells differentiate into mature oligodendrocytes and are maintained throughout life. Deficits in the generation, proliferation, or differentiation of these cells or their maintenance have been linked to neurological disorders ranging from developmental disorders to neurodegenerative diseases and limit repair after CNS injury. Understanding the regulation of these processes is critical for achieving proper myelination during development, preventing disease, or recovering from injury. Many of the key factors underlying these processes are epigenetic regulators that enable the fine tuning or reprogramming of gene expression during development and regeneration in response to changes in the local microenvironment. These include chromatin remodelers, histone-modifying enzymes, covalent modifiers of DNA methylation, and RNA modification–mediated mechanisms. In this review, we will discuss the key components in each of these classes which are responsible for generating and maintaining oligodendrocyte myelination as well as potential targeted approaches to stimulate the regenerative program in developmental disorders and neurodegenerative diseases.

Proper vascular formation is regulated by multiple signaling pathways. The vascular endothelial growth factor (VEGF) signaling promotes endothelial proliferation. Notch and its downstream targets act to lead endothelial cells toward an arterial fate through regulation of arterial gene expression. However, the mechanisms of how endothelial cells (ECs) in the artery maintain their arterial characteristics remain unclear. Here, we show that PRDM16 (positive regulatory domain-containing protein 16), a zinc finger transcription factor, is expressed in arterial ECs, but not venous ECs in developing embryos and neonatal retinas. Endothelial-specific deletion of Prdm16 induced ectopic venous marker expression in the arterial ECs and reduced vascular smooth muscle cell (vSMC) recruitment around arteries. Whole-genome transcriptome analysis using isolated brain ECs show that the expression of Angpt2 (encoding ANGIOPOIETIN2, which inhibits vSMC recruitment) is upregulated in the Prdm16 knockout ECs. Conversely, forced expression of PRDM16 in venous ECs is sufficient to induce arterial gene expression and repress the ANGPT2 level. Together, these results reveal an arterial cell-autonomous function for PRDM16 in suppressing venous characteristics in arterial ECs. [ABSTRACT FROM AUTHOR]

Evidencia arqueológica recuperada hace más de un siglo y los resultados de investigaciones más recientes sugieren que la isla de la Plata y cerro Jaboncillo, preeminentes lugares sagrados de la costa central del Ecuador, fueron seleccionados por los incas como sitios claves para establecer su presencia en la región y definir sus relaciones con el pueblo nativo manteño. En la isla de la Plata, se han registrado entierros sacrificiales inca del tipo capac hucha y una ofrenda masiva de Spondylus acompañada de un urpu inca; y en el cerro Jaboncillo, dos grupos de estructuras de piedra, identificables como observatorios dedicados al monitoreo de los solsticios, se componen de formas arquitectónicas claramente incas. Estas evidencias en conjunto con otras, examinadas en el contexto tanto de una presencia inca más amplia en la región como de intervenciones en otras provincias del imperio, conducen a una reevaluación de la estrategia inca con respecto a la costa ecuatoriana. Conducen, además, a una consideración de cómo, en práctica, los incas y los manteños hubieran interactuado, y, de ahí, cómo la presencia inca podría haber impactado a la sociedad manteña.

In Malawi, tetanus toxoid vaccination (TTV) is recommended in pregnancy, but few studies have assessed the prevalence of infant seroprotection against tetanus. Anti-TT levels from 84 6-week-old infants, born in 2019–2020 to mothers living with HIV (HEU: HIV-exposed-uninfected) infants and to HIV-negative women (HUU: HIV-unexposed-uninfected) infants were determined by ELISA assay. Although 94% of the infants (HEU=94.8%, HUU=92.3%) showed protective levels (>0.1 IU/mL), the mean titers observed (0.51 IU/mL) suggest an incomplete compliance with TT vaccination. The only factor positively correlated to anti-TT IgG levels was the duration of maternal antiretroviral therapy in HEU.

Abstract Background The Democratic Republic of the Congo (DRC) experienced its largest Ebola Virus Disease Outbreak in 2018–2020. As a result of the outbreak, significant funding and international support were provided to Eastern DRC to improve disease surveillance. The Integrated Disease Surveillance and Response (IDSR) strategy has been used in the DRC as a framework to strengthen public health surveillance, and full implementation could be critical as the DRC continues to face threats of various epidemic-prone diseases. In 2021, the DRC initiated an IDSR assessment in North Kivu province to assess the capabilities of the public health system to detect and respond to new public health threats. Methods The study utilized a mixed-methods design consisting of quantitative and qualitative methods. Quantitative assessment of the performance in IDSR core functions was conducted at multiple levels of the tiered health system through a standardized questionnaire and analysis of health data. Qualitative data were also collected through observations, focus groups and open-ended questions. Data were collected at the North Kivu provincial public health office, five health zones, 66 healthcare facilities, and from community health workers in 15 health areas. Results Thirty-six percent of health facilities had no case definition documents and 53% had no blank case reporting forms, limiting identification and reporting. Data completeness and timeliness among health facilities were 53% and 75% overall but varied widely by health zone. While these indicators seemingly improved at the health zone level at 100% and 97% respectively, the health facility data feeding into the reporting structure were inconsistent. The use of electronic Integrated Disease Surveillance and Response is not widely implemented. Rapid response teams were generally available, but functionality was low with lack of guidance documents and long response times. Conclusion Support is needed at the lower levels of the public health system and to address specific zones with low performance. Limitations in materials, resources for communication and transportation, and workforce training continue to be challenges. This assessment highlights the need to move from outbreak-focused support and funding to building systems that can improve the long-term functionality of the routine disease surveillance system.

