Hepatic iron overload in human due to over consumption of iron supplements or hereditary gene mutation and proteome complications, can cause a series of domino effect leading up to hepatic steatosis, nonalcoholic steatohepatitis and hepatocellular carcinoma. Here, we applied Perl's Prussian Blue stain to confirm iron overload in SK-Hep-1 cells after exogenous treatment. To differentiate intracellular from exogenous iron, we utilized soft X-ray tomography (SXT), at beamline 24A of Taiwan Photon Source (TPS 24A), and reveal subcellular localization or iron deposits, especially in the iron's L-edge absorption at 700 and 711 eV. Furthermore, we dissected deeper, via SXT, revealing some degree of morphological change in the Fe-treated cell's mitochondria, and likely conspiring various adverse responses due to this iron overload.
