2. Symbiosis in Convoluta roscoffensis

Abstract

1. Culture methods for the maintenance of symbiotic adult and juvenile Convoluta roscoffensis have been developed. The growth of adults but not juveniles is dependent on nitrogen enrichment of the medium. 2. Electron microscopic studies suggest that the algal symbionts in adult Convoluta are intracellular. 3. Structural studies of the gametes and early development of Convoluta are described. 4. Relevant aspects of the biology of the natural Convoluta symbiont, Platymonas convolutae, and a range of related algae have been studied. The growth of all Platymonas and Tetraselmis species tested except P.convolutae is stimulated by 0.1 M glucose. Glucose supports the growth of T.tetrathele, T.suecica and P.subcordiformis in darkness. P.convolutae and T.tetrathele have two uptake systems for uric acid, which conform to Michaelis-Menten kinetics; a high affinity system, operating in the concentration range 0.2-4.5 pM, and a low affinity system, operating at higher concentrations of uric acid. Uric acid uptake by P.convolutae is abolished by uncouplers of phosphorylation. Exogenous uric acid is accumulated in cells of Platymonas and Tetraselmis species up to internal concentrations of 0.3 M. P. convolutae metabolises (2-14C) uric acid to (14 C) carbon dioxide in the dark. 5. Aposymbiotic juvenile and symbiotic adult Convoluta, under standard culture conditions, contain solid uric acid. The uric acid content of juveniles declines on infection with P.convolutae and related species. Uric acid is utilised in the adult symbiosis, particularly under conditions of nitrogen demand. 6. Motile cells of P.convolutae and related species and Chlamydomonas coccoides aggregate around Convoluta eggs. These algae 'settle' on the eggs, by an interaction initiated by contact between the algal flagella and egg capsule surface. Living and killed algae are trapped by the capsules. No evidence that P.convolutae cells exhibit chemotaxis towards Convoluta eggs has been obtained. 7. Juvenile Convoluta ingest species of Platymonas and Tetraselmis, Prasinocladus marinus and Chlamydomonas coccoides but no other algae tested. Ingestion of P.convolutae is not affected by pretreatment of the cells with lectins or proteolytic enzymes, but is reduced if the algae are killed. 8. Cells of C.coccoidea, but not P.convolutae and related species, are disrupted in juvenile Convoluta. 9. Juvenile Convoluta form a viable symbiosis with P.convolutae, Pr.marinus, T.tetrathele and T.verrucosa under laboratory conditions. The structural changes of P.convolutae and T.verrucosa during the development of the symbiosis are described. 10. It is proposed that Convoluta discriminate against species unrelated to P.convolutae on initial contact and in the digestive region of the host. Evidence for a recognition mechanism discriminating between P.convolutae and related species is discussed. The nature of the recognition mechanism(s) has not been established.

Abstract

BackgroundAlthough native to North America, the invasion of the aphid-like grape phylloxera Daktulosphaira vitifoliae across the globe altered the course of grape cultivation. For the past 150 years, viticulture relied on grafting-resistant North American Vitis species as rootstocks, thereby limiting genetic stocks tolerant to other stressors such as pathogens and climate change. Limited understanding of the insect genetics resulted in successive outbreaks across the globe when rootstocks failed. Here we report the 294-Mb genome of D. vitifoliae as a basic tool to understand host plant manipulation, nutritional endosymbiosis, and enhance global viticulture.ResultsUsing a combination of genome, RNA, and population resequencing, we found grape phylloxera showed high duplication rates since its common ancestor with aphids, but similarity in most metabolic genes, despite lacking obligate nutritional symbioses and feeding from parenchyma. Similarly, no enrichment occurred in development genes in relation to viviparity. However, phylloxera evolved > 2700 unique genes that resemble putative effectors and are active during feeding. Population sequencing revealed the global invasion began from the upper Mississippi River in North America, spread to Europe and from there to the rest of the world.ConclusionsThe grape phylloxera genome reveals genetic architecture relative to the evolution of nutritional endosymbiosis, viviparity, and herbivory. The extraordinary expansion in effector genes also suggests novel adaptations to plant feeding and how insects induce complex plant phenotypes, for instance galls. Finally, our understanding of the origin of this invasive species and its genome provide genetics resources to alleviate rootstock bottlenecks restricting the advancement of viticulture.

