CYP106A2 from Bacillus megaterium ATCC 13368 is a bacterial cytochrome P450 that is capable of transforming steroid hormones. It can be easily expressed in Escherichia coli with a high yield. Its activity in vitro can be achieved by using the adrenal redox proteins adrenodoxin and adrenodoxin reductase. So far, it was not possible to crystallize CYP106A2 because of degradation during the crystallization process. Nevertheless, CYP106A2 is an interesting enzyme for biotechnological use. It hydroxylates pharmaceutically important steroids such as progesterone and 11-deoxycortisol. However, it will be necessary for efficient application of CYP106A2 in biotechnology to improve the hydroxylation activity and manipulate the regiospecificity. The present paper gives an overview of recent developments in protein engineering of CYP106A2.

Elevated plasma aldosterone levels play a detrimental role in certain forms of congestive heart failure and myocardial fibrosis. We proposed aldosterone synthase (CYP11B2) as a novel target for the treatment of these diseases. In this study, the synthesis and biological evaluation of substituted E- and Z-imidazolylmethylenetetrahydronaphthalenes and E- and Z-imidazolylmethyleneindanes (compounds 1a,b−9a,b) is described. The compounds were prepared by a Wittig-like reaction. They were tested for activity using bovine CYP11B and human CYP11B2 expressed in fission yeast and V79 MZh cells. Selectivity was determined toward human CYP11B1, CYP19, and CYP17. Especially in the case of CYP11B1 (steroid 11β-hydroxylase), selectivity is a crucial issue, since sequence homology between this enzyme and the target enzyme is very high (93%). On the basis of the X-ray structure of human CYP2C9, a protein model of CYP11B2 was developed and docking experiments with the title compounds were performed. The biological results revealed highly potent inhibitors of CYP11B2 (IC50 = 4−93 nM). The Z-isomers usually were more active than the corresponding E-isomers. Different inhibitory profiles could be observed:  rather selective inhibitors of CYP11B1, dual inhibitors of both enzymes, and rather selective inhibitors of CYP11B2. The chloro derivative 8b was found to be a highly potent CYP11B2 inhibitor (IC50 = 4 nM) showing a 5-fold selectivity for CYP11B1 (IC50 = 20 nM). This compound could be an interesting lead for further optimization as a therapeutic agent. It also could be used as well as the CYP11B1 selective compounds as a pharmacological tool.

This paper presents a novel Cobotic system with differential CVT. The new system is significantly cheaper, simpler to control and more efficient than Cobots with S-CVTs. Both path-guidance and power-assist functions can be simply realized with the new system. Basic structures, kinematic and dynamic models, as well as control algorithms, which are essential for design, control synthesis and control of the system, are briefly presented in the paper.

Transforming growth factor beta1 (TGFbeta1) has been shown to exert strong inhibitory effects on adrenocortical cell steroidogenesis. However, the molecular targets of TGFbeta1 in adrenocortical cells appear to differ between species. Here, we report the first characterization of the regulatory effects of TGFbeta1 on the steroidogenic functions of the human adrenocortical tumor cell line NCI-H295R. After treatment with 2 ng/ml TGFbeta1 for 24 h, basal production of corticosterone, cortisol and androstenedione was dramatically decreased. When TGFbeta1 was added simultaneously with forskolin, the production of cortisol and 11-hydroxyandrostenedione was decreased by 85% whereas that of deoxycortisol was increased. When TGFbeta1 was added simultaneously with angiotensin II, aldosterone production was reduced by 80%. We observed that TGFbeta1 strongly inhibits forskolin-induced steroid 11beta-hydroxylase activity and CYP11B1 mRNA levels, as well as angiotensin II-induced aldosterone synthase activity and CYP11B2 mRNA levels. CYP11B1 and CYP11B2 gene products thus appear as the major steroidogenic enzymes down-regulated by TGFbeta1 in the human adrenocortical tumor cell line NCI-H295R.

