Three visible imaging diagnostics, including a tangential view, an extreme wide-angle view, and a downward-looking diagnostic, are installed on the HL-2M tokamak for high-performance plasmas. The entire field of view (FOV) covers up to 80% of the vacuum vessel. They aim at meeting the requirements of safe machine operation, plasma position identification, magnetohydrodynamic (MHD) instabilities study, and so on. The tangential and extreme wide-angle view diagnostics are based on the bilateral telecentric relay optics, while the downward-looking view diagnostic utilizes the imaging guide bundle with large format size, realizing the long optical paths. Plasma movement trends observed from the tangential view are checked with the magnetic measurements, and the optical and magnetic measurements for the plasma positions are consistent. Some filamentary structures (blobs) wandering at the extreme plasma edge at the low field side are also snapped by the high-speed camera. A highlight in this article is the field of the extreme wide-angle view, which reaches 112°. Without showing the distortion of conventional wide-angle lenses, nearly one-third of the vacuum vessel is in its eye. The evolution process of multifaceted asymmetric radiation from the edge (MARFE) phenomenon is recorded by the three views. It occurs as a bright belt on the inner side of the torus under the limiter configuration. The underlying cause of the MARFE is discussed.

ABSTRACTMounting evidence indicates the potential involvement of ATP-citrate lyase (ACLY) in the modulation of various cancer types. Nevertheless, the precise biological significance of ACLY in gastric cancer (GC) remains elusive. This study sought to elucidate the biological function of ACLY and uncover its influence on peritoneal metastasis in GC. The expression of ACLY was assessed using both real-time quantitative PCR and western blot techniques. To investigate the impact of ACLY on the proliferation of gastric cancer (GC) cells, colony formation and 5-ethynyl-2’-deoxyuridine (EdU) assays were performed. The migratory and invasive abilities of GC were evaluated using wound healing and transwell assays. Additionally, a bioinformatics analysis was employed to predict the correlation between ACLY and HIF-1A. This interaction was subsequently confirmed through a chromatin immunoprecipitation (ChIP) assay. ACLY exhibited upregulation in gastric cancer (GC) as well as in peritoneal metastasis. Its overexpression was found to facilitate the proliferation and metastasis of GC cells in both in vitro and in vivo experiments. Moreover, ACLY was observed to play a role in promoting angiogenesis and epithelial-mesenchymal transition (EMT). Notably, under hypoxic conditions, HIF-1A levels were elevated, thereby acting as a transcription factor to upregulate ACLY expression. Under the regulatory influence of HIF-1A, ACLY exerts a significant impact on the progression of gastric cancer, thereby facilitating peritoneal metastasis.

Due to the deterioration of the crosswind-protection performance of the original earth embankment windbreak walls on the train with increased speed, researching the influence of the heightened windbreak walls on aerodynamic performances of trains as well as the protection of the catenary is necessary for optimizing the windproof performance. The improved delayed detached eddy simulation based on the shear stress transport (SST) κ-ω turbulence model was employed to simulate flow structures. Furthermore, the effect on the displacement of the catenary was investigated using the finite element method (FEM). The numerical case was validated against the field tests. The results show that the aerodynamic performances of the train were improved as the windbreak wall was heightened by 0.5 m. Whereas, further heightening the windbreak wall worsened the aerodynamic performance. When the windbreak wall was heightened by 0.5 and 1 m, the wind speed of the catenary was reduced by 31.17% and 53.87%, and the displacement of the catenary was reduced by 48.09% and 74.11%, respectively, compared with that of the original windbreak wall. These results reveal that the wind-protection performance on the train and catenary can be both improved by reasonably heightening windbreak walls.

Citrullination is a key post-translational modification (PTM) that affects protein structures and functions. Although it has been linked to various biological processes and disease pathogenesis, the underlying mechanism remains poorly understood due to a lack of effective tools to enrich, detect, and localize this PTM. Herein, we report the design and development of a biotin thiol tag that enables derivatization, enrichment, and confident identification of citrullination via mass spectrometry. We perform global mapping of the citrullination proteome of mouse tissues. In total, we identify 691 citrullination sites from 432 proteins which represents the largest data set to date. We discover novel distribution and functions of this PTM. This study depicts a landscape of protein citrullination and lays the foundation for further deciphering their physiological and pathological roles.

