Lung cancer is one of the most malignant tumors in the world. In China, the mortality rate of lung cancer has been in the first place for many years. Early screening and early diagnosis of lung cancer is the premise of prolonging the survival time of patients with lung cancer. In recent years, liquid biopsy technology, which is considered to have a bright future, has attracted more and more attention, and its value in the early diagnosis of lung cancer is worth discussing. This paper reviews the application of biomarkers in early screening and early diagnosis of lung cancer, looks for specific biomarkers from multi-omics, and discusses their significance in early diagnosis of lung cancer.

Ginsenoside Rh2 (Rh2), the major bio-active ginsenoside that originated from the root of Panax ginseng, has become a “hot topic” for playing multifunctional roles in both tumor treatment and tumor microenvironment (TME) immunomodulation. Up to now, emerging experimental research about Rh2 in tumor therapy and immuno-regulation has been published, however, the specific reviews focused on its role in the TME were limited. Hence, in this article, we briefly summarized existing evidence supporting the anticancer effects and potential mechanisms of Rh2 according to the tumor type, including anti-proliferation, anti-invasion, and metastasis, induction of cell cycle arrest, anti-angiogenesis, promotion of reactive oxygen species and differentiation. Meanwhile, we shed light on the role of Rh2 in TME, with an emphasis on regulating tumor immunogenicity, immune cells, cytokines, and physicochemical properties of TME. Finally, the current research limitation and prospects of Rh2 were discussed.

The fate of mesenchymal stem cells (MSCs) is regulated by biological, physical and chemical signals. Developments in biotechnology and materials science promoted the occurrence of bioactive materials which can provide physical and chemical signals for MSCs to regulate their fate. In order to design and synthesize materials that can precisely regulate the fate of MSCs, the relationship between the properties of materials and the fate of mesenchymal stem cells need to be clarified, in which the detection of the fate of mesenchymal stem cells plays an important role. In the past 30 years, a series of detection technologies have been developed to detect the fate of MSCs regulated by bioactive materials, among which high-throughput technology has shown great advantages due to its ability to detect large amounts of data at one time. In this review, the latest research progresses of detecting the fate of MSCs regulated by bone bioactive materials (BBMs) are systematically reviewed from traditional technology to high-throughput technology which is emphasized especially. Moreover, current problems and the future development direction of detection technologies of the MSCs fate regulated by BBMs are prospected. The aim of this review is to provide a detection technical framework for researchers to establish the relationship between the properties of BMMs and the fate of MSCs, so as to help researchers to design and synthesize BBMs better which can precisely regulate the fate of MSCs.

