The tumor microenvironment modulates cancer growth. Extracellular vesicles (EVs) have been identified as key mediators of intercellular communication, but their role in tumor growth is largely unexplored. Here, we demonstrate that EVs from sarcoma patients promote neoangiogenesis via a purinergic X receptor 4 (P2XR4) -dependent mechanism in vitro and in vivo. Using a proteomic approach, we analyzed the protein content of plasma EVs and identified critical activated pathways in human umbilical vein endothelial cells (HUVECs) and human progenitor hematopoietic cells (CD34+). We then showed that vessel formation was due to rapid mitochondrial activation, intracellular Ca2+ mobilization, increased extracellular ATP, and trafficking of the lysosomal P2XR4 to the cell membrane, which is required for cell motility and formation of stable branching vascular networks. Cell membrane translocation of P2XR4 was induced by proteins and chemokines contained in EVs (e.g. Del-1 and SDF-1). Del-1 was found expressed in many EVs from sarcoma tumors and several tumor types. P2XR4 blockade reduced EVs-induced vessels in angioreactors, as well as intratumor vascularization in mouse xenografts. Together, these findings identify P2XR4 as a key mediator of EVs-induced tumor angiogenesis via a signaling mediated by mitochondria-lysosome-sensing response in endothelial cells, and indicate a novel target for therapeutic interventions.

Optimal water quality is vital for the growth of Atlantic salmon aquaculture production. Recent data showed that Atlantic salmon feed intake and growth reduce linearly with increasing water carbon dioxide (CO2) concentrations, suggesting that even relatively low concentrations may impact fish performance. This study evaluated the molecular and physiological responses of Atlantic salmon (Salmo salar) to long-term CO2 exposure. For this purpose, Atlantic salmon post-smolts (N = 900; 67 ± 8 g) were exposed to six CO2 treatments (5, 12, 19, 26, 33 and 40 mg/L) for 12-weeks (RAS phase) followed by non-CO2 exposure for a (<5 mg/L) period of 6-weeks (seawaterphase). Results from blood analysis of fish exposed to CO2 for 12 weeks showed that CO2 lead to significantly higher pH, K+, HCO3 − and PCO2 and lower Na+ and Cl− plasma concentrations. Whereas, haematocrit, Ca+, Mg2+, urea and glucose concentrations were similar among all CO2 treatments. After 6 weeks in the seawater phase, all the parameters that were previously altered, became similar among all CO2 treatments. Gill microarray results analysis showed 88 differentially expressed genes, resulting from the CO2 exposure. At the end of the RAS phase (week 12), fish exposed to high CO2 (40 mg/L) in comparison to fish exposed to low CO2 (5 mg/L), showed 60 down-regulated genes, including genes encoding proteins involved in immune responses, differentiation, and maintenance of tissue structure. There was no evidence for stress and metabolic changes directed to neutralization of disturbance caused with high CO2. After 6 weeks in the seawater phase, a switch of expression from down regulated to up-regulated was observed. In conclusion, the present study brings new insights on the molecular and physiological responses of Atlantic salmon post-smolts to long-term CO2 exposure. High carbon dioxide (CO2) concentrations negatively impact fish, which makes data on its tolerance especially relevant for production systems that can accumulate CO2 such as recirculating aquaculture system (RAS). The current study evaluates the effect of CO2 on the growth performance, welfare, and health of Atlantic salmon post-smolts in RAS. This study consisted of two phases. The first was a CO2 exposure phase, where eighteen tanks were used with six treatments in triplicate: 5, 12, 19, 26, 33 and 40 mg/L of CO2 during 12 weeks in a 12 ppt salinity RAS (hereafter RAS phase). In the second phase, PIT-tagged fish were transferred to a 34 ppt salinity single flow-through tank at CO2 < 5 mg/L (hereafter seawater phase) for an additional 6-week experimental period mimicking a seawater phase. Overall, mortality of fish exposed to CO2 was low and not related to treatments. The mean final body weight was significantly higher in the 5 mg/L treatment compared to CO2 treatments ≥12 mg/L at the end of RAS phase and to CO2 treatments ≥33 mg/L at the end of seawater phase. Moreover, regressions showed that growth significantly decreased linearly with increasing CO2 in the water. Eye cataracts and visible external damage on skin, operculum, and fins were inexistent and similar among CO2 treatments. Kidneys showed no signs of mineral deposits in any of the structures of the tissue. However, skin analysis showed that fish exposed to high CO2 concentrations had a significantly thinner dermis layer (both at the end of RAS and seawater phase) and a significantly thinner epidermis layer and lower mucus cells count (at the end of seawater phase). In conclusion, Atlantic salmon post-smolts cultured in brackish water RAS showed a maximum growth performance at CO2 concentrations below 12 mg/L. [EN] In this letter, a three-dimensional antenna solution for automotive applications is presented. The proposed solution is formed by two independent antennas, perpendicularly placed inside a plastic cover with a shark-fin shape. The antennas, designed with low-cost materials, exhibit omnidirectional radiation patterns in the azimuthal plane. The first antenna is a doubleshorted monopole that provides service from 698 to 960 MHz (in the LTE700, GSM850, and GSM900 bands), whereas the second antenna is a drop-shaped monopole that operates from 1.7 to 2.7 GHz (in the DCS1800, PCS1900, WCDMA2100, WLAN2400, LTE2600, WiMAX2350, and Wi-Fi at 2.4-GHz bands), and from 5.1 to 6 GHz (in theWi-Fi at 5 GHz and C2C bands). Results show a good matching and radiation efficiency at all bands.

