There is increasing evidence that energy metabolism is disturbed in Amyotrophic Lateral Sclerosis (ALS) patients and animal models. Treatment with triheptanoin, the triglyceride of heptanoate, is a promising approach to provide alternative fuel to improve oxidative phosphorylation and aid ATP generation. Heptanoate can be metabolized to propionyl-CoA, which after carboxylation can produce succinyl-CoA and thereby re-fill the tricarboxylic acid (TCA) cycle (anaplerosis). Here we tested the hypothesis that treatment with triheptanoin prevents motor neuron loss and delays the onset of disease symptoms in female mice overexpressing the mutant human SOD1G93A (hSOD1G93A) gene. When oral triheptanoin (35% of caloric content) was initiated at P35, motor neuron loss at 70 days of age was attenuated by 33%. Understanding the combining ability effects, heterosis, and genetic relationships between parental lines would be helpful in the maize breeding program to develop high-yielding and genetically stable maize varieties for various contrasting environments. Hence, the objective of this study was to estimate combining ability effects, heterosis, genetic distance (GD) and associations for grain yield and other agronomic traits. Forty-five F1 diallel crosses, four commercial checks, and ten inbred lines were evaluated at Bako and Jima Agricultural Research Centers, Ethiopia. Significant differences were observed among environments, genotypes and genotype by environment interaction for grain yield and almost all studied agronomic traits. Combining ability analysis showed both additive and non-additive gene effects significantly controlled grain yield and all other studied traits. The preponderance of general combining ability (GCA) effects indicates the importance of additive gene action inheriting most agronomic traits. Lines L2, L7, and L9 were the best combiners for grain yield, whereas lines L3 and L4 had desirable GCA values to improve days to flowering and plant height. Among the top ten crosses, L6 × L9, L6 × L7 and L4 × L9 were good specific combiners and had 18.8 %, 17.2 % and 16.2 % grain yield advantage over the best check, BH546. These hybrids also had high mid and better-parent heterosis compared with other crosses. The associations of GD with mean of F1 and SCA effects were positive and highly significant for grain yield and some other traits. In contrast, correlations of GD with mid and better parent heterosis were non-significant for grain yield and most other traits. The results of this study are particularly useful for breeders who envisage combining conventional and molecular methods.

Abstract Organismal functional strategies form a continuum from slow- to fast-growing organisms, in response to common drivers such as resource availability and disturbance. However, whether there is synchronisation of these strategies at the entire community level is unclear. Here, we combine trait data for >2800 above- and belowground taxa from 14 trophic guilds spanning a disturbance and resource availability gradient in German grasslands. The results indicate that most guilds consistently respond to these drivers through both direct and trophically mediated effects, resulting in a ‘slow-fast’ axis at the level of the entire community. Using 15 indicators of carbon and nutrient fluxes, biomass production and decomposition, we also show that fast trait communities are associated with faster rates of ecosystem functioning. These findings demonstrate that ‘slow’ and ‘fast’ strategies can be manifested at the level of whole communities, opening new avenues of ecosystem-level functional classification.

Uterine corpus endometrial carcinoma (UCEC) poses significant clinical challenges due to its high incidence and poor prognosis, exacerbated by the lack of effective screening methods. The standard treatment for UCEC typically involves surgical intervention, with radiation and chemotherapy as potential adjuvant therapies. In recent years, immunotherapy has emerged as a promising avenue for the advanced treatment of UCEC. This study employs a multi-omics approach, analyzing RNA-sequencing data and clinical information from The Cancer Genome Atlas (TCGA), Gene Expression Profiling Interactive Analysis (GEPIA), and GeneMANIA databases to investigate the prognostic value of MutL Homolog 1 (MLH1) gene expression in UCEC. The dysregulation of MLH1 in UCEC is linked to adverse prognostic outcomes and suppressed immune cell infiltration. Gene Set Enrichment Analysis (GSEA) data reveal MLH1’s involvement in immune-related processes, while its expression correlates with tumor mutational burden (TMB) and microsatellite instability (MSI). Lower MLH1 expression is associated with poorer prognosis, reduced responsiveness to Programmed cell death protein 1 (PD-1)/Programmed death-ligand 1 (PD-L1) inhibitors, and heightened sensitivity to anti-cancer agents. This comprehensive analysis establishes MLH1 as a potential biomarker for predicting prognosis, immunotherapy response, and drug sensitivity in UCEC, offering crucial insights for the clinical management of patients.

Background and Purpose: Amyotrophic lateral sclerosis (ALS), a neurodegenerative disease characterized by the degeneration of upper and lower motor neurons, progressive wasting and paralysis of voluntary muscles and is currently incurable. Although considered to be a pure motor neuron disease, increasing evidence indicates that the sole protection of motor neurons by a single targeted drug is not sufficient to improve the pathological phenotype. We therefore evaluated the therapeutic potential of the multi‐target drug used to treatment of coronary artery disease, trimetazidine, in SOD1G93A mice. Experimental Approach As a metabolic modulator, trimetazidine improves glucose metabolism. Furthermore, trimetazidine enhances mitochondrial metabolism and promotes nerve regeneration, exerting an anti‐inflammatory and antioxidant effect. We orally treated SOD1G93A mice with trimetazidine, solubilized in drinking water at a dose of 20 mg kg−1, from disease onset. We assessed the impact of trimetazidine on disease progression by studying metabolic parameters, grip strength and histological alterations in skeletal muscle, peripheral nerves and the spinal cord. Key Results: Trimetazidine administration delays motor function decline, improves muscle performance and metabolism, and significantly extends overall survival of SOD1G93A mice (increased median survival of 16 days and 12.5 days for male and female respectively). Moreover, trimetazidine prevents the degeneration of neuromuscular junctions, attenuates motor neuron loss and reduces neuroinflammation in the spinal cord and in peripheral nerves. Conclusion and Implications: In SOD1G93A mice, therapeutic effect of trimetazidine is underpinned by its action on mitochondrial function in skeletal muscle and spinal cord. [ABSTRACT FROM AUTHOR]

