European canker, caused by the necrotrophic fungus Neonectria ditissima, is the most serious disease in apple production in Sweden. The disease is favored by a relatively cool and rainy climate. The canker damages have a significant economic impact due to reduced bearing surface and increased orchard management costs. The possibilities for chemical and biological control are very limited. Therefore, directed breeding for new resistant cultivars is urgently needed. Knowledge of inheritance of canker resistance and understanding of molecular mechanisms involved in resistant and susceptible responses to fungal attacks would facilitate breeding. In this study, we evaluated the tempo-spatial differences in plant-pathogen interactions in a set of partially resistant and susceptible cultivars by conducting metabolomic and lipidomic analyses. The major trends in metabolomics and lipidomic profiles were common among cultivars, irrespective of the degree of susceptibility. Several metabolites and lipids varied with time point and cultivar under N. ditissima infection. Putative key metabolites such as suberic acid and jasmonic acid were upregulated in all cultivars upon infection. Additionally, several lipids exhibited changes 30 to 45 days post-inoculation. Thus, the approach used seems to have resulted in a rich data set to be further analyzed in light of ongoing QTL-mapping efforts.

The present study quantifies the reduction in the levelized cost of electricity (LCoE) and capital expenditure (CAPEX) of a dish Stirling power plant (DSPP) through an increase in localization and unit production volume. Furthermore, the localization value of the plant is examined to determine how much investment is brought into the local economy. Ouarzazate, Morocco, was chosen as the location of the study due to the country's favorable regulatory framework with regards to solar power technologies and its established industry in the concentrating solar power (CSP) field. A detailed techno-economic model of a DSPP was developed using KTH's in-house modelling tool DYESOPT, which allows power plant evaluation by means of technical and economic performance indicators. Results on the basis of LCoE and CAPEX were compared between two different cases of production volume, examining both a minimum and maximum level of localization. Thereafter, the DSPP LCoE and localization value were compared against competing solar technologies to evaluate its competitiveness. In addition, a sensitivity analysis was conducted around key design parameters. The study confirms that the LCoE of a DSPP can be reduced to values similar to solar photovoltaic (PV) and lower than other CSP technologies. Furthermore, the investment in the local economy is far greater when compared to PV and of the same magnitude to other CSP technologies. Following the introduction, Chapter 2 provides an overview of mitigation in the forest sector, addressing the handling of forests under UNFCCC. Chapters 3 to 5 focus on forest-based mitigation options – afforestation, reforestation, REDD+ and forest management – and Chapters 6 and 7 focus on wood-product based options – wood energy and green building and furnishing. The publication describes these activities in the context of UNFCCC rules, assessing their mitigation potential and economic attrac tiveness as well as opportunities and challenges for implementation. Chapter 8 discusses the different considerations involved in choosing the right mix of options as well as some of the instruments and means for implementation. Chapter 8 also highlights the co-benefits generated by forest-based mitigation and emphasizes that economic assessment of mitigation options needs to take these benefits into account. The concluding chapter assesses national commitments under UNFCCC involving forest miti gation and summarizes the challenges and opportunities.

