OBJECTIVE: This study aimed to clarify the mechanism by which Krüppel-like factor 12 (KLF12) affects progesterone sensitivity in endometrial cancer (EC) through the progesterone receptor PGR signaling pathway. METHODS: The relationship of KLF12 with PGR in EC patients was examined by immunohistochemistry, and the expression of KLF12 and PGR in EC cell lines was detected by real-time PCR and western blotting. Cell proliferation assay, plate clone formation, cell apoptosis assay, and cell cycle analysis were conducted to determine the impact of KLF12 intervention on progesterone therapy. CUT&Tag analysis and the dual-luciferase reporter experiment were used to determine the underlying regulatory effect of KLF12 on the PGR DNA sequence. A subcutaneous xenograft nude mouse model was established to validate the in vivo effect of KLF12 on progesterone sensitivity via PGR expression modulation. RESULTS: KLF12 demonstrated decreased progesterone sensitivity and a negative correlation with PGR expression in EC tissues. Progesterone sensitivity was increased by KLF12 deficiency through PGR overexpression, a result that could be significantly reversed by PGR downregulation. PGR was identified as a target gene of KLF12, which could directly bind to the PGR promotor region and inhibit its expression. CONCLUSION: This study is the first to investigate the effect of KLF12 expression on EC cell resistance to progesterone. Our results offer important mechanistic insight into the direct regulation of the PGR promoter region, demonstrating that KLF12 expression strongly suppressed the PGR signaling pathway and, as a result, reduced progesterone sensitivity in EC patients.

Our previous studies have shown its protective effects against cerebral ischemia-reperfusion injury (IRI), but the underlying molecular mechanisms remain unclear.Purpose: The study delves into acacetin’s mechanism in mitigating cerebral IRI, with a focus on transcriptomic insights.Methods: We established the oxygen-glucose deprivation/re-oxygenation (OGD/R) model in BV2 microglia, treating them with 10μM acacetin. High-throughput RNA sequencing (RNA-seq) underpinned the analysis of differentially expressed genes (DEGs) and long non-coding RNAs (lncRNAs), functional enrichment, and alternative splicing events (ASEs), validated by reverse transcription–quantitative polymerase chain reaction (RT-qPCR).Results: OGD/R injury significantly impaired cell proliferation and increased LDH release, effects mitigated by acacetin. RNA-seq identified 2148 upregulated and 2135 downregulated DEGs post-OGD/R. In contrast, the acacetin-treated group showed 248 upregulated and 240 downregulated DEGs compared to the OGD/R group. The use of diselenides and alkyl halides in reductive conditions represents a potent approach for synthesizing diverse un-symmetrical selenides. Nonetheless, current methods suffer from drawbacks due to the requirement of stoichiometric re-ductants. In this research, we introduce a new and straightforward photoinduced N-heterocyclic nitrenium catalysis as an alternative. This catalytic process enables single electron reduction of Se-Se bonds, leading to the generation of nucleophilic selenolates. Consequently, unsymmetrical selenides can be synthesized efficiently from readily available diselenides and alkyl chlorides.

Therefore, this study aimed to validate the Chinese version of the Tuberculosis related-Stigma Scale (TSS) in medical students and to apply the scale to measure stigma and its determinants.Methods: A total of 1035 eligible medical students participated in the cross-sectional questionnaire. Exploratory factor analyses (EFA) and confirmatory factor analyses (CFA) were first conducted to test Chinese version of the TSS construct validity, and then internal consistency was assessed using Cronbach’s α. Finally, multiple linear regression analyses were conducted to identify predictors of stigma.Results: EFA extracted two factors (“Social distance” and “Exclusionary sentiments”). The CFA further confirmed the two-factor construct. The internal consistency, convergent and divergent validity of the adapted Chinese version of the TSS was acceptable. Furthermore, Whether one has received TB health education and whether know a person with TB are associated with all dimensions of TB stigma, while differences in sex and place of residence play different roles in influencing the different dimensions of stigma.Conclusion: The adapted Chinese version of the TSS can be used to assess the level of stigma among medical students in China towards tuberculosis patients. In our study, we found that the health status (OR: 0.540, 95% CI: 0.298– 0.976), self-esteem (OR: 0.395, 95% CI: 0.251– 0.619), PSS (OR: 0.621, 95% CI: 0.388– 0.994), organizational support (OR: 0.533, 95% CI. 0.333– 0.854) were protective factors for healthcare workers suffering from anxiety, and resistance to COVID-19 (OR: 1.703, 95% CI: 1.082– 2.681) was a risk factor for healthcare workers suffering from anxiety, while good quality of life (OR: 0.385, 95% CI: 0.206– 0.719) self-esteem (OR: 0.187, 95% CI: 0.110– 0.317), and PSS (OR: 0.475, 95% CI: 0.267– 0.847) were protective factors for healthcare workers suffering from depression, and at the age of 35– 40 years (OR: 2.475, 95% CI: 1.140– 5.369) and resistance to COVID-19 (OR: 2.219, 95% CI: 1.313– 3.751) were risk factors for healthcare workers suffering from depression.Conclusion: The anxiety and depression status of healthcare workers in China is poor, and hospital administrators should take positive measures to support healthcare workers and give positive expectations to alleviate negative emotions such as anxiety and depression.Keywords: anxiety, depressi

