ABSTRACTMost studies on remedial courses are based on their mandatory attendance. However, changes may have occurred in the attendance policy of developmental math courses since the state of Florida decided to overcome obligatory math remediation for unprepared students. Consequently, researchers have recently started focusing on voluntary math remedial courses. In general, literature that goes back to the 1980s suggests that developmental coursework should be mandatory for unprepared first-year students. Since most universities in the US have always followed these recommendations, hardly any empirical evidence exists for the participation of students in voluntary remediation. Conversely, the remedial education system in Europe and particularly Germany is primarily voluntary. Therefore, this study exploratively examines the participation of students in two optional developmental math courses: a so-called preparatory course and a so-called bridging course. The findings suggest that summer-school-like preparatory courses miss their target group of at-risk students, whereas semester-running bridging courses reach it.

Genetic alterations associated with prostate cancer (PCa) may be identified by sequencing metastatic tumour genomes to identify molecular markers at this lethal stage of disease. Previously, we characterized somatic alterations in metastatic tumours in the methylcytosine dioxygenase ten-eleven translocation 2(TET2), which is altered in 5–15% of myeloid, kidney, colon and PCas. Genome-wide association studies previously identified non-coding risk variants associated with PCa and melanoma. We perform fine-mapping of PCa risk across TET2using genotypes from the PEGASUS case-control cohort and identify six new risk variants in introns 1 and 2. Oligonucleotides containing two risk variants are bound by the transcription factor octamer-binding protein 1 (Oct1/POU2F1) and TET2and Oct1expression are positively correlated in prostate tumours. TET2is expressed in normal prostate tissue and reduced in a subset of tumours from the Cancer Genome Atlas (TCGA). Small interfering RNA-mediated TET2knockdown (KD) increases LNCaP cell proliferation, migration and wound healing, verifying loss drives a cancer phenotype. Endogenous TET2 bound the androgen receptor (AR) and AR-coactivator proteins in LNCaP cell extracts, and TET2KD increases prostate-specific antigen (KLK3/PSA) expression. Published data reveal TET2 binding sites and hydroxymethylcytosine proximal to KLK3. A gene co-expression network identified using TCGA prostate tumour RNA-sequencing identifies co-regulated cancer genes associated with 2-oxoglutarate (2-OG) and succinate metabolism, including TET2, lysine demethylase (KDM) KDM6A, BRCA1-associated BAP1, and citric acid cycle enzymes IDH1/2, SDHA/B, and FH. The co-expression signature is conserved across 31 TCGA cancers suggesting a putative role for TET2 as an energy sensor (of 2-OG) that modifies aspects of androgen-AR signalling. Decreased TET2mRNA expression in TCGA PCa tumours is strongly associated with reduced patient survival, indicating reduced expression in tumours may be an informative biomarker of disease progression and perhaps metastatic disease.

Solid organ transplant recipients have increased colorectal cancer (CRC) risk. We assessed CRC risk among transplant recipients and identified factors contributing to this association. The US transplant registry was linked to 15 population‐based cancer registries (1987–2010). We compared CRC risk in recipients to the general population by using standardized incidence ratios (SIRs) and identified CRC risk factors by using Poisson regression. Based on 790 cases of CRC among 224 098 transplant recipients, the recipients had elevated CRC risk (SIR 1.12, 95% confidence interval [CI] 1.04 to 1.20). The increase was driven by an excess of proximal colon cancer (SIR 1.69, 95% CI 1.53 to 1.87), while distal colon cancer was not increased (SIR 0.93, 95% CI 0.80 to 1.07), and rectal cancer was reduced (SIR 0.64, 95% CI 0.54 to 0.76). In multivariate analyses, CRC was increased markedly in lung recipients with cystic fibrosis (incidence rate ratio [IRR] 12.3, 95% CI 6.94 to 21.9, vs. kidney recipients). Liver recipients with primary sclerosing cholangitis and inflammatory bowel disease also had elevated CRC risk (IRR 5.32, 95% CI 3.73 to 7.58). Maintenance therapy with cyclosporine and azathioprine was associated with proximal colon cancer (IRR 1.53, 95% CI 1.05 to 2.23). Incidence was not elevated in a subgroup of kidney recipients treated with tacrolimus and mycophenolate mofetil, pointing to the relevance of the identified risk factors. Transplant recipients have increased proximal colon cancer risk, likely related to underlying medical conditions (cystic fibrosis and primary sclerosing cholangitis) and specific immunosuppressive regimens. Using data from the large U.S. Transplant Cancer Match Study, the authors show that solid organ transplant recipients have an increased risk of proximal colon cancer, in part related to underlying medical conditions and specific immunosuppressive regimens.

