Light-based gamma entrainment using sensory stimuli (GENUS) shows considerable potential for the treatment of Alzheimer's disease (AD) in both animal and human models. While the clinical efficacy of GENUS for AD is paramount, its effectiveness will eventually also rely on the barrier to treatment adherence imposed by the discomfort of gazing at luminance flickering (LF) light. Currently, there have been few attempts to improve the comfort of GENUS. Here we investigate if Invisible spectral flicker (ISF), a novel type of light-based 40 Hz GENUS for which the flicker is almost imperceptible, can be used as a more comfortable option. We found that whereas ISF, LF, and chromatic flicker (CF) all produce a 40 Hz steady-state visually evoked potential (SSVEP), ISF scores significantly better on measures of comfort and perceived flicker. We also demonstrate that, while there is a trend towards a lower SSVEP response, reducing the stimulation brightness has no significant effect on the 40 Hz SSVEP or perceived flicker, though it significantly improves comfort. Finally, there is a slight decrease in the 40 Hz SSVEP response when stimulating with ISF from increasingly peripheral angles. This may ease the discomfort of GENUS treatment by freeing patients from gazing directly at the light.

Background: Major depressive disorder (MDD) is a debilitating condition that affects more than 300 million people worldwide. Current treatments are based on a trial-and-error approach, and reliable biomarkers are needed for more informed and personalized treatment solutions. One of the potential biomarkers, gamma-frequency (30–80 Hz) brainwaves, are hypothesized to originate from the excitatory-inhibitory interaction between the pyramidal cells and interneurons. The imbalance between this interaction is described as a crucial pathological mechanism in neuropsychiatric conditions, including MDD, and the modulation of this pathological interaction has been investigated as a potential target. Previous studies attempted to induce gamma activity in the brain using rhythmic light and sound stimuli (GENUS–Gamma Entrainment Using Sensory stimuli) that resulted in neuroprotective effects in Alzheimer’s disease (AD) patients and animal models. Here, we investigate the antidepressant, cognitive, and electrophysiological effects of the novel light therapy approach using 40 Hz masked flickering light for patients diagnosed with MDD. Methods and design: Sixty patients with a current diagnosis of a major depressive episode will be enrolled in a randomized, double-blinded, placebo-controlled trial. The active treatment group will receive 40 Hz masked flickering light stimulation while the control group will receive continuous light matched in color temperature and brightness. Patients in both groups will get daily light treatment in their own homes and will attend four follow-up visits to assess the symptoms of depression, including depression severity measured by Hamilton Depression Rating Scale (HAM-D17), cognitive function, quality of life and sleep, and electroencephalographic changes. The primary endpoint is the mean change from baseline to week 6 in depression severity (HAM-D6 subscale) between the groups.

Hickson compact groups (HCGs) are dense configurations of four to ten galaxies, whose HI morphology appears to follow an evolutionary sequence of three phases, with gas initially confined to galaxies, then significant amounts spread throughout the intra-group medium, and finally with almost no gas remaining in the galaxies themselves. It has also been suggested that several groups may harbour a diffuse HI component that is resolved out by interferometric observations. The HI deficiency of HCGs is expected to increase as the HI morphological phase progresses along the evolutionary sequence. If this is the case, HI deficiency would be a rough proxy for the age and evolutionary state of a HCG. We aim to test this hypothesis for the first time using a large sample of HCGs and to investigate the evidence for diffuse HI in HCGs. We performed a uniform reduction of all publicly available VLA HI observations (38 HCGs) with a purpose-built pipeline that also maximises the reproducibility of this study. The resulting HI data cubes were then analysed with the latest software tools to perform a manual separation of emission features into those belonging to galaxies and those extending into the intra-group medium. We thereby classified the HI morphological phase of each group as well as quantified their HI deficiency compared to galaxies in isolation. We find little evidence that HI deficiency can be used as a proxy for the evolutionary phase of a compact group in either of the first two phases, with the distribution of HI deficiency being consistent in both. However, for the final phase, the distribution clearly shifts to high HI deficiencies, with more than 90% of the expected HI content typically missing. Across all HCGs studied, we identify a few cases where there is strong evidence for a diffuse gas component in the intra-group medium, which might be detectable with improved observations. Integrating bioenergy and carbon capture and storage (BECCS) presents a great opportunity for power production with negative global CO2 emissions. This work explores a novel synergetic system that integrates membranes, partial biomass oxycombustion and the calcium looping (CaL) process. Polymeric membranes generate oxygen-enriched air (OEA) with an O2 concentration of 39%v/v, which is used for partial oxycombustion of biomass waste. The CO2-enriched flue gas evolves from the waste-to-energy plant to the CaL unit, where CO2 concentration is increased up to 90–95%v/v, ready for purification and sequestration. Compared to only oxycombustion systems, the proposed concept presents fewer technological challenges in retrofitting boilers to waste-to-energy plants. Moreover, this new approach is highly efficient as integrating membranes to produce OEA instead of cryogenic distillation systems significantly reduces energy consumption. A novel integration concept is modelled to evaluate the whole process efficiency and the effect of key parameters on the system performance, such as the temperature of the reactors, the membrane surface area, and the partial oxy-combustion degree. The results show that the so-called mOxy-CaL system has an energy consumption associated with CO2 capture below 4 MJ/kg CO2 (a 31% lower than that for a conventional CaL process), with a higher CO2 capture efficiency than oxycombustion and the CaL process separately. On the other hand, the economic analysis shows a higher CO2 capture cost for the novel configuration than for the typical CaL configuration due to the additional investment cost of the membrane system. Improvements in membrane performance by increasing its permeance and diminishing the required surface area would significantly reduce the economic cost of this novel integration. Using membranes with permeance over 400 GPU would boost the system's competitiveness.

