Tissue and blood gas embolism (GE) associated with fisheries bycatch are likely a widespread, yet underestimated, cause of sea turtle mortality. Here, we evaluated risk factors associated with tissue and blood GE in loggerhead turtles caught incidentally by trawl and gillnet fisheries on the Valencian coastline of Spain. Of 413 turtles (303 caught by trawl, 110 by gillnet fisheries), 54% (n = 222) exhibited GE. For sea turtles caught in trawls, the probability and severity of GE increased with trawl depth and turtle body mass. In addition, trawl depth and the GE score together explained the probability of mortality (P[mortality]) following recompression therapy. Specifically, a turtle with a GE score of 3 caught in a trawl deployed at 110 m had a P[mortality] of ~50%. For turtles caught in gillnets, no risk variables were significantly correlated with either the P[GE] or GE score. However, gillnet depth or GE score, separately, explained P[mortality], and a turtle caught at 45 m or with a GE score between 3 and 4 had a P[mortality] of 50%. Differences in the fishery characteristics precluded direct comparison of GE risk and mortality between these gear types. Although P[mortality] is expected to be significantly higher in untreated turtles released at sea, our findings can improve estimates of sea turtle mortality associated with trawls and gillnets, and help guide associate conservation efforts

Management of gases during diving is not well understood across marine mammal species. Prior to diving, phocid (true) seals generally exhale, a behaviour thought to assist with the prevention of decompression sickness. Otariid seals (fur seals and sea lions) have a greater reliance on their lung oxygen stores, and inhale prior to diving. One otariid, the Antarctic fur seal (Arctocephalus gazella), then exhales during the final 50–85% of the return to the surface, which may prevent another gas management issue: shallow-water blackout. Here, we compare data collected from animal-attached tags (video cameras, hydrophones and conductivity sensors) deployed on a suite of otariid seal species to examine the ubiquity of ascent exhalations for this group. We find evidence for ascent exhalations across four fur seal species, but that such exhalations are absent for three sea lion species. Fur seals and sea lions are no longer genetically separated into distinct subfamilies, but are morphologically distinguished by the thick underfur layer of fur seals. Together with their smaller size and energetic dives, we suggest their air-filled fur might underlie the need to perform these exhalations, although whether to reduce buoyancy and ascent speed, for the avoidance of shallow-water blackout or to prevent other cardiovascular management issues in their diving remains unclear. Marine mammals such as northern elephant seals (NES) routinely experience hypoxemia and ischemia-reperfusion events to many tissues during deep dives with no apparent adverse effects. Adaptations to diving include increased antioxidants and elevated oxygen storage capacity associated with high hemoprotein content in blood and muscle. The natural turnover of heme by heme oxygenase enzymes (encoded by HMOX1 and HMOX2) produces endogenous carbon monoxide (CO), which is present at high levels in NES blood and has been shown to have cytoprotective effects in laboratory systems exposed to hypoxia. To understand how pathways associated with endogenous CO production and signaling change across ontogeny in diving mammals, we measured muscle CO and baseline expression of 17 CO-related genes in skeletal muscle and whole blood of three age classes of NES. Muscle CO levels approached those of animals exposed to high exogenous CO, increased with age, and were significantly correlated with gene expression levels. Muscle expression of genes associated with CO production and antioxidant defenses (HMOX1, BVR, GPX3, PRDX1) increased with age and was highest in adult females, while that of genes associated with protection from lipid peroxidation (GPX4, PRDX6, PRDX1, SIRT1) was highest in adult males. In contrast, muscle expression of mitochondrial biogenesis regulators (PGC1A, ESRRA, ESRRG) was highest in pups, while genes associated with inflammation (HMOX2, NRF2, IL1B) did not vary with age or sex. Blood expression of genes involved in regulation of inflammation (IL1B, NRF2, BVR, IL10) was highest in pups, while HMOX1, HMOX2 and pro-inflammatory markers (TLR4, CCL4, PRDX1, TNFA) did not vary with age. We propose that ontogenetic upregulation of baseline HMOX1 expression in skeletal muscle of NES may, in part, underlie increases in CO levels and expression of genes encoding antioxidant enzymes. However, evidence that marine mammals experience gas bubbles during diving is growing, causing concern that these bubbles may cause gas emboli pathology (GEP) under unusual circumstances. Marine mammal management, and usual avoidance, of gas emboli and GEP, or the bends, became a topic of intense scientific interest after sonar-exposed, mass-stranded deep-diving whales were observed with gas bubbles. Theoretical models, based on our current understanding of diving physiology in cetaceans, predict that the tissue and blood N2 levels in the bottlenose dolphin (Tursiops truncatus) are at levels that would result in severe DCS symptoms in similar sized terrestrial mammals. However, the dolphins appear to have physiological or behavioral mechanisms to avoid excessive blood N2 levels, or may be more resistant to circulating bubbles through immunological/biochemical