Injection of antigenic peptides has been widely used as a vaccine strategy to boost T cell immunity. However, the poor immunogenicity of single peptides can potentially be strengthened through modification of the tertiary structure and the selection of the accompanying adjuvant. Here, we generated antigenic peptides into non-linear trimers by solid phase peptide synthesis, thereby enhancing antigen presentation by dendritic cells to CD8+ T cells in vitro and in vivo. CD8+ T cells from mice vaccinated with trimers showed an KLRG1+ effector phenotype and were able to recognize and kill antigen-expressing tumor cells ex vivo. Importantly, trimers outperformed synthetic long peptide in terms of T cell response even when equal number of epitopes were used for immunization. To improve the synthesis of trimers containing difficult peptide sequences, we developed a novel small molecule that functions as conjugation platform for synthetic long peptides. This platform, termed Antigen MAtriX (AMAX) improved yield, purity and solubility of trimers over conventional solid phase synthesis strategies. AMAX outperformed synthetic long peptides in terms of both CD8+ and CD4+ T cell responses and allowed functionalization with DC-SIGN-binding carbohydrates for in vivo dendritic cell targeting strategies, boosting T cell responses even further. Here, we review the important roles of miRNAs in the regulation of CD8+ T cell biology and provide perspectives on the broader emerging principles of miRNA function.

Hepatocellular carcinoma (HCC) is the sixth most common type of cancer globally. Although many different kinds of treatment are performed for HCC according to the practical guidelines, the prognosis of patients is not still satisfactory because the effects of treatments are limited for advanced tumors and the recurrence rate of HCC, even in early stages, is very high. Therefore, immunotherapy is highly anticipated as a new treatment method for HCC. For the development of a new HCC therapy, we attempted to establish immunotherapy using dendritic cells (DCs) and peptide vaccine. In several clinical trials that we performed, we confirmed that the immunotherapy was safe and well-tolerated by HCC patients. We observed that DC therapy prolonged the recurrence-free survival of patients compared with that of patients without DC infusion, as well as observing the radiological anti-tumor effect in HCC patients with peptide vaccine. In this chapter, we summarize the results of previous studies using DC and peptide vaccine, including our own data, and describe the prospects of immunotherapy for HCC. © 2015, Springer Japan. Large granular lymphocyte leukemia (LGLL) has been morphologically characterized as a group of lymphoproliferative diseases that include T-cell large granular lymphocytic leukemia (T-LGLL) and chronic lymphoproliferative disorders of natural killer cells (CLPD-NK). We investigated mutations in the Src homology 2 (SH2) domain of the signal transducer and activator of transcription 3 (STAT3) gene in Asian cohorts of T-LGLL and CLPD-NK (n=42 and 11, respectively). Two mutations, Y640F and D661Y, were identified using direct sequencing or allele-specific (AS) PCR. Y640F and D661Y mutations were found in seven and 18 patients, respectively. Two patients were positive for both mutations. Frequencies of STAT3 mutations in T-LGLL and CLPD-NK were 47.6% and 27.2%, respectively. Pure red cell aplasia (PRCA) was associated with the mutations (P=0.005). The mutations were persistently found at stable levels in some patients after more than 5years using AS-quantitative PCR. The results of the present study indicate that the SH2 domain of the STAT3 gene is frequently mutated in Asian T-LGLL and CLPD-NK, and that PRCA is closely correlated with the mutations. SH2 domain of the STAT3 gene is frequently mutated in Asian T cell large granular lymphocyte leukemia and chronic lymphoproliferative disorders of NK cells. Pure red cell aplasia is closely associated with the mutations.

