Abstract

Background: In the last few years, new noninvasive strategies have emerged as rehabilitative treatments for patients with stroke. Action observation treatment (AOT) is a rehabilitation approach based on the properties of the mirror neuron system with a positive impact on modifying cortical activation patterns and improving the upper limb kinematics. AOT involves the dynamic process of observing purposeful actions with the intention of imitating and then practicing those actions. In recent years, several clinical studies suggested the effectiveness of AOT in patients with stroke to improve motor recovery and autonomy in activities of daily living. However, a deeper knowledge of the behavior of the sensorimotor cortex during AOT seems to be essential. Objective: The aim of this clinical trial, conducted in 2 neurorehabilitation centers and in patients’ homes, is to investigate the effectiveness of AOT in patients with stroke, confirming the translational power of a tailored treatment. Particular emphasis will be placed on the predictive value of neurophysiological biomarkers. In addition, the feasibility and impact of a home-based AOT program will be investigated. Methods: A 3-arm, assessor-blinded, randomized controlled trial will be performed by enrolling patients with stroke in the chronic stage. A total of 60 participants will be randomly allocated to receive 15 sessions of AOT with different protocols (AOT at the hospital, AOT at home, and sham AOT), 3 sessions per week. The primary outcome will be assessed using the Fugl-Meyer Assessment-Upper Extremity scores. Secondary outcomes will be clinical, biomechanical, and neurophysiological assessment. Results: The study protocol is part of a project (project code GR-2016–02361678) approved and funded by the Italian Ministry of Health. 3. Microfluidic flow injection immunoassay system for algal toxins determination:a case of study

Abstract

A novel flow injection microfluidic immunoassay system for continuous monitoring of saxitoxin, a lethal biotoxin, in seawater samples is presented in this article. The system consists of a preimmobilized G protein immunoaffinity column connected in line with a lab-on-chip setup. The detection of saxitoxin in seawater was carried out in two steps: an offline incubation step (competition reaction) performed between the analyte of interest (saxitoxin or Ag, as standard or seawater sample) and a tracer (an enzyme-conjugated antigen or Ag*) toward a specific polyclonal antibody. Then, the mixture was injected through a “loop” of a few μL using a six-way injection valve into a bioreactor, in line with the valve. The bioreactor consisted of a small glass column, manually filled with resin upon which G protein has been immobilized. When the mixture flowed through the bioreactor, all the antibody-antigen complex, formed during the competition step, is retained by the G protein. The tracer molecules that do not interact with the capture antibody and protein G are eluted out of the column, collected, and mixed with an enzymatic substrate directly within the microfluidic chip, via the use of two peristaltic pumps. When Ag* was present, a color change (absorbance variation, ΔAbs) of the solution is detected at a fixed wavelength (655 nm) by an optical chip docking system and registered by a computer. The amount of saxitoxin, present in the sample (or standard), that generates the variation of the intensity of the color, will be directly proportional to the concentration of the analyte in the analyzed solution. Indeed, the absorbance response increased proportionally to the enzymatic product and to the concentration of saxitoxin in the range of 3.5 × 10⁻⁷–2 × 10⁻⁵ ng ml⁻¹ with a detection limit of 1 × 10⁻⁷ ng ml⁻¹ (RSD% 15, S N⁻¹ equal to 3). Abstract

The synthesis of functional poly(2-alkyl-2-oxazoline) (PAOx) copolymers with complex nanoarchitectures using a graft-through ring-opening metathesis polymerization (ROMP) approach is described. First, well-defined norbornene-terminated poly(2-ethyl-2-oxazoline) (PEtOx) macromonomers (MM) were prepared by cationic ring-opening polymerization. ROMP of these MMs produced bottlebrush copolymers with PEtOx side chains. In addition, PEtOx-based branched MMs bearing a terminal alkyne group were prepared and conjugated to an azide-containing bis-spirocyclohexyl nitroxide via Cu-catalyzed azide-alkyne cycloaddition (CuAAC). ROMP of this branched MM, followed by in situ cross-linking, provided PEtOx-based brush-arm star polymers (BASPs) with nitroxide radicals localized at the core-shell interface. These PEtOx-based nitroxide-containing BASPs displayed relaxivity values on par with state-of-the-art polyethylene glycol (PEG)-based nitroxide materials, making them promising as organic radical contrast agents for metal-free magnetic resonance imaging (MRI).

