Salah satu pasar yang menjanjikan dengan pertumbuhan ekonomi yang pesat adalah Mesir karena menawarkan peluang signifikan bagi perusahaan internasional. UMKM asal Indonesia yang telah hampir masuk ke Mesir adalah rendang ratuju Indonesia, rendang katuju sendiri didirikan pada tahun 2017 oleh Ade Surianto. Adapun tujuan yang dilakukan pada penelitian ini untuk menentukan variabel dan mengidentifikasi preferensi masyarakat Mesir dengan menyesuaikan selera konsumen yang relevan seperti simbol, motif, warna, dan gaya visual. Setelah didapatkan variabel dilakukan perancangan kemasan dan membuat prototype kemasan rendang sesuai dengan preferensi dan selera konsumen. Perancangan kemasan dilakukan berdasarkan keinginan konsumen. Keinginan konsumen bersifat implisit, oleh karena itu perancangan kemasan ini dilakukan dengan menggunakan Kansei Engineering. Metode Kansei Engineering dapat mengetahui keinginan konsumen berdasarkan aspek psikologi, yaitu perasaan dan emosi konsumen, sehingga akan mempermudah dalam menerjemahkan keinginan konsumen menjadi desain produk. Hasil rancangan kemasan didapatkan dari hasil pengolahan data, dan bagaimana menghubungkan kata-kata Kansei dengan properti produk serta bagaimana hasil pemilihan berbagai jenis model kemasan yang sesuai dengan kriteria kata-kata Kansei. Alternatif rancangan kemasan yang diberikan ada 3 rancangan. yakni rancangan alternatif 1 (satu) berbentuk standing pouch, rancangan alternatif 2 (dua) berbentuk segitiga dan rancangan alternatif 3 (tiga) berbentuk persegi. Introduction: HIV drug resistance poses a challenge to the United Nation's goal of ending the HIV/AIDS epidemic. The integrase strand transfer inhibitor (InSTI) dolutegravir, which has a higher resistance barrier, was endorsed by the WHO in 2019 for first-line, second-line and third-line antiretroviral therapy (ART). This multiplicity of roles of dolutegravir in ART may facilitate the emergence of dolutegravir resistance. Methods and analysis: Nested within the International epidemiology Databases to Evaluate AIDS (IeDEA), DTG RESIST is a multicentre study of adults and adolescents living with HIV in sub-Saharan Africa, Asia, and South and Central America who experienced virological failure on dolutegravir-based ART. At the time of virological failure, whole blood will be collected and processed to prepare plasma or dried blood spots. Laboratories in Durban, Mexico City and Bangkok will perform genotyping. Analyses will focus on (1) individuals who experienced virological failure on dolutegravir and (2) those who started or switched to such a regimen and were at risk of virological failure. For population (1), the outcome will be any InSTI drug resistance mutations, and for population (2) virological failure is defined as a viral load >1000 copies/mL. Phenotypic testing will focus on non-B subtype viruses with major InSTI resistance mutations. Bayesian evolutionary models will explore and predict treatment failure genotypes. The study will have intermediate statistical power to detect differences in resistance mutation prevalence between major HIV-1 subtypes; ample power to identify risk factors for virological failure and limited power for analysing factors associated with individual InSTI drug resistance mutations. Soil contamination is one of the main threats to ecosystem health and sustainability. Yet little is known about the extent to which soil contaminants differ between urban greenspaces and natural ecosystems. Here we show that urban greenspaces and adjacent natural areas (i.e., natural/semi-natural ecosystems) shared similar levels of multiple soil contaminants (metal(loid)s, pesticides, microplastics, and antibiotic resistance genes) across the globe. We reveal that human influence explained many forms of soil contamination worldwide. Socio-economic factors were integral to explaining the occurrence of soil contaminants worldwide. We further show that increased levels of multiple soil contaminants were linked with changes in microbial traits including genes associated with environmental stress resistance, nutrient cycling, and pathogenesis. Taken together, our work demonstrates that human-driven soil contamination in nearby natural areas mirrors that in urban greenspaces globally, and highlights that soil contaminants have the potential to cause dire consequences for ecosystem sustainability and human wellbeing.