BackgroundDevelopmental delay is a public health problem in low- and middle-income countries. However, there is no summarized evidence in low- and middle-income countries on developmental delay, and primary studies on this issue show varied and inconclusive results. This systematic review and meta-analysis aimed to assess the pooled magnitude of confirmed developmental delay and its determinants among children in low- and middle-income countries.MethodsWe followed the Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) guidelines to write this systematic review and meta-analysis. Primary studies were searched from PubMed, PsycINFO, Hinari, Science Direct, African Journal of Online, Web of Science, and Google Scholar databases. The Newcastle–Ottawa Scale, adapted for the cross-sectional studies, was used to assess the quality of the included studies. Heterogeneity and publication bias were assessed by the I2 and Eggers tests, respectively. Due to the high heterogeneity, the random effects model was used for analysis. Odds ratios (ORs) with 95% confidence intervals (CIs) were used to show the association between developmental delay and its determinants.ResultsThe pooled prevalence of confirmed developmental delay was 18.83, 95% CI (15.53–22.12). In the subgroup analysis, a high prevalence of developmental delay [26.69% (95% CI, 15.78–37.60)] was observed in studies performed in Africa. Maternal education [3.04; 95% CI (2.05, 4.52)] and low birth weight [3.61; 95% CI (1.72, 7.57)] were significant determinants of developmental delay.ConclusionThe pooled prevalence of developmental delay in low- and middle-income countries was high as compared to that in high-income countries. Maternal education level and weight at birth were significantly associated with developmental delays. Therefore, strategies should be designed to decrease the rate of low birth weight and the number of illiterate mothers living in low- and middle-income countries.Systematic review registrationPROSPERO, CRD42024513060.

Corruption is a global problem that affects many countries by destroying economic, social, and political development. Therefore, we have formulated and analyzed a mathematical model to understand better control measures that reduce corruption transmission with optimal control strategies. To verify the validity of this model, we examined a model analysis showing that the solution of the model is positive and bounded. The basic reproduction number R0 was computed by using the next-generation matrix. The formulated model was studied analytically and numerically in the context of corruption dynamics. The stability analysis of the formulated model showed that the corruption-free equilibrium is locally and globally asymptotically stable for R0 < 1, but the corruption-endemic equilibrium is globally asymptotically stable for R0 > 1. Furthermore, the optimal control strategy was expressed through the Pontryagin Maximum Principle by incorporating two control variables. Finally, numerical simulations for the optimal control model were performed using the Runge-Kutta fourth order forward and backward methods. This study showed that applying both mass education and law enforcement is the most efficient strategy to reduce the spread of corruption.

Summary: Invasive earthworms colonize ecosystems around the globe. Compared to other species’ invasions, earthworm invasions have received little attention. Previous studies indicated their tremendous effects on resident soil biota representing a major part of the terrestrial biodiversity. We investigated effects of earthworm invasion on soil microbial communities in three forests in North America by conducting DNA sequencing of soil bacteria, fungi, and protists in two soil depths. Our study shows that microbial diversity was lower in highly invaded forest areas. While bacterial diversity was strongly affected compared to fungi and protists, fungal community composition and family dominance were strongly affected compared to bacteria and protists. We found most species specialized on invasion in fungi, mainly represented by saprotrophs. Comparably, few protist species, mostly bacterivorous, were specialized on invasion. As one of the first observational studies, we investigated earthworm invasion on three kingdoms showing distinct taxa- and trophic level-specific responses to earthworm invasion.

This paper aims to glean valuable insights from critical pedagogy in order to apply them to the field of African literary studies. Specifically, I am interested in how approaches inspired by culturally responsive education can help us revisit the important but under-researched topic of student agency as it features in fictional works that deal with colonial education in Africa. Although colonial education in literature has been the subject of intense focus in postcolonial theory, such theorizations largely examine how colonial education reproduces colonial rule through the dissemination of colonial discourse/ideology. When student agency in the colonial education systems is addressed, conventional postcolonial theory sees it as being overwhelmed or assimilated by colonial discourse and power. Ideas emanating from critical pedagogy and culturally responsive education are of value here in that they can elucidate how students interact with and even resist the pedagogical and political power of the colonial education system. Ngũgĩ wa Thiong’o’s novel Weep Not, Child is an illustrative example of how such a critical-pedagogy-inspired approach can help us reorient literary studies of colonial education systems.

AbstractEfficient conservation and subsequent utilization of genetic resources are primarily dependent on the strength in the assessment of variation among genotypes. An experiment was carried out aiming at determining the extent of phenotypic variability present in a panel of 320 barley genotypes and identifying candidate lines for further evaluation in improvement programs and successive utilization. It was conducted at two locations in Ethiopia, Aris Negelle and Holetta in the 2017/18 and 2018/19 cropping seasons. Among the best 30 lines for grain yield across all the environments, lines from farmers’ varieties constitute 73% and lines that mature in less than 85 days were identified. Based on the spike row number, the best-performing lines combined across all the environments were six-rowed types. Based on the two years’ data at Arsi Negelle the two-rowed spike type dominates, and at Holetta the six-rowed type. After principal component analysis, the first three PCs with an eigenvalue greater than one explained 70% of the variation. The correlation coefficient between grain and biomass yield was significant and though low (r = 0.38***). Significant, high, and negative correlation coefficient (−0.72***) was observed between 1000 kernel weight and the number of seeds per spike. A positive correlation between biomass and grain yield attracts farmers as a feed and food crop as it has also been signified in the current research. Having the improved barley gene pool largely from international sources, combining the improved materials with farmers’ varieties may minimize the existing gap between the local and improved barley gene pool.