Subtropical reefs provide an important habitat for flora and fauna, and proper monitoring is required for conservation. Monitoring these exposed and submerged reefs is challenging and available resources are limited. Citizen science is increasing in momentum, as an applied research tool and in the variety of monitoring approaches adopted. This paper aims to demonstrate an ecological assessment and mapping approach that incorporates both top-down (volunteer marine scientists) and bottom-up (divers/community) engagement aspects of citizen science, applied at a subtropical reef at Point Lookout, Southeast Queensland, Australia. Marine scientists trained fifty citizen scientists in survey techniques that included mapping of habitat features, recording of substrate, fish and invertebrate composition, and quantifying impacts (e.g., occurrence of substrate damage, presence of litter). In 2014 these volunteers conducted four seasonal surveys along semi-permanent transects, at five sites, across three reefs. The project presented is a model on how citizen science can be conducted in a marine environment through collaboration of volunteer researchers, non-researchers and local marine authorities. Significant differences in coral and algal cover were observed among the three sites, while fluctuations in algal cover were also observed seasonally. Differences in fish assemblages were apparent among sites and seasons, with subtropical fish groups observed more commonly in colder seasons. The least physical damage occurred in the most exposed sites (Flat Rock) within the highly protected marine park zones. The broad range of data collected through this top-down/bottom-up approach to citizen science exemplifies the projects' value and application for identifying ecosystem trends or patterns. The results of the project support natural resource and marine park management, providing a valuable contribution to existing scientific knowledge and the conservation of local reefs.

BACKGROUND: Several studies suggest beneficial effects of perioperative statin therapy on postoperative outcome after cardiac surgery. However, recent randomized controlled trials (RCTs) show potential detrimental effects. The objective of this systematic review is to examine the association between perioperative statin therapy and clinical outcomes in cardiac surgery patients. METHODS: Electronic databases were searched up to 1 November 2016 for RCTs of preoperative statin therapy versus placebo or no treatment in adult cardiac surgery. Postoperative outcomes were acute kidney injury, atrial fibrillation, myocardial infarction, stroke, infections, and mortality. We calculated odds ratios (ORs) and 95% confidence intervals (CIs) using fixed-effects meta-analyses. Primary analysis was restricted to trials with low risk of bias according to Cochrane methodology, and sensitivity analyses examined whether the risk of bias of included studies was associated with different results. We performed trial sequential analysis (TSA) to test the strength of the results. RESULTS: We included data from 23 RCTs involving 5102 patients. Meta-analysis of trials with low risk of bias showed that statin therapy was associated with an increase in acute kidney injury (314 of 1318 (23.82%) with statins versus 262 of 1319 (19.86%) with placebo; OR 1.26 (95%CI 1.05 to 1.52); p = 0.01); these results were supported by TSA. No difference in postoperative atrial fibrillation, myocardial infarction, stroke, infections, or mortality was present. On sensitivity analysis, statin therapy was associated with a slight increase in hospital mortality. Meta-analysis including also trials with high or unclear risk of bias showed no beneficial effects of statin therapy on any postoperative outcomes. CONCLUSIONS: There is no evidence that statin therapy in the days prior to cardiac surgery is beneficial for patients' outcomes. In order to get a compact solution, the profile of a slot Vivaldi antenna has been modified and a wideband antenna covering LTE700, GSM850, GSM900, DCS1800, PCS1900, WCDMA2100, Bluetooth, WiMAX2350, WLAN2400, and LTE2600 standards has been obtained, with a size reduction length of 30% over conventional monopole antennas. In contrast to traditional designs using Vivaldi antennas, a compact design with omnidirectional radiation pattern is proposed. Simulated and measured results are provided, which verify the broadband behavior of the antenna. In addition, the proposed design is perpendicularly mounted to a ground plane, being then suitable for mobile communications in vehicular applications.

A modal analysis of the antenna is carried out first by means of the Theory of Characteristic Modes, in order to identify the different radiating modes of the antenna. Then a set of feeding configurations is proposed so as to excite these modes. As the modes must operate in the same frequency band, a loading technique is used in the antenna in order to shift the resonance frequency of the modes to the proper band.