Background: Major histocompatibility complex (MHC) class I peptide binding and presentation are essential for antigen-specific activation of cytotoxic T lymphocytes (CTLs) and swine MHC class I molecules, also termed swine leukocyte antigens (SLA), thus play a crucial role in the process that leads to elimination of viruses such as swine influenza virus (SwIV). This study describes the identification of SLA-presented peptide epitopes that are targets for a swine CTL response, and further analyses multiple specificities expressed by SwIV activated CTL subsets. Findings: Four SwIV derived peptides were identified as T cell epitopes using fluorescent influenza: SLA tetramers. In addition, multiple CTL specificities were analyzed using peptide sequence substitutions in two of the four epitope candidates analyzed. Interestingly both conserved and substituted peptides were found to stain the CD4-CD8+ T cell subsets indicating multiple specificities. Conclusions: This study describes a timely and cost-effective approach for viral epitope identification in livestock animals. Analysis of T cell subsets showed multiple specificities suggesting SLA-bound epitope recognition of different conformations.

Charakterisierung des Zytokinprofils in Erwachsenen mit einer latenten oder aktiven Tuberkulose in einem hoch endemischen Gebiet Die Tuberkulose (TB) stellt mit rund 2 Milliarden Infizierten weltweit ein globales gesundheitliches Problem dar. Während die große Mehrheit der infizierten Personen in der Lage sind die Krankheit zu kontrollieren, entwickelt sich bei ungefähr 10 % die aktive Form der TB aus. Der zugrunde liegende immunologische Prozess für diese Verteilung ist bis heute nicht bekannt und im Fokus dieser Arbeit. Das adaptive Immunsystem spielt eine entscheidende Rolle in der Immunabwehr gegen Mycobacterium tuberculosis (M. tuberculosis), dem Erreger der TB. Hierbei sind besonders CD4+ T-Zellen für die erfolgreiche Eingrenzung der Erkrankung verantwortlich. Im Vorfeld konnte bereits mehrmals eine Assoziation zwischen polyfunktionalen CD4+ T-Zellen und einem Schutz gegen verschiedenste Krankheitserreger gezeigt werden. Im Rahmen dieser Doktorarbeit wird versucht die Frage zu beantworten, ob eine erhöhte Frequenz von polyfunktionalen CD4+ T-Zellen auch gegen die Ausbildung einer aktiven TB schützen kann. Zur Bearbeitung dieser Fragestellung wurde das TH1 Zytokinprofil von Patienten mit aktiver TB untersucht und mit dem von gesunden latent infizierten Probanden (LTBI) verglichen. Desweiteren wurden die TB Patienten während der antimikrobiellen Therapie begleitet um Änderungen im Zytokinprofil von CD4+ T-Zellen beobachten zu können. Im Rahmen dieser Arbeit wurde zum ersten Mal die simultane Expression der vier TH1 Zytokine IFNg, TNFa, IL-2 und GM-CSF mit Hilfe der multifarben Durchflusszytometrie untersucht. Nach antigenspezifischer Stimulation konnten sowohl in unbehandelten und behandelten Patienten mit aktiver TB ein großer Anteil an multifunktionale Gedächtnis-T-Zellen nachgewiesen werden, die alle vier Zytokine gleichzeitig exprimierten. Bemerkenswerterweise konnte diese Population ebenfalls in LTBI gezeigt werden. Since polyfunctional CD4+ T cells have been associated with protection against various pathogens, the question was raised if higher frequencies of polyfunctional CD4+ T cells can be linked to protection against reactivation of active TB. To address this the TH1 T cell cytokine profile of active TB patients was analyzed and compared with healthy latently infected individuals (LTBI). Furthermore TB patients were followed up under anti-microbial therapy to monitor changes in the cytokine pattern expressed by CD4+ T cells. Hereby, for the first time, the simultaneous expression of four TH1 cytokines, IFNg, TNFa, IL-2 and GM-CSF, was investigated using multi color flow cytometry. After antigen-specific stimulation multifunctional memory T cells (CD45RO+) co-expressing IFNg, TNFa, IL-2 and GM-CSF were strongly represented in both treated and untreated TB patients. Interestingly, this proportion of polyfunctional memory T cells was also found in LTBI. After the first two months of drug treatment the proportion of antigen-specific polyfunctional T cells was significantly increased, indicating a positive impact of these cells during therapy. Background Dendritic cells (DC) are the professional antigen-presenting cells of the immune system, fully equipped to prime naive T cells