The satellite monitoring of vegetation moisture content (VMC) and soil moisture content (SMC) in Southern European Atlantic mountains remains poorly understood but is a fundamental tool to better manage landscape moisture dynamics under climate change. In the Atlantic humid mountains of Portugal, we investigated an empirical model incorporating satellite (Sentinel-1 radar, S1; Sentinel-2 optical, S2) and ancillary predictors (topography and vegetation cover type) to monitor VMC (%) and SMC (%). Predictors derived from the S1 (VV, HH and VV/HH) and S2 (NDVI and NDMI) are compared to field measurements of VMC (n = 48) and SMC (n = 48) obtained during the early, mid and end of summer. Linear regression modelling was applied to uncover the feasibility of a landscape model for VMC and SMC, the role of vegetation type models (i.e. native forest, grasslands and shrubland) to enhance predictive capacity and the seasonal variation in the relationships between satellite predictors and VMC and SMC. Results revealed a significant but weak relationship between VMC and predictors at landscape level (R2 = 0.30, RMSEcv = 69.9 %) with S2_NDMI and vegetation cover type being the only significant predictors. The relationship improves in vegetation type models for grasslands (R2 = 0.35, RMSEcv = 95.0 % with S2_NDVI) and shrublands conditions (R2 = 0.52, RMSEcv = 45.3 %). A model incorporating S2_NDVI and S1_VV explained 52 % of the variation in VMC in shrublands. The relationship between SMC and satellite predictors at the landscape level was also weak, with only the S2_NDMI and vegetation cover type exhibiting a significant relationship (R2 = 0.28, RMSEcv = 18.9 %). Vegetation type models found significant associations with SMC only in shrublands (R2 = 0.31, RMSEcv = 9.03 %) based on the S2_NDMI and S1_VV/VH ratio. The seasonal analysis revealed however that predictors associated to VMC and SMC may vary over the summer. The relationships with VMC were stronger in the early summer (R2 = 0.31, RMSEcv = 90.1 %; based on S2_NDMI) and mid (R2 = 0.37, RMSEcv = 70.8 %; based on S2_NDVI), butnon-significant in the end of summer. Similar pattern was found for SMC, where the link with predictors decreases from the early summer (R2 = 0.33, RMSEcv = 16.0 %; based on S1_VH) and mid summer (R2 = 0.30, RMSEcv = 17.8 %; based on S2_NDMI) to the end of summer (non-significant). Overall, the hypothesis of a universal landscape model for VMC and SMC was not fully supported. Vegetation type models showed promise, particularly for VMC in shrubland conditions. Sentinel optical and radar data were the most significant predictors in all models, despite the inclusion of ancillary predictors. S2_NDVI, S2_NDMI, S1_VV and S1_VV/VH ratio were the most relevant predictors for VMC and, to a lesser extent, SMC. Future research should quantify misregistration effects using plot vs. moving window values for the satellite predictors, consider meteorological control factors, and enhance sampling to overcome a main limitation of our study, small sample size.

Abstract The mosquito species Aedes aegypti (L.) is known to act as a vector in the transmission of various diseases, including dengue fever and yellow fever. The use of insect repellents is one of precautionary measures used to mitigate the risk of these diseases in humans by reducing mosquito biting. Nepetalactone, a potent natural insect repellent primarily found in catnip (Nepeta cataria) essential oil, has emerged as a promising candidate for mosquito repellence. Here, we evaluated the potential of catnip essential oil (> 95% nepetalactone) for use as a mosquito repellent. Using a Y-tube olfactometer and human hands as an attractant, we analysed the effectiveness of catnip oil at repelling the mosquito species Aedes aegypti. We tested a range of dilutions of catnip essential oil and found that concentrations as low as 2% were effective at repelling > 70% of mosquitoes for between one and four hours after repellent application. These findings suggest that nepetalactone could potentially be used as a natural, effective alternative to synthetic mosquito repellents, thereby offering protection against vector-borne diseases.

Food production’s environmental, economic, and social challenges should be demystified through quantitative data. Therefore, the objective of this paper was to investigate the ecoregional sustainability of the Amazon biome from the perspective of the environmental life cycle, economic feasibility, and social life cycle analysis, emphasizing the pillars of sustainability in the production of three commodities: soybean, beef cattle, and Brazil nuts. Carbon footprint, net present value, and worker endpoint were the metrics evaluated. According to the results found in this study, the livestock presented greater environmental burdens in terms of carbon balance when compared to the production of Brazil nuts and soybean production with carbon balances in the order of 4.75 tCO2eq/ha, −0.02 tCO2eq/ha, and −1.20 tCO2eq/ha, respectively. From an economic viewpoint, the extractive production of Brazil nuts presented the highest net profit per hectare/year (USD 559.21), followed by the agricultural system (USD 533.94) and livestock (USD 146.19). Finally, in relation to the social aspect of the production systems analyzed, the negative impacts linked to beef cattle production are related to the subcategories of forced labor and equal opportunities, and the positive impacts linked to soybean production are related to the subcategories of salary and benefits. The results highlight a genuine and sustainable balance in Brazil nuts extraction, presenting it as an investment for a sustainable future while demystifying the multifaceted information related to food production as a whole, in order to assist in decision-making and the formulation of public policies.