Increasing the number of environmental stressors could decrease ecosystem functioning in soils. Yet this relationship has not been globally assessed outside laboratory experiments. Here, using two independent global standardized field surveys, and a range of natural and human factors, we test the relationship between the number of environmental stressors exceeding different critical thresholds and the maintenance of multiple ecosystem services across biomes. Our analysis shows that having multiple stressors, from medium levels (>50%), negatively and significantly correlates with impacts on ecosystem services and that having multiple stressors crossing a high-level critical threshold (over 75% of maximum observed levels) reduces soil biodiversity and functioning globally. The number of environmental stressors exceeding the >75% threshold was consistently seen as an important predictor of multiple ecosystem services, therefore improving prediction of ecosystem functioning. Our findings highlight the need to reduce the dimensionality of the human footprint on ecosystems to conserve biodiversity and function.

Synthetic pesticides are widely applied in modern agriculture, where they are used against diseases, pests, and weeds to secure crop yield and quality. However, their intensive application has led to widespread contamination of the environment, including soils. Due to their inherent toxicity, they might pose a risk to soil health by causing harm to non-target organisms and disrupting ecosystem services in both agricultural and other exposed soils. Following the Swiss National Action Plan on the reduction of pesticide risks, Agroscope has conducted several soil monitoring studies that are briefly presented here. All of them resort to different multi-residue trace analytical approaches to simultaneously quantify up to about 150 modern pesticides by either accelerated solvent, or Quick, Easy, Cheap, Efficient, Rugged, Safe (QuEChERS) extraction, followed by separation and detection with liquid chromatography-triple quadrupole mass spectrometry. While partly still in progress, our investigations led to the following major findings this far: Multiple pesticides are commonly present in soils, with individual concentrations in agricultural soils often reaching up to a few tens of µg/kg. Pesticide occurrence and concentrations in agricultural soils primarily depend on land use, land use history and cultivated crops. Pesticides can prevail much longer than predicted by their half-lives, and were found in soils even decades after conversion from conventional to organic farming. Corresponding residual fractions can be in the order of a few percent of the originally applied amounts. We further found negative associations of pesticide residues with the abundance of beneficial soil life, underpinning their potential risk to the fertility of agricultural soils. Traces of pesticides are also detected in soils to which they were never applied, indicating contamination, e.g., via spray drift or atmospheric deposition. Biowaste-derived substances isolated from green compost (BBS-GC) are environmentally friendly reactants similar to humic substances, which contain multiple functionalities, that are suitable for adsorbing different kinds of pollutants in wastewater. Herein, sodium alginate (derived from brown algae) cross-linked with both Ca2+ ions and BBS-GC in the form of hydrogels and dried films are proposed as green, easy-to-form, and handleable materials for tertiary water treatments. The results show that both hydrogels and films are mechanically stable and can effectively remove differently charged dyes through an adsorption mechanism that can be described by the Freundlich model. BBS-GC-containing gels always performed better than samples prepared without BBS-GC, revealing that such unconventional materials can integrate waste valorization and water decontamination, potentially providing social and environmental benefits.

Few studies have analyzed the presence of pesticides in soils of untreated areas that serve as ecological refuges (e.g. meadows). A lack of knowledge is also observed at the level of the processes of (unintentional) contamination of these untreated surfaces. This study, conducted in Switzerland, analyzed the presence and abundance of 46 pesticides in soils of extensively managed meadows, as well as in vegetable fields managed in organic and conventional agriculture (60 plots in total). Pesticides were found in all soils, including in extensive meadows, demonstrating a background and generalized contamination of soils by pesticides. The results obtained also suggest that fields initially managed in conventional agriculture, then converted to organic agriculture, reach pesticide levels as low as those of meadows only twenty years later, which illustrates the persistence of pesticides in soils. The analysis of the different pesticide compounds found in meadows and organic fields made it possible to differentiate the presence of pesticide residues due to long-term persistent molecules from presences due to diffuse contamination processes (spray drift during conventional field treatments and atmospheric deposition).

Therefore, new data analysis approaches are required to help uncover the complexity of such signals. Our Laser Ablation Ionization Mass Spectrometer (LIMS) is a compact time-of-flight mass spectrometer intended to investigate the elemental, isotope, and molecular composition of a wide range of solid samples, including e.g., low bulk density organic remains in microfossils. Here, we present an overview of the instrument and collected chemical spectrometric data at the micrometer level from a Precambrian chert sample (1.88 Ga Gunflint Formation, Ontario, Canada), which is considered to be a martian analogue. Data were collected from two distinct zones-a silicified host area and a carbon-bearing microfossil assemblage zone. We performed these measurements using an ultrafast pulsed laser system (pulse width of similar to 180 fs) with multiple wavelengths (infrared [IR]-775 nm, ultraviolet [UV]-387 nm, UV-258 nm) and using a pulsed high voltage on the mass spectrometer to reveal small organic signals. We investigated (1) the chemical composition of the sample and (2) the different laser wavelengths' performance to provide chemical depth profiles in silicified media. Our key findings are as follows: (1) microfossils from the Gunflint chert reveal a distinct chemical composition compared with the host mineralogy (we report the identification of 24 elements in the microfossils); (2) detection of the pristine composition of microfossils and co-occurring fine chemistry (rare earth elements) requires utilization of the depth profiling measurement protocol; and (3) our results show that, for analysis of heterogeneous material from siliciclastic deposits, siliceous sinters, and cherts, the most suitable wavelength for laser abla