A total of 245 patients were included. Data from January 2018 to December 2019 were used to construct the model, and data from January 2020 to December 2021 were used to evaluate the model. Data on maternal demographic characteristics, MRI cervical measurements were extracted. The prediction model was constructed with independent variables determined by multivariate logistic regression analyses. Through receiver-operating characteristic (ROC) curve analysis, the predictive ability of the model for subsequent second trimester pregnancy loss in women was evaluated, and internal validation was performed through validation data.Results: Thin cervix was observed in 77 (31.42%) women with prior second-trimester pregnancy loss, the mean longitudinal diameter of cervical canal on MRI was 11.76± 2.75mm. The model reached a sensitivity of 80%, specificity of 75.90%, positive predictive value (PPV) of 55.80% and negative predictive value of 90.90%; ROC characteristics proved that the model was superior to any single parameter with an AUC of 0.826.Conclusion: Our observati

We investigate the impact of the input dimension on the generalization error in generative adversarial networks (GANs). In particular, we first provide both theoretical and practical evidence to validate the existence of an optimal input dimension (OID) that minimizes the generalization error. Then, to identify the OID, we introduce a novel framework called generalized GANs (G-GANs), which includes existing GANs as a special case. By incorporating the group penalty and the architecture penalty developed in the paper, G-GANs have several intriguing features. First, our framework offers adaptive dimensionality reduction from the initial dimension to a dimension necessary for generating the target distribution. Second, this reduction in dimensionality also shrinks the required size of the generator network architecture, which is automatically identified by the proposed architecture penalty. Both reductions in dimensionality and the generator network significantly improve the stability and the accuracy of the estimation and prediction. Theoretical support for the consistent selection of the input dimension and the generator network is provided. Third, the proposed algorithm involves an end-to-end training process, and the algorithm allows for dynamic adjustments between the input dimension and the generator network during training, further enhancing the overall performance of G-GANs. Extensive experiments conducted with simulated and benchmark data demonstrate the superior performance of G-GANs. In particular, compared to that of off-the-shelf methods, G-GANs achieves an average improvement of 45.68% in the CT slice dataset, 43.22% in the MNIST dataset and 46.94% in the FashionMNIST dataset in terms of the maximum mean discrepancy or Frechet inception distance. Moreover, the features generated based on the input dimensions identified by G-GANs align with visually significant features.

Complex correlated states emerging from many-body interactions between quasiparticles (electrons, excitons and phonons) are at the core of condensed matter physics and material science. In low-dimensional materials, quantum confinement affects the electronic, and subsequently, optical properties for these correlated states. Here, by combining photoluminescence, optical reflection measurements and ab initio theoretical calculations, we demonstrate an unconventional excitonic state and its bound phonon sideband in layered silicon diphosphide (SiP2), where the bound electron-hole pair is composed of electrons confined within one-dimensional phosphorus-phosphorus chains and holes extended in two-dimensional SiP2 layers. The excitonic state and emergent phonon sideband show linear dichroism and large energy redshifts with increasing temperature. Our ab initio many-body calculations confirm that the observed phonon sideband results from the correlated interaction between excitons and optical phonons. With these results, we propose layered SiP2 as a platform for the study of excitonic physics and many-particle effects.

Revealing hydrothermal evolution from the early oxide to late carbonate stages for quartz–wolframite vein-type deposits is essential for understanding the ore-forming process. In this study, we choose the Baishitouwa tungsten polymetallic deposit located in the southern Great Xing'an Range tungsten belt as a case study, and present detailed deposit geology and in situ fluid inclusion (FI) analyses including microthermometry, laser Raman spectra, and LA-ICP-MS microanalysis to address this issue. Four stages of hydrothermal activity were identified: (1) quartz–wolframite (I), (2) quartz–wolframite (II)–pyrite–chalcopyrite, (3) quartz–polymetallic sulphides, and (4) quartz–carbonate. Four types of FIs were recognized: CO2-rich, CO2-bearing, liquid-rich, and brine inclusions. Microthermometric data showed that the homogenization temperatures and salinities from the early to late stages are 380–460°C, 7.4–17.3, and 29.3–43.2 wt% NaCl equiv., 300–390°C and 7.1–17.0 wt% NaCl equiv., 220–320°C and 2.7–8.1 wt% NaCl equiv., and 150–250°C and 0.5–4.8 wt% NaCl equiv., respectively, suggesting a decreasing trend. Geochemically, all stage fluids contained high Rb and Mn concentrations, high Rb/Na, Cs/Na, Li/Na, K/Na, Rb/Sr, low K/Rb, and consistent Cs/Rb and Cs/(Na + K) ratios, indicating that the mineralizing fluids originated from a common source—an underlying, geochemically uniform, and highly fractionated granitic magma. Fluid immiscibility and cooling are the main mechanisms for wolframite precipitation, whereas greisenization is subordinate; the incursion of meteoric water into the hydrothermal system initiated at the sulphide stage, and fluid mixing is the dominant mechanism for sulphide precipitation.

