Summary: Rutin, a flavonoid rich in buckwheat, is important for human health and plant resistance to external stresses. The hydrolysis of rutin to quercetin underlies the bitter taste of Tartary buckwheat. In order to identify rutin hydrolysis genes, a 200 genotypes mini‐core Tartary buckwheat germplasm resource was re‐sequenced with 30‐fold coverage depth. By combining the content of the intermediate metabolites of rutin metabolism with genome resequencing data, metabolite genome‐wide association analyses (GWAS) eventually identified a glycosyl hydrolase gene FtGH1, which could hydrolyse rutin to quercetin. This function was validated both in Tartary buckwheat overexpression hairy roots and in vitro enzyme activity assays. Mutation of the two key active sites, which were determined by molecular docking and experimentally verified via overexpression in hairy roots and transient expression in tobacco leaves, exhibited abnormal subcellular localization, suggesting functional changes. Sequence analysis revealed that mutation of the FtGH1 promoter in accessions of two haplotypes might be necessary for enzymatic activity. Co‐expression analysis and GWAS revealed that FtbHLH165 not only repressed FtGH1 expression, but also increased seed length. This work reveals a potential mechanism behind rutin metabolism, which should provide both theoretical support in the study of flavonoid metabolism and in the molecular breeding of Tartary buckwheat. Oats (Avena sativa L.) are a widely consumed functional cereal grain worldwide and have recently gained increasing attention due to their numerous health benefits. Oats are an excellent natural source of bioactive compounds, including β-glucan, and have broad application prospects for the development of functional foods or health products to prevent and treat of diabetes. In this review, oat bioactive compounds, including proteins, peptides, amino acids, β-glucans, resistant starch, dietary fibers, polyunsaturated fatty acids, vitamins, minerals, polyphenols, avenanthramides, oat saponins, and β-sitosterol are summarized, and their important biological functions are discussed, focusing on the relevant mechanisms of action. Due to their various bioactive ingredients, oats exhibit antioxidant, anti-diabetic, anti-microbial, anti-cancer, anti-hypertensive, immunomodulatory, anti-hyperlipidemic, anti-obesity, and cardioprotective properties. Sorghum (Sorghum bicolor (L.) Moench) is a raw material that can be used as food, feed, bioenergy, and wine; it is also a gramineous crop with drought, salt, waterlogging, and high temperature resistance. However, liquor-making sorghum faces the disadvantages of having a narrow genetic basis, poor resources, and few high-quality varieties. Ethyl methane sulfonate (EMS) is a common alkylating agent that can react with one or more bases to alkylate and cause changes in the molecular structure of DNA, thereby causing mutations. It has a minimum effect on organisms and the highest efficiency. The obtained mutant populations are of great significance for cultivating new plant varieties and enriching plant germplasm resources. Therefore, in this study, 'Hongyingzi' a liquor-making sorghum variety, was studied using seeds treated with 0.5% EMS and 415 M3 generation plants were obtained. (1) Investigation and statistical analysis of agronomic traits in mutant libraries showed that in the M3 generation, nine important phenotypic mutant lines were obtained, including plant type, leaf blade, spike, glume, growth period, fertility, plant height, and drought resistance. The variation frequency from high to low was as follows: glume color (75.42%) > spike type (54.70%) > spike shape (47.23%) > chaff coating degree (28%) > plant growth period (23.86%) > plant height (23.61%) > absorption degree (16.14%) > branchiness (10.84%) > leaf color (4.58%) > tillering (2.16%). (2) The PCR sequencing of SbGA2ox3 from 415 sorghum M3 plants revealed that the mutation frequency of SbGA2ox3 was 1/99.02 kb. Eight plants underwent mutations, but only one line experienced missense mutations of different amino acid types, changing Ser/Ala/Val/Leu/Gln/Ser/Pro/Ala to Asn/Thr/Gly/Val/Gln/Ala/Ser. The mutant line also had shorter plant height, reduced glume coating degree, and enhanced drought resistance. The constructions of