Abstract

Liquid Electron Ionization (LEI), is an innovative liquid chromatography-mass spectrometry (LC–MS)interface that converts liquid HPLC eluent to the gas-phase in a mass spectrometer equipped with anelectron ionization (EI) source. LEI extends the electronic spectra libraries access to liquid chromatog-raphy, providing a powerful tool in the untargeted approacssh. Negligible matrix effects allow accuratequantitative information. The purpose of this research was to evaluate the main aspects concerning theinterfacing process. These fundamental studies were necessary to understand the mechanism of LEI indetails, and improve the interfacing process, especially regarding robustness and sensitivity. Hardwarecomponents were installed to prevent analytes precipitation, reduce thermal decomposition of sensitivecompounds, and to stabilize the nano-flow delivery with different mobile-phase compositions. Partic-ular attention was devoted to insulating the heated vaporization area from the LC part of the system.Experiments were performed to optimize the interface inner capillary dimensions, and other operativeparameters, including temperature, gas and liquid flow rates. Test compounds of environmental interestwere selected based on molecular weight, thermal stability, volatility, and polarity. Robustness was eval-uated with a set of replicated injections and calibration experiments using a soil matrix as a test sample.MRM detection limits in the low-picogram range were obtained for five pesticides belonging to differentclasses in a soil sample. High-quality electron ionization mass spectra of a mixture of pesticides werealso obtained.

Abstract

Background: Psoriasis (Ps) is a chronic systemic autoimmune disease associated with pruritus in 64–98% of patients. However, few modestly sized studies assess factors associated with psoriatic pruritus. Objective: To investigate factors associated with Ps pruritus intensity. Methods: Psoriasis patients 18 years or older seen in one of 155 centres in Italy between September 2005 and 2009 were identified from the Italian PsoCare registry. Patients without cutaneous psoriasis and those with missed information on pruritus were excluded. Results: We identified 10 802 patients, with a mean age 48.8 ± 14.3 years. Mild itch was present in 33.2% of patients, moderate in 34.4%, severe in 18.7% and very severe in 13.7%. Higher itch intensity was associated with female gender, lower educational attainment compared to university degree, pustular psoriasis, psoriasis on the head, face, palmoplantar areas, folds and genitalia, more severe disease, disease duration <15 years, and no or few prior systemic treatments. Limitations: Effects of specific medication on itch were not assessed. Conclusions: Pruritus should be evaluated during psoriasis visits, and physicians should be aware of patients at higher risk for itch. Further studies are needed to assess the effects of medications on itch, and establish therapy for psoriasis patients with persistent itch.

Abstract

The `Alfv\'en Paradox' is that as resistivity decreases, the discrete eigenmodes do not converge to the generalized eigenmodes of the ideal Alfv\'en continuum. To resolve the paradox, the $\epsilon$-pseudospectrum of the RMHD operator is considered. It is proven that for any $\epsilon$, the $\epsilon$- pseudospectrum contains the Alfv\'en continuum for sufficiently small resistivity. Formal $\epsilon-pseudoeigenmodes$ are constructed using the formal Wentzel-Kramers-Brillouin-Jeffreys solutions, and it is shown that the entire stable half-annulus of complex frequencies with $\rho{|\omega|^2}=|\bf{v} \cdot \bf{B}(x)|^2$ is resonant to order $\epsilon$, i.e.~belongs to the $\epsilon-pseudospectrum$. The resistive eigenmodes are exponentially ill-conditioned as a basis and the condition number is proportional to $\exp(R_M^{1\over 2})$, where $R_M$ is the magnetic Reynolds number.

Abstract

Metal-free magnetic resonance imaging (MRI) agents could overcome the established toxicity associated with metal-based agents in some patient populations and enable new modes of functional MRI in vivo. Herein, we report nitroxide-functionalized brush-arm star polymer organic radical contrast agents (BASP-ORCAs) that overcome the low contrast and poor in vivo stability associated with nitroxide-based MRI contrast agents. As a consequence of their unique nanoarchitectures, BASP-ORCAs possess per-nitroxide transverse relaxivities up to ∼44-fold greater than common nitroxides, exceptional stability in highly reducing environments, and low toxicity. These features combine to provide for accumulation of a sufficient concentration of BASP-ORCA in murine subcutaneous tumors up to 20 h following systemic administration such that MRI contrast on par with metal-based agents is observed. Abstract

Stimuli-responsive multimodality imaging agents have broad potential in medical diagnostics. Herein, we report the development of a new class of branched-bottlebrush polymer dual-modality organic radical contrast agents—ORCAFluors—for combined magnetic resonance and near-infrared fluorescence imaging in vivo. These nitroxide radical-based nanostructures have longitudinal and transverse relaxation times that are on par with commonly used heavy-metal-based magnetic resonance imaging (MRI) contrast agents. Furthermore, these materials display a unique compensatory redox response: fluorescence is partially quenched by surrounding nitroxides in the native state; exposure to ​ascorbate or ​ascorbate/​glutathione leads to nitroxide reduction and a concomitant 2- to 3.5-fold increase in fluorescence emission. This behaviour enables correlation of MRI contrast, fluorescence intensity and spin concentration with tissues known to possess high concentrations of ​ascorbate in mice. Abstract