Thirty-nine leaf samples ofNarthecium ossifragumcollected from eight sites in More og Romsdal County, Norway, during June–September 1997 and 41 leaf samples collected at five sites in the same county during June–August 1998 were analysed for the concentrations of steroidal sapogenins using GC-MS. The 1998 samples were also examined for fungal elements (conidia and hyphae) after incubation in a moist chamber for 10–14 days. The highest 1997 and 1998 leaf sapogenin concentrations (4881 and 7115 mg/kg dry matter, respectively) were 13–14 times greater than the lowest sapogenin concentrations found (344 and 531 mg/kg dry matter, respectively). The results did not reveal systematic differences in sapogenin concentrations between the two seasons, or between samples harvested early or late in the same seasons, or between sapogenin concentrations in plants harvested at different sites.Cladosporium magnusianumwas the predominant fungus found in the samples. The degree of fungal infection on the samples was in generally low, but the number ofC. magnusianumcolonies in the moist chamber preparations and fungal elements (conidia and hyphae) in leaf washings and on leaves tended to increase with time. Factor analysis and multiple regression analysis performed on the chemical and fungal results suggest that sporulation may have occurred in the fungi in response to increase in sapogenin concentrations.

BackgroundThe improvement of quality of life gains increasing importance for the judgement of operative techniques. Besides the commonly used mechanical substitutes or bioprostheses for aortic valve replacement, the interest in the Ross procedure is growing. The aim of the study was to compare the quality of life after the Ross procedure with that after mechanical aortic valve replacement with two different anticoagulation regimes (self-management or conventional therapy). Methods and resultsClinical, echocardiographic and quality of life investigations (SF-36) were performed in patients with mechanical aortic valve replacement and self-management of anticoagulation (group A, n=20) or conventional anticoagulation therapy (group B, n=20) and in patients after the Ross procedure (group C, n=20). The mean ages were 59.5±9.2 (group A), 61.2±8.1 (group B) and 59.3±9 years (group C). Significantly lower values of quality of life (SF-36) were observed in group B compared with group A (5 of 9 subtests) and with group C (6 of 9 subtests) and also in the physical and mental health sum scales. ConclusionIn this study the quality of life in patients after the Ross procedure and similarly after mechanical valve replacement and self-management of anticoagulation is superior to the quality of life after mechanical valve replacement and conventional anticoagulation. Neben den herkömmlichen mechanischen und biologischen Substituten als Aortenklappenersatz wächst das Interesse für den pulmonalen Autograft (Ross-Operation). Ziel der vorliegenden Studie ist der Vergleich der Lebensqualität nach Ross-Operation mit der Lebensqualität nach mechanischem Aortenklappenersatz und zwei unterschiedlichen Verfahren der Antikoagulation (herkömmliches Regime bzw. Selbstmanagement). Methoden und ErgebnisseKlinische, echokardiographische und gesundheitsbezogene Lebensqualitätsuntersuchungen (SF-36) wurden bei Patienten mit mechanischem Aortenklappenersatz und Antikoagulationsselbstmanagement (Gruppe A, n=20) sowie konventionellem Antikoagulationsregime (Gruppe B, n=20) und bei Patienten nach Ross-Operation (Gruppe C, n=20) durchgeführt. Das mittlere Alter betrug 59,5±9,2 (Gruppe A) 61,2±8,1 (Gruppe B) und ¶59,3±9 Jahre (Gruppe C). Signifikant niedrigere Werte der Lebensqualität (SF-36) bestanden in Gruppe B im Vergleich zu Gruppe A (5 von 9 Subtests) und zu Gruppe C (6 von 9 Subtests) sowie in den Summenskalen physische und mentale Gesundheit. SchlussfolgerungIn der vorliegenden Arbeit wiesen die Patienten nach mechanischem Aortenklappenersatz mit Selbstmanagement der Antikoagulation eine vergleichbare Lebensqualität auf wie nach Ross-Operation und eine höhere Lebensqualität im Vergleich zur herkömmlichen Antikoagulationstherapie.

