Baby chicks administered a fecal transplant from adult chickens are resistant to Salmonella colonization by competitive exclusion. A two-pronged approach was used to investigate the mechanism of this process. First, Salmonella response to an exclusive (Salmonella competitive exclusion product, Aviguard®) or permissive microbial community (chicken cecal contents from colonized birds containing 7.85 Log₁ₒ Salmonella genomes/gram) was assessed ex vivo using a S. typhimurium reporter strain with fluorescent YFP and CFP gene fusions to rrn and hilA operon, respectively. Second, cecal transcriptome analysis was used to assess the cecal communities’ response to Salmonella in chickens with low (≤5.85 Log₁ₒ genomes/g) or high (≥6.00 Log₁ₒ genomes/g) Salmonella colonization. The ex vivo experiment revealed a reduction in Salmonella growth and hilA expression following co-culture with the exclusive community. The exclusive community also repressed Salmonella’s SPI-1 virulence genes and LPS modification, while the anti-virulence/inflammatory gene avrA was upregulated. Salmonella transcriptome analysis revealed significant metabolic disparities in Salmonella grown with the two different communities. Propanediol utilization and vitamin B12 synthesis were central to Salmonella metabolism co-cultured with either community, and mutations in propanediol and vitamin B12 metabolism altered Salmonella growth in the exclusive community. There were significant differences in the cecal community’s stress response to Salmonella colonization. Cecal community transcripts indicated that antimicrobials were central to the type of stress response detected in the low Salmonella abundance community, suggesting antagonism involved in Salmonella exclusion. This study indicates complex community interactions that modulate Salmonella metabolism and pathogenic behavior and reduce growth through antagonism may be key to exclusion.

Background: Infection rate associated with intravenous (IV) catheter placement is emerging as an important issue in small animal veterinary medicine, mostly because of the economic costs associated with these infections. Identification of possible associated factors may provide useful information for the surveillance and prevention of such infections. Objectives: To determine the incidence of positive bacterial cultures obtained from IV catheters used in dogs hospitalized for at least 48 hours and removed because of clinical complication. To identify the bacteria involved and factors associated with bacterial colonization. Animals: One-hundred eighty-two dogs that underwent IV catheterization from January 2015 to July 2015 at the Veterinary Teaching Hospital of Alfonso X el Sabio University of Madrid were enrolled in the study. Results: The bacterial colonization rate of all IV catheters removed in response to clinical complications was 39.6%, the cumulative proportion of catheters that remained in place at 24, 48, and 72 hours after placement was 89.5, 78, and 59.4%, respectively. Multivariable Cox proportional hazards regression indicated significant associations for staff who performed catheterization (junior, P = .002; student, P = .034) and use of steroidal anti-inflammatory drugs (P = .036). The most frequently isolated bacterium was Acinetobacter spp. (21.7%). Conclusions and clinical importance: The bacterial colonization incidence related to IV catheter placement was slightly higher than the incidence described in other veterinary studies. Associated factors not previously described in veterinary medicine were found. The genome of Candidatus Xenohaliotis californiensis was assembled from shotgun metagenomic sequencing of experimentally infected white abalone. Ninety-one percent genome completeness was achieved with low contamination. Sequencing this genome provides the opportunity to track pathogen evolution over time, conduct gene expression experiments, and study dynamics between this pathogen and its phage.