the sorghum mutant library widened the sorghum germplasm library and provided a method for sorghum breeding with a molecular basis for the functional verification of related genes and the analysis of related regulatory networks. Essential trace elements are required at very low quantities in the human body but are essential for various physiological functions. Each trace element has a specific role and a lack of these elements can easily cause a threat to health and can be potentially fatal. In this study, inductively coupled plasma mass spectrometry (ICP-MS) and inductively coupled plasma atomic emission spectrometry (ICP-AES) were used to determine the content of trace metal elements Ca, Fe, Cu, Mg, Zn, Se, Mo, Mn, and Cd in buckwheat flour. The content and distribution characteristics of trace metal elements were investigated using principal component and cluster analysis. The principal component analysis yielded a four-factor model that explained 73.64% of the test data; the cumulative contribution of the variance of the 1st and 2nd principal factors amounted to 44.41% and showed that Cu, Mg, Mo, and Cd are the characteristic elements of buckwheat flour. The cluster analysis divided the 28 buckwheat samples into two groups, to some extent, reflecting the genuineness of buckwheat flour. ABFs play a key role in regulating plant osmotic stress. However, in Tartary buckwheat, data on the role of ABF genes in osmotic stress remain limited and its associated mechanism in osmoregulation remain nebulous. Herein, a novel ABF family in Tartary buckwheat, FtbZIP12, was cloned and characterized. FtbZIP12 is a transcriptional activator located in the nucleus; its expression is induced by NaCl, mannitol, and abscisic acid (ABA). Atopic expression of FtbZIP12 in Arabidopsis promoted seed germination, reduced damage to primary roots, and improved the tolerance of seedlings to osmotic stress. The quantitative realtime polymerase chain reaction (RT-qPCR) results showed that the expressions of the typical genes related to stress, the SOS pathway, and the proline synthesis pathway in Arabidopsis were significantly (p < 0.05) upregulated under osmotic stress. FtbZIP12 improved the osmotic pressure resistance by reducing the damage caused by reactive oxygen species to plants and maintained plant homeostasis by upregulating the expression of genes related to stress, osmotic regulation, and ion homeostasis. This study identified a key candidate gene for understanding the mechanism underlying osmotic-stress-regulated function in Tartary buckwheat, thereby providing a theoretical basis for improving its yield and quality. The transcription factor (TFs) BES1, which mediates brassinosteroid (BR) signaling, regulates plant growth and development. However, BES1 genes have not yet been reported in Tartary buckwheat. Here, ten FtBES1 genes were identified in the Tartary buckwheat genome, and they were named FtBES1-1 to FtBES1-10. These genes were divided into four groups according to the classification in Arabidopsis thaliana. Multiple sequence alignment indicated that all BES1 gene members contained the BES1_N structural domain. Phylogenetic relationship FtBES1 genes in the same group had similar gene structures and motifs. An analysis of cis-acting elements demonstrated that the BES1 TFs contains many light-responsive, hormonal, and abiotic stress-responsive elements, etc. The 10 FtBES1 genes were located on four chromosomes of Tartary buckwheat, and gene distribution and synteny analysis revealed that segmental duplications have played important roles in FtBES1 gene family expansion. Tissue specificity revealed that all of the ten FtBES1 members expressed highly in two periods, and relatively high expression levels were observed in mature leaves. Gene expression profiles under different hormone treatments demonstrated that FtBES1 gene family participated in the hormone stress response. This study enriches our knowledge of the Tartary buckwheat BES1 gene family and provides a theoretical basis for analyzing the biological functions and stress tolerance mechanisms of the Tartary buckwheat BES1 transcription factors. GATA is a transcription factor that exerts a vital function in plant growth and development, physiological metabolism, and environmental responses. However, the GATA