Exploring how life is distributed in the universe is an extraordinary interdisciplinary challenge, but increasingly subject to testable hypotheses. Biology has emerged and flourished on at least one planet, and that renders the search for life elsewhere a scientific question. We cannot hope to travel to exoplanets in pursuit of other life even if we identify convincing biosignatures, but we do have direct access to planets and moons in our solar system. It is therefore a matter of deep astrobiological interest to study their histories and environments, whether or not they harbor life, and better understand the constraints that delimit the emergence and persistence of biology in any context. In this perspective, we argue that targeted chemistry- and biology-inspired experiments are informative to the development of instruments for space missions, and essential for interpreting the data they generate. This approach is especially useful for studying Venus because if it were an exoplanet we would categorize it as Earth-like based on its mass and orbital distance, but its atmosphere and surface are decidedly not Earth-like. Here, we present a general justification for exploring the solar system from an astrobiological perspective, even destinations that may not harbor life. We introduce the extreme environments of Venus, and argue that rigorous and observation-driven experiments can guide instrument development for imminent missions to the Venusian clouds. We highlight several specific examples, including the study of organic chemistry under extreme conditions, and harnessing the fluorescent properties of molecules to make a variety of otherwise challenging measurements.

Pesticides are widely applied in agriculture to combat disease, pests, and weeds, leading to long-lasting contamination of agricultural soils with pesticide residues. While classical risk assessment experiments have repeatedly addressed immediate pesticide effects, we employ an ecological approach to investigate how pesticide residues persisting in soils influence the soil microbiome under realistic agricultural conditions. We assessed a wide range of soil characteristics, including the occurrence of 48 widely-used pesticides in 60 fields under conventional, no-tillage and organic management. We then tested which factors best explain soil microbiome traits. Environmental factors, including climate, geography, and soil characteristics, were the soil microbiome's leading drivers. Remarkably, of all management factors, pesticide residues showed the strongest associations with soil microbiome traits, which were even more pronounced than the effects of cropping systems. Pesticide residues were almost exclusively positively associated with the relative abundance of 113 bacterial and 130 fungal taxa, many of them being assigned to taxa of known pesticide degraders. While fungal diversity and abundance were primarily positively associated with pesticide residues, bacterial diversity and abundance of the gene nifH - essential for biological nitrogen fixation - were negatively linked to the concentration of individual pesticide residues. Our results suggest that pesticide residues alter the soil microbiome, with potential long-term implications for the functioning of agricultural soils.

Extraction of useful information from unstructured, large and complex mass spectrometric signals is a challenge in many application fields of mass spectrometry. Therefore, new data analysis approaches are required to help uncover the complexity of such signals. In this contribution, we examined the chemical composition of the 1.88 Ga Gunflint chert using the newly developed high mass resolution laser ionization mass spectrometer (fs-LIMS-GT). We report results on the following: 1) mass-spectrometric multi-element imaging of the Gunflint chert sample; and 2) identification of multiple chemical entities from spatial mass spectrometric data utilizing nonlinear dimensionality reduction and spectral similarity networks. The analysis of 40 ' 000 mass spectra reveals the presence of chemical heterogeneity (seven minor compounds) and two large clusters of spectra registered from the organic material and inorganic host mineral. Our results show the utility of fs-LIMS imaging in combination with manifold learning methods in studying chemically diverse samples.

