As a novel environmental pollutant, the impacts of nanoplastics on crops have attracted more and more attention. To explore the response of soybean to polystyrene nanoplastics (PS-NPs) at physiological and gene expression levels, this experiment used hydroponics to determine the phenotype and physiological indexes of soybean seedlings after exposure to 50 mg/L 100 nm PS-NPs for 10 days, and carried out transcriptome sequencing analysis of soybean seedling root system. The results showed that the growth of the soybean seedlings was inhibited, the root morphology changed significantly, the activities of superoxide dismutase (SOD) and catalase (CAT) increased to a certain extent, the content of malondialdehyde (MDA) increased significantly, and the content of soluble sugar decreased after PS-NPs exposure. Transcriptome analysis showed that a total of 2112 differentially expressed genes were generated by PS-NPs exposure treatment, GO and KEGG analysis showed that they were mainly enriched in pathways related to redox reaction, reaction to ethylene and abscisic acid, amino acid metabolism and synthesis of other secondary metabolites. It was noted that 250 transcription factors were differentially expressed, mainly including the ERF, WRKY, NAC, bHLH and MYB transcription factor families. Objectives: This study aimed to investigate the long‐term sexual function of patients with cervical cancer who underwent treatment and to explore influential factors. Methods: This retrospective cross‐sectional study was conducted at Peking University First Hospital in (Beijing, China). A total of 207 patients, who were diagnosed with Stage IA‐IIA cervical cancer and had undergone surgical treatment (some patients had also been treated with adjuvant radiotherapy and chemotherapy) between January 2010 and August 2020, completed questionnaires via telephone. The median time since diagnosis was 54 (range, 13–138) months. Sexual function was assessed using the validated short form of Pelvic Organ Prolapse/Urinary Incontinence Sexual Questionnaire (PISQ‐12). The multivariate logistic regression analysis was performed to determine factors influencing sexual function after treatment. Results: The mean preoperative PISQ‐12 score was 39.42 ± 3.922, and the mean postoperative PISQ‐12 score was 32.60 ± 6.592, indicating a significant decrease in postoperative PISQ‐12 score compared with preoperation (p < 0.001). In total, 49.8% of the patients had sexual dysfunction after treatment. According to the results of the multivariate logistic regression analysis, longer follow‐up (months), ovariectomy, lack of hormone replacement therapy after ovariectomy and adjuvant radiotherapy were significantly associated with sexual dysfunction after treatment (p < 0.05). There was no significant correlation among surgical method, tumor stage, adjuvant chemotherapy, and sexual dysfunction after treatment. Conclusions: The sexual function of cervical cancer survivors significantly decreased after treatment, which was related to the length of follow‐up, ovariectomy, and adjuvant radiotherapy. Abstract Background Citrus tristeza virus (CTV), a member of the genus Closterovirus within the family Closteroviridae, is the causal agent of citrus tristeza disease. Previous studies revealed that the negative selection, RNA recombination and gene flow were the most important forces that drove CTV evolution. However, the CTV codon usage was not studied and thus its role in CTV evolution remains unknown. Results A detailed comparative analysis of CTV codon usage pattern was done in this study. Results of the study show that although in general CTV does not have a high degree of codon usage bias, the codon usage of CTV has a high level of resemblance to its host codon usage. In addition, our data indicate that the codon usage resemblance is only observed for the woody plant-infecting closteroviruses but not the closteroviruses infecting the herbaceous host plants, suggesting the existence of different virus-host interactions between the herbaceous plant-infecting and woody plant-infecting closteroviruses. Conclusion Based on the results, we suggest that in addition to RNA recombination, negative selection and gene flow, host plant codon usage selection can also affect CTV evolution.