The impacts of microsporidia on host individuals are frequently subtle and can be context dependent. A key example of the latter comes from a recently discovered microsporidian symbiont of Daphnia, the net impact of which was found to shift from negative to positive based on environmental context. Given this, we hypothesized low baseline virulence of the microsporidian; here, we investigated the impact of infection on hosts in controlled conditions and the absence of other stressors. We also investigated its phylogenetic position, ecology, and host range. The genetic data indicate that the symbiont is Ordospora pajunii, a newly described microsporidian parasite of Daphnia. We show that O. pajunii infection damages the gut, causing infected epithelial cells to lose microvilli and then rupture. The prevalence of this microsporidian could be high (up to 100% in the lab and 77% of adults in the field). Its overall virulence was low in most cases, but some genotypes suffered reduced survival and/or reproduction. Susceptibility and virulence were strongly host-genotype dependent. We found that North American O. pajunii were able to infect multiple Daphnia species, including the European species Daphnia longispina, as well as Ceriodaphnia spp. Given the low, often undetectable virulence of this microsporidian and potentially far-reaching consequences of infections for the host when interacting with other pathogens or food, this Daphnia-O. pajunii symbiosis emerges as a valuable system for studying the mechanisms of context-dependent shifts between mutualism and parasitism, as well as for understanding how symbionts might alter host interactions with resources.ImportanceThe net outcome of symbiosis depends on the costs and benefits to each partner. Most Campylobacter jejuni isolates carry the fucose utilization cluster (Cj0480c-Cj0489) that supports the metabolism of L-fucose and D-arabinose. In this study we quantified L-fucose and D-arabinose metabolism and metabolite production, and the impact on Caco-2 cell interaction and binding to fibronectin, using C. jejuni NCTC11168 and the closely related human isolate C. jejuni strain 286. When cultured with L-fucose and D-arabinose, both isolates showed increased survival and production of acetate, pyruvate and succinate, and the respective signature metabolites lactate and glycolic acid, in line with an overall upregulation of L-fucose cluster proteins. In vitro Caco-2 cell studies and fibronectin-binding experiments showed a trend towards higher invasion and a significantly higher fibronectin binding efficacy of C. jejuni NCTC11168 cells grown with L-fucose and D-arabinose, while no significant differences were found with C. jejuni 286. Both fibronectin binding proteins, CadF and FlpA, were detected in the two isolates, but were not significantly differentially expressed in L-fucose or D-arabinose grown cells. Comparative proteomics analysis linked the C. jejuni NCTC11168 phenotypes uniquely to the more than 135-fold upregulated protein Cj0608, putative TolC-like component MacC, which, together with the detected Cj0606 and Cj0607 proteins, forms the tripartite secretion system MacABC with putative functions in antibiotic resistance, cell envelope stress response and virulence in Gram negative pathogenic bacteria. Further studies are required to elucidate the role of the MacABC system in C. jejuni cell surface structure modulation and virulence.

Glabridin is an antimicrobial compound which can be extracted from plants, such as liquorice (Glycyrrhiza glabra) roots. Although its activity against foodborne pathogens and spoilage microorganisms has already been reported, the investigation of potential applications as a surface disinfectant is still largely unexplored. Hence, this study evaluated the disinfectant efficacy of glabridin against Listeria monocytogenes. The activity of glabridin was first tested in vitro in a nutrient-rich medium against eight strains of L. monocytogenes, including food isolates and the model strain EGDe. The tested strains showed similar susceptibility with minimal inhibitory and bactericidal concentrations of 12.5 µg/mL and 25 µg/mL, respectively. Subsequently, L. monocytogenes L6, FBR17 and EGDe were selected to assess the efficacy of glabridin against dried cells (according to the European standard EN 13697:2015 + A1:2019) and biofilm cells on stainless steel surfaces. Moreover, the impact of food residual organic matter was investigated using skim milk, cantaloupe and smoked salmon solution as soiling components. Our results showed that applying 200 µg/mL of glabridin resulted in a substantial reduction (>3 log10) of dried and biofilm cells of L. monocytogenes in standard conditions (i.e. low level of residual organic matter). Cantaloupe soiling components slightly reduced the activity of glabridin, while the efficacy of glabridin when tested with salmon and skim milk residuals was substantially affected. Comparative analysis using standardized protein contents provided evidence that the type of food matrices and type of proteins may impact the activity of glabridin as a disinfectant. Overall, this study showed low strain variability for the activity of glabridin against L. monocytogenes and shed light on the possible application of this natural antimicrobial compound as a surface disinfectant.

