Abstract

High data quality is a precondition for valid scientific conclusions. Indicators should therefore routinely be used to evaluate data quality within the life cycle of health studies. In this project, 15 representatives of seven German population-based cohort studies assessed 51 quality indicators that were proposed in a guideline for networked medical research. The applicability of the indicators to primary data collections was assessed. In addition, their importance was evaluated using a scale ranging from 1 (essential) to 4 (not important). Moreover, their implementation in data quality assessments in the participating studies was evaluated. Comments on potential improvements could be made. Forty-three indicators were rated as applicable. Of these, 29 received a mean importance score of 2 (important) or better, nine received a mean importance score of 1.5 or better. The latter represent a potential core set of data quality indicators for cohort studies. Most indicators that were rated as highly important were used in data quality assessments of the participating studies. Points of criticism regarding the guideline related to its structure and the understandability of some indicators. It was concluded that further improvement of the data quality indicator set will increase its usefulness and applicability in primary data collections. In practice, a small subset of data quality indicators may suffice to capture the most important aspects of data quality in cohort studies.

Abstract

High data quality is fundamental for valid inferences in health research. Metadata, i.e. “data that describe other data”, are essential to implement data quality assessments but more guidance on which metadata to use is needed. Similarly, the selection and use of variables describing the measurement process should be exemplified to improve the design and conduct of observational health studies. This work provides a conceptual framework and overview of metadata and process information for systematic data quality reports based on implementations within the population-based cohort Study of Health in Pomerania (SHIP). In previous years, a prerequisite for automated data quality checks has been established by the augmentation of the data dictionary; the added information of up to 20 different characteristics for each variable is used for data quality assessments and triggers diverse data quality checks. Conceptually we distinguish static metadata, variable metadata, and process variables. Examples for static metadata are the expected probability distribution, plausibility limits, and the data type. Variable metadata may be reference limits of a laboratory marker. Information inherent to these metadata allows for the detection of data quality flaws by comparing observed with expected data characteristics. In contrast, process variables, such as the observer or device ID, also allow for the identification of sources of data quality issues. This is the case even if characteristics defined in metadata were not violated. Metadata and process variables can be used alone or in combination to implement a versatile and efficient data quality assessment. A comprehensive setup of metadata and process variables is an extensive task, particularly in studies involving large data collections. Nonetheless, the gain in transparency and efficacy of data curation and quality reporting after this setup is considerable.

Abstract

In view of the rise of drug-resistant tuberculosis and difficult-to-treat related diseases caused by non-tuberculous mycobacteria, there is an urgent need for antimycobacterial drug discovery. Nα-aroyl-N-aryl-phenylalanine amides (AAPs) have been identified as antimycobacterial agents and are subject to lead optimization. The aim of the present study is to evaluate the impact of N-aryl ortho cyano substitution in a lead compound on the crystal and molecular structure and its in vitro activity against Mycobacterium abscessus. The title AAP can be conveniently synthesized from N-Boc-protected d-phenylalanine in two amide coupling steps using a previously established racemization-free method. Two polymorphic forms in the solid-state are described, as discovered by X-ray and electron diffraction. The introduction of a cyano group in the ortho position of the AAP N-aryl ring, however, leads to loss of in vitro activity against M. abscessus subsp. abscessus. [ABSTRACT FROM AUTHOR]

19. Health insurance uptake, poverty and financial inclusion in the Democratic Republic of Congo.

Abstract

This study estimates the prevalence of health insurance coverage and associated socioeconomic factors in the Democratic Republic of Congo (DRC). Using the nationally representative household survey of the 2017/2018 DRC Multiple Indicator Cluster Survey (MICS), we applied weighted logistic regression models to identify regions and subgroups with low health insurance coverage. The study's findings revealed a low insurance coverage of less than 5%, with significant disparities across provinces and socioeconomic status. Additionally, three factors were strongly associated with the low health insurance coverage rates: education, wealth, and financial inclusion proxied by bank account ownership. Consequently, we recommend that the government, the private sector, and donors prioritize programs targeting provinces with less coverage and individuals without formal education to increase health insurance coverage. This study encourages the government to establish national programs to improve financial inclusion, which could positively impact poverty reduction and health insurance coverage. We also propose that the government initiates pilot projects for premium exemptions and through subsidies for vulnerable populations in the short term, and ensure formal employment for the majority of the population in the long term to facilitate the proper collection of premiums for individuals. Overall, this study contributes to the literature on health insurance in the DRC and sub‐Saharan Africa by identifying the socioeconomic factors that explain the prevalence of health insurance coverage. The findings of this study have important policy implications for the government, the private sector, and donors to promote health insurance coverage and achieve universal health coverage. Abstract

