Purpose: The prevalence of non-alcoholic fatty liver disease (NAFLD) and its related mortality is increasing at an unprecedented rate. Traditional Chinese medicine (TCM) has been shown to offer potential for early prevention and treatment of NAFLD. The new mechanism of “Shenling Baizhu San” (SLBZS) is examined in this study for the prevention and treatment of NAFLD at the preclinical level.Methods: Male C57BL/6J mice were randomly divided into three groups: normal diet (ND), western diet + CCl4 injection (WDC), and SLBZS intervention (WDC + SLBZS). Body weights, energy intake, liver enzymes, pro-inflammatory factors, and steatosis were recorded in detail. Meanwhile, TPH1, 5-HT, HTR2A, and HTR2B were tested using qRT-PCR or ELISA. Dynamic changes in the gut microbiota and metabolites were further detected through the 16S rRNA gene and untargeted metabolomics.Results: SLBZS intervention for 6 weeks could reduce the serum and liver lipid profiles, glucose, and pro-inflammatory factors while improving insulin resistance and liver function indexes in the mice, thus alleviating NAFLD in mice. More importantly, significant changes were found in the intestinal TPH-1, 5-HT, liver 5-HT, and related receptors HTR2A and HTR2B. The 16S rRNA gene analysis suggested that SLBZS was able to modulate the disturbance of gut microbiota, remarkably increasing the relative abundance of probiotics (Bifidobacterium and Parvibacter) and inhibiting the growth of pro-inflammatory bacteria (Erysipelatoclostridium and Lachnoclostridium) in mice with NAFLD. Combined with metabolomics in positive- and negative-ion-mode analyses, approximately 50 common differential metabolites were selected via non-targeted metabolomics detection, which indicated that the targeting effect of SLBZS included lipid metabolites, bile acids (BAs), amino acids (AAs), and tryptophan metabolites. In particular, the lipid metabolites 15-OxEDE, vitamin D3, desoxycortone, and oleoyl ethanol amide were restored by SLBZS.Conclusion: Integrating the above results of multiple omics suggests that SLBZS ameliorates NAFLD via specific gut microbiota, gut-derived 5-HT, and related metabolites to decrease fat accumulation in the liver and inflammatory responses.

Purpose Owing to the mortality associated with metastatic prostate cancer and the shortcomings of the current parameters in predicting the disease prognosis, we require the identification of viable biomarkers, which would help in the diagnosis and prognosis of the disease. We aimed to determine whether the interleukin-8 level in the tumor microenvironment could serve as a potential clinical diagnostic marker and prognostic factor for prostate cancer. Methods The migration assay of prostate cancer cells was performed in an in vitro co-culture model. Cell lines PC3 and DU145 were divided into two groups and co-cultured with M0 and M2 macrophages, respectively. We used reverse transcription-quantitative polymerase chain reaction to detect M2 macrophage marker expression levels. Immunohistochemistry analyses of tissue microarrays were performed to analyze the correlation between the increased expression of interleukin-8 and the prognosis of prostate cancer. A retrospective analysis based on 142 residual serum specimens was performed to analyze the level of interleukin-8. Results We observed that M2 macrophages promoted the migration of prostate cancer cells and significantly increased the concentrations of interleukin-8 in the co-culture supernatants. We observed increased expression of CD163 and interleukin-8 in prostate cancer tissues. Furthermore, the levels of interleukin-8 in the serum of prostate cancer patients were higher than those in healthy controls. Untreated patients had higher levels of interleukin-8, which could be a predictor of a higher metastasis rate. Conclusion These results suggest that interleukin-8 produced via bidirectional communication between prostate cancer cells and M2 macrophages is a putative biomarker for prostate cancer diagnosis and treatment.