• The disturbance region update method (DRUM) is introduced to enhance the flow field simulation speed in the hypersonic CHT problem. • A new heat flux estimate method within a coupled time step is proposed to expand the available coupled time step. • DRUM reduces the time for flow field simulation by over 60 %, and the heat flux estimate method could effectively decrease the total coupled steps by increasing coupled time step while maintaining accuracy. This research proposes acceleration algorithms for the coupled numerical simulation of the long-time hypersonic conjugate heat transfer problem from two perspectives. First, the newly developed disturbance region update method (DRUM) is introduced to enhance the convergence speed of the steady flow field. The numerical investigations reveal that, in the context of hypersonic CHT issues, the disturbance prompted by variations in wall temperature remains confined post the detached shock wave. The implementation of DRUM has been shown to substantially diminish the computational demand per iteration and the total iterations required for convergence. DRUM could reduce the time needed to calculate the flow field by over 60 % for a typical CHT case. Second, a new aerodynamic heat flux estimate method is introduced based on the local wall temperature within a coupled time step to increase the coupled time step. Combining this method with the adaptive coupled time step method, a long-time CHT problem with a total time of 1100s requires only 25 coupled time steps, much less than the 220 steps needed for the existing method. Meanwhile, the average error of the obtained solid temperature field, when compared to the temperature obtained from an accurate simulation with a fixed small coupled time step, is maintained below 0.22 %. Wind tunnel tests for trains under large yaw angles are usually limited due to the width of the wind tunnel. Therefore, the leading car and a downstream dummy vehicle model are often employed instead of a real train, but there are no clear regulations regarding the shape of the end of the dummy vehicle. This paper studied the impact of the trailing edge shape of the downstream dummy vehicle on train aerodynamics subjected to crosswind based on the shear-stress-transport k-ωturbulence model of the delayed detached eddy simulation. Three types of end shapes, namely the rectangular end shape, the arc end shape, and the streamlined end shape were chosen for comparison, and the simulation results of the three-car-group train were selected as the benchmark. First, the reliability of the numerical method was validated by wind tunnel tests. Then, the aerodynamic coefficients under yaw angles of 0°–60° and the surface pressure distributions and flow structures around the train under the yaw angle of 60° of the head cars with different end shapes were compared and analyzed.

It is of great significance to reveal the microevolution mechanism of welded structures during thermo-mechanical coupling to improve the welding quality. In this paper, a random microcrystalline structure model for welds is established by the Voronoi tessellation method. According to the nanoindentation results, heterogeneous grains are produced. A welding workpiece model with statistical significance is established. On this basis, the Python script and the birth and death element method are used to realize the transient growth of a weld, and a thermo-mechanical coupling model for the SUS301L-HT stainless steel metal inert gas welding process is established. The temperature field and thermal stress field are calculated. The calculation shows that the thermal stresses along the growth direction of the weld area are in the form of a “trapezoid,” and the stresses at both ends are small. The stress in the vertical direction of the weld has a single peak, and the peak appears in the center of the weld. The stress distribution of the model that considers heterogeneous grains is obviously inhomogeneous compared with that of the traditional model. The thermal stress distribution in the weldment is obviously inhomogeneous due to the heterogeneous grains, the stresses at the boundaries of the adjacent grains in the weldment change abruptly. It is found that the greater the difference in the mechanical properties between grains is, the more obvious the change.

Nonalcoholic steatohepatitis (NASH) is rising in prevalence in the United States and is a growing cause of hepatocellular carcinomas (HCCs). Site-specific glycan heterogeneity on glycoproteins has been shown as a potential diagnostic biomarker for HCC. Herein, we have performed a comprehensive screening of site-specific N-glycopeptides in serum haptoglobin (Hp), a reporter molecule for aberrant glycosylation in HCC, to characterize glycopeptide markers for NASH-related HCCs. In total, 70 NASH patients (22 early HCC, 15 advanced HCC, and 33 cirrhosis cases) were analyzed, with Hp purified from 20 μL of serum in each patient, and 140 sets of mass spectrometry (MS) data were collected using liquid chromatography coupled with electron-transfer high-energy collisional dissociation tandem MS (LC-EThcD-MS/MS) for quantitative analysis on a novel software platform, Byos. Differential quantitation analysis revealed that five N-glycopeptides at sites N184 and N241 were significantly elevated during the progression from NASH cirrhosis to HCC (p< 0.05). Receiver operating characteristic (ROC) curve analysis demonstrated that the N-glycopeptides at sites N184 and N241 bearing a monofucosylated triantennary glycan A3G3F1S3 had the best diagnostic performance in detection of early NASH HCC, area under the curve (AUC) = 0.733 and 0.775, respectively, whereas α-fetoprotein (AFP) had an AUC of 0.692. When combined with AFP, the two panels improved the sensitivity for early NASH HCC from 59% (AFP alone) to 73% while maintaining a specificity of 70%, based on the optimal cutoff. Two-dimensional (2-D) scatter plots of the AFP value and N-glycopeptides showed that these N-glycopeptide markers detected 58% of AFP-negative HCC patients as distinct from cirrhosis. These site-specific N-glycopeptides could serve as potential markers for early detection of HCC in patients with NASH-related cirrhosis.