Homogeneous hybrids in which iron oxide nanoparticles are entrapped within polymer structure are of interest for their potential applications in biomedical field, such as diagnostic, therapeutic and theranostic purposes. For this reason, hybrid nanomaterials based on branched polyethyleneimine (PEI) and iron oxide with different ratios were synthesized in a single step by hydrothermal procedure at high pressure and low temperature. Iron oxide is formed in the presence of branched PEI and the interaction between them takes place in the reaction medium. The influence of synthesis parameters on the hybrid formation, as well as chemical and structural properties was studied by means of FTIR, DSC-TG, HRTEM, electron paramagnetic resonance (EPR), magnetic measurements (SQUID) and 57Fe M€ossbauer analyses.It has been shown that synthesis parameters influence thermal stability and morphology of the hybrids. FeO(OH) crystallites of 2e5 nm are formed. Iron oxyhydroxide nanoparticles strongly entrapped in PEI structure are obtained. The low and distributed values of the specific spontaneous magnetisation in samples prepared under the same pressure conditions support the presence of very fine FeO(OH) nanoparticles, which formation and magnetic properties are depending on the mass ratio between iron oxide and PEI.

Abstract

Herein we describe the synthesis and characterization of new sulfonated and carboxylated poly(propyleneimino) (PPI) dendrimers with the ethylenediamino (EDA) core, at generations 1, 2 and 3. By means of UV-Vis and EPR spectroscopy, using Cu(2+) as a probe, we concluded that these dendrimers show a specific pattern in the coordination of metal ions. In agreement with the UV-Vis studies, EPR spectra of carboxylated compounds are constituted by 3 different signals which appear and then disappear with increasing copper concentration, corresponding to the saturation of different copper complexation sites. At the lowest copper concentration up to a 1:1 molar ratio between Cu(II) and the dendrimer, the spectrum is characteristic of a CuN2O2 coordination at the core of the dendrimer. The spectrum appearing at higher Cu(II) concentrations indicates a peripheral location of the ions coordinating one nitrogen and 3 oxygen atoms in a square planar geometry in restricted mobility conditions. For the highest concentrations tested, copper ions are confined at the external dendrimer surface with CuO4 coordination. For sulfonate systems, the EPR results are in line with a weaker interaction of Cu(II) with the nitrogen sites and a stronger interaction with the oxygen (SO3(-)) groups with respect to the interactions measured by EPR for carboxylate systems., QC 20171218

21. An economic perspective on auctions

Abstract

The recent spectrum auctions in Europe have shown that serious problems can wise in auctions where multiple complementary objects are being sold (such as blocks of radio spectrum) that will subsequently be used by the wining bidders to compete against each other in downstream markets. Other important instances of such situations include take-off and landing slots at airports and rights for electricity and gas transmission. T I first review some of the theory describing multi-object auctions. I I next outline the importance of strategic effects arising,in auctions that are followed by competition between the bidders., and the tension arising between various goals such as efficiency and revenue maximization. Although more flexible auction formats can have virtues (particularly in taking into account complementarities), they, can also be manipulated by bidders to build market power to the detriment Of consumers. We next apply these insights to the recent European UMTS licence auctions. Finally, we draw the main conclusions and policy, implications.

22. Effects of dopamine on the in vivo binding of dopamine D2 receptor radioligands in rat striatum

Abstract

The effects of moderate changes in extracellular dopamine concentrations on the in vivo binding of specific dopaminergic D2 radioligands with different affinities and kinetics were investigated in rats. Either [125I]NCQ298 (Kd = 19 pM), or [25I]iodolisuride (Kd = 0.27 nM) or [3H]raclopride (Kd = 1.5 nM) were administered intravenously (IV) to animals 1 h after the intraperitoneal (IP) injection of either alpha-methyl-p-tyrosine (AMPT) (250 mg/kg) or nomifensine (15 mg/kg), or saline. The kinetics of radioactivity concentration in the striatum, cerebellum, and plasma were measured for up to 4 h after [125I]NCQ298 or [125I]iodolisuride injection and up to 1.5 h after [3H]raclopride injection. For each tracer, the striatum-to-cerebellum radioactivity concentration ratios (S/C) and the binding potential (BP), calculated as the association to dissociation binding rate constant ratios (k3/k4), were assessed and related to the changes in extracellular dopamine concentration induced by drug treatments. Results show that S/C and BP of [3H]raclopride were significantly diminished by pretreatment with nomifensine, a drug that increases extracellular dopamine concentration. Nomifensine pretreatment induced no changes in the in vivo binding indexes of the high affinity [125I]NCQ298 and a slight but not significant decrease of the binding indexes of 125I]iodolisuride. Treatment with AMPT, which induced a 40% reduction in dopamine concentration, did not change [125I]NCQ298 binding indexes but slightly increased those of [3H]raclopride and [125I]iodolisuride. In conclusion, the change of dopamine concentration induces modification of radiotracer kinetics. Thus, the combined use of tracers with high and low affinities could allow us to obtain information both on receptor density and neurotransmitter release in vivo. However, as indicated by the [3H]raclopride study with AMPT, small changes in the concentration of intrasynaptic dopamine cannot be easily detected