Research on the efficiency and quality issues faced in aircraft assembly was conducted in this article. A new method of human–machine collaborative riveting was proposed, which combined the flexibility of manual collaboration with the precise control of automatic riveting. The research works include: (1) a theoretical model of pneumatic hammer riveting was established to clarify the principle and parameters of riveting process. (2) A smart bucking bar was designed to support the data collection and extraction of manual collaborative riveting process. (3) An automatic riveting experimental platform was designed to test the automatic riveting process incorporating the extracted manual riveting process parameters, and further an optimization strategy was proposed for the automatic riveting process. (4) A human–machine collaborative riveting experimental platform was developed to conduct the verification work. Through the theoretical analysis, experimental research, system scheme design, and process parameters optimization, the application and verification of human–machine collaborative assembly technology have been achieved. This technology is expected to be comprehensively promoted in the field of aircraft manufacturing, and for breaking through the current difficulties of low production efficiency and poor assembly quality control.

The global pandemic COVID-19 has led to an increase in the number of people purchasing food online, which has brought to a higher demand on the food supply chain. Such as the need to collect more information related to food safety and quality in real-time. Strengthening management of food logistics information flow can reduce food loss and waste and bring better quality and safety of food to consumers. In this review, the importance and applicability of RFID (Radio Frequency Identification) technology to smart food packaging are described. This study emphasizes the recent advancement of the RFID tags in humidity, temperature, gas, pH, integrity, and traceability sensor applications in connection with food packaging. RFID sensors are more suitable for smart packaging both in terms of sensing ability and data transmission. A simpler, low-cost, more robust and less power-demanding sensors network is the development direction of smart packaging in the future. Chipless RFID sensors have the potential to achieve these functions. But it still faces many challenges to be overcome. For example, biocompatible, cost, reading range, multi-tag collision, multi-parameter sensors, recycling issues, security and privacy of RFID system should be solved.

Abstract Background Non-small cell lung cancer (NSCLC) is a common type of lung cancer. Extracellular vehicles (EVs) are nano-sized particles containing proteins, lipids, and miRNAs secreted by various cells, which play important roles in the development of lung cancer by regulating a wide range of signaling pathways. This study focused on elucidating a potential mechanism by which EVs promote the development of NSCLC. Methods Expression levels of miR-744, SUV39H1, Smad9, and BMP4 in clinical tissue samples of NSCLC patients and cell lines were quantified by RT-qPCR and/or western blot analysis. The interaction between SUV39H1 and miR-744 was identified by dual-luciferase reporter assay. miR-744, SUV39H1, and BMP4 expression levels were modulated in A549 and H460 cells, in order to evaluate their effects on cell proliferation, colony formation and cell cycle. A NSCLC xenograft mouse model was used to verify the in vitro findings. NSCLC cell-derived EVs and normal bronchial epithelial cell-derived EVs were isolated and their roles in NSCLC development were evaluated in vivo and in vitro. Results miR-744 expression was lower in cancer cell-derived derived EVs than in normal lung epithelial cell-derived EVs. Reduced miR-744 expression in EVs upregulated SUV39H1 in NSCLC cells and further increased BMP4 levels to promote NSCLC development. BMP4 was upregulated in NSCLC cells upon suppression of Smad9 mediated by SUV39H1. Reduced miR-744 expression transferred from cancer cell-derived EVs into NSCLC cells enhanced cancer development. Conclusion Overall, our findings unveiled a mechanism whereby miR-744 delivered by NSCLC-derived EVs upregulated SUV39H1, activating the Smad9/BMP9 axis and thus promoted cancer development.

Abstract A method with high sensitivity, good accuracy and fast response is of ever increasing importance for the simultaneous detection of AA, DA and UA. In this paper, a simple and sensitive electrochemical sensor, which based on the polyvinylpyrrolidone (PVP)-graphene composite film modified glassy carbon electrode (PVP-GR/GCE), was presented for detecting ascorbic acid (AA), dopamine (DA) and uric acid (UA) simultaneously. The PVP-GR/GCE has excellent electrocatalytic activity for the oxidation of AA, DA and UA. The second-order derivative linear sweep voltammetry was used for the electrochemical measurements. The peak potential differences of DA-AA, DA-UA, and UA-AA (measured on the PVP-GR/GCE) were 212, 130 and 342 mV respectively. Besides, the over potential of AA, DA and UA reduced obviously, so did the peak current increase. Under the optimum conditions, the linear ranges of AA, DA and UA were 4.0 μM–1.0 mM, 0.02–100 μM, and 0.04–100 μM, respectively. The detection limits were 0.8 μM, 0.002 μM and 0.02 μM for AA, DA, and UA. The electrochemical sensor presented the advantages of high sensitivity and selectivity, excellent reproducibility and long-term stability. Furthermore, the sensor was successfully applied to the analysis of real samples.

