Based on the experiments of utilization of garlic processing wastewater in a lotus pond, this study demonstrates that lotus pond wetlands have a remarkable ability to remove organic pollutants and decrease chemical oxygen demand (CODCr), biochemical oxygen demand (BOD5), and suspended substances (SS) in garlic processing wastewater. Results also show evident effects of lotus roots on absorption of NH3-N. The pH value in a lotus pond with wastewater discharged was relatively stable. The water quality in the lotus pond reached the class II emission standard, according to the Integrated Wastewater Discharge Standard (GB8978-1996), seven days after pretreated garlic processing wastewater had been discharged into the lotus pond. Garlic processing wastewater irrigation does not produce pollution in the pond sediment and has no negative effect on the growth of lotus roots. Due to utilization of garlic processing wastewater, the output of lotus roots increased by 3.0% to 8.3%, and the quality of lotus roots was improved. Therefore, better purification and utilization results can be achieved.

In order to explore the synergistic mechanisms of combinatorial treatment using curcumin and mitomycin C (MMC) for breast cancer, MCF-7 breast cancer xenografts were conducted to observe the synergistic effect of combinatorial treatment using curcumin and MMC at various dosages. The synergistic mechanisms of combinatorial treatment using curcumin and MMC on the inhibition of tumor growth were explored by differential gene expression profile, gene ontology (GO), ingenuity pathway analysis (IPA) and Signal–Net network analysis. The expression levels of selected genes identified by cDNA microarray expression profiling were validated by quantitative RT-PCR (qRT-PCR) and Western blot analysis. Effect of combinatorial treatment on the inhibition of cell growth was observed by MTT assay. Apoptosis was detected by flow cytometric analysis and Hoechst 33258 staining. The combinatorial treatment of 100 mg/kg curcumin and 1.5 mg/kg MMC revealed synergistic inhibition on tumor growth. Among 1501 differentially expressed genes, the expression of 25 genes exhibited an obvious change and a significant difference in 27 signal pathways was observed (p < 0.05). In addition, Mapk1 (ERK) and Mapk14 (MAPK p38) had more cross-interactions with other genes and revealed an increase in expression by 8.14- and 11.84-fold, respectively during the combinatorial treatment by curcumin and MMC when compared with the control. Moreover, curcumin can synergistically improve tumoricidal effect of MMC in another human breast cancer MDA-MB-231 cells. Apoptosis was significantly induced by the combinatorial treatment (p < 0.05) and significantly inhibited by ERK inhibitor (PD98059) in MCF-7 cells (p < 0.05). The synergistic effect of combinatorial treatment by curcumin and MMC on the induction of apoptosis in breast cancer cells may be via the ERK pathway.

Nitric oxide (NO), a bioactive signaling molecule, serves as an antioxidant and anti-stress agent under abiotic stress. A hydroponics experiment was conducted to investigate the effects of sodium nitroprusside (SNP), a NO donor, on tomato seedlings exposed to 50 μmol L−1 CuCl2. The results show that copper is primarily stored in the soluble cell sap fraction in the roots, especially after treatment with Cu+SNP treatment, which accounted for 66.2% of the total copper content. The copper concentration gradually decreased from the roots to the leaves. In the leaves, exogenous NO induces the storage of excess copper in the cell walls. Copper stress decreases the proportion of copper integrated with pectates and proteins, but exogenous NO remarkably reverses this trend. The alleviating effect of NO is blocked by hemoglobin. Thus, exogenous NO is likely involved in the regulation of the subcellular copper concentrations and its chemical forms under copper stress. Although exogenous NO inhibited the absorption and transport of excess copper to some extent, the copper accumulation in tomato seedlings significantly increased under copper stress. The use of exogenous NO to enhance copper tolerance in some plants is a promising method for copper remediation.