AbstractMajor advances have been made during the last decade in our understanding of adrenal steroid hormone biosynthesis. Two key players in these pathways are the human mitochondrial cytochrome P450 enzymes CYP11B1 and CYP11B2, which catalyze the final steps in the biosynthesis of cortisol and aldosterone. Using data from mutations found in patients suffering from steroid hormonerelated diseases, from mutagenesis studies and from the construction of threedimensional models of these enzymes, structural information could be deduced that provide a clue to the stereo and regiospecific steroid hydroxylation reactions carried out by these enzymes. In this review, we summarize the current knowledge on the physiological function and the biochemistry of these enzymes. Furthermore, the pharmacological and toxicological importance of these steroid hydroxylases, the means for the identification of their potential inhibitors and possible biotechnological applications are discussed.

Mitochondrial adrenodoxin (Adx) was found to cross-react with microsomal cytochrome P450 2B4 (CYP2B4) as the terminal electron acceptor. When compared with NADPH-cytochrome P450 reductase (P450R), the natural redox partner of CYP2B4, Adx was less efficient both in transferring the first electron and in coupling the system. The ferredoxin yielded an unusual reverse type I spectral change with low-spin CYP2B4, which underwent transformation to a typical type I optical perturbation upon deletion of the signal anchor sequence (Δ2-27) of the hemoprotein. Truncation of CYP2B4 slightly fostered electron transfer from Adx, but was deleterious to reduction of the engineered isozyme by P450R. Addition of manganese-substituted cytochrome b5, which failed to serve as an electron donor to CYP2B4, augmented the amount of hemoprotein existing in form of a low-spin complex with Adx and affected the ferredoxin-dependent reduction kinetics through causing a proportional rise in both Kmand Vmax. Conservative replacement of Asp-76 with glutamate in the Adx molecule was associated with a drastic drop in reductive efficiency toward CYP2B4, while spectral binding of the mutant to the hemoprotein was marginally changed. The results support the concept of an evolutionary relationship between the various cytochrome P450 forms as regards the conservation of surface regions participating in contacts with heterologous donor proteins.

The paper presents a concept and a first realization of a knowledge based off-line programming system. The design is based on an already developed system which was also applied for relevant production tasks. A characteristic feature of the system is a production task specific, dialog oriented user interface. For a flexible realization of the user interface knowledge based approaches have been applied. A concept for a first realization is presented.

The amino acid in position 54 of adrenodoxin is strongly conserved among ferredoxins, consisting of a threonine or serine. Its role was studied by analyzing mutants T54S and T54A of bovine adrenodoxin. Absorption, circular dichroism, fluorescence, and electron paramagnetic resonance spectra of mutant T54S show that this substitution has no influence on the formation and stability of the ferredoxin. The redox potential of this mutant, however, was lowered by 55 mV as compared with native adrenodoxin, indicating a role for this residue in redox potential modulation. Incorporation of the iron-sulfur cluster was not impaired in the T54A mutant, although structural features of the oxidized protein were considerably changed. The decreased stability of the T54A mutant as compared with the wild type and mutant T54S indicates that a hydrogen bond donor at this position stabilizes the protein. Both mutants have been shown to be functionally active. Replacement of threonine 54 by serine or alanine, however, leads to rearrangements at the recognition sites for its redox partners. This is reflected by decreased Km and Kd values of both mutants for the cytochromes P450, whereas only T54A displayed a decreased Km value in cytochrome c reduction. Substrate conversion was accelerated (2.2- and 2.4-fold for mutants T54A and T54S, respectively) in the CYP11B1-, but not in the CYP11A1-dependent reaction.

The interaction of bovine adrenodoxin with the chaperonin GroEL was investigated using sucrose density centrifugation and analytical ultracentrifugation. It could be clearly established that denatured mature adrenodoxin comigrated in a sucrose density gradient with the GroEL oligomer, indicating that a complex had been formed. Up to 2 moles of adrenodoxin/mol GroEL can be bound. From the partial concentrations, association constants of 4.3· 105M−1for the first adrenodoxin molecule and of 1.08· 105M−1for the second molecule to the complex, respectively, were calculated. Upon addition of the cochaperonin GroES and Mg-ATP to the adrenodoxin-GroEL complex, adrenodoxin was released,indicating a specific binding between GroEL and adrenodoxin.