Aptamers are special types of single-stranded DNA generated by a process called systematic evolution of ligands by exponential enrichment (SELEX). Due to significant advances in the chemical synthesis and biotechnological production, aptamers have gained considerable attention as versatile building blocks for the next generation of soft materials. Hydrogels are high water-retainable materials with a three-dimensional (3D) polymeric network. Aptamers, as a vital element, have greatly expanded the applications of hydrogels. Due to their biocompatibility, selective binding, and molecular recognition, aptamer-based hydrogels can be utilized for bioanalytical and biomedical applications. In this review, we focus on the latest strategies of aptamer-based hydrogels in bioanalytical and biomedical applications. We begin this review with an overview of the underlying design principles for the construction of aptamer-based hydrogels. Next, we will discuss some bioanalytical and biomedical applications of aptamer-based hydrogel including biosensing, target capture and release, logic devices, gene and cancer therapy. Finally, the recent progress of aptamer-based hydrogels is discussed, along with challenges and future perspectives.

A novel tantalum pentoxide nanoparticle-electrochemically reduced graphene oxide nanocomposite-modified glassy carbon electrode (Ta2O5-ErGO/GCE) was developed for the detection of oxytetracycline in milk. The composition, structure and morphology of GO, Ta2O5, and Ta2O5-ErGO were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM). Oxytetracycline electrochemical behavior on the bare GCE, GO/GCE, ErGO/GCE, and Ta2O5-ErGO/GCE was studied by cyclic voltammetry. The voltammetric conditions (including scan rate, pH, deposition potential, and deposition time) were systematically optimized. With the spacious electrochemical active area, the Ta2O5-ErGO/GCE showed a great magnification of the oxidation signal of oxytetracycline, while that of the other electrodes (GCE, GO/GCE, ErGO/GCE) could not reach the same level. Under the optimum conditions, the currents were proportional to the oxytetracycline concentration in the range from 0.2 to 10 μM, and a low detection limit of 0.095 μM (S/N = 3) was detectable. Moreover, the proposed Ta2O5-ErGO/GCE performed practically with satisfactory results. The preparation of Ta2O5-ErGO/GCE in the current work provides a minor outlook of detecting trace oxytetracycline in milk.

In this paper, an electrochemical method for the measurement of tryptophan (Trp) was developed based on a glassy carbon electrode modified with polyvinylpyrrolidonefunctionalized graphene (PVP-GR)/glassy carbon electrode (GCE). In 0.1 M phosphate buffer solution (PBS, pH = 2.2), compared with bare GCE, PVP/GCE, and GR/GCE, the oxidation peak current of Trp increased dramatically at PVP-GR/GCE. The oxidation mechanism of Trp on the PVP-GR/GCE was discussed and the experimental conditions were optimized. Under the best experimental conditions, the oxidation peak current of Trp was proportional to its concentration in the range of 0.06 µM−10.0 µM and 10.0−100.0 µM, and the limit of detection (LOD) was 0.01 µM (S/N = 3). The target modified electrode with excellent repeatability, stability and selectivity, was successfully applied to detectTrp in drugs and biological samples.

Due to multi-factor coupling behavior, the performance evaluation of an accelerometer subject to high-temperature and high-impact loads poses a significant challenge during its design phase. In this paper, the simulation-based method is applied to optimize the design of the accelerometer. The proposed method can reduce the uncertainties and improve the fidelity of the simulation in the sense that (i) the preloading conditions of fasteners are taken into consideration and modeled in static analysis; (ii) all types of loadings, including bolt preloads, thermal loads, and impact loads, are defined in virtual dynamic prototype of the accelerometer. It is our finding that from static and dynamic analysis, an accelerometer is exposed to the risk of malfunction and even a complete failure if the temperature rises to a certain limit; it has been proved that the thermal properties of sensing components are the most critical factors for an accelerometer to achieve its desired performance. Accordingly, we use a simulation-based method to optimize the thermal expansion coefficient of the sensing element and get the expected design objectives.