This paper provides a review of negative carbon capture technologies. Based on these technologies, here it is proposed an innovative negative emissions power plant combining the generation and storage of energy from biomass, photovoltaic, and concentrated solar power, capturing and recovering CO2 by producing H2 or CH4 as green energy carriers. The main features of the system are i) large-scale energy production system with negative CO2 emissions; ii) 100% renewable system based on biomass and solar energy with the possibility of integrating other renewables; iii) synergistic integration of processes and systems; iv) recovery of O2 generated by photovoltaic-driven electrolysis within the process of partial biomass oxycombustion and v) solar-driven limestone calcination. A detailed model of the entire plant is developed to evaluate the integration of the process. The model performance is assessed on an hourly basis throughout the whole year. The base case results show an energy consumption from 1 to 2.1 MJ/kg CO2 to capture 60–77% of CO2 emitted from the biomass plant and green methane production of more than 7500 tons/year. The negative emissions associated with the process are -612 kg CO2/MWh. It justifies the interest in the proposed negative emissions power plant.

Partial oxycombustion using Oxygen-Enriched Air (OEA), produced by air-gas separation with polymeric membranes, combined synergistically with CO2 capture technologies, can reduce the overall energy cost of CO2 capture, and it is a potential alternative to conventional CO2 capture technologies. An exhaustive review of polymeric membranes for this application is presented. The best membranes showed permeability values in the 450–25,100 barrer and selectivities higher than 3.6 for large-scale operations. These membranes can produce OEA with oxygen molar concentrations of up to 40% for retrofitting large-scale power plants (∼ 500 MWe) with partial oxycombustion. For OEA production, the polymeric membrane system is more efficient than cryogenic distillation since the specific power consumption of the former is 35.17 kWh/ton OEA. In comparison, that of the latter is 49.57 kWh/ton OEA. This work proposes that the OEA produced by the membranes feed a partial oxycombustion process integrated with calcium looping within a hybrid CO2 capture system. The power consumption of the hybrid CO2 capture system proposed here is 29.05% lower than in the case OEA is produced from cryogenic distillation, which justifies the potential interest in using polymeric membranes for OEA production.