ObjectiveThe aim of the present work was to investigate the expression of nitric oxide synthase 2 (iNOS/ NOS2) in colorectal and gastric cancers and evaluate its association with patient’s prognosis by integrated bioinformatics analysis.

Crustaceans are commonly used model organisms to study neuromodulation. Despite numerous reported crustacean neuropeptide families and their functions, there has been no report on neuropeptide glycosylation. This is in part due to a lack of sensitive methods that enable deciphering this intricate low-abundance post-translational modification, even though glycosylation has been shown to play an important role in neuromodulation. Here, we describe the discovery of glycosylated neuropeptides with an enrichment-free approach, taking advantage of signature oxonium ions produced in higher-energy collision dissociation (HCD) MS/MS spectra. The detection of the oxonium ions in the HCD scans suggests glycan attachment to peptides, allowing electron-transfer/higher-energy collision dissociation (EThcD) to be performed to selectively elucidate structural information of glycosylated neuropeptides that are buried in nonglycosylated peptides. Overall, 4 N-linked and 14 O-linked glycosylated neuropeptides have been identified for the first time in the crustacean nervous system. In addition, 91 novel putative neuropeptides have been discovered based on the collected HCD scans. This hybrid approach, coupling a shotgun method for neuropeptide discovery and targeted strategy for glycosylation characterization, enables the first report on glycosylated neuropeptides in crustaceans and the discovery of additional neuropeptides simultaneously. The elucidation of novel glycosylated neuropeptides sheds light on the crustacean peptidome and offers novel insights into future neuropeptide functional studies.

To reduce the impact of a mountain ridge on the crosswind flow field of a specific section of the Lanzhou-Xinjiang high-speed railway, a portion of the mountain ridge was removed to increase the distance from the windbreak. To verify the effects of this flow optimization measure, the wind speed and wind angle were tested in this mountain ridge region. In this paper, under the actual conditions of this specific case study, the average and transient wind characteristics were analyzed based on the test results. Then, based on the actual terrain model, using the computational fluid dynamics (CFD) method with two inlet boundary conditions, namely, constant wind and exponential wind, the results obtained from the two boundary conditions were validated. Furthermore, the visualized flow structures and wind speed distributions along the railway under both boundary conditions were compared. Finally, along the railway, the impacts of different terrain types on the flow field of the railway were compared.

ABSTRACTDouble unit trains running at high speeds may create additional aerodynamic challenges due to two streamlined structures with close proximity, exploring the aerodynamic performance of double unit trains is now critical. In this study, detached eddy simulation (DES) approach was employed to study the aerodynamic performance and the nearby flow patterns of a double unit train, whose results were compared and analyzed with that of a single-unit train with a same length. The results showed that the coupling method could change the aerodynamic drag on each car and tended to increase the overall drag of the double unit train. The lift force of the front car near the coupler was significantly increased. Similar slipstream distributions were found around the front half single and double-unit train except in a region close to the coupler. Due to the coupling structure, the slipstream of the rear half of double unit train was much stronger compared to single unit train. The vortex region behind the double-unit train was much wider than that of the single-unit train and was accompanied by greater vortex-shedding.

A certain gap spacing between adjacent vehicles is usually inevitable in wind tunnel force tests of high-speed trains under no crosswind, which may affect the wind tunnel test results. Thus, to understand the influence of gap spacings on the train aerodynamics, the aerodynamic drag, pressure distributions and airflow structures of 1/8th-scale high-speed train models with gap spacings of 0, 5, 8, 10, 20, and 30 mm were studied using RANS based on SST k-ω turbulence model. The simulation method was verified by the wind tunnel experiment data. The results show that the gap spacing significantly affects the airflow structure around inter-car gap and aerodynamic resistances of train models. For the high-speed train model scaled at 1/8th at zero yaw, compared with gap spacing of 0 mm, the gap spacings lead to a significant reduction in the aerodynamic drag of the head car and an increase in that of the tail car, whereas which of the middle car is not significant. The maximum difference of the drag coefficient of the entire train model is smaller than 2.0%. When the gap spacing does not exceed 8 mm, the discrepancies of the drag coefficients of three cars are within 6.15%.

