Pollen germination in vivo on wet stigmas is assisted by the receptive fluid—stigma exudate. Its exact composition is still unknown because only some components have been studied. For the first time, hormonal screening was carried out, and the fatty acid (FA) composition of lipid-rich (Nicotiana tabacum) and sugar-rich (Lilium longiflorum) exudates was studied. Screening of exudate for the presence of plant hormones using HPLC-MS revealed abscisic acid (ABA) in tobacco stigma exudate at the two stages of development, at pre-maturity and in mature stigmas awaiting pollination, increasing at the fertile stage. To assess physiological significance of ABA on stigma, we tested the effect of this hormone in vitro. ABA concentration found in the exudate strongly stimulated the germination of tobacco pollen, a lower concentration had a weaker effect, increasing the concentration did not increase the effect. GC-MS analysis showed that both types of exudate are characterized by a predominance of saturated FAs. The lipids of tobacco stigma exudate contain significantly more myristic, oleic, and linoleic acids, resulting in a higher unsaturation index relative to lily stigma exudate lipids. The latter, in turn, contain more 14-hexadecenoic and arachidic acids. Both exudates were found to contain significant amounts of squalene. The possible involvement of saturated FAs, ABA, and squalene in various exudate functions, as well as their potential relationship on the stigma, is discussed. [ABSTRACT FROM AUTHOR]

A pollen grain is a unique haploid organism characterized by a special composition and structure. The pollen of angiosperms and gymnosperms germinate in fundamentally similar ways, but the latter also have important features, including slow growth rates and lower dependence on female tissues. These features are, to some extent, due to the properties of pollen lipids, which perform a number of functions during germination. Here, we compared the absolute content and the fatty acid (FA) composition of pollen lipids of two species of flowering plants and spruce using GC-MS. The FA composition of spruce pollen differed significantly, including the predominance of saturated and monoene FAs, and a high proportion of very-long-chain FAs (VLCFAs). Significant differences between FAs from integumentary lipids (pollen coat (PC)) and lipids of gametophyte cells were found for lily and tobacco, including a very low unsaturation index of the PC. The proportion of VLCFAs in the integument was several times higher than in gametophyte cells. We found that the absolute content of lipids in lily pollen is almost three times higher than in tobacco and spruce pollen. For the first time, changes in the FA composition were analyzed during pollen germination in gymnosperms and angiosperms. The stimulating effect of H2O2 on spruce germination also led to noticeable changes in the FA content and composition of growing pollen. The effect of red (RL, 660 nm) and blue (BL, 450 nm) light on phy mutant tomato plants was studied. The rates of photosynthesis (Pn) and transpiration, the efficiency of the primary photochemical processes of photosynthesis, the contents of flavonoids and phenolic compounds, the low-molecular-weight antioxidant capacity (Trolox equivalent antioxidant capacity (TEAC)) of leaf extracts, and the expression of light-dependent genes were evaluated. Under RL, BL, and white fluorescent light (WFL), the Pn values decreased in the order: WT > phyb2 > phyaphyb2 > phyaphyb1phyb2, except for the Pn in phyb2 on BL. Phyb2 also had a larger number of stomata under BL and, as a result, it reached maximum transpiration. The noticeable accumulation of flavonoids and phenolic compounds was observed only in the phyb2 and phyaphyb2 mutants upon irradiation with BL, which agrees with the increased TEAC in the leaf extracts. We suggest that the increased antioxidant activity under PHYB2 deficiency and the maintenance of high photosynthesis under BL are based on an increase in the expression of the early signaling transcription factors genes BBX, HY5. The largest decrease in the content of flavonoids and TEAC was manifested with a deficiency in PHYB1, which is probably the key to maintaining the antioxidant status in BL plants. Simple Summary: Plants growing at temperate and polar latitudes are exposed to cold stress. With climate change, different durations of low temperatures and sometimes frost are being increasingly observed in regions at low latitudes. This can cause especially great harm to agricultural crops, the mortality of which can pose a serious challenge to the food supply for the global population. One of the factors affecting plant resistance to low and negative temperatures is specific changes in the fatty acid (FA) composition of lipids. It should be noted that most of the crops studied in this regard are angiosperms. It is known that the FA composition of angiosperms has undergone significant evolutionary changes compared to that of nonflowering vascular plants. Studying the FA composition of various taxonomic groups can shed light on and reveal new mechanisms of plant resistance. Therefore, in this paper, we focused on the rare evergreen fern Asplenium scolopendrium, whose fronds can tolerate freezing. A number of specific features of its FA composition were discovered, which, in combination with other resistance mechanisms, determine its ability to grow in temperate climate zones and safely undergo wintering. Ferns are one of the oldest land plants. Among them, there are species that, during the course of evolution, have adapted to living in temperate climates and under winter conditions. Asplenium scolopendrium is one such species whose fronds are able to tolerate low subzero temperatures in winter. It is known that the resistance of ferns to freezing is associated with their prevention of desiccation via unique properties of the xylem and effective photoprotective mechanisms. In this work, the composition of A. scolopendrium lipid fatty acids (FAs) at different times of the year was studied by gas–liquid chromatography with mass spectrometry to determine their role in the resistance of this species to low temperatures. During the growing season, the polyunsaturated FA content increased significantly. This led to increases in the unsaturation and double-bond indices by winter. In addition, after emergence from