Smoking is a well-known major risk factor in development of esophageal cancer, but few studies have reported the association between smoking status and prognosis of these patients. We conduct the present study to summarize current evidence. A computerized search of the PubMed and EMBASE was performed up to April 30, 2015. Eight studies, containing 4,286 patients, were analyzed. In the grouping analysis, among esophageal squamous-cell carcinoma patients, current and former smokers, compared to those who have never smoked, seemed to have a poorer prognosis (HR = 1.41, 95% CI 1.22–1.64, and HR = 1.35, 95% CI 0.92–1.97, resp.). In the subgroup analysis, adverse effects on current smoker compared with never smoker were also observed in China and the other countries (HR = 1.5, 95% CI 1.18–1.92, and HR = 1.36, 95% CI 1.12–1.65, resp.). In the group that ever smoked, we could not get a similar result. No significantly increased risk was found in esophageal adenocarcinoma patients compared to the squamous-cell histology ones. In the smoking intensity analysis, heavy smoking was associated with poor survival in esophageal squamous-cell carcinoma. Our pooled results supported the existence of harmful effects of smoking on survival after esophagus cancer diagnosis. [ABSTRACT FROM AUTHOR]

Abstract: The genetic diversity of the genus Dendrobium is not well known and the phylogenetic relationship of Dendrobium species are mainly determined by studies of the comparative vegetative anatomy and plant systematics. In the present study, we used the technique of inter-simple sequence repeats (ISSRs) to evaluate genetic diversity and phylogenetic relationship among 31 Dendrobium species from Yunnan region of China. In total, 2368 bands were amplified by 17 ISSR primers, resulting from 278 ISSR loci with 100% polymorphism at genus level. Thirty-one species were unequivocally distinguished based on ISSR fingerprinting. Species-specific ISSR markers were identified in nine of 31 tested Dendrobium species. Unweighted pair-group mean analysis (UPGMA) showed that 31 Dendrobium species were grouped into six clusters, indicating the genus was polyphyletic with several well-supported lineages. The high polymorphism and reliable amplification across species demonstrated the utility of ISSR marker for species diagnosis and genetic diversity study of the genus Dendrobium. Based on the study of the relationships between economic growth and industrial pollutant discharge of Suzhou City in 1991-2005, the environmental Kuznets curve (EKC) of the City was estimated by several kinds of function, with the cause analyzed. The results showed that the EKC of Suzhou was not a typical one. The industrial pollution of the City increased dramatically by scale effect of economic growth and the clear environmental benefit of industrial structure adjustment was not received obviously, while generalized discharge reduction technologies showed great results. In general terms, the pollution increase was comparatively slower than the GDP growth. As a whole, the environmental policy of Suzhou was in effect. However, the relationships between economic growth and industrial pollutant discharge had not been a benign development yet. In order to achieve win-win situation of economic growth and pollution reduction, Suzhou needed to optimize its industry structure, push the energy conservation and discharge reduction policy deeply, increase environmental investment, and enforce the strictest environmental and industrial policy. Plants use various secondary chemicals in their chemical defense against herbivores. While botanical insecticides are crucial for reducing the reliance on chemical pesticides, the development of plant-derived insecticides remains limited. In this study, we fed Henosepilachna vigintioctopunctata with three different host plants (Solanum nigrum, Solanum tuberosum, and Physalis floridana) and observed that feeding on P. floridana led to changes in the body size and a significantly high mortality rate. Through transcriptome analysis, it was found that the trehalose metabolism pathway of H. vigintioctopunctata changed significantly under different host feeding conditions, especially since the expression level of the trehalase gene was extremely different. We subsequently identified eight transcripts of HvTREs and analyzed their evolution and structure. Among them, significant differences are observed in the relative expression levels of HvTRE1-5 in H. vigintioctopunctata after the fourth instar and were affected by different plant diets. Compared with the natural host S. nigrum, the larvae that fed on P. floridana significantly reduced the contents of trehalose, glucose and glycogen and significantly affected the trehalase activity. Knockdown of HvTRE1-5 by RNAi increased mortality at the H. vigintioctopunctata prepupation stage, suggesting that HvTRE1-5 is important for H. vigintioctopunctata pupation. This study provides new insights into developing of green control methods for H. vigintioctopunctata and offers a valuable example for understanding the interaction between host plants and herbivorous insects. Chrysanthemum morifolium plants are important commercial herbs in China, widely used in ornamental horticulture and traditional medicine. In this study, start codon targeted (SCoT) polymorphic markers were applied to assess genetic relationships among 32 populations of C. morifolium . Thirty-two SCoT primers produced 872 loci, 90% (785) of which showed polymorphism. Average polymorphism information content of the SCoT primers was 0.987 (0.950–0.993), showing that plenty of genetic diversity exists among Chinese C. morifolium cultivars. Cluster analysis of SCoT markers (based on UPGMA and PCoA method) grouped all the cultivars of C. morifolium into two clusters. The partition of clusters in the dendrogram and PCoA plot was similar and large degree of grouping according to their origin and ecological distribution. Our results will provide much more useful information for C. morifolium germplasm resource protection and will also useful to improve the current plant breeding programs. Our results also indicated that SCoT markers are informative and could be used to evaluate genetic diversity of Chinese medicinal C. morifolium cultivars. [ABSTRACT FROM AUTHOR]

