35. Study of TRP Channels by Automated Patch Clamp Systems.

Abstract

Ion channels are responsible for the permeation of ions across the membrane and their central role in cellular physiology is well established. Historically, the direct study of ion channels has been considered technically challenging. As such, a significant barrier to drug discovery for ion channels has been the low throughput of high quality electrophysiological data. The emergence of automated high throughput platforms for studying ion channel kinetics and pharmacology has lowered this barrier. Ion channels are now recognized as increasingly important drug targets and a diverse range of ion channels are implicated in a variety of drug discovery and cardiac safety assessment programs. The TRP (Transient Receptor Potential) superfamily of ion channels play a crucial role in a broad range of sensory functions including vision, taste, olfaction, hearing, touch, pain and thermosensation. Many of the TRP channels are polymodal in their activation and deactivation mechanisms and even with conventional patch clamp electrophysiology, the TRP channels are considered to be a very complex target class. Abstract

Cancer rehabilitation is a process that assists the individual with a cancer diagnosis to obtain optimal physical, social, psychological, and vocational functioning within the limits created by the disease and its treatment. The functional autonomy of patients with cancer is compromised. The severity ranges from negligible to profound at the extremes. Owing to the nature of the cancer trajectory, rehabilitative goals have been divided into preventive, restorative, supportive, and palliative. Cancer not only affects the quality of life (QOL) of the individuals with the disease but also that of their caregivers. The primary aim of anticancer treatment has been to cure patients of their disease. However, the American Society of Clinical Oncology has emphasized to its members the importance of (I) identification and management of specific patient groups requiring rehabilitation and (II) recognition of the point in the illness trajectory when treatments should focus more on symptom management and psychosocial support for the patient. There is also recognition that psychosocial care is an essential component of a comprehensive approach to the treatment and rehabilitation of people with cancer, both in the short term and long term. The McGill Cancer Nutrition-Rehabilitation (CNR) Program was set up to use an interdisciplinary approach to empower individuals who are experiencing loss of function, fatigue, malnutrition, psychological distress, and other symptoms as a result of cancer or its treatment to improve their own quality of life. Abstract

Anxiety and depression can be viewed as part of a broader construct of psychologic distress. We chose to focus on anxiety and depression rather than on psychologic distress for several reasons. First, the literature on anxiety and depression can be more readily identified than the literature on the broader and less well defined construct of psychologic distress. Second, as described in the following, criteria have been established for the diagnosis of clinical syndromes of anxiety and depression; no similar criteria exist for the diagnosis of clinical syndromes of psychologic distress. Third, as described here, this approach allows us to build upon the findings of several previous systematic reviews and meta-analyses that also have focused on psychosocial and pharmacologic approaches to the management of anxiety and depression in adult cancer patients.