Exergy destructions in laminar premixed syngas flames, with varied fuel compositions and changed pressures, were numerically studied. Chemical reactions are found to be the dominant source for exergy destruction, followed by heat conduction and mass diffusion. As a result of the trade-off between the increased exergy destruction rates and narrowed flame thickness, the total exergy destruction increases by about 1% with hydrogen enrichment, but is nearly unchanged with pressure elevation. Specifically, with hydrogen enrichment, the exergy destruction from heat conduction decreases due to the narrowed flame thickness, whereas those from mass diffusion and chemical reactions increase because of higher diffusivity of H2, H and H2O and higher reactivity of H2than CO, respectively. In contrast, with pressure elevation, the higher temperature gradient and mole fraction gradients result in increased exergy destruction from heat conduction, but the exergy destruction from chemical reactions decrease due to the narrowed flame thickness.

Mannose-6-phosphate (M6P) glycosylation is an important post-translational modification (PTM) and plays a crucial role in transferring lysosomal hydrolases to lysosome, and is involved in several other biological processes. Aberrant M6P modifications have been implicated in lysosomal storage diseases and numerous other disorders including Alzheimer’s disease and cancer. Research on profiling of intact M6P glycopeptides remains challenging due to its extremely low stoichiometry. Here we propose a dual-mode affinity approach to enrich M6P glycopeptides by dual-functional titanium(IV) immobilized metal affinity chromatography [Ti(IV)-IMAC] materials. In combination with state-of-the-art mass spectrometry and database search engine, we profiled 237 intact M6P glycopeptides corresponding to 81 M6P glycoproteins in five types of tissues in mouse, representing the first large-scale profiling of M6P glycosylation in mouse samples. The analysis of M6P glycoforms revealed the predominant glycan substrates of this PTM. Gene ontology analysis showed that overrepresented M6P glycoproteins were lysosomal-associated proteins. However, there were still substantial M6P glycoproteins that possessed different subcellular locations and molecular functions. Deep mining of their roles implicated in lysosomal and nonlysosomal function can provide new insights into functional roles of this important yet poorly studied modification.

This paper investigates the effect of a wind barrier on the aerodynamic performance of a train–bridge system under crosswind using a numerical simulation method. The studied bridge is a long-span cable-stayed bridge with a flat steel box girder, located in Chongqing, China. The flow field around the train–bridge system with and without a wind barrier is numerically simulated. Wind barrier porosities varying from 10 to 60% are evaluated. The tricomponent coefficients of the train, bridge, and train–bridge system are obtained and investigated in detail. The effect of the wind barrier on the aerodynamics of the train–bridge system is revealed through the determination of the aerodynamic forces, pressure mapping, and flow visualization. The results show that a wind barrier successfully decreases the mean velocity above the girder and consequently decreases the drag force and moment on the train; however, the wind barrier also significantly increases the drag force on the girder. Therefore, installation of a wind barrier improves the running safety of the train but is detrimental to the wind resistance of the bridge. Additionally, the efficiency of the wind barrier depends on the porosity. A lower porosity improves the train safety but is more detrimental to the bridge safety. An optimal wind barrier porosity of 30% is obtained based on the aerodynamic forces of both the train and the bridge. Compared to a train–bridge system without a wind barrier, the drag force and moment on the train decrease by 66.1 and 62.9%, respectively; the drag force on the bridge girder increases to 0.86, and the drag force on the train–bridge system equals that without the wind barrier.

Absolute quantification in targeted proteomics is challenging due to a variety of factors, including low specificity in complex backgrounds, limited analytical throughput, and wide dynamic range. To address these problems, we developed a hybrid offset-triggered multiplex absolute quantification (HOTMAQ) strategy that combines cost-effective mass difference and isobaric tags to enable simultaneous construction of an internal standard curve in the MS1precursor scan, real-time identification of peptides at the MS2level, and mass offset-triggered accurate quantification of target proteins in synchronous precursor selection (SPS)-MS3spectra. This approach increases the analytical throughput of targeted quantitative proteomics by up to 12-fold. The HOTMAQ strategy was employed to verify candidate protein biomarkers in preclinical Alzheimer’s disease with high accuracy. The greatly enhanced throughput and quantitative performance, paired with sample flexibility, makes HOTMAQ broadly applicable to targeted peptidomics, proteomics, and phosphoproteomics.