Abstract: The genome organization of Dendrobium species remains largely unexplored. An F1 family derived from the controlled interspecific hybridization between Dendrobium moniliforme and D. officinale were used to create genetic linkage maps of parental Dendrobiums using double pseudo-testcross strategy. A total of 422 markers, including 66 EST-SSR, 126 SRAP, 74 ISSR, and 156 RAPD markers, showed expected Mendelian segregation ratio and were used for genetic map construction. Genetic linkage groups with Kosambi genetic distances were constructed using a LOD score≥4 and a recombination threshold of 0.4. The map of D. moniliforme was 1127.9cM in total length with 165 marker loci distributed in 17 linkage groups; and the D. officinale map consisted of 19 genetic linkage groups with a total length of 1210.9cM positioned by 169 marker loci. The genetic maps constructed in this study can serve as a reference linkage map of Dendrobium species for efficient studies of comparative genetics, genetic mapping of traits with economic interest, and marker-assisted selection in horticultural breeding. A species of Trichotylenchus nematode was isolated from the rhizosphere of banana root in Leizhou City, Guangdong Province, China. The species assumes the following characteristics: open C-shaped body; head offset from body; lateral field with three incisures, pharyngeal and tail regions irregularly areolated; stylet 18.6-20.7 µm long; pharyngeal gland not extending over intestine; fibrous tissue present in the intestine; post-anal intestinal sac present; elongate-subcylindroid tail, bluntly conoid terminus, lack of striations, and containing 34-44 annuli. In addition, scanning electron microscopy was used to elucidate some morphological details, but only some juveniles were observed. Partial 18S rRNA, ITS, and 28S D2-D3 expansion sequences were amplified with universal primers and deposited in GenBank under accession numbers ON622716, ON622717, and ON622714, respectively. Here, this species was identified as T. dispersus [(Siddiqi & Sharma, 1995) Geraert, 2011].