gene family has rarely been studied in Tartary buckwheat since the completion of its genome. This study used bioinformatics methods to identify GATA genes of Tartary buckwheat and to analyze their subfamily classification, structural composition, and developmental evolution, as well as to discuss the expression patterns of FtGATA genes in different subfamilies. The twenty-eight identified FtGATA genes in the Tartary buckwheat genome were divided into four subfamilies and distributed on eight chromosomes. One pair of tandem repeat genes and eight pairs of fragments were found in chromosome mapping. Spatiotemporal expression patterns of eight FtGATA genes in different subfamilies indicated that the FtGATA gene family has regulatory roles in tissue specificity, fruit development, abiotic stress, and hormonal responses. We aimed to elucidate the physiological and biochemical mechanism by which exogenous hydrogen peroxide (H2O2) alleviates salt stress toxicity in Tartary buckwheat (Fagopyrum tataricum (L.) Gaertn). Tartary buckwheat "Chuanqiao-2" under 150 mmol·L−1 salt (NaCl) stress was treated with 5 or 10 mmol·L−1 H2O2, and seedling growth, physiology and biochemistry, and related gene expression were studied. Treatment with 5 mmol·L−1 H2O2 significantly increased plant height (PH), fresh and dry weights of shoots (SFWs/SDWs) and roots (RFWs/RDWs), leaf length (LL) and area (LA), and relative water content (LRWC); increased chlorophyll a (Chl a) and b (Chl b) contents; improved fluorescence parameters; enhanced antioxidant enzyme activity and content; and reduced malondialdehyde (MDA) content. Expressions of all stress-related and enzyme-related genes were up-regulated. The F3′H gene (flavonoid synthesis pathway) exhibited similar up-regulation under 10 mmol·L−1 H2O2 treatment. Correlation and principal component analyses showed that 5 mmol·L−1 H2O2 could significantly alleviate the toxic effect of salt stress on Tartary buckwheat. Our results show that exogenous 5 mmol·L−1 H2O2 can alleviate the inhibitory or toxic effects of 150 mmol·L−1 NaCl stress on Tartary buckwheat by promoting growth, enhancing photosynthesis, improving enzymatic reactions, reducing membrane lipid peroxidation, and inducing the expression of related genes. Summary: Golden buckwheat (Fagopyrum dibotrys or Fagopyrum cymosum) and Tartary buckwheat (Fagopyrum tataricum) belong to the Polygonaceae and the Fagopyrum genus is rich in flavonoids. Golden buckwheat is a wild relative of Tartary buckwheat, yet golden buckwheat is a traditional Chinese herbal medicine and Tartary buckwheat is a food crop. The genetic basis of adaptive divergence between these two buckwheats is poorly understood.Here, we assembled a high‐quality chromosome‐level genome of golden buckwheat and found a one‐to‐one syntenic relationship with the chromosomes of Tartary buckwheat. Two large inversions were identified that differentiate golden buckwheat and Tartary buckwheat.Metabolomic and genetic comparisons of golden buckwheat and Tartary buckwheat indicate an amplified copy number of FdCHI, FdF3H, FdDFR, and FdLAR gene families in golden buckwheat, and a parallel increase in medicinal flavonoid content. Resequencing of 34 wild golden buckwheat accessions across the two morphologically distinct ecotypes identified candidate genes, including FdMYB44 and FdCRF4, putatively involved in flavonoid accumulation and differentiation of plant architecture, respectively.Our comparative genomic study provides abundant genomic resources of genomic divergent variation to improve buckwheat with excellent nutritional and medicinal value. Arbuscularmycorrhizal fungi (AMF) are a class of beneficial microorganisms that are widely distributed in soil ecosystems and can form symbionts with 80% of terrestrial higher plants, and improve the nutritional status of plants. The use of AMF as a biocontrol method to antagonize soil-borne pathogens has received increasing interest from phytopathologists and ecologists. In this paper, the mechanisms of resistance to diseases induced by AMF and the application of AMF to plant fungal, bacterial, and nematode diseases have been summarized. This study aimed to enhance the potential use of AMF as a biological control method to prevent plant diseases in the future. Root morphological alteration