Background: Although native to North America, the invasion of the aphid-like grape phylloxera Daktulosphaira vitifoliae across the globe altered the course of grape cultivation. For the past 150 years, viticulture relied on grafting-resistant North American Vitis species as rootstocks, thereby limiting genetic stocks tolerant to other stressors such as pathogens and climate change. Limited understanding of the insect genetics resulted in successive outbreaks across the globe when rootstocks failed. Here we report the 294-Mb genome of D. vitifoliae as a basic tool to understand host plant manipulation, nutritional endosymbiosis, and enhance global viticulture. Results: Using a combination of genome, RNA, and population resequencing, we found grape phylloxera showed high duplication rates since its common ancestor with aphids, but similarity in most metabolic genes, despite lacking obligate nutritional symbioses and feeding from parenchyma. Abstract

Aphids are important agricultural pests and also biological models for studies of insect-plant interactions, symbiosis, virus vectoring, and the developmental causes of extreme phenotypic plasticity. Here we present the 464 Mb draft genome assembly of the pea aphid Acyrthosiphon pisum. This first published whole genome sequence of a basal hemimetabolous insect provides an outgroup to the multiple published genomes of holometabolous insects. Pea aphids are host-plant specialists, they can reproduce both sexually and asexually, and they have coevolved with an obligate bacterial symbiont. Here we highlight findings from whole genome analysis that may be related to these unusual biological features. These findings include discovery of extensive gene duplication in more than 2000 gene families as well as loss of evolutionarily conserved genes. Gene family expansions relative to other published genomes include genes involved in chromatin modification, miRNA synthesis, and sugar transport. Gene losses include genes central to the IMD immune pathway, selenoprotein utilization, purine salvage, and the entire urea cycle. The pea aphid genome reveals that only a limited number of genes have been acquired from bacteria; thus the reduced gene count of Buchnera does not reflect gene transfer to the host genome. The inventory of metabolic genes in the pea aphid genome suggests that there is extensive metabolite exchange between the aphid and Buchnera, including sharing of amino acid biosynthesis between the aphid and Buchnera. The pea aphid genome provides a foundation for post-genomic studies of fundamental biological questions and applied agricultural problems.

7. A single bout of discontinuous exercise as a pragmatic approach to augment glycaemic control in pregnancy: a pilot study

Abstract

Introduction: Pregnancy is a state of naturally occurring insulin resistance; with hyperglycaemia being associated with maternal and foetal risk for adverse outcomes. For women with a healthy pregnancy, Australian guidelines recommend regular moderate intensity exercise. However, many pregnant women report significant barriers for regular exercise participation, including misconceptions with respect to the safety of engaging in moderate and higher intensities of exercise. Interval exercise at high intensity is efficacious for glycaemic control in people with metabolic conditions such as type 2 diabetes. Yet in pregnancy, the effectiveness of a bout of interval (discontinuous) exercise on enhancing daily glycaemic control has not yet been investigated.

Abstract

BackgroundThe whitefly Bemisia tabaci (Hemiptera: Aleyrodidae) is among the 100 worst invasive species in the world. As one of the most important crop pests and virus vectors, B. tabaci causes substantial crop losses and poses a serious threat to global food security.ResultsWe report the 615-Mb high-quality genome sequence of B. tabaci Middle East-Asia Minor 1 (MEAM1), the first genome sequence in the Aleyrodidae family, which contains 15,664 protein-coding genes. The B. tabaci genome is highly divergent from other sequenced hemipteran genomes, sharing no detectable synteny. A number of known detoxification gene families, including cytochrome P450s and UDP-glucuronosyltransferases, are significantly expanded in B. tabaci. Other expanded gene families, including cathepsins, large clusters of tandemly duplicated B. tabaci-specific genes, and phosphatidylethanolamine-binding proteins (PEBPs), were found to be associated with virus acquisition and transmission and/or insecticide resistance, likely contributing to the global invasiveness and efficient virus transmission capacity of B. tabaci. The presence of 142 horizontally transferred genes from bacteria or fungi in the B. tabaci genome, including genes encoding hopanoid/sterol synthesis and xenobiotic detoxification enzymes that are not present in other insects, offers novel insights into the unique biological adaptations of this insect such as polyphagy and insecticide resistance. Interestingly, two adjacent bacterial pantothenate biosynthesis genes, panB and panC, have been co-transferred into B. tabaci and fused into a single gene that has acquired introns during its evolution.ConclusionsThe B. tabaci genome contains numerous genetic novelties, including expansions in gene families associated with insecticide resistance, detoxification and virus transmission, as well as numerous horizontally transferred genes from bacteria and fungi. Abstract

