Nitrous oxide (N2O) is a potent greenhouse gas and a strong ozone-depleting substance. Ocean is an important natural source of atmospheric N2O, which has attracted increasing attention recently as to understand the biogeochemical cycles of nitrogen in marine systems. Nitrification is one of the major N2O production pathways during which N2O is produced as a side product and hydroxylamine (NH2OH) is produced as an obligate intermediate. However, few studies have reported the NH2OH variability in natural seawater and its relationship between N2O remains ambiguous. In summer 2022, spatial distribution and influencing factors of N2O and NH2OH in the northern South China Sea (SCS) were investigated. Dissolved N2O varied from 5.79 to 34.80 nmol L-1 in water column and generally increased with water depth above 800 m. N2O was correlated with dissolved oxygen and nitrate, inferring that N2O was mainly produced via nitrification. NH2OH varied from below detection limit to 3.91 nmol L-1. The adoption of multiple standard additions can largely reduce the bias in NH2OH measurement. Structural equation modeling showed that NH2OH has a potential effect favoring excessive N2O. Surface N2O saturation ranged between 102% and 278%, indicating that the northern SCS was a net source of atmospheric N2O in summer. Hydroxylamine (NH2OH) is an important intermediate product and a precursor of N2O in nitrification, but debates persist regarding NH2OH variability and N2O production. We investigated the distribution, variation and regulatory mechanism of dissolved N2O and NH2OH in the northern SCS, analyzed their potential relationship during nitrification, and provided a new perspective for the greenhouse gas research in the future. Yet, recent trends and drivers of lake volume change remain largely unknown globally. Here, we analyze the 1972 largest global lakes using three decades of satellite observations, climate data, and hydrologic models, finding statistically significant storage declines for 53% of these water bodies over the period 1992-2020. The net volume loss in natural lakes is largely attributable to climate warming, increasing evaporative demand, and human water consumption, whereas sedimentation dominates storage losses in reservoirs. We estimate that roughly one-quarter of the world's population resides in a basin of a drying lake, underscoring the necessity of incorporating climate change and sedimentation impacts into sustainable water resources management.

As China’s population ages rapidly, the health risks associated with a changing climate are becoming more threatening. The 2022 China report of the Lancet Countdown, led by Tsinghua University with the contributions of 73 experts from 23 leading global institutions, tracks progress in climate change and health in China through 27 indicators across five domains: (1) Climate change impacts, exposure, and vulnerability; (2) adaptation, planning, and resilience for health; (3) mitigation actions and health co-benefits; (4) economics and finance; and (5) public and political engagement. This report is the third China Lancet Countdown report, paying particular attention to the impacts on the elderly and highlighting the urgency of taking action. We selected the most urgent and relevant indicators to complete a policy brief that provides a better understanding of recent progress on climate change and health in China. We found heat-related health impacts increased from 2020 to 2021, increasing heat-related mortality, reducing labour capacity, and undermining the capacity to partake in physical activity due to rising temperature. In addition, exposure to wildfire, extreme drought, and extreme rainfall also increased in different regions across China. In 2021, compared with the 1986–2005 average, people in China had an average of 7.85 more heatwave days (which led to an extra 13185 heatwave-related deaths), and a loss of 0.67 more hours of safe outdoor physical exercise per day. The rising temperature also caused the annual average exposure to wildfire to increase by 60.0% between 2017–2021 compared with the 2001–2005 average. Normalized Difference Vegetation Index (NDVI) is an effective proxy for vegetation activity and terrestrial productivity, and the widely used global NDVI products include the third generation Global Inventory Modelling and Mapping Studies Advanced Very High-Resolution Radiometer NDVI (NDVI3g), Satellite Pour l’Observation de la Terre VEGETATION NDVI (NDVIsp) and Terra Moderate Resolution Imaging Spectroradiometer NDVI (NDVItr). Here, for the first time, we investigated the differences between the three NDVIs in response and sensitivity to water stress represented by root zone soil moisture, vapor pressure deficit, self-calibrating Palmer Drought Severity Index and Standardized Precipitation Evapotranspiration Index across climate zones, humidity gradients, vegetation types and tree cover gradients. Over the temperate and arid climates region, the three NDVIs show higher consistent response fractions (CRFs) to water stress in the water-limited regions, with their sensitivity being in the order of NDVI3g < NDVIsp < NDVItr. The three NDVIs tend to exhibit inconsistent responses to water stress in the areas with higher humidity and tree cover, and their sensitivities to water stress fall as humidity and tree cover increase. In addition, the three NDVIs display higher CRFs and sensitivities to water stress over the non-forested areas. Our results also show that, in the cold climates region, the three NDVIs respond in the opposite way to water stress as that in water-limited areas, and NDVI3g has higher sensitivity to water stress than NDVIsp and NDVItr. This study highlights the differences between three NDVIs products for qualifying and quantifying their response and sensitivity to water stress.