and thus essential components for cancer immunotherapy. Methods We tested the influence of several elements (cPPT, trip, WPRE, SIN) on the transduction efficiency of human DC. Human and murine DC were transduced with tNGFR-encoding lentiviruses to assess the effect of transduction on phenotype and function. Human DC were transduced with lentiviruses encoding huli80MAGE-A3 and murine DC with huIi80tOVA to test antigen presentation. Results A self-inactivating (SIN) lentiviral vector containing the trip element was most efficient in transducing human DC. The transduction of DC with trip/SIN tNGFR encoding lentiviral vectors at MOI 15 resulted in stable gene expression in up to 94.6% (murine) and 88.2% (human) of the mature DC, without perturbing viability, phenotype and function. Human huli80MAGE-A3-transduced DC were able to stimulate MAGE-A3-specific CD4(+) and CD8(+) T cell clones and could prime both MAGE-A3-specific CD4+ and CD8+ T cells in vitro. Murine huIi80tOVA-transduced DC were able to present OVA peptides in the context of MHC class I and class II in vitro and induced a strong OVA-specific cytotoxic T lymphocyte response in vivo, that was protective against subsequent challenge with OVA-expressing tumor cells. Conclusions We show that, using lentiviral vectors, efficient gene transfer in human and murine DC can be obtained and that these DC can elicit antigen-specific immune responses in vitro and in vivo. In previous studies we could show that a minor subset of monocytes, the Fc gamma receptor I-negative (CD64(-)) monocytes, exhibits significantly higher antigen-presenting capacity than the main population of monocytes (> 90%) which are Fc gamma receptor I-positive (CD64(+)). Therefore, we addressed the question whether they are also differentially susceptible to T cell-mediated killing. In the present study we demonstrate that the CD64(-) monocyte subset is more resistant to killing by antigen-activated T cells than CD64(+) monocytes, as indicated by a higher viability and recovery of CD64(-) monocytes. This mechanism involves CD95 (Fas) antigen, since monocyte death in co-cultures with antigen-activated T cells could be partially reduced by blocking anti-Fas monoclonal antibodies (mAb). In agreement with this finding, although CD95 antigen was expressed on CD64(+) and CD64(-) monocytes at comparable levels, killing of CD64(-) monocytes by activating anti-Fas mAb was lower than of CD64(+) monocytes.

Purpose: Hemophagocytic lymphohistiocytosis (HLH) is a life-threatening syndrome characterized by fever, hepatosplenomegaly, pancytopenia, and infiltration of vital organs by non-Langerhans histiocytes and is rapidly fatal without early diagnosis and institution of therapy. Immune dysregulation is thought to be responsible for the disease. Patients and Methods: Extensive immune evaluation was performed on 13 consecutive patients diagnosed with HLH over a 4-year period to characterize existing immunologic abnormalities in order to improve early diagnosis. Evaluation included quantitative immunoglobulins, immunophenotyping, mitogen-induced lymphoproliferation, natural killer (NK) cell function, and cytotoxic T cell lymphocytolysis (CTL). Results: Immunoglobulin levels showed no consistent abnormality. Immunophenotyping showed an absolute decrease in number of B cells but normal numbers and proportional distribution of T cell subsets and NK cells. Most patients demonstrated decreased proliferative responses to mitogens (10/13) and severely decreased to absent T cell cytotoxicity (11/12) and NK cytotoxic function (13/13). Conclusions: Our results show that while humoral immunity is essentially intact, cellular immune function is significantly impaired in the vast majority of patients with HLH. The coincident finding of profoundly decreased T cell cytotoxicity along with absent NK cytotoxicity suggests that patients with active HLH may have global cytotoxic dysfunction. Since the majority of our patients were studied prior to starting therapy, we feel that these findings reflect the pathophysiologic process and are not therapy related. Unclear from the present work is whether these findings represent primary or secondary dysfunction. We conclude from these studies that profoundly decreased CTL function and absence of NK cell function are characteristic immunologic features of HLH and may serve as additional laboratory data, in conjunction with currently proposed diagnost