The aim of the present study is to verify the feasibility and effectiveness of ICG fluorescence imaging used for guiding the biopsy and resection of skin squamous cell carcinoma (SSCC).Methods: Sixty patients were enrolled, including 18 patients of suspected SSCC and 42 patients of diagnosed SSCC on admission. The ICG fluorescence imaging-guided skin biopsy was performed preoperatively in the 18 cases of suspected SSCC. Fifty-three patients underwent ICG fluorescence imaging-guided radical excision.Results: The results showed that 138 skin tissue samples in 60 patients with preoperative or intraoperative ICG fluorescence imaging-guide biopsy were collected. For a total number of 138 biopsies, 122 specimens were squamous cell carcinoma, and the accuracy rate was 88.4%, which was significantly higher than that of the group without preoperative ICG fluorescence imaging (41/62, 66.1%, P < 0.05). This review aimed to summarize the characteristics, possible mechanisms, and management of tigecycline-induced AP.Methods: Case reports of tigecycline-induced AP published in Chinese or English were collected until February 2023 for retrospective analysis.Results: Thirty-four patients from 29 articles were included. Fifteen patients (46.9%) had solid organ transplantation, and 4 patients (12.5%) had malignant tumors. Twenty-five patients (89.3%) received a recommended maintenance dose of tigecycline (50 mg q12 h). The median age was 50 years (range 9– 87). Compared to the nontransplant patients, the median age of the transplant patients was significantly younger, 44 years (range 12.5– 61) versus 57.5 years (range 9– 87) (P=0.03). The median time of symptom onset was 7 days (range 2– 29), and 91.2% (31/34) were less than 14 days. Typical initial symptoms included abdominal pain (90.6%), nausea (46.9%), vomiting (43.8%), and abdominal distention (21.9%). Most cases were accompanied by elevated levels of pancreatic enzymes. We strengthen the usual stability theorem for Vietoris-Rips (VR) persistent homology of finite metric spaces by building upon constructions due to Usher and Zhang in the context of filtered chain complexes. The information present at the level of filtered chain complexes includes ephemeral points, i.e. points with zero persistence, which provide additional information to that present at homology level. The resulting invariant, called verbose barcode, which has a stronger discriminating power than the usual barcode, is proved to be stable under certain metrics which are sensitive to these ephemeral points. In some situations, we provide ways to compute such metrics between verbose barcodes. We also exhibit several examples of finite metric spaces with identical (standard) VR barcodes yet with different verbose VR barcodes thus confirming that these ephemeral points strengthen the discriminating power of the standard VR barcode.

The circular coordinates algorithm of de Silva, Morozov, and Vejdemo-Johansson takes as input a dataset together with a cohomology class representing a 1-dimensional hole in the data; the output is a map from the data into the circle that captures this hole, and that is of minimum energy in a suitable sense. However, when applied to several cohomology classes, the output circle-valued maps can be "geometrically correlated" even if the chosen cohomology classes are linearly independent. It is shown in the original work that less correlated maps can be obtained with suitable integer linear combinations of the cohomology classes, with the linear combinations being chosen by inspection. In this paper, we identify a formal notion of geometric correlation between circle-valued maps which, in the Riemannian manifold case, corresponds to the Dirichlet form, a bilinear form derived from the Dirichlet energy. We describe a systematic procedure for constructing low energy torus-valued maps on data, starting from a set of linearly independent cohomology classes. We showcase our procedure with computational examples. Our main algorithm is based on the Lenstra-Lenstra-Lovász algorithm from computational number theory.

Unconventional hydrates with differently bound water molecules and related mazy intermolecular interaction networks need systematic investigation. The assembly rules, interaction analysis, and dehydration behavior can be dramatically more complicated when compared with those of common hydrates. In this work, creatine phosphate sodium (CPS) was selected as a model compound representing unconventional hydrates. The packing mode and the role of water molecules related to the dehydration mechanism were explored by the combination of experimental (diffraction, thermal, microscopy) and quantum chemistry computational (interplay visualization, binding energy calculation) methods. It was observed that in the structure of CPS heptahydrate and heminonahydrate, channel, isolated-site, and ion-associated water molecules exist simultaneously, constructing the framework via the coordination bond and hydrogen bond. The binding energy of some ion-coordinated water molecules is lower than that of hydrogen-bonded ones while that of some isolated-site water molecules is lower when compared with that of channel ones, which is counterintuitive. Moreover, the diverse types and locations of water molecules, complicated H2O···H2O interactions, and the trade-off between CPS···H2O and H2O···H2O lead to multistage, variable, and binding-energy-independent dehydration behavior. This work sheds light on the novel structure and variable dehydration mechanism of complicated hydrate systems and inspires the particularity and further investigation of unconventional hydrates.