Background: Methane production as biofuels is a fast and strong growing technique for renewable energy. Substrates like waste (e.g. food, sludge fromwaste water treatment plants (WWTP), industrial wastes) can be used as a suitable resource for methane gas production, but in some cases, with elevated toxicity in the digestion residue. Former investigations have shown that co-digesting of contaminated waste such as sludge together with other substrates can produce a less toxic residue. In addition, wetlands and reed beds demonstrated good results in dewatering and detoxifying of sludge. The aim of the present study was to investigate if the toxicity may alter in industrial sludge co-digested with oat and post-treatment in reed beds. In this study, digestion of sludge from Bjorkborn industrial area in Karlskoga (reactor D6) and co-digestion of the same sludge mixed with oat (reactor D5) and post-treatment in reed beds were investigated in parallel. Methane production as well as changes in cytotoxicity (Microtox(R); ISO 11348-3), genotoxicity (Umu-C assay; ISO/13829) and AhR-mediated toxicity (7-ethoxyresorufin-O-deethylase (EROD) assay using RTW cells) were measured. Results: The result showed good methane production of industrial sludge (D6) although the digested residue was more toxic than the ingoing material measured using microtox30min and Umu-C. Co-digestion of toxic industrial sludge and oat (D5) showed higher methane production and significantly less toxic sludge residue than reactor D6. Furthermore, dewatering and treatment in reed beds showed low and non-detectable toxicity in reed bed material and outgoing water as well as reduced nutrients. Conclusions: Co-digestion of sludge and oat followed by dewatering and treatment of sludge residue in reed beds can be a sustainable waste management and energy production. Here, we studied the interaction of cationic uremic toxins with renal organic cation transport in a conditionally immortalized human proximal tubule epithelial cell line (ciPTEC). Transporter activity was measured and validated in cell suspensions by studying uptake of the fluorescent substrate 4-(4-(dimethylamino)styryl)-N-methylpyridinium-iodide (ASP(+)). Subsequently, the inhibitory potencies of the cationic uremic toxins, cadaverine, putrescine, spermine and spermidine (polyamines), acrolein (polyamine breakdown product), guanidine, and methylguanidine (guanidino compounds) were determined. Concentration-dependent inhibition of ASP(+) uptake by TPA, cimetidine, quinidine, and metformin confirmed functional endogenous organic cation transporter 2 (OCT2) expression in ciPTEC. All uremic toxins tested inhibited ASP(+) uptake, of which acrolein required the lowest concentration to provoke a half-maximal inhibition (IC50 = 44 +/- 2 muM). A Dixon plot was constructed for acrolein using three independent inhibition curves with 10, 20, or 30 muM ASP(+), which demonstrated competitive or mixed type of interaction (K i = 93 +/- 16 muM). Exposing the cells to a mixture of cationic uremic toxins resulted in a more potent and biphasic inhibitory response curve, indicating complex interactions between the toxins and ASP(+) uptake. In conclusion, ciPTEC proves a suitable model to study cationic xenobiotic interactions. Inhibition of cellular uptake transport was demonstrated for several uremic toxins, which might indicate a possible role in kidney disease progression during uremia.

Since the mid-1970s, Sweden has been depositing 1 million ton d.w sludge/year, produced at waste water treatment plants. Due to recent legislation this practice is no longer a viable method of waste management. It is necessary to improve existing and develop new sludge management techniques and one promising alternative is the dewatering and treatment of sludge in constructed wetlands. The aim of this study was to follow reduction of organic carbon, BOD and nutrients in an industrial sludge containing nitro-aromatic compounds passing through constructed small-scale wetlands, and to investigate any toxic effect such as growth inhibition of the common reed Phragmites australis. The result showed high reduction of all tested parameters in all the outgoing water samples, which shows that constructed wetlands are suitable for carbon and nutrient removal. The results also showed that P. australis is tolerant to xenobiotics and did not appear to be affected by the toxic compounds in the sludge. The sludge residual on the top of the beds contained low levels of organic carbon and is considered non-organic and could therefore be landfilled. Using energy systems analysis, we examine the potential to reduce CO 2 emissions and oil use by integrating motor biofuel production with pulp mills. BLG-DME (black liquor gasification with di-methyl ether production) is compared with solid biomass gasification with BIG-FT (solid biomass gasification with Fischer-Tropsch fuel production). The studied systems are expanded with stand-alone production of biomass-based electricity and motor fuel so that they yield the same functional unit in terms of motor fuel and electricity as well as pulp or paper product, in order to facilitate comparison. More motor biofuel can be produced in integration with the studied mills with BLG-DME than with BIG-FT because the black liquor flow is large compared with other fuel streams in the mill and the integration potential for BIG-FT is limited by the mill’s heat demand. When both systems are required to produce the same functional unit, the BLG-DME system achieves higher system efficiency and larger reductions in CO 2 emissions and oil use per unit of biomass consumed. In general, integration of motor biofuel production with a pulp mill is more efficient than stand-alone motor biofuel production. Larger reductions in CO 2 emissions or oil use can, however, be achieved if biomass replaces coal or oil in stationary applications. © 2012 Elsevier Ltd.