characteristics were explained, including the influence of AMF on root structure, function, and the regulation of AMF via secondary metabolites. AMF can improve the rhizosphere environment by influencing the physical and chemical proprieties of soil, enhancing the growth of other beneficial microorganisms, and by competing with pathogenic microorganisms. Two microorganism types may compete for the same invasive sites in root systems and regulate nutrition distribution. AMF can induce the host plant to form defense systems, including improving phytohormone concentrations, inducing signal substrate production, gene expression regulation, and enhancing protein production. The nutritional value of Tartary buckwheat is much higher than that of common buckwheat. Tartary buckwheat is rich in nutrients, some of them unique, and has good medicinal properties. It also serves as a good feed crop. This review aims to provide an up-to-date and comprehensive literature on Tartary buckwheat's active substances (proteins, polyphenols, D-chiro-inositol, fat-soluble substances, resistant starch, dietary fiber, minerals and vitamins), medicinal value (e.g., prevention of AIDS, and prevention and treatment of cancer, and cardiovascular and cerebrovascular diseases) and nutritional effects (as an antioxidant, in lowering blood pressure, blood fat, and blood sugar, for detoxification and weight loss), etc., and research activities connected to Tartary buckwheat and its health effects in China and abroad. A novel chymotrypsin inhibitor, which detected in the seed of wild emmer wheat ( Triticum dicoccoides ), was purified by ion-exchange chromatography, affinity chromatography and Ultracentrifugation. On the basis of its specificity, this inhibitor was named WeCI (wild emmer chymotrypsin inhibitor). SDS-PAGE analysis displayed that the purified WeCI is a single chain polypeptide with a molecular weight of approximately 13 kDa. The inhibition constants (K i ) for amylase and bovine pancreatic chymotrypsin were 1.12 × 10 − 9 M and 2.41 × 10 − 9 M, respectively. Automated sequencing and mass spectrometry analyses revealed that WeCI is a neutral monomeric protein consisting of 119 residues. In vitro, WeCI strongly suppressed bovine pancreatic chymotrypsin as well as chymotrypsin-like activities separated from the midgut of the beet armyworm Spodoptera exigua. No inhibitory activities were found against bovine pancreatic trypsin, bacterial subtilisin, or porcine pancreatic elastase. The primary structure of WeCI was markedly similar (46–95%) to those of several proteins belonging to the wheat crop chymotrypsin/α-amylase inhibitor superfamily and displayed the typical sequence motif of the α-amylase inhibitor–seed storage protein group. WeCI significantly inhibited the growth and development of Spodoptera exigua , dependent on inhibitor concentration. Soil salinization is one of the main abiotic stress factors impacting the growth of crops and the agricultural industry today. Thus, we aimed to investigate the effects of H2O2 pretreatment on seed germination in Tartary buckwheat (Fagopyrum tataricum) seeds under salt stress and to evaluate this species' salt tolerance. Through the preliminary experiment, this study used 50 mmol L−1 NaCl solution to induce seed stress. After soaking for 12 h in different H2O2 concentrations, seeds were laid in Petri dishes with 50 mmol L−1 NaCl for seven days and the germination parameters and physiological indicators were measured to screen the optimal H2O2 pretreatment concentration and the salt tolerance index. Our results indicated that pretreatment with 5–10 mmol L−1 H2O2 was most effective in alleviating NaCl's impacts on the seeds' germination parameters. Furthermore, the growth and material accumulation of seedlings was promoted; catalase, superoxide dismutase activity, and proline content were enhanced; and malondialdehyde content was reduced. Principal component analysis and stepwise regression revealed six key indicators that had a significant impact on the salt tolerance characteristics of F. tataricum, namely, germination potential, shoot fresh weight, root surface area, root average diameter, catalase activity, and superoxide dismutase activity. Jasmonic acid (JA) and its precursors and dervatives, referred as jasmonates (JAs) are important