Hickson Compact Groups (HCGs) are dense configurations of 4 to 10 galaxies, whose HI (neutral gas) morphology appears to follow an evolutionary sequence of three phases, with gas initially confined to galaxies, then significant amounts spread throughout the intra-group medium, and finally with almost no gas remaining in the galaxies themselves. The HI deficiency of HCGs is expected to increase as the HI morphological phase progresses along this sequence, potentially making it a useful proxy for evolutionary phase. We test this hypothesis for the first time with a large sample of 38 HCGs with VLA HI observations that are uniformly reduced and analysed with a purpose-built pipeline. However, we find little evidence that HI deficiency can be used as a proxy for the evolutionary phase of a HCG in either of the first two phases, with the distribution of HI deficiency being consistent in both, although it does greatly increase in the third phase. This appears to be the result to three factors: a) there is already a broad range of HI deficiencies in Phase 1 HCGs, possibly due to their differing locations relative to large scale structures; b) the timescale for major interactions and morphological changes is, in general, considerably shorter than the timescale for the destruction or consumption of HI gas; and c) some groups have their HI content rejuvenated by the late addition of a new gas-rich member (for which we added a new sub-phase, 3c, to the established evolutionary sequence). Finally, across all HCGs studied, we identify only a few cases where there is strong evidence for the existence of a previously proposed diffuse HI component in the intra-group medium, which might be detectable with improved observations. Marine citizen science is emerging with promising opportunities for science, policy and public but there is still no comprehensive overview of the current state in Europe. Based on 127 projects identified for the North Sea area we estimate there might be as much as 500 marine and coastal citizen science projects running in Europe, i.e., one marine citizen science project per 85 km of coastline, with an exponential growth since 1990. Beach-based projects are more accessible and hence most popular (60% of the projects), and the mean duration of the projects is 18–20 years. Current trends, topics, organizers, aims, and types of programme in terms of participation are presented in this overview. Progress in marine citizen science is specially enabled and promoted through technological developments. Recent technological advances and best practise examples are provided here, untapping the potential of smart mobile apps, do-it-yourself (DIY) technologies, drones, and artificial intelligence (AI) web services

Research involving the general population of people who use a spoken language to communicate has demonstrated that older adults experience cognitive and physical changes associated with aging. Notwithstanding the differences in the cognitive processes involved in sign and spoken languages, it is possible that aging can also affect cognitive processing in deaf signers. This research aims to explore the impact of aging on spatial abilities among sign language users. Results showed that younger signers were more accurate than older signers on all spatial tasks. Therefore, the age-related impact on spatial abilities found in the older hearing population can be generalized to the population of signers. Potential implications for sign language production and comprehension are discussed.

The recent sports science literature conveys a growing interest in robust statistical methods to analyze smooth, regularly-sampled functional data. This paper focuses on the inferential problem of identifying the parts of a functional domain where two population means differ. We considered four approaches recently used in sports science: interval-wise testing (IWT), statistical parametric mapping (SPM), statistical nonparametric mapping (SnPM) and the Benjamini-Hochberg (BH) procedure for false discovery control. We applied these procedures to both six representative sports science datasets, and also to systematically varied simulated datasets which replicated ten signal- and/or noise-relevant parameters that were identified in the experimental datasets. We observed generally higher IWT and BH sensitivity for five of the six experimental datasets. BH was the most sensitive procedure in simulation, but also had relatively high false positive rates (generally > 0.1) which increased sharply (> 0.3) in certain extreme simulation scenarios including highly rough data. SPM and SnPM were more sensitive than IWT in simulation except for (1) high roughness, (2) high nonstationarity, and (3) highly nonuniform smoothness. These results suggest that the optimum procedure is both signal and noise-dependent. We conclude that: (1) BH is most sensitive but also susceptible to high false positive rates, (2) IWT, SPM and SnPM appear to have relatively inconsequential differences in terms of domain identification sensitivity, except in cases of extreme signal/noise characteristics, where IWT appears to be superior at identifying a greater portion of the true signal.

Alzheimer's disease (AD) not only takes an emotional toll on the individual with the disease, their families, relatives, and caretakers, it also has immense socioeconomic consequences on the health care system and society. Moreover, the socioeconomic consequences are expected to increase significantly, thus reducing the social and economic cost of AD is of high importance. Recently, exposure to 40 Hz stroboscopic light therapy, for one hour a day, resulted in slowing the progression of AD in mice and has a considerable potential for treatment in humans. However, exposure to such stroboscopic light carries its own consequences being that it is difficult to implement in a patient's daily routine, irksome to use, and can cause visual discomfort which may result in a lack of patient adherence. Here, we demonstrate a novel technology based on controlling multiple single-color LEDs to produce white light where its spectral composition alternates at a given modulation frequency without visible flicker. We coin this technique as Invisible Spectral Flicker (ISF). We present 40 Hz invisible spectral flicker light as a potential alternative in reducing discomfort compared to 40 Hz stroboscopic light, whilst still entraining oscillations in various areas of the brain. Furthermore, we demonstrate a distinct way to generate a 40 Hz metameric light source with the presented color mixing scheme, and validate that the CIE 1931 (x, y) coordinates match for two different spectral power distributions. Finally, we illustrate the light characteristics of seven 40 Hz color fusion light sources and two 40 Hz stroboscopic light sources. The technology presented here will lead to new, and hopefully improved, designs of light therapy systems for the treatment of Alzheimer's and dementia.