Prefabricated construction has been well recognized for its benefits, including accelerated construction cycle time and improved sustainability. However, low efficiency in the production of precast concrete components due to component and production complexity has not been sufficiently addressed in previous research. This paper explores the complexity of precast concrete components by establishing complexity indices and evaluating their influence on production efficiency. First, referring to research on product complexity in the machinery manufacturing industry, we define the complexity of precast concrete components in the construction industry. Then, based on a literature review, field studies, and expert interviews, we systematically construct the complexity indices of precast concrete components using the three-stage coding method, Grounded Theory. The overall complexity index system for precast concrete components constructed comprises 16 constituent complexity indices in three dimensions (i.e., structural complexity of the component, production complexity, and management complexity). The relationship between complexity indices and production efficiency is then explored using structural equation model analysis based on the data collected through a questionnaire survey. The results reveal that complexity indices have a significant impact on the production efficiency for precast concrete components, where the number of embedded parts, waiting time in production, operating proficiency of workers, and degree of automation of the production line are found to be the most influential complexity indices. This study provides a foundation upon which production managers improve production efficiency for precast concrete components based on an analysis of their product complexity.

Despite their societal and environmental significance, spatial inventories of dams and reservoirs, even for the large ones, are insufficient. A dilemma of the existing georeferenced dam datasets is the polarized focus on either dam quantity and spatial coverage (e.g., GlObal geOreferenced Database of Dams, GOODD) or detailed attributes for a limited dam quantity or region (e.g., GRanD (Global Reservoir and Dam database) and national inventories). One of the most comprehensive datasets, the World Register of Dams (WRD), maintained by the International Commission on Large Dams (ICOLD), documents nearly 60g000 dams with an extensive suite of attributes. Unfortunately, the WRD records provide no geographic coordinates, limiting the benefits of their attributes for spatially explicit applications. To bridge the gap between attribute accessibility and spatial explicitness, we introduce the Georeferenced global Dams And Reservoirs (GeoDAR) dataset, created by utilizing the Google Maps geocoding application programming interface (API) and multi-source inventories. We release GeoDAR in two successive versions (v1.0 and v1.1) at 10.5281/zenodo.6163413 (Wang et al., 2022). GeoDAR v1.0 holds 22g560 dam points georeferenced from the WRD, whereas v1.1 consists of (a) 24g783 dam points after a harmonization between GeoDAR v1.0 and GRanD v1.3 and (b) 21g515 reservoir polygons retrieved from high-resolution water masks based on a one-to-one relationship between dams and reservoirs. Due to geocoding challenges, GeoDAR spatially resolved g1/4g40g% of the records in the WRD, which, however, comprise over 90g% of the total reservoir area, catchment area, and reservoir storage capacity. Pineoblastomas (PBs) are rare, aggressive pediatric brain tumors of the pineal gland with modest overall survival despite intensive therapy. We sought to define the clinical and molecular spectra of PB to inform new treatment approaches for this orphan cancer. Tumor, blood, and clinical data from 91 patients with PB or supratentorial primitive neuroectodermal tumor (sPNETs/CNS-PNETs), and 2 pineal parenchymal tumors of intermediate differentiation (PPTIDs) were collected from 29 centres in the Rare Brain Tumor Consortium. We used global DNA methylation profiling to define a core group of PB from 72/93 cases, which were delineated into five molecular sub-groups. Copy number, whole exome and targeted sequencing, and miRNA expression analyses were used to evaluate the clinico-pathologic significance of each sub-group. Tumors designated as group 1 and 2 almost exclusively exhibited deleterious homozygous loss-of-function alterations in miRNA biogenesis genes (DICER1, DROSHA, and DGCR8) in 62 and 100% of group 1 and 2 tumors, respectively. Recurrent alterations of the oncogenic MYC-miR-17/92-RB1 pathway were observed in the RB and MYC sub-group, respectively, characterized by RB1 loss with gain of miR-17/92, and recurrent gain or amplification of MYC. PB sub-groups exhibited distinct clinical features: group 1-3 arose in older children (median ages 5.2-14.0 years) and had intermediate to excellent survival (5-year OS of 68.0-100%), while Group RB and MYC PB patients were much younger (median age 1.3-1.4 years) with dismal survival (5-year OS 37.5% and 28.6%, respectively). We identified age < 3 years at diagnosis, metastatic disease, omission of upfront radiation, and chr 16q loss as significant negative prognostic factors across all PBs. Our findings demonstrate that PB exhibits substantial molecular heterogeneity with sub-group-associated clinical phenotypes and survival. BACKGROUND: Cardiac differentiation from human pluripotent stem cells provides a unique opportunity to study human heart development in vitro and offers a potential cell source for cardiac regeneration. Compared to the large body of studies investigating cardiac maturation and cardiomyocyte subtype-specific induction, molecular events underlying cardiac lineage commitment from pluripotent stem cells at early stage remain poorly characterized. RESULTS: In order to uncover key molecular events and regulators controlling cardiac lineage commitment from a pluripotent state during differentiation, we performed single-cell RNA-Seq sequencing and obtained high-quality data for 6879 cells collected from 6 stages during cardiac differentiation from human embryonic stem cells and identified multiple cell subpopulations with distinct molecular features. Through constructing developmental trajectory of cardiac differentiation and putative ligand-receptor interactions, we revealed crosstalk between cardiac progenitor cells and endoderm cells, which could potentially provide a cellular microenvironment supporting cardiac lineage commitment at day 5. In addition, computational analyses of single-cell RNA-Seq data unveiled ETS1 (ETS Proto-Oncogene 1) activation as an important downstream event induced by crosstalk between cardiac progenitor cells and endoderm cells. Consistent with the findings from single-cell analysis, chromatin immunoprecipitation followed by high-throughput sequencing (ChIP-Seq) against ETS1 revealed genomic occupancy of ETS1 at cardiac structural genes at day 9 and day 14, whereas ETS1 depletion dramatically compromised cardiac differentiation. CONCLUSION: Together, our study not only characterized the molecular features of different cell types and identified ETS1 as a crucial factor induced by cell-cell crosstalk contributing to cardiac lineage commitment from a pluripotent state, but may also have important implications for understanding human heart developm

