Background & objectives: This study reports observation on circulating serotypes and genotypes of Dengue Virus in North India. Methods: Serum samples were obtained from suspected cases of dengue referred to the virus diagnostic laboratory during 2014 to 2022. All samples were tested for anti-dengue virus IgM antibodies and NS1Ag by ELISA. NS1Ag positive samples were processed for serotyping and genotyping. Results: Total 41,476 dengue suspected cases were referred to the laboratory of which 12,292 (29.6%) tested positive. Anti-Dengue Virus IgM antibodies, NS1Ag, both IgM and NS1Ag, were positive in 7007 (57.4%); 3200 (26.0%) and 2085 (16.0%) cases respectively. Total 762 strains were serotyped during 9-year period. DENV-1, DENV-2, DENV-3 and DENV-4 serotypes were found in 79 (10.37%), 506 (66.40%), 151 (19.82%) and 26 (3.41%) cases respectively. DENV-1, DENV-2 and DENV-3 were in circulation throughout. Total 105 strains were genotyped. Genotype IV of DENV-1 serotype was circulating till 2014 which was later replaced by genotype V. A distinct seasonality with increase in number of cases in post-monsoon period was seen. Interpretation & conclusion: DENV-1, DENV-2 and DENV-3 were found to be in circulation in North India. Predominant serotype/genotype changed at times, but not at regular intervals.

Nowadays, effective cancer therapy is a global concern, and recent advances in nanomedicine are crucial. Cancer is one of the major fatal diseases and a leading cause of death globally. Nanotechnology provides rapidly evolving delivery systems in science for treating diseases in a site-specific manner using natural bioactive compounds, which are gaining widespread attention. Nanotechnology combined with bioactives is a very appealing and relatively new area in cancer treatment. Natural bioactive compounds have the potential to be employed as a chemotherapeutic agent in the treatment of cancer, in addition to their nutritional benefits. Alginate, pullulan, cellulose, polylactic acid, chitosan, and other biopolymers have been effectively used in the delivery of therapeutics to a specific site. Because of their biodegradability, biopolymeric nanoparticles (BNPs) have received a lot of attention in the development of new anticancer drug delivery systems. Biopolymer-based nanoparticle systems can be made in a variety of ways. These systems have developed as a cost-effective and environmentally friendly solution to boost treatment efficacy. Effective drug delivery systems with improved availability, increased selectivity, and lower toxicity are needed. Recent research findings and current knowledge on the use of BNPs in the administration of bioactive chemicals in cancer therapy are summarized in this review.

Abstract This study provides a combined analysis on the palynology, fossil charcoal and biomarkers of the subsurface coal deposits from a borehole RMB #2 drilled at the Dhulia Coal Block, Rajmahal Basin, India, in attempts to establish the chronology of sedimentation and to propose palaeobotanical as well as geochemical evidence for the occurrence of wildfires in these sediments. The palynological investigation suggests a Scheuringipollenites barakarensis palynoassemblage from the lower Barakar Formation, dated as Artinskian (Early Permian) in age. This assemblage reveals the dominance of Glossopteridales and sub-dominance of taxa belonging to Cordaitales and Coniferales. Fossil charcoal in sediments is usually recognized as a direct indicator for the occurrence of palaeo-wildfires. More data involving the anatomical features of fossil charcoal analyzed by Field Emission Scanning Electronic Microscope broaden our knowledge on Early Permian wildfires from the peninsula of India. The studied macroscopic charcoal fragments exhibit anatomical details such as homogenized cell walls, uniseriate simple and biseriate alternate pitting on tracheid walls and rays of varying heights pointing to a gymnospermous wood affinitity. The excellent preservation of charcoal fragments, shown by their large sizes and almost unabraded edges, suggests a parautochthonous origin. The embedded biomarker study performed for charcoal sediments and its characterization demonstrate the presence of n-alkanes, isoprenoids, terpenoids and aromatic compounds. A bimodal distribution pattern of n-alkanes with a Cmax at n-C25 is identified. Diterpenoids and pentacyclic terpenoids are identified, indicating the input of an early conifer vegetation and bacterial activity, respectively. The identified polyaromatic hydrocarbon (PAH) compounds, together with the charcoal fragments, clearly suggest that repeated wildfire events occurred during the deposition of these Artinskian sediments in the Rajmahal Basin.

Summary: BRAF fusions are detected in numerous neoplasms, but their clinical management remains unresolved. We identified six melanoma lines harboring BRAF fusions representative of the clinical cases reported in the literature. Their unexpected heterogeneous responses to RAF and MEK inhibitors could be categorized upon specific features of the fusion kinases. Higher expression level correlated with resistance, and fusion partners containing a dimerization domain promoted paradoxical activation of the mitogen-activated protein kinase (MAPK) pathway and hyperproliferation in response to first- and second-generation RAF inhibitors. By contrast, next-generation αC-IN/DFG-OUT RAF inhibitors blunted paradoxical activation across all lines and had their therapeutic efficacy further increased in vitro and in vivo by combination with MEK inhibitors, opening perspectives in the clinical management of tumors harboring BRAF fusions. : Botton et al. shed light on the heterogeneity of BRAF fusions encountered in melanocytic tumors and characterize features influencing their signaling and drug response. These findings unveil the singularities of BRAF fusions and establish general principles to guide their clinical management in melanoma and other malignancies.