In this work we focus on the problem of local inference for functional data. We describe a unified framework for testing hypotheses on functional data in a local perspective. The result of the testing procedures within the unified framework is an adjusted p-value function that can be used to select the areas of the domain responsible for the rejection of the null hypothesis. We discuss how different state of the art inferential procedures fall within the framework, and briefly describe a novel testing procedure with sound theoretical properties.

The functionality available on modern ‘smartphone’ mobile devices, along with mobile application software and access to the mobile web, have opened up a wide range of ways for volunteers to participate in environmental and biodiversity research by contributing wildlife and environmental observations, geospatial information, and other context-specific and time-bound data. This has brought about an increasing number of mobile phone based citizen science projects that are designed to access these device features (such as the camera, the microphone, and GPS location data), as well as to reach different user groups, over different project durations, and with different aims and goals. In this chapter we outline a number of key considerations when designing and developing mobile applications for citizen science, with regard to (1) Interoperability. The factors that influence the usability of the mobile application are covered in both (2) Participant Centred Design and Agile Development, and (3) User Interface and Experience Design. Finally, the factors that influence sustained engagement in the project are covered in (4) Motivational Factors for Participation.

We present here a one-tube-only standardized SDM approach that generates comprehensive collections of amino acid substitution variants, including scanning- and single site-multiple mutations. The approach combines unified mutagenic primer design with the mixing of multiple distinct primer pairs and/or plasmid templates to increase the yield of a single inverse-PCR mutagenesis reaction. Also, a user-friendly program for automatic design of standardized primers for Ala-scanning mutagenesis is made available. Experimental results were compared with a modeling approach together with stochastic simulation data. For single site-multiple mutagenesis purposes and for simultaneous mutagenesis in different plasmid backgrounds, combination of primer sets and/or plasmid templates in a single reaction tube yielded the distinct mutations in a stochastic fashion. For scanning mutagenesis, we found that a combination of overlapping primer sets in a single PCR reaction allowed the yield of different individual mutations, although this yield did not necessarily follow a stochastic trend. Double mutants were generated when the overlap of primer pairs was below 60%. Our results illustrate that one-tube-only SDM effectively reduces the number of reactions required in large-scale mutagenesis strategies, facilitating the generation of comprehensive collections of protein variants suitable for functional analysis.

We describe a standardized, rapid, and simple method of mutagenesis to perform single and multiple amino acid substitutions, as well as deletions of short nucleotide sequences, based on one-step inverse PCR and DpnI restriction enzyme treatment. This method of SDM is generally applicable to any other protein of interest.

Cloning and sequencing of the progesterone receptor gene in dogs have revealed 2 isoforms, A and B, transcribed from a single gene. Distribution of isoforms A and B in canine mammary lesions has hitherto been investigated only by Western blot analysis. This study analyzed progesterone receptor and its isoforms in formalin-fixed, paraffin-embedded tissue samples from canine mammary lesions (4 dysplasias, 10 benign tumors, and 46 carcinomas) using 1-step SYBR Green quantitative real-time polymerase chain reaction (RT-qPCR). Progesterone receptor was expressed in 75% of dysplasias, all benign tumors, and 59% of carcinomas. Carcinomas, and particularly simple epithelial-type carcinomas, displayed the lowest levels of expression. A high rate of agreement was recorded between RT-qPCR and immunohistochemical labeling. Isoforms A and B were successfully amplified, with correlation coefficients of 0.99 and amplification efficiencies close to 2, and were expressed in all lesion types analyzed. Predominance of A over B expression was observed in carcinomas and complex adenomas. Low-grade tumors exhibited higher progesterone receptor messenger RNA (mRNA) levels, but no difference was observed in the expression of isoform A versus B. Analysis of progesterone receptor mRNA isoforms by RT-qPCR was successful in routinely formalin-fixed, paraffin-embedded tissue samples and enabled the distribution of isoforms A and B to be identified for the first time in dysplasias, benign tumors, and malignant tumors of the canine mammary gland. These findings will facilitate future research into the role of progesterone receptor isoforms in the progression of canine mammary tumors.