Introduction: Daptomycin (DAP) is a cyclic lipopeptide that exhibits potent in vitro activity against many drug-resistant gram-positive organisms, including methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant enterococci (VRE). Despite substantial reports evaluating the clinical outcomes of DAP within the adult population, real-world data are lacking in children. The primary goal of this evaluation was to describe the clinical characteristics and outcomes of DAP use in pediatric patients across a wide range of infections. Methods: This retrospective evaluation included patients < 18 years of age who were treated with DAP from January 2014 to May 2023. The primary objective was to evaluate the composite clinical success, which was defined as a 30-day survival, the lack of a 30-day microbiological recurrence, and the resolution of signs and symptoms of an acute infection without therapy modifications based on clinical failures. Secondary objectives included adverse effects potentially attributable to DAP and reasons for DAP utilization. Results: Forty patients were included, which were predominately male (62.5%) and white (52.5%), with a median age of 8.7 [IQR, 4.4–16.0] years. DAP was used for a wide range of infections, including central line-associated bloodstream infections (CLABSIs; 32.5%), infective endocarditis (15.0%), surgical-site infections (12.5%), and osteomyelitis (12.5%). The most common pathogen isolated was MRSA (37.5%), and most patients were bacteremic (60.0%). The median DAP dose was 8 [IQR, 6–10] mg/kg, and the median duration of the DAP therapy was 11.5 [IQR, 4.8–18.8] days. Most patients achieved composite clinical success (75.0%). An adverse effect occurred in 5.0% of the patients. DAP was prescribed the most for its ease of use/ability to facilitate discharge (40.0%) and/or for issues with alternative therapies (37.5%). Conclusion: Most pediatric patients that received DAP demonstrated clinical success with a low incidence of adverse effects. Larger, real-world studies of DAP use are necessary to further assess clinical outcomes.

Abstract Background Gastrointestinal stromal tumors (GIST) represent a significant clinical challenge due to their metastatic potential and limited treatment options. Raf kinase inhibitor protein (RKIP), a suppressor of the MAPK signaling pathway, is downregulated in various cancers and acts as a metastasis suppressor. Our previous studies demonstrated low RKIP expression in GIST and its association with poor outcomes. This study aimed to expand on the previous findings and investigate the biological and therapeutic implications of RKIP loss on GIST. Methods To validate the RKIP prognostic significance, its expression was evaluated by immunohistochemistry in 142 bona fide GIST cases. The functional role of RKIP was evaluated in vitro, using the GIST-T1 cell line, which was knocked out for RKIP. The biological and therapeutic implications of RKIP were evaluated by invasion, migration, apoptosis, and 2D / 3D viability assays. Additionally, the transcriptome and proteome of RKIP knockout cells were determined by NanoString and mass spectrometry, respectively. Results Immunohistochemical analysis revealed the absence of RKIP in 25.3% of GIST cases, correlating with a tendency toward poor prognosis. Functional assays demonstrated that RKIP knockout increased GIST cells’ invasion and migration potential by nearly 60%. Moreover, we found that RKIP knockout cells exhibited reduced responsiveness to Imatinib treatment and higher cellular viability in 2D and 3D in vitro models, as assessed by apoptosis-related protein expression. Through comprehensive genetic and proteomic profiling of RKIP knockout cells, we identified several putative RKIP-regulated proteins in GIST, such as COL3A1. Conclusions Using a multidimensional integrative analysis, we identified, for the first time in GIST, molecules and pathways modulated by RKIP that may potentially drive metastasis and, consequently, poor prognosis in this disease.

Abstract Objectives: Evaluate system-wide antimicrobial stewardship program (ASP) update impact on intravenous (IV)-to-oral (PO) antimicrobial conversion in select community hospitals through pre- and postimplementation trend analysis. Methods: Retrospective study across seven hospitals: region one (four hospitals, 827 beds) with tele-ASP managed by infectious diseases (ID)-trained pharmacists and region two (three hospitals, 498 beds) without. Data were collected pre- (April 2022–September 2022) and postimplementation (April 2023–September 2023) on nine antimicrobials for the IV to PO days of therapy (DOTs). Antimicrobial administration route and (DOTs)/1,000 patient days were extracted from the electronical medical record (EMR). Primary outcome: reduction in IV DOTs/1,000 patient days. Secondary outcomes: decrease in IV usage via PO:total antimicrobial ratios and cost reduction. Results: In region one, IV usage decreased from 461 to 209/1,000 patient days (P = < .001), while PO usage increased from 289 to 412/1,000 patient days (P = < .001). Total antimicrobial use decreased from 750 to 621/1,000 patient days (P = < .001). In region two, IV usage decreased from 300 to 243/1,000 patient days (P = .005), and PO usage rose from 154 to 198/1,000 patient days (P = .031). The PO:total antimicrobial ratios increased in both regions, from .42–.52 to .60–.70 in region one and from .36–.55 to .46–.55 in region two. IV cost savings amounted to $19,359.77 in region one and $4,038.51 in region two. Conclusion: The ASP intervention improved IV-to-PO conversion rates in both regions, highlighting the contribution of ID-trained pharmacists in enhancing ASP initiatives in region one and suggesting tele-ASP expansion may be beneficial in resource-constrained settings.