Abstract Background Evaluation of the extent of genetic variation within and between the populations of crop genetic resources are of paramount importance in any breeding program. An experiment aimed at assessing the extent of variation among barley lines and the degree of association between hordein polypeptide and agronomic traits was hence executed. Methods Field experiment was conducted in six environments between 2017–2019 involving 19 barley lines. Hordein bands were separated using vertical Sodium Dodecyl Sulphate Poly- acrylamide Gel Electrophoresis (SDS-PAGE). Results The analysis of variance revealed significant variation among lines and wider range units were observed for the agronomic traits. The line (Acc# 16,811–6) was superior, producing the highest grain yield (2.97 ton ha−1) across environments, 3.6 ton ha−1 at Holleta, and 1.93 ton ha−1 at Chefedonsa. At Arsi Negelle a different line Acc# 17146–9 was the highest yielding (3.15ton ha−1). SDS-PAGE-based analysis of barley lines separated 12 hordein bands between C (four bands) and B (eight bands) subunits. Interestingly bands 52, 46a, and 46b were uniquely conserved in the four naked barley lines (Acc#16809–14,16956–11, 17240–3, 17244–19). A considerably high proportion of genetic diversity within the populations than among the populations could be a repercussion of high gene flow which substantiates the longstanding and dominant informal seed exchange system among the farmers. The significant positive association between grain yield and band 50 evocates the expression of this allele may code for higher grain yield. The negative association between days to maturity and band 52 perhaps stipulates earliness in barely lines upon the manifestation of the band. Band 52 and 60 appeared to be associated with more than one agronomic trait (days to maturity and thousand kernel weight; grain filling period and grain yield respectively) and could be the result of pleiotropic characteristics of the genes residing in these banding regions. Conclusion The barley lines exhibited substantial variation for hordein protein and agronomic traits. However, imparted the need for the implementation of decentralized breeding as a consequence of genotype-by-environment interaction. Significant hordein polypeptide and agronomic traits association advocated the utilization of hordein as a protein marker and perhaps consider them in the parental line selection.

Violence against women and girls is a major public problem globally. This study had conducted to assess women and girls safety and security in public transport and identify factors contributing to violence in Hawassa city, Ethiopia. Data used in the study have been obtained from 199 randomly selected respondents (36.7% women and 63.3% girls), and in addition, 24 focus group discussion members have also participated. Binary Logistic Regression had employed for quantitative data analysis. Data from focus group discussions were analyzed qualitatively using narrative analysis. The types of violence identified were physical, sexual, and psychological. The finding shows that 50.8% of women and girls have experienced more than one type of violence while using public transport. The results indicated that six variables significantly contributed to violence. These variables were age, marital status, type of public transport used, travel time, facilities, and management of public transport services. The result indicates that violence against women and girls in public transport is a serious threat to women’s safety and security to access their mobility in the city. Therefore, cities must focus on developing gender-sensitive public transport service plans and policies that consider women and girls unique needs in public transportation.

Abstract This paper aims to examine the major determinants and challenges of women's participation in income-generating activities focusing on rural women of Ethiopia. To carry out this study, both primary and secondary data were used. Randomly selected 161 households were the source of primary data for this study. Secondary data were collected from the review of related literature. A binary logistic regression econometric model was implemented to identify major determinants of women's participation in income-generating activities. The finding of this study revealed that in the study area, women are not allowed by their husbands to participate in high-income earning activities. They are considered a housewife and only husband are expected to participate in high income-generating activities due to the local customs. In the study area, women's participation in the income-generating activity is determined by age, husband’s education, women's education, family size, land size, market distance, livestock holding, and access to credit. This paper contributes to the literature on women's participation challenges in income-generating activities, giving emphasis to rural women's perspectives. It provides the basis for further studies aimed at challenges hindering women's participation in high-income earning activities, particularly in developing countries.

Maize is one of the most important staple food crops for most low-income households in the Southern African region. Erratic and inconsistent rainfall distribution across maize-growing areas is a major threat to maize production. Late rains in recent years have forced farmers to plant later than the optimal planting dates, leading to poor maize quality being reported by industry, which raised the question of the influence of later planting dates on grain yield and quality traits of maize. Three yellow and three white maize hybrids were evaluated at three planting dates in three different production environments for three consecutive seasons using a randomized complete block design with three replications. The second and third planting dates caused a significant yield decrease of 23.37 % and 53.73 % from the first planting date across environments, respectively. Planting date three was associated with decreased grain yield, starch content, and increased protein but no significant change in fat and fiber content. Some hybrids yielded relatively well at all planting dates. In conclusion, the early planting date was the most suitable for maize grain yield and starch production in the maize-growing areas of the country. However, planting in January should be avoided at all costs, as it leads to very low yield and poor grain quality.

Although alterations in energy metabolism are known in ALS, the specific mechanisms leading to energy deficit are not understood. We measured metabolite levels derived from injected [1-13C]glucose and [1,2-13C]acetate (i.p.) in cerebral cortex and spinal cord extracts of wild type and hSOD1G93A mice at onset and mid disease stages using high-pressure liquid chromatography, 1H and 13C nuclear magnetic resonance spectroscopy. Levels of spinal and cortical CNS total lactate, [3-13C]lactate, total alanine and [3-13C]alanine, but not cortical glucose and [1-13C]glucose, were reduced mostly at mid stage indicating impaired glycolysis. The [1-13C]glucose-derived [4-13C]glutamate, [4-13C]glutamine and [2-13C]GABA amounts were diminished at mid stage in cortex and both time points in spinal cord, suggesting decreased [3-13C]pyruvate entry into the TCA cycle. Lack of changes in [1,2-13C]acetate-derived [4,5-13C]glutamate, [4,5-13C]glutamine and [1,2-13C]GABA levels indicate unchanged astrocytic 13C-acetate metabolism. Reduced levels of leucine, isoleucine and valine in CNS suggest compensatory breakdown to refill TCA cycle intermediate levels. Unlabelled, [2-13C] and [4-13C]GABA concentrations were decreased in spinal cord indicating that impaired glucose metabolism contributes to hyperexcitability and supporting the use of treatments which increase GABA amounts. In conclusion, CNS glucose metabolism is compromised, while astrocytic TCA cycling appears to be normal in the hSOD1G93A mouse model at symptomatic disease stages. [ABSTRACT FROM AUTHOR]