År 2020 drabbades världen av Covid-19. Spridningen av viruset resulterade i att många länder upprättade en lockdown. Detta påverkade sjöfartsbranschen negativt och ställde till med problem när folk blev sjuka. Detta är på grund av den unika miljön som finns ombord på fartyg. Syftet med studien var att analysera och utvärdera hur olika fartyg och rederier agerade under covid-19-pandemin med fokus på effektivitet, framgång och problem när det kommer till smittskydd. Studiens frågeställningar berörde hur rederiers existerande smittskyddsplaner såg ut och huruvida de kan förbättras, vilka lärdomar som kan dras om smittspridning ombord på fartyg och huruvida en förbättrad smittskyddsplan kan konstrueras utifrån de som redan existerar. En kvalitativ metod i form av granskning och inläsning av studier i området smittspridning i allmänhet samt studier om utbrott av covid-19 ombord på fartyg. Smittskyddsplaner samlades in genom ett mailutskick till svenska rederier. Resultatet visar att rederierna efterlevde många av WHO:s riktlinjer där de även implementerade egna tillägg till den ombordhavna smittskyddsplanen. The spread of the virus resulted with many countries establishing lockdowns. This negatively affected the shipping industry and caused problems when people got sick. This is because of the unique environment that exists on board ships. The purpose of the study was to analyze and evaluate how different ships and shipping companies acted during the COVID-19 pandemic with a focus on efficiency, success and problems when it came to controlling the spread of infections. The study's questions concerned how shipping companies' infection control plans were complied with and whether they can be improved, what lessons can be learned about the spread of infections on board ships and whether an improved infection control plan can be constructed from those that already exist. A qualitative method of reviewing and reading studies in the field of infection control in general and studies on COVID-19 outbreaks on board ships. Infection control plans were collected through a mailing to Swedish shipping companies. The results showed that the shipping companies complied with many of the WHO's guidelines where they also implemented their own additions to the on-board infection control plan. We also came to the conclusion that land-based assistance is an important component for combating the spread of infections on board.

Vaccines have significantly reduced the impact of numerous deadly viral infections. However, there is an increasing need to expedite vaccine development in light of the recurrent pandemics and epidemics. Also, identifying vaccines against certain viruses is challenging due to various factors, notably the inability to culture certain viruses in cell cultures and the wide-ranging diversity of MHC profiles in humans. Fortunately, reverse vaccinology (RV) efficiently overcomes these limitations and has simplified the identification of epitopes from antigenic proteins across the entire proteome, streamlining the vaccine development process. Furthermore, it enables the creation of multiepitope vaccines that can effectively account for the variations in MHC profiles within the human population. The RV approach offers numerous advantages in developing precise and effective vaccines against viral pathogens, including extensive proteome coverage, accurate epitope identification, crossprotection capabilities, and MHC compatibility. With the introduction of RV, there is a growing emphasis among researchers on creating multiepitope-based vaccines aiming to stimulate the host's immune responses against multiple serotypes, as opposed to single-component monovalent alternatives. Regardless of how promising the RV-based vaccine candidates may appear, they must undergo experimental validation to probe their protection efficacy for real-world applications. The time, effort, and resources allocated to the laborious epitope identification process can now be redirected toward validating vaccine candidates identified through the RV approach. Modern taxonomic classification is often based on phylogenetic analyses of a few molecular markers, although single-gene studies are still common. Here, we leverage genome-scale molecular phylogenetics (phylogenomics) of species and populations to reconstruct evolutionary relationships in a dense data set of 710 fungal genomes from the biomedically and technologically important genus Aspergillus. To do so, we generated a novel set of 1,362 high-quality molecular markers specific for Aspergillus and provided profile Hidden Markov Models for each, facilitating their use by others. Examining the resulting phylogeny helped resolve ongoing taxonomic controversies, identified new ones, and revealed extensive strain misidentification (7.59% of strains were previously misidentified), underscoring the importance of population-level sampling in species classification. These findings were corroborated using the current standard, taxonomically informative loci. These findings suggest that phylogenomics of species and populations can facilitate accurate taxonomic classifications and reconstructions of the Tree of Life.

Salmonella spp. control in pork supply chains has always been a challenging issue and insufficient control can lead to high social and economic consequences. Conventional risk management and risk management approaches and models are not sufficient to address potential food safety shocks caused by Salmonella spp., as they mainly focus on assessing measures to reduce Salmonella spp. risks instead of developing the resilience capability (e.g., flexibility to adapt to sudden changes in the risks). Our study is the first that incorporated the resilience concept to the quantitative modeling of Salmonella spp. spread in the pork supply chain. The objective of this study was to explore the resilience performance of the pork supply chain under different food safety shocks caused by Salmonella spp., and to investigate the effectiveness of interventions on reducing the impact of these shocks on the resilience performance of the chain. Scenario analysis indicated that the effectiveness of the investigated resilience strategies or interventions depended on the risk profile (i.e., default, minimum, maximum level of Salmonella spp. contamination) of the pork supply chain. For pork supply chains with minimum and default risk profiles, more attention should be paid to increasing resilience of pigs towards Salmonella spp. infection. For supply chains with maximum risk profile, the focus should be on improving the performance of the slaughterhouse, such as careful evisceration, logistic slaughtering. To conclude, enhancing resilience performance of the pork supply chain can contribute to a safe pork supply.