Social media and crowdsourcing (SMCS) are increasingly proving useful for addressing the effects of natural and human-made hazards. SMCS allow different stakeholders to share crucial information during disaster management processes and to strengthen community resilience through engagement and collaboration. To harvest these opportunities there is a need for better knowledge on SMCS for diverse disaster scenarios. These challenges are being addressed within the LINKS Horizon 2020 project. The project aims at strengthening societal resilience by producing advanced learning on the use of SMCS in disasters. This is done through an in-depth study across three knowledge domains (disaster risk perception and vulnerability, disaster management processes, SMCS technologies), the establishment of an interactive framework, and an online platform in which a community of relevant stakeholders can learn and share knowledge and experiences. This paper provides an overview of the project objectives and approaches and a summary of the initial results.

Background: Cerebral palsy (CP) is one of the most common neurological disorders in children and results in lifelong physical impairments. Adults with CP have approximately the same life expectancy as their non-disabled peers, so helping them to stay healthy throughout the life course will have long-term cost benefits via reductions in hospital admissions, long-term care, and unemployment rates. Aim: To explore how adults with CP experience ageing. Design & setting: National online survey given to adults with CP in the UK. Method: The participants were adults with CP. Items for the online survey were taken from existing self-report measures, with additional items developed for the survey. Several domains of functioning were assessed including mobility, dexterity, fatigue, pain, speech, mental health, swallowing and health maintenance/self-care as well as healthcare usage. Data were analysed using χ2 to examine the relationships between the demographic variables and the survey responses. Cancer subtyping delivers valuable insights into the study of cancer heterogeneity and fulfills an essential step toward personalized medicine. For example, studies in breast cancer have shown that cancer subtypes based on molecular differences are associated with different patient survival and treatment responses. However, recent studies have suggested inconsistent breast cancer subtype classifications using alternative approaches, suggesting that current methods are yet to be optimized. Existing computation-based methods have also been limited by their dependency on incomplete prior knowledge and ineffectiveness in handling high-dimensional data beyond gene expression. Here, we propose a novel deep-learning-based algorithm, Moanna, that is trained to integrate multi-omics data for predicting breast cancer subtypes. Moanna’s architecture consists of a semi-supervised Autoencoder attached to a multi-task learning network for generalizing the combination of gene expression, copy number and somatic mutation data. We trained Moanna on a subset of the METABRIC breast cancer dataset and evaluated the performance on the remaining hold-out METABRIC samples and a fully independent cohort of TCGA samples. We evaluated our use of Autoencoder against other dimensionality reduction techniques and demonstrated its superiority in learning patterns associated with breast cancer subtypes. The overall Moanna model also achieved high accuracy in predicting samples’ ER status (96%), differentiating basal-like samples (98%), and classifying samples into PAM50 subtypes (85%). Moreover, Moanna’s predicted subtypes show a stronger correlation with patient survival when compared to the original PAM50 subtypes.

Abstract Increasing shares of volatile generation and non-steerable demand raise the need for automated control of the Energy Systems (ESs). Various solutions for management and schedule-based control of energy facilities exist today. However, the amount and diversity of applications lead to a multitude of different automated energy management solutions. Different optimization algorithms have proven more or less effective for energy management. The multitude of optimization algorithms and energy management solutions require flexible, modular, and scalable integrations. We present a novel Optimization Service (OS) for easily integrating optimization algorithms while evaluating candidate solutions in the context of ESs applications. We propose an adapter-based architecture using metadata and domain knowledge to bridge between clients, e.g. smart grid applications and optimization algorithms. The architecture interfaces different clients with optimizers in a flexible and modular way. The clients provide metadata-based descriptions of optimization jobs translated by OS. OS then interacts with optimizers and evaluates candidate solutions. A consistent definition of interfaces for clients and optimization algorithms facilitates the modular evaluation of candidate solutions. OS’s separation of client and optimization algorithms increases scalability by managing computational resources independently. We evaluate the presented architecture for scheduling a so-called Energy Hub (EH) as a test case describing a simulation scenario of a renewable EH embedded in grid scenarios from an industrial area in Karlsruhe, Germany. OS utilizes an Evolutionary Algorithm (EA) to optimize schedules for cost and strain on the electrical grid. The use case exemplifies OS’s advantages in a proof-of-concept evaluation.

Expensive, time-consuming and labour-intensive solvent-extraction and liquid-chromatography methods are the industry's current gold standard for antibiotic residue quantification. A novel immunoassay methodology and system for the rapid detection of clinically relevant levels of tetracycline residues found in food-producing animal tissues is described. Anti-tetracycline antibody-coated paramagnetic particles were used for the specific capture of tetracycline in spiked buffer (with and without a 1% pork muscle tissue suspension) and quantified via an analytically-sensitive in-house magnetometer instrument. Detection of tetracycline between 0.1 μg/mL - 1 μg/mL was achieved, with a readout time (including sample treatment) presented in 20 min. The magneto-immunoassay described provides a rapid, low-cost, de-skilled and analytically-sensitive solution for tetracycline screening at the point-of-sampling, with potential applications for other prevalent antibiotic families used in the international farming and food industry.

When a rectangular hyperbola is constructed from the image positions of a quadruply lensed quasar, as proposed by Witt, it passes very close to the lensing galaxy. The median measured perpendicular offset between the observed light center of the lens and Witt’s hyperbola is 0.″013 for a sample for 39 systems lensed by a relatively isolated galaxy. The family of lens models adopted by Witt predicts that the lens lies on the hyperbola, but its position is not used for its construction. The median offset corresponds to roughly 1% of the Einstein ring radius and suggests that the centers of the lensing potential are close to the light centers of the lens. By putting a restrictive prior on the perpendicular distance to Witt’s hyperbola (or on the distance between the galaxy and the potential), one reduces by one the dimensionality of a model space when fitting data. Taking the brightest pixel of a lensing galaxy as its center avoids a shortcoming of using the average light center for a constraint.