adaptations. Studies on behavior, anatomy and physiology of marine mammals have enhanced our understanding of the mechanisms that are thought to prevent excessive uptake of N2. This has led to the selective gas exchange hypothesis, which provides a mechanism how stress-induced behavioral change may cause failure of the normal physiology, which results in excessive uptake of N2, and in extreme cases may cause formation of symptomatic gas emboli. Studies on cardiorespiratory function have been integral to the development of this hypothesis, with work initially being conducted on excised tissues and cadavers, followed by studies on anesthetized animals or trained animals under human care. These studies enabled research on free-ranging common bottlenose dolphins in Sarasota Bay, FL, and off Bermuda, and have included work on the metabolic and cardiorespiratory physiology of both shallow- and deep-diving dolphins and have been integral to better understand how cetaceans can dive to extreme dept, Many of the studies that have resulted in the data in this review, and that have been integral to develop the selective gas exchange hypothesis have been funded by the Office of Naval Research (ONR Awards # N000141010159, N000141613088, N000141410563, N000140811220, and ONR YIP Award # N000141410563), and Dolphin Quest. Continuous measurements of haemodynamic and oxygenation changes in free living animals remain elusive. However, developments in biomedical technologies may help to fill this knowledge gap. One such technology is continuous-wave near-infrared spectroscopy (CW-NIRS)-a wearable and non-invasive optical technology. Here, we develop a marinized CW-NIRS system and deploy it on elite competition freedivers to test its capacity to function during deep freediving to 107 m depth. We use the oxyhaemoglobin and deoxyhaemoglobin concentration changes measured with CW-NIRS to monitor cerebral haemodynamic changes and oxygenation, arterial saturation and heart rate. Furthermore, using concentration changes in oxyhaemoglobin engendered by cardiac pulsation, we demonstrate the ability to conduct additional feature exploration of cardiac-dependent haemodynamic changes. Freedivers showed cerebral haemodynamic changes characteristic of apnoeic diving, while some divers also showed considerable elevations in venous blood volumes close to the end of diving. Some freedivers also showed pronounced arterial deoxygenation, the most extreme of which resulted in an arterial saturation of 25%. Freedivers also displayed heart rate changes that were comparable to diving mammals both in magnitude and patterns of change. Finally, changes in cardiac waveform associated with heart rates less than 40 bpm were associated with changes indicative of a reduction in vascular compliance. The success here of CW-NIRS to non-invasively measure a suite of physiological phenomenon in a deep-diving mammal highlights its efficacy as a future physiological monitoring tool for human freedivers as well as free living animals. Each year, marine mammals are exposed to increasing levels of anthropogenic disturbance. Some disturbances, such as boat strikes and entanglement, directly impact animals through injuries and mortality events. However, indirect effects from disturbances including over-fishing, noise and environmental pollution, declining sea ice cover, and changes in coastal habitats can have significant, though less apparent, impacts as well. These can affect both individuals and populations through declining prey availability, decreased reproductive and juvenile success, declines in body-condition, and increased mortality. Unfortunately, as a result of their cryptic lifestyle, it is difficult to measure the impact of these disturbances on many marine mammal species or predict how they will affect individuals or populations in the future. A better understanding of both maintenance and locomotor energetic demands for these species is needed to quantify the impacts of these disturbances, model the future effects, and predict the capacity of these species to adapt or respond. Using open-flow respirometry and submersible accelerometers, I undertook a comparative physiological study examining four marine mammal species from three different groups. In Chapter 1, I studied the interaction between maintenance and locomotor costs in two coastal marine mammal species living in tropical and subtropical environments, West Indian manatees and Hawaiian monk seals. Although these warm water species exhibited a lower resting metabolic rate (RMR) than their cold-water relatives, I found that this does not confer an energetic advantage during locomotion for these species due to decreased metabolic variability. In Chapter 2, I measured RMR and locomotor costs in beluga whales as the first step towards creating a population consequences of disturbance model to aid conservation of the Cook Inlet beluga whale population. To test the hypothesis that offshore bottlenose dolphins have evolved physiological adaptations to improve their ability for extended deep dives and as protection for lung barotrauma, we investigated the lung function and respiratory physiology of four wild common bottlenose dolphins (Tursiops truncatus) near the island of Bermuda. We measured blood hematocrit (Hct, %), resting metabolic rate (RMR, l O2 ⋅ min-1), tidal volume (VT, l), respiratory frequency (fR, breaths ⋅ min-1), respiratory flow (l ⋅ min-1), and dynamic lung compliance (CL, l ⋅ cmH2O-1) in air and in water, and compared measurements with