Objective - The aim is to identify the role and scope of Accredited Exercise Physiologist (AEP) services in the mental health sector and to provide insight as to how AEPs can contribute to the multidisciplinary mental health team. Methods - A modified Delphi approach was utilised. Thirteen AEPs with experience in mental health contributed to the iterative development of a national consensus statement. Six mental health professionals with expertise in psychiatry, mental health nursing, general practice and mental health research participated in the review process. Reviewers were provided with a template to systematically provide feedback on the language, content, structure and relevance to their professional group. Results - This consensus statement outlines how AEPs can contribute to the multidisciplinary mental health team, the aims and scope of AEP-led interventions in mental health services and examples of such interventions, the range of physical and mental health outcomes possible through AEP-led interventions and common referral pathways to community AEP services. Outcome - AEPs can play a key role in the treatment of individuals experiencing mental illness. The diversity of AEP interventions allows for a holistic approach to care, enhancing both physical and mental health outcomes.

10. Evolutionary conservation of candidate osmoregulation genes in plant phloem sap-feeding insects

Abstract

The high osmotic pressure generated by sugars in plant phloem sap is reduced in phloem-feeding aphids by sugar transformations and facilitated water flux in the gut. The genes mediating these osmoregulatory functions have been identified and validated empirically in the pea aphid Acyrthosiphon pisum: sucrase 1 (SUC1), a sucrase in glycoside hydrolase family 13 (GH13), and aquaporin 1 (AQP1), a member of the Drosophila integral protein (DRIP) family of aquaporins. Here, we describe molecular analysis of GH13 and AQP genes in phloem-feeding representatives of the four phloem-feeding groups: aphids (Myzus persicae), coccids (Planococcus citri), psyllids (Diaphorina citri, Bactericera cockerelli) and whiteflies (Bemisia tabaci MEAM1 and MED). A single candidate GH13-SUC gene and DRIP-AQP gene were identified in the genome/transcriptome of most insects tested by the criteria of sequence motif and gene expression in the gut. Exceptionally, the psyllid Ba. cockerelli transcriptome included a gut-expressed Pyrocoelia rufa integral protein (PRIP)-AQP, but has no DRIP-AQP transcripts, suggesting that PRIP-AQP is recruited for osmoregulatory function in this insect. This study indicates that phylogenetically related SUC and AQP genes may generally mediate osmoregulatory functions in these diverse phloem-feeding insects, and provides candidate genes for empirical validation and development as targets for osmotic disruption of pest species.

Abstract

In the last two decades, the widespread application of genetic and genomic approaches has revealed a bacterial world astonishing in its ubiquity and diversity. This review examines how a growing knowledge of the vast range of animal-bacterial interactions, whether in shared ecosystems or intimate symbioses, is fundamentally altering our understanding of animal biology. Specifically, we highlight recent technological and intellectual advances that have changed our thinking about five questions: how have bacteria facilitated the origin and evolution of animals; how do animals and bacteria affect each other's genomes; how does normal animal development depend on bacterial partners; how is homeostasis maintained between animals and their symbionts; and how can ecological approaches deepen our understanding of the multiple levels of animal-bacterial interaction. As answers to these fundamental questions emerge, all biologists will be challenged to broaden their appreciation of these interactions and to include investigations of the relationships between and among bacteria and their animal partners as we seek a better understanding of the natural world.

The health of animals, including humans, is dependent on their resident microbiota, but the complexity of the microbial communities makes these associations difficult to study in most animals. Exceptionally, the microbiology of the pea aphid Acyrthosiphon pisum is dominated by a single bacterium Buchnera aphidicola (B. aphidicola). A 1H NMR-based metabonomic strategy was applied to investigate metabolic profiles of aphids fed on a low essential amino acid diet and treated by antibiotic to eliminate B. aphidicola. In addition, differential gel electrophoresis (DIGE) with mass spectrometry was utilized to determine the alterations of proteins induced by these treatments. We found that these perturbations resulted in significant changes to the abundance of 15 metabolites and 238 proteins. Ten (67%) of the metabolites with altered abundance were amino acids, with nonessential amino acids increased and essential amino acids decreased by both perturbations. Over-represented proteins in the perturbed treatments included catabolic enzymes with roles in amino acid degradation and glycolysis, various cuticular proteins, and a C-type lectin and regucalcin with candidate defensive roles. This analysis demonstrates the central role of essential amino acid production in the relationship and identifies candidate proteins and processes underpinning the function and persistence of the association.