The title compound, 3‐[(benzo‐1,3‐dioxol‐5‐yl)amino]‐4‐methoxycyclobut‐3‐ene‐1,2‐dione, C12H9NO5 (3), is a precursor to an antimycobacterial squaramide. Block‐shaped crystals of a monoclinic form (3‐I, space group P21/c, Z = 8, Z′ = 2) and needle‐shaped crystals of a triclinic form (3‐II, space group P‐1, Z = 4, Z′ = 2) were found to crystallize concomitantly. In both crystal forms, R22(10) dimers assemble through N—H...O=C hydrogen bonds. These dimers are formed from crystallographically unique molecules in 3‐I, but exhibit crystallographic Ci symmetry in 3‐II. Twinning by pseudomerohedry was encountered in the crystals of 3‐II. The conformations of 3 in the solid forms 3‐I and 3‐II are different from one another but are similar for the unique molecules in each polymorph. Abstract

The study of insect morphology has recently benefited greatly from the emergence of new digital imaging and analysis technologies such as X-ray Microcomputed tomography (μ-CT), digital 3D reconstruction, and animation. Through interactive gaming and virtual reality, the external morphology of insects can be studied by a broad audience of both entomologists and non-specialists. EntomonVR is a serious game designed to investigate the external morphology of insects with adequate quality for the virtual reality platform. In this research, we discuss the advantages of virtual reality, introduce the EntomonVR new educational game, and conclude about future perspectives, validations, and cost-effectiveness. We tested this game on 25 participants with an entomological background for assessment and improved it based on their feedback. This study demonstrates the efficacy of virtual reality technology for an experimental learning environment in teaching the morphology of insects and the crucial need for advancing an efficient and interactive educational program. This study investigates the in vitro activity of Nα-aroyl-N-aryl-phenylalanine amides (AAPs), previously identified as antimycobacterial RNA polymerase (RNAP) inhibitors, against a panel of 25 non-tuberculous mycobacteria (NTM). The compounds, including the hit compound MMV688845, were selected based on their structural diversity and previously described activity against mycobacteria. Bacterial strains, including the M. abscessus complex, M. avium complex, and other clinically relevant NTM, were cultured and subjected to growth inhibition assays. The results demonstrate significant activity against the most common NTM pathogens from the M. abscessus and M. avium complexes. Variations in activity were observed against other NTM species, with for instance M. ulcerans displaying high susceptibility and M. xenopi and M. simiae resistance to AAPs. Comparative analysis of RNAP β and β′ subunits across mycobacterial species revealed strain-specific polymorphisms, providing insights into differential compound susceptibility. While conservation of target structures was observed, differences in compound activity suggested influences beyond drug–target interactions. This study highlights the potential of AAPs as effective antimycobacterial agents and emphasizes the complex interplay between compound structure, bacterial genetics, and in vitro activity. Abstract

Ants are a dominant family of eusocial terrestrial insects with a diversity of ecologies, lifestyles and morphologies. Ant diet preferences range from strict carnivory through omnivory to almost complete herbivory in species feeding on seeds or exudates of plant-sucking insects. While several studies have investigated ant feeding performance on different substrates, comparatively little is known about the functional morphology of the structures involved in food uptake or their diversification across the ants. To take stock of our current knowledge, we give an overview of how adult ants ingest food, followed by a morphological description of the mouthparts, preoral space and cephalic sucking pump. The mandibles are the most prominent mouthparts and have received considerable attention in the literature, so we focus on the maxillae and labium here. We present current hypotheses for the movement patterns of these parts and discuss morphological differences among ants that may be related to their ecological diversity. Finally, we give short comparisons of the ant condition with some other insects and vertebrates, as well as an outlook summarizing gaps in our knowledge. This sets the stage for future studies elucidating the connections between ant feeding mechanisms and mouthpart evolution. The title compound, C15H15F3N2O3S, crystallizes in the monoclinic system, space group I2/a, with Z = 8. As expected, the nine-membered heterobicyclic system is virtually planar and the cyclo­hexyl group adopts a chair conformation. There is structural evidence for intra­molecular N—SO chalcogen bonding between the benziso­thia­zolinone S atom and one O atom of the nitro group, approximately aligned along the extension of the covalent N—S bond [N—SO = 162.7 (1)°]. In the crystal, the mol­ecules form centrosymmetric dimers through C—HO weak hydrogen bonding between a C—H group of the electron-deficient benzene ring and the benzo­thia­zolinone carbonyl O atom with an R²2(10) motif. In contrast to the previously described N-acyl 7-nitro-5-(tri­fluoro­meth­yl)benzo[d]iso­thia­zol-3(2H)-ones, the title N-cyclo­hexyl­methyl analogue does not inhibit growth of Mycobacterium aurum and Mycobacterium smegmatis in vitro. Betaines of squaric acid have gained research interest because of their structural and spectral properties. We elucidated the crystal and molecular structure of the triethylammonium betaine of squaric acid (1) by X-ray crystallography, IR, and NMR spectroscopy augmented by Hirshfeld surface analysis and DFT calculations. The crystal structure determination using Hirshfeld atom refinement reveals that the resonance hybrid structure with partial enolate character of the two lateral squaric acid C=O groups describes 1 best. The solid-state supramolecular structure features weak intermolecular C−H···O hydrogen bonds. The number of C=O bands in the IR spectrum in the solid-state is consistent with local C2v symmetry of the squaric acid residue in 1. The 13C NMR signals of this group in solution were assigned based on 2D NMR experiments and computational prediction using the Gauge-Independent Atom Orbital (GIAO) method. Abstract