The modified protocol of continuous sacral block was divided into three steps: comprehensive lumbar and sacral vertebral canal scanning by ultrasound, catheterization and administration. The EVENDOL pain scales and pediatric anesthesia emergence delirium scales of the children were evaluated at 5 min after extubation (T3), 90 min (T4), and 4 h (T5) after the operation. Magnetic topological states refer to a class of exotic phases in magnetic materials with the non-trivial topological property determined by magnetic spin configurations. An example of such states is the quantum anomalous Hall (QAH) state, which is a zero magnetic field manifestation of the quantum Hall effect. Current research in this direction focuses on QAH insulators with a thickness of less than 10 nm. Here, molecular beam epitaxy (MBE) is employed to synthesize magnetic TI trilayers with a thickness of up to ≈106 nm. It is found that these samples exhibit well-quantized Hall resistance and vanishing longitudinal resistance at zero magnetic field. By varying the magnetic dopants, gate voltages, temperature, and external magnetic fields, the properties of these thick QAH insulators are examined and the robustness of the 3D QAH effect is demonstrated. The realization of the well-quantized 3D QAH effect indicates that the nonchiral side surface states of the thick magnetic TI trilayers are gapped and thus do not affect the QAH quantization. The 3D QAH insulators of hundred-nanometer thickness provide a promising platform for the exploration of fundamental physics, including axion physics and image magnetic monopole, and the advancement of electronic and spintronic devices to circumvent Moore's law. © 2023 The Authors. An axion insulator is a three-dimensional (3D) topological insulator (TI), in which the bulk maintains the time-reversal symmetry or inversion symmetry but the surface states are gapped by surface magnetization. The axion insulator state has been observed in molecular beam epitaxy (MBE)-grown magnetically doped TI sandwiches and exfoliated intrinsic magnetic TI MnBi2Te4 flakes with an even number layer. All these samples have a thickness of ~ 10 nm, near the 2D-to-3D boundary. The coupling between the top and bottom surface states in thin samples may hinder the observation of quantized topological magnetoelectric response. Here, we employ MBE to synthesize magnetic TI sandwich heterostructures and find that the axion insulator state persists in a 3D sample with a thickness of ~ 106 nm. Our transport results show that the axion insulator state starts to emerge when the thickness of the middle undoped TI layer is greater than ~ 3 nm. The 3D hundred-nanometer-thick axion insulator provides a promising platform for the exploration of the topological magnetoelectric effect and other emergent magnetic topological states, such as the high-order TI phase. © 2023, The Author(s).

Micro-expression recognition (MER) in low-resolution (LR) scenarios presents an important and complex challenge, particularly for practical applications such as group MER in crowded environments. Despite considerable advancements in super-resolution techniques for enhancing the quality of LR images and videos, few study has focused on investigate super-resolution for improving LR MER. The scarcity of investigation can be attributed to the inherent difficulty in capturing the subtle motions of micro-expressions, even in original-resolution MER samples, which becomes even more challenging in LR samples due to the loss of distinctive features. Furthermore, a lack of systematic benchmarking and thorough analysis of super-resolution-assisted MER methods has been noted. This paper tackles these issues by conducting a series of benchmark experiments that integrate both super-resolution (SR) and MER methods, guided by an in-depth literature survey. Specifically, we employ seven cutting-edge state-of-the-art (SOTA) MER techniques and evaluate their performance on samples generated from 13 SOTA SR techniques, thereby addressing the problem of super-resolution in MER. Through our empirical study, we uncover the primary challenges associated with SR-assisted MER and identify avenues to tackle these challenges by leveraging recent advancements in both SR and MER methodologies. Our analysis provides insights for progressing toward more efficient SR-assisted MER.

Cohomological ideas have recently been injected into persistent homology and have for example been used for accelerating the calculation of persistence diagrams by the software Ripser. The cup product operation which is available at cohomology level gives rise to a graded ring structure that extends the usual vector space structure and is therefore able to extract and encode additional rich information. The maximum number of cocycles having non-zero cup product yields an invariant, the cup-length, which is useful for discriminating spaces. In this paper, we lift the cup-length into the persistent cup-length function for the purpose of capturing ring-theoretic information about the evolution of the cohomology (ring) structure across a filtration. We show that the persistent cup-length function can be computed from a family of representative cocycles and devise a polynomial time algorithm for its computation. We furthermore show that this invariant is stable under suitable interleaving-type distances.

The risks associated with unsteady two-phase flows in pressurized pipe systems must be considered both in system design and operation. To this end, this paper summarizes experimental tests and numerical analyses that highlight key aspects of unsteady two-phase flows in water pipelines. The essential dynamics of air–water interactions in unvented lines are first considered, followed by a summary of how system dynamics change when air venting is provided. System behaviour during unsteady two-phase flows is shown to be counter-intuitive, surprising, and complex. The role of air valves as protection devices is considered as is the reasonableness of the usual assumptions regarding air valve behaviour. The paper then numerically clarifies the relevance of cavitation and air valve performance to both the predicted air exchanges through any installed air valves and their role in modifying system behaviour during unsteady flows

BackgroundThe neurotrophic factor Artemin (ARTN) is involved in tumor proliferation and metastasis. Nonetheless, ARTN's significance in cervical cancer (CC) has not been studied. In our study, we propose to investigate the biological function of ARTN in CC as well as its particular regulatory mechanism.MethodsImmunohistochemistry (IHC) was used to examine the degree of ARTN protein expression in CC patient tissue. Real-time PCR and Western blotting were performed to reveal related genes' levels in CC cells. The CCK-8 test, the colony formation assay, the wound-healing assay, and the transwell assay were utilized to determine the proliferation, migration, and invasion capabilities, respectively. To generate lung metastasis models, stable ARTN-expressing SiHa cells were injected into the caudal tail vein of mice. IHC was used to examine the protein levels in CC mice model tissues.ResultsARTN was overexpressed in CC tissues relative to normal cervical tissues and linked positively with lymph node metastases (P=0.012) and recurrence (P=0.015) in CC patients. In vitro, ARTN overexpression promoted the proliferation, invasion, and migration of CC cells. In contrast, the consequences of depleting endogenous ARTN were the opposite. Moreover, overexpression of ARTN increased lung metastasis of CC cells in vivo and shortened the lifespan of mice models. In addition, ARTN overexpression significantly enhanced AKT phosphorylation on Ser473 and mTOR phosphorylation on Ser2448 and promoted the epithelial-mesenchymal transition (EMT) cascade. In addition, rapamycin, a selective inhibitor of mTORC1, might rescue the EMT phenotype caused by ARTN.ConclusionOur findings suggested that ARTN may enhance CC metastasis through the AKT/mTORC1 pathway. ARTN is anticipated to be a novel potential therapeutic target for the treatment of CC metastases.