ABSTRACT Soil microbial communities play crucial roles in the earth’s biogeochemical cycles. Yet, their genomic potential for nutrient cycling in association with tree mycorrhizal type and tree-tree interactions remained unclear, especially in diverse tree communities. Here, we studied the genomic potential of soil fungi and bacteria with arbuscular (AM) and ectomycorrhizal (EcM) conspecific tree species pairs (TSPs) at three tree diversity levels in a subtropical tree diversity experiment (BEF-China). The soil fungi and bacteria of the TSPs’ interaction zone were characterized by amplicon sequencing, and their subcommunities were determined using a microbial interkingdom co-occurrence network approach. Their potential genomic functions were predicted with regard to the three major nutrients carbon (C), nitrogen (N), and phosphorus (P) and their combinations. We found the microbial subcommunities that were significantly responding to different soil characteristics. The tree mycorrhizal type significantly influenced the functional composition of these co-occurring subcommunities in monospecific stands and two-tree-species mixtures but not in mixtures with more than three tree species (here multi-tree-species mixtures). Differentiation of subcommunities was driven by differentially abundant taxa producing different sets of nutrient cycling enzymes across the tree diversity levels, predominantly enzymes of the P (n = 11 and 16) cycles, followed by the N (n = 9) and C (n = 9) cycles, in monospecific stands and two-tree-species mixtures, respectively. Fungi of the Agaricomycetes, Sordariomycetes, Eurotiomycetes, and Leotiomycetes and bacteria of the Verrucomicrobia, Acidobacteria, Alphaproteobacteria, and Actinobacteria were the major differential contributors (48% to 62%) to the nutrient cycling functional abundances of soil microbial communities across tree diversity levels. Our study demonstrated the versatility and significance of microbial subcommunities in different soil nutrient cycling processes of forest ecosystems. IMPORTANCE Loss of multifunctional microbial communities can negatively affect ecosystem services, especially forest soil nutrient cycling. Therefore, exploration of the genomic potential of soil microbial communities, particularly their constituting subcommunities and taxa for nutrient cycling, is vital to get an in-depth mechanistic understanding for better management of forest soil ecosystems. This study revealed soil microbes with rich nutrient cycling potential, organized in subcommunities that are functionally resilient and abundant. Such microbial communities mainly found in multi-tree-species mixtures associated with different mycorrhizal partners can foster soil microbiome stability. A stable and functionally rich soil microbiome is involved in the cycling of nutrients, such as carbon, nitrogen, and phosphorus, and their combinations could have positive effects on ecosystem functioning, including increased forest productivity. The new findings could be highly relevant for afforestation and reforestation regimes, notably in the face of growing deforestation and global warming scenarios.

Iron (Fe) and zinc (Zn) nutrient enrichment of staple crops through biofortification can contribute to alleviating micronutrient deficiency in sub-Saharan Africa. A line × tester mating design was used to determine the general combining ability (GCA), specific combining ability (SCA) and heterosis for grain yield, iron, Zn and phytic concentration of six lines crossed with three testers. Lines and testers were selected for high, intermediate and low mineral content. The F1 hybrids and parental lines were evaluated under low nitrogen (N) and optimum conditions across four environments over two seasons. Under low N conditions, Fe and Zn concentration in grain, and grain yield of genotypes were reduced by 9%, 9%, and 59%, respectively. However, phytic acid concentration in grain was increased by 10% under low N conditions. Both additive and non-additive gene effects were important in controlling Fe, Zn and phytic acid concentration in grain and grain yield of maize under both N conditions. The preponderance of GCA effects indicates the importance of additive gene effects in the inheritance of grain yield. Line GCA effects were more sensitive to N conditions across the environments than the tester GCA. High and significant positive SCA effects for grain yield, Fe and Zn content under low N conditions, would be a good indicator of possible heterosis in these traits. Hybrid CBY101 LM-1600 × CBY358 LM-1857 had high and significant positive SCA for grain yield under low N conditions and is a promising candidate for production in low N environments. CBY358 LM-1857 (tester) and CBY102 LM-1601 (line) are a good general combiners for Fe, Zn and GY can be used as parents in future maize hybrid breeding programs to develop high-yielding maize genotypes with high Fe and Zn content.

A study was conducted to determine the prevalence of bovine trypanosomosis in rainy (June 2019) and dry (February 2020) seasons in Jimma Horoo district, Kellem Wollega Zone, Oromia Regional State, Ethiopia. A total of 720 blood samples were examined using buffy coat and thin blood smear techniques. The packed cell volume (PCV) of each animal was determined. The overall bovine trypanosomosis prevalence was 4.3% (χ2 = 1.25, P = 0.26). The prevalence was 5% (95% CI = 4.1–8.3) and 3.3% (95% CI = 2.7–6.3) in the dry and rainy reasons, respectively (P > 0.05). Trypanosoma congolense, T. vivax and T. b. brucei were detected in (60%), (33.3%) and (6.7%) of infected cattle, respectively. The highest trypanosome prevalence was observed in Burka Gudina (7.6%), and the lowest in Melka Nega village (2.1%). There were significant variations between trypanosome prevalence in relation to body condition scores (χ2 = 23.16; P = 0.0.00) of examined cattle. No significant difference (P > 0.05) was observed between seasons, age, and sex categories of cattle. Climate change has a devastating effect on wheat production; therefore, crop production might decline by 2030. Phosphorus (P) nutrient deficiency is another main limiting factor of reduced yield. Hence, there is a dire need to judiciously consider wheat yield, so that human requirements and nutrition balance can be sustained efficiently. Despite the great significance of biostimulants in sustainable agriculture, there is still a lack of integrated technology encompassing the successful competitiveness of inoculated phosphate-solubilizing bacteria (PSB) in agricultural systems in the context of climatic conditions/meteorological factors and soil nutritional status. Therefore, the present study reveals the modulation of an integrated P nutrient management approach to develop potential PSB consortia for recommended wheat varieties by considering the respective soil health and agro-climatic conditions. The designed consortia were found to maintain adequate viability for up to 9 months, verified through field emission scanning electron microscopy and viable count. Furthermore, a significant increase in grain yield (5%–8%) and seed P (4%) content was observed in consortia-inoculated wheat plants with 20% reduced Diammonium phosphate (DAP) application under net house conditions. Fluorescence in situ hybridization analysis of roots and amplification of the gcd gene of Ochrobactrum sp. SSR indicated the survival and rhizosphere competency of the inoculated PSB. Categorical principal component analysis (CAT-PCA) showed a positive correlation of inoculated field-grown wheat varieties in native soils to grain yield, soil P content, and precipitation for sites belonging to irrigated plains and seed P content, soil organic matter, and number of tillers for sites belonging to Northern dry mountains. However, the impact of inoculation at sites belonging to the Indus delta was found significantly correlated to soil potassium (K) content, electrical conductivity (EC), and temperature. Additionally, a significant increase in grain yield (15%) and seed P (14%) content was observed in inoculated wheat plants. Thus, the present study demonstrates for the first time the need to integrate soil biological health and agro-climatic conditions for consistent performance of augmented PSB and enhanced P nutrient uptake to curtail soil pollution caused by the extensive use of agrochemicals. This study provides innovative insights and identifies key questions for future research on PSB to promote its successful implementation in agriculture.