In this paper, a tryptophan (Trp) molecularly imprinted chitosan film was prepared on the surface of an acetylene black paste electrode using chitosan as the functional polymer, Trp as the template molecule and sulfuric acid as the crosslinking agent. The surface morphologies of non-imprinted and imprinted electrodes were characterized by scanning electron microscopy (SEM). The formation of hydrogen bonds between the functional polymer and the template molecule was confirmed by infrared spectroscopy. Some factors affecting the performance of the imprinted electrode such as the concentration of chitosan, the mass ratio of chitosan to Trp, the dropping amount of the chitosan-Trp mixture, the solution pH, and the accumulation potential and time were discussed. The experimental results show that the imprinted electrode exhibit good affinity and selectivity for Trp. The dynamic linear ranges of 0.01−4 μM, 4−20 μM and 20−100 μM were obtained by second derivative linear sweep voltammetry, and the detection limit was calculated to be 8.0 nM. The use of the imprinted electrode provides an effective method for eliminating the interference of potentially interfering substances. In addition, the sensor has high sensitivity, reproducibility and stability, and can be used for the determination of Trp in pharmaceutical preparations and human serum samples.

In this paper, the preparation, characterization, and electrochemical application of Cu2O nanoparticles and an electrochemical reduced graphene oxide nanohybrid modified glassy carbon electrode (denoted as Cu2O NPs‒ERGO/GCE) are described. This modified electrode was used as an electrochemical sensor for the catalytic oxidation of rhodamine B (RhB), and it exhibited an excellent electrochemical performance for RhB. The oxidation potential of RhB was decreased greatly, and the sensitivity to detect RhB was improved significantly. Under optimum conditions, a linear dynamic range of 0.01−20.0 μM and a low detection limit of 0.006 μM were obtained with the Cu2O NPs‒ERGO/GCE by using second‒order derivative linear sweep voltammetry. In addition, the selectivity of the prepared modified electrode was analyzed for the determination of RhB. The practical application of this sensor was investigated for the determination of RhB in food samples, and satisfactory results were obtained.

In this paper, a nanocomposite of cuprous oxide and electrochemically reduced graphene oxide (Cu2O−ERGO) was prepared by a simple and low-cost method; hereby, a new method for the electrochemical determination of tryptophan (Trp) by this composite modified glassy carbon electrode (GCE) is proposed. The prepared materials and modified electrodes were characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD), and cyclic voltammetry (CV). The results showed that Cu2O−ERGO/GCE had good electrocatalytic activity for Trp. The effects of supporting electrolyte, scanning rate, accumulation potential, and accumulation time on the determination of Trp were studied. Under the optimum experimental conditions, Trp was quantitatively analyzed by square-wave voltammetry (SWV). The oxidation peak current of Trp had a good linear relationship with its concentration in the range of 0.02−20 μM, and the detection limit was 0.01 μM (S/N = 3). In addition, the modified electrode has high sensitivity, good repeatability, and long-term stability. Finally, the proposed method has been successfully applied in the determination of Trp concentration in practical samples.