This study aimed to estimate the patterns and trends of DR-TB in patients with suspected DR-TB. In addition, risk factors for multidrug-resistant tuberculosis (MDR-TB) were identified among suspected DR-TB patients in Dalian, China.Patients and Methods: A total of 5661 patients with suspected DR-TB from Jan 1, 2013 to Dec 31, 2020 were included in the final analysis. The resistance pattern of all resistant strains was determined by drug susceptibility testing (DST) using the conventional Lowenstein-Jensen Proportion Method (LJ). DR-TB trends were estimated from 2013 to 2020. During the research period, the chi-square test was employed to analyze the significance of linear drug-resistance trends across time. Bivariate and multivariate logistic regression were performed to assess factors associated with MDR-TB.Results: From 2013 to 2020, the resistance rates of rifampicin (RFP) and isoniazid (INH) decreased significantly, whereas the resistance rates of ethambutol (EMB) and streptomycin (SM) increased in patients with suspected DR-TB. From 2013 to 2020, the prevalence of DR-TB decreased in all patients from 34.71% to 28.01% with an average annual decrease of 3.02%. Among new cases, from 2013 to 2020, the prevalence of DR-TB (from 26.67% to 24.75%), RFP-resistant TB (RR-TB) (from 15.09% to 3.00%) and MDR-TB (from 6.08% to 2.62%) showed a significant downward trend. Among patients with a previous treatment history, DR-TB (from 54.70% to 37.50%), RR-TB (from 44.16% to 11.49%)

The circular coordinates algorithm of de Silva, Morozov, and Vejdemo-Johansson takes as input a dataset together with a cohomology class representing a $1$-dimensional hole in the data; the output is a map from the data into the circle that captures this hole, and that is of minimum energy in a suitable sense. However, when applied to several cohomology classes, the output circle-valued maps can be "geometrically correlated" even if the chosen cohomology classes are linearly independent. It is shown in the original work that less correlated maps can be obtained with suitable integer linear combinations of the cohomology classes, with the linear combinations being chosen by inspection. In this paper, we identify a formal notion of geometric correlation between circle-valued maps which, in the Riemannian manifold case, corresponds to the Dirichlet form, a bilinear form derived from the Dirichlet energy. We describe a systematic procedure for constructing low energy torus-valued maps on data, starting from a set of linearly independent cohomology classes. Background: The rechallenge of immune checkpoint inhibitors (ICI) is now an optional strategy for patients who discontinued ICI due to immune-related adverse events (irAEs) or disease progression. However, little data is available for the prognosis and prognostic factors of patients receiving ICI rechallenge treatment in advanced lung cancer patients. Our study aimed to explore the efficacy, prognosis and safety of patients who received anti-programmed cell death-1/programmed cell death ligand 1 (anti-PD-1/PD-L1) inhibitor rechallenge. Methods: In our retrospective cohort study, data of advanced lung cancer patients who received anti-PD-1/PD-L1 inhibitor and discontinued due to irAEs or disease progression were collected from December 2016 to August 2021. Enrolled patients were categorized into two groups: rechallenge group (R group) and non-rechallenge group (NR group). Progression-free survival (PFS), overall survival (OS), disease control rate (DCR) and safety data were analyzed. Cox model and subgroup analysis were analyzed according to baseline characteristics, ICI type, the reason for discontinuing ICI, etc. According to Response Evaluation Criteria in Solid Tumors Version 1.1 (RECIST 1.1), evaluation was performed routinely every 6-8 weeks after initiating treatment with the PD-1/PD-L1 inhibitor. The last follow-up in the study was on September 20, 2021. Results: Eighty-one patients who met our inclusion criteria were enrolled. In the whole cohort, the R group achieved better OS than the NR group [hazard ratio (HR) =0.176; 95% confidence interval (CI): 0.065-0.477; P=0.001). In the irAEs group, the survival analyses showed a trend toward improved OS in the rechallenge subgroup (HR =0.287; 95% CI: 0.081-1.025; P=0.055), and a promising DCR of 75% after an ICI rechallenge. Fusing regression coefficients into homogenous groups can unveil those coefficients that share a common value within each group. Such groupwise homogeneity reduces the intrinsic dimension of the parameter space and unleashes sharper statistical accuracy. We propose and investigate a new combinatorial grouping approach called $L_0$-Fusion that is amenable to mixed integer optimization (MIO). On the statistical aspect, we identify a fundamental quantity called grouping sensitivity that underpins the difficulty of recovering the true groups. We show that $L_0$-Fusion achieves grouping consistency under the weakest possible requirement of the grouping sensitivity: if this requirement is violated, then the minimax risk of group misspecification will fail to converge to zero. Moreover, we show that in the high-dimensional regime, one can apply $L_0$-Fusion coupled with a sure screening set of features without any essential loss of statistical efficiency, while reducing the computational cost substantially. On the algorithmic aspect, we provide a MIO formulation for $L_0$-Fusion along with a warm start strategy. Simulation and real data analysis demonstrate that $L_0$-Fusion exhibits superiority over its competitors in terms of grouping accuracy.