snow, medium-chain FAs were found in the fronds. Thus, it can be speculated that the FA composition plays an important role in the adaptation of A. scolopendrium to growing conditions and preparation for successful wintering. The composition of fatty acids (FAs) of total lipids of pericarp, seeds, and leaves of Pyrus caucasica Fed. and Pyrus communis L. growing in mountain ecosystems at different altitudes (300, 700 and 1200 m) was studied. It was found that the greatest differences in the relative content of FAs within a species, depending on the altitudes above sea level, were characteristic of the outer tissues of the pericarp (peel) and leaves, which were in direct contact with the external environment. Pericarp parenchyma to a lesser extent, and seeds practically did not differ in FA composition at different heights. At altitudes with increased UV radiation, conjugated octadecadienoates: rumenic acid (9,11–18:2) and 10,12–18:2 were registered in the pericarp and leaf of Purys L., the functions of which in plants were practically not studied. The wild P. caucasica at all growing altitudes was characterized by more very-long-chain FAs (VLCFAs) than the P. communis cultivar. At 700 m, most likely when exposed to fungal infections, the relative number of VLCFAs increased significantly, and new species of individual odd-chaine FAs appeared in their composition in both representatives. It was especially worth noting the appearance in peel and leaf melissic acid (30:0), which was rarely recorded in the plant. A characteristic feature of only P. communis at an altitude of 700 m was the large number of unsaturated individual VLCFAs. Based on the data obtained, a scheme of possible pathways for VLCFA biosynthesis in P. communis were proposed. Ferns are the second most diverse group of higher plants on the planet. However, the metabolic processes of these plants, as well as their reaction to the influence of various environmental factors, benefit from new investigation. In seed plants, an important role of the lipid fatty acid (FA) composition in plant response to a variety of environmental conditions has been repeatedly demonstrated. In this paper, the composition of FAs in the epiphytic ferns, Platycerium bifurcatum and Asplenium nidus, and the terrestrial fern, Asplenium trichomanes was analyzed by gas-liquid chromatography with mass spectrometry. The data obtained clearly indicate that epiphytic ferns have several distinctive features in the composition of their FAs. They are characterized by a high level of saturated FAs (about 50%) and a great variety of very-long-chain FAs (VLCFA). This strongly contrasts with the FAs composition in the terrestrial fern in which unsaturated FAs prevail. High levels of saturated fatty acids, in particular palmitic acid (16:0), together with a high VLCFA content, can be a necessary component in complex interactions with microorganisms. Thus, the specific FA composition of P. bifurcatum and A. nidus may act both as a protective mechanism against the penetration of bacteria and pathogenic fungi, and as a mechanism for the interaction of the epiphytes with saprophytic fungi. Pollen is a highly specialized structure for sexual plant reproduction. Early stages of pollen germination require the transition from dormant state to active metabolism. In particular, an important role during this early phase of angiosperm pollen germination is played by H+-ATPase. Very little is known about pollen activation in gymnosperm species, and information on the involvement of H+-ATPase is lacking. We tracked four indicators characterizing the physiological state of pollen: membrane potential, intracellular pH, anion efflux and oxygen uptake, in order to monitor the dynamics of activation in Picea pungens. Based on pH dynamics during activation, we assumed the important role of H+-ATPase in spruce pollen germination. Indeed, germination was severely suppressed by P-type ATPase inhibitor orthovanadate. In spruce pollen tubes, a pronounced pH gradient with a maximum in the apical zone was found, which was different from the pollen tubes of flowering plants. Using orthovanadate and fusicoccin, we found that the proton pump is largely responsible for maintaining the gradient. Immunolocalization of the enzyme in pollen tubes showed that the distribution of H+-ATPase generally coincides with the shape of the pH gradient: its maximum accumulation is observed in the apical zone. This pilot study aimed at comparing zinc (Zn) and nickel (Ni) effects on the fatty acid (FA) profiles, oxidative stress and desaturase activity in the Zn hyperaccumulator Arabidopsis halleri and the excluder Arabidopsis lyrata to allow a better picture of the physiological mechanisms which may contribute to metal tolerance or acclimation. The most significant changes in the FA composition were observed in the shoots of the hyperaccumulator and in the roots of the excluder, and were not only metal-dependent, but also species-specific, since the most significant changes in the shoots of A. halleri were observed under Ni treatment, though Ni, in contrast to Zn, was accumulated mainly in its roots. Several FAs appeared in the roots and shoots of A. lyrata only upon metal exposure, whereas they were already found in control A. halleri. In both species, there was an increase in oleic acid under Ni treatment in both organs, whereas in Zn-treated plants the increase was shown only for the shoots. A rare conjugated α-parinaric acid was identified only in the shoots of metal-treated A. halleri. In the shoots of the hyperaccumulator, there was an increase in the content of saturated FAs and a decrease in the content of unsaturated FAs, while in the roots of the excluder, the opposite pattern was observed. These metal-induced changes in FA composition in the shoots of A. halleri can lead to a decrease in the fluidity of membranes, which could diminish the penetration of ROS into the membrane and thus maintain its stability. • In shoots of metal-treated Arabidopsis halleri saturated fatty acid content increased. • In roots of metal-treated Arabidopsis lyrata unsaturated fatty acid content decreased. • α-parinaric acid was identified only in shoots of metal-treated Arabidopsis halleri. • In both species the content of oleic acid increased under Ni and Zn treatments. • Many changes in fatty acid composition were metal-dependent and species-specific.