Nitrogen deficiency is the most limiting abiotic stress factor affecting the grain yield of cowpea (Vigna unguiculata L.) in sub-Saharan Africa (SSA) countries. Breeding for high-yielding potential in association with high nitrogen fixation performance is the principal objective of cowpea breeding programs to improve both the productivity and production of this orphan crop in the region. Therefore, the present study was undertaken to determine the diversity of genotypes for nodulation capacity in two environments and to understand the root architecture and morphology of genotypes as well as grain yield and yield-related traits in 324 cowpea genotypes. The experiment was conducted in two different environments during the 2017 cropping season. The experiments were laid out in 18 × 18 simple lattice design. Grain yield exhibited highly significant and positive genotypic correlations with stem diameter, basal root length of 1st whorls, basal root branching density, taproot length and adventitious root length, demonstrating that selection of cowpea genotypes based on these traits could be effective to capitalize on grain yield under low nitrogen conditions. Days to flowering, days to maturity, stem diameter, 1st whorl angle, basal root length of 1st whorls, basal root branching density, taproot length, adventitious root diameter and adventitious root length had significant and positive genotypic correlations with days to maturity. The first ten principal components (PC) explained 65.13% of the total variation. Stem diameter, taproot width, and taproot length traits were important contributors to the variability in the first PC. The highest inter-cluster distance (D2) was recorded between clusters III and IV. However, clusters II and V recorded the minimum inter-cluster distance (78.96 units). The range of intra-cluster distance was 24.22–5,112.92 units, indicating that the high genetic distance displayed within and between clusters has to be exploited via crossing and selecting the most divergent parents for future cowpea improvement. Five clusters of cowpea genotypes were evident, and within the clusters, the genotypes had good nodulation potential with high grain yield traits, which could significantly contribute to SSA food and nutritional security. Moreover, it can contribute to resilience and improve crop production and sustainability under marginal environmental conditions.

Abstract Herbivorous insects acquire microorganisms from host plants or soil, but it remains unclear how the diversity and functional composition of host plants contribute to structuring herbivore microbiomes. Within a controlled tree diversity setting, we used DNA metabarcoding of 16S rRNA to assess the contribution of Lepidoptera species and their local environment (particularly, tree diversity, host tree species, and leaf traits) to the composition of associated bacterial communities. In total, we obtained 7,909 bacterial OTUs from 634 caterpillar individuals comprising 146 species. Tree diversity was found to drive the diversity of caterpillar‐associated bacteria both directly and indirectly via effects on caterpillar communities, and tree diversity was a stronger predictor of bacterial diversity than diversity of caterpillars. Leaf toughness and dry matter content were important traits of the host plant determining bacterial species composition, while leaf calcium and potassium concentration influenced bacterial richness. Our study reveals previously unknown linkages between trees and their characteristics, herbivore insects, and their associated microbes, which contributes to developing a more nuanced understanding of functional dependencies between herbivores and their environment, and has implications for the consequences of plant diversity loss for trophic interactions.

Abstract Amyotrophic lateral sclerosis (ALS) is a fatal progressive neurodegenerative disorder primarily characterized by selective degeneration of both the upper motor neurons in the brain and lower motor neurons in the brain stem and the spinal cord. The exact mechanism for the selective death of neurons is unknown. A growing body of evidence demonstrates abnormalities in energy metabolism at the cellular and whole-body level in animal models and in people living with ALS. Many patients with ALS exhibit metabolic changes such as hypermetabolism and body weight loss. Despite these whole-body metabolic changes being observed in patients with ALS, the origin of metabolic dysregulation remains to be fully elucidated. A number of pre-clinical studies indicate that underlying bioenergetic impairments at the cellular level may contribute to metabolic dysfunctions in ALS. In particular, defects in CNS glucose transport and metabolism appear to lead to reduced mitochondrial energy generation and increased oxidative stress, which seem to contribute to disease progression in ALS. Here, we review the current knowledge and understanding regarding dysfunctions in CNS glucose metabolism in ALS focusing on metabolic impairments in glucose transport, glycolysis, pentose phosphate pathway, TCA cycle and oxidative phosphorylation. We also summarize disturbances found in glycogen metabolism and neuroglial metabolic interactions. Finally, we discuss options for future investigations into how metabolic impairments can be modified to slow disease progression in ALS. These investigations are imperative for understanding the underlying causes of metabolic dysfunction and subsequent neurodegeneration, and to also reveal new therapeutic strategies in ALS.