A previous paper proposed an accurate and efficient matched-field processing method for leak localization in looped pipe networks based on the known discharge and pressure head (i.e., hydraulic properties) at all internal nodes. However, the computation of these internal hydraulic properties from boundary measurements is an ill-conditioned inverse problem: for a small perturbation in the boundary measurements there is a huge change in the internal hydraulic properties. We prove and clarify the invalidity of the classic Tikhonov regularization for this ill-conditioned problem. As an alternative, this paper derives the theoretical upper bounds of noise-induced errors, based on which a threshold is determined for identifying ill-conditioned frequencies. The leak localization is implemented by discarding wave information at all ill-conditioned frequencies. This results in an efficient and robust algorithm for leak localization in noisy environments. Numerical experiments demonstrate that an unknown leak anywhere in looped pipe networks can be accurately localized by the proposed algorithm.

It is necessary to optimize the design of the missile aerodynamic shape for better performance while meeting tactical specifications. However, current design methods for aerodynamic shape are based on manual design and physical model simulations, which are very time-consuming. Therefore, we propose an optimization framework based on conditional Wasserstein Gan-GP (CWGAN-GP), convolutional neural network (CNN), multi-task learning with multi-gate mixture-of-Experts-3D (MMoE-3D) and differential evolution (DE). This method consists of four stages. In the first step, CWGAN-GP can learn the relationship between existing missile shape designs and shape conditions, generating diverse missile shapes as required. In the second step, CNN is used for feature extraction of missile design drawing, and the missile shape generated by CWGAN-GP is transformed into missile shape parameters. In the third step, the MMoE-3D model is trained in the subsonic and supersonic ranges to efficiently generate aerodynamic data corresponding to the missile shape. In the fourth step, DE is used to select the optimal missile shape by adjusting the potential variables of CWGAN-GP. The efficiency of the proposed optimization framework is verified by optimizing the rate of change of the center of pressure and the lift-to-drag ratio, with the neural network-based optimization framework achieving almost the same optimization results in a shorter time compared to conventional optimization with DATCOM.

Polarized neutron reflectometry is a powerful technique to interrogate the structures of multilayered magnetic materials with depth sensitivity and nanometer resolution. However, reflectometry profiles often inhabit a complicated objective function landscape using traditional fitting methods, posing a significant challenge for parameter retrieval. In this work, we develop a data-driven framework to recover the sample parameters from polarized neutron reflectometry data with minimal user intervention. We train a variational autoencoder to map reflectometry profiles with moderate experimental noise to an interpretable, low-dimensional space from which sample parameters can be extracted with high resolution. We apply our method to recover the scattering length density profiles of the topological insulator–ferromagnetic insulator heterostructure Bi2Se3/EuS exhibiting proximity magnetism in good agreement with the results of conventional fitting. We further analyze a more challenging reflectometry profile of the topological insulator–antiferromagnet heterostructure (Bi,Sb)2Te3/Cr2O3 and identify possible interfacial proximity magnetism in this material. We anticipate that the framework developed here can be applied to resolve hidden interfacial phenomena in a broad range of layered systems.

A 3-knotted restricted cubic spline (RCS) fitted for Cox proportional hazard regression models is used to analyse the effects of cut-off points of treatment delay on incident poor treatment outcomes. To explore the moderating effects of age, gender and diabetes, we added the interaction terms of these moderating variables and treatment delay to Cox proportional hazard regression models.Results: The median time of treatment initiation was 30 days (IQR: 14– 59 days). The risk of incident poor treatment outcomes increased when the time was greater than cut-off point 1 (53 days; adjusted HR: 1.26; 95% CI: 1.00– 1.60) of treatment delay, and the risk of incident death events increased when the time was greater than cut-off point 2 (103 days; adjusted HR: 1.56; 95% CI: 1.00– 2.44) of delay. Descriptive statistics were employed to calculate the frequencies and percentages of TB strains, and the average annual growth rates (AAGRs) for each strain were calculated. The Chi-square test was applied to examine the significance of linear drug-resistance trends over time during the study period.Results: Over the eight-year study period, the percentages of both initial (from 9.01% to 4.82%) and acquired (from 40.85% to 9.09%) MDR-TB cases decreased significantly, AAGRs of 8.55% and 19.32%, respectively. [Background and Aim] There is debate among the hepatology community regarding the simple non-invasive scoring systems and histological scores (even it was developed for histological classification) in predicting clinical outcomes in patients with non-alcoholic fatty liver disease (NAFLD). This study aimed to determine whether the presence of simple non-invasive scoring systems and histological scores could predict all-cause mortality, liver-related mortality, and liver disease decompensation (liver failure, cirrhosis, hepatocellular carcinoma, or decompensated liver disease)., [Methods] The pooled hazard ratio of prognostic factors and incidence rate per 1000 person-years in patients with NAFLD was calculated and further adjusted by two different models of handling the duplicated data., [Results] A total of 19 longitudinal studies were included. Most simple non-invasive scoring systems (Fibrosis-4 Score, BARD, and aspartate aminotransferase-to-platelet ratio index ) and histological scores (NAFLD activity score, Brunt, and "steatosis, activity, and fibrosis" ) failed in predicting mortality, and only the NAFLD fibrosis score > 0.676 showed prognostic ability to all-cause mortality (four studies, 7564 patients, 118 352 person-years followed up, pooled hazard ratio 1.44, 95% confidence interval [CI] 1.05–1.96). The incidence rate per 1000 person-years of all-cause mortality, liver-related mortality, cardiovascular-related mortality, and liver disease decompensation resulted in a pooled incidence rate per 1000 person-years of 22.65 (14 studies, 95% CI 9.62–53.31), 3.19 (7 studies, 95% CI 1.14–8.93), 6.02 (6 studies, 95% CI 4.69–7.74), and 11.46 (4 studies, 95% CI 5.33–24.63), respectively., [Conclusion] Non-alcoholic fatty liver disease fibrosis score showed promising prognostic value to all-cause mortality. Most present simple non-invasive scoring systems and histological scores failed to predict clinical outcomes.