In the present experiment, effects of sodium hydrosulfide (NaHS), a H2S donor, on the oxidative damage, antioxidant capacity and the growth of cucumber hypocotyls and radicles were studied under 100 mmol L−1 NaCl stress. NaCl treatment significantly induced accumulation of H2O2 and thiobarbituric acid-reactive substances (TBARS) in cucumber hypocotyls and radicles, and application of NaHS dramatically reduced the accumulation of H2O2 and lipid peroxidation. However, the alleviating effects greatly depended on the concentrations of NaHS, and 400 μmol L−1 NaHS treatment showed the most significant effects. Corresponding to the change of lipid peroxidation, higher activities of antioxidant enzymes as well as the antioxidant capacity indicated as DPPH scavenging activity, chelating activity of ferrous ions and hydroxyl radical (·OH) scavenging activity were induced by NaHS treatment under NaCl stress, especially by 400 μmol L−1 NaHS treatment. With the alleviating lipid peroxidation, the amylase activities in cotyledons were increased, and the length of cucumber hypocotyls and radicles were significantly promoted by NaHS treatment under NaCl stress. Unlike the effects of NaHS, pretreatment with other sodium salts including Na2S, Na2SO4, NaHSO4, Na2SO3, NaHSO3 and NaAc did not show significant effects on the growth of cucumber hypocotyls and radicles. These salts do not release H2S. Based on above results, it can be concluded that the effects of NaHS in the experiment depended on the H2S rather than other compounds derived from NaHS, and the alleviating effects might related with its function in modulating antioxidant capacity and amylase activities.

Chinese chive (A. tuberosum Rottler ex Spr.) is one of the most widely cultivated Allium species in China. However, minimal transcriptomic and genomic data are available to reveal its evolution and genetic diversity. In this study, de novo transcriptome sequencing was performed to produce large transcript sequences using an Illumina HiSeq 2000 instrument. We produced 51,968,882 high-quality clean reads and assembled them into 150,154 contigs. A total of 60,031 unigenes with an average length of 631 bp were identified. Of these, 36,523 unigenes were homologous to existing database sequences, 35,648 unigenes were annotated in the NCBI non-redundant (Nr) sequence database, and 23,509 unigenes were annotated in the Swiss-Prot database. A total of 26,798 unigenes were assigned to 57 Gene Ontology (GO) terms, and 13,378 unigenes were assigned to Cluster of Orthologous Group categories. Using the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway database, we mapped 21,361 unigenes onto 128 pathways. Furthermore, 2,125 sequences containing simple sequence repeats (SSRs) were identified. This new dataset provides the most comprehensive resource currently available for gene expression, gene discovery, and future genomic research on Chinese chive. The sequence resources developed in this study can be used to develop molecular markers that will facilitate further genetic research on Chinese chive and related species.

Enteropathy-associated T-cell lymphoma (EATL) is a rare gastrointestinal non-Hodgkin's lymphoma, originating from intraepithelial T-lymphocyte, which is specifically associated with celiac disease. EATL most commonly presents in the sixth and seventh decades of life. We report a unique case of type I EATL in the colon with liver metastasis, which was presented with nonspecific radiological findings and at a very young age (29 years old) compared with previously published data. We suggest that EATL should be regarded as part of differential diagnosis in any patient presenting with abdominal pain, diarrhea, weight loss, and malabsorption because delay in treatment can result in an irreversible clinical outcome. Cardiomyocytes apoptosis can lead to heart failure. Conventional and alternative drugs, such as Chinese herbal remedies, have been developed to target cardiomyoblast cells apoptosis. In this study, we investigated the effects of ginsenoside Rbl (Rbl), an active compound, which is isolated from Notoginseng and Ginseng on isoproterenol-(ISO-) induced apoptosis in rat cardiomyocytes and its mechanism in vivo and in vitro. Rbl reduced the ISO-induced apoptosis in rat cardiomyocytes and H9c2 cells. The effect of Rbl was significantly suppressed by H89 (inhibitor for PKA), but not by C-l (inhibitor for PKC). Based on in-cell blot analysis, the ISO-induced PKA and PKC expressions were decreased by Rbl, which was inhibited by H89, but not by C-l. The expressions of caspase-3 and caspase-9 were decreased after treatment with both ISO and Rbl, but with no change for caspase-8. Our results indicated that Rbl reducing ISO-induced rat cardiomyocytes apoptosis may be involved in PKA and caspase-9 pathways. [ABSTRACT FROM AUTHOR]