27. Leading indigenous education in a remote location : reflections on teaching to be 'proud and deadly'

Abstract

This thesis is a critical reflection of the author’s time as a Principal of an Indigenous state school from 2003-2004. The purpose is to reassess the impact of her principalship in terms of the staff, students and Community change that affected learning outcomes at the school and to reanalyse to what actions and to whom positive changes could be attributed. This thesis reflects and identifies, in light of the literature, strategies which were effective in enhancing student learning outcomes. The focus of this thesis was the Doongal State School*, its students, staff and facilities. The author will attempt to draw out theoretical frameworks in terms of: (1) what changed educationally in Doongal State School, (2) what seemed to be important in the Principal’s role, (3) the processes that took place, and (4) the effect of being non- Indigenous and a female. Overall, the author undertook this critical reflection in order to understand and embrace educational practices that will (a) lessen the gap between the academic outcomes achieved by Indigenous and non-Indigenous students, and (b) enhance life choices for Indigenous children. The findings indicate that principal leadership is critical for success in Indigenous schools and is the centrepiece of the models developed to explain improvement at Doongal State School. School factors, Principal Leadership factors, Change factors and factors relating to being a non-Indigenous female principal, which, when implemented, will lead to improved educational outcomes for Indigenous students, have evolved as a result of this thesis. Principal Leadership factors were found to be the enablers for the effective implementation of the key components for success.

28. The secreted salivary proteome of the pea aphid Acyrthosiphon pisum characterised by mass spectrometry

Abstract

Nine proteins secreted in the saliva of the pea aphid Acyrthosiphon pisum were identified by a proteomics approach using GE-LC-MS/MS and LC-MS/MS, with reference to EST and genomic sequence data for A. pisum. Four proteins were identified by their sequences: a homolog of angiotensin-converting enzyme (an M2 metalloprotease), an M1 zinc-dependant metalloprotease, a glucose-methanol-choline (GMC)-oxidoreductase and a homolog to regucalcin (also known as senescence marker protein 30). The other five proteins are not homologous to any previously described sequence and included an abundant salivary protein (represented by ACYPI009881), with a predicted length of 1161 amino acids and high serine, tyrosine and cysteine content. A. pisum feeds on plant phloem sap and the metalloproteases and regucalcin (a putative calcium-binding protein) are predicted determinants of sustained feeding, by inactivation of plant protein defences and inhibition of calcium-mediated occlusion of phloem sieve elements, respectively. The amino acid composition of ACYPI009881 suggests a role in the aphid salivary sheath that protects the aphid mouthparts from plant defences, and the oxidoreductase may promote gelling of the sheath protein or mediate oxidative detoxification of plant allelochemicals. Further salivary proteins are expected to be identified as more sensitive MS technologies are developed.

29. The impact of chlortetracycline on Drosophila melanogaster and Aedes aegypti

Abstract

Microbial symbionts of insects have been demonstrated to play an important role in the nutrition and protection of the host; these include aphids and tsetse. Studies often use antibiotics to eliminate the symbionts but the deleterious impact of using these antibiotics is not commonly addressed. The impact of chlortetracycline treatment on Aedes aegypti and Drosophila melanogaster was investigated by assessing life-span, fecundity, development time, survival, nutrition and metabolism. The impact on microbial numbers and diversity was also determined. With Drosophila, treatment with 50 µg ml-1 and above showed a significant extension in development time and life-span, reduction in fecundity and change in nutritional content. Microbial numbers were significantly reduced at 50 µg ml-1 and above. Culturable techniques and 454 pyrosequencing, demonstrated that the microbial diversity of Drosophila was predominantly Acetobacter. Bacterial elimination through egg dechorionation yielded some similar results to chlortetracycline treatment. However, fecundity and life-span was not significantly affected. Microarray analysis established a significant reduction in the abundance of transcripts associated with immunity, particularly antimicrobial peptides. With Aedes aegypti, treatment significantly reduced the survival and also affected the life-span and nutrition of the insect. Microbial numbers of mosquito larvae were reduced at 30 and 100 µg ml-1. Colonies grew on plates supplemented with 50 µg ml-1 of chlortetracycline, indicating that the larvae bore chlortetracycline-resistant bacteria. 454 pyrosequencing demonstrated a change in diversity of bacteria found in mosquitoes +/- chlortetracycline, switching from Elizabethkingia meningoseptica to Raoultella sp with chlortetracycline. It is concluded that chlortetracycline significantly impacts the performance of the 2 insects through bacterial depletion, changes to bacterial diversity and toxicity. Nevertheless, different responses were observed with Aedes aegypti and Drosophila melanogaster. Moreover, experiments with Drosophila using egg dechorionation, emphasised the toxic impact of using antibiotics to eliminate microbes in the insect host.