The emergence of infectious diseases presents a significant global health, economic, and security risk. Climate change can unexpectedly lead to the spread of pathogens, vectors, or hosts into new areas, contributing to the rise of infectious diseases. Surveillance plays a crucial role in monitoring disease trends and implementing control strategies. In this study, we document the first discovery of Heterobilharzia americana, a parasitic schistosome of mammals and its intermediate hosts Galba cubensis and Galba humilis along the banks of the Colorado River in California. We conducted multiple samplings of snails from various locations in the region with a previous history of canine schistosomiasis. Nucleotide sequencing of the multiple regions of the snails and parasites DNA revealed the coexistence of G. cubensis and G. humilis, both infected with H. americana. Phylogenetic analyses further validate the presence of H. americana in California, suggesting a wider distribution than previously reported. Our findings have implications for public health, veterinary medicine, and biodiversity conservation, contributing to developing effective control strategies to prevent the spread of this emerging infectious disease.

Acinetobacter baumannii is highly invasive, resistant to multiple drugs bacteria that are primary source of nosocomial illness in the modern hospital systems. It has been linked to a significant death rate or has been identified as a causative of meningitis, pneumonia; a condition called urine tract illnesses, or wound diseases. Many virulence variables, such as as porins, capsules, including cell wall a substance called lip digestive enzymes, biofilm formation, movement, or iron-acquisition structures, amongst other people, contribute to severity in A. baumannii illnesses. These virulence factors aid in the organism's ability to withstand harsh ecological circumstances also permit the growth of serious diseases. For tandem to the rise for A. baumannii diseases, difficult varied resistant pathways for this pathogen are effectively known, leading to the low efficacy of main antibiotics groups. A. baumannii has a distinct capacity to sustain a resistant to multiple drugs phenotype via a diverse range of antibiotic-hydrolyzing digestive enzymes, modifications to the efflux pumps, impermeability, or alterations in pharmaceutical targets, making therapy even more intricate. Understanding of A. baumannii's transmissible diseases revolves on a comprehension of the processes underlying illness, pathogenicity, or resistant development. Listeria monocytogenes is a facultative anaerobe which can cause a severe food-borne infection known as listeriosis. L. monocytogenes is capable of utilizing various nutrient sources including rhamnose, a naturally occurring deoxy sugar abundant in foods. L. monocytogenes can degrade rhamnose into lactate, acetate and 1,2-propanediol. Our previous study showed that addition of vitamin B12 stimulated anaerobic growth of L. monocytogenes on rhamnose due to the activation of bacterial microcompartments for 1,2-propanediol utilization (pdu BMC) with concomitant production of propionate and propanol. Notably, anaerobic 1,2-propanediol metabolism has been linked to virulence of enteric pathogens including Salmonella spp. and L. monocytogenes. In this study we investigated the impact of B12 and BMC activation on i) aerobic and anerobic growth of L. monocytogenes on rhamnose and ii) the level of virulence. We observed B12-induced pdu BMC activation and growth stimulation only in anaerobically grown cells. Comparative Caco-2 virulence assays showed that these pdu BMC-induced cells have significantly higher translocation efficiency compared to non-induced cells (anaerobic growth without B12; aerobic growth with or without B12), while adhesion and invasion capacity is similar for all cells. Comparative proteome analysis showed specific and overlapping responses linked to metabolic shifts, activation of stress defense proteins and virulence factors, with RNA polymerase sigma factor SigL, teichoic acid export ATP-binding protein TagH, DNA repair and protection proteins, RadA and DPS, and glutathione synthase GshAB, previously linked to activation of virulence response in L. monocytogenes, uniquely upregulated in anaerobically rhamnose grown pdu-induced cells. Our results shed light on possible effects of B12 on L. monocytogenes competitive fitness and virulence activation when utilizing rhamnose in anaerobic conditions encountered during transmission and the human intestine.

