The Cantang grouper (Epinephelus sp.) is one of the export commodities in which the production is keep increasing. One of the diseases that often attack the cantang grouper is Viral Nervous Necrosis caused by the Betanodavirus genus. Vaccination is an administration of vaccinesto increase the fish body's immune system against the Viral Nervous Necrosis virus infection. This study aims to develop a Chlorella Vulgaris recombinant protein nanovaccine based on adjuvant polymer nanoparticles to improve the immunity performance against hematological responses (total erythrocytes, total leukocytes, differential leukocytes) in the Cantang grouper infected with VNN. The method used was experimental with a completely randomized design (CRD) consisting of eight treatments and three replications, namely K+ (cantang grouper infected with VNN), K-(Healthy cantang grouper), P1 (grouper Healthy cantang grouper was given 33 ul of vaccine), P2 (Healthy cantang grouper was given vaccine of 66 ul), P3 (Healthy cantang grouper was given 112 ul of vaccine), P4 (Healthy cantang grouper was given 33 ul of vaccine and then tested against Viral Nervous Necrosis) P5 (Healthy cantang grouper was given 66 ul vaccine and then tested against Viral Nervous Necrosis), P6 (Healthy cantang grouper was given 112 ul vaccine and then tested against Viral Nervous Necrosis). The fish used were 8-10 cm in size, with a maintenance period of 28 days. The results showed that the administration of Chlorella Vulgaris recombinant protein vaccine based on adjuvant polymer nanoparticles had an effect on the hematological response of cantang grouper infected with VNN. The best dose of recombinant p-PERCv vaccine based on adjuvant polymer nanoparticles is 33 ul (P4), judging from the hematological status such as total red blood cells 2.14x10