Microorganisms in anaerobic digestion (AD) are easily affected by ammonia, especially acetoclastic methanogens. Thus, in ammonia-suppressed AD systems, acetate degradation is reported to be carried out mainly by the cooperation of syntrophic acetate oxidizers and hydrogenotrophic methanogens. Previous studies have revealed ammonia inhibition on microbial flora by AD performance, but the effect mechanism of ammonia on microbial metabolism remains poorly understood. In this study, we constructed a mesophilic chemostat fed with acetate as the sole carbon source, gradually increased the total ammonia nitrogen (TAN) concentration from 1 g L -1 to 6 g L -1 , and employed the 16S rRNA gene, metagenomics, and metatranscriptomics analysis to characterize the microbial community structure and metabolic behavior. The results showed that even at the TAN of 6 g L -1 (pH 7.5), the methanogenesis kept normal, the biogas production was approximately 92% of that at TAN of 1 g L -1 and the acetate degradation ratio reached 99%, suggesting the strong TAN tolerance of the microbial community enriched. 16S rRNA gene analysis suggested that the microbial community structure changed along with the TAN concentration. Methanothrix predominated in methanogens all the time, in which the dominant species was gradually replaced from M. soehngenii to M. harundinacea with the increased TAN. Dominant bacterial species also changed and Proteiniphilum showed a significant positive correlation with increased TAN. Meta-omics analysis showed that the absolute dominant microorganisms at TAN of 6 g L -1 were M. harundinacea and Proteiniphilum , both of which highly expressed genes for anti-oxidative stress. M. harundinacea and the second dominant methanogen Methanosarcina highly expressed both acetate cleavage and CO 2 reduction pathways, suggesting the possibility that these two pathways contributed to methanogenesis together. Proteiniphilum and some other species in Firmicutes and Synergistetes were likely acetate oxidizers in the community as they highly expressed genes for syntrophic acetate oxidization, H 2 generation, and electron transfer. These results suggested that Proteiniphilum as well as M. harundinacea have strong ammonia tolerance and played critical roles in acetate degradation under ammonia-suppressed conditions. Acetate is a significant intermediate of anaerobic fermentation. There are two pathways for converting acetate to CH 4 and CO 2 : acetoclastic methanogenesis by acetoclastic methanogens, and syntrophic acetate oxidation by acetate-oxidizing bacteria (AOB) and hydrogenotrophic methanogens. Detailed investigations of syntrophic acetate-oxidizing bacteria (SAOB) should contribute to the elucidation of the microbial mechanisms of methanogenesis. In this study, we investigated the major phylogenetic groups of acetate-utilizing bacteria (AUB) in a mesophilic methanogenic chemostat fed with acetate as the sole carbon source by using DNA stable isotope probing (SIP) technology. The results indicated that acetoclastic methanogenesis and acetate oxidization/hydrogenotrophic methanogenesis coexisted in the mesophilic chemostat fed with acetate, operated at a dilution rate of 0.1 d -1 . OTU Ace13(9-17) (KU869530), Ace13(9-4) (KU667241), and Ace13(9-23) (KU667236), assigned to the phyla Firmicutes and Bacteroidetes, were probably potential SAOB in the chemostat, which needs further investigation. Species in the phyla Proteobacteria, Deferribacteres, Acidobacteria, Spirochaetes and Actinobacteria were probably capable of utilizing acetate for their growth. Methanoculleus was likely to be the preferred hydrogenotrophic methanogen for syntrophy with AOB in the chemostat.

Objective: To study the mechanisms on drug susceptibility and resistance of clinically multidrug-resistant Escherichia coli isolates, to provide information on related treatment., Methods: The susceptibility of E. coli strains that isolated from different kinds of samples in the last 3 years on drugs was analyzed by agar dilution test, with strains that exhibiting resistances to cefotaxime, ciprofloxacin and amikacin simultaneously collected for further analysis. Resistant genes which mediate resistance to β-lactamases, fluoroquinolone and aminoglycoside as well as phylogenic type were detected by PCR amplification while genetic relation was analyzed by PFGE. Transferability of resistant plasmids was identified by conjugation test., Results: In total, 137 multidrug-resistant E. coli isolates were collected. Only 1% of the isolates exhibited resistance to both imipenem and meropenem while 4% of the strains were resistant to piperacillin/tazobactam. Most (85%) of the isolates were positive to ESBL and majority of them produced CTX-M. Target substitution and production of methylases were the main mechanisms causing resistance to fluoroquinolones and aminoglycosides respectively., Conclusion: The main source of clinical multidrug-resistance was collected from urine samples. Carbapenem and enzyme inhibitor-containing antibiotics seemed to be the available antibiotics that were sensitivity to the clinically multidrug-resistant E. coli isolates.

Premise of the Study: Expressed sequence tag (EST)-derived simple sequence repeat (SSR) markers were developed in Dendrobium officinale by screening a cDNA library. The loci were verified by sequencing and explored for polymorphism among 19 genotypes and transferability among 30 other distantly related Dendrobium species. •, Methods and Results: One hundred ten EST-SSRs were developed, and a set of 20 amplified two to six nucleotide repeats with a mean number of 2.5 alleles per locus and with an observed heterozygosity and polymorphism information content per locus ranging from 0.3463 to 0.9003 and 0.0997 to 0.6537 in 19 D. officinale genotypes, respectively. Furthermore, 92 of these markers have cross-taxa transferability, ranging from 6.45% to 100% among 30 other distantly related Dendrobium species. •, Conclusions: The developed markers have potential for application in germplasm appraisal, genetic diversity study, genetic mapping, and molecular breeding in D. officinale and other congeneric species.