Many Chinese rivers are polluted with multiple pollutants. Therefore, this study aims to quantify the annual inputs of nutrients, macro-and microplastics, and Cryptosporidium (a pathogen) to 395 Chinese rivers from agriculture and urbanization-related sources during 2010–2050 following the storyline of Shared Socio-economic Pathway 3 and Representative Concentration Pathway 8.5. Our model estimates that multiple pollutants in Chinese rivers are projected to increase by 41–88 % between 2010 and 2050. Central, eastern, and southern Chinese sub-basins may be more polluted in the future. In half of the sub-basins, at least two types of pollutants are expected to increase by over 50 % during 2010–2050. By 2050, nutrients and plastics in rivers are projected to increase by 70 % due to urbanization. For nutrients and Cryptosporidium in rivers increases of 49–88 % are projected, due to agricultural activities. In contrast, water is expected to get cleaner in some western sub-basins by 2050 because of fewer human activities. Our insights about multiple pollutants in Chinese rivers could help prioritize water pollution reduction strategies for sub-basins.

Multiple stress resistant variants of Listeria monocytogenes with mutations in rpsU encoding ribosomal protein RpsU have previously been isolated after a single exposure to acid stress. These variants, including L. monocytogenes LO28 variant V14 with a complete deletion of the rpsU gene, showed upregulation of the general stress sigma factor Sigma B-mediated stress resistance genes and had a lower maximum specific growth rate than the LO28 WT, signifying a trade-off between stress resistance and fitness. In the current work V14 has been subjected to an experimental evolution regime, selecting for higher fitness in two parallel evolving cultures. This resulted in two evolved variants with WT-like fitness: 14EV1 and 14EV2. Comparative analysis of growth performance, acid and heat stress resistance, in combination with proteomics and RNA-sequencing, indicated that in both lines reversion to WT-like fitness also resulted in WT-like stress sensitivity, due to lack of Sigma B-activated stress defense. Notably, genotyping of 14EV1 and 14EV2 provided evidence for unique point-mutations in the ribosomal rpsB gene causing amino acid substitutions at the same position in RpsB, resulting in RpsB22Arg-His and RpsB22Arg-Ser, respectively. Combined with data obtained with constructed RpsB22Arg-His and RpsB22Arg-Ser mutants in the V14 background, we provide evidence that loss of function of RpsU resulting in the multiple stress resistant and reduced fitness phenotype, can be reversed by single point mutations in rpsB leading to arginine substitutions in RpsB at position 22 into histidine or serine, resulting in a WT-like high fitness and low stress resistance phenotype. This demonstrates the impact of genetic changes in L. monocytogenes’ ribosomes on fitness and stress resistance.

An unusual mass mortality event (MME) of harbour seals (Phoca vitulina) and harbour porpoises (Phocoena phocoena) occurred in Denmark and Sweden in June 2007. Prior to this incident, the region had experienced two MMEs in harbour seals caused by Phocine Distemper Virus (PDV) in 1988 and 2002. Although epidemiology and symptoms of the 2007 MME resembled PDV, none of the animals examined for PDV tested positive. Thus, it has been speculated that another – yet unknown – pathogen caused the June 2007 MME. To shed new light on the likely cause of death, we combine previously unpublished veterinary examinations of harbour seals with novel analyses of algal toxins and algal monitoring data. All harbour seals subject to pathological examination showed pneumonia, but were negative for PDV, influenza and coronavirus. Histological analyses revealed septicaemia in multiple animals, and six animals tested positive for Klebsiella pneumonia. Furthermore, we detected the algal Dinophysis toxin DTX-1b (1–115 ng g−1) in five seals subject to toxicology, representing the first time DTX-1b has been detected in marine vertebrates. However, no animals tested positive for both Klebsiella and toxins. Thus, while our relatively small sample size prevent firm conclusions on causative agents, we speculate that the unexplained MME may have been caused by a chance incidence of multiple pathogens acting in parallel in June 2007, including Dinophysis toxin and Klebsiella. Our study illustrates the complexity of wildlife MMEs and highlights the need for thorough sampling during and after MMEs, as well as additional research on and monitoring of DTX-1b and other algal toxins in the region.