Summary: Space-based remote sensing can make an important contribution toward monitoring greenhouse gas emissions and removals from the agriculture, forestry, and other land use (AFOLU) sector, and to understanding and addressing human-caused climate change through the UNFCCC Paris Agreement. Space agencies have begun to coordinate their efforts to identify needs, collect and harmonize available data and efforts, and plan and maintain a long-term roadmap for observations. International cooperation is crucial in developing and realizing the roadmap, and the Committee on Earth Observation Satellites (CEOS) is a key coordinating driver of this effort. Here, we first identify the data and information that will be useful to support the global stocktake (GST) of the Paris Agreement. Then, the paper explains how existing and planned space-based capabilities and products can be used and combined, particularly in the land use sector, and provides a workflow for their harmonization and contribution to greenhouse gas inventories and assessments at the national and global level.

Se realizaron nuevos descubrimientos significativos durante las excavaciones de rescate conducidas en 2015–2016 en Calle 22, Salango, en la costa sur de la provincia de Manabí, Ecuador. Entre estos estaba la presencia de dos montículos funerarios que pueden atribuirse a una fase Guangala muy temprana previamente no documentada del Período Desarrollo Regional. Los entierros, en su mayoría de bebés, mostraban marcadas patologías indicativas de una variedad de problemas de salud. Y en lo que parece ser un ritual de entierro único a nivel mundial, dos bebés portaron como cascos los cráneos modificados de otros niños. La investigación en curso sugiere que los montículos funerarios y los entierros en Calle 22 eran parte de un programa de entierro más grande centrado en una plataforma ceremonial Engoroy Tardío situada 150 m. al sur, y que este programa siguió y fue una respuesta a una caída catastrófica de ceniza volcánica. Este artículo describe la evidencia arqueológica descubierta, el proceso de investigación y análisis realizado hasta el momento, y las razones para suponer que el programa de entierro se llevó a cabo en respuesta a la caída de ceniza.

Abstract Background The impaired transplacental passage of IgG from mothers living with HIV to their infants could be one of the causes of the high vulnerability to infections of HIV-exposed uninfected (HEU) infants, but controversial results have been obtained in different settings. The aim of this study was to assess in 6-week old HEU and HIV-unexposed, uninfected (HUU) Malawian infants the total IgG levels, the subclasses profile and the concentrations of global anti-pneumococcal capsular polysaccharide (anti-PCP) IgG and IgG2. Methods Dried blood spots were collected from 80 infants (40 HEU, 40 HUU) and antibodies concentrations determined by nephelometric method (total IgG and subclasses), or using ELISA (anti-PCP total IgG and IgG2). Results are expressed as median levels with IQR, while the proportions of each subclass out of the total IgG are used to describe the subclasses profile. Results At 6 weeks HEU infants had higher median levels of total IgG and IgG1 and a significantly lower level of IgG2 [0.376 (0.344–0.523) g/l vs 0.485 (0.374–0.781) g/l, p = 0.037] compared to the HUU counterparts. The IgG subclasses distribution confirmed the underrepresentation of IgG2 (IgG2 represented 5.82% of total IgG in HEU and 8.87% in HUU). Abstract The shortage of intensive care unit (ICU) resources, including equipment and supplies for renal replacement therapy (RRT), is a critical problem in several countries. This study aimed to assess hospital mortality and associated factors in patients treated in public hospitals of the Federal District, Brazil, who requested admission to ICU with renal replacement therapy support (ICU-RRT) in court. Retrospective cohort study that included 883 adult patients treated in public hospitals of the Federal District who requested ICU-RRT admission in court from January 2017 to December 2018. ICU-RRT was denied to 407 patients, which increased mortality (OR 3.33, 95% CI 2.39–4.56, p ≪ 0.01), especially in patients with priority level I/II (OR 1.02, 95% CI 1.01–1.04, p ≪ 0.01). Of the requests made in court, 450 were filed by patients with priority levels III/IV, and 44.7% of these were admitted to ICU-RRT. Tick-borne encephalitis virus is an enveloped, pathogenic, RNA virus in the family Flaviviridae, genus Flavivirus. Viral particles are formed when the nucleocapsid, consisting of an RNA genome and multiple copies of the capsid protein, buds through the endoplasmic reticulum membrane and acquires the viral envelope and the associated proteins. The coordination of the nucleocapsid components to the sites of assembly and budding are poorly understood. Here, we investigate the interactions of the wild-type and truncated capsid proteins with membranes with biophysical methods and model membrane systems. We show that capsid protein initially binds membranes via electrostatic interactions with negatively-charged lipids, which is followed by membrane insertion. Additionally, we show that membrane-bound capsid protein can recruit viral genomic RNA. We confirm the biological relevance of the biophysical findings by using mass spectrometry to show that purified virions contain negatively-charged lipids. Our results suggest that nucleocapsid assembly is coordinated by negatively-charged membrane patches on the endoplasmic reticulum and that the capsid protein mediates direct contacts between the nucleocapsid and the membrane.

