Maternal stress during pregnancy can cause alterations to the fetal hypothalamus-pituitary-adrenal (HPA) axis, a phenomenon known as fetal programming that may have lasting effects on offspring outcomes, including depression. Evidence suggests that these effects may vary with respect to the offspring's genetic risk. Nonetheless, few studies have examined these effects into adulthood, when risk for depression onset is highest. The present study builds upon the extant literature by examining the interaction of maternal prenatal perceived stress (MPPS) and offspring HPA-axis polygenic risk to predict offspring depression in early adulthood. A total of 381 mother-child dyads participated in a prospective, longitudinal study that spanned from pregnancy until offspring were 20 years of age. Polygenic risk was defined by a multilocus genetic profile score (MGPS) that reflected the additive risk of three HPA-axis candidate genes. The results indicated that the interaction of MPPS and HPA-axis MGPS confers risk for offspring depression at age 20, in line with the differential susceptibility model. This interaction may be specific to prenatal stress, as maternal stress during early childhood did not interact with genetic risk to predict depression. These findings provide the first evidence that genetic variants that are associated with the HPA axis may act in a polygenic, additive fashion to moderate the association between fetal programming and adult depression.

IntroductionLittle is known about onward HIV transmissions from people living with HIV (PLWH) in care. Antiretroviral therapy (ART) has increased in potency, and treatment as prevention (TasP) is an important component of ending the epidemic. Syndemic theory has informed modelling of HIV risk but has yet to inform modelling of HIV transmissions.MethodsData were from 61,198 primary HIV care visits for 14,261 PLWH receiving care through the Centers for AIDS Research (CFAR) Network of Integrated Clinical Systems (CNICS) at seven United States (U.S.) sites from 2007 to 2017. Patient-reported outcomes and measures (PROs) of syndemic conditions - depressive symptoms, anxiety symptoms, drug use (opiates, amphetamines, crack/cocaine) and alcohol use - were collected approximately four to six months apart along with sexual behaviours (mean = 4.3 observations). Counts of syndemic conditions, HIV sexual risk group and time in care were modelled to predict estimated HIV transmissions resulting from sexual behaviour and viral suppression status (HIV RNA < 400/mL) using hierarchical linear modelling.ResultsPatients averaged 0.38 estimated HIV transmissions/100 patients/year for all visits with syndemic conditions measured (down from 0.83, first visit). The final multivariate model showed that per 100 patients, each care visit predicted 0.05 fewer estimated transmissions annually (95% confidence interval (CI): 0.03 to 0.06; p < 0.0005). Cisgender women, cisgender heterosexual men and cisgender men of undisclosed sexual orientation had, respectively, 0.47 (95% CI: 0.35 to 0.59; p < 0.0005), 0.34 (95% CI: 0.20 to 0.49; p < 0.0005) and 0.22 (95% CI: 0.09 to 0.35; p < 0.005) fewer estimated HIV transmissions/100 patients/year than cisgender men who have sex with men (MSM). Psychosocial syndemic conditions have received more attention regarding their deleterious effects on HIV acquisition risk than for their potential impact on HIV treatment and viral suppression. To examine syndemic conditions' impact on the HIV care continuum, we analyzed data collected from people living with HIV (N = 14,261) receiving care through The Centers for AIDS Research Network of Integrated Clinical Systems at seven sites from 2007 to 2017 who provided patient-reported outcomes ∼4-6 months apart. Syndemic condition count (depression, anxiety, substance use, and hazardous drinking), sexual risk group, and time in care were modeled to predict antiretroviral therapy (ART) adherence and viral suppression (HIV RNA <400 copies/mL) using multilevel logistic regression. Comparing patients with each other, odds of ART adherence were 61.6% lower per between-patient syndemic condition [adjusted odds ratio (AOR) = 0.384; 95% confidence interval (CI), 0.362-0.408]; comparing patients with themselves, odds of ART adherence were 36.4% lower per within-patient syndemic condition (AOR = 0.636 95% CI, 0.606-0.667). Odds of viral suppression were 29.3% lower per between-patient syndemic condition (AOR = 0.707; 95% CI, 0.644-0.778) and 27.7% lower per within-patient syndemic condition (AOR = 0.723; 95% CI, 0.671-0.780). Controlling for the effects of adherence (AOR = 5.522; 95% CI, 4.67-6.53), each additional clinic visit was associated with 1.296 times higher odds of viral suppression (AOR = 1.296; 95% CI, 1.22-1.38), but syndemic conditions were not significant. Deploying effective interventions within clinics to identify and treat syndemic conditions and bolster ART adherence and continued engagement in care can help control the HIV epidemic, even within academic medical settings in the era of increasingly potent ART.