Intact N-glycopeptide analysis remains challenging due to the complexity of glycopeptide structures, low abundance of glycopeptides in protein digests, and difficulties in data interpretation/quantitation. Herein, we developed a workflow that involved advanced methodologies, the EThcD-MS/MS fragmentation method and data interpretation software, for differential analysis of the microheterogeneity of site-specific intact N-glycopeptides of serum haptoglobin between early hepatocellular carcinoma (HCC) and liver cirrhosis. Haptoglobin was immunopurified from 20 μL of serum in patients with early HCC, liver cirrhosis, and healthy controls, respectively, followed by trypsin/GluC digestion, glycopeptide enrichment, and LC-EThcD-MS/MS analysis. Identification and differential quantitation of site-specific N-glycopeptides were performed using a combination of Byonic and Byologic software. In total, 93, 87, and 68 site-specific N-glycopeptides were identified in early HCC, liver cirrhosis, and healthy controls, respectively, with high confidence. The increased variety of N-glycopeptides in liver diseases compared to healthy controls was due to increased branching with hyper-fucosylation and sialylation. Differential quantitation analysis showed that 5 site-specific N-glycopeptides on sites N184 and N241 were significantly elevated in early HCC compared to cirrhosis (p< 0.05) and normal controls (p≤ 0.001). The result demonstrates that the workflow provides a strategy for detailed profiles of N-glycopeptides of patient samples as well as for relative quantitation to determine the level changes in site-specific N-glycopeptides between disease states.

Ultraviolet (UV) radiation has a variety of impacts including the health of humans, the production of crops, and the lifetime of buildings. Based on the photovoltaic effect, self-powered UV photodetectors can measure and monitor UV radiation without any power consumption. However, the current low photoelectric performance of these detectors has hindered their practical use. In our study, a super-high-performance self-powered UV photodetector based on a GaN/Sn:Ga2O3pn junction was generated by depositing a Sn-doped n-type Ga2O3thin film onto a p-type GaN thick film. The responsivity at 254 nm reached up to 3.05 A/W without a power supply and had a high UV/visible rejection ratio of R254 nm/R400 nm= 5.9 × 103and an ideal detectivity at 1.69 × 1013cm·Hz1/2·W–1, which is well beyond the level of previous self-powered UV photodetectors. Moreover, our device also has a low dark current (1.8 × 10–11A), a high Iphoto/Idarkratio (∼104), and a fast photoresponse time of 18 ms without bias. These outstanding performance results are attributed to the rapid separation of photogenerated electron–hole pairs driven by a high built-in electric field in the interface depletion region of the GaN/Sn:Ga2O3pn junction. Our results provide an improved and easy route to constructing high-performance self-powered UV photodetectors that can potentially replace traditional high-energy-consuming UV detection systems.

Ga2−xFexO3(GFO) bulks with xfrom 0.7 to 1.3 have been fabricated using the classic solid-state route. The structural, optical, and magnetic properties have been investigated systematically. X-ray diffraction spectra and FULLPROF profile fitting indicate that GFO bulks belong to the orthorhombic structure with the space group Pc21n. Phase separation appears at the Fe content of x= 1.3. The optical bandgap decreases almost linearly with the increase of iron content, which means that the bandgap of GFO bulks can be controlled by adjusting the Fe content in the samples. The magnetic property measurements suggest that GFO is ferromagnetic, and the magnetic properties are enhanced compared with other reported works, exhibiting the application in ferromagnetic semiconductors devices.