Traditional farming systems have been associated with the occurrence of intervarietal and interspecific natural crosses in many crop plants, thus contributing to the genetic diversity of the crop through genetic exchange. In this study, a combination of farmerpreferred morphological traits and RAPD markers were used to evaluate the dynamics of genetic diversity in 43 watermelon accessions collected at a single village level with a traditional farming system. The molecular variability assessed with RAPD markers and analyzed with multidimensional scaling and cluster analysis, demonstrated a substantial differentiation among the accessions. Population structure analysis also demonstrated the existence of three major forms of watermelon, identified by a set of alleles predominant within each form. Dendrograms based on RAPD data and on farmer-preferred traits data were positively correlated according to a Mantel test. Although cultivated cow-melons were genetically most similar to wild-weedy plants at molecular level, they grouped more similar to sweet watermelons based on farmer-preferred traits. In this paper, we examine how an increased use of biomass could efficiently meet Swedish energy policy goals of reducing carbon dioxide (CO2) emissions and oil use. In particular, we examine the trade-offs inherent when biomass use is intended to pursue multiple objectives. We set up four scenarios in which up to 400 PJ/year of additional biomass is prioritised to reduce CO2 emissions, reduce oil use, simultaneously reduce both CO2 emission and oil use, or to produce ethanol to replace gasoline. Technologies analysed for using the biomass include the production of electricity, heat, and transport fuels, and also as construction materials and other products. We find that optimising biomass use for a single objective (either CO2 emission reduction or oil use reduction) results in high fulfilment of that single objective (17.4 Tg C/year and 350 PJ oil/year, respectively), at a monetary cost of 130–330 million €/year, but with low fulfilment of the other objective. A careful selection of biomass uses for combined benefits results in reductions of 12.6 Tg C/year and 230 PJ oil/year (72% and 67%, respectively, of the reductions achieved in the scenarios with single objectives), with a monetary benefit of 45 million €/year. Prioritising for ethanol production gives the lowest CO2 emissions reduction, intermediate oil use reduction, and the highest monetary cost.

An industrial sludge containing a complex mixture of nitroaromatic compounds was treated in industrial large-scale aerobic and anaerobic biodegradation processes, performed in compost sacs. The goal was to study changes in genotoxicity during the two different oxygen regimes using the umuC genotoxicity assay. The composting sac was actively aerated during 3 months and allowed to mature for another 3 months. The anaerobic sac was not aerated for 5 months and aerated during the last month in order to enhance degradation of remaining organic carbon. The sludge was obtained from the wastewater treatment plant at an industrial area in Karlskoga, Sweden. The biodegradation study was performed at a commercial waste treatment plant in Stockholm, according to the company routine procedure when treating household waste in sealed sacs. The material from the non-aerated system showed increased genotoxicity in the acetone-soluble fraction after treatment, as did the water-soluble fraction. The subsequent aeration period did not decrease the toxicity below the genotoxicity limit. The increase in the water-soluble genotoxic compounds may pose an environmental problem during secondary storage or use of sludge treated this way, since leakage of water-dissolved genotoxic compounds may occur. The composting process also generated genotoxicity, but this was restricted to acetone-soluble compounds, while the water-soluble compounds remained low in genotoxicity. The aerated process therefore seems more favorable in term of risk reduction of this industrial sludge, although it is necessary to optimize the aerated process in order to achieve non-toxic levels of potential genotoxic compounds extractable by organic solvents.