Today industrial enterprises are often decentrally organized. The objective of this is to concentrate resources and capacities for the elaboration of specific tasks and the effective solution of problems. On the other hand, an effective information exchange is required between the various departments which are located at different sites. Existing networks and communication systems are not suitable to fulfil the necessary tasks. This paper reports on R&D activities which aim at overcoming the impediments to the integration of decentrally stuctured enterprises. The basis for the integration is a broadband communication system based on an optical fiber network which has already been installed in Berlin by the Deutsche Bundespost. The BERCIM project has been launched with the purpose of a conception and realization of distributed, fault tolerant CIM structures using such a communication system. The main tasks of the projekt are the conception and prototypical realization of distributed CIM applications and their informational linkage via this communication system. Therefor the communication kernel, transport system and a management information system have to be realized. For the communication system and the distributed CIM applications local and network overlapping fault tolerant procedures are to be considered.

The accessibility of the zebrafish embryo offers unique possibilities to study the mechanisms that guide growing axons in the developing vertebrate central nervous system. This review examines the current understanding of the pathfinding decisions by the growing axons, their substrates, and the recognition molecules that mediate axon–substrate interactions. The detailed analysis of pathfinding at the level of individual axons demonstrates that growing axons chose their paths unerringly. To do so, they rely on cues presented by their environment, in particular by neuroepithelial cells. Our understanding of the molecular bases of axon–substrate interactions is increasing. Members of most classes of recognition molecules have been identified in fish. Experimental evidence for the functions of these molecules in the zebrafish nervous system is accumulating. It has recently been shown that high molecular weight microtubule-associated proteins (HMWP) in the brain are present in dendrites and are absent from axons (Matus et al., 1981, Proc. Natl. Acad. Sci. U. S. A. 78:3010-3014). In this study we followed the appearance of both HMWP and tubulin in the neonatal rat cerebellum by immunoperoxidase staining, concentrating particularly on comparing Purkinje cell dendrites with adjacent granule cell axons. In the axons both immunohistochemically demonstrable tubulin and structurally distinct microtubules are present at all stages of development. By contrast the Purkinje cell dendrites contain better neither tubulin nor microtubules at early stages of their growth. However, immunoperoxidase staining showed that these developing dendrites are rich in HMWP which are particularly concentrated in the dendritic distal regions. HMWP are also present as patches beneath the surface membrane of the cell body before the emergence of dendrites. Based on this data and the well-documented ability of HMWP to promote microtubule assembly, we propose the hypothesis that during the initial phase of Purkinje neuron differentiation HMWP form part of a specialized cytoskeletal structure which acts as a specifier for the development of dendrites as opposed to axons.

The steady increase in the employment of robots in industrial production raised the demand for comfortable computer aided planning and off-line programming tools. Domain specific tools collect and compile expert’s knowledge. They are a step towards a specification and realisation of an integrated CIM concept.

To the radiometric thickness gage robotron 24024 manufactured by the VEB Robotron-Meβclektronik “Otto Schon” Dresden new sensors basing on absorption of beta and gamma radiation were developed. Altogether 21 modifications of one base type suitable for the solution of different measuring problems are available. In comparison to previous sensors the stability of the measuring signal has been advanced. Even, there are a diminished response time and enlarged thickness respectively mass per unit area ranges. Profile measurements with a higher resolution can, be carried out. Moreover, the radiation protection was improved.

An analysis of the electron transfer kinetics from the reduced [2Fe-2S] center of bovine adrenodoxin and its mutants to the natural electron acceptors, cytochromes P450scc and P45011beta, is the primary focus of this paper. A series of mutant proteins with distinctive structural parameters such as redox potential, microenvironment of the iron-sulfur cluster, electrostatic properties, and conformational stability was used to provide more detailed insight into the contribution of the electronic and conformational states of adrenodoxin to the driving forces of the complex formation of reduced adrenodoxin with cytochromes P450scc and P45011beta and electron transfer. The apparent rate constants of P450scc reduction were generally proportional to the adrenodoxin redox potential under conditions in which the protein-protein interactions were not affected. However, the effect of redox potential differences was shown to be masked by structural and electrostatic effects. In contrast, no correlation of the reduction rates of P45011beta with the redox potential of adrenodoxin mutants was found. Compared with the interaction with P450scc, however, the hydrophobic protein region between the iron-sulfur cluster and the acidic site on the surface of adrenodoxin seems to play an important role for precise complementarity in the tightly associated complex with P45011beta.