Plastic pollution in aquatic environments poses significant concerns due to its potential to serve as a refuge for aquatic pathogens. However, the role of plastic surfaces and microbial biofilm interfaces in facilitating pathogen development remains poorly understood. In this study, a microcosm setup was employed to investigate the interactions between plastics and the microbial community and examine the differences in bacterial community composition and potential pathogen occurrences between the plastisphere-biofilm and surrounding seawater. Community composition analysis combined with SEM observations over time indicated that biofilm extracellular polymeric substance formation over 14 days had a link with the relative abundance and succession patterns of pathogen taxa. Colony clusters were observed on biofilms from day 7 and coincided with higher bacterial pathogen dominance. On day 14, pathogen abundance overall decreased with a potentially degrading biofilm. Pseudomonas and Pseudoalteromonas were the dominant potential pathogen groups observed in the microcosm. When further subjected to chemical treatment as an imposed environmental stress over time, biofilm-associated Psuedoalteromonas sharply increased in abundance after three days of exposure, but quickly diminished by 14 days in favor of genera such as Acinetobacter, Pseudomonas, and Staphylococcus. These results suggest that environmental plastisphere-biofilms can promote the early selection, enrichment, and spread of pathogenic bacteria in the aquatic environment and could be later worsened under chemical and long-term pressure. This study provided new insights into the succession of pathogens in plastisphere biofilms, contributing to the understanding of pathogen risks involved in emerging plastisphere biofilms in light of global plastic pollution.

This paper gives an account of the history of veterinary bacteriology including clinical veterinary bacteriology as well as the area of veterinary public health in Denmark from the 1880s to 2022. We describe key persons, including B. Bang, C.O. Jensen, K.A. Jensen and others who made important contributions to the development of these areas of microbiological expertise, and we discuss how challenges ranging from bovine tuberculosis to bacterial antimicrobial resistance have been met. Further, we describe progress in research on important bacterial pathogens both with regard to animal clinical aspects and zoonotic food-related aspects. Finally, we describe current issues in relation to One Health and research organization.

We have extended previously published sets of simple sequence repeat markers for Synchytrium endobioticum, selected to be polymorphic for the German-standard isolates of pathotypes P1, P2, P6, P8, and P18. These markers also complement the extensive published information on DNA polymorphisms for the mitogenomes of Synchytrium endobioticum. This extended set of 35 markers representing 73 alleles differentiated 51 isolates from Europe and North America into three large, well-separated clusters and subclusters using dendrogram analysis, principal coordinates analysis (PCoA), and population substructure analysis using STRUCTURE 2.3.4 software. This suggests a limited number of introgressions of the wart disease pathogen into current potato growing areas, followed by recombination and admixture of populations through human activities. The new markers extend the published marker sets and are useful tools for future analyses of population structure and dynamics in Synchytrium endobioticum, which are necessary to understand the biology of the interaction between the pathogen and its potato host and to develop future control strategies.

Clean water is essential for supporting human livelihoods and maintaining ecosystem health. However, our knowledge of water quality is severely impaired by a lack of quantitative information. Being under-monitored and often imperceptible to the human eye, water pollution has been branded an “invisible crisis”. Protecting and improving the quality of surface waters globally is contingent upon an improved understanding of the problem and its drivers. Process-based models are tools that can supplement our knowledge of water quality beyond what is possible using in situ measurements alone. This thesis introduces and applies the Dynamical Surface Water Quality (DynQual) model, a high-resolution global surface water quality model for simulating water temperature and concentrations of salinity (total dissolved solids; TDS), organic (biological oxygen demand; BOD) and pathogen (fecal coliform; FC) pollution. DynQual was used to provide a global assessment of past and current surface water quality. Modelled results demonstrate that surface water quality issues are globally relevant, with exceedances of key concentration thresholds for TDS, BOD and FC pollution occurring across all world regions albeit with different frequencies and magnitudes. Current year-round and multi-pollutant hotspots are located across northern India and eastern China, whereas trends towards surface water quality deterioration in the last ~40 years are most profound in Sub-Saharan Africa and southern Asia. Process-based models provide unique opportunities to quantitatively assess the impact of future change on the availability and quality of water resources. This includes exploring the effectiveness of management strategies for improving water quality. Plant pathogens secrete effector proteins to support host colonization through a wide range of molecular mechanisms, while plant immune systems evolved receptors to recognize effectors or their activities to mount immune responses to halt pathogens. Importantly, plants do not act as single organisms, but rather as holobionts that actively shape their microbiota as a determinant of health. The soil-borne fungal pathogen Verticillium dahliae was recently demonstrated to exploit the VdAve1 effector to manipulate the host microbiota to promote vascular wilt disease in the absence of the corresponding immune receptor Ve1. We identify a multiallelic V. dahliae gene displaying c. 65% sequence similarity to VdAve1, named VdAve1-like (VdAve1L), which shows extreme sequence variation, including alleles that encode dysfunctional proteins, indicative of selection pressure to overcome host recognition. We show that the orphan cell surface receptor Ve2, encoded at the Ve locus, does not recognize VdAve1L. Additionally, we demonstrate that the full-length variant VdAve1L2 possesses antimicrobial activity, like VdAve1, yet with a divergent activity spectrum, that is exploited by V. dahliae to mediate tomato colonization through the direct suppression of antagonistic Actinobacteria in the host microbiota. Our findings open up strategies for more targeted biocontrol against microbial plant pathogens.