Micro-expression recognition (\textbf{MER}) has attracted lots of researchers' attention in a decade. However, occlusion will occur for MER in real-world scenarios. This paper deeply investigates an interesting but unexplored challenging issue in MER, \ie, occlusion MER. First, to research MER under real-world occlusion, synthetic occluded micro-expression databases are created by using various mask for the community. Second, to suppress the influence of occlusion, a \underline{R}egion-inspired \underline{R}elation \underline{R}easoning \underline{N}etwork (\textbf{RRRN}) is proposed to model relations between various facial regions. RRRN consists of a backbone network, the Region-Inspired (\textbf{RI}) module and Relation Reasoning (\textbf{RR}) module. More specifically, the backbone network aims at extracting feature representations from different facial regions, RI module computing an adaptive weight from the region itself based on attention mechanism with respect to the unobstructedness and importance for suppressing the influence of occlusion, and RR module exploiting the progressive interactions among these regions by performing graph convolutions. Experiments are conducted on handout-database evaluation and composite database evaluation tasks of MEGC 2018 protocol. Experimental results show that RRRN can significantly explore the importance of facial regions and capture the cooperative complementary relationship of facial regions for MER. The results also demonstrate RRRN outperforms the state-of-the-art approaches, especially on occlusion, and RRRN acts more robust to occlusion.

Cohomological ideas have recently been injected into persistent homology and have for example been used for accelerating the calculation of persistence diagrams by the software Ripser. The cup product operation which is available at cohomology level gives rise to a graded ring structure that extends the usual vector space structure and is therefore able to extract and encode additional rich information. The maximum number of cocycles having non-zero cup product yields an invariant, the cup-length, which is useful for discriminating spaces. In this paper, we lift the cup-length into the persistent cup-length function for the purpose of capturing ring-theoretic information about the evolution of the cohomology (ring) structure across a filtration. We show that the persistent cup-length function can be computed from a family of representative cocycles and devise a polynomial time algorithm for its computation. Accurately estimating personalized treatment effects within a study site (e.g., a hospital) has been challenging due to limited sample size. Furthermore, privacy considerations and lack of resources prevent a site from leveraging subject-level data from other sites. We propose a tree-based model averaging approach to improve the estimation accuracy of conditional average treatment effects (CATE) at a target site by leveraging models derived from other potentially heterogeneous sites, without them sharing subject-level data. To our best knowledge, there is no established model averaging approach for distributed data with a focus on improving the estimation of treatment effects. Specifically, under distributed data networks, our framework provides an interpretable tree-based ensemble of CATE estimators that joins models across study sites, while actively modeling the heterogeneity in data sources through site partitioning. Micro-Expression Recognition has become challenging, as it is extremely difficult to extract the subtle facial changes of micro-expressions. Recently, several approaches proposed several expression-shared features algorithms for micro-expression recognition. However, they do not reveal the specific discriminative characteristics, which lead to sub-optimal performance. This paper proposes a novel Feature Refinement ({FR}) with expression-specific feature learning and fusion for micro-expression recognition. It aims to obtain salient and discriminative features for specific expressions and also predict expression by fusing the expression-specific features. FR consists of an expression proposal module with attention mechanism and a classification branch. First, an inception module is designed based on optical flow to obtain expression-shared features. Second, in order to extract salient and discriminative features for specific expression, expression-shared features are fed into an expression proposal module with attention factors and proposal loss. Last, in the classification branch, labels of categories are predicted by a fusion of the expression-specific features. Experiments on three publicly available databases validate the effectiveness of FR under different protocol. Results on public benchmarks demonstrate that our FR provides salient and discriminative information for micro-expression recognition. The results also show our FR achieves better or competitive performance with the existing state-of-the-art methods on micro-expression recognition.

Urea (CO(NH2)2) is conventionally synthesized through two consecutive industrial processes, N2 + H2 → NH3 followed by NH3 + CO2 → urea. Both reactions operate under harsh conditions and consume more than 2% of the world’s energy. Urea synthesis consumes approximately 80% of the NH3 produced globally. Here we directly coupled N2 and CO2 in H2O to produce urea under ambient conditions. The process was carried out using an electrocatalyst consisting of PdCu alloy nanoparticles on TiO2 nanosheets. This coupling reaction occurs through the formation of C–N bonds via the thermodynamically spontaneous reaction between *N=N* and CO. Products were identified and quantified using isotope labelling and the mechanism investigated using isotope-labelled operando synchrotron-radiation Fourier transform infrared spectroscopy. A high rate of urea formation of 3.36 mmol g–1 h–1 and corresponding Faradic efficiency of 8.92% were measured at –0.4 V versus reversible hydrogen electrode. [Figure not available: see fulltext.]