The addition of supplementary fat to pig diets above current dietary recommendations has generally been viewed in a negative manner because it is considered that the surplus energy will be deposited into adipose tissue, and may therefore be detrimental to carcass value. The current study sought to investigate the effects of adding supplementary fat, in place of starch, to increase the energy density of male and female pig diets on growth performance and carcass quality. A total of 288 Large White × Landrace male and female pigs (boars and gilts) were randomly allocated to pens of 12 pigs each. The effects of (i) sex (gilts v. boars), (ii) added dietary fat level (as tallow; 0 v. 40 and 80 g/kg) and (iii) within added dietary fat level (40 v. 80 g fat/kg), were tested for a 5-week period (Period 1) (n = 4). These diets were formulated to contain 13·5, 14·3 and 15·2 MJ digestible energy/kg for 0, 40 and 80 g added fat/kg, respectively; the calculated standardized ileal digestible lysine content was kept constant at 8·8 g/kg of diet. Thereafter the experiment was designed to test for the same effects over a second 5-week period (Period 2), during which pigs from the initial three treatments were offered the diets with 0 and 40 g added fat/kg. Treatment effects were also assessed over the whole 10 weeks of the study. There was a positive linear effect of dietary fat supplementation in Period 1 on average daily weight gain, whereas dietary fat level in Period 1 had no effect on performance in Period 2, overall, or on carcass weight and P2 backfat thickness (i.e. thickness of fat at 65 mm down the left side from the midline, at the level of the head of the last rib) at 10 weeks. In contrast, supplementation of the diet with 40 g fat/kg during Period 2, regardless of the diet offered in Period 1, increased ADG in the second period and over the entire experiment. Multipotent mesenchymal stromal cells derived from human placenta (pMSCs), and unrestricted somatic stem cells (USSCs) derived from cord blood share many properties with human bone marrow-derived mesenchymal stromal cells (bmMSCs) and are currently in clinical trials for a wide range of clinical settings. Here we present gene expression profiles of human cord blood-derived unrestricted somatic stem cells (USSCs), human placental-derived mesenchymal stem cells (hpMSCs), and human bone marrow-derived mesenchymal stromal cells (bmMSCs), all derived from four different donors. The microarray data are available on the ArrayExpress database (www.ebi.ac.uk/arrayexpress) under accession number E-TABM-880. Additionally, the data has been integrated into a public portal, www.stemformatics.org. Our data provide a resource for understanding the differences in MSCs derived from different tissues.

Mesenchymal stem cells (MSC) are emerging as a leading cellular therapy for a number of diseases. However, for such treatments to become available as a routine therapeutic option, efficient and cost-effective means for industrial manufacture of MSC are required. At present, clinical grade MSC are manufactured through a process of manual cell culture in specialized cGMP facilities. This process is open, extremely labor intensive, costly, and impractical for anything more than a small number of patients. While it has been shown that MSC can be cultivated in stirred bioreactor systems using microcarriers, providing a route to process scale-up, the degree of numerical expansion achieved has generally been limited. Furthermore, little attention has been given to the issue of primary cell isolation from complex tissues such as placenta. In this article we describe the initial development of a closed process for bulk isolation of MSC from human placenta, and subsequent cultivation on microcarriers in scalable single-use bioreactor systems. Based on our initial data, we estimate that a single placenta may be sufficient to produce over 7,000 doses of therapeutic MSC using a large-scale process.

Supplemental fat is routinely included in young pig diets but often removed with increasing weight to match the perceived declining need for high energy as pigs grow and thus to prevent excessive fat deposition during finishing. However, recent results suggest that early positive effects on growth may be lost if fat is removed from subsequent diets and that, for reasons yet to be established, older pigs may be more responsive to supplemental fat than younger pigs (Campbell, 2005). This project was designed to characterize the effects of added fat on performance, carcass quality and the possible mechanisms of why the pig responds like it does to added fat.

A normal mode model is used to predict signal coherence for a sound source moving horizontally in shallow water. In the model, modal amplitude or modal phase are assumed to fluctuate. The fluctuations are ascribed to the effect of the surface or bottom roughness on sound propagation. For frequencies at which only a few modes are important, namely water depths of the order of 1.5 wavelengths, model coherence was calculated for sensor configurations in the plane containing source and receivers. For source motion towards the receivers, it was found that for phase and amplitude fluctuations the coherence obtained are identical and that the coherence, which can drop to zero over a few wavelengths, is independent of the fluctuation distribution width. For source motion towards the receivers, coherence is controlled by mode strengths and mode propagation constants. In contrast, if the source is in motion in a circle about a vertical array, coherence depends upon the distribution width, and for the same distribution width coherences for phase fluctuations are proportionately lower than those for amplitude fluctuations.