l-tryptophan is one of the eight kinds of essential amino acids for sustainable human life activity. It is common to detect the concentration of tryptophan in human serum for diagnosing and preventing brain related diseases. Herein, in this study, GCE (glassy carbon electrode) modified by Ta2O5-reduced graphene oxide (-rGO) composite (Ta2O5-rGO-GCE) is synthesized by the hydrothermal synthesis-calcination methods, which is used for detecting the concentration of tryptophan in human serum under the as-obtained optimal detection conditions. As a result, the obtained Ta2O5-rGO-GCE shows larger electrochemical activity area than other bare GCE and rGO-GCE due to the synergistic effect of Ta2O5 NPs and rGO. Meanwhile, Ta2O5-rGO-GCE shows an excellent response to tryptophan during the oxidation process in 0.1 M phosphate buffer solution (pH = 6). Moreover, three wide linear detection range (1.0−8.0 μM, 8.0−80 μM and 80−800 μM) and a low limit of detection (LOD) of 0.84 μM (S/N = 3) in the detection of tryptophan are also presented, showing the larger linear ranges and lower detection limit by employing Ta2O5-rGO-GCE. Finally, the as-proposed Ta2O5-rGO-GCE with satisfactory recoveries (101~106%) is successfully realized for the detection of tryptophan in human serum. The synthesis of Ta2O5-rGO-GCE in this article could provide a slight view for the synthesis of other electrochemical catalytic systems in detection of trace substance in human serum.

4-nitrophenol (4-NP) is a hazardous waste and a priority toxic pollutant identified by US Environmental Protection Agency (EPA). Hence, in this paper, a voltammetric sensor was proposed for the direct and sensitive detection of 4-nitrophenol (4-NP) at nanomolar level in complex matrices by using graphene and acetylene black paste hybridized electrode (GR/ABPE). Under optimal conditions, the calibration curve demonstrates a linear relationship for 4-NP in the range from 20 nM to 8.0 μM and 8.0 μM to 0.1 mM separately with the detection limit of 8.0 nM. In addition to it, the performance of the GR/ABPE in practical applications was evaluated by detecting 4-NP in various water samples, and satisfactory recoveries were realized. Therefore, GR/ABPE may have a great potential application for facile and sensitive detection of 4-NP in complex matrices at nanomolar level.

The nanohybrid of electrochemically-reduced graphene oxide (ERGO) nanosheets decorated with MnO2 nanorods (MnO2 NRs) was modified on the surface of a glassy carbon electrode (GCE). Controlled potential reduction was applied for the reduction of graphene oxide (GO). The characterization was performed by scanning electron microscopy, X-ray diffraction and cyclic voltammetry. Compared with the poor electrochemical response at bare GCE, a well-defined oxidation peak of sunset yellow (SY) was observed at the MnO2 NRs-ERGO/GCE, which was attributed to the high accumulation efficiency as well as considerable electrocatalytic activity of ERGO and MnO2 NRs on the electrode surface. The experimental parameters for SY detection were optimized in detail. Under the optimized experiment conditions, the MnO2 NRs-ERGO/GCE showed good linear response to SY in concentration range of 0.01–2.0 μM, 2.0–10.0 μM and 10.0–100.0 μM with a detection limit of 2.0 nM. This developed method was applied for SY detection in soft drinks with satisfied detected results.

A new electrochemical sensor for nanomolar rutin detection based on amine-functionalized Fe3O4 nanoparticles and electrochemically reduced graphene oxide nanocomposite modified glassy carbon electrode (NH2-Fe3O4 NPs-ErGO/GCE) was fabricated through a simple method, and the X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscope (SEM), transmission electron microscope (TEM), vibrating sample magnetometer (VSM) and electrochemical technique were used to characterize the modified electrode. The electrochemical behavior of rutin on the Fe3O4 NPs-ErGO/GCE was studied in detail. The electrochemical response of rutin at this modified electrode was remarkably higher than that of the bare GCE or other modified GCE (GO/GCE, Fe3O4 NPs-GO/GCE, and ErGO/GCE). Under the optimum determination conditions, Fe3O4 NPs-ErGO/GCE provided rutin with a broader detection range of 6.0 nM–0.1 µM; 0.1–8.0 µM and 8.0–80 µM, a minimum detectable concentration of 4.0 nM was obtained after 210 s accumulation. This novel method was applied in determination of rutin in pharmaceutical tablets and urine samples with satisfactory results.