Slipstreams are generated when high-speed trains pass through the open air causing safety threat to passengers, trackside workers and infrastructure. This study calculates the slipstream induced by trains with different longitudinal section lines using a detached-eddy simulation. The slipstream velocities and pressure at various lateral distances from the centre of the rail position and various vertical distances from the top of the rail position are calculated at a Reynolds number of 1.8 × 106, and the flow field around the trains is analysed. The results of the calculation are compared with the results of a full-scale test to validate the numerical method adopted in this work. The results demonstrate that the variations in the slipstream velocities induced by the four types of trains are similar as are the variations in the trackside pressures. The amplitudes of the slipstream velocities and trackside pressures are different due to the influence of the longitudinal section line, and both the slipstream velocity and the trackside pressure increase with the slope of the longitudinal section line. The slipstream velocity and trackside pressure decrease with increasing distance from the centre of the rail and the top of the rail. The large difference in the slipstream induced by the four types of trains occurs in regions where the distance from the centre of the rail is greater than 2.5 m and the distance from the top of the rail is greater than 1.5 m, and those regions are also the areas where platform passengers and track infrastructure are located. The results demonstrate that the slipstream in those regions can be reduced by adopting relatively lower slopes of the longitudinal section line.

High-quality cerium-doped β-Ga_2O_3 (Ga_2O_3:Ce) thin films could be achieved on (0001)α-Al_2O_3 substrates using a pulsed-laser deposition method. The impact of dopant contents concentration on crystal structure, optical absorption, photoluminescence, and photoelectric properties has been intensively studied. X-ray diffraction analysis results have shown that Ga_2O_3:Ce films are highly (2¯01) oriented, and the lattice spacing of the (4¯02) planes is sensitive to the Ce doping level. The prepared Ga_2O_3:Ce films show a sharp absorption edge at about 250 nm, meaning a high transparency to deep ultraviolet (DUV) light. The photoluminescence results revealed that the emissions were in the violet–blue–green region, which are associated with the donor–acceptor transitions with the Ce^3+ and oxygen vacancies related defects. A simple DUV photodetector device with a metal–semiconductor–metal structure has also been fabricated based on Ga_2O_3:Ce thin film. A distinct DUV photoresponse was obtained, suggesting a potential application in DUV photodetector devices.

ABSTRACTIn areas with strong wind, windbreaks are built along railways to reduce the impact of wind on trains. However, because of the restrictions imposed by actual terrain, windbreak structures are often not uniform, such as from a cutting to an embankment, resulting in a discontinuous transition region. When a train runs through this region, a distinct yawing phenomenon occurs. This study numerically explored the aerodynamic features of a train running through a rectangular windbreak transition region. The variations in the pressure, side force, and moment of a train were analyzed, and the flow field features were clarified. Furthermore, the yawing motion of the car body with time was described. Finally, based on EN14067-6, the critical wind speed was obtained using the safety assessment of a train running through a windbreak transition region.

Rapid identification of edible oil and swill-cooked dirty oil is a challenging and important task in the field of food safety. The main object of this investigation was to distinguish edible oil (QO) and swill-cooked dirty oil (SO) by employing near-infrared (NIR) spectroscopy and the sparse representation classification (SRC) method. Because of the diversity and uncertainty of the species in swill-cooked dirty oil, building a classification model based on NIR spectroscopy faces the problems of complex systems and small numbers of samples. To improve the stability and accuracy of the identification, in the SRC method, the redundant dictionaries for QO and SO were trained, and the sparse representation coefficients for spectra in a validation set under both dictionaries were calculated. Then the spectra in the validation set were reconstructed with the sparse representation coefficients and the corresponding dictionary. Finally, the reconstruction errors under the QO and SO dictionaries were used as indicators for classification. Moreover, a simplified SRC algorithm (SRC-S) that directly uses the calibration set spectra as redundant dictionaries was proposed, and this removed the dictionary training process and avoided information loss during training. Compared with linear discriminant analysis (LDA) and partial least squares discriminant analysis (PLS-DA), the experimental results showed that the SRC-S outperformed SRC, and it reached a maximum classification accuracy of 95.37%, which proved that SRC-S and NIR spectroscopy can distinguish QO and SO.

An extended canonical variates analysis (ECVA) method dealing with multicollinear data of infrared spectrum and a singular within-group covariance matrix is proposed for wine origin discrimination. The wine-origin is classified by infrared spectrum and the chemometrics methods. Comparing the classification results of the k-nearest neighbor algorithm (KNN), partial least-squares discriminant analysis (PLS-DA), and principal component analysis (PCA) methods, the ECVA-KNN method reaches the optimal correctness of 97.73%. The experiment results prove that the ECVA method is fit for wine origin discrimination and can be effectively applied to the qualitative analysis of the collinear infrared spectrum.