An enhancement-mode GaN power switch with monolithically integrated gate driver is demonstrated on a 650-V GaN-on-Si power device platform. The integrated GaN-based gate driver features advanced designs such as bootstrapped gate-charging current source that enables high current driving capability during the entire turn-on process and rail-to-rail output. The GaN power transistor with integrated gate driver was characterized up to 300 V/15 A switching operations using a double pulse tester, and exhibits suppressed gate ringing and fast switching speed. The peak drain voltage slew rate d V/dt is above 125 V/ns during turn-on, and 336 V/ns during turn-off.

Endorheic (hydrologically landlocked) basins spatially concur with arid/semi-arid climates. Given limited precipitation but high potential evaporation, their water storage is vulnerable to subtle flux perturbations, which are exacerbated by global warming and human activities. Increasing regional evidence suggests a probably recent net decline in endorheic water storage, but this remains unquantified at a global scale. By integrating satellite observations and hydrological modelling, we reveal that during 2002–2016 the global endorheic system experienced a widespread water loss of about 106.3 Gt yr−1, attributed to comparable losses in surface water, soil moisture and groundwater. This decadal decline, disparate from water storage fluctuations in exorheic basins, appears less sensitive to El Niño–Southern Oscillation-driven climate variability, which implies a possible response to longer-term climate conditions and human water management. In the mass-conserved hydrosphere, such an endorheic water loss not only exacerbates local water stress, but also imposes excess water on exorheic basins, leading to a potential sea level rise that matches the contribution of nearly half of the land glacier retreat (excluding Greenland and Antarctica). Given these dual ramifications, we suggest the necessity for long-term monitoring of water storage variation in the global endorheic system and the inclusion of its net contribution to future sea level budgeting.

In the mass-conserved hydrosphere, such an endorheic water loss not only exacerbates local water stress, but also imposes excess water on exorheic basins, leading to a potential sea level rise that matches the contribution of nearly half of the land glacier retreat (excluding Greenland and Antarctica). Given these dual ramifications, we suggest the necessity for long-term monitoring of water storage variation in the global endorheic system and the inclusion of its net contribution to future sea level budgeting.

