Abstract:This article assesses a collaborative digital storytelling project between Chief Leschi Schools, a Puyallup Tribal K-12 school, and University of Washington students, teachers, and institutional leaders. This assessment emphasizes the importance of and examines how to build trust and facilitate co-planning in digital storytelling workshops centering Indigenous voices. Collaboration between Indigenous and non-indigenous students and teachers offered unconventional learning experiences and community celebrations of Indigenous youth. Inductive, qualitative research drawing from personal memos, interviews, focus groups, and surveys revealed the significance of co-planning and trust-building between participants of distinct social and formal education backgrounds. This analysis showcases how these different factors shaped and sustained the project, resulting in what the authors hope will be an enduring commitment. Recommendations include: (1) Emphasis on consistent and ongoing participation to cultivate trust and facilitate continuous communication; (2) purposeful engagement of students’ fundamental skills, presenting digital narratives that align with their interests and competencies; and (3) adopting an approach for non-indigenous academics and academic institutions of prioritizing and actively engaging in collaborative partnerships with Indigenous communities. The article underscores the significance of recognizing and resolving complex power and cultural dynamics between universities and tribes for successful project co-planning and implementation.

Background. Cathepsin D is a lysosomal protease overexpressed and abnormally secreted in most breast cancer cells. Several retrospective clinical studies have shown that cathepsin D is an independent prognostic factor in breast cancer that is associated with a higher risk of recurrence and a shorter overall survival. Methods. To the authors' knowledge, this is the first prospective study in which the prognostic value of cathepsin D was studied in 123 patients with primary breast cancer who were followed for 5 years between March 1985 and December 1990. Cathepsin D concentrations in breast cancer cytosol were measured using a solid-phase sandwich immunoenzymatic assay. The most significant prognostic factors were identified by multivariate analysis using the Cox proportional-hazards method. Results. The median value of cathepsin D was 20.8 pmol/mg of protein, which was approximately half than the median value found in subsequent assays done using a commercially available kit and reported in most retrospective studies. The cathepsin D status or level was correlated only with axillary lymph node involvement. A univariate analysis showed that high levels of cathepsin D (> 20 pmol/mg of protein) were correlated with a higher risk of recurrence and a shorter overall survival (P < 0.01 and P < 0.03, respectively). Using multivariate analysis, a high cathepsin D level, a negative progesterone receptor status, and lymph node involvement were the most important factors for predicting relapse-free survival (P = 0.02, P < 0.01, and P < 0.05, respectively). The cathepsin D level had prognostic value in patients with node-positive disease (P = 0.001) and appeared to be particularly useful in association with the progesterone receptor status by isolating a high-risk subgroup of patients (high cathepsin D level; negative progesterone receptor status). Conclusions. This first prospective study confirmed the prognostic value of the cathepsin D level in association with other major prognostic factors. The next step will be to determine whether the subset of patients with high cathepsin D levels would benefit from adjuvant therapy. Cancer 1993; 71:2006-12.

Heat stress is expected to intensify, since temperatures are projected to increase during the 21st century. We investigate the assumed co‐occurrence of annual extremes of temperature and one heat stress metric and assess the effect of relative humidity variations on the heat stress changes. We show in CMIP5 simulations that both extremes tend to co‐occur in Europe and northern America, when the conditions are the hottest and the driest. However, extreme seasons occur later for heat stress than temperature by up to 2 months within the tropics. By 2100, models project a drying that hampers the increase in heat stress extremes. Within northeastern Africa, the slight projected wettening enhances the warming effect on the heat stress changes and induces the maximum heat stress intensification. Models are generally able to represent the phasing of both extremes compared to observations, with large uncertainties over regions experiencing the greatest future heat stress intensification. Temperature and heat stress extremes are projected to intensify in the future. However, the occurrence and the changes in heat stress extremes also depend on humidity variations. We show that both extremes tend to co‐occur during the year in mid‐latitudes, but extremes of heat stress tend to occur later than temperature extremes in the tropics, when the relative humidity is annually higher. We also show in future climate simulations that the global projected drying that strengthens the increase in temperature extremes weakens the intensification of heat stress extremes. Nevertheless, this does not prevent risk for health, particularly in tropical areas. Abstract:As part of an initiative to enhance cultural ties between Quebec and Haiti, Brother Marie-Victorin (founder of the Jardin Botanique de Montréal) led a French Canadian mission that visited Quisqueya (island home to Haiti and the Dominican Republic) between December 1937 and January 1938, less than four years after the end of the US occupation of Haiti (1915–1934). The mission's primary aim was to reinforce Quebec and Haiti's positions as important centers for "French Civilization" in the Americas. Through his travelogue and 305 photos, Brother Marie-Victorin created a detailed record of, for the most part, Haiti's plants and society. He greatly appreciated the vitality of Haiti, its people, and various aspects of its society; however, as a member of the Brothers of La Salle Catholic Congregation, he also held negative perceptions of Vodou. Brother Marie-Victorin's journal entries reflect a certain naïveté, as well as uninformed reflections on Haitian society's ethnic and racial structures that seem to indicate that he and the other members of his delegation failed to examine the importance of Haiti's African heritage, particularly in the post-occupation period of nation-(re)building. In spite of these failings, the mission succeeded in offering Brother Marie-Victorin an introduction to Caribbean plants and critical botanical information needed for the seven subsequent expeditions he made to Cuba between 1938 and 1944.