This study investigated the aerosol particle spreading characteristic under transient state at different location released by an assailant and determine the virus reproductive number over time at different location of virus released by an assailant. The spreading during coughing process will be validated with previous literature review using Computational Fluid Dynamics (CFD) simulation study. Mesh sensitivity study was done on the model to get better accuracy results and optimum computational load for the CFD simulation. The model involved in this study is the mosque and the prayers during the congregation prayers. The particle spreading characteristics are found to be influenced by the velocity distribution and velocity vector inside the mosque which is due to fan and air conditioner. Particles less than 10 µm are mostly deposited on the wall and ceiling and particles greater than 30 µm deposited on the ground or the prayers body. The location of assailant at the center are found to cause the most infection among the prayers which is 52% of the total prayer with the virus reproductive number (Ro) of 0.83. The assailant at top right and bottom right also produced high Ro of 0.73 and 0.6 while top left produced the lowest which is 0.32. The existence of partition is found to reduce the particle spreading from the assailant at bottom left.

OBJECTIVES To identify the pathogens in the ascitic fluids of patients with spontaneous bacterial peritonitis and then to determine their sensitivity pattern to ceftriaxone. METHODOLOGY The cross-sectional study was conducted at the Medical Unit-A, Department of Medicine, Hayatabad Medical Complex, Peshawar, from November 2021 to April 2022. Before ceftriaxone treatment was started, a minimum of 10 ml of ascitic fluid was introduced into a blood culture vial. Only patients with a positive culture were registered, and their information was gathered using a proforma. For statistical analysis, SPSS version 23 was used. RESULTSA total of 96 patients were enrolled in our study. There were 62 (59.52%) male and 34 (40.48%) female patients. Based on the isolation and identification of bacteria, the most prevalent bacteria isolated was Escherichia coli in 36 (37.5%) patients, followed by Acinetobacter Spp in 13 (13.54%) patients, Streptococcus spp in 14 (14.58%), Enterococcus spp in 11 (11.45%), Staphylococcus aureus in 9 (9.39%), MRSA in 8(8.33%) and K. Pneumonia in 5(5.21%) patients. The overall sensitivity of ceftriaxone to gram-positive bacteria was observed in 12 (42.85%) isolates, whereas the overall sensitivity of ceftriaxone to gram-negative bacteria was observed in 25 (36.76%) isolates. (p=0.091) (Figure 6). CONCLUSION Our study concludes that gram-negative bacteria were more prevalent than gram-positive bacteria in ascitic fluids of patients with spontaneous bacterial peritonitis. The most common isolated pathogen was E.coli. Gram-negative was more resistant to ceftriaxone as compared to gram-positive bacteria.