Abstract Background Forests perform various important ecosystem functions that contribute to ecosystem services. In many parts of the world, forest management has shifted from a focus on timber production to multi-purpose forestry, combining timber production with the supply of other forest ecosystem services. However, it is unclear which forest types provide which ecosystem services and to what extent forests primarily managed for timber already supply multiple ecosystem services. Based on a comprehensive dataset collected across 150 forest plots in three regions differing in management intensity and species composition, we develop models to predict the potential supply of 13 ecosystem services. We use those models to assess the level of multifunctionality of managed forests at the national level using national forest inventory data. Results Looking at the potential supply of ecosystem services, we found trade-offs (e.g. between both bark beetle control or dung decomposition and both productivity or soil carbon stocks) as well as synergies (e.g. for temperature regulation, carbon storage and culturally interesting plants) across the 53 most dominant forest types in Germany. No single forest type provided all ecosystem services equally. Some ecosystem services showed comparable levels across forest types (e.g. decomposition or richness of saprotrophs), while others varied strongly, depending on forest structural attributes (e.g. phosphorous availability or cover of edible plants) or tree species composition (e.g. potential nitrification activity). Variability in potential supply of ecosystem services was only to a lesser extent driven by environmental conditions. However, the geographic variation in ecosystem function supply across Germany was closely linked with the distribution of main tree species. Conclusions Our results show that forest multifunctionality is limited to subsets of ecosystem services. The importance of tree species composition highlights that a lack of multifunctionality at the stand level can be compensated by managing forests at the landscape level, when stands of complementary forest types are combined. These results imply that multi-purpose forestry should be based on a variety of forest types requiring coordinated planning across larger spatial scales.

Abstract Land management is known to have consequences for biodiversity; however, our synthetic understanding of its effects is limited due to highly variable results across studies, which vary in the focal taxa and spatial grain considered, as well as the response variables reported. Such synthetic knowledge is necessary for management of agroecosystems for high diversity and function. To fill this knowledge gap, we investigated the importance of scale‐dependent effects of land management (LM) (pastures vs. meadows), on plant and soil microbe diversity (fungi and bacteria) across 5 study sites in Central Germany. Analyses included diversity partitioning of species richness and related biodiversity components (i.e., density of individuals, species‐abundance distribution, and spatial aggregation) at two spatial grains (α‐ and γ‐scale, 1 m2 and 16 km2, respectively). Our results show scale‐dependent patterns in response to LM to be the norm rather than the exception and highlight the importance of measuring species richness and its underlying components at multiple spatial grains. Our outcomes provide new insight to the complexity of scale‐dependent responses within and across taxonomic groups. They suggest that, despite close associations between taxa, LM responses are not easily extrapolated across multiple spatial grains and taxa. Responses of biodiversity to LM are often driven by changes to evenness and spatial aggregation, rather than by changes in individual density. High‐site specificity of LM effects might be due to a variety of context‐specific factors, such as historic land management, identity of grazers, and grazing regime. Synthesis and applications: Our results suggest that links between taxa are not necessarily strong enough to allow for generalization of biodiversity patterns. These findings highlight the importance of considering multiple taxa and spatial grains when investigating LM responses, while promoting management practices that do the same and are tailored to local and regional conditions.

Soil organism biodiversity contributes to ecosystem function, but biodiversity and function have not been equivalently studied across the globe. Here the authors identify locations, environment types, and taxonomic groups for which there is currently a lack of biodiversity and ecosystem function data in the existing literature.

