Abstract

This book is a joint publication of the National Science Teachers Association (NSTA) and the Society of Protozoologists and is the result of efforts to find a way in which protozoa research can be used to teach biology. This program puts cutting edge science into the hands of science teachers and enables students to experience a variety of hands-on activities. There are 28 investigations grouped into five sections: (1) "Protozoology Laboratory Skills"; (2) "Comparative Physiology"; (3) "Organism Interdependence"; (4) "Comparative Ecology"; and (5) "Adaptive Strategies." Each of the investigations has a student section, and a teacher section that provides materials to be photocopied for students and background information on the topic to be investigated. Appendices contain student evaluation forms; details about making lab ware, caring for microscopes, and collecting, culturing, and maintaining microorganisms; and a bibliography of reference and resource materials. Abstract

Dataset: RV Atlantic Explorer BATS BV50: CO2 and PO4, Bulk and cell-specific CO2 fixation and PO4 uptake from Atlantic Explorer cruise AE1524 (BATS validation cruise BV50), September 2015. Phosphate uptake rates were measured in Prochlorococcus, Synechococcus, pigmented eukaryotes, and unpigmented eukaryotes. Also reported are CO2 fixation rate by Prochlorococcus, Synechococcus, and pigmented eukaryotes. Abstract

Protists are the most diverse eukaryotes. These microbes are keystone organisms of soil ecosystems and regulate essential processes of soil fertility such as nutrient cycling and plant growth. Despite this, protists have received little scientific attention, especially compared to bacteria, fungi and nematodes in soil studies. Recent methodological advances, particularly in molecular biology techniques, have made the study of soil protists more accessible, and have created a resurgence of interest in soil protistology. This ongoing revolution now enables comprehensive investigations of the structure and functioning of soil protist communities, paving the way to a new era in soil biology. Instead of providing an exhaustive review, we provide a synthesis of research gaps that should be prioritized in future studies of soil protistology to guide this rapidly developing research area. Based on a synthesis of expert opinion we propose 30 key questions covering a broad range of topics including evolution, phylogenetics, functional ecology, macroecology, paleoecology, and methodologies. These questions highlight a diversity of topics that will establish soil protistology as a hub discipline connecting different fundamental and applied fields such as ecology, biogeography, evolution, plant-microbe interactions, agronomy, and conservation biology. We are convinced that soil protistology has the potential to be one of the most exciting frontiers in biology.

8. Effects of variations in light intensity on life processes of the planktonic foraminifer Globigerinoides Sacculifer in laboratory culture

Abstract

The planktonic foraminifer Globigerinoides sacculifer (Brady) was cultured under two different light intensities and in continuous darkness. High light intensity (HLI = 4oo-soo einsteins/m2/s) resulted in a longer lifespan, a greater number of chambers formed, and a larger final shell size compared with individuals cultured under low light intensity (LLI = 20-50 einsteins/m2/s) or in continuous darkness. Shell growth rates were unaffected by increasing light intensity, but gametogenesis was delayed. Continuous darkness induced a rapid onset of gametogenesis in organisms with shell lengths larger than 250 m. Feeding frequency had a greater effect on growth and reproduction than light intensity under conditions of LLI and HLI, but continuous darkness had an overriding effect on growth and reproduction owing to the rapid onset of gametogenesis which terminated the life of the mother cell. Our previous data indicated that the longevity of G. sacculifer was dependent on feeding frequency, and that G. sacculifer cultured under LLI had a lifespan of approximately 2-4 weeks. Present results suggest that the lifespan can vary from a minimum of 8 days for organisms fed daily in continuous darkness to a maximum of 54 days for organisms fed once every 7 days and maintained in HLI. It is concluded that individual G. sacculifer attain a shell size greater than 6oo ,urn only if they maintain their position in the euphotic zone. Prolonged existence below the euphotic zone would result in premature death or gametogenesis following stunted shell growth.

Abstract

The majority of amoeboid lineages with flattened body forms are placed under a taxonomic hypothetical class Discosea sensu Smirnov et al. (2011), which encompasses some of the most diverse morphs within Amoebozoa. However, its taxonomy and phylogeny is poorly understood. This is partly due to lack of support in studies that are based on limited gene sampling. In this study we use a phylogenomic approach including newly-generated RNA-Seq data and comprehensive taxon sampling to resolve the phylogeny of Discosea. Our analysis included representatives from all orders of Discosea and up to 550 genes, the largest gene sampling in Amoebozoa to date. We conducted extensive analyses to assess the robustness of our resulting phylogenies to effects of missing data and outgroup choice using probabilistic methods. All of our analyses, which explore the impact of varying amounts of missing data, consistently recover well-resolved and supported groups of Amoebozoa. Our results neither support the monophyly nor dichotomy of Discosea as defined by Smirnov et al. (2011). Rather, we recover a robust well-resolved clade referred to as Eudiscosea encompassing the majority of discosean orders (seven of the nine studied here), while the Dactylopodida, Thecamoebida and Himatismenida, previously included in Discosea, are non-monophyletic. We also recover novel relationships within the Eudiscosea that are largely congruent with morphology. Our analyses enabled us to place some incertae sedis lineages and previously unstable lineages such as Vermistella, Mayorella, Gocevia, and Stereomyxa. We recommend some phylogeny-based taxonomic amendments highlighting the new findings of this study and discuss the evolution of the group based on our current understanding.