published results from coastal, shallow-diving dolphins. We found that offshore dolphins had greater Hct (56 ± 2%) compared to shallow-diving bottlenose dolphins (range: 30–49%), thus resulting in a greater O2 storage capacity and longer aerobic diving duration. Contrary to our hypothesis, the specific CL (sCL, 0.30 ± 0.12 cmH2O-1) was not different between populations. Neither the mass-specific RMR (3.0 ± 1.7 ml O2 ⋅ min-1 ⋅ kg-1) nor VT (23.0 ± 3.7 ml ⋅ kg-1) were different from coastal ecotype bottlenose dolphins, both in the wild and under managed care, suggesting that deep-diving dolphins do not have metabolic or respiratory adaptations that differ from the shallow-diving ecotypes. Considerable research has been done to understand how bottlenose dolphins manage O2 during diving, but little information exists on other gases or how pressure affects gas exchange. Here we used a dynamic multi-compartment gas exchange model to estimate blood and tissue O2, CO2, and N2 from high-resolution dive records of two different common bottlenose dolphin ecotypes inhabiting shallow (Sarasota Bay) and deep (Bermuda) habitats. The objective was to compare potential physiological strategies used by the two populations to manage shallow and deep diving life styles. We informed the model using species-specific parameters for blood hematocrit, resting metabolic rate, and lung compliance. The model suggested that the known O2 stores were sufficient for Sarasota Bay dolphins to remain within the calculated aerobic dive limit (cADL), but insufficient for Bermuda dolphins that regularly exceeded their cADL. By adjusting the model to reflect the body composition of deep diving Bermuda dolphins, with elevated muscle mass, muscle myoglobin concentration and blood volume, the cADL increased beyond the longest dive duration, thus reflecting the necessary physiological and morphological changes to maintain their deep-diving life-style. The results indicate that cardiac output had to remain elevated during surface intervals for both ecotypes, and suggests that cardiac output has to remain elevated during shallow dives in-between deep dives to allow sufficient restoration of O2 stores for Bermuda dolphins. However, studies of beached and bycaught cetaceans and sea turtles imply that air-breathing marine vertebrates may, under unusual circumstances, develop GE that result in decompression sickness (DCS) symptoms. Theoretical modelling of tissue and blood gas dynamics of breath-hold divers suggests that changes in perfusion and blood flow distribution may also play a significant role. The results from the modelling work suggest that our current understanding of diving physiology in many species is poor, as the models predict blood and tissue N2 levels that would result in severe DCS symptoms (chokes, paralysis and death) in a large fraction of natural dive profiles. In this review, we combine published results from marine mammals and turtles to propose alternative mechanisms for how marine vertebrates control gas exchange in the lung, through management of the pulmonary distribution of alveolar ventilation (Embedded Image) and cardiac output/lung perfusion (Embedded Image), varying the level of Embedded Image in different regions of the lung. Man-made disturbances, causing stress, could alter the Embedded Image mismatch level in the lung, resulting in an abnormally elevated uptake of N2, increasing the risk for GE. The definitive version was published in Royal Society Open Science 5 (2018): 171280, doi:10.1098/rsos.171280., We measured respiratory flow rates, and expired O2 in 32 (2–34 years, body mass [Mb] range: 73–291 kg) common bottlenose dolphins (Tursiops truncatus) during voluntary breaths on land or in water (between 2014 and 2017). The data were used to measure the resting O2 consumption rate (V˙O2, range: 0.76–9.45ml O2min−1 kg−1) and tidal volume (VT, range: 2.2–10.4 l) during rest. For adult dolphins, the resting VT, but not V˙O2, correlated with body mass (Mb, range: 141–291 kg) with an allometric mass-exponent of 0.41. These data suggest that the mass-specific VT of larger dolphins decreases considerably more than that of terrestrial mammals (mass-exponent: 1.03). The average resting sV˙O2 was similar to previously published metabolic measurements from the same species. Air-breathing divers are assumed to have evolved to apportion their time between surface and underwater periods to maximize the benefit gained from diving activities. However, whether they change their time allocation depending on the aim of the dive is still unknown. This may be particularly crucial for 'surfacers' because they dive for various purposes in addition to foraging. In this study, we counted breath events at the surface and estimated oxygen consumption during resting, foraging, and other dives in 11 green turtles (Chelonia mydas) in the wild. Breath events were counted by a head-mounted acceleration logger or direct observation based on an animal-borne video logger, and oxygen consumption was estimated by measuring overall dynamic body acceleration. Our results indicate that green turtles maximized their submerged time, following this with 5-7 breaths to replenish oxygen for resting dives. However, they changed their dive tactic during foraging and other dives; they surfaced without depleting their oxygen content, followed by only a few breaths for effective foraging and locomotion. These dichotomous surfacing tactics would be the result of behavioral modifications by turtles depending on the aim of each dive.