Site-directed mutagenesis of a domain (amino acids 299-338) aligning to the I-helix region of P450cam, P450BM3 and P450terp was used to investigate the different regioselectivities displayed in the hydroxylation reactions performed by human aldosterone synthase (P450aldo) and 11beta-hydroxylase (P45011beta). The two enzymes are 93% identical and are essential for the synthesis of mineralocorticoids and glucocorticoids in the human adrenal gland. Single replacement of P450aldo residues for P45011 beta-specific residues at positions 296, 301, 302, 320, and 335 only gave rise to slightly increased 11beta-hydroxylase activities. However, a L301P/A320V double substitution increased 11beta-hydroxylase activity to 60% as compared with that of P45011 beta. Additionally substituting Ala-320 for Val-320 of P45011 beta further enhanced this activity to 85%. The aldosterone synthase activities of the mutant P450aldo proteins were suppressed to a varying degree, with triple replacement mutant L301P/E302D/A320V retaining only 10% and double replacement mutant L301P/A320V retaining only 13% of the P450aldo wild type activity. These results demonstrate a switch in regio- and stereoselectivities of the engineered P450aldo enzyme due to manipulation of residues at three critical positions, and we attribute the determination of these features in P450aldo to the structure of a region analogous to the I-helix in P450cam.

A proline residue in the C-terminal part of the polypeptide chain is highly conserved among many [2Fe-2S] ferredoxins. To investigate the requirement for proline at this position, we constructed steric (4-108W), charged (4-108K), polar (4-108S) and non-polar (4-108A) truncated mutants of adrenodoxin and studied them for biological function and stability. Although the variants were expressed in Escherichia coli with a significantly lower yield compared with wild-type adrenodoxin, successful incorporation of the iron-sulfur cluster suggested their proper folding. Similar absorption, cD and EPR spectra indicated that the cluster environment was not affected by the mutations. No evidence for an essential role of Pro108 in determining the redox potential of adrenodoxin or its interactions with the redox partners was found. However, replacement of this residue results in a dramatic decrease in the overall protein stability. The differences in the Gibbs energy of unfolding at 37°C, Δ[ΔdG(37°C)], are -5.0, -7.8, -10.1 and -10.7 kJ/mol for 4-108A, 4-108S, 4-108W and 4-108K mutants, respectively, compared with 4-108P as a control. We conclude that the principle function of Pro108 is to stabilize adrenodoxin threefold: (i) through limitation of the conformation of the polypeptide chain in this region, (ii) through a hydrogen bond to Arg14 and (iii) favorable hydrophobic contacts.Keywords: ferredoxin/iron-sulfur cluster/protein unfolding/site-directed mutagenesis/thermodynamic stability

Computer modelling and site-directed mutagenesis were employed to investigate the structural basis for the regioselectivity of steroid hydroxylation and to determine whether point mutations of CYP11B1 and CYP11B2 can result in changes in the ratio of glucocorticoids/mineralocorticoids. Single replacement of CYP11B2 residues for CYP11B1-specific residues at positions 296, 301, 302, 320, and 335, belonging to the putative I-helix, gave rise to slightly increased 11β-hydroxylase activities. Replacement of 3 amino acids of CYP11B2 by the corresponding residues in CYP11B1 was sufficient to increase cortisol formation from about 5 to 85 of the CYP11B1 wild type level. The aldosterone synthase activities of the mutants were decreased to varying degrees indicating that point mutations at positions 296, 301, 302, and 335 could potentially cause hypoaldosteronism. Replacement of Val-320 of CYP11B1 by alanine, the corresponding residue found in CYP11B2, led to production of aldosterone by this mutant enzyme. This observation suggests that glucocorticoid-remediable hyperaldosteronism could also be due to point mutations located in the CYP11B1 sequence.

Summary Some 2045 male Chinese industrial workers aged 40–59 years living in the city of Wuhan in the People's Republic of China were examined for coronary risk factors in the year 1983. The investigation included a patient history, clinical examination, and ECG and laboratory tests, with special attention to serum lipids. After 5 years, a follow-up investigation of the study group was carried out. The results were compared to the similarly designed German GRIPS project.