ABSTRACT Dalbavancin is a second-generation lipoglycopeptide antibiotic with activity against Gram-positive organisms. Dalbavancin is Food and Drug Administration (FDA)-approved for acute bacterial skin and soft tissue infections (ABSSTIs). There is a lack of substantial data on dalbavancin in more invasive infections, particularly in high-risk populations (patients with intravenous drug use and unstable living conditions). In this retrospective observational study, we reviewed all patients that received at least one dose of dalbavancin in an inpatient or outpatient setting at Parkland Hospital from February of 2019 to August of 2021. The demographics, type of infection, and rationale for dalbavancin were collected at the baseline. Clinical failure was measured by an avoidance of emergency department (ED) visits or hospital readmission at 30, 60, and 90 days. A separate analysis was conducted to estimate hospital, rehabilitation, or nursing facility days saved based on the projected length of treatment. 40 patients were included, and the majority were uninsured (85%), experiencing homelessness (60%), or had intravenous drug use (IDU) (57.5%). Indications for use included ABSSTIs (45%), bloodstream infection (67.5%), osteomyelitis (40%), infective endocarditis (10%), and septic arthritis (10%). Clinical failure was observed in 5 of the 40 patients (12.5%). Nonadherence to medical recommendations, a lack of source control, and ongoing IDU increased the risk of failure. Dalbavancin saved a total of 566 days of inpatient, rehabilitation, and nursing facility stays. Dalbavancin is a reasonable alternative to the standard of care in an at-risk population, offering decreased lengths of stays and cost savings. The uses of second-generation lipoglycopeptides are desirable alternatives to traditional outpatient parenteral antibiotic therapies for patients who otherwise would not qualify or for patients who desire less hospital contact in light of the COVID-19 pandemic. IMPORTANCE This study contributes additional experience to the literature of dalbavancin use in off-label indications, particularly for patients who do not qualify for outpatient parenteral antimicrobial therapy. The majority of the patient population were people who inject drugs and the uninsured. There is difficulty in tracking outcomes in this patient population, given their outpatient follow-up rates; however, we were able to track emergency room visits and readmissions throughout the majority of the local metroplex. The clinical use of dalbavancin at our institution also increased in the midst of the COVID-19 pandemic in an effort to preserve hospital resources and limit health care exposure. In addition, we are able to provide institution-specific cost-saving data with the use of dalbavancin.

In Uganda, due to the criminalization of same-sex sexual practices, men who have sex with men (MSM) experience barriers to accessing HIV care. To retain patients within the HIV Care Continuum, some health interventions have used patient navigators as an ancillary support service. To understand the potential care benefits of using patient navigators for marginalized populations experiencing challenges to HIV care and treatment access in a Ugandan context, this qualitative study explored the experiences of newly diagnosed MSM using patient navigators for ARV retention in care in Kampala. Additionally, to gain insight into the feasibility of patient navigator interventions, this study also aimed to understand the perspectives and experiences of patient navigators working with HIV positive MSM. Individual in-depth, semi structured interviews were conducted with 24 HIV positive MSM and four patient navigators that were part of a patient navigator pilot program from January 2019 -December 2020. Analysis was done using manifest and latent qualitative content analysis. Results showed that HIV positive MSM in Uganda experienced a variety of social, emotional, and financial challenges that placed them at risk for dropping off the HIV Care Continuum. Patient navigators provided HIV positive MSM with the skills, support, and resources necessary to overcome these challenges. Based on study results, we conclude that within the patient navigator pilot program, patient navigators improved MSM participants' quality of life by helping them to achieve the HIV Care Continuum stages: diagnosis, linked to care, receiving HIV treatment, and retention in care. Study results suggest future research is needed on the psychosocial support needs of patient navigators, how the support needs of MSM change throughout their lifetime on the HIV Care Continuum, and how potential benefits of patient navigators may differ in rural Ugandan contexts.

Despite the growing evidence of the benefits of total-intravenous anaesthesia using propofol compared to conventional volatile-based anaesthesia, both in terms of environmental impact and patient outcomes, the majority of administered general anaesthetics use volatile agents. A significant reason for this is the lack of suitable methods for continuous, real-time propofol monitoring. Here we present a cytochrome P450 2B6/carbon nanotube/graphene oxide/metal oxide nanocomposite sensor for propofol monitoring. The enzyme prevents electrode fouling by converting the propofol into a quinone/quinol redox pair and the nanocomposite enables rapid and sensitive detection. The nanocomposite was synthesised via a simple ‘green synthesis’-based approach using an extract of common bay laurel. It was found that composites containing iron oxide nanoparticles resulted in the best performance, with a limit of detection of 7.0 ± 0.7 ng/ml and a sensitivity of 29.9 ± 6.4 nA/μg/ml/mm2. The sensor demonstrated good specificity with respect to several common perioperative drugs, propofol detection was demonstrated in a ‘serum-like’ solution and produced a linear response across the therapeutic range of propofol (1–10 μg/ml).

Abstract Anal cancer is a rare disease that has doubled in incidence over the last four decades. Current treatment and survival of patients with this disease has not changed substantially over this period of time, due, in part, to a paucity of preclinical models to assess new therapeutic options. To address this hiatus, we set-out to establish, validate and characterise a panel of human anal squamous cell carcinoma (ASCC) cell lines by employing an explant technique using fresh human ASCC tumour tissue. The panel of five human ASCC cell lines were validated to confirm their origin, squamous features and tumourigenicity, followed by molecular and genomic (whole-exome sequencing) characterisation. This panel recapitulates the genetic and molecular characteristics previously described in ASCC including phosphoinositide-3-kinase (PI3K) mutations in three of the human papillomavirus (HPV) positive lines and TP53 mutations in the HPV negative line. The cell lines demonstrate the ability to form tumouroids and retain their tumourigenic potential upon xenotransplantation, with varied inducible expression of major histocompatibility complex class I (MHC class I) and Programmed cell death ligand 1 (PD-L1). We observed differential responses to standard chemotherapy, radiotherapy and a PI3K specific molecular targeted agent in vitro, which correlated with the clinical response of the patient tumours from which they were derived. We anticipate this novel panel of human ASCC cell lines will form a valuable resource for future studies into the biology and therapeutics of this rare disease.