The use of pesticides used in the agricultural sector for pest control has caused the deterioration of Colombian arable soils and environmental problems due to their excessive use. As a consequence, interest has arisen in the use of biological agents as an alternative method for disease control. Through qualitative research, literature review and implementation of a survey of 65 farmers in the municipality of Monterrey Casanare as an information collection technique, the objective of this article is answered, which consisted of determining the biological control techniques that exert a antagonistic against the control of phytopathogenic fungi, in agricultural crops in the municipality of Monterrey Casanare. Likewise, it is intended to design a protocol where each of the phases that must be implemented for the use of antagonistic microorganisms in the biological control of phytopathogenic organisms for the study area are observed. The results obtained reflect a high use of chemical methods by farmers that affect their well-being and the environment, likewise, with regard to the selection of strains through environmental, economic and technical criteria, it was obtained that Trichoderma harzianum Rifai is the strain that best fits the selection criteria and the study area., El uso de plaguicidas utilizados en el sector agrícola para control de plagas ha causado el deterioro de los suelos cultivables colombianos y problemas ambientales debido a su uso excesivo. Como consecuencia ha surgido el interés por la utilización de agentes biológicos, como método alternativo para el control de enfermedades. A través de una investigación cualitativa, revisión bibliográfica e implementación de una encuesta a 65 agricultores del municipio de Monterrey Casanare como técnica de recolección de información, se responde al objetivo de la presente investigación el cual consistió en determinar las técnicas de control biológico que ejercen un efecto antagónico frente al control de hongos fitopatógenos, en cultivos agrícolas del municipio de Monterrey Casanare. Así mismo se diseñó y se propone para implementación en el municipio un protocolo donde se relacionan cada una de las fases que se deben implementar para la utilización de microorganimos antagonitas en el control biológico de organismos fitopatógenos para la zona de estudio. Los resultados obtenidos reflejan una alta utilización de métodos químicos por los agricultores que repercuten en su bienestar y en el ambiente, así mismo, en lo que respecta a la selección de las cepas mediante los criterios ambientales, económicos y técnicos, se obtuvo que Trichoderma harzianum Rifai es la cepa que más se ajusta a los criterios de selección y a la zona de estudio.

Polymicrobial diseases significantly impact the health of humans and animals but remain understudied in natural systems. We recently described the Pacific Oyster Mortality Syndrome (POMS), a polymicrobial disease that impacts oyster production and is prevalent worldwide. Analysis of POMS-infected oysters on the French North Atlantic coast revealed that the disease involves co-infection with the endemic ostreid herpesvirus 1 (OsHV-1) and virulent bacterial species such asVibrio crassostreae. However, it is unknown whether consistentVibriopopulations are associated with POMS in different regions, howVibriocontribute to POMS, and how they interact with the OsHV-1 virus during pathogenesis. We resolved theVibriopopulation structure in oysters from a Mediterranean ecosystem and investigated their functions in POMS development. We find thatVibrio harveyiandVibrio rotiferianusare the predominant species found in OsHV-1-diseased oysters and show that OsHV-1 is necessary to reproduce the partition of theVibriocommunity observed in the field. By characterizing the interspecific interactions between OsHV-1,V. harveyiandV. rotiferianus, we find that onlyV. harveyisynergizes with OsHV-1. When co-infected, OsHV-1 andV. harveyibehave cooperatively by promoting mutual growth and accelerating oyster death.V. harveyishowed high virulence potential in oysters and dampened host cellular defenses, making oysters a more favorable niche for microbe colonization. Metabolism is central to maintaining health and homeostasis during all challenges a mammal may face. This is true for infections as well and the field of immunometabolism has emerged for determining how metabolism influences the immune system during disease. Lipids are a major source of carbon and energy for cells while having been found to influence immune cells in a variety of systems. Here we explore novel roles for triglycerides through the interactions of adaptive immune and adipose tissue along with fatty acid utilization in myeloid cells. This dissertation describes original work demonstrating how adaptive immune system leads to sickness-induced anorexia and fat wasting. We found that fat wasting was dependent on lipolysis and that this fat wasting had no impact on inflammation within the adipose tissue. Further, we found that the fat wasting and sickness-induced anorexia are likely linked and CD4+ T cells are necessary to mediate both processes. Surprisingly, these metabolic perturbations do not have any tangible benefit to the host and preventing them did not change parasitemia or survival. Indeed, we found that CD4+ T cells were entirely dispensable in the infection, with mice lacking these immune cells surviving as long as wild type mice with similar parasitemia. Only B cells seemed necessary for mediating survival and parasitemia in wild type mice and CD8+ T cells were detrimental to survival. When looking specifically at lipid utilization in myeloid cells rather than lipid availability, we found beta oxidation in myeloid cells is detrimental to survival. There were no differences between knockout mice and wild type mice in parasitemia, indicating that immunopathology caused by the myeloid cells likely contributes to death and knocking out beta oxidation may alleviate the immunopathology.