The correlations between the relaxing effect of papaverine derivatives, inhibition of low Km-phosphodiesterase (cAMP-PDE=EC 3.1.4.17) activity and cyclic 3',5'-AMP (cAMP) levels in isolated rabbit ileum were investigated. There was a strong correlation between the relaxing effect, inhibition of PDE activity and cAMP content for eupaverine, ethylpapaverine and papaverine. Eupaverine was the most effective relaxing agent (I50=7.5 µM) and the most potent inhibitor of PDE activity (Ki=0.6 µM), followed by ethylpapaverine (I50=10 µM); Ki=0.8 µM) and papaverine (I50=20 µM; Ki=2 µM). In contrast, there was a strong relaxing effect (I50=6 µM) but only slight inhibition of PDE activity (Ki=350 µM) by tetrahydropapaveroline (THP). The adenylate cyclase stimulating effect of THP which was shown by others is most likely the reason for comparatively higher cAMP levels, which were found to be elevated about seven times over basal levels of 0.35 nmoles/g wet weight, and effective relaxation. Relaxation could be induced by exogenously added cAMP (I50=45 µM) and dibutyryl-cAMP (I50=450 µM). Our results support the assumption that smooth muscle relaxation in rabbit ileum is mediated by cAMP. Some of these observations have been published in abstract form (Schulz and Berndt, 1972).

The effects of p,p'-DDT and o,p'-DDT on adrenal cortex function have been studied in rats and dogs. After oral administration (200 mg/kg) to rats both isomers of DDT accumulated in higher concentrations in the adrenals than in adipose tissue. Repeated doses lead to cumulative storage in adipose tissue but not in the adrenals.

Summary The effects of methylxanthines, papaverine, dipyridamole and imipramine on lipolysis and phosphodiesterase activity of rat adipose tissue were investigated. Lipolysis in isolated fat cells was stimulated by theophylline and caffeine whereas papaverine, dipyridamole and imipramine had no substantial effect on the basal lipolytic rate. Lipolysis induced by noradrenaline was potentiated by theophylline, but blocked by papaverine, dipyridamole and imipramine at concentrations between 0.02 to 0.2 mM. These agents also depressed lipolysis induced by theophylline and dibutyryl cyclic AMP and reduced the lipolytic activity of homogenates of adipose tissue. The activity of phosphodiesterase assayed over a wide range of substrate concentrations revealed two different Michaelis constants. Both types of phosphodiesterase were inhibited by theophylline, papaverine, dipyridamole and imipramine in a competitive manner, the low Km enzyme being more sensitive for inhibition than the high Km enzyme. On both types of phosphodiesterase papaverine and dipyridamole proved to be 10 to 100 times more potent inhibitors than theophylline and imipramine. To explain the antilipolytic effect of phosphodiesterase inhibitors it is assumed that they do not only affect substrate binding of cyclic AMP to phosphodiesterase but also displace cyclic AMP from the binding site on protein kinase, thus acting as inhibitors of the activation process within the lipolytic system.