La population dans le bassin versant du Bandama blanc au nord de la Côte d’Ivoire est à majorité rurale et vit essentiellement de l’agriculture. Ce travail analyse les tendances des variables hydro-climatiques du bassin et leurs niveaux de significativité. Les méthodes utilisées sont l’indice standardisé et le test statistique non paramétrique de Mann-Kendall. Les résultats révèlent une tendance à la baisse, bien que peu significative, de la pluviométrie annuelle. Alors que, les précipitations de la saison pluvieuse et du mois le plus pluvieux ont fortement baissé. On constate une tendance à la hausse significative de la température et de l’évapotranspiration potentielle, alors que les écoulements sont en forte baisse. Ces changements dans les séries hydro-climatiques se sont produits dans les années 1970 et 1980. Cette étude se différencie de celles déjà réalisées sur la variabilité climatique au Nord de la Côte d’Ivoire, par les tests statistiques utilisés et la détermination du niveau de significativité des tendances dans les variables hydro-climatiques.

GaN-based digital integrated circuits (ICs) are realized on a 6-inch GaN-on-Si power high-electron-mobility transistor (HEMT) platform by monolithic integration of enhancement/depletion-mode HEMTs using a 0.5-mu m gate technology. A direct-coupled FET logic inverter and a 101-stage ring oscillator are fabricated and characterized. The inverter exhibits a large input voltage swing, wide noise margin, and high temperature stability, while the ring oscillator features a small propagation delay of 0.1 ns/stage under a supply voltage of 4 V. These digital ICs can operate properly up to at least 200 degrees C and show great potential for GaN smart power IC applications.

BACKGROUND: A key question when analyzing high throughput data is whether the information provided by the measured biological entities (gene, metabolite expression for example) is related to the experimental conditions, or, rather, to some interfering signals, such as experimental bias or artefacts. Visualization tools are therefore useful to better understand the underlying structure of the data in a 'blind' (unsupervised) way. A well-established technique to do so is Principal Component Analysis (PCA). PCA is particularly powerful if the biological question is related to the highest variance. Independent Component Analysis (ICA) has been proposed as an alternative to PCA as it optimizes an independence condition to give more meaningful components. However, neither PCA nor ICA can overcome both the high dimensionality and noisy characteristics of biological data. RESULTS: We propose Independent Principal Component Analysis (IPCA) that combines the advantages of both PCA and ICA. It uses ICA as a denoising process of the loading vectors produced by PCA to better highlight the important biological entities and reveal insightful patterns in the data. The result is a better clustering of the biological samples on graphical representations. In addition, a sparse version is proposed that performs an internal variable selection to identify biologically relevant features (sIPCA). CONCLUSIONS: On simulation studies and real data sets, we showed that IPCA offers a better visualization of the data than ICA and with a smaller number of components than PCA. Furthermore, a preliminary investigation of the list of genes selected with sIPCA demonstrate that the approach is well able to highlight relevant genes in the data with respect to the biological experiment.IPCA and sIPCA are both implemented in the R package mixomics dedicated to the analysis and exploration of high dimensional biological data sets, and on mixomics' web-interface.

The exchange between the Persian (Arabian) Gulf and the Indian Ocean is investigated using hydrographic and moored acoustic Doppler current profiler data from the Straits of Hormuz during the period December 1996 to March 1998. The moored time series records show a relatively steady deep outflow through the strait from 40 m to the bottom with a mean speed of approximately 20 cm/s. A variable flow is found in the upper layer with frequent reversals on timescales of several days to weeks. The annual mean flow in the near-surface layer is found to be northeastward (out of the Persian Gulf) in the southern part of the strait, suggesting a mean horizontal exchange with the Indian Ocean that is superimposed on the vertical overturning exchange driven by evaporation over the gulf. The salinity of the deep outflow varies from 39.3 to 40.8 psu with highest outflow salinities occurring in the winter months (December–March). The annual mean deep outflow through the strait is estimated to be 0.15 ± 0.03 Sv. Calculation of the associated heat and freshwater fluxes through the strait yields estimates for the annual heat loss over the surface of the gulf of −7 ± 4 W/m2 and an annual water loss (E-P-R) of 1.68 ± 0.39 m/yr. These values are shown to be in relatively good agreement with climatological surface fluxes derived from the Southampton Oceanography Centre global flux climatology after known regional biases in the radiative budget are taken into account.