ISSR was applied to detect the relationship between 16 Cymbidium species, and 836 bands were amplified with 15 primers, including 227 polymorphic bands. The polymorphic percentage is 27.2%. UPGMA results showed that the genetic distance were closest between C.goeringii (Rchb.f.) Rchb.f. and C. goeringii var. longibracteatum, and C.lancifolium Hook. was far away from the other 15 species. This result is quite similar to the traditional classification, indicating that the technique could supplement some information to traditional taxonomy in the molecular level.

Cymbidium is one of the Orchidaceae genera. There are 70 Cymbidium species in the world, mainly distributed over the tropical and the subtropical area of Asia and northern Australia. In China, approximately 49 species and some variations are distributed over the provinces to the south of Qinling. The orchid market of China is quite wide, not only because of their view and admiration value, but also their officinal value as well. Some species of Cymbidium are morphologically similar, so the identification of Cymbidium species and the understanding of the relationships between them are very important for new varieties breeding of Cymbidium. This study attempts to apply RMAPD to detect the relationships between the Cymbidium species. 456 pairs of RMAPD combinations were screened, but only 12 pairs of them were used to analyze the genetic polymorphism of 16 Cymbidium. 362 bands were produced by 12 pairs of RMAPD primers in total, including 90 polymorphic bands (24.8%). The polymorphic bands of each primer combination ranged from 3 to 11 with the average of 8. Among all species, genetic similarity coefficient ranged from 0.63 to 0.87. The result showed that the genetic distance was closest between C. floribundum and C. aestivum, C. ensifolium and C. sinense, C. maguanense and C. hookerianum, which is almost the same as the traditional classification. It was the first time that species of Cymbidium were detected by RMAPD technique. This study shows RMAPD is an appropriate and effective molecular marker technique to identify the relationships of Cymbidium.

The techniques of random amplified polymorphic DNA (RAPD) and amplified fragment length polymorphisms (AFLPs) were used to amplify the genomic DNA of 10 species of Ilex, so as to reveal the polymorphism in different species of Ilex. In RAPD analysis, a total of 301 lines were amplified with 11 kinds of primers selected from 100 kinds of 10 bp random primers, and the polymorphism was 98.63%. In AFLPs analysis,a huge diversity was showed by 3 primers combinations carried out on these species. Both RAPD and AFLPs analysis were clustered by UPGMA and the results showed that the genetic distances were the closest between I. purpurea and I. vomitoria Ait, I. Litseaefolia and I. zhejiangensis, I. hylonoma var. glabra and I. triflora var. kanehirai.

To explore a new approach to generating reproductive sterility in transgenic plants, the barnase gene from Bacillus amyloliquefaciens was placed under the control of an 1853-bp nucleotide sequence from the 3'end of the second intron of Arabidopsis AGAMOUS and CaMV 35S (-60) minimal promoter [AG-I-35S (-60)::Barnase], and was introduced into tobacco through transformation mediated by Agrobacterium tumefaciens. All AG-I-35S (-60)::Barnase transgenic plants showed normal vegetative growth and 28% of the transgenic lines displayed complete ablation of flowering. Two transgenic lines, Bar-5 and Bar-15, were 98.1 and 98.4% sterile, respectively, as determined by seed production and germination. When controlled by AG-I-35S (-60) chimeric promoter, barnase mRNA was detected in the reproductive tissues of transgenic tobacco plants, but not in vegetative parts. This study presents the first application of an AG intron sequence in the engineered ablation of sexual reproduction in plants. The AG-I-35S (-60)::Barnase construct can be useful in diminishing pollen and seed formation in plants, providing a novel bisexual sterility strategy for interception of transgene escape and has other potentially commercial use for transgenic engineering.