IntroductionPeach (Prunus persica (L.) Batsch,) and nectarine fruits (Prunus persica (L.) Batsch, var nectarine), are characterized by a rapid deterioration at room temperature. Therefore, cold storage is widely used to delay fruit post-harvest ripening and extend fruit commercial life. Physiological disorders, collectively known as chilling injury, can develop typically after 3 weeks of low-temperature storage and affect fruit quality.MethodsA comparative transcriptomic analysis was performed to identify regulatory pathways that develop before chilling injury symptoms are detectable using next generation sequencing on the fruits of two contrasting cultivars, one peach (Sagittaria) and one nectarine, (Big Top), over 14 days of postharvest cold storage.ResultsThere was a progressive increase in the number of differentially expressed genes between time points (DEGs) in both cultivars. More (1264) time point DEGs were identified in ‘Big Top’ compared to ‘Sagittaria’ (746 DEGs). Both cultivars showed a downregulation of pathways related to photosynthesis, and an upregulation of pathways related to amino sugars, nucleotide sugar metabolism and plant hormone signal transduction with ethylene pathways being most affected. Expression patterns of ethylene related genes (including biosynthesis, signaling and ERF transcription factors) correlated with genes involved in cell wall modification, membrane composition, pathogen and stress response, which are all involved later during storage in development of chilling injury.DiscussionOverall, the results show that common pathways are activated in the fruit of ‘Big Top’ nectarine and ‘Sagittaria’ peach in response to cold storage but include also differences that are cultivar-specific responses.

Background: Very limited information is available on SARS-CoV-2 seroprevalence in infants in sub-Saharan countries. Objective: In this study, we aimed to determine the rate and the temporal evolution of SARS CoV-2 seropositivity in breastfed Malawian infants. Study design: Blood samples (n = 250) from 158 infants, born to HIV-negative women and women living with HIV, collected from February 2020 to May 2021, were first tested using an Anti-IgG/A/M SARS CoV 2 ELISA assay against trimeric spike protein, and then, if positive, confirmed using a second ELISA assay detecting IgG against Receptor Binding Domain. Results: The confirmed prevalence of anti-SARS CoV-2 antibodies was 31.0% (95% CI: 23.7%-38.3%) with no significant difference between HIV-exposed and HIV-unexposed infants (29.3% and 37.1% respectively, P = 0.410). The presence of anti-SARS-CoV-2 IgG was not associated with maternal socioeconomic or demographic indices. Conclusions: Our data underline the wide spread of the SARS-CoV-2 infection in the pediatric population in sub-Saharan Africa. Design of more specific serological tests for African samples and improvements in serosurveillance programs are needed for more rigorous monitoring of the dynamics of SARS-CoV-2 infection in Africa.

Immunocompromised patients can experience prolonged SARS-CoV-2 infections in the setting of a lack of protectivity immunity despite vaccination. As circulating SARS-CoV-2 strains become more heterogeneous, concomitant infection with multiple SARS-CoV-2 variants has become an increasing concern. Immunocompromised patient populations represent potential reservoirs for the emergence of novel SARS-CoV-2 variants through mutagenic change or coinfection followed by recombinatory events. Identification of SARS-CoV-2 coinfections is challenging using traditional next generation sequencing pipelines; however, targeted genotyping approaches can facilitate detection. Here we describe five COVID-19 cases caused by coinfection with different SARS-CoV-2 variants (Delta/Omicron BA.1 and Omicron BA.1/BA.2) as identified by multiplex fragment analysis.

To accommodate surplus energy, the adipose tissue expands by increasing adipocyte size (hypertrophy) and number (hyperplasia). The presence of hypertrophic adipocytes is a key characteristic of adipose tissue dysfunction. High-fat diet (HFD) fed C57BL/6J mice are a commonly used model to study obesity and obesity-related complications. In the present study, we have characterized adipose plasticity, at both the cellular and tissue level, by examining the temporal development of systemic insulin resistance and adiposity in response to HFD-feeding for 4, 8, and 12 weeks (4w, 8w, and 12w). Within the same time frame, we examined systemic metabolic flexibility and adipose plasticity when switching from HFD- to chow-diet during the last 2 weeks of diet intervention (referred to as the reverse (REV) group: 4wREV (2w HFD+2w chow), 8wREV (6w HFD+2w chow), 12wREV (10w HFD+2w chow)). In response to HFD-feeding over time, the 12w group had impaired systemic insulin sensitivity compared to both the 4w and 8w groups, accompanied by an increase in hypertrophic inguinal adipocytes and liver triglycerides. After reversing from HFD- to chow-feeding, most parameters were completely restored to chow control levels for 4wREV and 8wREV groups. In contrast, the 12wREV group had a significantly increased number of hypertrophic adipocytes, liver triglycerides accumulation, and impaired systemic insulin sensitivity compared to chow-fed mice. Further, image analysis at the single-cell level revealed a cell-size dependent organization of actin filaments for all feeding conditions. Indeed, the impaired adipocyte size plasticity in the 12wREV group was accompanied by increased actin filamentation and reduced insulin-stimulated glucose uptake compared with chow-fed mice. In summary, these results demonstrate that the C57BL/6J HFD-feeding model has a large capacity to restore adipocyte cell size and systemic insulin sensitivity, and that a metabolic tipping point occurs between 8 and 12w of HFD-feeding where this plasticity deteriorates. We believe these findings provide substantial understanding of C57BL/6J mice as an obesity model, and that an increased pool of hypertrophic ING adipocytes could contribute to aggravated insulin resistance.