There are little data available on the structural and functional adaptations of the marine mammal respiratory system. The aim of this research was to investigate the anatomical (gross) and structural (compliance) characteristics of excised marine mammal tracheas. Here we defined different types of tracheal structures, categorizing pinniped tracheas by varying degrees of continuity of cartilage (categories 1-4) and cetacean tracheas by varying compliance values (categories 5A and 5B). Some tracheas fell into more than one category, along their length, for example, the harbor seal (Phoca vitulina) demonstrated complete rings cranially, and as the trachea progressed caudally tracheal rings changed morphology. Dolphins and porpoises had less stiff, more compliant spiraling rings while beaked whales had very stiff, less compliant spiraling rings. The pressure-volume (P-V) relationships of isolated tracheas from different species were measured to assess structural differences between species. Dive duration generally increases with body size in animals including wildfowl. Therefore, diving behaviour may vary between the sexes in sexually size dimorphic species, such as the extremely sexually size dimorphic Musk Duck Biziura lobata. However, a previous study reports longer dives in the smaller sex (females) when breeding. In this study, non-breeding male Musk Ducks dived for significantly longer periods than females and tended to have longer inter-dive intervals, conforming to the general patterns described for other species. The differences in dive behaviour we describe may be explained by niche partitioning or differential oxygen requirements or uptake rates by the sexes.

In this study, the relationship between dive duration, activity level during the dive and the number of breaths during post-dive surface duration of one free-ranging nesting green turtle, Chelonia mydas, was examined using two acceleration data loggers. Breathing was extracted from the static acceleration and depth data of the head-mounted data logger. The overall dynamic body acceleration (ODBA), which has been used as an index of activity levels per unit time of a dive, was calculated from tri-axis dynamic acceleration data of the carapace-attached data logger. The number of breaths during post-dive surface duration divided by ODBA significantly increased with longer dive duration. The number of breaths during post-dive surface duration divided by dive duration significantly increased with larger ODBA during a dive. The results suggest that the number of breaths was regulated by dive duration and activity level during a dive., March 18-19, 2013

There is a dire need to determine the normal cardiovascular dive response of deep-diving odontocetes like beaked whales, and to examine how that response might be altered during exposure to anthropogenic sound. However, no one has ever recorded the electrocardiogram of a wild, freeranging odontocete. Recent advances in the miniaturization of multi-channel datalogging devices now make it possible to conceive of remotely attaching a device to a deep-diving odontocete to record physiological variables such as heart rate and body temperature, which would then permit studies of the cardiovascular response to diving and advance many of the discussions about the susceptibility of beaked whales to gas bubble disease beyond theory and speculation. We will modify our existing technology for making physiological recordings and demonstrate its utility on beaked and pilot whales so that diving physiology studies could be conducted to further our understanding of the susceptibility of these whales to adverse physiological effects of exposure to anthropogenic sounds., The original document contains color images.