Trehalose conjugates of 3-hydroxychromone (3HC) dyes have previously been utilized as fluorescence labels to detect metabolically active mycobacteria with a view to facilitating point-of-care detection of mycobacterial pathogens, especially Mycobacterium tuberculosis. We subjected the 3HC dye 2-(6-(diethylamino)benzofuran-2-yl)-3-hydroxy-4H-chromen-4-one (3HC-2) to a combined X-ray crystallography and density functional theory (DFT) study, and conducted preliminary fluorescence labelling experiments with the model organism Mycobacterium aurum. In the crystal, 3HC-2 exhibits an s-cis conformation of the chromone and the benzofuran moieties about the central C–C bond. According to DFT calculations, the s-cis conformer is about 1.8 kcal mol−1 lower in energy than the s-trans conformer. The solid-state supramolecular structure features hydrogen-bonded dimers and π...π stacking. Fluorescence microscopy revealed fluorescence of M. aurum cells treated with the dye trehalose conjugate 3HC-2-Tre in the GFP channel. It was concluded that s-cis is the preferred conformation of 3HC-2 and that the generally considered non-pathogenic M. aurum can be labelled with the fluorescence probe 3HC-2-Tre for convenient in vitro drug screening of new antimycobacterial agents. Tuberculosis is the leading bacterial killer worldwide. 8-Nitro-4H-benzo[e][1,3]thiazin-4-ones are a potent class of antitubercular agents with a new mechanism of action. BTZ043 and PBTZ169 (macozinone) have progressed to clinical studies. Herein, we give a comprehensive account of this important class of potential new drugs to treat tuberculosis. We present an overview of recent developments in the field of antitubercular benzothiazinones (BTZs) and summarize our own contributions. The review covers synthesis, structures and reactivity, mechanism of action, in vitro activity and structure activity relationships (SARs), physicochemical and pharmacokinetic properties as well as a brief summary of in vivo models and clinical studies. We address bioavailability issues and the challenge of the potentially toxic nitroaromatic moiety, including reactivity towards nucleophiles in vivo and highlight possible directions of further research into BTZs through chemical modification. Abstract