For practical engineering structures, fatigue is one of the main factors affecting their safety and durability. Under long-term service conditions, the minor damage will be affected by fatigue loading and expand to macroscopic cracks, affecting the structure's service performance. Based on the sensitivity of Lamb waves to minor and initial damage, a damage monitoring method for fatigue crack propagation is proposed. By carrying out fatigue crack propagation tests under constant amplitude loading, the Paris equation of 316L steel and damage signals at different crack growth stages were obtained. Combined with damage monitoring tests and finite element analysis, the relationship between the phase damage index (PDI), amplitude damage index (ADI), signal correlation coefficient, and fatigue crack propagation length was studied. Compared with PDI and ADI, the signal correlation coefficient is more sensitive to crack initiation, which can be selected as the damage monitoring index in the initial stage of crack growth. With the increase of fatigue crack propagation length, the peak time of the direct wave signal gradually moves backward, which shows an obvious phase change. In the whole fatigue crack growth stage, PDI and crack length show a monotonically changing trend. By using the stress intensity factor as the conversion parameter, a prediction model of the fatigue crack propagation rate based on PDI was established. Compared to the fatigue crack propagation rate measured by experiments, the relative error of the predicted results is 10%, which verifies the accuracy of the proposed damage monitoring method. While RNA therapeutics hold great promise for combating pancreatic ductal adenocarcinoma (PDAC), the limited therapy outcome due to the insufficient delivery efficiency of vehicles and inadequate innate therapeutic potency for single siRNA hinders the broader application. Herein, report bispecific, exosome‐mimetic lipid nanoparticles (EM‐LNPs) carrying high loads of siKras and siTP53, which can efficiently overcome layers of barriers to accumulate cytoplasm in vitro and in vivo and achieve synergistic treatment for PDAC tumors. EM‐LNPs inherit features of synthetic LNPs and the protein characteristics of exosomes with outer CD47 proteins, membrane‐inserted integrin α6β4, and connexin 43. The siRNAs‐loaded EM‐LNPs can efficiently evade the host immune clearance in the circulation, considerably accumulate and penetrate dense tumors, and simultaneously target EGFR/VEGF of PDAC cells, following leakage into the cytoplasm. The intracellular siKras and siTP53 achieved synergistic therapeutic effects for PDAC cells with as high as 20 times enhancement in vitro and 3.5 times increase in vivo, significantly suppressed xenograft PDAC tumors, considerably reduced lung metastasis, and vastly extended mice survival at a low injection dose. Bacterial infections pose a major concern for the medical community, especially regarding wound healing. Traditional passive antibiotic therapies can be cytotoxic and lead to bacterial resistance, posing a continuing challenge to treat. Based on precision therapy, a novel targeted‐delivery nanosystem is developed to efficiently eliminate bacteria and promote bacteria‐infected wound healing. Macrophage membranes pre‐activated by Staphylococcus aureus (MMSa) are prepared. In doing so, Toll‐like receptors (TLRs), a typical pathogen‐associated molecular pattern (PAMP), are significantly higher than normal macrophage membranes (MM0). Subsequently, MMSa are coated onto ultrasound‐triggered piezocatalytic nano‐barium titanate (BaTiO3, BTO) surfaces, and these two components are assembled to form a novel targeting delivery nanosystem, namely BTO@MMSa. The in vitro and in vivo results demonstrate that the biocompatible BTO@MMSa nanosystem can target infected areas and rapidly generate reactive oxygen species (ROS) to kill bacteria under ultrasound (US) irradiation, as well as accelerate wound healing. Furthermore, prokaryotic RNA‐seq transcriptomics reveals that changes in bacterial membrane function and substance, and energy metabolism are responsible for the targeted antibacterial ability of BTO@MMSa with US. Compared to widely reported unselective antibacterial agents, this novel targeted delivery nanosystem has potential for precise bacterial infection treatment by using macrophage membrane functions. Enlightened by the great success of the drug repurposing strategy in the pharmaceutical industry, in the current study, material repurposing is proposed where the performance of carbonyl iron powder (CIP), a nutritional intervention agent of iron supplement approved by the US FDA for iron deficiency anemia in clinic, was explored in anti-cancer treatment. Besides the abnormal iron metabolic characteristics of tumors, serving as potential targets for CIP-based cancer therapy under the repurposing paradigm, the efficacy of CIP as a catalyst in the Fenton reaction, activator for dihydroartemisinin (DHA), thus increasing the chemo-sensitivity of tumors, as well as a potent agent for NIR-II photothermal therapy (PTT) was fully evaluated in an injectable alginate hydrogel form. The CIP-ALG gel caused a rapid temperature rise in the tumor site under NIR-II laser irradiation, leading to complete ablation in the primary tumor. Further, this photothermal-ablation led to the significant release of ATP, and in the bilateral tumor model, both primary tumor ablation and inhibition of secondary tumor were observed simultaneously under the synergistic tumor treatment of nutritional-photothermal therapy (NT/PTT). Thus, material repurposing was confirmed by our pioneering trial and CIP-ALG-meditated NT/PTT/immunotherapy provides a new choice for safe and efficient tumor therapy. [ABSTRACT FROM AUTHOR]