Cancer metastasis is refractory to most forms of chemotherapy. Conventional and alternative drugs, such as Chinese herbal remedies, have been developed to target metastatic cancer cells. In this study, we investigated the effects of PC-SPESII, an herbal formulation, on the migration, invasion, and metastasis of an experimental human breast cancer cell line in vivo and in vitro. PC-SPESII suppressed pulmonary metastasis and tumor growth of MDA-MB-231 human breast cancer xenografts without affecting body weight, liver function, and kidney function. PC-SPESII also inhibited MDA-MB-231 cell migration and invasion in vitro in a dose-dependent manner. Based on ELISA analysis, secretion of MMP-2 and MMP-9, proteins associated with extracellular matrix degradation, was reduced in response to PC-SPESII treatment. Western blot analysis of whole-cell extracts revealed that the levels of proteolytic proteins associated with matrix and base membrane degradation (MMP-2, MMP-9, and uPA) were decreased and the levels of their endogenous inhibitors (TIMP1 and TIMP2) were increased. Moreover, the p38MAPK and SAPK/JNK signaling pathway, which stimulates proteolytic enzymes and matrix degradation, was inhibited by PC-PSESII. Remarkably, cotreatment with PC-PSESII and p38MAPK or SAPK/JNK inhibitors magnified the antimetastatic phenotype. Our results indicate that PC-PSESII impairs human breast cancer metastasis by regulating proteolytic enzymes and matrix dynamics through the p38MAPK and SAPK/JNK pathway. [ABSTRACT FROM AUTHOR]

Herbal formulas based on the traditional Chinese medicine (TCM) syndrome (ZHENG) have been used as alternative treatments for breast cancer. However, there is a lack of the experimental animal ZHENG model for the evaluation of the herbal formulas. In this study, we have established 4T1 mouse breast cancer with Liver Fire Invading Stomach Syndrome model (4T1 LFISS mice) and investigated the effects of the herbal formula, Zuo-Jin Wan (ZJW). Our results showed that 4T1 LFISS mice have the features of LFISS including irritability, loss of appetite, yellow urine, chow, and a tail hot. Compared to untreated 4T1 LFISS mice, ZJW significantly reduced tumor weight and volume (P < 0.05), although it was weaker than Cisplatin. However, ZJW significantly increased the body weight and food intake of 4T1 LFISS mice and decreased serum ALT, AST, Cr, and BUN levels and ZHENG score (P < 0.05), while Cisplatin reduced the food intake, and body weight and increased serum ALT, AST, Cr, and BUN levels in 4T1 LFISS mice. Our study has provided a mouse breast cancer ZHENG model and showed that ZJW suppresses tumor growth and improves LFISS and kidney and liver functions in the 4T1 LFISS mice. [ABSTRACT FROM AUTHOR]

The bioglass modified HA-Al2O3 composites were successfully fabricated by mixing HA, synthesized by wet chemical method between precursor materials H3PO4 and Ca(OH)2, with 25wt% Al2O3 and different content of bioglass (5%, 25%, 45%, 65wt%) respectively, with a mole fraction of 53.9%SiO2, 22.6%Na2O, 21.8%CaO, and 1.7wt%P2O5, sintered in air at various temperatures (750-950°C) for 2h. when the content of bioglass is below 45wt% in the composite, HA decomposes completely and transforms to β-TCP. The main phase in this case are β-TCP, Al2O3 and Ca3(AlO3)2.When the content of bioglass is above 45wt% in the composite, the decomposition of HA to β-TCP is suppressed and the main phases in this case are Al2O3 and HA, DCP CaHPO4 β-TCP, which almost have the same chemical composition, forming ternary-glass phase, and have better bioactive than pure HA. It can also be found that at the certain addition of bioglass, the higher sintered temperature, the bigger volume density and flexural strength of the composite are, but when the sintered temperature reaches 950°C, they decrease. Activated pores in the glass-alumina composite during isothermal sintering at 710°C by colloidal processing were extracted by the threshold conversion of the micrograph obtained with a field emission scanning microscope. The relative ratio of the activated pores in the final stage of sintering to that in the initial stage of sintering was calculated. The kinetics equation of the evolution of the activated pores during isothermal sintering was established. The pores evolution confirms the viscous flow of the glass matrix, indicating the validity of the predominant liquid-phase sintering of the composite. [ABSTRACT FROM AUTHOR]