Summary In Asia coronary heart disease mortality is almost 10 fold less frequent than in European countries. These findings attract interest to search for different risk factor patterns. From 1982 to 1985 epidemiologic surveys were carried out in China (n=2047), Japan (n=7580) and Germany (n=6052). Healthy, male subjects, aged 30 to 59 years were enrolled. The prevalence rate of hypertension for the Germans was 20% versus 18% for the Japanese, and 11% for the Chinese. About 69% of Chinese, 55% of Japanese, and 37% of Germans were smokers. About 66% of the Germans were overweight (BMI>25), 17% of the Japanese, and 11% of the Chinese. The highest risk group with cholesterol levels of >300 mg/dl included no Chinese subject, 0.1% of the Japanese, but 5% of the Germans. The lipoprotein profiles among the Japanese and the Chinese collectives typically showed antiatherosclerotic characteristics, whereas most Germans exhibited profiles which support development of atherosclerosis. About 36% of the participants from Germany showed 3 or more risk factors accumulated per person (Japan and China 5%). Multifactorial risk factor reduction for Germany is recommended.

One traditional use of radionuclides is unit area weight determination to the transmission and the backscattering methods. Noticeable progress in advancement of radiometric unit area weight measuring techniques has been and is being made above all thanks to the application of microelectronics. The use of microcomputers rendered possible the solution of the main problem of data processing - the determination of the unit area weight value from the detector signal with nonlinear calibration functions - as well as considerable increase of the efficiency of measuring systems.

The distributions of tubulin and high molecular weight microtubule-associated proteins (HMWPs) in brain were determined by immunoperoxidase histochemistry with specific antisera. Tubulin was found in microtubules of both neurons and glia and both axons and dendrites. HMWPs were found only in neurons where, in all cases examined, they were associated with dendritic microtubules but not those in axons. Both tubulin and HMWPs were also found in postsynaptic densities. These results indicate that brain microtubules vary in chemical composition. The preferential association of HMWPs with dendritic microtubules suggests that they may play a role in distinguishing between dendritic and axonal export routes from the cell body.

We have isolated cDNA clones for two forms of P-450s, P-450ka-1 and P-450ka-2, from a rabbit kidney cDNA library, using the cDNA for rabbit pulmonary cytochrome P-450p-2, a prostaglandin omega-hydroxylase (Matsubara, S., Yamamoto, S., Sogawa, K., Yokotani, N., Fujii-Kuriyama, Y., Haniu, M., Shively, J.E., Gotoh, O., Kusunose, E., and Kusunose, M. (1987) J. Biol. Chem.262, 13366-13371), as a hybridization probe. The cDNAs for P-450ka-1 and P-450ka-2 encode polypeptides of 510 and 511 amino acids, respectively, with sequence similarity of 85% and 87% to P-450p-2. The two deduced primary structures have 87% identity. RNA blot analysis demonstrated that the mRNAs for P-450ka-1 and P-450ka-2 formed single bands at approximately 3.0- and 2.6-kilobase positions, respectively. The mRNA for P-450ka-1 was expressed only in the liver and kidney and was increased remarkably in these tissues by the administration of clofibrate. In contrast, the mRNA for P-450ka-2 was expressed constitutively in the liver, kidney, and small intestine, but its transcription was enhanced only in the liver by clofibrate treatment. Thus, in spite of their high sequence similarity, these P-450 species have different modes of regulatory expression. Comparison of the nucleotide sequences among P-450ka-1, P-450ka-2, and P-450p-2 shows about 90% overall sequence similarity in any pair of the three sequences. Nucleotide replacements are not evenly distributed, but are rather biased. There is a region of approximately 500 base pairs of exceptionally high homology among the three sequences. These results indicate that the gene conversion event occurred during the evolutionary process of these genes.

Simulation of industrial robots has become an important means for the increased efficient application of robots. However, the simulations still do not reflect a desired reality. A breakthrough has been achieved by defining the realistic robot simulation (RRS) interface. It enables the integration of original controller software into simulation systems. Now the motion behaviour of simulated robots can match reality with high precision.