Maize is the staple food crop for millions of people in sub-Saharan Africa. Iron (Fe) and zinc (Zn) deficiency is a significant health risk that mainly affects low-income populations who rely solely on maize-based diets. This problem can be alleviated by developing micronutrient-rich maize grain. The aim of this study was to determine the adaptation and performance of hybrids for Fe and Zn concentration and grain yield under low soil nitrogen (N) and optimal conditions. Eighteen hybrids derived from lines and testers with low, medium, and high Fe and Zn concentration were grown during the summer rainy seasons of 2017 and 2018 at three locations under low and optimal N conditions. There were significant genotype and environment effects for grain yield, and Fe and Zn concentration, but the genotype by environment interaction effects were the largest, accounting for between 36% and 56% of variation under low N conditions. Low N levels significantly reduced grain yield, and Fe and Zn concentration. Hybrids G1, G2, G4, G7, G10, G11, and G16 were relatively stable, with relatively high mean Fe and Zn concentrations, and low additive main effects and multiplicative interaction (AMMI) stability values and iron stability index (FSI) and zinc stability index (ZSI) under low N conditions. These genotypes can be considered for production under low N stress conditions. Two environments (E4 and E3) were identified for good discriminatory power for genotype performance in terms of Fe and Zn content, respectively. Stable and high-yielding genotypes with high Fe and Zn concentration can be used as biofortified hybrids, which can contribute to a sustainable solution to malnutrition in the region, especially under low N conditions.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease primarily characterized by loss of motor neurons in brain and spinal cord. The death of motor neurons leads to denervation of muscle which in turn causes muscle weakness and paralysis, decreased respiratory function and eventually death. Growing evidence indicates disturbances in energy metabolism in patients with ALS and animal models of ALS, which are likely to contribute to disease progression. Particularly, defects in glucose metabolism and mitochondrial dysfunction limit the availability of ATP to CNS tissues and muscle. Several metabolic approaches improving mitochondrial function have been investigated in vitro and in vivo and showed varying effects in ALS. The effects of metabolic approaches in ALS models encompass delays in onset of motor symptoms, protection of motor neurons and extension of survival, which signifies an important role of metabolism in the pathogenesis of the disease. There is now an urgent need to test metabolic approaches in controlled clinical trials. In addition, more detailed studies to better characterize the abnormalities in energy metabolism in patients with ALS and ALS models are necessary to develop metabolically targeted effective therapies that can slow the progression of the disease and prolong life for patients with ALS. Aims To assess the influence of non- Saccharomyces yeasts on the pyranoanthocyanins and polymeric pigments formation after the addition of (+)-catechin and procyanidin B2 to fresh red grape must. Methods and Results The fermentation of red grape musts was done with non- Saccharomyces yeasts either alone or in sequential fermentations with the Saccharomyces cerevisiae species. The characterization of both pyranoanthocyanin and polymeric pigments has been carried out with liquid chromatography coupled to mass spectroscopy ( HPLC- DAD- ESI/ MS). Red wines were also characterized by infrared spectroscopy ( FTIR), gas chromatography ( GC- FID) and spectrophotometry ( UV-Vis). It has been observed that fermentation with the species Schizosaccharomyces pombe led to higher concentrations of pigments of all types: anthocyanins, polymeric pigments and pyranoanthocyanins, particularly vitisin A. Conclusions The use of non- Saccharomyces yeasts improve the formation of stable pigments in red wines thanks to the differences in the microbial metabolism from among the yeasts studied. There is increasing evidence that energy metabolism is disturbed in Amyotrophic Lateral Sclerosis (ALS) patients and animal models. Treatment with triheptanoin, the triglyceride of heptanoate, is a promising approach to provide alternative fuel to improve oxidative phosphorylation and aid ATP generation. Heptanoate can be metabolized to propionyl-CoA, which after carboxylation can produce succinyl-CoA and thereby re-fill the tricarboxylic acid (TCA) cycle (anaplerosis). Here we tested the hypothesis that treatment with triheptanoin prevents motor neuron loss and delays the onset of disease symptoms in female mice overexpressing the mutant human SOD1G93A (hSOD1G93A) gene. When oral triheptanoin (35% of caloric content) was initiated at P35, motor neuron loss at 70 days of age was attenuated by 33%. In untreated hSOD1G93A mice, the loss of hind limb grip strength began at 16.7 weeks. Triheptanoin maintained hind limb grip strength for 2.8 weeks longer (p<0.01). Loss of balance on the rotarod and reduction of body weight were delayed by 13 and 11 days respectively (both p<0.01). Improved motor function occurred in parallel with alterations in the expression of genes associated with muscle metabolism. In gastrocnemius muscles, the mRNA levels of pyruvate, 2-oxoglutarate and succinate dehydrogenases and methyl-malonyl mutase were reduced by 24–33% in 10 week old hSOD1G93A mice when compared to wild-type mice, suggesting that TCA cycling in skeletal muscle may be slowed in this ALS mouse model at a stage when muscle strength is still normal. At 25 weeks of age, mRNA levels of succinate dehydrogenases, glutamic pyruvic transaminase 2 and the propionyl carboxylase β subunit were reduced by 69–84% in control, but not in triheptanoin treated hSOD1G93A animals. Taken together, our results suggest that triheptanoin slows motor neuron loss and the onset of motor symptoms in ALS mice by improving TCA cycling. [ABSTRACT FROM AUTHOR]

Opencast limestone mines or limestone quarries are considered challenging ecosystems for soil fungi as they are highly degraded land with specific conditions, including high temperature, prolonged sunlight exposure, and a lack of organic matter, moisture, and nutrients in soil. In such ecosystems, certain fungi can survive and have a crucial function in maintaining soil ecosystem functions. Unfortunately, we know very little about taxonomic diversity, potential functions, and the ecology of such fungi, especially for a limestone quarry in a tropical region. Here, we characterized and compared the living soil fungal communities in an opencast limestone mine, including mining site and its associated rehabilitation site (9 months post-rehabilitation), with the soil fungal community in a reference forest, using the amplicon sequencing of enrichment culture. Our results showed that living fungal richness in the quarry areas was significantly lower than that in the reference forest, and their community compositions were also significantly different. Living fungi in the mining sites mostly comprised of Ascomycota (Eurotiomycetes and Sordariomycetes) with strongly declined abundance or absence of Basidiomycota and Mucoromycota. After nine months of rehabilitation, certain taxa were introduced, such as Hypoxylon spp. and Phellinus noxius, though this change did not significantly differentiate fungal community composition between the mining and rehabilitation plots. The majority of fungi in these plots are classified as saprotrophs, which potentially produce all fifteen soil enzymes used as soil health indicators. Network analysis, which was analyzed to show insight into complex structures of living fungal community in the limestone quarry, showed a clear modular structure that was significantly impacted by different soil properties. Furthermore, this study suggests potential taxa that could be useful for future rehabilitation.

Little is known about microbial communities of aquatic plants despite their crucial ecosystem function in aquatic ecosystems. Here, we analyzed the microbiota of an aquatic rheophyte, Hanseniella heterophylla, growing at three areas differing in their degree of anthropogenic disturbance in Thailand employing a metabarcoding approach. Our results show that diverse taxonomic and functional groups of microbes colonize H. heterophylla. Proteobacteria, Actinobacteria, Dothideomycetes, and Sordariomycetes form the backbone of the microbiota. Surprisingly, the beneficial microbes reported from plant microbiomes in terrestrial habitats, such as N-fixing bacteria and ectomycorrhizal fungi, were also frequently detected. We showed that biofilms for attachment of H. heterophylla plants to rocks may associate with diverse cyanobacteria (distributed in eight families, including Chroococcidiopsaceae, Coleofasciculaceae, Leptolyngbyaceae, Microcystaceae, Nostocaceae, Phormidiaceae, Synechococcaceae, and Xenococcaceae) and other rock biofilm-forming bacteria (mainly Acinetobacter, Pseudomonas, and Flavobacterium). We found distinct community compositions of both bacteria and fungi at high and low anthropogenic disturbance levels regardless of the study areas. In the highly disturbed area, we found strong enrichment of Gammaproteobacteria and Tremellomycetes coupled with significant decline of total bacterial OTU richness. Bacteria involved with sulfamethoxazole (antibiotic) degradation and human pathogenic fungi (Candida, Cryptococcus, Trichosporon, and Rhodotorula) were exclusively detected as indicator microorganisms in H. heterophylla microbiota growing in a highly disturbed area, which can pose a major threat to human health. We conclude that aquatic plant microbiota are sensitive to anthropogenic disturbance. Our results also unravel the potential use of this plant as biological indicators in remediation or treatment of such disturbed ecosystems.