Multifunctionalized biomaterials with enhanced bone antiresorptive properties were obtained through adsorption of a bisphosphonate, risedronate, on hydroxyapatite (HA) nanocrystals functionalized with zinc ions and polyethylenimine (PEI). Zn incorporation into the HA structure amounts to about 8 atom %, whereas the PEI content of the bifunctionalized material ZnHAPEIBP is about 5.9 wt %. The mechanism of adsorption and release of the bisphosphonate on ZnHAPEI is compared with that on ZnHA: risedronate adsorption isotherm on ZnHA is a Langmuir type, whereas the isotherm of adsorption on ZnHAPEI is better fitted with a Freundlich model and involved a higher amount of adsorbed risedronate. In vitro cell tests were carried out with a coculture model of osteoblasts and osteoclasts using a model simulating oxidative stress and consequent cellular senescence and osteoporosis by the addition of H2O2. The conditions utilized in the coculture model strongly affect osteoblast behavior. The results show that the composite materials allow an increase in osteoblast viability and recover impairment, revealing a novel characteristic of risedronate that is able to counteract the negative effects of oxidative stress when associated with differently functionalized samples. Both PEI and the bisphosphonate reduce osteoclast viability. Moreover, PEI, and even more risedronate, exerts an inhibitory effect on osteoclast activity.

ObjectiveSustained inflammation originating from macrophages is a driving force of fibrosis progression and resolution. Monoacylglycerol lipase (MAGL) is the rate-limiting enzyme in the degradation of monoacylglycerols. It is a proinflammatory enzyme that metabolises 2-arachidonoylglycerol, an endocannabinoid receptor ligand, into arachidonic acid. Here, we investigated the impact of MAGL on inflammation and fibrosis during chronic liver injury.DesignC57BL/6J mice and mice with global invalidation of MAGL (MAGL-/-), or myeloid-specific deletion of either MAGL (MAGLMye-/-), ATG5 (ATGMye-/-) or CB2 (CB2Mye-/-), were used. Fibrosis was induced by repeated carbon tetrachloride (CCl4) injections or bile duct ligation (BDL). Studies were performed on peritoneal or bone marrow-derived macrophages and Kupffer cells.ResultsMAGL-/-or MAGLMye-/-mice exposed to CCl4or subjected to BDL were more resistant to inflammation and fibrosis than wild-type counterparts. Therapeutic intervention with MJN110, an MAGL inhibitor, reduced hepatic macrophage number and inflammatory gene expression and slowed down fibrosis progression. MAGL inhibitors also accelerated fibrosis regression and increased Ly-6Clowmacrophage number. Antifibrogenic effects exclusively relied on MAGL inhibition in macrophages, since MJN110 treatment of MAGLMye-/-BDL mice did not further decrease liver fibrosis. Cultured macrophages exposed to MJN110 or from MAGLMye-/-mice displayed reduced cytokine secretion. These effects were independent of the cannabinoid receptor 2, as they were preserved in CB2Mye-/-mice. They relied on macrophage autophagy, since anti-inflammatory and antifibrogenic effects of MJN110 were lost in ATG5Mye-/-BDL mice, and were associated with increased autophagic flux and autophagosome biosynthesis in macrophages when MAGL was pharmacologically or genetically inhibited.ConclusionMAGL is an immunometabolic target in the liver. MAGL inhibitors may show promising antifibrogenic effects during chronic liver injury.

Cette technologie accroît les possibilités de traitement personnalisé pour les patients tout en abaissant les coûts de fabrication. Dans le cadre des reconstructions de mandibule par lambeau libre de fibula, des sociétés proposent, à l’heure actuelle, des guides de coupe obtenus par moulage à partir de scanner. Outre les délais de réalisation importants i.e. minimum 21jours, ces guides sont vendus à des coûts prohibitifs (2000 à 6000 euros). Conjointement avec le CNRS, des étudiants ingénieurs, une société biomédicale, les auteurs ont mis au point des guides de coupe et des fantômes de mandibule imprimés en 3 dimensions, livrables en 7jours et à moindre coût. La nouveauté de ce projet réside dans la rapidité d’élaboration des produits et la réduction importante de leur prix. Les auteurs proposent de décrire dans cette note technique la chaîne logistique de réalisation des fantômes et des guides de coupe ainsi que les résultats apportés. L’objectif est de permettre dans un avenir proche l’accès à cette technologie pour tous les patients.