For over a century, the metapleural gland, an exocrine gland above the hind coxa, has been thought to be a unique structure for ants (Hymenoptera: Formicidae), and regarded as a catalyst for the ecological and evolutionary success of the family. This gland is one of the most researched exocrine glands in arthropods and its anatomy, ultrastructure, and chemistry are well documented. Herein, we describe an exocrine gland from the proctotrupoid wasp Pelecinus polyturator (Hymenoptera: Pelecinidae) with a similar position, structure, and chemistry to the ant metapleural gland: it is located just above the hind coxa, corresponds to an externally concave and fenestrated atrium, is composed of class 3 gland cells, and its extract contains relatively strong acids. We discover that the pelecinid gland is associated with the dilator muscle of the first abdominal spiracle, a trait that is shared with ants, but remained overlooked, possibly due to its small diameter, or obfuscation by the extensive metapleural gland. We also provide a biomechanical argument for passive emptying of the gland in both taxa. Pelecinids and ants with metapleural glands share a close association with soil. The pelecinid metapleural gland might therefore also have an antiseptic function as suggested for ants. We examined 44 other Hymenoptera families and found no glands associated with the oclusor apodeme or any signs of external modification. Our results strongly indicate that this complex trait (anatomical & chemical) evolved independently in ants and pelecinid wasps providing an exceptional system to better understand exocrine gland evolution in Hymenoptera. However, reusable software tools from major studies, underlying such needs, are not necessarily known to other researchers. Therefore, this work gives an overview on the main tools used to conduct the internationally highly networked populationbased project Study of Health in Pomerania (SHIP), as well as approaches taken to improve its FAIRness. Deep phenotyping, formalizing processes from data capture to data transfer, with a strong emphasis on cooperation and data exchange have laid the foundation for a broad scientific impact with more than 1500 published papers to date. X-ray crystallography revealed the structure of the azoxy compound to be comprised of two benzothiazinone moieties linked by a Z-configured azoxy group in an almost coplanar arrangement. Cysteine plays a versatile role in cellular physiology and has previously been shown to be instrumental to Mycobacterium tuberculosis (M.tb) pathophysiology. In this study, we have generated mutants deficient in CysK 2 and CysH, the key Cysteine, biosynthetic enzymes. In contrast to the Δ cys H mutant, the Δ cysK 2 mutant is not an auxotroph and as such not essential for cysteine biosynthesis. Interestingly, the Δ cysK 2 mutant shows increased sensitivity to cumene hydroperoxide, vitamin C, diamide, rifampicin and Vancomycin and shows alterations in phospholipid profile of Mtb cell wall. Our findings suggest that alteration in phospholipids content of M.tb cell wall by CysK 2 may form a mode of defence against selected antibiotics and oxidative stress. • CysK 2 is not essential for cysteine biosynthesis. • Cysteine-deficiency in the Δ cysH mutant leads to thiols deficiency, however is restored by methionine supplementation. • CysK 2 is involved in the modulation of the cell wall lipid profile, mainly phospholipids. • The Δ cysK 2 mutant is sensitive to oxidative stress generated by vitamin C and Cumene hydroperoxide. • Modulation of cell wall phospholipids, is a potential defence mechanism by M.tb against oxidative stress. Abstract

The fossil record allows a unique glimpse into the evolutionary history of organisms living on Earth today. We discovered a specimen of the stem group ant † Gerontoformica gracilis (Barden and Grimaldi, 2014) in Kachin amber with near-complete preservation of internal head structures, which we document employing µ-computed-tomography-based 3D reconstructions. We compare † Gerontoformica to four outgroup taxa and four extant ant species, employing parsimony and Bayesian ancestral state reconstruction to identify morphological differences and similarities between stem and crown ants and thus improve our understanding of ant evolution through the lens of head anatomy. Of 149 morphological characters, 87 are new in this study, and almost all applicable to the fossil. † Gerontoformica gracilis shares shortened dorsal tentorial arms, basally angled pedicels, and the pharyngeal gland as apomorphies with other total clade Formicidae. Retained plesiomorphies include mandible shape and features of the prepharynx. Implications of the reconstructed transitions especially for the ant groundplan are critically discussed based on our restricted taxon sampling, emphasizing the crucial information derived from internal anatomy which is applied to deep time for the first time. Based on the falcate mandible in † Gerontoformica and other Aculeata, we present hypotheses for how the shovel-shaped mandibles in crown Formicidae could have evolved. Our results support the notion of † Gerontoformica as 'generalized' above-ground predator missing crucial novelties of crown ants which may have helped the latter survive the end-Cretaceous extinction. Abstract

It is generally assumed that Cretaceous stem ants were obligately eusocial, because of the presence of wingless adult females, yet the available evidence is ambiguous. Here, we report the syninclusion of a pupa and adult of a stem ant species from Mid-Cretaceous amber. As brood are immobile, the pupa was likely to have been transported by an adult. Therefore, the fossil substantiates the hypothesis that wingless females were cooperators, thus these were true 'workers'. Re-examination of all described Cretaceous ant species reveals that winged–wingless diphenism – hence a variable dispersal capacity – may have been ancestral to the total clade of the ants, and that highly specialized worker-specific phenotypes evolved in parallel between the stem and crown groups. The soft-tissue preservation of the fossil is exceptional, demonstrating the possibility of analysing the development of the internal anatomy in stem ants. Based on the highest-resolution µ-CT scans of stem ants to date, we describe † Gerontoformica sternorhabda sp. nov. , redescribe † G. gracilis , redefine the species group classification of † Gerontoformica , and provide a key to the species of the genus. Our work clarifies the species boundaries of † Gerontoformica and renders fossils relevant to the discussion of eusocial evolution in a way that has heretofore been intractable.