Amplified fragment length polymorphism (AFLP) was assessed in 13 Chinese species of Dendrobium. Three primer combinations E+ACT/M+CAC, E+AAC/M+CAC and E+ACA/M+CAC were used for selective amplification. Between 100 bp and 300 bp, a totle of 346 fragments were detected, in which 342 are polymorphic. The rate of polymorphism was 98.8%. The AFLP data were analyzed using Nei & Li similarity coefficient and UPGMA Cluster method. The result indicated that the 13 species were clustered into three groups on coefficient level 0.54. Group I included D. falconeri, D. officinale, D. wilsonii, D. hercoglossum, D. crystallinum. D. hancockii, D. guangxiense, D. primulinum, D. crepidatum, D. thyrsiflorum. Group II included D. chrysotoxum. Group III included D. loddigesii. This outcome was corresponding to the classification result by using the traditional method. It is concluded that AFLP markers can be used on the studies of genetic relationships and classification of Dendrobium.

Filamentous and rod-shaped virions, and aggregated crystals were observed in infected leaves of by negative staining and ultramicrotomy. Histologic study synchronously showed typical crystal forms of the two virions: the crystals from filamentous particles congregated in strips, arrayed in multilayer and piled in certain angles or helix between layers; while the crystals from rod-shaped particles arrayed in parallel, angle-layer or helix. The two kinds of crystals both presented in parenchyma cells, plasmodesma, and vascular bundles; as an evidence that indicates the existence of short distance transport of viruses between cells, plasmodesmata were produced through piercing the membrane around the reproducing viral crystals; the chloroplasts in the infected cells were hypoplastic, and the filamentous virion were observed within the chloroplasts; the mitochondrions were over-developed, swelled or even cavitated; the nucleus were also swelled and cavitated. Further multiplex RT-PCR and sequencing that the coat protein genes simultaneously expanded to Cymbidium mosaic potexvirus (CymMV) and Odontoglossum ringspot tobamovirus (ORSV) showed homology with available abstracts from GenBank, and the respective percentages of identity are 98% and 99%-100%. The instant and direct identification evidences of CymMV and ORSV complex infection on phalaenopsis are revealed at both cellular and molecular levels, and the character of its pathological ultrastructural alteration as the gist in cellular pathology for pathogenesis are also presented.

Objective: To evaluate the role of adiponectin in regulating tumor necrosis factor alpha (TNF alpha) production and preventing fulminant autoimmunological damage of hepatocytes following concanavalin A (Con A) injection into mice., Methods: Three days after recombinant plasmids pAA-neo-mAd were injected into the mice via the tail veins, Con A was injected into the mice. Mice transfected with empty pAA-neo vector served as controls. The serum levels of alanine aminotransferase (ALT), TNF alpha and adiponectin were detected, and histological examination of livers was carried out at different time points after the Con A injection. All results were subjected to statistical analyses., Results: Histological examinations showed that the damage in livers of mice with high serum adiponectin levels was milder than that of the controls. The serum levels of ALT and TNF alpha were both lower than those of the controls (P less than 0.01, respectively). Statistical analyses showed the serum levels of ALT was negatively related to the levels of adiponectin in the sera (r=-0.5034)., Conclusion: Adiponectin is effective in protecting hepatocytes from Con A-induced immunological injury. The mechanism of this protective effect may be caused by inhibiting the synthesis and/or release of TNF alpha.

A sacB mutant was obtained by transposon IS10 inactivation of a plasmid pXT3sacB carrying the sacB gene. Sequencing of this mutant plasmid DNA (GenBank accession No. AY580883.1) showed that the IS10 flanking the 22 bp inverted repeats were 5'-CTGAGAGATCCCCTCATAATTT-3' and 5'-AAATCATTAGGGGATTCATCAG-3', which were the similar to those published in reports previously. However, the target sequence adjacent to IS10 was 5'-TGCTTGGTT-3' instead of the previously reported 5'-NGCTNAGCN-3'. To our knowledge, this is the first report on the novel insertion site of IS10. In addition, Southern blot hybridization confirmed that the mobile IS10 originated from the chromosomal DNA of the host strain Escherichia coli DH5alpha and that there were two copies in the DH5alpha genome.

RAPDs and AFLPs were used to investigate the genetic variation of 14 species of Cymbidium. A great diversity was shown by 12 random primers and three primer combinations. UPGMA cluster analysis showed that the results from the RAPD and AFLP analysis were similar and the genetic distances were closest between C. ensifolium and C. sinense, C. kanran and C. faberi var. omeiense, C. mastersii and C. eburneum respectively.