To date, few studies assessed the occurrence of pesticides in organically managed agricultural soils, and it is unresolved whether these pesticide residues affect soil life. We screened 100 fields under organic and conventional management with an analytical method containing 46 pesticides (16 herbicides, 8 herbicide transformation products, 17 fungicides, seven insecticides). Pesticides were found in all sites, including 40 organic fields. The number of pesticide residues was two times and the concentration nine times higher in conventional compared to organic fields. Pesticide number and concentrations significantly decreased with the duration of organic management. Even after 20 years of organic agriculture, up to 16 different pesticide residues were present. Microbial biomass and specifically the abundance of arbuscular mycorrhizal fungi, a widespread group of beneficial plant symbionts, were significantly negatively linked to the amount of pesticide residues in soil. This indicates that pesticide residues, in addition to abiotic factors such as pH, are a key factor determining microbial soil life in agroecosystems. This comprehensive study demonstrates that pesticides are a hidden reality in agricultural soils, and our results suggest that they have harmful effects on beneficial soil life.

Background: Extreme terrestrial, analogue environments are widely used models to study the limits of life and to infer habitability of extraterrestrial settings. In contrast to Earth’s ecosystems, potential extraterrestrial biotopes are usually characterized by a lack of oxygen. Methods: In the MASE project (Mars Analogues for Space Exploration), we selected representative anoxic analogue environments (permafrost, salt-mine, acidic lake and river, sulfur springs) for the comprehensive analysis of their microbial communities. We assessed the microbiome profile of intact cells by propidium monoazide-based amplicon and shotgun metagenome sequencing, supplemented with an extensive cultivation effort. Results: The information retrieved from microbiome analyses on the intact microbial community thriving in the MASE sites, together with the isolation of 31 model microorganisms and successful binning of 15 high-quality genomes allowed us to observe principle pathways, which pinpoint specific microbial functions in the MASE sites compared to moderate environments. The microorganisms were characterized by an impressive machinery to withstand physical and chemical pressures. All levels of our analyses revealed the strong and omnipresent dependency of the microbial communities on complex organic matter. Moreover, we identified an extremotolerant cosmopolitan group of 34 poly-extremophiles thriving in all sites. Conclusions: Our results reveal the presence of a core microbiome and microbial taxonomic similarities between saline and acidic anoxic environments. Our work further emphasizes the importance of the environmental, terrestrial parameters for the functionality of a microbial community, but also reveals a high proportion of living microorganisms in extreme environments with a high adaptation potential within habitability borders.

Analysis of LIMS spectra acquired from the individual microfossils reveals complex mineralization, which can reflect the metabolic diversity of the Gunflint microbiome. We present chemical depth profiling studies on mineralogical inclusions embedded in amygdale calcium carbonate by our Laser Ablation Ionisation Mass Spectrometer designed for in situ space research. An IR femtosecond laser ablation is employed to generate ions that are recorded by a miniature time-of-flight mass spectrometer. The mass spectra were measured at several locations on the sample surface and yield chemical depth profiles along the depth length of about 30 mu m. The presence of oxides and sulphides within inclusion material allows us to derive elemental abundance calibration factors (relative sensitivity coefficients, RSCs) for major and minor elements. These are obtained from the atomic intensity correlations performed on the depth profiling data. With the RSCs corrections the quantitative analysis of more complex mineralogical phases within the inclusion is conducted by correlating atomic abundance fractions in ternary diagrams, typically used in geology. The spatial resolution of the depth profiles was sufficient to study chemically distinct micrometre-sized objects, such as mineralogical grains and thin layers of minerals including micrometre-sized filamentous structures. The method presented here is well-suited for the quantitative chemical analyses of highly heterogeneous materials where the ablation condition can vary locally with the material composition making the application of standard reference materials less accurate. The presented method is developed to distinguish between abiotic and biological material while searching for micrometre-sized extinct or extent life forms on the surfaces of Solar System bodies.

Pesticides are applied in large quantities to agroecosystems worldwide. To date, few studies assessed the occurrence of pesticides in organically managed agricultural soils, and it is unresolved whether these pesticide residues affect soil life. We screened 100 fields under organic and conventional management with an analytical method containing 46 pesticides (16 herbicides, 8 herbicide transformation products, 17 fungicides, seven insecticides). Pesticides were found in all sites, including 40 organic fields. The number of pesticide residues was two times and the concentration nine times higher in conventional compared to organic fields. Pesticide number and concentrations significantly decreased with the duration of organic management. Even after 20 years of organic agriculture, up to 16 different pesticide residues were present. Microbial biomass and specifically the abundance of arbuscular mycorrhizal fungi, a widespread group of beneficial plant symbionts, were significantly negatively linked to the amount of pesticide residues in soil. This indicates that pesticide residues, in addition to abiotic factors such as pH, are a key factor determining microbial soil life in agroecosystems. This comprehensive study demonstrates that pesticides are a hidden reality in agricultural soils, and our results suggest that they have harmful effects on beneficial soil life.