molecules in the regulation of many physiological processes in plant growth and development, and especially the mediation of plant responses to biotic and abiotic stresses. JAs biosynthesis, perception, transport, signal transduction and action have been extensively investigated. In this review, we will discuss the initiation of JA signaling with a focus on environmental signal perception and transduction, JA biosynthesis and metabolism, transport of signaling molecules (local transmission, vascular bundle transmission, and airborne transportation), and biological function (JA signal receptors, regulated transcription factors, and biological processes involved). The GATA family of transcription factors is zinc finger DNA binding proteins involved in a variety of biological processes, including plant growth and development and response to biotic/abiotic stresses, and thus play an essential role in plant response to environmental changes. However, the GATA gene family of Sorghum ( SbGATA ) has not been systematically analyzed and reported yet. Herein, we used a variety of bioinformatics methods and quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) to explore the evolution and function of the 33 SbGATA genes identified. These SbGATA genes, distributed on 10 chromosomes, are classified into four subfamilies (I-IV) containing one pair of tandem duplications and nine pairs of segment duplications, which are more closely related to the monocot Brachypodium distachyon and Oryza sativa GATA genes. The physicochemical properties of the SbGATAs are significantly different among the subfamilies, while the protein structure and conserved protein motifs are highly conserved in the subfamilies. In addition, the transcription of SbGATAs is tissue-specific during Sorghum growth and development, which allows for functional diversity in response to stress and hormones. The second-largest transcription factor superfamily in plants is that of the basic helix-loop-helix (bHLH) family, which plays an important complex physiological role in plant growth, tissue development, and environmental adaptation. Systematic research on the Chenopodium quinoa bHLH family will enable a better understanding of this species. Herein, authors used a variety of bioinformatics methods and quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) to explore the evolution and function of the 218 CqbHLH genes identified. A total of 218 CqbHLH transcription factor genes were identified in the whole genome, located on 18 chromosomes. A phylogenetic tree was constructed using the CqbHLH and AtbHLH proteins to determine their homology, and the members were divided into 20 subgroups and one unclustered gene. Authors also analyzed 218 CqbHLH genes, conservative motifs, chromosome diffusion, and gene replication. The author constructed one Neighbor-Joining (NJ) tree and a collinearity analysis map of the bHLH family in C. quinoa and six other plant species to study the evolutionary relationship and homology among multiple species. In addition, the expression levels of 20 CqbHLH members from different subgroups in various tissues, different fruit developmental stages, and six abiotic stresses were analyzed. Trichoderma is mainly used to control soil-borne diseases as well as some leaf and panicle diseases of various plants. Trichoderma can not only prevent diseases but also promotes plant growth, improves nutrient utilization efficiency, enhances plant resistance, and improves agrochemical pollution environment. Trichoderma spp. also behaves as a safe, low-cost, effective, eco-friendly biocontrol agent for different crop species. In this study, we introduced the biological control mechanism of Trichoderma in plant fungal and nematode disease, including competition, antibiosis, antagonism, and mycoparasitism, as well as the mechanism of promoting plant growth and inducing plant systemic resistance between Trichoderma and plants, and expounded on the application and control effects of Trichoderma in the control of various plant fungal and nematode diseases. Tartary buckwheat (Fagopyrum tataricum, Polygonaceae) is an annual plant originating in Southwest China. It has a short growth cycle, barren soil tolerance, and strong stress resistance (Zhang et al. 2021). Because of its high content of