The basic pathology and cause are relatively well known to human divers. Breath-hold diving marine mammals were thought to be relatively immune to DCS owing to multiple anatomical, physiological and behavioural adaptations that reduce nitrogen gas (N2) loading during dives. However, recent observations have shown that gas bubbles may form and tissue injury may occur in marine mammals under certain circumstances. Gas kinetic models based on measured time-depth profiles further suggest the potential occurrence of high blood and tissue N2 tensions. We review evidence for gas-bubble incidence in marine mammal tissues and discuss the theory behind gas loading and bubble formation. We suggest that diving mammals vary their physiological responses according to multiple stressors, and that the perspective on marine mammal diving physiology should change from simply minimizing N2 loading to management of the N2 load. This suggests several avenues for further study, ranging from the effects of gas bubbles at molecular, cellular and organ function levels, to comparative studies relating the presence/absence of gas bubbles to diving behaviour. To evaluate live dolphins for bubbles, liver, kidneys, eyes and blubber–muscle interface of live-stranded and capture-release dolphins were scanned with B-mode ultrasound. Gas was identified in kidneys of 21 of 22 live-stranded dolphins and in the hepatic portal vasculature of 2 of 22. Nine then died or were euthanized and bubble presence corroborated by computer tomography and necropsy, 13 were released of which all but two did not re-strand. Bubbles were not detected in 20 live wild dolphins examined during health assessments in shallow water. Off-gassing of supersaturated blood and tissues was the most probable origin for the gas bubbles. In contrast to marine mammals repeatedly diving in the wild, stranded animals are unable to recompress by diving, and thus may retain bubbles. Since the majority of beached dolphins released did not re-strand it also suggests that minor bubble formation is tolerated and will not lead to clinically significant decompression sickness., Funding for this work was provided by the US Office of Naval Research Award no. Our data indicate that mass-specific TLC (sTLC, l • kg-1) does not differ between species or groups (odontocete vs. phocid) and agree with that estimated (TLCest) from body mass (Mb) by: TLCest = 0.135 • Mb 0.92. Measured MAV was on average 7% of TLC, with a range from 0% to 16%. The pressure-volume curves were similar among species on inflation but diverged during deflation in phocids as compared with odontocetes. Changes in metabolic rates were measured in 3 captive female Steller sea lions Eumetopias jubatus that experienced fasts during summer and winter. We measured metabolic rates (via O2 consumption) before (MRs, surface) and after (DMR, dive + surface interval) the sea lions dove to 10–50 m depths. Measurements were obtained prior to and immediately after 9 to 10 d fasts, and during a 14 d recovery period. The sea lions lost significantly more body mass (Mb) during the winter fast (10.6%), compared with the summer (9.5%). Mass-corrected dive metabolic rate (cDMR = DMR × Mb–0.714) was not affected by dive depth or duration, but increased significantly following the winter fasts (13.5 ± 8.1%), but did not change during summer (–1.1 ± 3.2%). However, mass-corrected surface metabolic rate (cMRs) decreased significantly after both the summer (–16.4 ± 4.7%) and winter (–8.0 ± 9.0%) fasts. Consequently, the ratio between cDMR and cMRs was significantly higher in winter, suggestive of an increased thermal challenge and convective heat loss while diving. Increased cMRs following the fast indicated that digestion began during foraging and was not deferred, implying that access to ingested energy was of higher priority than optimizing diving ability. cDMR was elevated throughout the recovery period, independent of season, resulting in a 12% increase in foraging cost in winter and a 3% increase in summer. Our data suggest that Steller sea lions are more sensitive to changes in body condition due to food shortages in the winter compared with the summer.

The metabolic costs of foraging and the management of O2 and CO2 stores during breath-hold diving was investigated in three female Steller sea lions (Eumetopias jubatus) trained to dive between 10 and 50 m (N=1142 dives). Each trial consisted of two to eight dives separated by surface intervals that were determined by the sea lion (spontaneous trials) or by the researcher (conditioned trials). During conditioned trials, surface intervals were long enough for O2 to return to pre-dive levels between each dive. The metabolic cost of each dive event (dive+surface interval; DMR) was measured using flow-through respirometry. The respiratory exchange ratio (O2/CO2) was significantly lower during spontaneous trials compared with conditioned trials. DMR was significantly higher during spontaneous trials and decreased exponentially with dive duration. A similar decrease in DMR was not as evident during conditioned trials. DMR could not be accurately estimated from the surface interval (SI) following individual dives that had short SIs (<50 s), but could be estimated on a dive by dive basis for longer SIs (>50 s). DMR decreased by 15%, but did not differ significantly from surface metabolic rates (MRS) when dive duration increased from 1 to 7 min. Overall, these data suggest that DMR is almost the same as MRS, and that Steller sea lions incur an O2 debt during spontaneous diving that is not repaid until the end of the dive bout. This has important consequences in differentiating between the actual and `apparent' metabolic rate during diving, and may explain some of the differences in metabolic rates reported in pinniped species.

Gas mixture and pressure modify the spectral characteristics of diver's speech. Additionally, constraints imposed on jaw movements by wearing a facial mask affect the speech production process. The auditory feedback loop is equally concerned. Furthermore, underwater adverse working conditions are characterised by noise from different sources. As a result, divers' speech is poorly intelligible and communications between divers and surface control need to be enhanced. To this end, "voice unscramblers" are being used. However, the technological state of commercially available equipment is dated and the quality of speech remains insufficient. To help with the design, testing and qualification (NORM) of new communication devices, a bilingual (French-English) Data-base is currently being set up. It consists of phonetically balanced lists of 200 words read by 17 divers under sea and in chambers at operational levels from the surface to -300 m. These recordings will be edited, labelled and stored for further distribution on a CD-ROM.