Information obtained by atomic force microscopy (AFM) depends strongly on the kind of probe or tip used; therefore, probe and tip effects have to be taken into account when verifying or interpreting the data acquired. In many papers, double‐tip effects have been mentioned while other research was done; however, there are only a few special reports on double‐ or triple‐tip effects, especially double‐probe effects. In our paper, metaphase chromosomes of Chinese hamster ovary (CHO) cells, aggregates of pectin molecules, membrane surface of mouse embryonic stem cells, and R‐phycoerythrin‐conjugated immunoglobulin G complexes were imaged by AFM with high‐quality probes, double‐probe cantilever, and doubletip and triple‐tip probes, respectively, in order to determine double‐probe, double‐tip, and triple‐tip effects during AFM scanning. We found that the double‐probe, double‐tip, and triple‐tip effects share the same principle, and that these effects correlate with distance and height differences between probes of double‐probe cantilever or tips of double‐tip or multiple‐tip probes. Since many other factors influence double‐probe or double‐tip effects, more in‐depth studies must be undertaken. However, this initial research will make all users of AFM techniques aware of double‐probe and double‐tip or triple‐tip effects during AFM scanning and aid in verifying or interpreting the data acquired.

•We found the circAFF2/miR-6894-5p/ANTXR 1/axis in gastric cancer by dual-luciferase reporter assay.•We detected the expression and functions of circAFF2, miR-6894-5p, and ANTXR 1 in gastric cancer.•CircAFF2 may act as an oncogene in gastric cancer and exert its effects via miR-6894-5p/ANTXR 1 signaling.

Site-specific O-glycoproteome mapping in complex biological systems provides a molecular basis for understanding the structure–function relationships of glycoproteins and their roles in physiological and pathological processes. Previous O-glycoproteome analysis in cerebrospinal fluid (CSF) focused on sialylated glycoforms, while missing information on other glycosylation types. In order to achieve an unbiased O-glycosylation profile, we developed an integrated strategy combining universal boronic acid enrichment, high-pH fractionation, and electron-transfer and higher-energy collision dissociation (EThcD) for enhanced intact O-glycopeptide analysis. We applied this strategy to analyze the O-glycoproteome in CSF, resulting in the identification of 308 O-glycopeptides from 110 O-glycoproteins, covering both sialylated and nonsialylated glycoforms. To our knowledge, this is the largest data set of O-glycoproteins and O-glycosites reported for CSF to date. We also developed a peptidomics workflow that utilized the EThcD and a three-step database searching strategy for comprehensive PTM analysis of endogenous peptides, including N-glycosylation, O-glycosylation, and other common peptide PTMs. Interestingly, among the 1411 endogenous peptides identified, 89 were O-glycosylated, and only one N-glycosylated peptide was found, indicating that CSF endogenous peptides were predominantly O-glycosylated. Analyses of the O-glycoproteome and endogenous peptidome PTMs were also conducted in the CSF of MCI and AD patients to provide a landscape of glycosylation patterns in different disease states. Our results showed a decreasing trend in fucosylation and an increasing trend of endogenous peptide O-glycosylation, which may play an important role in AD progression.

The fatigue life of the bearing can be predicted effectively by combining the multifield coupling condition of the working process with the internal microstructure. In this paper, a bearing fluid lubrication model is established based on the computational fluid dynamics method, and the calculation result is coupled to the thermal-solid model by the grid mapping method. Through this method, a bearing thermal-fluid-solid multifield coupling dynamics model is established to obtain the evolution information of bearings. Simultaneously, the micro-characterization experiments of SKF6406 bearing (M50 bearing steel) were carried out by electron backscatter diffraction, scanning electron microscope and nano-indentation. The microstructure model of bearing containing micro-inclusions was established by the Voronoi method. On this basis, the microscopic stress distribution characteristics and the motion law of bearing microstructure under the multifield coupling action are solved. In the end, the fatigue life prediction method of rolling bearing was established.

