Lukas fish (Puntius bramoides) is one of the common fishery resources in the Banyumaswaters. Lukas fish was domesticated and has an economic value. Some species of Cyprinidae family like Lukas fish are become difficult to find in Serayu river. Accordingly, the reproductive technology study, such as cryopreservation are needed to keep these species exist in the wild. The purpose of this study was to determine the fertility of sperm Lukas fish in captivity. Fish sperm analysis was carried out by analyzing the sperm motility, viability and abnormalities, respectively. The results showed that the highest percentage of sperm motility, viability at 100x sperm dilution 84.71 ± 3.53%, 84.78 ± 1.41%, and the lowest percentage abnormalities 16 ± 2.07%. This result indicates that Lukas sperm has high fertility. The LSD test expresses the optimum sperm motility and viability showed significant differences at 100x milt dilution (p<0.05).