Currently, the world population is increasing, and humanity is facing food and nutritional scarcity. Climate change and variability are a major threat to global food and nutritional security, reducing crop productivity in the tropical and subtropical regions of the globe. Cowpea has the potential to make a significant contribution to global food and nutritional security. In addition, it can be part of a sustainable food system, being a genetic resource for future crop improvement, contributing to resilience and improving agricultural sustainability under climate change conditions. In malnutrition prone regions of sub-Saharan Africa (SSA) countries, cowpea has become a strategic dryland legume crop for addressing food insecurity and malnutrition. Therefore, this review aims to assess the contribution of cowpea to SSA countries as a climate-resilient crop and the existing production challenges and perspectives. Cowpea leaves and immature pods are rich in diverse nutrients, with high levels of protein, vitamins, macro and micronutrients, minerals, fiber, and carbohydrates compared to its grain. In addition, cowpea is truly a multifunctional crop for maintaining good health and for reducing non-communicable human diseases. However, as a leafy vegetable, cowpea has not been researched and promoted sufficiently because it has not been promoted as a food security crop due to its low yield potential, susceptibility to biotic and abiotic stresses, quality assurance issues, policy regulation, and cultural beliefs (it is considered a livestock feed). The development of superior cowpea as a leafy vegetable can be approached in different ways, such as conventional breeding and gene stacking, speed breeding, mutation breeding, space breeding, demand-led breeding, a pan-omics approach, and local government policies. The successful breeding of cowpea genotypes that are high-yielding with a good nutritional value as well as having resistance to biotics and tolerant to abiotic stress could also be used to address food security and malnutrition-related challenges in sub-Saharan Africa.

Opencast mining removes topsoil and associated bacterial communities that play crucial roles in soil ecosystem functioning. Understanding the community composition and functioning of these organisms may lead to improve mine-rehabilitation practices. We used a culture-dependent method, combined with Illumina sequencing, to compare the taxonomic richness and composition of living bacterial communities in opencast mine substrates and young mine-rehabilitation plots, with those of soil in adjacent remnant forest at a limestone mine in northern Thailand. We further investigated the effects of soil physico-chemical factors and ground-flora cover on the same. Although, loosened subsoil, brought in to initiate rehabilitation, improved water retention and facilitated plant re-establishment, it did not increase the population density of living microbes substantially within 9 months. Planted trees and sparse ground flora in young rehabilitation plots had not ameliorated the micro-habitat enough to change the taxonomic composition of the soil bacteria compared with non-rehabilitated mine sites. Viable microbes were significantly more abundant in forest soil than in mine substrates. The living bacterial community composition differed significantly, between the forest plots and both the mine and rehabilitation plots. Proteobacteria dominated in forest soil, whereas Firmicutes dominated in samples from both mine and rehabilitation plots. Although, several bacterial taxa could survive in the mine substrate, soil ecosystem functions were greatly reduced. Bacteria, capable of chitinolysis, aromatic compound degradation, ammonification and nitrate reduction were all absent or rare in the mine substrate. Functional redundancy of the bacterial communities in both mine substrate and young mine-rehabilitation soil was substantially reduced, compared with that of forest soil. Promoting the recovery of microbial biomass and functional diversity, early during mine rehabilitation, is recommended, to accelerate soil ecosystem restoration and support vegetation recovery. Moreover, if inoculation is included in mine rehabilitation programs, the genera: Bacillus, Streptomyces and Arthrobacter are likely to be of particular interest, since these genera can be cultivated easily and this study showed that they can survive under the extreme conditions that prevail on opencast mines.

In social-ecological systems, natural resource management can be characterized by trade-offs across sectors and sustainability targets. The water-energy-food (WEF) nexus concept makes explicit various trade-offs in order to maximize synergies of interventions. However, there are few successful examples of its operationalization in research settings. Here, we explore in a learning setting if sustainability impact assessment (SIA) protocols can be a useful process to be used to adopt a systemic, interdisciplinary perspective to operationalize WEF nexus in research for sustainable development. The process and method adopted of SIA protocol, evaluated for five exemplary WEF nexus cases in Central Asia during a week-long international workshop, adequately addressed the complexity of WEF interrelationships and associated sustainability issues, and facilitated a comparative case study analysis across scales. Results within this process highlight that water governance was critical for large-scale transboundary WEF nexus management, while land and soil management were decisive for minimizing trade-offs at local levels. Issues of interdisciplinarity, complexity, uncertainty, and reflection on impacts were adequately addressed, but challenges remain in the consideration of ethics and the design of transparent, multi-actor cooperation. Most importantly, this exercise showed that employment of the process of SIA protocol supported disciplinary experts to work across disciplines and take a systemic approach for analyzing WEF nexus.

Abstract Background Land use changes and related land management practices significantly alter soil physicochemical properties; however, their effects on the soil microbial community structure are still unclear. In this study, we used automated ribosomal intergenic spacer analysis to determine the fungal and bacterial community composition in soils from different land use areas in the Ethiopian highlands. Soil samples were collected from five areas with different land uses, natural forest, eucalyptus plantation, exclosure, grassland and cropland, which had all historically been natural forest. Results Our results showed a significant shift in the soil bacterial and fungal community composition in response to land use change. We also identified soil physicochemical factors corresponding to the changes in bacterial and fungal communities. Although most soil attributes, including soil organic carbon, total soil nitrogen, labile P, soil pH and soil aggregate stability, were related to the change in bacterial community composition, the total soil nitrogen and soil organic carbon had the strongest relationships. The change in fungal community composition was correlated with soil nutrients, organic carbon, soil nitrogen and particularly the labile P concentration. Conclusions The fungal community composition was likely affected by the alteration of vegetation cover in response to land use change, whereas the bacterial communities were mainly sensitive to changes in soil attributes. The study highlights the higher sensitivity of fungal communities than bacterial communities to land use changes.