Background: The evaluation of seroprotection rates against vaccine-preventable infectious diseases allows for the identification of risk populations. HIV-exposed infants, even if not infected with HIV, have higher morbidity and mortality in comparison to unexposed counterparts. The aim of this study was to compare the specific IgG levels against Haemophilus influenzae type-B (HiB), Hepatitis-B (HBV), and Streptococcus pneumoniae (Spn) in two groups of infants (HIV-exposed and HIV-unexposed) living in Malawi. Methods: Blood samples from 62 infants, 49 HIV-exposed, uninfected (HEU), and born to women living with HIV and 13 HIV-unexposed and uninfected (HUU), were collected at 6 months, and specific IgG levels were determined using ELISA tests. Results: The antibody levels against HiB, HBV, and Spn were similar in the two groups. At six months, all HUU infants and 81.6% of HEU infants showed seroprotective levels against HiB, while a percentage of protection varying from 80.6 to 84.6% was observed for HBV and Spn regardless of HIV exposure. Only 59.2% of HEU and 69.2% of HUU infants showed antibody protection against all three pathogens. Conclusions: These results indicate similar rates of seroprotection among HEU and HUU infants but also suggest that a consistent fraction of infants received incomplete vaccinations. Strategies to enforce participation in immunization programs in Malawi should be a health priority.

This study explores the potential use of graphene-based electrodes in the electrochemical determination of estradiol using amperometric techniques as a simple, enzyme-free approach. Graphene, a carbon-based nanomaterial, has been extensively investigated in materials science as a sensing material. Its remarkable properties, such as its high electron mobility and conductivity, robust mechanical characteristics, and good surface-to-volume ratio, have led to its adoption in numerous applications, including electrochemical sensing. Estradiol is a crucial sex hormone that affects metabolism and reproduction. However, excessive amounts may disrupt endocrine functions. Electrochemical sensors suffer from electrode fouling, leading to passivation that ultimately affects performance. We exploit the inherent properties of various types of graphene-based electrodes, including graphene screen-printed electrodes (GHSPE), electrochemically exfoliated graphene-modified electrodes (EEFGHSPE), and 3D graphene foam screen-printed electrodes (3D-GFSPE), for the amperometric studies. The electrochemical properties and structural characteristics of these sensors are evaluated using cyclic voltammetry and scanning electron microscopy. The analytical performance of these sensors is at an applied potential of +0.65 V (vs. Ag/AgCl) over the concentration range 0.83 to 4.98 μM estradiol. Sensitivities of 0.151 µAµM−1 cm−2, 0.429 µAµM−1 cm−2, and 0.273 µA µM−1 cm−2, with detection limits of 0.0041 µM, 0.097 µM, and 0.018 µM (S/N = 3), are found for GHPSPE, 3D-GFSPE and EEFGHSPE, respectively. The possibility of amperometrically determining the estradiol levels in a potable tap water sample are then investigated over the concentration range 0.83–4.98 µM.

Abstract Background In sub-Saharan African countries Epstein Barr virus (EBV) infection occurs in early childhood. We aim to investigate the factors associated with EBV acquisition and the impact of EBV infection on the humoral response to HBV vaccination in infants born from HIV-positive, antiretroviral-treated mothers in Malawi. Methods A total of 149 HIV-exposed infants were included in this longitudinal study. EBV anti-VCA IgG were measured using an ELISA assay. The EBV seroconversion was correlated with the maternal viro-immunological conditions, with infant growth and immunological vulnerability, and with the humoral response to the HBV vaccine. Results No infant was EBV-positive at 6 months (n. 52 tested). More than a third of infants (49/115 or 42.6 %) on study beyond 6 months seroconverted at 12 months. At 24 months, out of 66 tested infants, only 13 remained EBV-uninfected, while 53 (80.3 %) acquired EBV infection, rising the total proportion of EBV seroconversion to 88.7 % (102/115 infants). EBV seroconversion was significantly associated with a low maternal educational status but had no impact on infant growth or vulnerability to infections. Reduced HBsAb levels and accelerated waning of antibodies were associated with early EBV seroconversion. Conclusions We found a heterogeneous timing of acquisition of EBV with the majority of infants born from HIV + mothers acquiring infection after 6 months. Anti-HBs levels were lower and appeared to wane faster in infants acquiring EBV infection.

The use of dried goat rumen content (DGRC) as a partial replacement for fish meal in layer diets was investigated. A total of 90 H&N Brown Nick layer chickens were offered diets in which DGRC were incorporated at 0, 5 and 10% levels. Iso-caloric and nitrogenous diets were formulated to meet the recommended nutritional requirements for laying hens. Experimental birds were assigned to 9 cages (10 birds/cage) and experimental diets offered in a completely randomized design (CRD) with three replications.