As the body fluid that directly interchanges with the extracellular fluid of the central nervous system (CNS), cerebrospinal fluid (CSF) serves as a rich source for CNS-related disease biomarker discovery. Extensive proteome profiling has been conducted for CSF, but studies aimed at unraveling site-specific CSF N-glycoproteome are lacking. Initial efforts into site-specific N-glycoproteomics study in CSF yield limited coverage, hindering further experimental design of glycosylation-based disease biomarker discovery in CSF. In the present study, we have developed an N-glycoproteomic approach that combines enhanced N-glycopeptide sequential enrichment by hydrophilic interaction chromatography (HILIC) and boronic acid enrichment with electron transfer and higher-energy collision dissociation (EThcD) for large-scale intact N-glycopeptide analysis. The application of the developed approach to the analyses of human CSF samples enabled identifications of a total of 2893 intact N-glycopeptides from 511 N-glycosites and 285 N-glycoproteins. To our knowledge, this is the largest site-specific N-glycoproteome dataset reported for CSF to date. Such dataset provides molecular basis for a better understanding of the structure–function relationships of glycoproteins and their roles in CNS-related physiological and pathological processes. As accumulating evidence suggests that defects in glycosylation are involved in Alzheimer's disease (AD) pathogenesis, in the present study, a comparative in-depth N-glycoproteomic analysis was conducted for CSF samples from healthy control and AD patients, which yielded a comparable N-glycoproteome coverage but a distinct expression pattern for different categories of glycoforms, such as decreased fucosylation in AD CSF samples. Altered glycosylation patterns were detected for a number of N-glycoproteins including alpha-1-antichymotrypsin, ephrin-A3 and carnosinase CN1 etc., which serve as potentially interesting targets for further glycosylation-based AD study and may eventually lead to molecular elucidation of the role of glycosylation in AD progression.

The metastable cubic ?-(Ga1?xMnx)2O3thin films are successfully obtained at high growth temperature by laser molecular beam epitaxy technology. The optoelectronic properties of the solar blind Schottky-type photodetectors (PDs) based on ?-(Ga1?xMnx)2O3thin films are reported for the first time. In this experimental system, the ?-(Ga0.96Mn0.04)2O3PD exhibits the highest light-to-dark ratio (LDR) of 6.89 × 103, which is two orders of magnitude higher than the pure ?-Ga2O3PD prepared under the same condition. In addition, it shows a fast photoresponse decay speed of about 0.081 s. The results suggest that Mn element is expected to be one of the promising dopants to induce and stabilize the metastable ?-Ga2O3, as well as optimize the optoelectronic performance of photodetectors.

ABSTRACTSequence variants (SVs) resulting from unintended amino acid substitutions in recombinant therapeutic proteins have increasingly gained attention from both regulatory agencies and the biopharmaceutical industry given their potential impact on efficacy and safety. With well-optimized production systems, such sequence variants usually exist at very low levels in the final protein products due to the high fidelity of DNA replication and protein biosynthesis process in mammalian expression systems such as Chinese hamster ovary cell lines. However, their levels can be significantly elevated in cases where the selected production cell line has unexpected DNA mutations or the manufacturing process is not fully optimized, for example, if depletion of certain amino acids occurs in the cell culture media in bioreactors. Therefore, it is important to design and implement an effective monitoring and control strategy to prevent or minimize the possible risks of SVs during the early stage of product and process development. However, there is no well-established guidance from the regulatory agencies or consensus across the industry to assess and manage SV risks. A question frequently asked is: What levels of SVs can be considered acceptable during product and process development, but also have no negative effects on drug safety and efficacy in patients? To address this critical question, we have taken a holistic approach and conducted a comprehensive sequence variant analysis. To guide biologic development, a general SV control limit of 0.1% at individual amino acid sites was proposed and properly justified based on extensive literature review, SV benchmark survey of approved therapeutic proteins, and accumulated experience on SV control practice at Regeneron.

In order to establish the rapid traffic economic circle in Xinjiang province, there is a plan to operate the EMU (Electric Multiple Units) in Nanjiang normal-speed railway line. But there is few standard and experience that the EMU running on the normal-speed railway under strong wind. Therefore, the present work conducts a full-scale test to research the safety of the EMU running on the normal-speed railway to obtain the train aerodynamic and system dynamic performance. Furthermore, the part of deteriorate reasons of train aerodynamic performance is studied by the CFD (Computational Fluid Dynamics) method. In the next step, based on the experimental and CFD results, the disadvantage of current windproof facility will be found and improved to satisfy the requirement of the operation safety of the EMU.

Red LEDs based on Eu:Ga2O3/GaAs heterojunctions were fabricated using a pulsed laser deposition method. Eu-related luminescence originating from the 5D0-7F2transition of Eu3+was observed at about 611 nm. Bright red light can be observed with the naked eye at 6.0 V. Under forward bias, the electrons will first transit to the defect-related energy levels and then recombine with holes. The indirect recombination of carriers in the Ga2O3host could transfer energy to the Eu ions. This work will provide an outlook for future low driven voltage and small-scale displays.