AbstractAim:To examine whether the cell growth inhibitory effect of the combination of baicalin and baicalein is related to apoptosis. Moreover, to determine whether the expression of some apoptosis-related proteins is regulated by the ERK/p38 MAPK pathway.Methods:Cell viability was measured using a 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Apoptosis was detected by acridine orange (AO) staining, DNA ladder assay and flow cytometric analysis. Apoptosis-related proteins were observed using Western blot analysis.Results:Compared with baicalin or baicalein alone, the combination treatment of baicalin (50 μmol/L) and baicalein (25 μmol/L) had an anti-proliferative effect in a time-dependent manner. Isobologram analysis demonstrated that the combination treatment had a synergistic effect. Moreover, apoptosis in MCF-7 cells was increased by 12% and 20% with the combination treatment at 24 h and 48 h, respectively. With the combination treatment in MCF-7 cells, cleaved caspase-3 and caspase-9 were observed, and the level of bcl-2 expression was decreased approximately 20% and 40% at 24 h and 48 h, respectively. The expression of bax and p53 were increased about 25% and 15% at 48 h, respectively. Moreover, the activation of caspase-3, -9 and the regulation of bcl-2, bax and p53 were related to ERK /p38 MAPK activation.Conclusion:In this study, apoptosis was enhanced by the combination treatment of baicalin and baicalein, which activated caspases-3 and caspase-9, downregulated the level of bcl-2 and upregulated the level of bax or p53 via the ERK/p38 MAPK pathway. Cut flower chrysanthemum (Dendranthema grandiflora 'Jinba') plants were treated with 40 °C/35 °C or 33 °C/28 °C (day/night) for 11 days and then transferred to 23 °C/18 °C for 5 days to study the changes in their photosysnthesis and flourescence parameters under high temperature stress and normal temperature recovery. The results showed that on the 5th day of 33 °C/28 °C treatment, net photosynthesis (Pn) decreased gradually and stomatal conductance (Gs) decreased evidently; while after recovery for 5 days, both P n and G s resumed to 80% of the control. At 40 °C/35 °C, Pn and Gs decreased dramatically. The increase of intercellular CO2 concentration (Ci) at the early stage under given high temperatures that the photosynthesis inhibition by high temperature stress was resulted from non-stomatal limitations. However, 9 days later, stomatal limitation became the mainly cause of photosynthesis inhibition by high temperature stress was resulted from non-stomatal limitations. The photochemical quenching (qp) at 33 °C/28 °C descended first and turned to rise then, suggesting that the election transfer was firstly restrained by the stress. Contrastively, qp rose continuously at 40 °C/35 °C, indicating that oxygen-evolving complex (OEC) was the location in chrysanthemum photosynthesis apparatus most sensitive to extreme high temperature. Based on the density functional calculations, the structural and electronic properties of the WS2/graphene heterojunction under different strains are investigated. The calculated results show that unlike the free mono-layer WS2, the monolayer WS2 in the equilibrium WS2/graphene heterojunctionis characterized by indirect band gap due to the weak van der Waals interaction. The height of the schottky barrier for the WS2/graphene heterojunction is 0.13 eV, which is lower than the conventional metal/MoS2 contact. Moreover, the band properties and height of schottky barrier for WS2/graphene heterojunction can be tuned by strain. It is found that the height of the schottky barrier can be tuned to be near zero under an in-plane compressive strain, and the band gap of the WS2 in the heterojunction is turned into a direct band gap from the indirect band gap with the increasing schottky barrier height under an in-plane tensile strain. Our calculation results may provide a potential guidance for designing and fabricating the WS2-based field effect transistors. [ABSTRACT FROM AUTHOR]