Air expulsion from an end-of-pipe orifice in a rapidly filling horizontal pipe is investigated experimentally and analytically in order to more completely characterize the system's transient response. In particular, images of air-water patterns, air-volume variations, orifice flow regimes, and measured pressure histories are synchronized to elucidate the process of air expulsion. Air expulsion typically undergoes an early stage involving pressurization, expansion, and release of a portion of the initial air, events that generally occur even before the advancing water column reaches the pipe end. The next stage depends strongly on the orifice size. For a small discharge orifice, an oscillation of the residual air occurs with the discharge orifice being intermittently choked by water; by contrast, larger discharge orifices rapidly and completely expelled the air, often leading to high water-hammer pressures. Three distinct patterns of pressure oscillation are typically observed. With small orifices, the cushioning effect of the initial air tends to dominate, whereas slightly larger orifices lead to a more complex process of expulsion and more persistent and larger pressure oscillations. Even larger orifices often lead to severe water-hammer pressures. Thus, smaller orifices tend to result in smaller pressure fluctuations. As expected, both the initial-air volume and the inlet pressure significantly influence the transient response. A derived analytical model accurately captures the measured pressure oscillations during the intermittent release of residual air, including the water hammer that can arise due to suddenly arresting the liquid water column.

Objectives Diverse evidence including clinical, genetic and microbiome studies support a major role of the gut microbiome in the common immune-mediated arthropathy, ankylosing spondylitis (AS). We set out to (1) further define the key microbial characteristics driving disease, and (2) examine the effects of tumour necrosis factor-inhibitor (TNFi) therapy upon the microbiome. Methods The stools from a case–control cohort of 250 Han-Chinese subjects underwent shotgun metagenomic sequencing. All subjects were genotyped using the Illumina CoreExome SNP microarray. Results Previous reports of gut dysbiosis in AS were reconfirmed and several notable bacterial species and functional categories were differentially abundant. TNFi therapy was correlated with a restoration the perturbed microbiome observed in untreated AS cases to that of healthy controls, including several important bacterial species that have been previously associated with AS and other related diseases. Enrichment of bacterial peptides homologous to HLA-B27-presented epitopes was observed in the stools of patients with AS, suggesting that either HLA-B27 fails to clear these or that they are involved in driving HLA-B27-associated immune reactions. TNFi therapy largely restored the perturbed microbiome observed in untreated AS cases to that of healthy controls, including several important bacterial species that have been previously associated with AS and other related diseases. TNFi therapy of patients with AS was also associated with a reduction of potentially arthritogenic bacterial peptides, relative to untreated patients. Conclusion These findings emphasise the key role that the gut microbiome plays in driving the pathogenesis of AS and highlight potential therapeutic and/or preventative targets.

The purpose of present study was to explore the roles of medication adherence, doctor–patient trust and communication on treatment effects, and its inequality between urban and rural areas.Methods: There were 564 eligible TB patients, from four tuberculosis hospitals in China, participating in this cross-sectional study. They filled out questionnaires regarding socio-demographic characteristics, medication adherence, treatment effect, doctor–patient trust, and communication. The structural equation model (SEM) was applied to explore the hypotheses in this study. All statistical analysis was done by SPSS 25.0 and Mplus 7.0 statistical software.Results: This study included 267 (47.34%) urban and 297 (52.66%) rural eligible TB patients. The data fitted the research model well, and the urban TB patients reported better treatment effect than the rural ones (P=0.027). Overall, treatment adherence positively predicted treatment effect (Est.=0.353, P< 0.001); doctor–patient communication positively influenced treatment adherence (Est.=0.214, P=0.002); and treatment adherence positively mediated the role of communication on treatment effect (Est.=0.076), 95% CI (0.026, 0.152). Medication adherence was measured using the eight-item Chinese version of the Morisky Medication Adherence Scale, which divides adherence into three levels. The independent variables consisted of sociodemographic characteristics, treatment factors, knowledge about TB, mental health, and behavioral characteristics. Descriptive statistics, the chi-square test, and multivariate ordinal logistic regression were applied to analyze the data using Stata/MP 14.0.Results: Among the 564 eligible participants, 236 (41.84%) and 183 (32.45%) exhibited high and medium medication adherence, respectively, but 145 (25.71%) exhibited low medication adherence. Multivariate ordinal logistic regression showed that patients who were older (OR: 1.02, p=0.013) were employed (OR: 1.61, p=0.011), had better tuberculosis knowledge (OR: 1.34, p< 0.001), and did not consume alcohol (OR: 1.84, p=0.032) exhibited higher medication adherence. Micro-Expression Recognition (MER) is a challenging task as the subtle changes occur over different action regions of a face. Changes in facial action regions are formed as Action Units (AUs), and AUs in micro-expressions can be seen as the actors in cooperative group activities. In this paper, we propose a novel deep neural network model for objective class-based MER, which simultaneously detects AUs and aggregates AU-level features into micro-expression-level representation through Graph Convolutional Networks (GCN). Specifically, we propose two new strategies in our AU detection module for more effective AU feature learning: the attention mechanism and the balanced detection loss function. With those two strategies, features are learned for all the AUs in a unified model, eliminating the error-prune landmark detection process and tedious separate training for each AU. Moreover, our model incorporates a tailored objective class-based AU knowledge-graph, which facilitates the GCN to aggregate the AU-level features into a micro-expression-level feature representation. Extensive experiments on two tasks in MEGC 2018 show that our approach significantly outperforms the current state-of-the-arts in MER. Additionally, we also report our single model-based micro-expression AU detection results.