proteins, starch, trace elements, phenols, and dietary fiber, Tartary buckwheat is beneficial to the human body and hence has received widespread attention (Joshi et al. 2019; Dc ja, B, et al. 2020). In the period from September to November 2020, a diseased plant infected with gray mold was found among M2 generation plants treated using ethyl methanesulfonate (EMS) in a location with potted Tartary buckwheat plants in Huaxi District, Guiyang City, Guizhou Province, China. The diseased plant started to show symptoms during the initial flowering stage; water-soaked spots appeared at first, that the spots increased in size and turned into light brown patches, with the leaf edges scorched brown. In severe cases, the leaves turned yellow, the diseased spots became dry, and finally the leaves necrotic (Figure 1A). Among the leaves that showed disease symptoms, severely susceptible leaves were selected; a piece of tissue (2×2 mm) was removed at the junction of the diseased and healthy tissues. The tissue was then soaked in 75% ethanol for 2 to 3 s, transferred to 1% sodium hypochlorite solution and soaked for 3 min, rinsed three times with sterile water, and placed on sterilized filter paper to dry. Sterile tweezers were used to transfer the tissue blocks to Potato Dextrose Agar medium (Bio-Rad Ltd. Com, USA) containing a Streptomyces-Penicillium mixture (100 μg/mL), and they were incubated on this medium for 7 to 10 days at 25°C and 70% humidity under 16 h light and 8 h dark conditions. The colonies were white at the early stages, with developed aerial hyphae; subsequently, they gradually turned gray-green (Figure 1B). In the later stages, the back of the colony was black and piles of conidia could be seen (Figure 1C). The conidia are scattered, which were colorless and transparent, fusiform or fusiform, with a size of 8.02-11.13 μm×2.06-3.22 μm (average=9.51 μm×2.69 μm, n=50) (Figure 1D). Based on their morphological characteristics, These cultural and morphological characteristics were consistent with the descriptions of as B. dothidea (Fan et al. 2021). The ITS1/ITS4 (Mills et al. 1992), Bt-2a/Bt-2b primers (Glass and Donaldson 1995), and EF1-728F/EF1-986R (Slippers et al. 2004) were amplified and sequenced to analyze the ITS region, β-tubulin genes translation elongation factor 1-α (TEF1-α), and translation elongation factor 1-α (TEF1-α), respectively. According to BLAST search in GenBank, the sequences of ITS (MZ326853), TUB2 (MZ399162) and TEF1-α (MZ399163) had 99.40%, 100% and 100% similarity to sequences NR111146.1, AY236927.1, and AY236898.1 of B. dothidea ex-type strain CMW8000, respectively. The three nucleotide sequences were concatenated together, and MEGA-X (with the neighbor-joining method) with 1,000 bootstraps was used to construct a phylogenetic tree. The results showed that our isolate was closely related to B. dothidea (Figure 2). Healthy Tartary buckwheat from the M2 generation was used for the pathogenicity test. Disinfect with 75% alcohol and 1×105 mL-1 of spore suspension was sprayed on the leaves. Each treatment included three plants, and it was repeated three times with sterile water as control. The treatments were kept in a houseat25°C for 24 h, then transferred it to the natural environment of 22℃ to 28℃,and sterile water was sprayed every morning and evening to keep the leaves moist. After 10 days, the symptoms seen in the field appeared on the treated plants (Figure 1E), but the control plants did not show any symptoms (Figure 1F). The diseased parts of the leaves were isolated and cultured again, and the isolates were consistent with the original inoculum. Thus, the study conformed to Koch's postulates. B. dothidea is a fungus with no host preference in the genus Botryosphaeria (Botryosphaeriaceae, Botryosphaeriales). It can cause canker, leaf spots, trunk diseases, fruit rot and die-back of many important wood plants all over the world (Marsberg et al.2017). Recently, it was reported that B. dothidea caused soybean canker in China (Chen et al.2021), but there have been no reports of B. dothidea causing Tartary buckwheat gray mold. To the best of our knowledge, this is the first report of B. dothidea causing gray mold on Tartary buckwheat. This finding will provide a basis for the prevention and treatment of Tartary buckwheat gray mold.