Purpose: To investigate the therapeutic effects of Jiaotai pill (JTP) on rats with insomnia induced by pchlorophenylalanine (PCPA). Methods: Rats with PCPA-induced insomnia were divided into 5 groups (n = 10), made up of control group, positive treatment group (estazolam 0.1 mg/kg), and 3 JTP treatment groups (0.6, 1.2 and 2.4 g/kg). Another group of 10 rats were treated as normal group. Rats in normal and control groups were treated with normal saline (10 mL/kg). After 14 days of drug treatment, the rats were injected intraperitoneally with sodium pentobarbital (45 mg/kg) and thereafter, latent period and sleeping time were recorded, while contents of γ-aminobutyric acid (GABA) and glutamic acid (Glu) in hypothalamus were determined by high performance liquid chromatography (HPLC). Furthermore, the expressions of glutamate decarboxylase 65 (GAD-65), glutamate decarboxylase 67 (GAD-67), GABAaminotransferase (GABA)-T, anti-GABA transporter 1 (GAT)-1, anti-GABA transporter (GAT)-3, and GABA receptors (GABA-A and GABA-B) in the hypothalamus were analyzed by western blotting assay. Results: The results showed that JTP (0.6, 1.2 and 2.4 g/kg) significantly shortened latent period and prolonged sleeping time (p < 0.01). JTP also increased GABA level (p < 0.01), but decreased Glu contents of the rat hypothalamus (p < 0.01). Western blotting data indicate that JTP significantly upregulated the levels of GAD-65 (p < 0.01), GAD-67 (p < 0.05), GAT-1 (p < 0.01), GAT-3 (p < 0.01), GABA-A (p < 0.01) and GABA-B (p < 0.01), while the level of GABA-T was down-regulated. Conclusion: The results demonstrate that JTP possesses significant sedative effects on insomnia in rats, most probably through a mechanism involving GABA signal pathway. [ABSTRACT FROM AUTHOR]

Based on the salt-tolerance identification of a series of cucumber rootstock varieties, the cucumber cultivar 'Xintaimici' grafted on different salt-tolerant rootstock varieties 'Sherpa', 'Shintosa' , 'Tielizhen' , and 'Figleaf gourd' was selected to study its seedlings physiological responses to NaCl stress, taking the self-rooted ones as the control. Under the stress of 100 mmol NaCl x L(-1), the leaf electrolyte leakage rate and malondialdehyde (MDA) content of the grafted seedlings were significantly lower than those of self-rooted seedlings, and the seedlings grafted on 'Figleaf gourd' showed the lowest electrolyte leakage rate and malondialdehyde (MDA) content, followed by the seedlings grafted on 'Tielizhen', 'Shintosa' , and 'Sherpa'. The leaf proline and soluble sugar contents and peroxidase (POD), superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX) activities of the grafted seedlings were significantly higher than those of the self-rooted seedlings, and the POD, SOD, CAT, and APX activities were the highest for the seedlings grafted on 'Figleaf gourd' and the lowest for the seedlings grafted on 'Sherpa', but had no significant differences for the seedlings grafted on 'Tielizhen' and 'Shintosa'. The leaf Na+ content of the seedlings grafted on different rootstock varieties ranked as 'Figleaf gourd' < 'Tielizhen' < 'Shintosa' < 'Sherpa', while the leaf K+ content had little difference for the seedlings grafted on 'Figleaf gourd', 'Tielizhen', and 'Shintosa' but was significantly higher than that for the seedlings grafted on 'Sherpa'. The self-rooted seedlings had the highest leaf Na+ content but the lowest leaf K+ content. The leaf Na+/K+ ratio of grafted seedlings was significantly lower than that of self-rooted ones, and the seedlings grafted on 'Figleaf gourd' had the lowest leaf Na+/K+ ratio.