The mosquito Aedes aegypti is the main vector of Dengue and Yellow Fever flaviviruses. The organophosphate insecticide temephos is a larvicide that is used globally to control Ae. aegypti populations; many of which have in turn evolved resistance. Target site alteration in the acetylcholine esterase of this species has not being identified. Instead, we tracked changes in transcription of metabolic detoxification genes using the Ae. aegypti ‘Detox Chip’ microarray during five generations of temephos selection. We selected for temephos resistance in three replicates in each of six collections, five from México, and one from Perú. The response to selection was tracked in terms of lethal concentrations (LC50). Uniform upregulation was seen in the epsilon class glutathione-S-transferase genes (eGSTs) in strains from México prior to laboratory selection, while eGSTs in the Iquitos Perú strain became upregulated following five generations of temephos selection. While expression of many esterase genes (CCE) increased with selection, no single esterase was consistently upregulated and this same pattern was noted in the cytochrome P450 genes (CYP) and in other genes involved in reduction or oxidation of xenobiotics. Bioassays using GST, CCE and CYP inhibitors suggest that various CCE instead of GSTs are the main metabolic mechanism conferring resistance to temephos. We show that temephos selected strains show no cross resistance to permethrin and that genes associated with temephos selection are largely independent of those selected with permethrin in a previous study.

The mosquito Aedes aegypti is the main vector of Dengue and Yellow Fever flaviviruses. The organophosphate insecticide temephos is a larvicide that is used globally to control Ae. aegypti populations; many of which have in turn evolved resistance. Target site alteration in the acetylcholine esterase of this species has not being identified. Instead, we tracked changes in transcription of metabolic detoxification genes using the Ae. aegypti ‘Detox Chip’ microarray during five generations of temephos selection. We selected for temephos resistance in three replicates in each of six collections, five from México, and one from Perú. The response to selection was tracked in terms of lethal concentrations (LC50). RESULTS: Our best model, which uses December precipitation and mink (Neovison vison, formerly Mustela vison) bagging figures, successfully predicted every major increase or decrease in TBE during the past quarter century, with a minimum of false alarms. However, this model was not efficient in predicting small increases and decreases. CONCLUSIONS: Predictions from our models can be used to determine when preventive and adaptive programmes should be implemented. For example, in years when the frequency of TBE in humans is predicted to be high, vector control could be intensified where infested ticks have a higher probability of encountering humans, such as at playgrounds, bathing lakes, barbecue areas and camping facilities. Because our models use only wildlife and climate data, they can be used even when the human population is vaccinated. Another advantage is that because our models employ data from previously-established databases, no additional funding for surveillance is required.

[EN] Background: The objective of this study was to estimate the association between sleep quality (SQ) and improvements in low back pain (LBP) and disability, among patients treated for LBP in routine practice. Methods: This prospective cohort study included 461 subacute and chronic LBP patients treated in 11 specialized centres, 14 primary care centres and eight physical therapy practices across 12 Spanish regions. LBP, leg pain, disability, catastrophizing, depression and SQ were assessed through validated questionnaires upon recruitment and 3 months later. Logistic regression models were developed to assess: (1) the association between the baseline score for SQ and improvements in LBP and disability at 3 months, and (2) the association between improvement in SQ and improvements in LBP and disability during the follow-up period. Results: Seventy-three per cent of patients were subacute. Median scores at baseline were four points for both pain and disability, as assessed with a visual analog scale and the Roland-Morris Questionnaire, respectively. Regression models showed (OR [95% CI]) that baseline SQ was not associated with improvements in LBP (0.99 [0.94; 1.06]) or in disability (0.99 [0.93; 1.05]), although associations existed between ‘improvement in SQ’ and ‘improvement in LBP’ (4.34 [2.21; 8.51]), and ‘improvement in SQ’ and ‘improvement in disability’ (4.60 [2.29; 9.27]). Conclusions: Improvement in SQ is associated with improvements in LBP and in disability at 3-month follow-up, suggesting that they may reflect or be influenced by common factors. However, baseline SQ does not predict improvements in pain or disability. Significance: In clinical practice, sleep quality, low back pain and disability are associated. However, sleep quality at baseline does not predict improvement in pain and disability.