In this paper, MnO2 nanowires-decorated reduced graphene oxide composite (MnO2 NWs-rGO) was synthesized by physical adsorption and controlled potential reduction. Means of X-ray diffraction (XRD), scanning electron microscopy (SEM), and cyclic voltammetry (CV) were used to characterize this material. Glassy carbon electrode (GCE) modified with MnO2 NWs-rGO composite was fabricated by drop-coating method for the sensitive determination of bisphenol A (BPA). Electrochemical behaviors of BPA at the MnO2 NWs-rGO/GCE were studied using CV and chronocoulometry (CC). Experimental results found that MnO2 NWs-rGO/GCE exhibited excellent electrocatalytic activity, sensitivity, stability, and selectivity towards BPA. Under the optimized experimental conditions, quantitative analysis was carried out using the second derivative linear sweep voltammetry (SDLSV), and the MnO2 NWs-rGO/GCE revealed a good linear response to BPA concentration ranging from 0.02–20 μM and 20–100 μM, and the detection limit was 6.0 nM (S/N = 3). This new method possessed some obvious advantages, such as high determination sensitivity, simple preparation process, rapid response and low cost. The sensing performance of MnO2 NWs-rGO/GCE was demonstrated for the detection of BPA in plastic samples, and satisfactory results have been obtained. Nitrite (NO2-) has been extensively applied in agricultural and industrial products and is often found in various foodstuff, tap water, biological samples and environmental systems. However, NO2- as a toxic contaminant probably threaten the human health by producing highly carcinogenic N-nitrosamines. Compared with the traditional analytical techniques, electroanalytical method has considerable advantages such as cost-effective, rapidness, facile operation, and easy miniaturization. Graphene nanocomposites have significant synergistic electrocatalytic effect toward the nitrite redox, which could eventually amplify the electrochemical response signals, and improve the selectivity, sensitivity, and practicability for the nitrite detection in various real samples. The recent developments on graphene-based nitrite electrochemical sensors are reviewed from the view of sensing materials, including graphene, metal nanoparticle/graphene composites, nanostructured metal compound/graphene composites, polymer/graphene composites, MOF/graphene composites, enzyme/graphene composites, MWCNT/graphene composites, and electron mediator/graphene composites. Moreover, the sensing performances including detection ranges, limit of detection (LOD) and sensitivity are tabulated. Finally, the major drawbacks, opportunities and challenges of graphene-based nitrite electrochemical sensors are also discussed. [ABSTRACT FROM AUTHOR]

A facile voltammetric sensor based on MnO2 nanorods/electro-reduced graphene oxide composite modified glassy carbon electrode (MnO2NRs-ERGO/GCE) was proposed for the detection of Rhodamine B (RhB). The MnO2NRs-ERGO/GCE was fabricated by a facile drop-casting approach followed by a potentiostatic reduction technique. A well-shaped oxidation peak of RhB was observed at the MnO2NRs-ERGO/GCE, and the response peak current was 17-fold and 2-fold higher than that of bare GCE and ERGO/GCE respectively, attributing to the synergistic electrocatalytic effect from ERGO sheets and MnO2NRs. Under the optimal analytical conditions, the response peak currents linearly increased with the RhB concentrations varying from 0.02 µM to 1.0 µMand from 1.0 µMto 20 µM. The limit of detection (LOD, S/N=3) was 6.0 nM. Furthermore, the MnO2NRs-ERGO/GCE showed good selectivity, repeatability, reproducibility and stability. Finally, the MnO2NRs-ERGO/GCE was successfully employed to detect RhB in different commercial chili-containing foodstuffs, and the results were in good accordance with the values that detected by high-performance liquid chromatography. Combining with low cost and portable electrochemical apparatus, the proposed sensor is especially suitable for the in-situ detection of RhB in actual food samples. [ABSTRACT FROM AUTHOR]

We investigate fluorescence (FL) and spontaneous parametric four-wave mixing (SP-FWM) signals in V-, Λ- and N-type energy level systems by changing the frequency detuning or power of the dressing field in Pr3+ doped Y2SiO5 at 77 K. The lifetime of the two kinds of signals has been calculated, and the desired experimental results have been obtained. We successfully control the intensity and lifetime of fluorescence and SP-FWM signals by changing the power or frequency detuning of the dressing field.
